Long noncoding RNAs HAND2-AS1 ultrasound microbubbles suppress hepatocellular carcinoma progression by regulating the miR-873-5p/tissue inhibitor of matrix metalloproteinase-2 axis

BACKGROUND Increasing data indicated that long noncoding RNAs(lncRNAs)were directly or indirectly involved in the occurrence and development of tumors,including hepatocellular carcinoma(HCC).Recent studies had found that the expression of lncRNA HAND2-AS1 was downregulated in HCC tissues,but its role in HCC progression is unclear.Ultrasound targeted microbubble destruction mediated gene transfection is a new method to overexpress genes.AIM To study the role of ultrasound microbubbles(UTMBs)mediated HAND2-AS1 in the progression of HCC,in order to provide a new reference for the treatment of HCC.METHODS In vitro,we transfected HAND2-AS1 siRNA into HepG2 cells by UTMBs,and detected cell proliferation,apoptosis,invasion and epithelial-mesenchymal transition(EMT)by cell counting kit-8 assay,flow cytometry,Transwell invasion assay and Western blotting,respectively.In addition,we transfected miR-837-5p mimic into UTMBs treated cells and observed the changes of cell behavior.Next,the UTMBs treated HepG2 cells were transfected together with miR-837-5p mimic and tissue inhibitor of matrix metalloproteinase-2(TIMP2)overexpression vector,and we detected cell proliferation,apoptosis,invasion and EMT.In vivo,we established a mouse model of subcutaneous transplantation of HepG2 cells and observed the effect of HAND2-AS1 silencing on tumor formation ability.RESULTS We found that UTMBs carrying HAND2-AS1 restricted cell proliferation,invasion,and EMT,encouraged apoptosis,and HAND2-AS1 silencing eliminated the effect of UTMBs.Additionally,miR-873-5p targets the gene HAND2-AS1,which also targets the 3’UTR of TIMP2.And miR-873-5p mimic counteracted the impact of HAND2-AS1.Further,miR-873-5p mimic solely or in combination with pcDNA-TIMP2 had been transformed into HepG2 cells exposed to UTMBs.We discovered that TIMP2 reversed the effect of miR-873-5p mimic caused by the blocked signalling cascade for matrix metalloproteinase(MMP)2/MMP9.In vivo results showed that HAND2-AS1 silencing significantly inhibited tumor formation in mice.CONCLUSION LncRNA HAND2-AS1 promotes TIMP2 expression by targeting miR-873-5p to inhibit HepG2 cell growth and delay HCC progression.

Non-use value assessment for wetland ecosystem service of Hongxing National Nature Reserve in northeast China

By the contingent value method, we studied the non-use value of wetland ecosystem service of Hongxing National Nature Reserve （HNNR） in Heilongjiang Pro- vince, northeast China. The proportion of respondents willing to pay （WTP） for protection of HNNR was 63 %. The WTP ratio was affected by geographical area, contact nature, personal preferences, and familiar degree of the respondents. The WTP value was affected by age, education level and career of the respondents. The mainly reasons for people rejecting to pay for protecting HNNR were ＂I am not familiar to HNNR＂ and ＂I had no capacity for additional spending because of low income＂. Weighted average individual WTP value was CNY 59.26 Yuan ind.^-1 year^-1 for all the respondents with WTP. The total non-use ecosystem service value of HNNR was CNY 1430 million Yuan in 2013. The heritage value was highest followed by existence value and option value. From the high ecosystem service value in HNNR, it is very important to construct nature reserve for protecting natural ecosystems and human sustainable use of natural resources.

A Compact Difference Scheme for Multi-point Boundary Value Problems of Heat Equations

In this paper,a compact difference scheme is established for the heat equations with multi-point boundary value conditions.The truncation error of the difference scheme is O(τ2+h^4),where t and h are the temporal step size and the spatial step size.A prior estimate of the difference solution in a weighted norm is obtained.The unique solvability,stability and convergence of the difference scheme are proved by the energy method.The theoretical statements for the solution of the difference scheme are supported by numerical examples.

Jacobi-Sobolev Orthogonal Polynomialsand Spectral Methods for Elliptic Boundary Value Problems

Generalized Jacobi polynomials with indexes α,β∈ R are introduced and some basic properties are established. As examples of applications,the second- and fourth-order elliptic boundary value problems with Dirichlet or Robin boundary conditions are considered,and the generalized Jacobi spectral schemes are proposed. For the diagonalization of discrete systems,the Jacobi-Sobolev orthogonal basis functions are constructed,which allow the exact solutions and the approximate solutions to be represented in the forms of infinite and truncated Jacobi series. Error estimates are obtained and numerical results are provided to illustrate the effectiveness and the spectral accuracy.

