Mechanism and Clinical Observation of Effect of Growgx Yeast Fermentation Product Extract on Photo-aged HFF-1 Cells

[Objectives]To explore the effect of the extract of Growgx yeast fermentation product on ultraviolet A(UVA)-induced photo-aging of human skin fibroblasts(HFF-1)and its specific mechanisms from the level of cell biology,and discussed the application of the Growgx yeast fermentation product extract for skin aging.[Methods]A photo-aging model of HFF-1 cells was induced by UVA radiation meter,and it was given different concentrations of Growgx for intervention.The proliferation activity of the HFF-1 cells was detected with MTT method,and their migration and invasion ability was measured by scratch test.The SOD,HYP and MDA levels were detected with corresponding kits.After oxygen infusion apparatus assisted facial skin administration,the repair of the skin was observed through the VISIA skin tester.[Results]The survival rate of HFF-1 cells was reduced significantly by UVA with an irradiation dose of 10.8 J/cm2(P<0.05),while Growgx significantly increased their survival rate(P<0.001)and sped up the repair of damaged cells.Growgx promoted the production of SOD(P<0.05),reduced the level of MDA(P<0.05),and increased the level of HYP(P<0.05).Growgx effectively inhibited UVA-induced photo-aging of HFF-1 cells.The mechanism may be related to accelerating cell damage repair,up-regulating SOD and HYP expression levels,and reducing MDA expression level.The clinical observation showed that Growgx effectively improved skin spots and pores,making the skin smoother and more delicate.[Conclusions]Growgx can effectively fight against photo-aging caused by ultraviolet rays,and can significantly improve skin wrinkles. 展开更多

This study aims to determine the effect of adding feed supplements of fermented shrimp waste extract in the ration on the digestibility of local chicken rations in the growth phase.The research was carried out in Jatinangor District as well as the Laboratory of Ruminant Animal Nutrition and Animal Feed Chemistry,Faculty of Animal Husbandry,Padjadjaran University,Sumedang.The method used in this study was experimental with a Complete Randomized Design(RAL).The data were analyzed by fingerprint test(ANOVA)with further tests using the Dunnet test.The object of this study consisted of 20 Sentul chickens raised from the age of 1 day to 12 weeks,divided into 5 treatments and 4 tests.The treatment consists of P0=Basal ration without the addition of feed supplement fermented shrimp waste extract,P1=Basal ration+0.5%feed supplement fermented shrimp waste extract,P2=Basal ration+1.0%feed supplement fermented shrimp waste extract,P3=Basal ration+1.5%feed supplement fermented shrimp waste extract,and P4=Basal ration+2.0%feed supplement fermented shrimp waste extract.The changes observed are the digestibility of dry matter,the digestibility of organic matter,and the digestibility of proteins.The results showed that the addition of feed supplements for fermented shrimp waste extract had a significantly different influence on the digestibility of dry matter,the digestibility of organic matter,and the digestibility of protein.This study concludes that the addition of a feed supplement of 2%fermented shrimp waste extract in the ration can produce a high ration digestibility value for Sentul chicken in the growth phase. 展开更多

Objective A subcutaneous transplantation tumor model of human HT-29 cells in nude mice was established to evaluate anticarcinogenic activities, and the apoptosis-regulated mechanism effect of aqueous extract of fermented wheat germ with Lactobacillus plantarum dy-1 (LFWGE). Methods The HT-29 cells were transplanted via subcutaneous injection of 1×10^7cells into the right flank of each nude mouse. Then, nude mice were treated for 30 d with LFWGE (high-dose 2 g/kg/d; low-dose 1 g/kg/d) and for 7 d with 5-fluorouracil (5-FU, 25 mg/kg/d) by gavage and intraperitoneal injection, respectively. An inhibition of tumor growth was observed. Results Tumor volume and weights decreased significantly in both groups of nude mice treated with LFWGE. In addition, the cell apoptosis rate of the LFWGE group (2 g/kg/d, 60.2%+4.4%; 1 g/kg/d, 58.6%+6.9%) was significantly higher than that of the control group (11.5%+1.6%) and 5-FU group (32.1%+3.5%) as measured by the TUNEL assay. Moreover, the real-time fluorescent quantitative PCR and Western blot method further confirmed these enhancing apoptosis and growth inhibition effects. The involvement of LFWGE in inducing apoptosis was confirmed by the expression of Bax, Bcl-2, Caspase-3, and CyclinD1. Conclusion The results showed that LFWGE could induce subcutaneous transplantation tumor apoptosis in nude mice and could be as a natural nutrient supplements or chemopreventive agent in the treatment of human colon cancer. 展开更多

