The results show that the available B in western Jilin is in a middle level,the content in Nong'an is higher than that in the others; the available Mn is extremely abundant; the available Cu in Nong'an is obviously higher than other areas,and the content in Da'an differs greatly; the available Zn is in a middle level; the severe shortage of available Mo differs between different areas; and the available Fe is extremely abundant. The available Mn,Cu and Fe have significantly positive correlation with their total content; the available B has significantly positive correlation with pH,and Zn has obviously negative correlation with pH; the available Mn and Fe had significantly positive correlation with soil organic matter.展开更多

However, it is difficult to classitythe high dimensional feature matrix directly because of too large dimensions for many classifiers. This paper combines the concepts of time-frequency distribution(TFD) with non-negative matrix factorization(NMF), and proposes a novel TFD matrix factorization method to enhance representation and identification of bearing fault. Throughout this method, the TFD of a vibration signal is firstly accomplished to describe the localized faults with short-time Fourier transform(STFT). Then, the supervised NMF mapping is adopted to extract the fault features from TFD. Meanwhile, the fault samples can be clustered and recognized automatically by using the clustering property of NMF. The proposed method takes advantages of the NMF in the parts-based representation and the adaptive clustering. The localized fault features of interest can be extracted as well. To evaluate the performance of the proposed method, the 9 kinds of the bearing fault on a test bench is performed. The proposed method can effectively identify the fault severity and different fault types. Moreover, in comparison with the artificial neural network(ANN), NMF yields 99.3% mean accuracy which is much superior to ANN. This research presents a simple and practical resolution for the fault diagnosis problem of rolling element bearing in high dimensional feature space.展开更多

Based on multi-purpose regional geochemical survey, regional eco-geochemical assessment, local eco-geochemical assessment and comprehensive appraisal, the eco-geochemical survey and assessment in Pearl River Delta Economic Zone of 41,698 km2?were completed. Samples from soils were collected in accordance with the two-layer grid method. Totally 54 elements and indicators for soils were determined. Compared to deep soils, the sampled surface soils are enriched in OrgC, N, P, Cd, S,Hg, Ag, B, Au, S and poor with As, Ni, I, Co, Cr, V, MgO, Sc, Al2O3, Fe2O3?etc. The characteristics of geochemical reference value of element in soil that inherited soil parent material and regional elements combined features reflected that the elements enriched in the soil was interrelated with acid rock, sandstone and shale. The spatial distribution characteristics of element regional geochemistry were conditional by regional environmental geological conditions,and effected by human activities. The Pearl River Delta plain is a typical geochemical landscape area with regional anomaly of multiple-elements. The north, western and eastern parts of the Pearl River Delta Economic Zone are quite different in geochemical features due to regional geological background, soil parent materials, geomorphic characteristics and human activities. Environment quality evaluation results show that the grade I and grade II soil accounted for 19.9% and 57.3% of the total area. Many samples that widely distributed in the economic developed of Pearl River Delta Plain area reached the third-grade of national soil environment standard. The soil enriched in Cd, Hg, As, and the area ratio accounting for 22.8% of the total area. It is mainly controlled by the geochemical background, the Pearl River Delta formation evolution process, especially the marine transgression process lead to Cd, Cu, Zn and Pb enrichment in Pearl River Delta plain. At the same time, under the influence of higher pressure of human activities, all kinds of exogenous input material carrying heavy metal pollutants on soil environmental quality also could not to ignore.展开更多

Firstly, by comparing the rigorous solution against the asymptotic solution for a problem of an infinite plate embedded a central crack, we found that the stresses in a restrictive interval near the crack tip given by the rigorous solution can be used to determine the stress intensity factor, which is nearly equal to the stress intensity factor given by the asymptotic solution. Secondly, the crack problem is solved numerically by the finite element method. Depending on the modeling capability of the software, we designed an adaptive mesh model to simulate the stress singularity. Thus, the stress result in an appropriate interval near the crack tip is fairly approximated to the rigorous solution of the corresponding crack problem. Therefore, the stress intensity factor may be calculated from the stress distribution in the appropriate interval, with a high accuracy.展开更多

The incident solar...A spectrum-splitting and beam-concentrating (SSBC) diffractive optical element (DOE) for three-junction pho- tovoltaics (PV) system is designed and fabricated by five-circ/e micro-fabrication. The incident solar light is efficiently split into three sub-spectrum ranges and strongly concentrated on the focal plane, which can be di- rectly utilized by suitable spectrum-matching solar cells. The system concentration factor reaches 12x. Moreover, the designed wavelengths (450nm, 550nm and 65Onto) are spatially distributed on the focal plane, in good agree- ment with the theoretical results. The average optical effic/ency of all the cells over the three designed wavelengths is 60.07%. The SSBC DOE with a high concentration factor and a high optical efficiency provides a cost-effective approach to achieve higher PV conversion efficieneies.展开更多

