A method to evaluate the bioactive function of fruit extracts of Chinese wild Citrus with microtubular activity

Due to the high content of secondary metabolites, mining wild Chinese Citrus for novel medical applications is promising. In this study, extracts of Chinese wild species from different taxonomical groups were screened for potential effects on microtubules (MTs) in vitro. MT density as a readout for nucleation, and frequency distribution over MT lengths as a readout for elongation and decay were determined by quantitative image analysis via a standardized coverslJp assay using fluorescently labelled neurotubulin. Extract from peels of Citrus ichangensis Swing. strongly increased the density of MTs; whereas, extract from peels of Cfurs limon (L.) Burm.f. exerted the opposite effect. Extract from pulp of Citrus limonia Osbeck promoted MT elongation, and in addition induced a small population of very long MTs. These data suggest that wild Chinese Citrus harbour compounds that act specifically on different aspects of MT nucleation, elongation, and decay. 展开更多

To evaluate the potential of both jamun (Syzygium cumini) seed and fruit extracts against hyperglycemia.MethodsMale Sprague Dawley rats were used to evaluate hypoglycemic potential of jamun extracts. Purposely, jamun fruit and seed’s ethanolic extracts based diets were provided to normal and high sucrose diet induced hyperglycemic/diabetic rats for sixty days. The serum glucose and insulin levels were monitored at monthly intervals to evaluate hypoglycemic effect of jamun extracts.ResultsThe results of instant research depicted that both seed and fruit extracts reduce the blood glucose level significantly and also regulate the insulin levels in hyperglycemic rats. It was noted that jamun fruit extract attenuated serum glucose levels to 5.35% and 12.29% in normal and hyperglycemic rats, respectively; while insulin levels were improved by 2.82% and 6.19%, correspondingly. Whereas, jamun seed extract reduced glucose to 7.04% & 14.36% and showed 3.56% & 7.24% higher insulin levels in normal & hyperglycemic rats, respectively.ConclusionsThe present research revealed that both jamun fruit and seeds have potent prophylactic role against hyperglycemia. In this respect, diet based regimen may be tailored using jamun fruit/seed and their extracts to alleviate hyperglycemia. 展开更多

This study investigated the anti-anxiety/anti-depression potential of a formula containing red pomegranate fruit extract(RPFE;40%,m/m),Lactobacillus rhamnosus(JB-1)(34%,m/m),magnesium gluconate(25%,m/m)and vanillin(1%,m/m).The RPFE formula(dose:2.0,1.5 or 1.0 mg/g·day)reversed behavioral dysfunctions and body weight gain induced by chronic restraint stress combined with corticosterone injection in C57BL/6 male mice.The RPFE formula exhibited the abilities to normalize the levels of serum infl ammatory cytokines(NF-κB,TNF-α,IL-6,IL-1βand IFN-γ)and malondialdehyde(MDA),and activities of superoxide dismutase(SOD),catalase(CAT)and nitric oxide synthase(NOS),as well as relieve the injury of hippocampal neurons.The serotonin(5-HT)levels in hippocampus were increasingly enhanced,which might be mediated by reducing the activity of indoleamine-2,3-dioxygenase(IDO)and increasing the activity of tryptophan hydroxylase(TPH).Thus,the neuroprotective and ameliorating effects on anxiety/depression-like behaviors resulting from the RPFE formula ingestion were possibly related to serotonergic activation,which might be mediated via anti-infl ammatory and anti-oxidant actions. 展开更多

