Globozoospermia is diagnosed by the presence of 100% round-headed spermatozoa on semen analysis, and patients with this condition are absolutely infertile. The objective of this study was to investigate the differences in protein expression between human round- headed and normal spermatozoa. Two-dimensional (2-D) fluorescence difference gel electrophoresis (DIGE) coupled with mass spectrometry (MS) was used in this study. Over 61 protein spots were analysed in each paired normal/round-headed comparison, using DIGE technology along with an internal standard. In total, 35 protein spots identified by tandem mass spectrometry (MS/MS) exhibited significant changes (paired t-test, P 〈 0.05) in the expression level between normal and round-headed spermatozoa. A total of nine proteins were found to be upregulated and 26 proteins were found to be downregulated in round-headed spermatozoa compared with normal spermatozoa. The differentially expressed proteins that we identified may have important roles in a variety of cellular processes and structures, including spermatogenesis, cell skeleton, metabolism and spermatozoa motility.展开更多

Around the Lembeh Strait, there exist some north-south differences in terms of water mass properties. NPTIW is only found in the southern Lembeh Strait. Water mass with the salinity of 34.6 is only detected at 200–240 m between NPTW and NPTIW in the southern Lembeh Strait, and results from the process of mixing between the saltier water transported from the South Pacific Ocean and the lighter water from the North Pacific Ocean and Sulawesi Sea. According to the analysis on mixing layer depth, it is indicated that there exists an onshore surface current in the northern Lembeh Strait and the surface current in the Lembeh Strait is southward. These dramatic differences of water masses demonstrate that the less water exchange has been occurred between the north and south of Lembeh Strait. In 2015, the positive wind stress curl covering the northern Lembeh Strait induces the shoaling of thermocline and deepening of NPIW, which show that the north-south difference of airsea system is possible of inducing north-south differences of seawater properties.展开更多

Phys. 218, 353(2006)]is favored due to its good stability, high efficiency, and large density ratio. However, in terms of mass conservation, this model is not satisfactory during the simulation computations. In this paper, a mass correction is introduced into the ZSC model to make up the mass leakage, while a high-order difference is used to calculate the gradient of the order parameter to improve the accuracy. To verify the improved model, several three-dimensional multiphase flow simulations are carried out, including a bubble in a stationary flow, the merging of two bubbles, and the bubble rising under buoyancy. The numerical simulations show that the results from the present model are in good agreement with those from previous experiments and simulations. The present model not only retains the good properties of the original ZSC model, but also achieves the mass conservation and higher accuracy.展开更多

However, little information is presently available regarding the use of 2D-DIGE to investigate mechanisms of ischemic cerebrovascular diseases. Plasma and body fluids have been utilized in proteomic technology to study ischemic cerebrovascular diseases. OBJECTIVE: To perform proteomic analysis of fresh rat brain tissue in peripheral ischemic regions using 2D-DIGE 6 hours after middle cerebral artery occlusion (MCAO), and to identify specific proteins closely associated with early ischemic cerebrovascular diseases. DESIGN, TIME AND SETTING: Proteomics-based, randomized, controlled, animal experiment was performed at the Laboratories of Neurology and Proteomics, Jilin University between January and April 2006. MATERIALS: 2, 3, 5-triphenyl tetrazolium chloride was purchased from Sigma, USA. Ettan DALTSix system, DeCyder DIA V5.0 differential analysis software, and Ettan matrix-assisted laser desorption/ionization time-of-flight mass spectrometer (MALDI-TOF-MS) were purchased from Amersham Bioscience, Sweden. METHODS: Eight healthy, male, Wistar rats were randomized to experimental and control groups, with four rats in each group. In the experimental group, rat models of focal cerebral ischemia were established by MCAO. In the control group, the internal and external carotid arteries were exposed and then immediately sutured, and the remaining procedures were identical to the experimental group. MAIN OUTCOME MEASURES: At 6 hours after cerebral ischemia, protein expression in the peripheral ischemia region of the experimental group was compared with the control group using 2D-DIGE. Protein spots that exhibited statistical differences between experimental and control groups with 〉 1.4 attributable risk were screened using DeCyder DIA V5.0 differential analysis software. Differential proteins were identified using MALDI-TOF-MS. RESULTS: Triphenyl tetrazolium chloride staining results revealed pink, normal brain tissue and white, ischemic brain tissue, suggesting successful MCAO establishment. The average matching rate of four 2D-DIGE gels was 92.4%. There were (1 758 ± 43) protein spots on each gel, with similar distribution modes. At 6 hours after focal cerebral ischemia, 13 protein spots exhibited marked expression changes, including significantly increased (n = 7) and decreased (n = 6) expression (P 〈 0.05). MALDI-TOF-MS results revealed two differential protein spots: a-tubulin and heat shock protein 27, which were significantly decreased in the experimental group compared with the control group (P 〈 0.05). CONCLUSION: Thirteen protein spots with expression changes were revealed by 2D-DIGE proteomics technology. Of them, a-tubulin and heat shock protein 27 expressions were markedly decreased during the early stage of cerebral ischemia. These two proteins were presumed to be proteins associated with early ischemic cerebrovascular diseases.展开更多