Objective To investigate the effect of fermented barley extracts with Lactobacillus plantarum dy-1(LFBE) for modulating glucose consumption in HepG2 cells via miR-212 regulation. Methods Hepatocellular carcinoma(HepG2) cells were treated with palmitate. After 12 h, palmitate-induced HepG2 cells were treated with LFBE and its main components. Changes in glucose consumption, proinflammatory cytokine secretion, and miRNA-212 expression in HepG2 cells was observed. Results Treatment with LFBE rich in vanillic acid(VA) increased glucose consumption and reduced proinflammatory cytokine secretion in HepG2 cells. LFBE and VA normalized the upregulation of miR-212, which led to the upregulation of dual-specificity phosphatase-9(DUSP9), a direct target of miR-212, at both protein and mR NA levels. Downregulation of miR-212 markedly increased glucose consumption and reduced proinflammatory cytokine secretion by enhancing DUSP9 expression. Conclusion The results showed the benefit of LFBE and miR-212 downregulation in modulating glucose consumption and reducing proinflammatory cytokine secretion by targeting DUSP9. VA in LFBE was a strong regulator of palmitate-induced abnormal glucose consumption in HepG2 cells and can be a primary mediator. 展开更多

Effect of Food Waste Compost (FWC) and its Non-Aerated Fermented Extract (NFCE) on Seeds Germination and Plant Growth 被引量:1

The aim of this study was to investigate the effect of the food wastes compost (FWC) and its non-aerated fermented extract (NFCE) on seed germination and growth of tomato (<i>Solanum</i> <i>lycopersicum</i> L.), watercress (<i>Nasturtium</i> <i>officinale</i>), chili pepper (<i>Capsicum</i> <i>annuum</i>), peas (<i>Pisum</i> <i>sativum</i> L.), chickpea (<i>Cicer</i> <i>arietinum</i>) and beans (<i>Vicia</i> <i>faba</i>) under greenhouse conditions. The FWC and NFCE were physico-chemically and microbiologically characterized. The NFCE effect was evaluated on tomato, watercress, and chili pepper seeds germination and seedling growth. However, for leguminous, pea, chickpea and bean seedlings, the FWC amended soils and irrigated with NFCE were tested for plants growth. The results of FWC analyses revealed that FWC has neutral pH, low EC and C/N ratio, with fertilizing elements (N, P, K and Mg) and lack of phytotoxic effect. The NFCE was characterized by low EC and relatively high carbon content (COD = 9700 mg/l), and intense microbial activity, notably mesophilic bacteria. Therefore, in fermented compost extract, mesophilic bacteria were increased by 225, yeasts by 25 and molds by 10 times compared to those of the investigated compost. In greenhouse, the diluted NFCE increased significantly (<i>p</i>< 0.05) germination and growth of the tested seedlings. Used alone, the FWC amended soil or the NFCE irrigated soil, improved the growth of tested seedlings. The use of soil amended with compost and irrigated by fermented compost extract decreased significantly the growth of the same experimented seedlings. Therefore, the FWC and its fermented extract were a suitable substrate for germination and growth of the studied seeds. 展开更多

Objective: To investigate the adjuvant therapeutic effects of fermented red ginseng(FRG) extract on non-small cell lung cancer(NSCLC) patients treated with chemotherapy. Methods: A total of 60 patients with advanced NSCLC were assigned to two groups using a random number table, i.e., the gemcitabine plus cisplatin(GP) chemotherapy alone group(26 patients) and the FRG + GP chemotherapy group(34 patients), for 60-day treatment. Patients were then assessed according to the Fatigue Symptom Inventory, Chinese medicine symptoms score, Self-Rating Anxiety Scale, Self-Rating Depression Scale, Karnofsky Performance Status Scale, and Functional Assessment of Cancer Therapy-Lung. In addition, chemotherapy toxicity and tumor biomarkers were measured. Results: For NSCLC patients after chemotherapy, FRG extract significantly improved the FSI score, CM symptoms score, psychological status, physical conditions, and quality of life and reduced chemotherapy toxicity(P〈0.01), but the expression levels of carcinoembryonic antigen, cytokeratin-19 fragments, and neuron-specific enolase were not significantly different between the chemotherapy alone and the FRG + chemotherapy groups or between pre-and post-treatments(P〉0.05). Conclusions: This study demonstrated that FRG extract had an adjuvant effect on advanced NSCLC patients treated with chemotherapy. Further studies with a larger sample size will verify the current findings. 展开更多