Clinical Value of Musculoskeletal Ultrasound in Rehabilitation After Flexor Tendon Rupture Repair of the Hand

Objective:To explore the application effect and clinical value of musculoskeletal ultrasound in the rehabilitation of hand function after flexor tendon rupture repair.Methods:In this study,72 patients were selected from among patients who underwent flexor tendon rupture repair of the hand in Yancheng Third People’s Hospital from May 2018 to May 2020;the patients were randomly divided into the control group(routine hand rehabilitation training)and the experimental group(musculoskeletal ultrasound and targeted hand rehabilitation training based on examination results)by die roll,with 34 cases in each group;the hand rehabilitation of the two groups were compared.Results:The excellent and good rate of the total active motion(TAM)of the experimental group(94.44%)was significantly higher than that of the control group(69.44%)(P<0.05);before treatment,there was no significant difference in the diameter and degree of stenosis of the artery in the finger between the two groups(P>0.05);after treatment,the degree of stenosis and the diameter of the artery of the experimental group were significantly better than those of the control group(P<0.05).Conclusion:For patients treated with flexor tendon rupture repair of the hand,the use of musculoskeletal ultrasound in the rehabilitation process can significantly improve the functional recovery of the hand;therefore,it is worthy of in-depth research,promotion,and application in clinical rehabilitation.

Reflection of Liberal Values in the Georgian Press in 1918-1921

This study deals with the values and orientation of Georgian social and political publications in the times of the Georgian Democratic Republic （1918-1921）. It also shows how the press of that time reflected the idea of ＂Georgian Europeanism＂ based on liberal values that took shape as an anti-colonial concept in the journalism of the late 19th century. The opposed dominant ideological projects of that era comprised on the one hand, National Democratic and Socialist Federalist parties that aspired to preserve national identity and carry out changes and on the other hand, ruling Social Democrats （who initially neglected national identity, but later shared the idea）. This opposition was clearly reflected in the media discourse of that time, which implied the following rights based on the principles of classical liberalism： freedom of conscience and expression, rule of law, personal independence, and individual rights. Other elements were added to this list later. The issue of religious tolerance also became topical. Journalist texts of the period reflected and rationalised the values recognised in today＇s democratic and liberal communities. The research was carried out in the archives of the periodicals of the aforementioned period （newspapers Sakhalkho Sakme, Sakartvelo, Ertoba, and Sakartvelos Respublika） by means of systematising and classifying retrieved materials and making a substantive and varietal analysis of individual media texts.

Abnormal Secondary Growth and Histochemical Localization of Alkaloids in Root System of Aconitum flavum Hand.-Mazz.

[Objective] The purpose of this study was to clarify the structure,growth pattern and histochemical localization of alkaloids in root system of Aconitum flavum Hand.-Mazz.[Method] Paraffin sectioning and histochemistry were employed for performing the analysis in this study.[Result] The root system of Aconitum flavum Hand.-Mazz.consists of taproot,lateral root and adventitious root.The primary structure of root system is normal,but secondary structure shows abnormal.The cambium and the extra cambium of taproot form a ＂U＂-shaped secondary vascular bundle and tertiary bundle in abnormal secondary structure.The sieve tube group is made of little sieve tube group which is differentiated from primary phloem and cambium.Meanwhile,the secondary xylem in tuberous root also appears to be a ＂U＂ shape.Parenchyma cells of secondary phloem occupy most of the tuberous root.The sieve tube group of tuberous root is mainly differentiated from parenchyma cell of secondary phloem.[Conclusion] The difference in abnormal secondary structure of taproot and tuberous root are attributed to their varied cambium compose and activity pattern.Alkaloids are mainly accumulated in parenchyma cell of the inside cortex and between bundle in taproot,while parenchyma of secondary phloem and pith in tuberous root.

Risk measurement of international oil and gas projects based on the Value at Risk method

International oil and gas projects feature high capital-intensity, high risks and contract diversity. Therefore, in order to help decision makers make more reasonable decisions under uncertainty, it is necessary to measure the risks of international oil and gas projects. For this purpose, this paper constructs a probabilistic model that is based on the traditional economic evaluation model, and introduces value at risk(VaR) which is a valuable risk measure tool in finance, and applies Va R to measure the risks of royalty contracts, production share contracts and service contracts of an international oil and gas project. Besides, this paper compares the influences of different risk factors on the net present value(NPV) of the project by using the simulation results. The results indicate:(1) risks have great impacts on the project's NPV, therefore, if risks are overlooked, the decision may be wrong.(2) A simulation method is applied to simulate the stochastic distribution of risk factors in the probabilistic model. Therefore, the probability is related to the project's NPV, overcoming the inherent limitation of the traditional economic evaluation method.(3) VaR is a straightforward risk measure tool, and can be applied to evaluate the risks of international oil and gas projects. It is helpful for decision making.

