Modified constraint-induced movement therapy is an effective treatment for neurological and motor impairments in patients with stroke by increasing the use of their affected limb and limiting the contralateral limb.Ho...Modified constraint-induced movement therapy is an effective treatment for neurological and motor impairments in patients with stroke by increasing the use of their affected limb and limiting the contralateral limb.However,the molecular mechanism underlying its efficacy remains unclear.In this study,a middle cerebral artery occlusion(MCAO)rat model was produced by the suture method.Rats received modified constraint-induced movement therapy 1 hour a day for 14 consecutive days,starting from the 7^th day after middle cerebral artery occlusion.Day 1 of treatment lasted for 10 minutes at 2r/min,day 2 for 20 minutes at 2 r/min,and from day 3 onward for 20 minutes at 4 r/min.CatWalk gait analysis,adhesive removal test,and Y-maze test were used to investigate motor function,sensory function as well as cognitive function in rodent animals from the 1st day before MCAO to the 21^st day after MCAO.On the 21^st day after MCAO,the neurotransmitter receptor-related genes from both contralateral and ipsilateral hippocampi were tested by micro-array and then verified by western blot assay.The glutamate related receptor was shown by transmission electron microscopy and the glutamate content was determined by high-performance liquid chromatography.The results of behavior tests showed that modified constraint-induced movement therapy promoted motor and sensory functional recovery in the middle cerebral artery-occluded rats,but had no effect on cognitive function.The modified constraint-induced movement therapy upregulated the expression of glutamate ionotropic receptor AMPA type subunit 3(Gria3)in the hippocampus and downregulated the expression of the beta3-adrenergic receptor gene Adrb3 and arginine vasopressin receptor 1 A,Avprla in the middle cerebral artery-occluded rats.In the ipsilateral hippocampus,only Adra2 a was downregulated,and there was no significant change in Gria3.Transmission electron microscopy revealed a denser distribution the more distribution of postsynaptic glutamate receptor 2/3,which is an a-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid receptor,within 240 nm of the postsynaptic density in the contralateral cornu ammonis 3 region.The size and distribution of the synaptic vesicles within 100 nm of the presynaptic active zone were unchanged.Western blot analysis showed that modified constraint-induced movement therapy also increased the expression of glutamate receptor 2/3 and brain-derived neurotrophic factor in the hippocampus of rats with middle cerebral artery occlusion,but had no effect on Synapsin I levels.Besides,we also found modified constraint-induced movement therapy effectively reduced glutamate content in the contralateral hippocampus.This study demonstrated that modified constraint-induced movement therapy is an effective rehabilitation therapy in middle cerebral artery-occluded rats,and suggests that these positive effects occur via the upregulation of the postsynaptic membrane a-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid receptor expression.This study was approved by the Institutional Animal Care and Use Committee of Fudan University,China(approval No.201802173 S)on March 3,2018.展开更多

In this work, a detailed pollen analysis of it is carried out to provide a preliminary insight into the paleo-climate and paleo-altimetry change in the central Tibetan Plateau. It can be concluded that the pollen assemblage can be obviously divided into two pollen zones, Pollen zone I (251.1 - 314 m in depth, 120.0 - 345.8 ka BP.) and Pollen zone II (200 - 251.1 m in depth, 105.4 - 120 ka BP.). The paleo-climate during pollen zone I deposition period was comparatively colder and wetter than it during the pollen zone II deposition period. After Gonghe Movement, the center of Tibetan Plateau was uplifted about 300 m (from 3500 - 3700 m to 3800 - 4000 m in elevation). The wind was changed from horizontal or downward direction to upward direction, in the study area. In the central of Tibetan Plateau, the climate change seems to be mainly driven by global climate change, and that tectonic uplift may have been a subordinate influence at the middle-late Pleistocene.展开更多

