Semiconductor QDs have emerged as a novel class of fluorophore with unique photoluminescence properties, in particular, CdSe/ZnS core-shell QDs have been successfully used as biocompatible fluorescence resonance energ...Semiconductor QDs have emerged as a novel class of fluorophore with unique photoluminescence properties, in particular, CdSe/ZnS core-shell QDs have been successfully used as biocompatible fluorescence resonance energy transfer donors. Here we report FRET between CdSe/ZnS core-shell QDs (donor) and organic dye fluorescein 27 (F27) (acceptor). The results demonstrate the occurrence of efficient energy transfer in the system and the FRET efficiency is not only influenced by the spectral overlap between the QD donor emission and acceptor absorption, it might depend on QDs surface effect also. Efforts are made to correlate quantitatively spectral dependence of FRET rate with acceptor absorption spectrum, Forster distance, transfer efficiency (E) obtained employing steady-state & time-resolved technique.展开更多
Enzymes are the major players for many biological processes.Fundamental studies of the enzymatic activity at the single-molecule level provides important information that is otherwise inaccessible at the ensemble leve...Enzymes are the major players for many biological processes.Fundamental studies of the enzymatic activity at the single-molecule level provides important information that is otherwise inaccessible at the ensemble level.Yet,these single-molecule experiments are technically di±cult and generally require complicated experimental design.Here,we develop a Holliday junction(HJ)-based platform to study the activity of restriction endonucleases at the single-molecule level using single-molecule FRET(sm-FRET).We show that the intrinsic dynamics of HJ can be used as the reporter for both the enzyme-binding and the substrate-release events.Thanks to the multiple-arms structure of HJ,the fluorophore-labeled arms can be different from the surface anchoring arm and the substrate arm.Therefore,it is possible to independently change the substrate arm to study different enzymes with similar functions.Such a design is extremely useful for the systematic study of enzymes from the same family or enzymes bearing different pathologic mutations.Moreover,this method can be easily extended to study other types of DNA-binding enzymes without too much modi fication of the design.We anticipate it can find broad applications in single-molecule enzymology.展开更多
In the course of pushing forward the overall growth capacities of the mining industry and getting adapted to the tense competition after China抯 entry into WTO, it is quite necessary to speed up the process of fosteri...In the course of pushing forward the overall growth capacities of the mining industry and getting adapted to the tense competition after China抯 entry into WTO, it is quite necessary to speed up the process of fostering large state-owned coal group enter-prises. This strategical proposal is made on the basis of a comprehensive analysis of the main problems challenges faced by the coal mining industry is made on. The main prob-lems with Chinese coal mining industries discussed include: over-competition with a low convergence, severe waste and loss of mine resources with poor exploitation techniques, disordered operation with few industrial entry barriers, heavy welfare burdens from low socialization. The accession of WTO provide Chinese coal mining industry with chances to push forward the reforms and structural shift in the enterprises, expedite the estab-lishment and perfect of coal mining market mechanism, expand their international market share as well as introduce foreign investment, and with challenges from high technology, knowledge economy, and information revolution. The solutions to the foster of large-scale coal mining group enterprises, as well as the possible points to note and the policy pro-posal are advanced.展开更多

In this novel, Forster explores the difficulty to unify human relations,and exposes the eternal gaps between two nations. This paper will center on the eternal gaps of a Passage to India, which are represented by the misunderstandings between"India"and"British". By exploring and analyzing those gaps, the paper reaches the conclusion that those gaps can not be eliminated and the long-existing communication proposed by Forster is still a long way to go.展开更多
