Doubled haploid(DH)technology is an efficient method used in commercial maize breeding.Chromosome doubling is a vital step of DH technology;however,the underlying processes regulating chromosome doubling of haploid is...Doubled haploid(DH)technology is an efficient method used in commercial maize breeding.Chromosome doubling is a vital step of DH technology;however,the underlying processes regulating chromosome doubling of haploid is still not well understood,which is key to optimize the technology.In this study,the immature haploid embryos of the maize inbred line Zheng58 treated with amiprophos-methyl(APM)or colchicine were used to analyze transcriptomic and metabolomic changes,75 and 60 differential expressed metabolites(DEMs)were identified between control treatment,respectively.Most differentially expressed genes(DEGs)related to artificial chromosome doubling were down regulated;these were mainly involved in mitosis process.Both DEMs and DEGs co-expression analyses showed that,compared to controls,zeatin biosynthesis and cofactor and vitamin metabolism were significantly enriched in both APM and colchicine treatments.In a parallel experiment,exogenous vitamins including thiamine,nicotinic acid,vitamin B6,or trans-zeatin were added to colchicine treatment;there were synergistic effects between vitamins or zeatin and colchicine in haploid artificial chromosome doubling.These results provide novel insights in exploring the molecular responses to antimitotic reagents at both the transcriptomic and metabolomic levels.In addition,the application efficiency of haploid breeding will be greatly improved by the key factors for artificial chromosome doubling.展开更多

D. Chu seedlings under CO 2 doubling was studied by measuring soil electric conductance to survey soil water profiles and comparing it with root distribution surveyed by soil coring and root harvesting in Changbai Mountain in 1999. The results were: (1) The profiles of soil water content were adjusted by root activity. The water content of the soil layer with abundant roots was higher. (2) When CO 2 concentration was doubled, water transport was more active at the root/soil interface and the roots were distributed into deeper layer. It was shown in this work that the method of measuring electric conductance is an inexpensive, non_destructive and relatively sensitive way for underground water transport process.展开更多
Potential changes in precipitation extremes in July–August over China in response to CO 2 doubling are analyzed based on the output of 24 coupled climate models from the Twentieth-Century Climate in Coupled Models (...Potential changes in precipitation extremes in July–August over China in response to CO 2 doubling are analyzed based on the output of 24 coupled climate models from the Twentieth-Century Climate in Coupled Models (20C3M) experiment and the 1% per year CO 2 increase experiment (to doubling) (1pctto2x) of phase 3 of the Coupled Model Inter-comparison Project (CMIP3). Evaluation of the models’ performance in simulating the mean state shows that the majority of models fairly reproduce the broad spatial pattern of observed precipitation. However, all the models underestimate extreme precipitation by ~50%. The spread among the models over the Tibetan Plateau is ~2–3 times larger than that over the other areas. Models with higher resolution generally perform better than those with lower resolutions in terms of spatial pattern and precipitation amount. Under the 1pctto2x scenario, the ratio between the absolute value of MME extreme precipitation change and model spread is larger than that of total precipitation, indicating a relatively robust change of extremes. The change of extreme precipitation is more homogeneous than the total precipitation. Analysis on the output of Geophysical Fluid Dynamics Laboratory coupled climate model version 2.1 (GFDL-CM2.1) indicates that the spatially consistent increase of surface temperature and water vapor content contribute to the large increase of extreme precipitation over contiguous China, which follows the Clausius–Clapeyron relationship. Whereas, the meridionally tri-polar pattern of mean precipitation change over eastern China is dominated by the change of water vapor convergence, which is determined by the response of monsoon circulation to global warming.展开更多