Objective:To investigate effect and possible mechanisms of silencing human WFDC2(HE4) gene on biological behavior changes as cell proliferation,apoplosis,movement and invasion of human serous ovarian cancer cell lin...Objective:To investigate effect and possible mechanisms of silencing human WFDC2(HE4) gene on biological behavior changes as cell proliferation,apoplosis,movement and invasion of human serous ovarian cancer cell line SKOV3.Methods:Lentiviral WFDC2 gene sequence of small interfering siRNA was stablely transfected into SKOV3 identified by Q-PCR and western-blot. Obtained SKOV3 stable strains with silenced HE4 were measured by proliferation,apoplosis, migration,and invasion.Results:Gene sequencing showed that the oligonucleotides were successfully inserted into the expected site.After silencing HE4 in the SKOV3,proliferation was significandy inhibited(P【0.05).G<sub>0</sub>/G<sub>1</sub> phase was arrested by the cell cycle(P【0.01) and capacity of the migration and invasion decreased significandy(P【0.01).Slight early apoptosis ratio and no change of late apoplosis were found without change of Caspase-3 or Bcl-2 protein.Proteins involed in ERK pathway as phosphorylated protein as p-EGFR,p- ERK decreased and protease protein involved in tissue remoding as matrix metalloproteinases MMP-9,MMP-2 and cathepsin B decreased compared with control group.Conclusions:HE4 gene plays an important role in regulating proliferation,apoptosis,migration,invasion of serous ovarian cancer cells by ERK pathway and protease system.Its role in apoptosis needs to be further explored,and it may be a potential target for serous ovarian cancer.展开更多
The effects of two different nitrogen fertilizers (urea and NH4C1) with monocalcium phosphate (MCP) on the movement and transformation of fertilizer P in soil microsites along with soil pH changes at different dis...The effects of two different nitrogen fertilizers (urea and NH4C1) with monocalcium phosphate (MCP) on the movement and transformation of fertilizer P in soil microsites along with soil pH changes at different distances from the fertilizer application site were studied in an incubation experiment. A highly acidic red soil (Ultisol, pH 4.57) from south China with MCP fertilizer alone or in combination with NH4C1 or urea was added to the surface of soil cylinders and packed in wax blocks. After 7 and 28 days, the extraction and analysis of each 2 mm layer from the interface of the soil and fertilizer showed that added NH4C1 or urea did not change the movement distance of fertilizer P. However, P transformation was significantly affected (P 〈 0.05). After 7 days, at 0-8 mm distance from the fertilizer site the addition of urea significantly decreased the water-extractable P concentration; however, after 28 days the effect of N addition had disappeared. Also,at limited distances close to the fertilizer site NH4Cl application with MCP significantly increased acid-extractable P and available P, while with the addition of urea they significantly decreased. Compared with application of MCP alone,addition of urea significantly increased soil pH in fertilizer microsites, whereas the addition of NH4Cl significantly decreased soil pH.展开更多

However, most of them could only realize simple response to constant conditions(e.g. a stationary magnetic field) while carrying out humdrum motions. By controlling distribution of metal organic framework obtained carbon-enriched Fe304 nanoparticles in self-assembly reduced graphene oxide(RGO) monoliths, we could achieve two distinctive RGO-Fe_3 O_4 stirrers that could dynamically respond to the rapidly changing magnetic field while executing designed movements precisely: rotating with lying down posture or standing straight posture. These stirrers can not only be applied in environmental remediation(e.g.suction skimmer), but also be recycled as electrode active materials for supercapacitors after fulfilling their destiny, realizing transformation of trash to treasure, which will inspire other dynamically responsive monoliths for various applications.展开更多

METHODS: Male Wistar rats received three weekly injections of CCl4 for 11 wk, and were analyzed before and during the induction of cirrhosis. Rats were im- planted with electrodes to record their sleep patterns. Polygraph recordings were made weekly over 11 wk for 8 h, during the light period. After a basal recording, rats received three weekly injections of CCl4. Histological confirmation of cirrhosis was performed after 11 wk. RESULTS: The results showed a progressive decrease in total wake time that reached statistical significance from the second week of treatment. In addition, there was an increase in total time of slow wave sleep (SWS)Ⅱ and rapid eye movement sleep (REM sleep) in most of the 11 wk. SWS I showed no significant variations. During the final weeks, a significant increase in REM sleep frequency was also observed. Histological analyses of the livers showed unequivocal signs of cirrhosis. CONCLUSION: These data suggest that hepatic failure produced by CCh administration is capable of modifying the sleep pattern even after only a few doses.展开更多

