Macroalgae serve as a potential feedstock for fucoxanthin extraction.Fucoxanthin,a bioactive pigment found in the chloroplasts of marine algae,exhibits significant pharmacological properties.As a member of the caroten...Macroalgae serve as a potential feedstock for fucoxanthin extraction.Fucoxanthin,a bioactive pigment found in the chloroplasts of marine algae,exhibits significant pharmacological properties.As a member of the carotenoid family,fucoxanthin plays a crucial role in both the food and pharmaceutical industries.This research explores the effects of ultrasonics on the extraction of fucoxanthin from the marine macroalga Padina australis.In addition,various extraction techniques and the influence of solvents on the efficient separation of fucoxanthin from algae have been studied and compared.Using methanol,chloroform,and a combination of methanol and chloroform(1:1,v/v),conventional fucoxanthin extraction from Padina australis yielded 8.12 mg of fucoxanthin per gram of biomass.However,the ultrasonic-assisted extraction resulted in a significantly higher yield of 16.9 mg of fucoxanthin per gram of biomass,demonstrating that the use of ultrasonics enhances the extraction rate compared to conventional methods.Therefore,the efficient separation of fucoxanthin from Padina australis is highly dependent on ultrasonic-assisted extraction.The process conditions for the extraction were optimized to maximize the yield of fucoxanthin from seaweeds.The following parameters were selected for optimization studies:moisture content,particle size,mixing speed,extraction temperature,extraction duration,and solid-to-solvent ratio.The extracted fucoxanthin exhibited various biological activities,including antimicrobial and antioxidant properties,and its structure was elucidated through FTIR and NMR spectroscopy.Additionally,thin-layer chromatography of the crude algae extracts confirmed the presence of fucoxanthin in the marine algae.Given these findings,the optimized extraction process holds the potential for scaling up to large-scale fucoxanthin production.Fucoxanthin,as a potent pharmacological agent,offers promising applications in the treatment of various ailments.展开更多

It also aimed to identify phenolic acids present in the extracts. The 2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and ferric thiocyanate lipid peroxidation antioxidant assays were performed. High performance liquid chromatography was used to identify phenolic acids. There was no solvent effect on TPC nor on scavenging activities, and inhibition of lipid peroxidation (p > 0.05) among solvent extracts. On the other hand, 1:1:3 water: acetone: methanol extract (10.22 mg GAE/g sample) had significantly higher reducing potential than 50% ethanol extract (EE) (9.259 mg GAE/g sample) (p < 0.05);but EE was not significantly different from 80% methanol extract (9.781 mg GAE/g sample) (p > 0.05). Phenolic fraction designated as fraction 4 had the highest antioxidant activity (p < 0.05) with 69.49% ABTS scavenging activity and FRAP reducing potential, 22.26 mg of GAE/g sample. [Objectives]To establish the chromatographic fingerprint of Gancao Qinlian Extracts(GQE)and reveal the possible material basis of the anti-inflammatory effect of GQE by the correlation analysis between the fingerprint...[Objectives]To establish the chromatographic fingerprint of Gancao Qinlian Extracts(GQE)and reveal the possible material basis of the anti-inflammatory effect of GQE by the correlation analysis between the fingerprint chromatographic peaks of different components of GQE and its anti-inflammatory activity.[Methods]Ultra-performance liquid chromatography(UPLC)was used to detect the different ingredients of GQE to establish its chromatographic fingerprint and analyze the differences among the three medicine components;LPS stimulated RAW264.7 cells to construct an inflammatory cell model.The NO secretion of cells was detected by the Griess method.ELISA was used to detect the changes in TNF-αand IL-10 contents.RT-qPCR tested the mRNA expression levels of TNF-αand IL-10.Grey relational analysis was carried out by combining fingerprint chromatographic peak data and anti-inflammatory activity data.[Results]The GQE fingerprint was established,34 fingerprint characteristic peaks were calibrated,and 33 related chromatographic peaks were screened out.The corresponding chromatographic peaks in the three components were obtained,and the content of the components was calculated;the anti-inflammatory results showed that the content of NO,TNF-α,and the expression of TNF-αmRNA in the high and medium-dose groups of GQE were significantly lower than those in the blank group(P<0.01).The NO content and TNF-αmRNA expression in the high-dose group of GQE I was considerably lower than those in the blank group(P<0.01).The secretion of NO,TNF-α,and the expression of TNF-αmRNA in the high,medium,and low dose groups of GQE II were significantly lower than those in the blank group(P<0.01);the results of grey relational analysis showed that the correlation degree of the three components was GQE II>GQE>GQE I,and the characteristic fingerprint peaks 12,15,22,23,28,31,33 may be closely related to the anti-inflammatory effect.[Conclusions]The best component of the anti-inflammatory effect in GQE is water-soluble component,and its main components are flavonoids and alkaloids.These components can alleviate cellular inflammatory damage by inhibiting the excessive secretion of NO and reducing the expression of TNF-αmRNA.展开更多