Its promoter driving the expression of a reporter gene (d2EGFP) has been used to analyse the role of certain defined putative elements and a downstream enhancer element in gene expression. These experiments were carried out in vitro using a line of pre-pubertal mouse Sertoli cells, transienly transfected with circular DNA constructs with variously mutated promoter elements. A downstream enhancer element, situated immediately 3’ of the polyadenylation (PA) signal for Amh, has been inserted in an equivalent position in the d2EGFP construct. When the Amh promoter is unmodified, the downstream enhancer (DE) is positively associated with a large increase in EGFP expression. This is at least partly the consequence of an increased rate of expression by individual cells. Experiments using variously truncated Amh promoters indicate that an upstream region (-214 to -336) may play a minor role in facilitating enhancement. However mutation of the Wilms tumour factor-1 element, situated between the tata box and the start of translation, results in an almost complete suppression of enhancement.展开更多

Gene expression of transfected myoblasts was detected by RT-PCR, Western blot and fluorescence microscope under different oxygen concentrations and different hypoxia time. The results showed that in hypoxia group, the VEGF gene bands were seen and with the decrease of oxygen concentrations and prolongation of hypoxia time, the expression of VEGF mRNA was obviously increased. Under hypoxic environment, the expression of VEGF protein in the transfected myoblasts was significantly increased. EGFP was expressed only under hypoxic environment but not under normoxic environment. It was concluded that hypoxia promoter could be constructed with HRE and regulate the expression of VEGF gene under hypoxic and normoxic environment, which could enhance the re- liability of gene therapy.展开更多

CREB activation initiates a series of intracellular signaling pathways that promote neuronal survival. OBJECTIVE: To investigate the regulatory effects of basic fibroblast growth factor (bFGF) on cerebral neuronal CREB expression following ischemia/reperfusion injury. DESIGN, TIME AND SETTING: An immunohistochemical detection experiment was performed at the Department of Anatomy, Shenyang Medical College, between October 2006 and April 2008. MATERIALS: A total of 60 healthy, adult, Wistar rats were randomly divided into three groups: sham-operated (n =12), ischemia/reperfusion (n = 24), and bFGF-treated (n = 24). Rabbit anti-rat CREB (1: 100) and biotin labeled goat anti-rabbit IgG were purchased from the Wuhan Boster Company, China. MetaMorph-evolution MP5.0-BX51 microscopy imaging system was provided by China Medical University, China. METHODS: Rat models of cerebral ischemia/reperfusion injury were developed using the suture method for right middle cerebral artery occlusion. Two-hour ischemia was followed by reperfusion. Rats from the bFGF-treated and ischemia/reperfusion groups were intraperitoneally administered endogenous bFGF (500 IU/mL, 2 000 IU/kg) or an equal amount of physiological saline. Rats from the sham-operated group underwent a similar surgical procedure, without induction of ischemia/reperfusion injury and drug administration. MAIN OUTCOME MEASURES: After 48-hour reperfusion, hippocampal and parietal cortical neuronal CREB expression was detected by immunohistochemistry, and the absorbance of hippocampal CREB-positive products was determined using MetaMorph-evolutionMP5.0-BX51 microscopy imaging system. RESULTS: The sham-operated group exhibited noticeable CREB expression in hippocampal and parietal cortical neurons. In the ischemia/reperfusion group, the CREB expression was discrete and neurons were poorly arranged. The bFGF-treated group exhibited increased CREB expression and better neuronal arrangement compared with the ischemia/reperfusion group. The mean absorbance of CREB-immunoreactive products in the hippocampus and parietal cortex was significantly higher in the ischemia/reperfusion group than in the sham-operated group (P 〈 0.05), and significantly higher in the bFGF-treated group than in the ischemia/reperfusion group (P 〈 0.05). CONCLUSION: bFGF significantly upregulates CREB expression in hippocampal and parietal cortical neurons following ischemia/reperfusion injury.展开更多

The circular regions surrounding notch/crack tips are taken as the complementary energy region in which a number of leading terms of singular solutions for stresses are used, with the sought SIFs being among the unknown coefficients. The rest of the arbitrary domain is taken as the potential energy region in which FEMOL is applied to obtain approximate displacements. A mixed system of ordinary differential equations (ODEs) and algebraic equations is derived via the sub-region generalized variational principle. A singularity removal technique that eliminates the stress parameters from the mixed equation system eventually yields a standard FEMOL ODE system, the solution of which is no longer singular and is simply and efficiently obtained using a standard general-purpose ODE solver. A number of numerical examples, including bi-material notches/cracks in anti-plane and plane elasticity, are given to show the generally excellent performance of the proposed method.展开更多