Jambul (Syzygium cumini) contain several biological activities including anti diabetic, anti-inflammatory, gastroprotective and antibacterial activity. In this study, we investigated antibacterial activity of Jambul juice extract against some common enteric pathogens like Salmonella typhimurium, Shigella flexneri, Staphylococcus aureus, and ETEC (Entero toxigenic E. coli). Growth inhibition of these entero pathogenic bacteria was measured by growing in Nutrient Broth media supplemented with 0%, 5%, 10% and 25% juice extract and then plating on Nutrient Agar plate for colony count at 0, 24 and 48 hours time points. The effect of Jambul juice on the growth of Lactobacillus acidophilus and Lactobacillus bulgaricus were also investigated. We observed that the growth of Salmonella typhimurium, Shigella flexneri, Staphylococcus aureus, and ETEC were significantly inhibited by Jambul juice by 1 to 6 logs (p Lactobacillus acidophilus and Lactobacillus bulgaricus) were not affected significantly. These findings indicate that Jambul juice have selective bactericidal effects against several enteric pathogens while beneficial species remain unaffected. To far our knowledge, this is the first report about the antibacterial activity of fruit juice in Bangladesh. 展开更多

Alcoholic liver injury is a liver disease caused by excessive alcohol consumption,which can lead to chronic liver disease death.Solanum Nigrum Linn taste bitter,cold,has the effect of clearing heat and detoxification,promoting blood and detumescence.Solanum Nigrum Linn fruit contains a variety of antioxidant enzymes,can remove the body produced by aerobic metabolism harmful substances.In this paper,a model of alcohol-induced liver injury in C57BL/6 mice was established to evaluate the protective effect of Solanum Nigrum Linn green fruit(SNGF)ethanolic extract on alcohol-induced liver injury.H&E staining and oil red O(ORO)staining showed that hepatic lobules were clearly demarcated,vacuoles were significantly reduced and lipid droplets were reduced in SNGF ethanolic extract treatment group.Serum levels of TC,TG,LDH,TBA,AKP,ALT and AST were decreased in the SNGF ethanolic extract treatment group,and SNGF ethanolic extract could clear reactive oxygen species(ROS)in time.MDA content was signifi cantly decreased after SNGF ethanolic extract treatment,while superoxide dismutase(SOD)and GSH-Px contents were increased after SNGF ethanolic extract treatment.These results suggest that SNGF ethanolic extract has a protective effect on alcohol-induced liver injury. 展开更多

Objective:To investigate the effect of the ethanolic extract of Rosa laevigata Michx.fruit on rats with mesangial proliferative glomerulonephritis based on the NLRP3 inflammasome pathway.Methods:Thirty Wistar rats were divided into three groups,a blank control group,a diabetic nephropathy(DN)model group,and an ethanolic extract intervention group,according to the random number table method,with 10 rats in each group.One day before the experiment,basic feeding was initiated for all the rats;the changes in activity and weight of each group of rats were observed and recorded after 7 d,and a rat model of renal function injury was established after 1 d.Results:Compared with the control group,the model group had significantly higher kidney/body ratio,24 h urine protein,serum creatinine(SCr),blood urea nitrogen(BUN),glomerular mesangial cell(GMC)count,and extracellular matrix(ECM)positive area ratio(P<0.05);the same indicators were significantly lower in the intervention group than in the model group(P<0.05).The NLRP3 inflammasome pathway in renal intrinsic cells was activated in the intervention group.The overactivation of NLRP3 inflammasome is known to promote interleukin(IL)-1βrelease,which was inhibited in the intervention group.Conclusion:The ethanolic extract of Rosa laevigata Michx.fruit has a protective effect on renal intrinsic cells and may be related to NLRP3 inflammasome pathway,suggesting that the fruit of Rosa laevigata Michx.has a potential role in protecting renal intrinsic cells from inflammatory damage.NLRP3 inflammasomes are involved in the development of various chronic inflammatory diseases,such as acute and chronic glomerulonephritis and renal fibrosis. 展开更多

By compounding the cranberry extract,lavender flower extract,Kakadu plum extract,edelweiss flower/leaf extract and aloe vera extract into a composite plant extract(BIRC-Rosée WT100),the whitening effects of this composite plant extract in antioxidation,anti-inflammation,skin barrier maintenance,and inhibition of melanin production and transport were evaluated.By comparing the inhibitory effects on tyrosinase activity between the composite plant extract andα-arbutin,and then analyzing the decline of apparent chromaticity,apparent brightness and melanin content through 3D melanin skin model platform,the comprehensive whitening effect was evaluated and compared with that of kojic acid.The results showed that,the composite plant extract(BIRC-Rosée WT100)had a tyrosinase inhibition rate of 98.99%at the original concentration,which was much higher than that ofα-arbutin.In 3D melanin skin model,the apparent chromaticity and brightness could be effectively improved,and moreover,the total amount of melanin in the skin could be reduced,showing remarkable whitening effect. 展开更多

