Establishment and characterization of a canine chondrosarcoma cell line:Mango

In the global progress of bone tumor research,established stable and long-lasting transgenic chondrosarcoma(CSA)cell lines are rare,mainly of murine and human origin,while the establishment of canine CSA cell lines has yet to be reported.This study established a canine CSA cell line to facilitate the basic clinical study of canine CSA.Fifty fve cases of canine osteolytic disease were collected,and more than 10 bone tumor samples from dogs with typical clinical signs were used for primary cell culture.A cell line with stable passaging for more than 100 generations and mouse tumorigenic ability was successfully cultured.According to the clinical characteristics of the dog and the histopathological results of the primary tumor,CSA was diagnosed,and the CSA cell line was designated Mango.Immunohistochemical(IHC)results showed that the immunoreactivity of bone gamma-carboxyglutamate protein(BGLAP),secreted protein acidic and rich in cysteine(SPARC),alkaline phosphatase(ALPL),vimentin(VIM)and S100 were positive.However,the immunoreactivity of pan-cytokeratin(PCK),chromogranin A(CGA),and platelet endothelial cell adhesion molecule-1(CD31)was negative.Immunofuorescence(IF)results showed that the protein expressions in the Mango cell line were consistent with the IHC identifcation of the primary tumor.The Mango cell line’s doubling time was 43.92 h,and the cell formation rate exceeded 20%.There were abnormal chromosome numbers,hetero staining with toluidine blue,and certain calcifcation abilities.It could be passaged stably and continuously without changing the cell morphology and characteristics.In vivo,the cells were successfully injected into the nude mice model with a tumorigenic rate of 100%.The immunophenotype of the xenograft tumor was consistent with that of the primary tumor.Therefore,we efectively established a canine CSA cell line.As a promising cell material,this cell line can be used to construct a tumor-bearing model conducive to the subsequent basic research of canine CSA.Moreover,because of its similarity to human CSA,the animal model of CSA is also indispensable for investigating human CSA.

Principle for the Installation of Accommodation for Sick guards in Public Health Establishments in Brazzaville in the Republic of Congo

The results of scientific studies of human social facts in the field of health show that the management of a patient should involve the patient’s entourage,whatever the status or size of the health establishment.In healthcare establishments in the Congo,the following are recognised as being responsible for medical care:specialist doctors,doctors,midwives,nurses and care assistants.The patient’s family and close friends are responsible for looking after the patient and financing care.The hospital infrastructure does not provide any space for the patient warden who accompany the patient during reception and hospitalisation.This makes Congolese hospitals inefficient for patient care.How can we integrate the function of the Sick guard and the assistance of the family,in order to reduce the mortality rate and repair the harm caused to patients requiring the presence of relatives during their stay in hospital,which is considered to be a dangerous place?This article examines the functional principles for configuring the space that patient warden would occupy in the patient care system.On the basis of a documentary analysis of sociological and architectural studies of existing facilities,this article proposes a typical accommodation model with the spaces needed to ensure the well-being and effectiveness of the patient warden with the patient.These are rooms with minimum space for 2 to 4 individual beds,equipped with toilets and showers.The accommodation has a dining area,kitchen and laundry facilities.In the future,this accommodation will become part of the hospital estate and may be occupied by orderlies and patient warden recruited by the hospital administration.

The Translation of Allusions in Political Discourse:An Intertextuality Perspective

drawn from ancient classics are the treasures of traditional Chinese culture.The allusions used in Xi Jinping:The Governance of China illustrate a distinctive feature of political discourse,and the publication of its English version provides a valuable opportunity for the study of allusions in translated political discourse.With the English translations of 241 allusions collected from two volumes of the book,we classified these allusions into three categories of history,culture,and literature based on their original sources.Then we analyzed the strategies used in translating the allusions,and provided typical examples to illustrate the use of such translation strategies for each category.Finally,we tried to account for the use of these strategies in light of the intertextuality theory.