The impacted slope is around 200 meters high, with two terraces that have different slope inclinations. The lower bench, located above the basement, consistently fails and sets others up for failure. The fluctuating water level of the slope, which travels down the slope masses, exacerbates the slide problem. The majority of these rocks are Amalpata landslide area experiences several structural disruptions. The area’s stability must be evaluated in order to prevent and control more harm from occurring to the nearby agricultural land and people living along the slope. The slopes’ failures increase the damages of house existing in nearby area and the erosion of the slope. Two modeling techniques the finite element approach and the limit equilibrium method were used to simulate the slope. The findings show that, in every case, the terrace above the basement is where the majority of the stress is concentrated, with a safety factor of near unity. Using probabilistic slope stability analysis, the failure probability was predicted to be between 98.90% and 100%.展开更多

In the present review we summarize the known roles of extracellular signal-regulated kinase, cAMP response element-binding protein and brain-derived neurotrophic factor in the pathogenesis of depression and in the mechanism of action of antidepressant medicines. The extracellular signal-regulated kinase/cAMP response element-binding protein/brain-derived neurotrophic factor pathway has potential to be used as a biological index to help diagnose depression, and as such it is considered as an important new target in the treatment of depression.展开更多

The stress intensity factor (SIF) is an important parameter for estimating the life of the cracked structure. In this paper, the stress intensity factors of a slant-cracked plate, which is made of 6061-T651 aluminum, have been calculated using extended finite element method (XFEM) and finite element method (FEM) in ABAQUS software and the results were compared with theoretical values. Numerical values obtained from these two methods were close to the theoretical values. In simulations of crack growth at different crack angles, the crack propagation angle values were closer to the theoretical values in XFEM method. Also, the accuracy and validity of fatigue crack growth curve were much closer to the theoretical graph in XFEM than the FEM. Therefore, in this paper the capabilities of XFEM were realized in analyzing issues such as cracks.展开更多

This paper explores a novel approach to structural repair by utilizing piezoelectric actuators. The primary focus of this study is to investigate the influence of an adhesively bonded piezoelectric actuator patch placed above a circular hole on the stress intensity factor (SIF) in an aluminium plate. The plate is subjected to uniaxial tensile stress, while the piezoelectric actuator is excited with varying voltage levels. The analysis is conducted using the finite element method (FEM), a powerful numerical technique for simulating complex structures. The study assesses the stress distribution and employs the SIF as an adequate criterion for evaluating the impact of different patch configurations. The results indicate a strong correlation between the applied voltage and the SIF. Whether the SIF increases or decreases depends on the polarization of the piezoelectric actuator. Particularly noteworthy is the finding that rectangular patches in a horizontal orientation significantly reduce the SIF compared to other patch geometries. Moreover, double-sided patches exhibit a pronounced decrease in the SIF compared to single-sided patches. In summary, this research underscores the potential of piezoelectric actuators in mitigating stress intensity in structures with circular hole with crack initiation. It offers valuable insights into the influence of applied voltage, patch geometry, and patch placement on the SIF, thereby contributing to developing effective strategies for enhancing structural integrity.展开更多

The square-root stress singularity around the corner crack front is simulated using the collapsed 20-node quarter point singular elements. The contact interaction between the bolt and the hole boundary is considered in the finite element analysis. The stress intensity factors (SIFs) along the crack front are evaluated by using the displacement correlation technique. The effects of the amount of clearance between the hole and the bolt on the SIFs are investigated. The numerical results indicate that the SIF for mode I decrease with the decreases in clearance, and in the cases of clearance being present, the corner crack is in a mix-mode, even if mode I loading is dominant.展开更多

The trace element composition of aerosol particles(TSP and PM ...An one-year of sampling aerosol program was carried out during the period of April 1995 to April 1996 at coastal region, Cape D′Aguilar, in Hong Kong. The trace element composition of aerosol particles(TSP and PM 10 ) were measured and analyzed by using the Instrumental Neutron Activation Analysis(INAA) and Inductive Couple Plasma-Mass Spectrometry(ICP-MS) techniques. The results showed that the polluted elements such as: V, Zn, As, Pb, Cd, Sb, I, Se, Bi, with high enrichment factor(E.F) values, are derived from anthropogenic sources, while Sc, Al, Fe, Mn, Sm, La, Sr are less than 10 in E.F, these elements are related the crust and soil. The concentration of the trace elements are present seasonal variation i.e. summer low and winter high, whereas the V or noncrustal V(V *) present summer high and winter low, this phenomenon might be explained by local or regional oil combustion sources. Use three type of elemental ratio, e.g. La/Sm, Mn */V * and Bi/Al indicate that both TSP and PM 10 of La/Sm ratios are mainly related to soil and dust, Mn */V * are probably represent fossil combustion and Bi/Al are possibly associated with refuse incineration, ferromanganese alloys and aluminum production. The varimax rotation factor analysis for trace elements was performed. By means of the absolute principal component analysis(APCA) and multiple regression, the contribution of trace elements to possible sources and overall means of trace elements were estimated. The results showed that the present station is impacted by the anthropogenic species, although the quantities are different during the summertime and wintertime.展开更多

