Based on a combination of morphology and molecular data of ribosomal DNA genes,a new diatom genus Lineaperpetua gen.nov.Yu,You,Kociolek&Wang is described.The features that help define Lineaperpetua at the level of genus include:a tangentially undulated valve face;continuous cribra areolae on the valve interior consisting of pores arranged as strips;single rimoportula located inside the ring of marginal fultoportulae.Additionally,phylogenetic analysis based on nuclear small subunit(SSU)rDNA sequences and nuclear large subunit(LSU)rDNA gene placed the three strains of L.lacustris in a single,monophyletic clade at a considerable sequence distance from the other genera(Thalassiosira,Conticribra,Planktoniella,Shinodiscus,and other genera)belonging to Thalassiosirales.Despite the similarities with some species of Thalassiosira,Conticribra,and Spicaticribra,the suite of features found in Lineaperpetua differentiate it from these other genera.These molecular data and morphological characters suggest an affinity of the new genus to the Thalassiosiraceae. 展开更多

During our investigation of diatom biodiversity in Xizang,two species exhibited unique morphological features discriminative from all previously known genera.Herein we describe these two species and describe as new the genus,Spargeria gen.nov.The new genus features narrow to wide rectangular valves,narrow valve mantles,filiform raphe branches that occur on the valve face only,terminal raphe fissures straight or slightly deflected to same side,bow-tie shaped central areas,chambered striae present on the valve face only,being absent from the mantle,wider striae near the axial area and very narrow near the margin,multiseriate striae with small and round areolae that are occluded externally.Comparatively,Spargeria zhuii sp.nov.has larger and robust valves,radiate striae,with one divergent stria near the apices,while Spargeria chenia sp.nov.is smaller,with narrow valves,striae slightly radiate in the middle,becoming convergent or parallel near apices.This new genus belongs to the family Pinnulariaceae,and it was compared and contrasted with other genera of this family.Our work suggests the need for continued studies to document the biodiversity of diatoms in Xizang. 展开更多

The flexed frustules in pennate diatoms are usually associated with monoraphid diatoms. Interestingly, we found a biraphid diatom species with flexed frustules in an offshore intertidal beach environment on Weizhou Island,Beihai City, Guangxi Zhuang Autonomous Region, China. Therefore, based on morphological characteristics, we described a new genus of diatoms Yuzaoea sinensis gen. et sp. nov. CH Li, HH Liu, YH Gao & CP Chen. The frustule of this genus is characterized by heterogeneous frustule with one concave valve and one convex valve,complete raphe on both valves, straight and moderately eccentric raphe, uniseriate striae and girdle bands with a single row of areolae. The most identifying feature of this genus was the flexed frustule, which is rare in biraphid diatoms and common in monoraphid diatoms. We compared the morphometric characteristics of genus Yuzaoea with genus Rhoikoneis and several genera within the family Rhoicospheniaceae, including Rhoicosphenia,Campylopyxis, and Cuneolus. Phylogenetic analyses based on SSU rRNA and rbc L showed that the genus Yuzaoea was the sister group to the clade of Rhoicosphenia with a high support value(bootstrap values = 100%), and the clade “Yuzaoea+Rhoicosphenia” was sister to the clade of monoraphid diatoms, in which the genera Achnanthidium, Planothidium and some Cocconeis with high support values(bootstrap = 100%).Morphologically, the genus Yuzaoea shares many morphological features with monoraphid diatoms like genera Achnanthidium and Planothidium and the members within the Rhoicospheniaceae. Therefore, based on a combined morphological studies and phylogenetic results we suggested that this branch may represented the evolution of one kind monoraphid diatoms, from biraphid diatoms(e.g. genus Yuzaoea), to incompleted biraphid diatoms(e.g. genera Rhoicosphenia, Campylopyxis), to monoraphid diatoms(e.g. genera Achnanthidium and Planothidium). 展开更多