The study focuses on the potential changes of July–August temperature extremes over China. The pattern correlation coefficients of the simulated temperature with the observations are 0.6–0.9, which are higher than the results for precipitation. However, most models have cold bias compared to observation, with a larger cold bias over western China (5°C) than over eastern China (2°C). The multi-model ensemble (MME) exhibits a significant increase of temperature under the 1pctto2x scenario. The amplitude of the MME warming shows a northwest–southeast decreasing gradient. The warming spread among the models (~1°C– 2°C) is less than MME warming (~2°C–4°C), indicating a relatively robust temperature change under CO 2 doubling. Further analysis of Geophysical Fluid Dynamics Laboratory coupled climate model version 2.1 (GFDL-CM2.1) simulations suggests that the warming pattern may be related to heat transport by summer monsoons. The contrast of cloud effects also has contributions. The different vertical structures of warming over northwestern China and southeastern China may be attributed to the different natures of vertical circulations. The deep, moist convection over southeastern China is an effective mechanism for "transporting" the warming upward, leading to more upper-level warming. The purpose of this study was to determine the relative time during induction culture when chromosome doubling may occur in wheat, and to identify early indicators for doubled haploid microspores. Microspore nuclei were stained with 4’,6-diamidino-2-phenylindole (DAPI) and observed under a fluorescent microscope on the day of isolation, three days after isolation, and six days after isolation. The change in the percentage of microspores containing a single small nucleus, two small nuclei, a single enlarged nucleus, and three or more nuclei was then tracked throughout the six-day period. Ploidy levels were estimated by determining the cross-sectional area and number of nucleoli in microspores containing small and large nuclei then comparing the results of each respective cell-type. The percentage of microspores containing enlarged nuclei increased throughout the six-day test period, and the percentage of binucleated microspores containing small nuclei decreased. Comparison of the changes in average percentage of microspores containing a single small nucleus, binucleated microspores, microspores containing a single large nucleus, and multinucleate microspores on days 0, 3, and 6 indicates that nuclei classified as “small” are likely haploids and nuclei classified as “large” are doubled haploids. In order to obtain doubled haploid, two chromosome doubling methods were used. Results showed that: After adding colchicine solution directly into a medium for young embryos that had been cultured 7 days, frequencies of embryo germination in colchicine concentrations of 50mg/L, 100mg/L and 200mg/L were 32.1% , 26.4% and 16.3% , respectively, and frequencies of chromosome doubling were 85.3% , 100% and 50.0% , respectively. But in the control without colchicine, the frequency of embryo germination was 67.4% and no seed was setting. As the time of colchicine treatment increased from 24 to 72 hours, the frequency of embryo germination was reduced, and 24 hours had better results. After soaking seeding crowns and roots with colchicine solution of 500mg/L, 750mg/L and 1 000mg/L for 5 hours, the frequencies of doubling were 89.6%, 76.0% and 73.3%, respectively. By soaking crowns and roots of strong seedings with 500mg/L colchicine solution, the frequency and efficiency of doubling were 98.2% and 93.2% , respectively.展开更多

Nine models show t...Based on simulations of the IPCC 20C3M and SRES A1B experiments in ten coupled models, the Asian summer mon-soon (ASM) response to CO2 doubling and the different responses among models are examined. Nine models show the similar results that the weakening of land-ocean thermal contrast caused by the CO2 doubling contributes to a weaker large-scale ASM circulation. Further analysis in this study also shows that the major ASM components,such as the Somali cross-equatorial flow,the low level India-South China Sea monsoon trough,and the upper level tropical easterly jet stream, weaken as CO2 doubles.However,the ASM rainfall increases as a result of the increased moisture from the warmer Indian Ocean and the South China Sea, and the enhanced northward moisture transport over the ASM region.For the response of enhanced northward moisture transport over South Asia, the positive contribution of moisture content increase in the Indian Ocean is dominant and the negative contribution of the weaker mon-soon circulation is secondary at 850 hPa,but both have positive contribution to the total moisture transport along the East China coast.The paradox of the weaker ASM circulation and the increasing precipitation in CO2 doubling is confirmed.It is found that strengthening of northward moisture transport could intensify the precipitation and atmospheric heat source over the north Arabian Sea and East China,and result in enhanced southwesterly at 850 hPa as global warming occurs.All ten models show significant enhanced southwesterly response over the north Arabian Sea,and six of them show enhanced southwesterly response along the East China coast.展开更多