Si levels are highest, followed by Al, and the d...The major and trace elements in 110 surface sediment samples collected from the middle of the Bay of Bengal(mid-Bay of Bengal) are analyzed to investigate provenance. Si levels are highest, followed by Al, and the distributions of these two elements are identical. The average CIA*(chemical index of alteration) value is 72.07,indicating that the degree of weathering of the sediments in the study area is intermediate between those of sediments of the Himalayan and Indian rivers. Factor analyses and discrimination function analyses imply that the two main provenances are the Himalayan and the Indian continent. The inverse model calculation of the Tinormalized element ratios of the Bay of Bengal sediments indicate an estimated average contribution of 83.5%and 16.5% from the Himalayan and peninsular Indian rivers to the study area, respectively. The Himalayan source contributes more sediment to the eastern part of the study area, whereas the western part receives more sediment from the Indian Peninsula than did the eastern part. The primary mechanisms for deposition of sediments in the study area are the transport of Himalayan matter by turbidity currents and river-diluted water and the transport of Indian matter to the study area by a surface circulation in the Bay of Bengal, particularly the East India Coastal Current.展开更多

Gastric cancer cases were selected from the authors′ hospital, occiput hair was collected to measure its molybdenum content with an atomic absorption spectrograph and controls were selected from the same hospital for comparison. Gastric cancer cases were selected from three hospitals at the same time, blood samples were taken on an empty stomach and serum molybdenum contents were measured with the atomic absorption spectrograph, and controls were selected from the same hospitals. Blind method was used in the whole course (chemical analysts did not know the source and nature of samples).RESULTS A negative correlation existed between soil available molybdenum content and mortality rate of gastric cancer (r=-0285, P<005); hair molybdenum contents of gastric cancer cases were lower than those of healthy controls (0308μg/g±0673μg/g and 0707μg/g±0561μg/g respectively, P<001); serum molybdenum contents of patients were also lower than those of healthy controls (2184μg/L±749μg/L and 2538μg/L±858μg/L respectively, P<005).CONCLUSION Deficiency of molybdenum may be one of the risk factors in gastric cancer.展开更多
Cryoconite samples were collected from two different climatic domains i.e.,the Sutri Dhaka glacier,western Himalaya India and Svalbard glaciers,the Spitsbergen,Arctic,to understand the elemental source and elemental d...Cryoconite samples were collected from two different climatic domains i.e.,the Sutri Dhaka glacier,western Himalaya India and Svalbard glaciers,the Spitsbergen,Arctic,to understand the elemental source and elemental deposition patterns.The data of geochemical analysis suggest that the Himalayan cryoconite samples accumulate higher concentrations as compared to the cryoconite samples of the Arctic glaciers.The concentration of lithophile elements(Cs,Li,Rb and U)was recorded higher in the cryoconite holes of the Himalayas,especially,in the lower to the higher parts of the glacier,whereas,lower concentrations were recorded in the Arctic samples.Chalcophile elements in the Himalayan cryoconites are enriched in As and Bi while the Arctic cryoconite samples show a higher concentration of Bi,Pb and As.The higher concentrations are responsible for influencing the ecosystem and in human health related issues.Siderophile elements(Co,Fe,Mn and Ni)show high concentrations in the Himalayan samples,whereas,the Arctic samples show minor variations and low elemental concentration in these elements,respectively.In addition,a few elements,such as Ag,Mg,and Ca show higher concentration in the Himalayan glacier samples.Ca also occurs in high concentrations in Arctic glacier samples.R-mode factor analysis of the Himalayas(Arctic)samples indicate that the elements are distributed in four(three)factors,explaining 89%(90%)of the variance in their elemental distribution.The Factor 1 suggests statistically significant positive loadings for most of the lithophile,chalcophile and siderophile elements of the "Himalayan" and the Arctic cryoconite samples.The sample-wise factor score distribution shows a considerable variation in the sampling locations along the glaciers of both the regions.Factors 2 and 3,demonstrate insignificant loading for most of the elements,except statistically significant positive loading in some of the elements of the both,Himalayan and Arctic "cryoconites".The higher elemental concentration in the cryoconites of the Himalayan region may be an indicator of the natural processes and/or attributed to the rapid industrialization in the Asian countries.展开更多

文摘Based on the assay of the total content and available content of the trace nutrient elements and the soil pH and organic matter of the soil samples,the characteristic of the available content of the trace nutrient elements and their affecting factors are studied. The results show that the available B in western Jilin is in a middle level,the content in Nong'an is higher than that in the others; the available Mn is extremely abundant; the available Cu in Nong'an is obviously higher than other areas,and the content in Da'an differs greatly; the available Zn is in a middle level; the severe shortage of available Mo differs between different areas; and the available Fe is extremely abundant. The available Mn,Cu and Fe have significantly positive correlation with their total content; the available B has significantly positive correlation with pH,and Zn has obviously negative correlation with pH; the available Mn and Fe had significantly positive correlation with soil organic matter.