Personification in Chinese Diplomatic Discourse and the Construction of National Identity Based on Corpus:A Case Study of the Regular Press Conference of Ministry of Foreign Affairs

With the development of the major country diplomacy with Chinese characteristics,diplomatic discourse,as an important part of building China’s international discourse power,has attracted much attention.Personification is a common means in diplomatic discourse.By giving anthropomorphic expressions to countries,regions,institutions and policies,the purpose of building national identity and establishing national image can be achieved.Based on conceptual metaphor theory and national identity theory,this paper focuses on personification in diplomatic discourse with a case study of the regular press conference of Ministry of Foreign Affairs.It is found that the spokesperson skillfully constructed China’s positive national identity,such as peace-loving,openness and inclusiveness by using a large number of body metaphor,kinship metaphor and role metaphor.

Ecological Discourse Analysis Based on the Corpus-A Case Study on the Environmental Discourse of 21st Century

Language learning materials not only provide language input for learners but also subtly disseminate ideology and influence readers’behavior.This study,based on the framework of ecological discourse analysis,quantitatively analyzes and qualitatively describes the ecological significance of environmental texts in the 21st Century.The aim is to provide references for the compilation of environmental discourse in language learning materials,promote the construction of more ecologically beneficial discourse,and facilitate the widespread dissemination of the ecological philosophy of“Diversity and Harmony,Interaction and Co-existence”among young readers.The initial findings of this study are as follows:In the environmental texts of the publication,a large number of relational processes and action processes are used to objectively represent the natural environment;the discourse tends to be ambiguous and beneficial,generally aligning with the ecological philosophy of“Diversity and Harmony,Interaction and Co-existence.”This can guide readers to correctly understand and face environmental issues,which is conducive to the healthy development of natural ecosystems.

The Youth’s View of Marriage in Chinese Mainstream Media Discourse-A Corpus-assisted Three-dimensional Discourse Analysis

The rising of aging and the declining of birth rates have forced the public to focus on the youth’s view on marriage.Based on critical discourse analysis and combined with Fairclough’s three-dimensional discourse analysis model,this paper builds a“Chinese media News Report Corpus on the topic of‘marriage’”whose news are collected from China Daily.It is found that the discourses are neutral and objective with regard to the advantages and disadvantages of marriage,but in general,it is still a traditional view of marriage that is inevitable and closely related to fertility.Although this is controlled by the policies and the social reasons including declining fertility rate,it deviates from the current view of the youth towards marriage,resulting in many serious consequences such as young people’s rejection.In addition,this research found that male and female have great differences in their views on marriage,and men’s resistance to marriage is far greater than that of women,which is departure from the public’s cognition.The reasons behind this need to be explored in order to solve the marriage and love problems of young people in today’s era and realize the healthy development of young marriage.

English Translation of Chinese Natural Ecological Discourse:A Corpus-based Case Study on Xi Jinping:The Governance of China

Amid the increasingly severe global natural eco-environment,it is necessary to build a natural ecological civilization by constructing an ecological civilization discourse.Against this background,this study compiles a corpus of natural ecological discourse in the English translation of Xi Jinping:The Governance of China(short for Xi).By using Wordsmith and AntConc,this study explores the linguistic features of the ecological discourse in English translation of Xi in the following dimensions,including high-frequency words,keywords,word collocations,concordance lines.This study aims to analyze the concepts and attitudes towards natural ecology,so as to provide certain valuable insights for the construction of China’s discourse on natural ecological civilization.The study found that the natural ecology discourse involving in the English translation of Xi turned out to be ecologically beneficial.

A Multimodal Critical Discourse Analysis of Lingnan Cultural Promotional Videos on Social Media

In recent years,more and more directors of culture and tourism have taken part in the promotion of local cultural tourism by cross-dressing,talent shows,and pushing their limits on self-media platforms.This study investigates short videos of Lingnan culture promoted by directors general and deputy directors general of the Culture,Radio,Television,Tourism,and Sports Bureau of counties and cities in Guangdong Province on social media by the method of multimodal critical discourse analysis.The analysis of 33 videos shows that Lingnan culture is a domineering and confident culture,historical culture,graceful and elegant culture,and vibrant and active culture.Domineering and confident culture is embedded in the utterances and behaviors of the directors general or deputy directors general in the video.Historical culture is realized through the conversation with historical figures through time travel.Graceful and elegant culture is constructed in the depiction of sceneries and the depiction of characters’manners.Vibrant and active culture is represented in the depiction of the characters’actional process and analytical process.