AIM To evaluate the efficacy of doubling time(DT) of gastrointestinal submucosal tumors(GIST).METHODS From April 1987 through November 2012, a total of 323 patients were given a final histopathological diagnosis of GI...AIM To evaluate the efficacy of doubling time(DT) of gastrointestinal submucosal tumors(GIST).METHODS From April 1987 through November 2012, a total of 323 patients were given a final histopathological diagnosis of GISTs on surgical resection or endoscopic ultrasound-guided fine-needle aspiration(EUS-FNA) in Kitasato University East Hospital or Kitasato University Hospital. We studied 53 of these patients(34 with resected tumors and 19 with unresected tumors) whose tumors could be measured on EUS on at least two successive occasions. The histopathological diagnosis was GIST in 34 patients, leiomyoma in 5, schwannoma in 3, ectopic pancreas in 1, hamartoma in 1, cyst in 1, Brunner's adenoma in 1, and spindle-cell tumor in 7. We retrospectively calculated the DT of GISTs on the basis of the time course of EUS findings to estimate the growth rate of such tumors.RESULTS The DT was 17.2 mo for GIST, as compared with 231.2 mo for leiomyoma, 104.7 mo for schwannoma, 274.9mo for ectopic pancreas, 61.2 mo for hamartoma, 49.0 mo for cyst, and 134.7 mo for Brunner's adenoma. The GISTs were divided into risk classes on the basis of tumor diameters and mitotic figures(Fletcher's classification). The classification was extremely low risk or low risk in 28 patients, intermediate risk in 3, and high risk in 3. DT of GIST according to risk was 24.0 mo for extremely low-risk plus low-risk GIST, 17.1 mo for intermediate-risk GIST, and 3.9 mo for high-risk GIST. DT of GIST was significantly shorter than that of leiomyoma plus schwannoma(P < 0.05), and DT of high-risk GIST was significantly shorter than that of extremely low-risk plus low-risk GIST(P < 0.05).CONCLUSION For GIST, a higher risk grade was associated with a significantly shorter DT. Small SMTs should initially be followed up within 6 mo after detection.展开更多

This phenomenon can be attributed to the narrowing of the 1.56 μm laser linewidth and explained by our presented theoretical model. After soaking seeding crowns and roots with colchicine solution of 500mg/L, 750mg/L and 1 000mg/L for 5 hours, the frequencies of doubling were 89.6%, 76.0% and 73.3%, respectively. By soaking crowns and roots of strong seedings with 500mg/L colchicine solution, the frequency and efficiency of doubling were 98.2% and 93.2% , respectively.

文摘Based on simulations of the IPCC 20C3M and SRES A1B experiments in ten coupled models, the Asian summer mon-soon (ASM) response to CO2 doubling and the different responses among models are examined. Nine models show the similar results that the weakening of land-ocean thermal contrast caused by the CO2 doubling contributes to a weaker large-scale ASM circulation. Further analysis in this study also shows that the major ASM components,such as the Somali cross-equatorial flow,the low level India-South China Sea monsoon trough,and the upper level tropical easterly jet stream, weaken as CO2 doubles.However,the ASM rainfall increases as a result of the increased moisture from the warmer Indian Ocean and the South China Sea, and the enhanced northward moisture transport over the ASM region.For the response of enhanced northward moisture transport over South Asia, the positive contribution of moisture content increase in the Indian Ocean is dominant and the negative contribution of the weaker mon-soon circulation is secondary at 850 hPa,but both have positive contribution to the total moisture transport along the East China coast.The paradox of the weaker ASM circulation and the increasing precipitation in CO2 doubling is confirmed.It is found that strengthening of northward moisture transport could intensify the precipitation and atmospheric heat source over the north Arabian Sea and East China,and result in enhanced southwesterly at 850 hPa as global warming occurs.All ten models show significant enhanced southwesterly response over the north Arabian Sea,and six of them show enhanced southwesterly response along the East China coast.