The new genus Parathrausta gen.n. in the subfamily Spilomelinae (Lepidoptera, Crambidae) is erected based on a series of specimens collected in the Afrotropical region of Saudi Arabia. The new genus is monotypical, with Parathrausta internervalis sp.n. its type species. The adults of the type species, the male genitalia, the female genitalia and the tympanal organs are described and figured. The new genus is recognized as related by characters in the male and female genitalia with genera in the tribes Steniini Guine, 1854 and Nomophilini Kuznetzov & Stekolnikov, 1979, namely with Bradina Lederer, 1863, Perisyntrocha Meyrick, 1894, Diathrausta Lederer, 1863, Diasemiopsis Munroe, 1957, Diasemiodes Munroe, 1957 and with the genera in the Duponchelia Zeller, 1847 group comprising Duponchelia Zeller, 1847, Penestola Mschler, 1890, Tatobotys Butler, 1880 and Hymenoptychis Zeller, 1852. The character states differentiating the new genus from the comparative genera are discussed. Potential autapomorphies of the new genus are listed. The tribal assignment of the new genus to the Nomophilini Kuznetzov & Stekolnikov, 1979 based on morphological characters is discussed. The support of the tribal and cladistic assignment of the new genus as a sister clade to Diathrausta Lederer, 1863 based phylogenetic analyses (NJ, MP and ML) is shown. An unusually strong genetic divergence of the genus in the COI segment of the mt-DNA based on the Tamura-Nei distance measure from the morphologically related genera is recognized and addressed. 展开更多

Association between childhood obesity and gut microbiota:16S rRNA gene sequencing-based cohort study

BACKGROUND This study aimed to identify characteristic gut genera in obese and normal-weight children(8-12 years old)using 16S rDNA sequencing.The research aimed to provide insights for mechanistic studies and prevention strategies for childhood obesity.Thirty normal-weight and thirty age-and sex-matched obese children were included.Questionnaires and body measurements were collected,and fecal samples underwent 16S rDNA sequencing.Significant differences in body mass index(BMI)and body-fat percentage were observed between the groups.Analysis of gut microbiota diversity revealed lowerα-diversity in obese children.Differences in gut microbiota composition were found between the two groups.Prevotella and Firmicutes were more abundant in the obese group,while Bacteroides and Sanguibacteroides were more prevalent in the control group.AIM To identify the characteristic gut genera in obese and normal-weight children(8-12-year-old)using 16S rDNA sequencing,and provide a basis for subsequent mechanistic studies and prevention strategies for childhood obesity.METHODS Thirty each normal-weight,1:1 matched for age and sex,and obese children,with an obese status from 2020 to 2022,were included in the control and obese groups,respectively.Basic information was collected through questionnaires and body measurements were obtained from both obese and normal-weight children.Fecal samples were collected from both groups and subjected to 16S rDNA sequencing using an Illumina MiSeq sequencing platform for gut microbiota diversity analysis.RESULTS Significant differences in BMI and body-fat percentage were observed between the two groups.The Ace and Chao1 indices were significantly lower in the obese group than those in the control group,whereas differences were not significant in the Shannon and Simpson indices.Kruskal-Wallis tests indicated significant differences in unweighted and weighted UniFrac distances between the gut microbiota of normal-weight and obese children(P<0.01),suggesting substantial disparities in both the species and quantity of gut microbiota between the two groups.Prevotella,Firmicutes,Bacteroides,and Sanguibacteroides were more abundant in the obese and control groups,respectively.Heatmap results demonstrated significant differences in the gut microbiota composition between obese and normal-weight children.CONCLUSION Obese children exhibited lowerα-diversity in their gut microbiota than did the normal-weight children.Significant differences were observed in the composition of gut microbiota between obese and normal-weight children. 展开更多