A literature analysis has shown that object search,recognition,and tracking systems are becoming increasingly popular.However,such systems do not achieve high practical results in analyzing small moving living objects...A literature analysis has shown that object search,recognition,and tracking systems are becoming increasingly popular.However,such systems do not achieve high practical results in analyzing small moving living objects ranging from 8 to 14 mm.This article examines methods and tools for recognizing and tracking the class of small moving objects,such as ants.To fulfill those aims,a customized You Only Look Once Ants Recognition(YOLO_AR)Convolutional Neural Network(CNN)has been trained to recognize Messor Structor ants in the laboratory using the LabelImg object marker tool.The proposed model is an extension of the You Only Look Once v4(Yolov4)512×512 model with an additional Self Regularized Non–Monotonic(Mish)activation function.Additionally,the scalable solution for continuous object recognizing and tracking was implemented.This solution is based on the OpenDatacam system,with extended Object Tracking modules that allow for tracking and counting objects that have crossed the custom boundary line.During the study,the methods of the alignment algorithm for finding the trajectory of moving objects were modified.I discovered that the Hungarian algorithm showed better results in tracking small objects than the K–D dimensional tree(k-d tree)matching algorithm used in OpenDataCam.Remarkably,such an algorithm showed better results with the implemented YOLO_AR model due to the lack of False Positives(FP).Therefore,I provided a new tracker module with a Hungarian matching algorithm verified on the Multiple Object Tracking(MOT)benchmark.Furthermore,additional customization parameters for object recognition and tracking results parsing and filtering were added,like boundary angle threshold(BAT)and past frames trajectory prediction(PFTP).Experimental tests confirmed the results of the study on a mobile device.During the experiment,parameters such as the quality of recognition and tracking of moving objects,the PFTP and BAT,and the configuration parameters of the neural network and boundary line model were analyzed.The results showed an increased tracking accuracy with the proposed methods by 50%.The study results confirmed the relevance of the topic and the effectiveness of the implemented methods and tools.展开更多

文摘A literature analysis has shown that object search,recognition,and tracking systems are becoming increasingly popular.However,such systems do not achieve high practical results in analyzing small moving living objects ranging from 8 to 14 mm.This article examines methods and tools for recognizing and tracking the class of small moving objects,such as ants.To fulfill those aims,a customized You Only Look Once Ants Recognition(YOLO_AR)Convolutional Neural Network(CNN)has been trained to recognize Messor Structor ants in the laboratory using the LabelImg object marker tool.The proposed model is an extension of the You Only Look Once v4(Yolov4)512×512 model with an additional Self Regularized Non–Monotonic(Mish)activation function.Additionally,the scalable solution for continuous object recognizing and tracking was implemented.This solution is based on the OpenDatacam system,with extended Object Tracking modules that allow for tracking and counting objects that have crossed the custom boundary line.During the study,the methods of the alignment algorithm for finding the trajectory of moving objects were modified.I discovered that the Hungarian algorithm showed better results in tracking small objects than the K–D dimensional tree(k-d tree)matching algorithm used in OpenDataCam.Remarkably,such an algorithm showed better results with the implemented YOLO_AR model due to the lack of False Positives(FP).Therefore,I provided a new tracker module with a Hungarian matching algorithm verified on the Multiple Object Tracking(MOT)benchmark.Furthermore,additional customization parameters for object recognition and tracking results parsing and filtering were added,like boundary angle threshold(BAT)and past frames trajectory prediction(PFTP).Experimental tests confirmed the results of the study on a mobile device.During the experiment,parameters such as the quality of recognition and tracking of moving objects,the PFTP and BAT,and the configuration parameters of the neural network and boundary line model were analyzed.The results showed an increased tracking accuracy with the proposed methods by 50%.The study results confirmed the relevance of the topic and the effectiveness of the implemented methods and tools.
