Symptom presentation and evolution in the first 48 hours after injury are associated with return to play after concussion in elite Rugby Union

Background:Return to play(RTP)in elite rugby is managed using a 6-stage graduated RTP protocol,which can result in clearance to play within 1 week of injury.We aimed to explore how symptom,cognitive,and balance presentation and evolution during concussion screens 2 h(head injury assessment(HIA2)and 48 h(HIA3)after injury were associated with time to RTP)to identify whether a more conservative graduated RTP may be appropriate.Methods:A retrospective cohort study was conducted in 380 concussed rugby players from elite men’s rugby over 3 consecutive seasons.Players were classified as shorter or longer returns,depending on whether RTP occurred within 7 days(allowing them to be considered to play the match 1 week after injury)or longer than 8 days,respectively.Symptom,cognitive,and balance performance during screens was assessed relative to baseline(normal or abnormal)and to the preceding screen(improving or worsening).Associations between sub-test abnormalities and RTP time were explored using odds ratios(OR,longer vs.shorter).Median day absence was compared between players with abnormal or worsening results and those whose results were normal or improving.Results:Abnormal symptom results during screens 2 h and 48 h after concussion were associated with longer return time(HIA2:OR=2.21,95%confidence interval(95%CI):1.39-3.50;HIA3:OR=3.30,95%CI:1.89-5.75).Worsening symptom number or severity from the time of injury to 2 h and 48 h post-injury was associated with longer return(HIA2:OR=2.49,95%CI:1.36-4.58;HIA3:OR=3.34,95%CI:1.10-10.15).Median days absence was greater in players with abnormal symptom results at both HIA2 and HIA3.Cognitive and balance performance were not associated with longer return and did not affect median days absence.Conclusion:Symptom presentation and evolution within 48 h of concussion were associated with longer RTP times.This may guide a more conservative approach to RTP,while still adhering to individualized concussion management principles.

Multiple-stacked Hybrid Plays of lacustrine source rock intervals：Case studies from lacustrine basins in China

Hydrocarbon-producing lacustrine basins are widely developed in the world, and China has a large number of lacustrine basins that have developed since the early Permian. The organic-rich shale-dominated heterogeneous source rock intervals in Chinese lacustrine basins generally contain frequent thin interbeds of stratigraphically associated sandstone, siltstone, marl, dolomite, and limestone. The concept of ‘‘Hybrid Plays＇ ＇ as put forth in this article recognizes this pattern of alternating organicrich shale and organic-lean interbeds and existence of mixed unconventional and conventional plays. Hybrid Plays in lacustrine source rock intervals present a unique closed petroleum system hosting continuous hydrocarbons.The interbedded organic-lean siliciclastic and/or carbonateplays are efficiently charged with hydrocarbons via short migration pathways from the adjacent organic-rich shale that is often also a self-sourced play. We assert ‘‘Hybrid Plays＇ ＇ provide the most realistic exploration model for targeting multiple-stacked and genetically related very tight shale, tight and conventional plays together in the entire source rock interval rather than individual plays only. The Hybrid Play model has been proven and works for a wide variety of lacustrine rift, sag and foreland basins in China.

Defining stratigraphic oil and gas plays by modifying structural plays:A case study from the Xihu Sag,east China Sea Shelf Basin

The study focuses on an area of relatively mature oil and gas exploration located in the western slope of the Xihu Sag,East China Sea Shelf Basin.Three stratigraphic oil and gas plays are introduced to replenish the shrinking prospect inventory in the area.The reservoirs of these plays are traditional pay intervals of the Eocene Pinghu Formation and Baoshi Formation composed of tide-dominated delta front,tidal flat and delta front sand bar facies.By investigating conventional seismic data,seismic inversion volumes,and well log data,multiple tidal-influenced delta front channels,beach sand packages have been mapped out in different reservoir sand intervals of the Pinghu Formation and Baoshi Formation.We recognize four types of stratigraphic terminations:channel flank pinch-outs,axial pinch-outs of sand-fills,channel mouth sand bar pinch-outs,and onlap pinch-outs for the Baoshi Formation.Both the depositional facies and relatively low sand-shale ratio of the reservoir formations favor the occurrence of stratigraphic terminations.The closure mechanism of all these plays are similar to the traditional structure plays in the area:fault blocks or fault-bounded rollover anticlines,except that one or more closing faults are replaced by these stratigraphic terminations.Several small-sized closures should exist but the relatively large ones are centered around a major intra-slope rise,raising the exploration potential in this maturely explored area.The new exploration drilling campaign has so far tested the play of delta front channel facies,with three out of four wells commercially successful.These play concepts open up a novel approach to identify new traps in the study area and many other rift basins with similar settings in eastern China.