文摘Due to the non-stationary characteristics of vibration signals acquired from rolling element bearing fault, thc time-frequency analysis is often applied to describe the local information of these unstable signals smartly. However, it is difficult to classitythe high dimensional feature matrix directly because of too large dimensions for many classifiers. This paper combines the concepts of time-frequency distribution(TFD) with non-negative matrix factorization(NMF), and proposes a novel TFD matrix factorization method to enhance representation and identification of bearing fault. Throughout this method, the TFD of a vibration signal is firstly accomplished to describe the localized faults with short-time Fourier transform(STFT). Then, the supervised NMF mapping is adopted to extract the fault features from TFD. Meanwhile, the fault samples can be clustered and recognized automatically by using the clustering property of NMF. The proposed method takes advantages of the NMF in the parts-based representation and the adaptive clustering. The localized fault features of interest can be extracted as well. To evaluate the performance of the proposed method, the 9 kinds of the bearing fault on a test bench is performed. The proposed method can effectively identify the fault severity and different fault types. Moreover, in comparison with the artificial neural network(ANN), NMF yields 99.3% mean accuracy which is much superior to ANN. This research presents a simple and practical resolution for the fault diagnosis problem of rolling element bearing in high dimensional feature space.
文摘Soil heavy metal pollution is one of the main environmental problems in Pearl River Delta Economic?Zone of China. Based on multi-purpose regional geochemical survey, regional eco-geochemical assessment, local eco-geochemical assessment and comprehensive appraisal, the eco-geochemical survey and assessment in Pearl River Delta Economic Zone of 41,698 km2?were completed. Samples from soils were collected in accordance with the two-layer grid method. Totally 54 elements and indicators for soils were determined. Compared to deep soils, the sampled surface soils are enriched in OrgC, N, P, Cd, S,Hg, Ag, B, Au, S and poor with As, Ni, I, Co, Cr, V, MgO, Sc, Al2O3, Fe2O3?etc. The characteristics of geochemical reference value of element in soil that inherited soil parent material and regional elements combined features reflected that the elements enriched in the soil was interrelated with acid rock, sandstone and shale. The spatial distribution characteristics of element regional geochemistry were conditional by regional environmental geological conditions,and effected by human activities. The Pearl River Delta plain is a typical geochemical landscape area with regional anomaly of multiple-elements. The north, western and eastern parts of the Pearl River Delta Economic Zone are quite different in geochemical features due to regional geological background, soil parent materials, geomorphic characteristics and human activities. Environment quality evaluation results show that the grade I and grade II soil accounted for 19.9% and 57.3% of the total area. Many samples that widely distributed in the economic developed of Pearl River Delta Plain area reached the third-grade of national soil environment standard. The soil enriched in Cd, Hg, As, and the area ratio accounting for 22.8% of the total area. It is mainly controlled by the geochemical background, the Pearl River Delta formation evolution process, especially the marine transgression process lead to Cd, Cu, Zn and Pb enrichment in Pearl River Delta plain. At the same time, under the influence of higher pressure of human activities, all kinds of exogenous input material carrying heavy metal pollutants on soil environmental quality also could not to ignore.

文摘In this article, a direct stress approach based on finite element analysis to determine the stress intensity factor is improved. Firstly, by comparing the rigorous solution against the asymptotic solution for a problem of an infinite plate embedded a central crack, we found that the stresses in a restrictive interval near the crack tip given by the rigorous solution can be used to determine the stress intensity factor, which is nearly equal to the stress intensity factor given by the asymptotic solution. Secondly, the crack problem is solved numerically by the finite element method. Depending on the modeling capability of the software, we designed an adaptive mesh model to simulate the stress singularity. Thus, the stress result in an appropriate interval near the crack tip is fairly approximated to the rigorous solution of the corresponding crack problem. Therefore, the stress intensity factor may be calculated from the stress distribution in the appropriate interval, with a high accuracy.