This study aimed to investigate the flavonoid profiles and antioxidant activity of Paeonia lactiflora petal flavonoid extract(PPF). The UHPLC-ESI-Q-Exactive HF MS/MS method was established for characterization, and 21 predominant flavonoid compounds were tentatively identified in PPF. Among them, isoscutellarein-7-(6’-acetylallosyl-(1->2)-glucoside) and scutellarin methylester were discovered in PPF for the first time. Pretreatment with PPF significantly reduced H2O2-induced cell damage, ROS accumulation, and malondialdehyde content and increased the activity of SOD, CAT, and GSH-Px in buffalo rat liver 3A(BRL3A) cells. Moreover, the expression of nuclear Factor E2-related factor(Nrf2) was upregulated by PPF, whose expression trend was consistent with that of heme oxygenase-1(HO-1), glutamate-cysteine ligase catalytic subunit(GCLC), and NAD(P)H quinone oxidoreductase-1(NQO1). These findings suggested that herbaceous peony flavonoids can be used as a natural bioactive agent to prevent oxidative stress.展开更多

The obtained plant extracts were then diluted to determine their nematici- dal activities. [Result] The Chuanxiong extract had the highest nematicidal activity, followed by cocklebur extract, agrimony extract, basil extract, garlic extract, ragweed extract. [Method] Fibronectin, immunoglobulin, and hemoglobin were isolated from porcine blood, and SOD was extracted. Trace pyrogallol self-oxidation method to determine SOD activity was modified by optimizing the volume of pyrogallol and SOD samples, reaction temperature, and buffer pH. The specific activity of SOD was determined with the optimized extraction conditions. [ Result] The improved experimental conditions of SOD activity detection were as follows: 7 pyrogallol (50 mmol/L), 3 ml Tris-HCI (50 mmol/L, pH 8.2), reactive temperature at 25(3, and 10 pl SOD sample solution. The specific activity of extracted SOD was 5 056 U/mg protein. [ Conclusion] Four kinds of bioactive substance can be isolated from porcine blood by modern biological engi- neering integration technology, and the extracted SOD has better activity.展开更多
[Objective] The antifungal activity of the extracts from,Atractylodes macracephal Koidz and Pulsatilla chinensis Bunge Regel,against Botrytis cinerea and Alternaria solani were studied under the condition of laborator...[Objective] The antifungal activity of the extracts from,Atractylodes macracephal Koidz and Pulsatilla chinensis Bunge Regel,against Botrytis cinerea and Alternaria solani were studied under the condition of laboratory,in order to develop and utilize these two plants.[Method] The mycelium growth rate test was applied to measure the antifungal activities of extracts against fungi.[Result] the extracts of all the two plants showed strong antifungal activity against the target pathogenic fungi,especially the antifungal activity of the extract from Pulsatilla chinensis Bunge Regel was stronger and more stable.The inhibition rate to the mycelium growth of Botrytis cinerea was 80.25%.At the same concentration,the extract from Atractylodes macracephal Koidz showed little inhibition to Botrytis cinerea and Alternaria solani.The petrolelum ether extract of Atractylodes macracephal Koidz showed stronger antifungal activities and the EC50 was 5.31 mg/ml,and the n-butanol extract of Pulsatilla chinensis Bunge Regel showed stronger antifungal activities and the EC50 was 2.93 mg/ml.[Conclusion] The extracts from Pulsatilla chinensis Bunge Regel showed the stronger antifungal activity against Botrytis cinerea and Alternaria solani.展开更多