Comprehensive analysis of the full-length transcripts and alternative splicing involved in clubroot resistance in Chinese cabbage

Chinese cabbage is an economically important Brassica vegetable worldwide, and clubroot, which is caused by the soilborne protist plant pathogen Plasmodiophora brassicae is regarded as a destructive disease to Brassica crops. Previous studies on the gene transcripts related to Chinese cabbage resistance to clubroot mainly employed RNA-seq technology,although it cannot provide accurate transcript assembly and structural information. In this study, PacBio RS II SMRT sequencing was used to generate full-length transcriptomes of mixed roots at 0, 2, 5, 8, 13, and 22 days after P. brassicae infection in the clubroot-resistant line DH40R. Overall, 39 376 high-quality isoforms and 26 270 open reading frames(ORFs) were identified from the SMRT sequencing data. Additionally, 426 annotated long noncoding RNAs(lncRNAs),56 transcription factor(TF) families, 1 883 genes with poly(A) sites and 1 691 alternative splicing(AS) events were identified. Furthermore, 1 201 of the genes had at least one AS event in DH40R. A comparison with RNA-seq data revealed six differentially expressed AS genes(one for disease resistance and five for defensive response) that are potentially involved in P. brassicae resistance. The results of this study provide valuable resources for basic research on clubroot resistance in Chinese cabbage.

Full-Length Transcriptome Analysis of Cultivated and Wild Tetraploid Peanut

The high-quality genomes and large-scale full-length cDNA sequences of allotetraploid peanuts have been sequenced and released,which has accelerated the functional genomics and molecular breeding research of peanut.In order to understand the difference in the transcriptional levels of wild and cultivated peanuts.In this study,we integrated of second-and third-generation sequencing technologies to sequence full-length transcriptomes in peanut cv.Pingdu9616 and its putative ancestor Arachis monticola.The RNA extracted from six different tissues(i.e.,roots,stems,leaves,flowers,needles and pods)were sampled at 20 days after flowering.A total of 31,764 and 33,981 high-quality transcripts were obtained from Monticola and Pingdu9616,respectively.The number of alternative splicing,the unit point mutation of variable adenylation,the number of open reading frames and the two-site mutation were identified in Pingdu9616 more than in Monticola,but the three-site mutation in Pingdu9616 was lower than in Monticola.1,691 LncRNAs,and 4,000 bp of maximum length of LncRNA was identified in Monticola and Pingdu9616.Furthermore,comparative analysis between transcript data shown that 56 transcription factor families were involved in Monticola,and Pingdu9616 and the number of transcription factors in Pingdu9616 was higher than that in Monticola,the number of expressed genes estimated in flower,root,young pod and leaf organs was higher in Monticola than Pingdu9616.Over all,our study provided a valuable resource of large-scale full-length transcripts for further research of the molecular breeding and functional analysis of genes.

From Cultural Utilization to Cultural Understanding:Two Phases of the English-Speaking World’s Reception of Traditional Chinese Plays

The article analyzes the translations,performances and the reception of traditional Chinese plays in the English-speaking world from the mid-and-late eighteenth century to the early twentieth century.We found that the reception underwent two phases featuring self-centered and multicultural respectively.In the first phase,the English-speaking world acknowledged the moral values of traditional Chinese plays but it did not accept their aesthetic values while in the second phase,aesthetic values became the focus and were received through translations,studies and performances.The two phases differed significantly.The first phase was characterized by the initial contacts and exchanges between the plays and the English culture as well as obvious cultural misunderstandings.During the second phase,moral and aesthetic values were gradually accepted.Text translations,theatrical studies and performances were flourishing while cultural misunderstandings decreased remarkably.The two stages demonstrate the general result of the Chinese culture’s historical process of“going global”.The reception of Chinese culture by other cultures often started with cultural utilization,accompanied by obvious cultural misunderstandings.With the deepening of contacts and exchanges between the two cultures,however,the host culture began to gain a better understanding of the intrinsic values of Chinese culture and therefore cultural misunderstandings decreased.Cultural misunderstandings are an inevitable historical process that the Chinese culture would experience during its“going global”.With continuing and expanded exposures,the intrinsic values of Chinese culture will continue to manifest themselves.

Full-length transcriptome sequence and SSR marker development for genetic diversity research in yellowfin seabream Acanthopagrus latus

Yellowfin seabream Acanthopagrus latus is an important economic fish in Chinese coastal areas.Given its narrow distribution and overfishing,the genetic diversity of yellowfin seabream has been restricted for artificial breeding and reproduction.We performed full-length transcriptome sequencing and assembly of the genome of yellowfin seabream.A total of 68086 unigenes were obtained,with an N50 of 3391 bp on average length of 2933 bp.A total number of 50593 expressed sequence tags linked to simple sequence repeats(EST-SSR)were identified,among them dinucleotide repeats(40.6%)and AC/GT motifs(38.5%)were the most frequent.Of the 190 EST-SSRs for which PCR primer pairs were designed,150 primer pairs successfully amplified target loci and 15 SSRs showed high polymorphism.The alleles per locus ranged 6-50 on average of 25.3.The expected and observed heterozygosity varied from 0.632 to 0.969 and from 0.519 to 0.953,respectively.The polymorphic index content(PIC)values of each locus ranged 0.587-0.966 on average of 0.851.Among six yellowfin seabream population samples preliminarily tested for genetic diversity and differentiation,the Fangchenggang(FCG)population in Guangxi Province had the highest mean observed heterozygosity(H_(o))value(0.786),whereas the Zhangzhou(ZZ)population in Fujian Province had the lowest(0.678).The pairwise fixation index(Fst)values indicated significant population differentiation among six yellowfin seabream populations.This study provided evidence for the usefulness of the transcriptomic resource information and EST-SSR markers for natural resource conservation,population genetics,and breeding studies of yellowfin seabream in South China.