Discourse Analysis of the Belt and Road Initiative From the Perspective of Central Asian Countries: A Case Study of the Republic of Kazakhstan

This study employs Norman Fairclough’s Critical Discourse Analysis(CDA)three-dimensional model,using the Republic of Kazakhstan as a case study,to delve into the discourse construction of China’s Belt and Road Initiative(BRI)in Central Asian countries.Through detailed analysis of policy documents,media reports,and public discussions in Central Asian countries,this paper reveals how the BRI constructs specific social practices,discourse events,and textual meanings within these nations.The findings indicate that through this global development strategy,China has not only strengthened its economic ties with Central Asian countries but has also exerted profound influences on political,cultural,and social levels.

First report on establishment and characterization of the extrahepatic cholangiocarcinoma sarcoma cell line CBC2T-2

BACKGROUND Extrahepatic cholangiocarcinoma sarcoma is extremely rare in clinical practice.These cells consist of both epithelial and mesenchymal cells.Patient-derived cell lines that maintain tumor characteristics are valuable tools for studying the molecular mechanisms associated with carcinosarcoma.However,cholangiocarcinoma sarcoma cell lines are not available in cell banks.AIM To establish and characterize a new extrahepatic cholangiocarcinoma sarcoma cell line,namely CBC2T-2.METHODS We conducted a short tandem repeat(STR)test to confirm the identity of the CBC2T-2 cell line.Furthermore,we assessed the migratory and invasive properties of the cells and performed clonogenicity assay to evaluate the ability of individual cells to form colonies.The tumorigenic potential of CBC2T-2 cells was tested in vivo using nonobese diabetic/severe combined immunodeficient(NOD/SCID)mice.The cells were injected subcutaneously and tumor formation was observed.In addition,immunohistochemical analysis was carried out to examine the expression of epithelial marker CK19 and mesenchymal marker vimentin in both CBC2T-2 cells and xenografts.The CBC2T-2 cell line was used to screen the potential therapeutic effects of various clinical agents in patients with cholangiocarcinoma sarcoma.Lastly,whole-exome sequencing was performed to identify genetic alterations and screen for somatic mutations in the CBC2T-2 cell line.RESULTS The STR test showed that there was no cross-contamination and the results were identical to those of the original tissue.The cells showed round or oval-shaped epithelioid cells and mesenchymal cells with spindle-shaped or elongated morphology.The cells exhibited a high proliferation ratio with a doubling time of 47.11 h.This cell line has migratory,invasive,and clonogenic abilities.The chromosomes in the CBC2T-2 cells were polyploidy,with numbers ranging from 69 to 79.The subcutaneous tumorigenic assay confirmed the in vivo tumorigenic ability of CBC2T-2 cells in NOD/SCID mice.CBC2T-2 cells and xenografts were positive for both the epithelial marker,CK19,and the mesenchymal marker,vimentin.These results suggest that CBC2T-2 cells may have both epithelial and mesenchymal characteristics.The cells were also used to screen clinical agents in patients with cholangiocarcinoma sarcoma,and a combination of paclitaxel and gemcitabine was found to be the most effective treatment option.CONCLUSION We established the first human cholangiocarcinoma sarcoma cell line,CBC2T-2,with stable biogenetic traits.This cell line,as a research model,has a high clinical value and would facilitate the understanding of the pathogenesis of cholangiocarcinoma sarcoma.