Purpose: The genus Pseudomonas is a ubiquitous microorganism frequently detected from immunocompromised patients. The inherent resistance to numerous antimicrobial agents contributes to the opportunistic character of this pathogen exhaustive monitoring of this pathogen is considered of critical importance to public health organizations. The reliable identification method able to distinguish genetic close Pseudomonas species is needed, because these organisms are difficult to differentiate by phenotypic or biochemical methods. The purpose of the present study was to design species-specific primers in order to identify and detect four Pseudomonas species which are frequently detected from the human oral cavities, and to investigate the distribution of these organisms in the living environment using a multiplex PCR. Methods: Polymerase chain reaction (PCR) primers were designed based on partial sequences of the rpoD gene of four Pseudomonas species. Swab samples were collected from fifty washstands, and the distribution of Pseudomonas species was investigated using a conventional PCR at genus level and a multiplex PCR at species level. Results: Multiplex PCR method developed in this study was able to distinguish four Pseudomonas species clearly. The genus Pseudomonas was detected from all samples (100%), whereas P. putida, P, aeruginosa, P. stutzeri and P. fluorescens were detected at 44%, 8%, 4% and 2% in fifty swab samples, respectively. Conclusion: Our developed one-step multiplex PCR method is accurate, specific, cost-effective, time-saving, and works without requiring DNA extraction. It was indicated that washstands were the uninhabitable environment for P. putida, P, aeruginosa, P. stutzeri and P. fluorescens. 展开更多

Taxonomic and faunistic results are presented for three genera of the tribe Pyraustini Meyrick, 1890 of the subfamily Pyraustinae Meyrick, 1890 based on material from the southern Arabian Peninsula. The new genus Dysgrammodes gen.n. is established and placed in the tribe Pyraustini Meyrick, 1890. It shares synapomorphies in the male genitalia with the genera Loxostege Hübner, 1825, Circobotys Butler, 1897, Anania Hübner, 1823, and Pagyda Walker, 1859. Character states differentiating the new genus from these genera are listed. The genus is monotypical, with Dysgrammodes rubrifascialis sp.n. as its type species. Taxonomic and faunistic updates are given for the genera Pyrausta Schrank, 1802 and Anania Hübner, 1823. Pyrausta flavicilialis sp.n. is newly described and placed close to the Afrotropical species Pyrausta centralis Maes, 2009 and Pyrausta grisealis Maes, 2009. Pyrausta phoenicealis Hübner, 1818 is reported for the first time in Saudi Arabia. The presence of the genus Anania Hübner, 1823 is reported as new to the entomofauna of the Arabian Peninsula. The records are attributed to the new species Anania interruptalis sp.n. The new species is recognized as unique among the Afrotropical species of the genus in wing maculation and placed close to Anania murcialis (Ragonot, 1895), Anania hortulata (Linnaeus, 1758) and Anania shanxiensis (Yang & Landry, 2019). The adults of each of the new species and of Pyruasta phoenicealis (Hübner, 1818), the male genitalia of Dysgrammodes rubrifascialis gen.n., sp.n. and of Pyrausta flavicilialis sp.n., the female genitalia of Dysgrammodes rubrifascialis gen.n., sp.n., Anania interruptalis sp.n. and of Pyruasta phoenicealis (Hübner, 1818), and the tympanal organs of Pyrausta flavicilialis sp.n. and of Anania interruptalis sp.n. are described and figured. 展开更多

The genus-level recognition of monophyletic short-legged toads(Brachytarsophrys)has been recently implicated in the taxonomic debate of Megophrys sensu lato.In the present study,Brachytarsophrys is reasonably regarded as a distinct genus based on significant morphological differentiations and recent molecular analyses.Furthermore,a comprehensive review of this genus is performed,with two species groups proposed based on morphological differences and phylogenetic relationships.Particularly,Brachytarsophrys platyparietus is removed as a synonym of Brachytarsophrys carinense and considered a valid species due to significant genetic divergence and distinct morphological differences.In addition,a new species,Brachytarsophrys orientalis sp.nov.,is described based on a series of specimens collected from southeastern China.This work takes the member species of the genus Brachytarsophrys to seven,suggesting that the diversity of Brachytarsophrys is underestimated.In addition,the genus levels of other monophyletic groups within the subfamily Megophryinae are discussed. 展开更多