超敏C反应蛋白、降钙素原联合白细胞介素-6对手足口重症患儿的诊断价值研究

目的探讨超敏C反应蛋白(hypersensitive C-reactive protein,hs-CRP)、降钙素原(procalcitonin,PCT)联合白细胞介素-6(interleukin-6,IL-6)在手足口重症患儿中诊断价值及临床意义。方法收集2022年1月到2022年12月在嘉兴市第一医院住院治疗的62例手足口患儿为研究对象,根据患儿感染的严重程度分为观察组(重型感染组)29例及对照组(轻型感染组)33例,通过分析对比两组一般资料、白细胞总数、中性粒细胞计数、淋巴细胞计数、血小板计数、hs-CRP、PCT、IL-6及肌酸激酶同工酶(creatine kinase isoenzyme,CK-MB)等的差异及临床应用。结果观察组白细胞总数、中性粒细胞计数、淋巴细胞计数、hs-CRP、PCT、IL-6较对照组有升高,差异有统计学意义(P<0.05);受试者工作特征(receiver operator characteristic,ROC)曲线分析hs-CRP预测手足口重型感染敏感度为79.3%,特异性为93.9%(95%CI:0.852~0.985,P<0.05);PCT敏感度为93.1%,特异性84.8%(95%CI:0.907~1,P<0.05);IL-6敏感度为96.6%,特异性87.9%(95%CI:0.945~1,P<0.05)。结论在手足口分型诊断中,PCT、IL-6敏感度高,虽hs-CRP敏感度较前者低,但特异性较前者高,因此hs-CRP、PCT、IL-6三者联合对手足口诊断分型价值更高。

Role of 18F-fluorodeoxyglucose positron emission tomography imaging in surgery for pancreatic cancer

AIM： To evaluate the role of positron emission tomography using 18F-fluorodeoxyglucose （FDG-PET） in the surgical management of patients with pancreatic cancer, including the diagnosis, staging, and selection of patients for the subsequent surgical treatment. METHODS： This study involved 53 patients with proven primary pancreatic cancer. The sensitivity of diagnosing the primary cancer was examined for FDG-PET CT, cytological examination of the bile or pancreatic juice, and the serum levels of carcinoembrionic antigens （CEA） and carbohydrate antigen 19-9 （CA29-9）. Next, the accuracy of staging was compared between FDG-PET and CT. Finally, FDG-PET was analyzed semiquantitatively using the standard uptake value （SUV）. The impact of the SUV on patient management was evaluated by examining the correlations between the SUV and the histological findings of cancer. RESULTS： The sensitivity of FDG-PET, CT, cytological examination of the bile or pancreatic juice, and the serum levels of CEA and CA19-9 were 92.5%, 88.7%, 46.4%, 37.7% and 69.8%, respectively. In staging, FDG-PET was superior to CT only in diagnosing distant disease （bone metastasis）. For local staging, the sensitivity of CT was better than that of FDG-PEr. The SUV did not correlate with the pTNM stage, grades, invasions to the vessels and nerve, or with the size of the tumor. However, there was a statistically significant difference （4.6 ± 2.9 vs 7.8 ± 4.5, P = 0.024） in the SUV between patients with respectable and unresectable disease. CONCLUSION： FDG-PET is thus considered to be useful in the diagnosis of pancreatic cancer. However, regarding the staging of the disease, FDG-PET is not considered to be a sufficiently accurate diagnostic modality. Although the SUV does not correlate with the patho-histological prognostic factors, it may be useful in selecting patients who should undergo subsequent surgical treatment.

Relationship between serum calcium and CA 19-9 levels in colorectal cancer

AIM:To examine the calcium metabolism of colorectal cancer (CRC)in patients with colorectal cancer and control patients. METHODS:Seventy newly diagnosed CRC patients were included.The healthy control group was age and gender matched(n=32).Particular attention was devoted to the relationship between serum calcium of patients,and levels of AFP,CEA,carbohydrate antigen 19-9(CA 19-9)(that could be considered as prognostic factors).Furthermore,the Ca-sensing receptor(CaSR)gene A986S polymorphism was investigated in these patients,as well as the relationship between different CaSR genotypes and the data stated above. RESULTS:A lower level of ionized calcium(also corrected for albumin)was found in the serum of CRC patients with normal 25(OH)vitamin D levels.The ionized calcium concentration was inversely correlated with the serum level of CA.19-9.There was no difference in the distribution of CaSR genotypes,between CRC patients and general population.The genotypes did not correlate with other data examined. CONCLUSION:Based on these results,lower levels of serum calcium might be a pathogenic and prognostic factor in colorectal cancer.

AIM: To investigate the prognostic significance of pretreatment standardized maximum uptake value (SUVmax) and serum carbohydrate antigen (CA)19-9 in pancreatic cancer.

