Involvement of chromatin and histone acetylation in the regulation of HIV-LTR by thyroid hormone receptor 被引量:4

The HIV-1 LTR controls the expression of HIV-1 viral genes and thus is critical for viral propagation and pathology. Numerous host factors have been shown to participate in the regulation of the LTR promoter. Among them is the thyroid hormone (T3) receptor (TR). TR has been shown to bind to the critical region of the promoter that contain the NFbB and Sp1 binding sites. Interestingly, earlier transient transfection studies in tissue culture cells have yielded contradicting conclusions on the role of TR in LTR regulation, likely due to the use of different cell types and/or lack of proper chromatin organization. Here, using the frog oocyte as a model system that allows replication-coupled chromatin assembly, mimicking that in somatic cells, we demonstrate that unliganded heterodimers of TR and RXR (9-cis retinoic acid receptor) repress LTR while the addition of T3 relieves the repression and further activates the promoter. More importantly, we show that chromatin and unliganded TR/RXR synergize to repress the promoter in a histone deacetylase-dependent manner. 展开更多

Elemental Compositions and Chemical Mass Closure of Fine Particulate in an Animal Feeding Operation Facility and Its Vicinity

Particulate matter (PM) emissions from animal feeding operations (AFOs) have been considered as an important contributor to ambient PM in rural areas. Investigation of the chemical compositions of PM2.5 inside and in the vicinity of AFOs can enhance our understanding of the AFO emissions impact on ambient PM characteristics. This year-long field study was conducted on a commercial egg production farm to investigate ambient PM chemical compositions as impacted by the air emissions from the production houses. The PM2.5 samples were collected from five sampling stations (one in-house station and four ambient locations in four wind directions). The trace elements, major ions, organic carbon (OC) and element carbon (EC) were analyzed by X-ray florescence (XRF), ion chromatography (IC), and thermo-optical analyzer, respectively. There were significant differences in elemental compositions between PM samples from in-house station (ST1) and ambient stations (ST2-ST5). The chemical mass balance analysis revealed that OC accounted for above 50% of PM2.5 mass at in-house and ambient stations;NH4+, SO42-, and NO3- accounted for about 40.0% of the total PM2.5 mass in ambient locations and for only 12% of the total PM2.5 mass in house. The measured PM2.5 masses agreed with the sums of the masses of chemical compositions at all stations except for the in-house station. Knowledge gained from this study, with additional consideration of NH3 concentrations and emissions, will lead to better understanding of PM2.5 source and formation, fate and transport, and their atmospheric dynamics. 展开更多

The Negative Effect of Heavy Metal Salts on the Body of Mammal Animals

The main purpose of this presented article was to explain the need to study the amount of heavy metal salts in the environment where animals live, in the water, in air, and in the food and fodder consumed. This article presents materials from the literature on the effects of heavy metal salts on the body of animals and the environment in which they live. The cited analytical data showed that the general information on the negative effects of heavy metal salts on the body is sufficient, but their effects on the digestive tract and morpho-functional properties of rabbits should be studied in depth. Therefore, we planned to focus our scientific work on this topic. The article mainly refers to salts of heavy metals cadmium, lead, and mercury (Cd, Pb, Hg). It is noted in the literature that heavy metal salts have a negative effect on the body of animals. We focused mainly on data on the effects of heavy metals on farm animals, including rabbits. But it is clear that the authors referred to were referring to experimental animals. These negative effects are manifested in the form of disorders of digestive functions, disorders of neurovegetative processes, increasing incidence of cardiovascular disease, rapid heart failure, deterioration of calcium metabolism, as well as impaired haemoglobin metabolism. Disorders of protein metabolism manifest themselves in the form of cases of hyperproteinaemia and dysproteinaemia. The results of the evaluation of the organism of healthy animals in chemically and radioactively contaminated areas showed the accumulation of significant levels of chemical elements in their organism. We mainly looked at the effects of heavy metal salts on farm animals. The cited analytical data showed that the general information on the negative effects of heavy metal salts on the body is sufficient, but the effects on the activity of organ systems in the body (respiration, blood and blood circulation, digestion, reproduction, productivity and immunological systems) have not been comprehensively studied. 展开更多

为分析动物源营养液在空气凤梨上的应用效果,以‘卷叶霸王’空气凤梨为研究对象,以清水作对照,研究动物源营养液、磷酸二氢钾、尿素喷施后对空气凤梨新叶生长、单株增重、光泽度及矿质元素含量等的影响。结果表明,动物源营养液中含有钙... 为分析动物源营养液在空气凤梨上的应用效果,以‘卷叶霸王’空气凤梨为研究对象,以清水作对照,研究动物源营养液、磷酸二氢钾、尿素喷施后对空气凤梨新叶生长、单株增重、光泽度及矿质元素含量等的影响。结果表明,动物源营养液中含有钙、镁、铁、锌、硼和锰等矿质元素,空气凤梨在各种处理后3月内都没有新根萌出,均未出现肥害与微量元素中毒现象,光泽度没有明显差异。2.50‰营养液处理的第1小叶生长率增加41.58%,增加幅度最大。0.10‰动物源营养液处理单株增重率最高,2.50‰、0.50‰营养液处理次之;不同浓度动物源营养液处理间单株增重率差异不显著。与CK相比,3.33‰营养液处理、1.00‰营养液处理显著增加空气凤梨新叶内钙、镁、铁和锰含量,3.33‰动物源营养液处理的钙含量增加27.80%,镁含量增加23.71%,铁含量增加10.75%,锰含量增加104.41%。3.33‰营养液处理新叶内钙、镁、锰和硼含量显著高于1.00‰营养液处理,但铁和锌含量显著低于1.00‰营养液处理,生产上可以将动物源营养液作为卷叶霸王空气凤梨新型叶面营养肥进行合理开发与利用。 展开更多

背景:髌腱末端病中髌骨髌腱结合部是损伤的高发区域。兔髌骨髌腱损伤模型是常用动物实验研究对象,目前对损伤机制和造模方法的应力分析尚不全面。目的:在过往组织学和力学测量的基础上,对兔整个髌骨、髌骨髌腱结合区和股四髌腱止点结合... 背景:髌腱末端病中髌骨髌腱结合部是损伤的高发区域。兔髌骨髌腱损伤模型是常用动物实验研究对象,目前对损伤机制和造模方法的应力分析尚不全面。目的:在过往组织学和力学测量的基础上,对兔整个髌骨、髌骨髌腱结合区和股四髌腱止点结合区进行扫描建模,分析其应力变化特点,探讨不同运动形式对髌腱退行性变的潜在影响,为损伤预防以及动物模型建立提供理论依据。方法:以成年健康雌性新西兰大白兔左侧膝关节的髌骨和两端肌腱为实验对象,取材处理后经Micro-CT扫描,Mimics、Geomagic Studio、SolidWorks等软件建模,应用Ansys Workbench进行有限元分析。分析不同加载条件下(载荷大小和方向改变,不同动作模式)目标区域的等效位移、应力、应变情况。结果与结论:①载荷方向改变影响趋势和数值变化,载荷大小改变影响数值变化;②与其他区域相比,髌腱腱止点处应力集中较明显,应变值最低;③在髌腱腱止点处,外翻比内翻、内旋比外旋产生更大的应力集中;④提示膝关节屈伸运动中所产生的应力集中和应力屏蔽效应是导致髌腱腱止点损伤的主要原因;内旋和外翻状态的应力同时是导致损伤的潜在因素,有待进一步研究;以跳跃、跑动状态建立动物模型更加符合运动损伤特点。 展开更多