Effects of extraction conditions including extraction temperature, ethanol concentration, extraction time and liquid-to-solid ratio on extraction were explored with single-factor experiment, and the extrac- tion process was optimized with orthogonal experiment. [Result] Based on the single- factor experiment and orthogonal experiment, the optimal extraction condition for crude anti-allergic active extracts of citrus peels were determined to be under extraction temperature of 60℃, ethanol concentration of 70%, extraction time of 60 min, liquid-to-solid ratio of 1:25 and twice extraction. [Concluslon] This process condition is of simple operation and high extraction efficiency, which is environment-friendly and suitable for the extraction of crude anti-allergic active extracts from citrus peels. The fermentation effect of the isolate was the best under the condition of temperature 30 ~C, pH 7, liquid volume 75 ml/250 ml, inoculation volume 20% and culture time 48 h. The antibacterial effect of the isolate was the best when extract- ed by 80% saturated ammonium sulfate. The antibacterial crude protein had strong resistance to heat and acid. Organic solvent and UV irradiation had some influences on antibacterial crude protein. Proteases had hydrolytic effects on antibacterial crude protein. The isolated B. subti/is can be used to prevent and control the diseases caused by S. aureus, streptococcus and swine erysipelas, and can regulate intesti- nal microecology by adding into expanded feeds.展开更多

The toxicity and antifeedant activity on wheat aphid were determined, [ Result ] Walnut green husk extracts in the solvent of ethanol, acetone, ethyl acetate had higher toxicity and antifeedant activity on wheat aphid. When the concentration was 100 mg/ml, after 24 h, the toxicity and antifeedant activity of three kinds extracts on wheat aphid were greater than 84.35% and 66.98%, respectively. After 48 h, the toxicity and antifeedant activity were greater than 85.91% and 79.34%, respectively. Among 5 kinds extracts,ethanol extracts had the highest toxicity and antifeedant activity on wheat aphid. After 24 h, the corrected mortality rate was 93.64% and the antifeedant rate was 83.61%. After 48 h,the corrected mortality rate was 94.95% and antifeedant rate was 92.21%. Petroleum ether extract had the smallest toxicity and antifeedant activity on wheat aphid. After 24 h,the corrected mortality rate and antifeedant rate were 19.54% and 34.65%, respectively. After 48 h,the corrected mortality rate and antifeedant rate were 20.13% and 39. 03%, respectively. [ Conclsion] The extracts from walnut green husk with larger polarity solvent had better toxicity and higher antifeedant activity on wheat aphid.展开更多