文摘A spectrum-splitting and beam-concentrating (SSBC) diffractive optical element (DOE) for three-junction pho- tovoltaics (PV) system is designed and fabricated by five-circ/e micro-fabrication. The incident solar light is efficiently split into three sub-spectrum ranges and strongly concentrated on the focal plane, which can be di- rectly utilized by suitable spectrum-matching solar cells. The system concentration factor reaches 12x. Moreover, the designed wavelengths (450nm, 550nm and 65Onto) are spatially distributed on the focal plane, in good agree- ment with the theoretical results. The average optical effic/ency of all the cells over the three designed wavelengths is 60.07%. The SSBC DOE with a high concentration factor and a high optical efficiency provides a cost-effective approach to achieve higher PV conversion efficieneies.
文摘Amh (anti-Müllerian hormone) is a single copy gene which is expressed strongly in Sertoli cells in the foetal testis and participates in the onset of sexual differentiation. Its promoter driving the expression of a reporter gene (d2EGFP) has been used to analyse the role of certain defined putative elements and a downstream enhancer element in gene expression. These experiments were carried out in vitro using a line of pre-pubertal mouse Sertoli cells, transienly transfected with circular DNA constructs with variously mutated promoter elements. A downstream enhancer element, situated immediately 3’ of the polyadenylation (PA) signal for Amh, has been inserted in an equivalent position in the d2EGFP construct. When the Amh promoter is unmodified, the downstream enhancer (DE) is positively associated with a large increase in EGFP expression. This is at least partly the consequence of an increased rate of expression by individual cells. Experiments using variously truncated Amh promoters indicate that an upstream region (-214 to -336) may play a minor role in facilitating enhancement. However mutation of the Wilms tumour factor-1 element, situated between the tata box and the start of translation, results in an almost complete suppression of enhancement.
文摘The present work presents an iin silicoi analysis of Upstream Regulatory Modules (URMs) of genes expressed in tapetum specific manner in dicotyledon and monocotyledon plants. In the current analysis, we identified several motifs conserved in these URMs of which ten were observed to be part of known icisi-elements using tools and databases like MEME, PLACE, MAST and TFSEARCH. We also identified that binding sites for two transcription factors, DOF and WRKY71 were found to be present in majority of the URMs.

文摘The regulation of hypoxic response elements on the expression of vascular endothelial growth factor (VEGF) gene transfected to primary cultured rat skeletal myoblasts under hypoxic environment was investigated. pEGFP-C3-9HRE-CMV-VEGF vector was constructed with molecular biology technique and transfected to primary cultured rat skeletal myoblasts by lipofectamine in vitro. Gene expression of transfected myoblasts was detected by RT-PCR, Western blot and fluorescence microscope under different oxygen concentrations and different hypoxia time. The results showed that in hypoxia group, the VEGF gene bands were seen and with the decrease of oxygen concentrations and prolongation of hypoxia time, the expression of VEGF mRNA was obviously increased. Under hypoxic environment, the expression of VEGF protein in the transfected myoblasts was significantly increased. EGFP was expressed only under hypoxic environment but not under normoxic environment. It was concluded that hypoxia promoter could be constructed with HRE and regulate the expression of VEGF gene under hypoxic and normoxic environment, which could enhance the re- liability of gene therapy.

文摘BACKGROUND: cAMP-response element binding protein (CREB) is a key modulator of various signaling pathways. CREB activation initiates a series of intracellular signaling pathways that promote neuronal survival. OBJECTIVE: To investigate the regulatory effects of basic fibroblast growth factor (bFGF) on cerebral neuronal CREB expression following ischemia/reperfusion injury. DESIGN, TIME AND SETTING: An immunohistochemical detection experiment was performed at the Department of Anatomy, Shenyang Medical College, between October 2006 and April 2008. MATERIALS: A total of 60 healthy, adult, Wistar rats were randomly divided into three groups: sham-operated (n =12), ischemia/reperfusion (n = 24), and bFGF-treated (n = 24). Rabbit anti-rat CREB (1: 100) and biotin labeled goat anti-rabbit IgG were purchased from the Wuhan Boster Company, China. MetaMorph-evolution MP5.0-BX51 microscopy imaging system was provided by China Medical University, China. METHODS: Rat models of cerebral ischemia/reperfusion injury were developed using the suture method for right middle cerebral artery occlusion. Two-hour ischemia was followed by reperfusion. Rats from the bFGF-treated and ischemia/reperfusion groups were intraperitoneally administered endogenous bFGF (500 IU/mL, 2 000 IU/kg) or an equal amount of physiological saline. Rats from the sham-operated group underwent a similar surgical procedure, without induction of ischemia/reperfusion injury and drug administration. MAIN OUTCOME MEASURES: After 48-hour reperfusion, hippocampal and parietal cortical neuronal CREB expression was detected by immunohistochemistry, and the absorbance of hippocampal CREB-positive products was determined using MetaMorph-evolutionMP5.0-BX51 microscopy imaging system. RESULTS: The sham-operated group exhibited noticeable CREB expression in hippocampal and parietal cortical neurons. In the ischemia/reperfusion group, the CREB expression was discrete and neurons were poorly arranged. The bFGF-treated group exhibited increased CREB expression and better neuronal arrangement compared with the ischemia/reperfusion group. The mean absorbance of CREB-immunoreactive products in the hippocampus and parietal cortex was significantly higher in the ischemia/reperfusion group than in the sham-operated group (P 〈 0.05), and significantly higher in the bFGF-treated group than in the ischemia/reperfusion group (P 〈 0.05). CONCLUSION: bFGF significantly upregulates CREB expression in hippocampal and parietal cortical neurons following ischemia/reperfusion injury.
文摘The present paper is addressed to the finite element method combined with dynamic photoelastic analysis of propagating cracks, that is, on the basis of [1] by Chien Wei-zang, finite elements which incorporate the propagating crack-tip singularity intrinsic to two-dimensional elasticity are employed. THe relation between crack opening length and time step obtained from dynamic photoelaslie analysis is used as a definite condition for solving the dynamic equations and simulating the crack propagations as well As an example, the impact response of dynamie-bending-test specimen is investigated and the dynamic stress-intensity factor obtained from the mentioned finite element analysis and dynamic photoelasticity is in reasonable agreement with each other.