Comparison of Antioxidant Properties of Water and Ethanol Extracts Obtained from Dried Boxthorn (<i>Lycium chinensis</i>) Fruit 被引量:4

Antioxidant activities of W and E extracts obtained from dried boxthorn (Lycium chinensis) fruit were measured based on DPPH radical scavenging and reducing powers, and their relationships with total phenolics, flavonoid content, and antioxidant activity were investigated. A linear correlation among antioxidant activity, total phenolics, and flavonoid content was observed in concentration-dependent mode. Both extracts showed > 95% DPPH radical-scavenging activity and the higher reducing power of 3200 ppm at the same concentration. The antioxidant potential of both extracts were compared with those of commercial antioxidants such as BHA, BHT, TBHQ, ferulic acid, and α-tocopherol using H2O2 scavenging activity, inhibition of linoleic acid peroxidation, inhibition of hemolysis of rat erythrocyte induced by peroxyl radicals, and inhibition of Fe2+-induced lipid peroxidation using rat brain tissue. In the H2O2 scavenging activity, E extract showed a comparable significant antioxidant power, comparable to commercial antioxidants, and no signifi-cant difference (P > 0.05) was found between W and E extracts on inhibition of the linoleic acid peroxidation. Whereas W extract exhibited a significant power in the hemolysis of rat erythrocytes, none was observed in E extract. In the Fe-induced lipid peroxidation using rat brain tissue, no significant difference (P > 0.05) was found between both ex-tracts, showing a comparable activity with those of synthetic antioxidants. Both W and E extracts of dried boxthorn (Lycium chinensis) fruit may have a potential as natural antioxidants to replace synthetic antioxidants. 展开更多

One of them is doum fruit extract. The study was conducted to evaluate effect of doum fruit extract on some biochemical parameters, including enzyme activities and histopathological changes of pancreas in alloxan induced diabetic rats. Animals were allocated into five groups of rats, two groups, several as control (one normal and one diabetic rats) and the other three groups were induced diabetic with doum extract (0.5, 1 and 2 g/kg b&middot;wt) which were given every other day by stomach tube for 6 weeks. Evaluation was made for glucose, insulin, lipid profile, enzyme activities, antioxidant activity and histopathological changes of pancreas. The results showed that doum fruit extract decreased blood glucose, insulin, total cholesterol (TC), triglyceride (TG), total lipid (TL), low-density lipoprotein cholesterol (LDL-C), lactate dyhadreygenase (LDH), alanine and aspartate amino transferase (ALT, AST), alkaline phosphatase (ALP) and Malondialdehyde (MDA) while plasma catalase activity and high density lipoprotein cholesterol (HDL-C) were significantly (p 0.05) increased. Therefore, the results of this study demonstrated that the active compounds present in the doum extract have a positive effect on the biochemical parameters of diabetic rats. 展开更多

Objective:Aim To analyze the chemical constituents of essential oil from the fruits of Bupleurum chinense DC.Methods:Essential oil from the fruits of Bupleurum chinense DC.was extracted by using supercritical fluid extraction (SFE),and the chemical constituents to be identified by GC/MS spectrum.Results:The yield of essential oil from the fruits of Bupleurum chinense DC.by SFE was 6.60%(V/W)and 73 components were identified in this oil.Conclusions:The essential oil from the fruits of Bupleurum chinense DC.was produced for the first time,and its yield was 3 times more than its root,the relative contents of linoleic acid,cetyl acetate,ethyl linoleate,(-)-caryophyllene oxide are 34.85%,16.63%,2.64%,1.81% apart in this essential oil.Phytol,cetyl acetate,ethyl linoleate,(-)-caryophyllene oxide were confirmed for the first time from the genus of Bupleurum. 展开更多

