Evaluation of in vitro digestibility of Aspergillus oryzae fungal biomass grown on organic residue derived‑VFAs as a promising ruminant feed supplement

Background As demand for high quality animal feed continues to raise,it becomes increasingly important to mini-mize the environmental impact of feed production.An appealing sustainable approach to provide feed fractions is to use organic residues from agro-food industry.In this regard,volatile fatty acids(VFAs)such as acetic,propionic and butyric acids,derived from bioconversion of organic residues can be used as precursors for production of micro-bial protein with ruminant feed inclusion potential.This study aims to investigate the in vitro digestibility of the Asper-gillus oryzae edible fungal biomass cultivated on VFAs-derived from anaerobic digestion of residues.The produced fungal protein biomass,along with hay clover silage and rapeseed meal were subjected to various in vitro assays using two-stage Tilley and Terry(TT),gas,and bag methods to evaluate and compare its digestibility for application in ruminant feed.Results The produced fungal biomass contained a higher crude protein(CP)(41%–49%)and rather similar neutral detergent fiber(NDF)(41%–56%)compared to rapeseed meal.The rumen in vitro dry matter digestibility(IVDMD)of the fungal biomass in the TT method ranged from 82%to 88%(statistically similar to that of the gas method(72%to 85%)).The IVDMD of fungal biomass were up to 26%and 40%greater than that of hay clover silage and rapeseed meal,respectively.The type of substrate and bag method had pronounced effect on the fermentation products(ammonium-N(NH4+-N),total gas and VFAs).Fungal biomass digestion resulted in the highest release of NH4+-N(340–540 mg/L)and the ratio of acetate to propionate ratio(3.5)among subjected substrates.Conclusion The results indicate that gas method can be used as a reliable predictor for IVDMD as well as fermenta-tion products.Furthermore,the high IVDMD and fermentation product observed for Aspergillus oryzae fungal biomass digestion,suggest that the supplementation of fungal biomass will contribute to improving the rumen digestion by providing necessary nitrogen and energy to the ruminant and microbiota. 展开更多

This study aims to determine the effect of adding feed supplements of fermented shrimp waste extract in the ration on the digestibility of local chicken rations in the growth phase.The research was carried out in Jatinangor District as well as the Laboratory of Ruminant Animal Nutrition and Animal Feed Chemistry,Faculty of Animal Husbandry,Padjadjaran University,Sumedang.The method used in this study was experimental with a Complete Randomized Design(RAL).The data were analyzed by fingerprint test(ANOVA)with further tests using the Dunnet test.The object of this study consisted of 20 Sentul chickens raised from the age of 1 day to 12 weeks,divided into 5 treatments and 4 tests.The treatment consists of P0=Basal ration without the addition of feed supplement fermented shrimp waste extract,P1=Basal ration+0.5%feed supplement fermented shrimp waste extract,P2=Basal ration+1.0%feed supplement fermented shrimp waste extract,P3=Basal ration+1.5%feed supplement fermented shrimp waste extract,and P4=Basal ration+2.0%feed supplement fermented shrimp waste extract.The changes observed are the digestibility of dry matter,the digestibility of organic matter,and the digestibility of proteins.The results showed that the addition of feed supplements for fermented shrimp waste extract had a significantly different influence on the digestibility of dry matter,the digestibility of organic matter,and the digestibility of protein.This study concludes that the addition of a feed supplement of 2%fermented shrimp waste extract in the ration can produce a high ration digestibility value for Sentul chicken in the growth phase. 展开更多

Dewey) is a cool-season, perennial grass native to southeastern United States. Recently, there is a growing interest in its development as a grazing and haying forage crop due to its wide area of adaptation across this region. Consequently, there is a great need for the evaluation of its forage quality by rapid, but accurate analytical methods like Near-Infrared Reflectance Spectroscopy (NIRS). In this study, acceptable NIRS calibration models were developed for: dry matter, DM (n = 113, R<sup>2</sup> = 0.904, RSCD = 2.54, RSCIQ = 4.65);crude protein, CP (n = 113, R<sup>2</sup> = 0.974, RSCD = 5.16, RSCIQ = 5.92);acid detergent fiber, ADF (n = 116, R<sup>2</sup> = 0.896, RSCD = 2.35, RSCIQ = 1.28);neutral detergent fiber, NDF (n = 118, R<sup>2</sup> = 0.934, RSCD = 2.53, RSCIQ = 3.38);digestible dry matter, DDM (n = 116, R<sup>2</sup> = 0.895, RSCD = 2.36, RSCIQ = 1.35);dry matter intake, DMI (n = 115, R<sup>2</sup> = 0.924, RSCD = 2.40, RSCIQ = 2.53);and relative feed value, RFV (n = 114, R<sup>2</sup> = 0.932, RSCD = 2.94, RSCIQ = 2.81). Prediction of independent validation sets yielded good agreement between the NIRS predicted values and the laboratory reference values for each of: DM (n = 53, R<sup>2</sup> = 0.831, RPD = 2.45, RPIQ = 4.24);CP (n = 57, R<sup>2</sup> = 0.967, RPD = 5.37, RPIQ = 7.16);ADF (n = 49, R<sup>2</sup> = 0.895, RPD = 2.97, RPIQ = 1.51);NDF (n = 53, R<sup>2</sup> = 0.928, RPD = 3.75, RPIQ = 4.22);digestible dry matter, DDM (n = 55, R<sup>2</sup> = 0.860, RSCD = 265, RSCIQ = 1.15);dry matter intake, DMI (n = 156, R<sup>2</sup> = 0.845, RSCD = 2.48, RSCIQ = 2.11);and relative feed value, RFV (n = 55, R<sup>2</sup> = 0.916, RSCD = 3.45, RSCIQ = 3.04) contents, indicating that all seven calibration models had good quantitative information. We studied species of plants foraged by red deer in the Muling forest region of northeastern China during January 2014 and 2015. We focused on nutritional composition, energy content, and the digestibility of these plants. Crude protein content varied by plant species.Average crude protein content was 7.5 ± 1.7% of dry matter, ranging from 4.5 to 10.3%. Crude lipid content did not vary by species and averaged 7 ± 1.5%(range5.4–9.8%). Non-fiber carbohydrate(NFC) and neutral detergent fiber(NDF) content varied broadly with average values being 20.1 ± 4.8%(range 13.5–28.8%) and64.7 ± 5.9%(range 53.5–71.8%), respectively. Energy content ranged from 16.7 to 19.7 k J/g. Average dry matter digestibility(DMD) was 46.1 ± 4.8%, with a range from38.7 to 54.6%. Only three plants, Saussurea manshurica,Aralia elata and Equisetum hyemale exhibited DMD [ 50%. NFC content explained most of the variation in energy content(r = 0.451). In contrast, energy content was inversely related to NDF(r =-0.443). There was no correlation between crude protein, crude lipid, and energy content(r= 0.004, r=-0.190). S. manshurica,A. elata and E. hyemale had higher NFC and lower NDF content. Thus, these species offered the greatest nutritional value for wintering red deer in the study area. Along with high CP and NFC content, Taxus cuspidate also had high lignin content, and was thus, considered a good forage species for red deer. 展开更多