In China,the main discourse on human rights can be classified into four categories:political discourse,policy discourse,institutional discourse,and academic discourse.These four categories show significant differences in terms of the context,content,mode of expression,characteristics,and functions of the discourse.They cannot be simply equated or interchangeable with one another.However,they also rely on,restrict,and promote each other,and under certain conditions,they can be transformed into one another.It is needed to prevent imbalances,mismatches in context,isolation,and inadequate translation among human rights discourses.Meanwhile,it is essential to promote balanced development among different discourses,where each discourse maintains its own boundaries,refers to one another,and undergoes accurate translation,in order to construct their healthy interrelationships.Exploring appropriate methods of translation between discourses is an important and worthwhile topic for research in Chinese human rights discourse.It holds significant practical significance and academic value in constructing the Chinese human rights discourse system.

Since the early 1970s,discourse analysis has garnered significant attention across various academic disciplines,marked by the proliferation of numerous theoretical frameworks.Then the profound impact of discourse analysis on the field of translation research has become increasingly apparent,with a substantial body of translation studies embracing the discourse analysis approach.The disciplinary significance of discourse analysis has become undeniable.Contemporary China is in a relatively weak position in global cultural communication.To advance Chinese culture on the global stage,Chinese literary works serve as a precious channel.To effectively showcase the unique qualities of Chinese literature,translators must consider a multitude of influencing factors and address them judiciously to enable target language readers to fully immerse themselves in the essence of Chinese literary artistry.This paper takes Yang Xianyi and Gladys Yang’s translation of Kong Yiji as an example to elucidate the function of discourse analysis in translation of Chinese literature.

Voice and Heart of Darkness:Centrality and Discourse in Colonialism

My essay on Heart of Darkness explores Conrad’s dramatization of the voice and heart dynamic in the novella as a penetration of the deep-lying rationale of colonial operation and more generally the human condition at the turn of the 20th century.Three different forms of assumed centrality in the novella-capital,Eurocentrism,and the African interior as detrimental void symbolized by the heart of darkness form concentric circles that interconnect and work in concert as the core colonial ideology and produce a sense of incongruity and absurdity.The voice of this colonial ideological apparatus disguised as the universal subject of humanity and rationality finds its best expression in the eloquence of Kurtz,the specimen of culture seduced by its very opposite-the wilderness-right in the heart of darkness.“The horror”,as his final verdict of life as a European with real intent of living on his own terms at the turn of the 20th century,sums up the predicament of a civilization with too much culture and self-absorption for its own good.

A Critical Discourse Analysis of Cyberbullying Language Based on Text-mining Techniques——A Case Study of Prince Harry and Meghan Markle

With the development of AI,a large amount of cyberbullying language flooded into the Internet.Cyberbullying language damages the online environment and causes mental harm to the victims of online bullying.Many researchers,therefore,have paid much attention to the problem.This study collected online comments targeted at Prince Harry and Meghan Markle as a corpus and then analyzed text data based on Critical Discourse Analysis by using text-mining tools to explore the factors that contribute to the social ideological effects of the cyberbullying language.The research results show that cultural differences,prejudice,or social exclusion due to race or gender form cyberbullying on social media.

The Construction of PLA Image in China’s Official Discourse From the Perspective of Positive Discourse Analysis——Based on Government Work Reports Over the Last 15 Years

Government Work Report is essential for domestic and international understanding of China’s national conditions.This paper selects the national defense section in Government Work Reports released by the State Council of the People’s Republic of China over the last fifteen years from 2009 to 2023 as an example of China’s official discourse.From the perspective of positive discourse analysis,the theory of three meta-functions of language proposed by M.A.K.Halliday in Systemic Functional Linguistics is used in this article in terms of transitivity,appraisal,mood,modality,and theme to explore the constructing of the People’s Liberation Army(PLA)image.This study finds that,by using the goals and circumstances with optimistic meaning in material processes,adopting more appreciation in attitude,applying more declaratives in mood,utilizing modal operators with high modulation,and implicit objective modal adjuncts,and employing government-focused themes,China’s official discourse objectively and authentically conveys a positive image of the brave and just PLA both at home and abroad which advocates itself to peace-keeping,and mission-fulfilling.