Bond.et Sing)为湖北等地习用的桑黄品种,也是土家族常用药,多糖是其主要活性成分。为了研究忍冬木层孔菌子实体多糖(Phellinus lonicerinus polysaccharide, PLP)的提取工艺及其体外抗氧化活性,采用冷凝回流提取法在单因素实验结果的基础上,进行Box-Behnken响应面分析,确定PLP的最佳提取工艺,并通过测定忍冬木层孔菌多糖(PLP30、PLP60、PLP85、PLPA1和PLPA2)的还原能力、对DPPH、ABTS、羟基自由基的清除能力、对酪氨酸酶活性的抑制能力。结果显示PLP最佳提取工艺的参数为:料液比1∶20 g/mL、提取时间120 min、提取温度100℃,此条件下PLP提取率为(3.16±0.05)%;五种多糖对DPPH、ABTS和羟基自由基均具有较强的清除作用,且具有较强的还原力,其对ABTS自由基清除作用和还原力均与浓度呈量效关系。表明忍冬木层孔菌子实体多糖具有良好的体外抗氧化和一定的酪氨酸酶活性抑制能力。 展开更多

为研究百香果果壳提取物对热应激小鼠的保护作用,本试验选取健康清洁级ICR雄性小鼠40只,随机分为对照组、热应激组、低剂量组和高剂量组共4个组,每组10只。所有小鼠饲喂基础颗粒饲料,正常饮水。低剂量组和高剂量组分别按100和200 mg/(kg... 为研究百香果果壳提取物对热应激小鼠的保护作用,本试验选取健康清洁级ICR雄性小鼠40只,随机分为对照组、热应激组、低剂量组和高剂量组共4个组,每组10只。所有小鼠饲喂基础颗粒饲料,正常饮水。低剂量组和高剂量组分别按100和200 mg/(kg·bw·d)剂量灌胃百香果果壳提取物,对照组和热应激组小鼠每天灌胃等量生理盐水。持续灌胃14 d后,除对照组外,其余3个组小鼠每天热处理2 h,连续7 d,期间持续给药。试验结束时,空腹称量小鼠体重;分析小鼠体重变化;采集血液检测血清中谷氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、肌酐(Cr)、尿素氮(BUN)和热休克蛋白70(HSP70)生化指标,以及肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、白介素2(IL-2)和γ干扰素(IFN-γ)细胞因子含量;收集小鼠肝脏和肾脏,观察其组织病理学变化。结果显示,热应激结束后,4个组小鼠体重无显著差异(P>0.05),但热应激组减重最多;与对照组比较,热应激组血清生化和细胞因子各指标均显著升高(P<0.05);与热应激组相比,低剂量组和高剂量组小鼠血清生化和细胞因子各指标均显著降低(P<0.05),小鼠肝脏和肾脏组织损伤有所减轻,且高剂量组优于低剂量组。结果表明,百香果果壳提取物对热应激小鼠具有一定的保护作用。 展开更多

为促进油茶蒲的高值化开发利用,推动油茶产业高质量发展,以油茶蒲为原料,采用超声辅助醇提法制备油茶蒲粗黄酮,并采用AB-8大孔树脂吸附法纯化。另外,对纯化的油茶蒲黄酮的结构进行了紫外光谱和红外光谱表征,利用UPLC-Q-TOF-MS对其组分... 为促进油茶蒲的高值化开发利用,推动油茶产业高质量发展,以油茶蒲为原料,采用超声辅助醇提法制备油茶蒲粗黄酮,并采用AB-8大孔树脂吸附法纯化。另外,对纯化的油茶蒲黄酮的结构进行了紫外光谱和红外光谱表征,利用UPLC-Q-TOF-MS对其组分进行了鉴定。结果表明:采用超声辅助醇提法制备的油茶蒲粗黄酮中黄酮含量为32.41%,采用AB-8大孔树脂纯化后的黄酮含量为70.14%,黄酮总回收率为60.12%;紫外光谱与红外光谱分析表明,纯化物具有明显的黄酮类紫外、红外特征光谱,证明该纯化物主要成分为黄酮类化合物;UPLC-Q-TOF-MS从该纯化物中共鉴定出44种多酚类和黄酮类化合物,其中14种黄酮类化合物、30种多酚类化合物,油茶蒲黄酮多以黄酮苷类化合物形式存在,且多为山奈酚的糖苷类化合物,油茶蒲黄酮中含有原花青素、儿茶素、表儿茶素及没食子酸等抗氧化活性成分。根据油茶蒲黄酮的组成成分,推测其具有开发成为抗糖尿病食品以及与抗癌药物联用治疗疾病的潜力。 展开更多