An appropriate finite difference method was used to calculate the size dissolution rate, dissolution time and mass of alumina dissolved employing commercial software and custom algorithm based on the shrinking sphere assumption. The effects of some convection and thermal condition parameters on the dissolution process were studied. The calculated results show that the decrease of alumina content or the increase of alumina diffusion coefficient is beneficial for the increase of size dissolution rate and the decrease of dissolution time of non-agglomerated particles. The increase of bath superheat or alumina preheating temperature results in the increase of size dissolution rate and the decrease of dissolution time of agglomerated particles. The calculated dissolution curve of alumina(mass fraction of alumina dissolved) for a 300 k A aluminum reduction cell is in well accordance with the experimental results. The analysis shows that the dissolution process of alumina can be divided into two distinct stages: the fast dissolution stage of non-agglomerated particles and the slow dissolution stage of agglomerated particles, with the dissolution time in the order of 10 and 100 s, respectively. The agglomerated particles were identified to be the most important factor limiting the dissolution process.展开更多

To achieve a better un- derstanding of the difference in OA in summer and autumn, a high-resolution time-of-flight aerosol mass spectrometer (HR- ToF-AMS, Aerodyne Research Inc., USA) was deployed in urban Beijing in August and October 2012. The mean OA mass concentration in autumn was 30 4-30 μg m-3, which was higher than in summer (13 4-6.9 μg m-3). The elemental anal- ysis found that OA was more aged in summer (oxygen-to-carbon (O/C) ratios were 0.41 and 0.32 for summer and autumn, respectively). Positive matrix factorization (PMF) analysis identified three and five components in summer and autumn, re- spectively. In summer, an oxygenated OA (OOA), a cooking-emission-related OA (COA), and a hydrocarbon-like OA (HOA) were indentified. Meanwhile, the OOA was separated into LV-OOA (low-volatility OOA) and SV-OOA (semi-volatile OOA); and in autumn, a nitrogen-containing OA (NOA) was also found. The SOA (secondary OA) was always the most important OA component, accounting for 55% of the OA in the two seasons. Back trajectory clustering analysis found that the origin of the air masses was more complex in summer. Southerly air masses in both seasons were associated with the highest OA loading, while northerly air masses were associated with the lowest OA loading. A preliminary study of OA components, especially the POA (primary OA), in different periods found that the HOA and COA all decreased during the National Day holiday period, and HOA decreased at weekends compared with weekdays.展开更多

METHODS: Five hundred and fifty-six consecutive patients positive for HCV antibody and treatment-nafive HCC diagnosed between 1995 and 2004 were analyzed. Patients were classified into three groups according to age at HCC onset: < 60 years (n = 79), 60-79 years (n = 439), or ≥ 80 years (n = 38). Differences among groups in terms of sex, body mass index (BMI), lifestyle characteristics, and liver function were assessed. Factors associated with HCC onset in patients < 60 or ≥ 80 years were analyzed by logistic regression analysis. RESULTS: Significant differences emerged for sex, BMI, degree of smoking and alcohol consumption, mean bilirubin, alanine aminotransferase (ALT), and γ-glutamyl transpeptidase (GGT) levels, prothrombin activity, and platelet counts. The mean BMI values of male patients > 60 years old were lower and mean BMI values of female patients < 60 years old were higher than those of the general Japanese population. BMI > 25 kg/m2 [hazard ratio (HR), 1.8, P = 0.045], excessive alcohol consumption (HR, 2.5, P = 0.024), male sex (HR, 3.6, P = 0.002), and GGT levels > 50 IU/L (HR, 2.4, P = 0.014) were independently associated with HCC onset in patients < 60 years. Low ALT level was the only factor associated with HCC onset in patients aged ≥ 80 years. CONCLUSION: Increased BMI is associated with increased risk for early HCC development in HCV-infected patients. Achieving recommended BMI and reducing alcohol intake could help prevent hepatic carcinogenesis.展开更多