On the feasibility of re-stimulation of shale wells

As a result of advances in horizontal comple- tions and multi-stage hydraulic fracturing, the U.S. has been able to economically develop several decades of worth of natural gas. However, a considerable concern has risen on the economic viability of shale gas development for reasons associated with the fast production declines as well as recent down-turns of natural gas prices besides rises in the costs of new technologies. Therefore, an economic analysis is required to investigate the profitability of the re- fracturing treatment of unconventional gas resources. Net present value of cash flows and internal rate of return are calculated for a range of gas prices considering 20 years of natural gas production from a typical unconventional shale gas reservoir. A systematic comparison is then accom- plished for three scenarios： （1） re-fracturing versus no re- fracturing, （2） combination of re-fracturing and drilling new wells, and （3） time-dependent re-fracturing treatment. Further, this paper incorporates the cost of re-fracturing treatment, the cost of drilling a new horizontal well, the water treatment cost, as well as the current and future price of natural gas in the model. The findings of this work would help the future re-stimulation development plans of the emerging unconventional shale gas plays.

手足口病患儿皮质醇β-内啡肽和部分细胞因子监测价值

目的明确手足口病患儿皮质醇、β-内啡肽和部分细胞因子监测的临床价值。方法选取2011年1-6月该院收治的手足口病患儿（普通组30例、重型组30例）及健康儿童（对照组30例）,比较皮质醇、β-内啡肽、白细胞介素10（IL-10）、白细胞介素13（IL-13）、γ干扰素（IFN-γ）及肿瘤坏死因子-α（TNF-α）水平,并对其治疗前后各项指标水平进行对比。结果与对照组比较,普通组与重型组的皮质醇、β-内啡肽、IL-10、IL-13、IFN-γ及TNF-α水平均有所升高,组间比较,差异有统计学意义（P〈0.05）。随着病情的加重,皮质醇、β-内啡肽及细胞因子水平逐渐增加,且重型组与普通组的各项指标差异有统计学意义（P〈0.05）。普通组患儿治疗后,皮质醇、β-内啡肽、IL-10、IL-13、IFN-γ及TNF-α水平下降,接近对照组。重型组患儿治疗后,皮质醇、β-内啡肽、IL-10、IL-13、IFN-γ及TNF-α水平升高。两组患儿治疗前后皮质醇、β-内啡肽及细胞因子水平组内比较,差异有统计学意义（P〈0.05）。结论血清皮质醇、β-内啡肽及部分细胞因子水平对手足口病的病情判断及治疗具有重要的临床价值。

降钙素原、C-反应蛋白及血糖检测在手足口病患儿诊断中的应用价值

目的探讨降钙素原、C-反应蛋白及血糖检测在手足口病患儿诊断中的应用价值。方法选取我院2016年3月～2017年10月收治的80例手足口病患儿作为研究组,按照不同病情分为研究1组(40例)与研究2组(40例)。研究1组为急性期手足口病患儿,研究2组为恢复期手足口病患儿,同期选取健康儿童40例作为对照组,对所有手足口病患儿、健康儿童使用全自动生化分析仪AU640及配套试剂检测C-反应蛋白及血糖,使用双抗夹心法和化学发光分析检测降钙素原。比较各组的降钙素原、C-反应蛋白及血糖差异结果。结果研究1组患儿的降钙素原、C-反应蛋白及血糖水平高于研究2组及对照组,差异有统计学意义(P<0.05)。研究2组的降钙素原、C-反应蛋白及血糖浓度均高于对照组,差异有统计学意义(P<0.05)。80例研究组患儿中,降钙素原检测阳性25例,C反应蛋白检测阳性32例,血糖检测阳性26例,联合检测阳性65例;40例对照组中,降钙素原检测阳性2例,C反应蛋白检测阳性4例,血糖检测阳性3例,联合检测6例。联合诊断的灵敏度、特异度高于单独诊断,差异有统计学意义(P<0.05)。结论对手足口病患儿予以降钙素原、C-反应蛋白及血糖检测,其阳性率较高,使用三项联合检测能够将手足口病患儿的诊断灵敏度有效提高,这为临床治疗提供了可靠的诊断依据。

Study on Ba_(6-3x)Sm_(8+2x)Ti_(18)O_(54) Microwave Dielectric Ceramics

The effects of different x values on the dielectric properties in the Ba6-3xSm8+2xTi18O54 microwave dielectric ceramics(0.3≤x≤0.7) were studied. The results showed that the major crystal phase was BaSm2Ti4O12 and the sintering temperature of the ceramics was 1340 ℃ when the x value was 0.67. The dielectric properties were better: εr≈79, tanδ≈1×10-4(1 MHz); εr≈80, Q·f=9230 GHz(3.97 GHz), τf=-17 ppm·℃-1.