以人参果为原料,采用水提醇沉法提取人参果水溶性总多糖(the total water-soluble polysaccharide of ginseng fruit,WPGF),并以超滤-纤维素柱层析-凝胶柱层析三级分离模式,对人参果水溶性总多糖WPGF进行全面系统的分级分离纯化,得到人... 以人参果为原料,采用水提醇沉法提取人参果水溶性总多糖(the total water-soluble polysaccharide of ginseng fruit,WPGF),并以超滤-纤维素柱层析-凝胶柱层析三级分离模式,对人参果水溶性总多糖WPGF进行全面系统的分级分离纯化,得到人参果多糖一级级分(the high-polymer polysaccharide of ginseng fruit,WPGFH)1个、二级级分4个(WPGFH-0、WPGFH-0.04、WPGFH-0.08、WPGFH-0.12)、三级级分1个(WPGFH-0.08-1)。采用1-苯基-3-甲基-5-吡唑啉酮柱前衍生化高效液相色谱法(high performance liquid chromatography for pre-column derivatization of 1-phenyl-3-methyl-5-pyrazolone,PMPA-HPLC)和高效凝胶渗透色谱法(high performance gel permeation chromatography,HPGPC)初步分析人参果多糖4个二级级分和1个三级级分的单糖组成、分子质量均一性、分子质量范围,结果表明5个级分均主要含有葡萄糖、半乳糖、阿拉伯糖和鼠李糖及少量甘露糖,WPGFH-0和WPGFH-0.04不含糖醛酸,而其他级分含半乳糖醛酸和葡萄糖醛酸。结合红外光谱分析(Fourier transform infrared spectroscopic investigation,IR)对WPGFH-0.08-1级分进行了结构分析,初步推断其为阿拉伯半乳聚糖型果胶。为人参果多糖的研发提供参考及试验基础。 展开更多

以沙棘果为原料,乙醇溶液为主要溶剂,沙棘果总黄酮提取量为评价指标,采用超声波辅助法在单因素实验基础上结合响应面分析法,综合考察了超声提取时间、提取温度、提取液(乙醇)体积分数、液料比4因素对沙棘果总黄酮的提取量影响.结果表明... 以沙棘果为原料,乙醇溶液为主要溶剂,沙棘果总黄酮提取量为评价指标,采用超声波辅助法在单因素实验基础上结合响应面分析法,综合考察了超声提取时间、提取温度、提取液(乙醇)体积分数、液料比4因素对沙棘果总黄酮的提取量影响.结果表明,沙棘果中总黄酮的最佳提取条件为:超声提取时间34 min、提取温度77℃、提取液(乙醇)体积分数60%、料液比22.8∶1.在此工艺条件下沙棘果总黄酮得率为5.694 mg/g,与理论预测值5.770 mg/g的RSD值为1.33%,小于5%,模型拟合性良好.对制得的沙棘果总黄酮提取物粗品,采用清除DPPH自由基法与羟自由基法,以同浓度的维生素C溶液作为阳性对照,进行体外抗氧化活性测定.按清除DPPH自由基法得到的维生素C、沙棘果提取物的IC 50值分别为0.13、2.32 mg/mL,按羟自由基法得到的IC 50值分别为0.11、0.45 mg/mL,说明沙棘果总黄酮具有较好抗氧化活性. 展开更多