Genetic improvement of the digestibility of rice straw for increasing the utilization of the whole rice plant as feedstuffs is an important way to solve the feedstuffs shortage in southeastern China. To elucidate the genetic basis of the traits affecting the digestibility of rice straw, a rice population of 111 recombinant inbred lines (RILs) derived from a cross of Nekken 2xGaya was used to map the quantitative traits loci (QTLs) for in vitro dry matter digestion (IVDMD), the percentages of nonstructural carbohydrate (NSC) and acid detergent fiber (exclusive of residual ash, ADFom) in 2005 and 2006. IVDMD was positively correlated with NSC, and negatively correlated with ADFom. A total of 16 QTLs were detected in the two years, and the amounts of variation explained by individual QTLs ranged from 6.9% to 15.5%. Some QTLs for IVDMD and ADFom on chromosome 2 were detected in the two years. On chromosome 2, the interval RM475-RM3515 contained QTLs for IVDMD in the two years and for NSC in 2006 only, while the interval RM3874-RM5305 influenced ADFom in the two years and NSC in 2005 only. At those loci, Gaya contributed favorable alleles to IVDMD and NSC, while Nekken 2 contributed positive alleles to ADFom. 展开更多

Batch cultures of mixed rumen micro-organisms were conducted to evaluate the effects of encapsulated yeast(+EY)and encapsulated enzyme(+EE)using plant proteins(barley and oats grain)on rumen fermentation in vitro,investigate the abilities of encapsulated yeast and encapsulated enzyme to prevent rumen digestion in vitro.Treatments of the study were the control,+EY,+EE products(3.33 mg·mL^(-1) of the incubation medium),unencapsulated yeast(-EY)and enzyme(-EE)products(0.17 and 0.17μL·mL^(-1) of the incubation medium,respectively).+EY group increased dry matter disappearance(DMD,P<0.01)and the total volatile fatty acids(TVFA,P<0.01)at 3 h of the incubation compared with the control,regardless of encapsulation of yeast.Gas production(GP)of+EY group was higher(P=0.05,29.94 mL·mL^(-1) organic matter,OM)than that of the control(25.08 mL·g^(-1) OM)at 3 h of the incubation.Supplementation+EY increased DMD(P=0.04,0.394 vs 0.352,respectively)and acetic proportion(P=0.04,52.6 vs 49.8 mol•100 mL^(-1),respectively)at 6 h of the incubation and increased A:P ratio(P<0.01,3.11 and 2.86,respectively)at 24 h of the incubation,as compared to unencapsulation of yeast.Supplementation of enzyme had higher(P≤0.04)GP,DMD and TVFA at 3 and 6 h of the incubation compared with the control,regardless of encapsulation.Moreover,the addition of+EE produced greater GP at 6(P<0.01,92.35 vs 78.21 mL·g^(-1) OM,respectively),12(218.47 vs 159.18 mL·g^(-1) OM)and 24 h(380.97 vs 297.78 mL·g^(-1) OM,respectively)of the incubation,higher DMD(0.347 vs 0.313,respectively)at 3 h of the incubation as compared to-EE group.The study showed that the encapsulation might protect part of yeast and enzyme from releasing to the rumen throughout the digestion in vitro,resulting in higher or no difference of rumen fermentation parameters compared with unencapsulated groups at any incubation times.In comparison with-EY and-EE,the higher rumen fermentation parameters at the early incubation time were observed,which could be attributed to the higher concentration of yeast or enzyme.However,regardless of the encapsulation,the results indicated that both yeast and enzyme only improved the speed rather than the extent of rumen fermentation in vitro. 展开更多
