Establishment of a TLC Identification Method for Ensete wilsonii

[Objectives]The paper was to establish a TLC identification method for Ensete wilsonii.[Methods]Usingβ-sitosterol as the reference,the effects of preparation methods of test solutions,developing solvents,developing distances and color developing agents on TLC analysis were investigated,and the best TLC conditions for E.wilsonii were determined.[Results]The test solution prepared with 90%methanol solvent was dotted on TLC silica gel G plate,and developed with dichloromethane-toluene-methanol=10:5:1.5 as the developing solvent.Then the plate was sprayed with 10%sulfuric acid ethanol solution,and dried with hot blast for color development.Finally,the plate was examined under an ultraviolet lamp at 365 nm.The TLC results of E.wilsonii obtained showed good separation and color development effect,and the spots were clear and characteristic.[Conclusions]This method is safe,specific,and easy to operate,and can be used as a TLC identification method for E.wilsonii. 展开更多

[Objective] This study aimed to establish the standards for quality control of Qishen oral solution. [Method] Astragalus membranaceus, Atractylodes macro- cephala Koidz. and Glycyrrhiza uralensis in the preparation were identified by using thin layer chromatography (TLC), and Codonopsis pilosula was identified by using high-performance liquid chromatography (HPLC). [Result] By using the developed TLC system, experimental solution and reference solution showed clear spots, while negative control presented no interference. By using the developed HPLC system, the chromatographic peak of Iobetyolin was detected in experimental solution. [Con- clusion] The developed TLC and HPLC systems presented high specificity and good repeatability for identification of these four components and could be used in the quality control of Qishen oral solution. 展开更多

[Objectives] The moisture, ash and extract content of Dracocephalum tanguticum Maxim. were measured.[Methods] The moisture, total ash, acid-insoluble ash and extract content of D. tanguticum Maxim collected from 16 producing areas were detected by using the methods in Chinese Pharmacopoeia (2015 edition, volume I), and D. tanguticum Maxim. was identified by thin layer chromatography in Chinese Pharmacopoeia (2015 edition, volume I).[Results] The moisture content of D. tanguticum Maxim collected from 16 producing areas ranged from 8.68% to 10.36%, averaging 9.28%. The content of total ash was between 7.21% and 11.60%, averaging 9.89%. The content of acid-insoluble ash was 0.21%-2.71%, averaging 1.51%. The extract content (water-soluble) was 26.67%-42.91%, averaging 32.65%. According to the results of TLC identification, the 16 samples and reference substance had the same characteristic spots at the corresponding positions.[Conclusions] It was recommended that the moisture, total ash and acid-insolue ash content of D. tanguticum Maxim were not be more than 11%, 12%, and 3% respectively, while the extract content was not less than 26%. It provides reference for filling the gaps in the quality standards of D. tanguticum Maxim. 展开更多

Objective:To develop and validate an image analysis method for quantitative analysis ofγ-oryzanol in cold pressed rice bran oil.Methods:TLC-densitometric and TLC-image analysis methods were developed,validated,and used for quantitative analysis of γ-oryzanol in cold pressed rice bran oil.The results obtained by these two different quantification methods were compared by paired t-test.Results:Both assays provided good linearity,accuracy,reproducibility and selectivity for determination of γ-oryzanol.Conclusions:The TLC-densitomelric and TLC-image analysis methods providett a similar reproducibility,accuracy and selectivity for the quantitative determination of γ-oryzanol in cold pressed rice bran oil.A statistical comparison of the quantitative determinations of γ-oryzanol in samples did not show any statistically significant difference between TLC-densitometric and TLC-image analysis methods.As both methods were found to be equal,they therefore can be used for the determination of γ-oryzanol in cold pressed rice bran oil. 展开更多