Polysaccharides are the important biologically active components found in the peel of Dioscorea opposita Thunb.(DTTP).The influences of 4 extraction methods,namely hot water extraction(W),acidic extraction(HA),hot-com...Polysaccharides are the important biologically active components found in the peel of Dioscorea opposita Thunb.(DTTP).The influences of 4 extraction methods,namely hot water extraction(W),acidic extraction(HA),hot-compressed water extraction(HCW)and enzyme-assisted extraction(EAE),on the yields,physicochemical properties,hypoglycemic and antioxidant activities of polysaccharides from DTTP were studied and compared.Among these DTTP polysaccharides,DTTP-HA was outstanding in several respects.DTTP-HA was the most water soluble;it had the highest total carbohydrate content(85.08%),the highest uronic acid(13.20%)and the highest thermal stability.DTTP-HA and DTTP-W possessed a triple-helix structure.These 4 kinds of polysaccharides have the same types of monosaccharides,but in different molar percentages.Extraction method had a significant impact on the microstructures of the extracted polysaccharides.DTT-HA exhibited irregular structure with many holes.Among the 4 extracted methods,the DTTP-HA and DTTP-W initially exhibited higher hypoglycemic and antioxidant activities.The better bioactivities of DTTP-HA may be related to the above factors.The findings indicated that acid extraction is an effective method to extract polysaccharides with high biological activities from DTTP.展开更多
Human Action Recognition(HAR)and pose estimation from videos have gained significant attention among research communities due to its applica-tion in several areas namely intelligent surveillance,human robot interaction...Human Action Recognition(HAR)and pose estimation from videos have gained significant attention among research communities due to its applica-tion in several areas namely intelligent surveillance,human robot interaction,robot vision,etc.Though considerable improvements have been made in recent days,design of an effective and accurate action recognition model is yet a difficult process owing to the existence of different obstacles such as variations in camera angle,occlusion,background,movement speed,and so on.From the literature,it is observed that hard to deal with the temporal dimension in the action recognition process.Convolutional neural network(CNN)models could be used widely to solve this.With this motivation,this study designs a novel key point extraction with deep convolutional neural networks based pose estimation(KPE-DCNN)model for activity recognition.The KPE-DCNN technique initially converts the input video into a sequence of frames followed by a three stage process namely key point extraction,hyperparameter tuning,and pose estimation.In the keypoint extraction process an OpenPose model is designed to compute the accurate key-points in the human pose.Then,an optimal DCNN model is developed to classify the human activities label based on the extracted key points.For improving the training process of the DCNN technique,RMSProp optimizer is used to optimally adjust the hyperparameters such as learning rate,batch size,and epoch count.The experimental results tested using benchmark dataset like UCF sports dataset showed that KPE-DCNN technique is able to achieve good results compared with benchmark algorithms like CNN,DBN,SVM,STAL,T-CNN and so on.展开更多
文摘Macroalgae serve as a potential feedstock for fucoxanthin extraction.Fucoxanthin,a bioactive pigment found in the chloroplasts of marine algae,exhibits significant pharmacological properties.As a member of the carotenoid family,fucoxanthin plays a crucial role in both the food and pharmaceutical industries.This research explores the effects of ultrasonics on the extraction of fucoxanthin from the marine macroalga Padina australis.In addition,various extraction techniques and the influence of solvents on the efficient separation of fucoxanthin from algae have been studied and compared.Using methanol,chloroform,and a combination of methanol and chloroform(1:1,v/v),conventional fucoxanthin extraction from Padina australis yielded 8.12 mg of fucoxanthin per gram of biomass.However,the ultrasonic-assisted extraction resulted in a significantly higher yield of 16.9 mg of fucoxanthin per gram of biomass,demonstrating that the use of ultrasonics enhances the extraction rate compared to conventional methods.Therefore,the efficient separation of fucoxanthin from Padina australis is highly dependent on ultrasonic-assisted extraction.The process conditions for the extraction were optimized to maximize the yield of fucoxanthin from seaweeds.The following parameters were selected for optimization studies:moisture content,particle size,mixing speed,extraction temperature,extraction duration,and solid-to-solvent ratio.The extracted fucoxanthin exhibited various biological activities,including antimicrobial and antioxidant properties,and its structure was elucidated through FTIR and NMR spectroscopy.Additionally,thin-layer chromatography of the crude algae extracts confirmed the presence of fucoxanthin in the marine algae.Given these findings,the optimized extraction process holds the potential for scaling up to large-scale fucoxanthin production.Fucoxanthin,as a potent pharmacological agent,offers promising applications in the treatment of various ailments.
文摘This study aimed to compare the total phenolic content (TPC) and antioxidant activities of A. rigidula extracts. It also aimed to identify phenolic acids present in the extracts. The 2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and ferric thiocyanate lipid peroxidation antioxidant assays were performed. High performance liquid chromatography was used to identify phenolic acids. There was no solvent effect on TPC nor on scavenging activities, and inhibition of lipid peroxidation (p > 0.05) among solvent extracts. On the other hand, 1:1:3 water: acetone: methanol extract (10.22 mg GAE/g sample) had significantly higher reducing potential than 50% ethanol extract (EE) (9.259 mg GAE/g sample) (p < 0.05);but EE was not significantly different from 80% methanol extract (9.781 mg GAE/g sample) (p > 0.05). Phenolic fraction designated as fraction 4 had the highest antioxidant activity (p < 0.05) with 69.49% ABTS scavenging activity and FRAP reducing potential, 22.26 mg of GAE/g sample. 文摘[Objectives]To establish the chromatographic fingerprint of Gancao Qinlian Extracts(GQE)and reveal the possible material basis of the anti-inflammatory effect of GQE by the correlation analysis between the fingerprint chromatographic peaks of different components of GQE and its anti-inflammatory activity.[Methods]Ultra-performance liquid chromatography(UPLC)was used to detect the different ingredients of GQE to establish its chromatographic fingerprint and analyze the differences among the three medicine components;LPS stimulated RAW264.7 cells to construct an inflammatory cell model.The NO secretion of cells was detected by the Griess method.ELISA was used to detect the changes in TNF-αand IL-10 contents.RT-qPCR tested the mRNA expression levels of TNF-αand IL-10.Grey relational analysis was carried out by combining fingerprint chromatographic peak data and anti-inflammatory activity data.[Results]The GQE fingerprint was established,34 fingerprint characteristic peaks were calibrated,and 33 related chromatographic peaks were screened out.The corresponding chromatographic peaks in the three components were obtained,and the content of the components was calculated;the anti-inflammatory results showed that the content of NO,TNF-α,and the expression of TNF-αmRNA in the high and medium-dose groups of GQE were significantly lower than those in the blank group(P<0.01).The NO content and TNF-αmRNA expression in the high-dose group of GQE I was considerably lower than those in the blank group(P<0.01).The secretion of NO,TNF-α,and the expression of TNF-αmRNA in the high,medium,and low dose groups of GQE II were significantly lower than those in the blank group(P<0.01);the results of grey relational analysis showed that the correlation degree of the three components was GQE II>GQE>GQE I,and the characteristic fingerprint peaks 12,15,22,23,28,31,33 may be closely related to the anti-inflammatory effect.[Conclusions]The best component of the anti-inflammatory effect in GQE is water-soluble component,and its main components are flavonoids and alkaloids.These components can alleviate cellular inflammatory damage by inhibiting the excessive secretion of NO and reducing the expression of TNF-αmRNA.