文摘Based on the sub-region generalized variationM principle, a sub-region mixed version of the newly-developed semi-analytical 'finite element method of lines' (FEMOL) is proposed in this paper for accurate and efficient computation of stress intensity factors (SIFs) of two-dimensional notches/cracks. The circular regions surrounding notch/crack tips are taken as the complementary energy region in which a number of leading terms of singular solutions for stresses are used, with the sought SIFs being among the unknown coefficients. The rest of the arbitrary domain is taken as the potential energy region in which FEMOL is applied to obtain approximate displacements. A mixed system of ordinary differential equations (ODEs) and algebraic equations is derived via the sub-region generalized variational principle. A singularity removal technique that eliminates the stress parameters from the mixed equation system eventually yields a standard FEMOL ODE system, the solution of which is no longer singular and is simply and efficiently obtained using a standard general-purpose ODE solver. A number of numerical examples, including bi-material notches/cracks in anti-plane and plane elasticity, are given to show the generally excellent performance of the proposed method.
文摘The stability study of the ongoing and recurring Amalpata landslide in Baglung in Nepal’s Gandaki Province is presented in this research. The impacted slope is around 200 meters high, with two terraces that have different slope inclinations. The lower bench, located above the basement, consistently fails and sets others up for failure. The fluctuating water level of the slope, which travels down the slope masses, exacerbates the slide problem. The majority of these rocks are Amalpata landslide area experiences several structural disruptions. The area’s stability must be evaluated in order to prevent and control more harm from occurring to the nearby agricultural land and people living along the slope. The slopes’ failures increase the damages of house existing in nearby area and the erosion of the slope. Two modeling techniques the finite element approach and the limit equilibrium method were used to simulate the slope. The findings show that, in every case, the terrace above the basement is where the majority of the stress is concentrated, with a safety factor of near unity. Using probabilistic slope stability analysis, the failure probability was predicted to be between 98.90% and 100%.

文摘The extracellular signal-regulated kinase/cAMP response element-binding protein/brain-derived neurotrophic factor signal transduction pathway plays an important role in the mechanism of action of antidepressant drugs and has dominated recent studies on the pathogenesis of depression. In the present review we summarize the known roles of extracellular signal-regulated kinase, cAMP response element-binding protein and brain-derived neurotrophic factor in the pathogenesis of depression and in the mechanism of action of antidepressant medicines. The extracellular signal-regulated kinase/cAMP response element-binding protein/brain-derived neurotrophic factor pathway has potential to be used as a biological index to help diagnose depression, and as such it is considered as an important new target in the treatment of depression.
文摘The 6061-T651 aluminium alloy is one of the most common aluminium alloys for marine components and general structures. The stress intensity factor (SIF) is an important parameter for estimating the life of the cracked structure. In this paper, the stress intensity factors of a slant-cracked plate, which is made of 6061-T651 aluminum, have been calculated using extended finite element method (XFEM) and finite element method (FEM) in ABAQUS software and the results were compared with theoretical values. Numerical values obtained from these two methods were close to the theoretical values. In simulations of crack growth at different crack angles, the crack propagation angle values were closer to the theoretical values in XFEM method. Also, the accuracy and validity of fatigue crack growth curve were much closer to the theoretical graph in XFEM than the FEM. Therefore, in this paper the capabilities of XFEM were realized in analyzing issues such as cracks.
文摘Circular holes are commonly employed in engineering designs;however, they often serve as locations where cracks initiate and propagate. This paper explores a novel approach to structural repair by utilizing piezoelectric actuators. The primary focus of this study is to investigate the influence of an adhesively bonded piezoelectric actuator patch placed above a circular hole on the stress intensity factor (SIF) in an aluminium plate. The plate is subjected to uniaxial tensile stress, while the piezoelectric actuator is excited with varying voltage levels. The analysis is conducted using the finite element method (FEM), a powerful numerical technique for simulating complex structures. The study assesses the stress distribution and employs the SIF as an adequate criterion for evaluating the impact of different patch configurations. The results indicate a strong correlation between the applied voltage and the SIF. Whether the SIF increases or decreases depends on the polarization of the piezoelectric actuator. Particularly noteworthy is the finding that rectangular patches in a horizontal orientation significantly reduce the SIF compared to other patch geometries. Moreover, double-sided patches exhibit a pronounced decrease in the SIF compared to single-sided patches. In summary, this research underscores the potential of piezoelectric actuators in mitigating stress intensity in structures with circular hole with crack initiation. It offers valuable insights into the influence of applied voltage, patch geometry, and patch placement on the SIF, thereby contributing to developing effective strategies for enhancing structural integrity.