As a consequence, they only apply when the rock mass is homogeneous and so exclude many cases of practical interest. We describe a robust and fast numerical technique that solves the tunnel unloading problem and estimates the ground reaction curve for a cylindrical cavity excavated in a rock mass with properties depending on the radial coordinate, where the solution is no longer self-similar. The solution is based on a continuation-like approach(associated with the unloading and with the incremental formulation of the elasto-plastic behavior), fnite element spatial discretization and a combination of explicit sub-stepping schemes and implicit techniques to integrate the constitutive law, so as to tackle the diffculties associated with both strong strain-softening and elasto-brittle behaviors. The developed algorithm is used for two practical ground reaction curve computation applications. The frst application refers to a tunnel surrounded by an aureole of material damaged by blasting and the second to a tunnel surrounded by a ring-like zone of reinforced(rock-bolted) material.展开更多

MEE has been widely identified in all large mountains, but how it could be measured and what its main forming-factors are still remain open. This paper, supposing that the local mountain base elevation (MBE) is the main factor of MEE, takes the Qinghai-Tibet Plateau (QTP) as the study area, defines MEE as the temperature difference (AT) between the inner and outer parts of mountain massifs, identifies the main forming factors, and analyzes their contributions to MEE. A total of 73 mountain bases were identified, ranging from 708 m to 5081 m and increasing from the edges to the central parts of the plateau. Climate data (1981-2010) from 134 meteorological stations were used to acquire ATby comparing near-surface air temperature on the main plateau with the free-air temperature at the same altitude and simi- lar latitude outside of the plateau. The AT for the warmest month is averagely 6.15 ~C, over 12~C at Lhatse and Baxoi. A multivariate linear regression model was developed to simulate MEE based on three variables (latitude, annual mean precipitation and MBE), which are all significantly correlated to AT. The model could explain 67.3% of MEE variation, and the contribution rates of three independent variables to MEE are 35.29%, 22.69% and 42.02%, respectively. This confirms that MBE is the main factor of MEE. The intensive MEE of the QTP pushes the 10~C isotherm of the warmest month mean temperature 1300-2000 m higher in the main plateau than in the outer regions, leading the occurrence of the highest timberline (4900 m) and the highest snowline (6200 m) of the Northern Hemisphere in the southeast and southwest of the plateau, respectively.展开更多

In this study, we consider both strong concentrations (n = 0) and weak concentrations (n = 1/2). The governing equations have been transformed into nonlinear ordinary differential equations by applying the similarity transformation and have been solved numerically by using the finite difference method (FDM) and analytically by using (DTM). The effects of various governing parameters, namely, material parameter, radiation parameter, magnetic parameter, Prandtl number, Schmidt number, chemical reaction parameter and Soret number on the velocity, microrotation, temperature and concentration have been computed and discussed in detail through some figures and tables. In order to verify the accuracy of the present results, we have compared these results with the analytical solutions by using the differential transform method (DTM) and the multi-step differential transform method (MDTM). It is observed that this approximate numerical solution is in good agreement with the analytical solution.展开更多

文摘Surface mass anomalies estimated by mass concentration(mascon)approach using Gravity Recovery and Climate Experiment(GRACE)observations with regularization constraints generally present higher spatial resolution than the spheric harmonic(SH)solutions. To analyze the influence of different types of constraints on the estimation of mascon solutions,we carried out a closed-loop simulation experiment to estimate surface mass anomalies over South America based on simulated GRACE intersatellite geopotential differences.Tikhonov regularization with spatial constraint(SC),uniform weighting constraint(UWC),and a prior information constraint(APC)were employed to stabilize the mascon solutions, and the corresponding optimal regularization parameters were determined based on the minimum residual root-mean-square(RMS)criterion.The results show that mascon solutions estimated under different types of constraints are consistent and equivalent when the optimal regularization parameters are selected. The spatial distributions and main characteristics of regional surface mass anomalies estimated by the three types of constraints agree well, and the values of residual RMS with different constraints are very close. But due to the smoothing effect of regularization, the signal strength of mascon solutions is a bit weaker than that of original true signal, especially in the regions with strong signals. In addition, due to the ill-conditioned problem is more serious for higher grid resolution, the relative contribution of the three types of constraints to the final mascon solutions would be stronger. The results show that the averages of relative contribution percentages of these constraints for 2°×2° mascon grids are 80%-90%,while the corresponding values for 4°×4° mascon grids are 30%-60%.However,based on the minimum residual RMS criterion, the accuracy of estimation results is not affected by the type of constraints and their relative contribution to the final mascon solutions.

