舒胃汤对FD肝郁脾虚证模型大鼠胃肠平滑肌Beclin-1、mTOR、Cx43蛋白与mRNA表达的影响 被引量:11

目的通过研究舒胃汤(柴胡、香附、白术,等)对功能性消化不良(FD)肝郁脾虚证模型大鼠胃窦和幽门区平滑肌中自噬调控基因Beclin-1、雷帕霉素靶蛋白mTOR、连接蛋白Cx43蛋白与mRNA表达的影响,从自噬与细胞间缝隙连接通道探讨舒胃汤对功能性... 目的通过研究舒胃汤(柴胡、香附、白术,等)对功能性消化不良(FD)肝郁脾虚证模型大鼠胃窦和幽门区平滑肌中自噬调控基因Beclin-1、雷帕霉素靶蛋白mTOR、连接蛋白Cx43蛋白与mRNA表达的影响,从自噬与细胞间缝隙连接通道探讨舒胃汤对功能性消化不良的治疗作用及机制。方法取60只Wistar大鼠随机分为空白组,模型组,莫沙必利组(1.37 mg/kg),舒胃汤低、中、高剂量组(7.67、15.34、30.68 g/kg)。除空白组外,各组大鼠按复合病因造模法造模21 d复制FD肝郁脾虚证大鼠模型。于造模结束后第1天开始给药,空白组与模型组大鼠给予等量蒸馏水,其余各组大鼠按剂量要求灌胃给药,每天1次,连续给药14 d。末次给药2 h后,颈静脉放血处死大鼠,摘取各组大鼠胃窦与幽门组织,Western blot法检测Beclin-1、mTOR、Cx43蛋白表达,RT-qPCR法检测Beclin-1、mTOR、Cx43 mRNA表达。结果 Western blot结果显示,舒胃汤低、中、高剂量组Beclin-1表达明显降低(P<0.01),舒胃汤高剂量组Cx43表达明显升高(P<0.05);RT-qPCR结果显示,舒胃汤中、高剂量组Beclin-1表达明显降低(P<0.01),mTOR表达明显升高(P<0.01),舒胃汤低、中、高剂量组Cx43表达明显升高(P<0.01)。结论舒胃汤可通过上调FD模型大鼠胃肠平滑肌细胞mTOR mRNA、Cx43蛋白及mRNA表达,下调Beclin-1蛋白及mRNA表达,抑制细胞自噬及增强细胞间缝隙连接治疗功能性消化不良。 展开更多

目的:探讨助运除满汤治疗功能性消化不良(FD)的免疫学机理。方法:将60只雄性SD大鼠随机分为空白对照组、模型组、助运除满汤低剂量组(低剂量组)、助运除满汤中剂量组(中剂量组)、助运除满汤高剂量组(高剂量组)及半夏泻心汤组6组,每组10... 目的:探讨助运除满汤治疗功能性消化不良(FD)的免疫学机理。方法:将60只雄性SD大鼠随机分为空白对照组、模型组、助运除满汤低剂量组(低剂量组)、助运除满汤中剂量组(中剂量组)、助运除满汤高剂量组(高剂量组)及半夏泻心汤组6组,每组10只。除空白对照组外余均用夹尾刺激法制作FD动物模型。造模成功后,各组给予相应药物治疗。连续用药14天后,分别检测各组大鼠脾脏指数、胸腺指数及血清IL-2的含量。结果:模型组与空白对照组相比,脾脏指数、胸腺指数及血清IL-2含量均显著下降(P<0.05);与模型组相比,半夏泻心汤组、中剂量组脾脏指数显著升高(P<0.05);对大鼠血清IL-2含量的影响:与模型组大鼠相比,中、高剂量组及半夏泻心汤组大鼠血清IL-2含量显著升高(P<0.05);中、高剂量组比半夏泻心汤组大鼠血清IL-2含量升高更显著(P<0.05)。结论:助运除满汤增强机体免疫功能的作用主要是增强机体体液免疫,而对细胞免疫的影响不明显。 展开更多

Influence of Functional Groups and Modification Sites of Metal-Organic Frameworks on CO2/CH4 Separation:A Monte Carlo Simulation Study

In order to explore the influence of modification sites of functional groups on landfill gas (CO2/CH4) separation performance of metal-organic frameworks (MOFs), six types of or- ganic linkers and three types of functional groups (i.e. -F, -NH2, -CH3) were used to construct 36 MOFs of pcu topology based on copper paddlewheel. Grand canonical Monte Carlo sim- ulations were performed in this work to evaluate the separation performance of MOFs at low (vacuum swing adsorption) and high (pressure swing adsorption) pressures, respectively. Simulation results demonstrated that CO2 working capacity of the unfunctionalized MOFs generally exhibits pore-size dependence at 1 bar, which increases with the decrease in pore sizes. It was also found that -NH2 funetionalized MOFs exhibit the highest CO2 uptake due to the enhanced Coulombic interactions between the polar -NH2 groups and the quadrupole moment of CO2 molecules, which is followed by -CH3 and -F functionalized ones. Moreover, positioning the functional groups -NH2 and -CH3 at sites far from the metal node (site b) exhibits more significant enhancement on CO2/CH4 separation performance compared to that adjacent to the metal node (site a). 展开更多

