The situation of tuberculosis and M. tuberculosis (MTB) drug resistance is severe in China.Among the MTB isolates, 27.8% were resistant to one primary antituberculosis drug at least. Isoniazid (INH) is one of the first-line anti-TB drugs. The rates of primary resistance and acquired resistance to INH were 11.0% and 31.0%, respectively. 目的研究已证实Oct4基因可调控肿瘤细胞上皮间质转化(epithelial-mesenchymal transition,EMT)。本研究探讨沉默Oct4的亚型Oct4B1能否逆转结直肠癌耐奥沙利铂细胞株SW620/OxR的EMT。方法分别采用带G418抗性的Oct4B1-shRNA质粒和阴性对... 目的研究已证实Oct4基因可调控肿瘤细胞上皮间质转化(epithelial-mesenchymal transition,EMT)。本研究探讨沉默Oct4的亚型Oct4B1能否逆转结直肠癌耐奥沙利铂细胞株SW620/OxR的EMT。方法分别采用带G418抗性的Oct4B1-shRNA质粒和阴性对照质粒转染SW620/OxR细胞,转染成功后通过G418筛选建立稳定转染细胞株,分别称为实验组(RNAi组)及对照组(Ctrl组)。对两组细胞作如下检测,(1)RT-qPCR检测Oct4B1的mRNA水平;(2)划痕实验检测细胞迁移能力;(3)Transwell小室检测细胞侵袭能力变化;(4)蛋白质印迹法检测EMT标记物E-钙粘蛋白(E-cadherin)、N-钙粘蛋白(N-cadherin)和波形蛋白(Vimentin)表达水平。结果RNAi组与Ctrl组比较,(1)Oct4B1mRNA表达明显下调,P<0.05;(2)细胞迁移能力明显减弱,P<0.05;(3)细胞侵袭能力显著降低,P<0.01;(4)上皮标记物E-cadherin明显增高(P<0.05),间质标记物N-cadherin(P<0.01)和Vimentin(P<0.05)则明显下调。结论沉默Oct4B1基因可逆转SW620/OxR细胞EMT。 展开更多