文摘Flavonoids have attracted considerable attention due to their health benefits. This study aimed to investigate the flavonoid profiles and antioxidant activity of Paeonia lactiflora petal flavonoid extract(PPF). The UHPLC-ESI-Q-Exactive HF MS/MS method was established for characterization, and 21 predominant flavonoid compounds were tentatively identified in PPF. Among them, isoscutellarein-7-(6’-acetylallosyl-(1->2)-glucoside) and scutellarin methylester were discovered in PPF for the first time. Pretreatment with PPF significantly reduced H2O2-induced cell damage, ROS accumulation, and malondialdehyde content and increased the activity of SOD, CAT, and GSH-Px in buffalo rat liver 3A(BRL3A) cells. Moreover, the expression of nuclear Factor E2-related factor(Nrf2) was upregulated by PPF, whose expression trend was consistent with that of heme oxygenase-1(HO-1), glutamate-cysteine ligase catalytic subunit(GCLC), and NAD(P)H quinone oxidoreductase-1(NQO1). These findings suggested that herbaceous peony flavonoids can be used as a natural bioactive agent to prevent oxidative stress.

文摘[Objectives]Peptides was extracted from Haemadipsa hainana and its activity was studied.[Methods]Electric stimulation,water extraction and ultrasonic extraction were used to extract the peptides from H.hainana.Then the protein content and molecular weight distribution of H.hainana peptides were detected by the BCA method and SDS-PAGE method,respectively.The antithrombin activity and analgesic activity of the three peptide extracts of H.hainana were detected by Markwardt thrombin titration method and mouse hot plate experiment,respectively.[Results]There extraction methods of electric stimulation,water extraction and ultrasonic extraction were used to extract the peptide extract of H.hainana,and the yields were as follows:water extraction>electrical stimulation>ultrasonic extraction.The three peptide extracts from H.hainana had antithrombin activity,and the antithrombin activity was as follows:water extraction>ultrasonic extraction>electrical stimulation.Through the hot plate experiment in mice,it was verified that the three peptide extracts of H.hainana had analgesic activity,and the analgesic activity was water extraction>electric stimulation>ultrasonic extraction.The analgesic activity of high-dose(100 mg/kg)group of H.hainana obtained by water extraction was slightly weaker than that of tramadol.[Conclusions]This study confirmed that the peptide extract of H.hainana had certain antithrombin and analgesic activity,laying a foundation for the subsequent development and application of H.hainana.

文摘The aim of this study was to investigate the extraction process and antioxidant activity of polyphenols from Yunnan Mengzi pomegranate peel.Single factor and orthogonal test were used as optimize the extract rate(ER)of polyphenols from pomegranate peel(PGP).The polyphenols antioxidant activity was evaluated by DPPH·,·OH and total reducing ability,and compared with that of Vc.The optimum extraction conditions of polyphenols were as follows:ethanol concentration of 50%,liquid-solid ratio of 50:1,extraction temperature of 90°C,extraction for 4 h.Under these conditions,the ER of polyphenols from GPG was 8.15%.The results showed that the scavenging effects of polyphenols and Vc on DPPH·,·OH increased with increasing concentration,and the scavenging effect of polyphenols on DPPH·was higher than Vc in rang of 0.1-1.6µg/mL.When the concentration is greater than 0.6µg/mL,the scavenging effects of plyphenols on·OH is higher than Vc.The total reducing ability of polyphenols from PGP improved with the increase of concentration,and when the concentration was 8µg/mL,the total reducing ability was 114 U/mL.

