The sufficient conditions of existence of order-1 periodic solution are obtained in view of the geometrical theory of the semi-continuous dynamical system and the qualitative properties of the corresp on ding continuous system. The stability of the order-1 periodic solution is proved by means of [H. J. Guo, L. S. Chen and X. Y. Song, Qualitative analysis of impulsive state feedback control to an algae-fish system with bistable property, Appl. Math. Comput. 271 (2015) 905-922]. Methods: RT-PCR and immunocytochemistry were used to investigate the expression of cyclooxygenase-2 in 3 clones drug-sensitive and 5 clones drug-resistant ovarian cancer cell. Results: Strong COX-2 mRNA expressions were detected in 3 clones of drug-sensitive cell and weak expressions were detected in 5 clones of drug-resistant cell. The protein expression of COX-2 in drug-sensitive cell was strongly positive reaction in immunocytochemistry stain and there was a weak positive reaction in 5 clones of drug-resistant cell. Conclusion: The expression of COX-2 mRNA in drug-sensitive cell strains is much higher than that in drugresistant strains of ovarian cancer cell lines, providing a basis of the chemoprevention for ovarian cancer.展开更多

With this method, we observed the in vitro anticancer efficiency of several antineoplastics on human solid tumors and acute non-lymphatic leukemia (ANLL) specimen. Comparing with the conventional two-layer agar culture assay (2-LAC assay). The CAP-assay had a higher culture success rate, higher plating efficiency (PE) and shorter experimental period. The preliminary data suggesting and that the CAP assay has the potential to replace the conventional 2- LAC assay for in vitro drug- sensitivity testing and may be applied clinically.展开更多

METHODS siRNA,which is a leading sequence selected by previous experiments,was...OBJECTIVE To study whether siRNA targeting against the Bcl-2 gene can enhance sensitivity of HL-60 cells to all trans retinoic acid(ATRA). METHODS siRNA,which is a leading sequence selected by previous experiments,was transferred into HL-60 cells.At 6 h after transfection,the cells were cultured with ATRA.The cell growth of the HL-60 cells was measured by the MTT assay at 24, 48,72 h.The level of the Bcl-2 protein and ROS(reactive oxygen species)as well as membrane potential of the mitochondria were determined by flowcytometry. RESULTS siRNA significantly increased the inhibitory effect of ATRA on growth of the HL-60 cells.The combination of siRNA with ATRA resulted in a decrease in the Bcl-2 protein level and an increase in the ROS level as well as significantly lowering the mitochondrial membrane potential of the HL-60 cells(P<0.05). CONCLUSION Effective siRNA targeting of Bcl-2 increases the sensitivity of HL-60 leukemic cells to ATRA by inhibiting the expression of the Bcl-2 protein.展开更多
India has the highest burden of both tuberculosis(TB)and multidrug-resistant TB(MDRTB)based on the WHO Global TB Report 2019.Although the available data suggest that the total TB incidence has declined,the absolute nu...India has the highest burden of both tuberculosis(TB)and multidrug-resistant TB(MDRTB)based on the WHO Global TB Report 2019.Although the available data suggest that the total TB incidence has declined,the absolute number of new cases is still increasing.The number of reported TB cases in India in 2018 was 2.2 million,which was 1.5 million in 2009.About 47%increment in TB case notification in India within a decade shows a persistent public health problem.India contributes about 22%of the World's TB burden.Indian National Strategic Plan 2017e2025,sets out the government plans to eliminate TB by 2025.However,the milestone seems unrealistic to achieve the TB eradication goal by 2025.We developed a five-dimensional mathematical model to understand the TB dynamics in India and investigate the possibility of the earliest TB eradication time frame.The model stratifies the entire TB class into three different classes as drug-sensitive(DS),MDR,and isolated classes.The effective reproduction number,equilibrium points,and stability analysis of the model were carried out.This model predicts the total estimated cases of DS-TB and MDR-TB from 2018 to 2035 through numerical simulation and suggests that TB may be eliminated by 2035 in India if the treatment success rate could be achieved to 95%,by contact tracing and isolating at least 50%of MDR-TB.展开更多