文摘The three-dimensional finite element method is used to solve the problem of the quarter-elliptical comer crack of the bolt-hole in mechanical joints being subjected to remote tension. The square-root stress singularity around the corner crack front is simulated using the collapsed 20-node quarter point singular elements. The contact interaction between the bolt and the hole boundary is considered in the finite element analysis. The stress intensity factors (SIFs) along the crack front are evaluated by using the displacement correlation technique. The effects of the amount of clearance between the hole and the bolt on the SIFs are investigated. The numerical results indicate that the SIF for mode I decrease with the decreases in clearance, and in the cases of clearance being present, the corner crack is in a mix-mode, even if mode I loading is dominant.
文摘An one-year of sampling aerosol program was carried out during the period of April 1995 to April 1996 at coastal region, Cape D′Aguilar, in Hong Kong. The trace element composition of aerosol particles(TSP and PM 10 ) were measured and analyzed by using the Instrumental Neutron Activation Analysis(INAA) and Inductive Couple Plasma-Mass Spectrometry(ICP-MS) techniques. The results showed that the polluted elements such as: V, Zn, As, Pb, Cd, Sb, I, Se, Bi, with high enrichment factor(E.F) values, are derived from anthropogenic sources, while Sc, Al, Fe, Mn, Sm, La, Sr are less than 10 in E.F, these elements are related the crust and soil. The concentration of the trace elements are present seasonal variation i.e. summer low and winter high, whereas the V or noncrustal V(V *) present summer high and winter low, this phenomenon might be explained by local or regional oil combustion sources. Use three type of elemental ratio, e.g. La/Sm, Mn */V * and Bi/Al indicate that both TSP and PM 10 of La/Sm ratios are mainly related to soil and dust, Mn */V * are probably represent fossil combustion and Bi/Al are possibly associated with refuse incineration, ferromanganese alloys and aluminum production. The varimax rotation factor analysis for trace elements was performed. By means of the absolute principal component analysis(APCA) and multiple regression, the contribution of trace elements to possible sources and overall means of trace elements were estimated. The results showed that the present station is impacted by the anthropogenic species, although the quantities are different during the summertime and wintertime.

文摘The major and trace elements in 110 surface sediment samples collected from the middle of the Bay of Bengal(mid-Bay of Bengal) are analyzed to investigate provenance. Si levels are highest, followed by Al, and the distributions of these two elements are identical. The average CIA*(chemical index of alteration) value is 72.07,indicating that the degree of weathering of the sediments in the study area is intermediate between those of sediments of the Himalayan and Indian rivers. Factor analyses and discrimination function analyses imply that the two main provenances are the Himalayan and the Indian continent. The inverse model calculation of the Tinormalized element ratios of the Bay of Bengal sediments indicate an estimated average contribution of 83.5%and 16.5% from the Himalayan and peninsular Indian rivers to the study area, respectively. The Himalayan source contributes more sediment to the eastern part of the study area, whereas the western part receives more sediment from the Indian Peninsula than did the eastern part. The primary mechanisms for deposition of sediments in the study area are the transport of Himalayan matter by turbidity currents and river-diluted water and the transport of Indian matter to the study area by a surface circulation in the Bay of Bengal, particularly the East India Coastal Current.
文摘IM To study the relationship between trace element Mo and gastric cancer.MATERIALS AND METHODS Soil samples were collected according to its type in different areas of Jiangxi Province; available molybdenum content in soil was measured by catalytic polarography and rank correlation method was used to analyse correlation between the mean of soil available molybdenum and mortality rate of gastric cancer in each county and city in Jiangxi Province. Gastric cancer cases were selected from the authors′ hospital, occiput hair was collected to measure its molybdenum content with an atomic absorption spectrograph and controls were selected from the same hospital for comparison. Gastric cancer cases were selected from three hospitals at the same time, blood samples were taken on an empty stomach and serum molybdenum contents were measured with the atomic absorption spectrograph, and controls were selected from the same hospitals. Blind method was used in the whole course (chemical analysts did not know the source and nature of samples).RESULTS A negative correlation existed between soil available molybdenum content and mortality rate of gastric cancer (r=-0285, P<005); hair molybdenum contents of gastric cancer cases were lower than those of healthy controls (0308μg/g±0673μg/g and 0707μg/g±0561μg/g respectively, P<001); serum molybdenum contents of patients were also lower than those of healthy controls (2184μg/L±749μg/L and 2538μg/L±858μg/L respectively, P<005).CONCLUSION Deficiency of molybdenum may be one of the risk factors in gastric cancer.