文摘[Objective] The nematicidal activity of plant extracts was investigated. We aimed to explore a kind of botanical pesticide to control Hirschmanniella spp. [Method] The Chuanxiong (root), agrimony (whole plant), cocklebur (grain), basil (leaves), ragweed (stems and leaves), green onion (root), garlic (tuber), Yuzhu (leaves), marshpeper smartweed (whole plant), horseweed (aboveground stems and leaves) were dried, grinded and sieved. The powders were then extracted by ethanol at room temperature for 24 h. The extract solutions were filtered through fil- ter pater. The obtained plant extracts were then diluted to determine their nematici- dal activities. [Result] The Chuanxiong extract had the highest nematicidal activity, followed by cocklebur extract, agrimony extract, basil extract, garlic extract, ragweed extract. The horseweed extract had the lowest nematicidal activity. [Conclusion] The Chuanxiong and other plants were natural, and they had a good environmental compatibility. They could be developed as botanical pesticides against Hirschman- niella spp.

文摘Present mature plants hydroponic technology was used,combined with some excellent characteristics,such as growth conditions was easy to control and process of root growth was easy to continuously observe,the nematicidal activity of 5 kinds of Chinese herbs extracts and the compound solution of Avermectin,with strong contact toxicity effect indoor,was systematically studied and investigated the affection on the root-knot nematode parasitized on the cucumber seeding stage. It is found that under the premise of no influence on root growth of cucumber,extracts from Picrorhiza scrophulariiflora and Punica granatum showed strong prevention and nematicidal activity,and had the similar efficacy of Avermectin; while the extracts from Cibotium barometz,Aucklandia lappa Decne and Fructus cnidii showed low nematicidal activity and various degrees inhibition effect on plant growth.

文摘[ Objective] The paper was to study the contact toxicity and antifeedant activity of Aconitum flavum against cabbage worm. [ Method ] In- sect dipping method was adopted to determine the contact toxicity of the extracts of A. fiavum extracted from five polar solvents including ethanol, petroleum ether, ether, ethyl acetate, n-butanol and water; leaf dish method was adopted to determine the antifeedant activities of five solvent ex- tracts including ethanol, petroleum ether, ether, ethyl acetate, n-butanol and water against cabbage worm, [ Result] Extracts of A. flavum had high contact toxicity against cabbage worm. When the concentration was 100.00 mg/ml, the corrected mortality at 48 h roached 97.24%, and the insec- ticidal activities of five solvent extracts against cabbage worm in sequence were water 〉 n-butanol 〉 ethyl acetate 〉 ether 〉 petroleum ether, the cor- rected mortality of water extract at 48 h was 95.87% ; the antifeedant activities of five solvent extracts in sequence were water 〉 n-butanol 〉 ethyl ac- etate 〉 ether 〉 petroleum ether. [ Conclusion] Extracts of A. flavum had strong contact toxicity and antifeedant activity against cabbage worm, and the active ingredients with contact toxicity and antifeedant activity might be a kind of polar compound.
文摘[Objective] TO study the combined extracting process of porcine blood superoxide dismutase (SOD) and other bioactive substances and thus to provide technical basis for making full use of blood resources and large-scale production of SOD. [Method] Fibronectin, immunoglobulin, and hemoglobin were isolated from porcine blood, and SOD was extracted. Trace pyrogallol self-oxidation method to determine SOD activity was modified by optimizing the volume of pyrogallol and SOD samples, reaction temperature, and buffer pH. The specific activity of SOD was determined with the optimized extraction conditions. [ Result] The improved experimental conditions of SOD activity detection were as follows: 7 pyrogallol (50 mmol/L), 3 ml Tris-HCI (50 mmol/L, pH 8.2), reactive temperature at 25(3, and 10 pl SOD sample solution. The specific activity of extracted SOD was 5 056 U/mg protein. [ Conclusion] Four kinds of bioactive substance can be isolated from porcine blood by modern biological engi- neering integration technology, and the extracted SOD has better activity.