Objective To study the activation changes of the brain in patients with amyotrophic lateral sclerosis (ALS) while executing sequential finger tapping movement using the method of blood oxygenation level dependent (BOLD) functional magnetic resonance imaging (tMRI). Methods Fifteen patients with definite or probable ALS and fifteen age and gender matched normal controls were enrolled. MRI was performed on a 3.0 Tesla scanner with standard headcoiL The functional images were acquired using a gradient echo single shot echo planar imaging (EPI) sequence. All patients and normal subjects executed sequential finger tapping movement at the frequency of 1-2 Hz during a block-design motor task. Structural MRI was acquired using a three-dimensional fast spoiled gradient echo (3D-FSPGR) sequence. The tMRI data were analyzed by statistical parametric mapping (SPM). Results Bilateral primary sensorimotor cortex ( PSM), bilateral premotor area ( PA), bilateral supplementary motor area (SMA), bilateral parietal region ( PAR), contralateral inferior lateral premotor area ( ILPA), and ipsilateral cerebellum showed activation in both ALS patients and normal controls when executing the same motor task. The activation areas in bilateral PSM, bilateral PA, bilateral SMA, and ipsilateral cerebellum were significantly larger in ALS patients than those in normal controls ( P 〈 0.05 ). Extra activation areas including ipsilateral ILPA, bilateral posterior limb of internal capsule, and contralateral cerebellum were only detected in ALS patients. Conclusions Similar activation areas are activated in ALS patients and normal subjects while executing the same motor task. The increased activation areas in ALS patients may represent neural reorganization, while the extra activation areas in ALS patients may indicate functional compensation. 展开更多

The use of BOLD-contrast imaging in f MRI has been refined over the years, for example, the inclusion of a spin echo pulse and increased magnetic strength were shown to produce better recorded images. Taking careful precautions to control variables during measurement, comparisons between different specimen groups can be illustrated by f MRI imaging using both quantitative and qualitative methods. Differences have been observed in comparisons of active and resting, developing and aging, and defective and damaged brains in various studies. However, cognitive studies using f MRI still face a number of challenges in interpretation that can only be overcome by imaging large numbers of samples. Furthermore, f MRI studies of brain cancer, lesions and other brain pathologies of both humans and animals are still to be explored. 展开更多

These motor i... Parkinson's disease is a neurodegenerative disorder caused by loss of dopamine neurons in the substantia nigra pars compacta. Tremor, rigidity, and bradykinesia are the major symptoms of the disease. These motor impairments are often accompanied by affective and emotional dysfunctions which have been largely studied over the last decade. The aim of this study was to investigate emotional processing organization in the brain of patients with Parkinson's disease and to explore whether there are differences between recognition of different types of emotions in Parkinson's disease. We examined 18 patients with Parkinson's disease(8 men, 10 women) with no history of neurological or psychiatric comorbidities. All these patients underwent identical brain blood oxygenation level-dependent functional magnetic resonance imaging for emotion evaluation. Blood oxygenation level-dependent functional magnetic resonance imaging results revealed that the occipito-temporal cortices, insula, orbitofrontal cortex, basal ganglia, and parietal cortex which are involved in emotion processing, were activated during the functional control. Additionally, positive emotions activate larger volumes of the same anatomical entities than neutral and negative emotions. Results also revealed that Parkinson's disease associated with emotional disorders are increasingly recognized as disabling as classic motor symptoms. These findings help clinical physicians to recognize the emotional dysfunction of patients with Parkinson's disease. 展开更多

Alteration of functional connectivity in patients with Alzheimer’s disease revealed by resting-state functional magnetic resonance imaging 被引量:5

Functional connectivity reflects the synchrony of function... The main symptom of patients with Alzheimer’s disease is cognitive dysfunction. Alzheimer’s disease is mainly diagnosed based on changes in brain structure. Functional connectivity reflects the synchrony of functional activities between non-adjacent brain regions, and changes in functional connectivity appear earlier than those in brain structure. In this study, we detected resting-state functional connectivity changes in patients with Alzheimer’s disease to provide reference evidence for disease prediction. Functional magnetic resonance imaging data from patients with Alzheimer’s disease were used to show whether particular white and gray matter areas had certain functional connectivity patterns and if these patterns changed with disease severity. In nine white and corresponding gray matter regions, correlations of normal cognition, early mild cognitive impairment, and late mild cognitive impairment with blood oxygen level-dependent signal time series were detected. Average correlation coefficient analysis indicated functional connectivity patterns between white and gray matter in the resting state of patients with Alzheimer’s disease. Functional connectivity pattern variation correlated with disease severity, with some regions having relatively strong or weak correlations. We found that the correlation coefficients of five regions were 0.3–0.5 in patients with normal cognition and 0–0.2 in those developing Alzheimer’s disease. Moreover, in the other four regions, the range increased to 0.45–0.7 with increasing cognitive impairment. In some white and gray matter areas, there were specific connectivity patterns. Two metrics, sup port and strength, are presented for measuring the degree of approximate XFD. A basic algorithm is designed for extracting minimal set of approximate XFDs, and then two optimized strategies are proposed to improve the performance. Finally, the experimental results show that the optimized algorithms are correct and effective. 展开更多