[Methods]According to the method described in Chinese Veterinary Pharmacopoeia,thin layer chromatography and high performance liquid chromatography were used respectively for qualitative and quantitative identification of berberine hydrochloride contained in Jin Weng Zhili Particle by improving the conditions of extraction and purification. [Results]After optimizing TLC conditions of Jin Weng Zhili Particle,the separation effect of TLC of test samples was favorable. The spot color was clear,reproducibility was good,and precision was high. Under the conditions of mobile phase constituted by acetonitrile-water-glacial acetic acid-triethylamine( 28∶ 70∶ 1∶ 1),detection wavelength of 265 nm,flow rate of 1. 0 m L min^(-1)and column temperature of 35℃,the resolution of berberine hydrochloride was good; the injection volume in the range of 3. 15-50 μg· m L^(-1)had good linear relationship with the peak area( R^2= 0. 998); the average sample recovery rate(n=6) was 5. 672%,and RSD was 0. 864%; the content of berberine hydrochloride contained in Jin Weng Zhili Particle was 4. 023 mg. [ Method] With ethyl ace- tate-acetone-formic acid-water (10:7:2:0.5) as the developer and 10% sulfuric acid ethanol solution as the chromogenic reagent, the samples were heated at 100 ~C until the spots were clearly visible, and the existence of gardenoside was checked under natural light. With toluene-acetone(9:l ) as the developer and 5% vanillin sulfuric acid solution as the chromogenic reagent, the samples were heated at 105 ~C until the spots were clearly visible, and the existence of sarsasapogenin was checked under natural light. [Result] Qingwen baidu granules had the same spots with gardenoside and sarsasapogenin at the same Rfvalue under natural light. [ Conclusion] A TLC method detecting gardenoside and sarsasapogenin in Qingwen baidu granules was established, which had good specificity and repeatability, suitable for rapid detection of gardenoside and sarsasapogenin. 展开更多

[ Method ] With benzene-ethyl acetate-methanol-isopropanol-cancentrated ammonium liquid ( 12:6:3:3:1 ) as the developer, the samples were outspread in the cylinder with sat- urated ammonia steam by thin layer chromatography (TLC), and the existence of C. chinensis and berberine hydrochloride was checked under UV lamp. With tri- chloromethane-methanol (5:1) as the developer and 10% sulfuric acid ethanol solution as the chromogenic reagent, the samples were heated at 105 ℃ for 5 min to check the existence of F. suspense under natural light. [Result] Qingwen baidu granules had the same spots with C. chinensis control and berberine hydrochloride at the same Rf value under 365 nm UV lamp; Qingweu baidu granules had the same spots with F. suspensa control at the same Rfvalue under natural light. [ Condusion] A TLC method detecting C. chinensis and F. suspensa in Qingwen baidu granules was established, which had good specificity and repeatability, suitable for rapid detection of C. chinensis and F. suspensa. Anatomical and TLC study of three Chinese peonies were conducted to make a comparative analysis. Peonies (Paeonia suffruticosa (tree peony), Paeonia lactiflora (Chinese peony) and Paeonia veitchii (Chinese peony)) are traditionally used on the Qinghai-Tibet Plateau in China. Recent studies have shown that the peonies have different pharmacological activities and clinical applications. To distinguish these three species of peonies and ensure the safety and effectiveness in their use, the microscopic characteristics and chromatographic profile of their roots and the corresponding powder were studied. Plant materials sectioned and stained and the raw powder were studied with an optical microscope using standard techniques in microscopy. The results of microscopic features and TLC were described and illustrated. The three species have different microscopic characteristics and TLC profiles, which allow us to distinguish them. In fact, with the help of features semi-quantitative and qualitative, an identification key was developed in our work and illustrated with photos and a table. The aim of our work was to show that the optical microscopy and related techniques provide a achievable practicality, which can be applied without ambiguity to the authentication of species peonies. 展开更多

Methods: Polydatin was extracted by methanol and separated with chloroform-acetone-formic acid-water (4∶4∶0.5∶0.2) by thin layer chromatography. The excitation wavelength and emission wavelength were 284 nm and 384 nm, respectively. Results The linear regression equation of the calibration graph was y=7.02179x+4.5143, a linear regression correlative coefficient r=0.9936. Conclusion This method was proved simple, stable and sensitive. It can be used in quality control of herbs. 展开更多