文摘[Objective] The antifungal activity of the extracts from,Atractylodes macracephal Koidz and Pulsatilla chinensis Bunge Regel,against Botrytis cinerea and Alternaria solani were studied under the condition of laboratory,in order to develop and utilize these two plants.[Method] The mycelium growth rate test was applied to measure the antifungal activities of extracts against fungi.[Result] the extracts of all the two plants showed strong antifungal activity against the target pathogenic fungi,especially the antifungal activity of the extract from Pulsatilla chinensis Bunge Regel was stronger and more stable.The inhibition rate to the mycelium growth of Botrytis cinerea was 80.25%.At the same concentration,the extract from Atractylodes macracephal Koidz showed little inhibition to Botrytis cinerea and Alternaria solani.The petrolelum ether extract of Atractylodes macracephal Koidz showed stronger antifungal activities and the EC50 was 5.31 mg/ml,and the n-butanol extract of Pulsatilla chinensis Bunge Regel showed stronger antifungal activities and the EC50 was 2.93 mg/ml.[Conclusion] The extracts from Pulsatilla chinensis Bunge Regel showed the stronger antifungal activity against Botrytis cinerea and Alternaria solani.

文摘[Objective] This study aimed to determine the optimal process conditions for extraction of anti-allergic active ingredient from citrus peels. [Method] Crude anti- allergic active extracts of citrus peels were extracted. Effects of extraction conditions including extraction temperature, ethanol concentration, extraction time and liquid-to-solid ratio on extraction were explored with single-factor experiment, and the extrac- tion process was optimized with orthogonal experiment. [Result] Based on the single- factor experiment and orthogonal experiment, the optimal extraction condition for crude anti-allergic active extracts of citrus peels were determined to be under extraction temperature of 60℃, ethanol concentration of 70%, extraction time of 60 min, liquid-to-solid ratio of 1:25 and twice extraction. [Concluslon] This process condition is of simple operation and high extraction efficiency, which is environment-friendly and suitable for the extraction of crude anti-allergic active extracts from citrus peels. This study provided a new approach and research ideas for the effective and rapid extraction of anti-allergic active ingredient in citrus peels.
文摘In order to get biological drugs with no resistance or toxic side effects and to reduce the use of antibiotics, a strain of Baci//us subtilis was isolated from animal intestine, and the isolate was identified by molecular biological method; in vitro an- tibacterial test of the isolate was performed using agar diffusion method; the optimal fermentation condition of the isoJate was screened by conventional culture method; the antibacterial crude protein of the isolate was extracted by saturated ammonium sulfate method; the physicochemical properties of antibacterial crude protein was de- tected by comparison method; The results showed that the isolate was B. subti/is, which had antibacterial effects on Staphy/ococcus aureus, streptococcus and swine erysipelas. The fermentation effect of the isolate was the best under the condition of temperature 30 ~C, pH 7, liquid volume 75 ml/250 ml, inoculation volume 20% and culture time 48 h. The antibacterial effect of the isolate was the best when extract- ed by 80% saturated ammonium sulfate. The antibacterial crude protein had strong resistance to heat and acid. Organic solvent and UV irradiation had some influences on antibacterial crude protein. Proteases had hydrolytic effects on antibacterial crude protein. The isolated B. subti/is can be used to prevent and control the diseases caused by S. aureus, streptococcus and swine erysipelas, and can regulate intesti- nal microecology by adding into expanded feeds.