文摘BACKGROUND: The use of fluorescent two-dimensional difference gel electrophoresis (2D-DIGE) has been shown to compensate for the shortcomings of conventional two-dimensional gel electrophoresis, such as poor repeatability and large systematic errors. These two proteins were presumed to be proteins associated with early ischemic cerebrovascular diseases.

文摘Tungsten(W)is an important material in tokamak walls and divertors. The W ion charge state distribution and the dynamic behavior of ions play important roles in the investigation of plasma–wall interactions using laser-ablation-based diagnostics such as laser-induced breakdown spectroscopy and laser-induced ablation spectroscopy. In this work,we investigate the temporal and spatial evolutions of differently charged ions in a nanosecond-laser-produced W plasma in vacuum using time-of-flight mass spectroscopy.Ions with different charge states from 1 to 7(W+to W7+)are all observed. The temporal evolutions of the differently charged ions show that ions with higher charge states have higher velocities, indicating that space separation occurs between the differently charged ion groups.Spatially-resolved mass spectroscopy measurements further demonstrate the separation phenomenon. The temporal profile can be accurately fitted by a shifted Maxwell–Boltzmann distribution, and the velocities of the differently charged ions are also obtained from the fittings. It is found that the ion velocities increase continuously from the measured position of 0.75 cm to 2.25 cm away from the target surface, which indicates that the acceleration process lasts through the period of plasma expansion. The acceleration and space separation of the differently charged ions confirm that there is a dynamic plasma sheath in the laser-produced plasma, which provides essential information for the theoretical laser-ablation model with plasma formation and expansion.

文摘Using the technique of integration within an ordered product of operators we present a convenient approach for introducing the squeezing operator for the entangled states of two entangled particles with different masses. We also introduce one-sided squeezing operators.
文摘We study a multispecies one-dimensional Calogero model with two- and three-body interactions. Here, we factorize the ground stateout of the Hamiltonian H in order to get the new operatorwhich preserves some spaces of polynomialsin the case of equal masses, i.e. (the usual Calogero model) and in the case with different masses. The spectrum of these both cases is found easily.

文摘A comprehensive heat and mass transfer model of dissolution process of non-agglomerated and agglomerated alumina particles was established in an aluminum reduction cell. The agglomerated particles were identified to be the most important factor limiting the dissolution process.

文摘Fine particle of organic aerosol (OA), mostly arising from pollution, are abundant in Beijing. A preliminary study of OA components, especially the POA (primary OA), in different periods found that the HOA and COA all decreased during the National Day holiday period, and HOA decreased at weekends compared with weekdays.
文摘AIM: To identify factors associated with the age at onset of hepatitis C virus (HCV)-related hepatocellular carcinoma (HCC). Achieving recommended BMI and reducing alcohol intake could help prevent hepatic carcinogenesis.

文摘Fast methods to solve the unloading problem of a cylindrical cavity or tunnel excavated in elasto-perfectly plastic, elasto-brittle or strain-softening materials under a hydrostatic stress feld can be derived based on the self-similarity of the solution. The frst application refers to a tunnel surrounded by an aureole of material damaged by blasting and the second to a tunnel surrounded by a ring-like zone of reinforced(rock-bolted) material.

文摘Mass elevation effect (MEE) refers to the thermal effect of huge mountains or plateaus, which causes the tendency for tem- perature-related montane landscape limits to occur at higher elevations in the inner massifs than on their outer margins. The intensive MEE of the QTP pushes the 10~C isotherm of the warmest month mean temperature 1300-2000 m higher in the main plateau than in the outer regions, leading the occurrence of the highest timberline (4900 m) and the highest snowline (6200 m) of the Northern Hemisphere in the southeast and southwest of the plateau, respectively.
文摘The paper investigates the numerical solution of problem of magnetohydrodynamic (MHD) micropolar fluid flow with heat and mass transfer towards a stagnation point on a vertical plate. It is observed that this approximate numerical solution is in good agreement with the analytical solution.