文摘AIM To evaluate the efficacy of doubling time(DT) of gastrointestinal submucosal tumors(GIST).METHODS From April 1987 through November 2012, a total of 323 patients were given a final histopathological diagnosis of GISTs on surgical resection or endoscopic ultrasound-guided fine-needle aspiration(EUS-FNA) in Kitasato University East Hospital or Kitasato University Hospital. We studied 53 of these patients(34 with resected tumors and 19 with unresected tumors) whose tumors could be measured on EUS on at least two successive occasions. The histopathological diagnosis was GIST in 34 patients, leiomyoma in 5, schwannoma in 3, ectopic pancreas in 1, hamartoma in 1, cyst in 1, Brunner's adenoma in 1, and spindle-cell tumor in 7. We retrospectively calculated the DT of GISTs on the basis of the time course of EUS findings to estimate the growth rate of such tumors.RESULTS The DT was 17.2 mo for GIST, as compared with 231.2 mo for leiomyoma, 104.7 mo for schwannoma, 274.9mo for ectopic pancreas, 61.2 mo for hamartoma, 49.0 mo for cyst, and 134.7 mo for Brunner's adenoma. The GISTs were divided into risk classes on the basis of tumor diameters and mitotic figures(Fletcher's classification). The classification was extremely low risk or low risk in 28 patients, intermediate risk in 3, and high risk in 3. DT of GIST according to risk was 24.0 mo for extremely low-risk plus low-risk GIST, 17.1 mo for intermediate-risk GIST, and 3.9 mo for high-risk GIST. DT of GIST was significantly shorter than that of leiomyoma plus schwannoma(P < 0.05), and DT of high-risk GIST was significantly shorter than that of extremely low-risk plus low-risk GIST(P < 0.05).CONCLUSION For GIST, a higher risk grade was associated with a significantly shorter DT. Small SMTs should initially be followed up within 6 mo after detection.
文摘It is revealed in frictional experiments on medium-scale samples that period doubling bifurcation of stress drop for stick-slip occurs due to macroscopic heterogeneity of the sliding surface under conditions for typical stick-slip.The observed data show that the period doubling bifurcation of stress drop results from the alternate occurrence of strain release along the whole fault and along part of fault.This implies that complicated nonlinear behavior corresponds to clear physical implication in some cases.

文摘We study the doubling property of binomial measures on generalized ternary Cantor subsets of [0, 1]. We find some new phenomena. There are three different cases. In the first case, we obtain an equivalent condition for the measure to be doubling. In the other cases, we show that the condition is not necessary. Then facts and partial results are discussed.

文摘The factors influencing pollen chromosome doubling in Zenjimaru persimmon were studied, based on the knowledge of meiosis process of pollen mother cells. The results showed: (1) the suitable concentration of colchicine was 0.3-0. 5%; (2) the best doubling effect could be obtained during the period from the diplotene stage to the diakinasis stage, and the highest proportion was up to 40.6%; (3) considering giant pollen ratio and proportion of staminate-flower collection, the better treating frequency was 3 times; (4) giant pollen, which formed mainly from dyad and had bigger cell nucleus volume, was unreduced and hexaploid.
文摘We report a case of metastatic castration-resistant prostate cancer, who received prior treatment with docetaxel and was then given cabazitaxel as salvage therapy. The patient was monitored by prostate-specific antigen doubling time and prostate-specific antigen absolute value. The prostate-specific antigen doubling time was found to be a good response predictor in the patient.

文摘By using an external-cavity frequency-doubling master oscillator fiber power amplifier (MOPA), a 700 mW continuous-wave single-frequency laser source at 780 nm is produced. It is shown that the frequency doubling efficiency is improved when the seed diode laser is optically locked to a resonant frequency of a confocal Fabry-Perot (F-P) cavity. This phenomenon can be attributed to the narrowing of the 1.56 μm laser linewidth and explained by our presented theoretical model. The experimental results are found to be in good agreement with the theoretical predictions.