文摘[Objective] The research aimed to study the toxicity and antifeedant activity of walnut green husk extract on wheat aphid. [Method] Using ultrasonic extraction, 5 kinds of different polarity solvents were used to prepare the extracts from walnut green husk. The toxicity and antifeedant activity on wheat aphid were determined, [ Result ] Walnut green husk extracts in the solvent of ethanol, acetone, ethyl acetate had higher toxicity and antifeedant activity on wheat aphid. When the concentration was 100 mg/ml, after 24 h, the toxicity and antifeedant activity of three kinds extracts on wheat aphid were greater than 84.35% and 66.98%, respectively. After 48 h, the toxicity and antifeedant activity were greater than 85.91% and 79.34%, respectively. Among 5 kinds extracts,ethanol extracts had the highest toxicity and antifeedant activity on wheat aphid. After 24 h, the corrected mortality rate was 93.64% and the antifeedant rate was 83.61%. After 48 h,the corrected mortality rate was 94.95% and antifeedant rate was 92.21%. Petroleum ether extract had the smallest toxicity and antifeedant activity on wheat aphid. After 24 h,the corrected mortality rate and antifeedant rate were 19.54% and 34.65%, respectively. After 48 h,the corrected mortality rate and antifeedant rate were 20.13% and 39. 03%, respectively. [ Conclsion] The extracts from walnut green husk with larger polarity solvent had better toxicity and higher antifeedant activity on wheat aphid.
文摘[Objective] The experiment studied on insecticidal and antifeedant action of on Portulaca oleracea L.to provide theoretical basis for developing a new botanical insecticide by taking Portulaca oleracea L.as a raw material.[Method] The insecticidal activities of the ethanol extracts and 5 kinds of extracts with petroleum ether,chloroform,ethyl acetate,n-butanol,water from Portulaca oleracea L..against Aphis sp.was studied;Leaves are using drug treatments of 5 extracts anti-feedant activity of Aphis sp.[Result] The results showed that 95% ethanol extract displayed high contact activity to Aphis sp.The corrected mortality after 48 h reached 95.4 % under the concentration of 50.00 mg·ml-1 and the insecticidal activities of 5 extracts against Aphis sp.was in the order that petroleum ether > chloroform > n-butanol > ethyl acetate >water.Five kinds of anti-feeding activity in the extract of the order for the water> ethyl acetate> n-butanol > chloroform >petroleum ether.[Conclusion] The extract from Portulaca oleracea L.has a strong contact toxicity of pesticide active on Aphis sp.substances and for a small polar compounds,the strong anti-feeding activity is a kind of larger polar compounds.

文摘Polysaccharides are the important biologically active components found in the peel of Dioscorea opposita Thunb.(DTTP).The influences of 4 extraction methods,namely hot water extraction(W),acidic extraction(HA),hot-compressed water extraction(HCW)and enzyme-assisted extraction(EAE),on the yields,physicochemical properties,hypoglycemic and antioxidant activities of polysaccharides from DTTP were studied and compared.Among these DTTP polysaccharides,DTTP-HA was outstanding in several respects.DTTP-HA was the most water soluble;it had the highest total carbohydrate content(85.08%),the highest uronic acid(13.20%)and the highest thermal stability.DTTP-HA and DTTP-W possessed a triple-helix structure.These 4 kinds of polysaccharides have the same types of monosaccharides,but in different molar percentages.Extraction method had a significant impact on the microstructures of the extracted polysaccharides.DTT-HA exhibited irregular structure with many holes.Among the 4 extracted methods,the DTTP-HA and DTTP-W initially exhibited higher hypoglycemic and antioxidant activities.The better bioactivities of DTTP-HA may be related to the above factors.The findings indicated that acid extraction is an effective method to extract polysaccharides with high biological activities from DTTP.
文摘Human Action Recognition(HAR)and pose estimation from videos have gained significant attention among research communities due to its applica-tion in several areas namely intelligent surveillance,human robot interaction,robot vision,etc.Though considerable improvements have been made in recent days,design of an effective and accurate action recognition model is yet a difficult process owing to the existence of different obstacles such as variations in camera angle,occlusion,background,movement speed,and so on.From the literature,it is observed that hard to deal with the temporal dimension in the action recognition process.Convolutional neural network(CNN)models could be used widely to solve this.With this motivation,this study designs a novel key point extraction with deep convolutional neural networks based pose estimation(KPE-DCNN)model for activity recognition.The KPE-DCNN technique initially converts the input video into a sequence of frames followed by a three stage process namely key point extraction,hyperparameter tuning,and pose estimation.In the keypoint extraction process an OpenPose model is designed to compute the accurate key-points in the human pose.Then,an optimal DCNN model is developed to classify the human activities label based on the extracted key points.For improving the training process of the DCNN technique,RMSProp optimizer is used to optimally adjust the hyperparameters such as learning rate,batch size,and epoch count.The experimental results tested using benchmark dataset like UCF sports dataset showed that KPE-DCNN technique is able to achieve good results compared with benchmark algorithms like CNN,DBN,SVM,STAL,T-CNN and so on.