This new genus differs from AraripelibeUula in the following characters: origins of RP and MA distinctly separated at arculus in both pairs of wings; anal loop wider and shorter, with Y-shaped veins inside; MA and IR2 not zigzag; several small intercalary veins present in the postdiscoidal area of hindwing; cells smaller and much more dense, especially in the apex and hind margin; bigger in size. The flying squirrels(Pteromyini,Rodentia)are the most diverse and widely distributed group of gliding mammals.Taxonomic boundaries and relationships within flying squirrels remain an area of active research in mammalogy.The discovery of new specimens of Pteromys(Hylopetes)leonardi Thomas,1921,previously considered a synonym of Hylopetes alboniger,in Yunnan Province,China allowed a morphological and genetic reassessment of the status of this taxon.Phylogenetic reconstruction was implemented using sequences of two mitochondrial(12S ribosomal RNA and 16S ribosomal RNA)and one nuclear(interphotoreceptor retinoid-binding protein)gene fragments.Morphological assessments involved examinations of features preserved on skins,skulls,and penises of museum specimens,supplemented with principal component analysis of craniometric data.Together these assessments revealed that this taxon should be recognized not only as a distinct species,but should also be placed within a new genus,described here as Priapomys gen.nov. 展开更多

To assesse the genetic diversity among wild and cultivated accessions of 8 taxonomic groups in 2 species, and 5 subspecies under Pisum genus, and to analyze population structure and their genetic relationships among various groups of taxonomy, the study tried to verify the fitness of traditionally botanical taxonomic system under Pisum genus and to provide essential information for the exploration and utilization of wild relatives of pea genetic resources. 197 Pisum accessions from 62 counties of 5 continents were employed for SSR analysis using 21 polymorphic primer pairs in this study. Except for cultivated field pea Pisum sativum ssp. sativum var. sativum (94 genotypes), also included were wild relative genotypes that were classified as belonging to P. fulvum, P. sativum ssp.abyssinicum, P. sativum ssp. asiaticum, P. sativum ssp. transcaucasicum, P. sativum ssp. elatius var. elatius, P. sativum ssp. elatius var. pumilio and P. sativum ssp. sativum var. arvense (103 genotypes). The PCA analyses and 3-dimension PCA graphs were conducted and drawn by NTSYSpc 2.2d statistical package. Nei78 genetic distances among groups of genetic resources were calculated, and cluster analysis using UPGMA method was carried out by using Popgene V1.32 statistical package, the dendrogram was drawn by MEGA3.1 statistical package. Allelic statistics were carried out by Popgene V1.32. The significance test between groups of genotypes was carried out by Fstat V2.9.3.2 statistical package. 104 polymorphic bands were amplified using 21 SSR primer pairs with unambiguous unique polymorphic bands. 4.95 alleles were detected by each SSR primer pair in average, of which 65.56% were effective alleles for diversity. PSAD270, PSAC58, PSAA18, PSAC75, PSAA175 and PSAB72 were the most effective SSR pairs. SSR alleles were uniformly distributed among botanical taxon units under Pisum genus, but significant difference appeared in most pairwise comparisons for genetic diversity between taxon unit based groups of genetic resources. Genetic diversity level of wild species P. fulvum was much lower than the cultivated species P. sativum. Under species P. sativum, P. sativum ssp. sativum var. sativum and P. sativum ssp. asiaticum were the highest in gentic diversity, followed by P. sativum ssp. elatius var. elatius and P. sativum ssp. transcaucasicum, P. sativum ssp. elatius var. pumilio, P. sativum ssp. sativum vat. arvense and P. sativum ssp. abyssinicum were the lowest. Four gene pool clusters were detected under Pisum genus by using PCA analysis. Gene pool "fulvum" mainly consisted of wild species Pisum fulvum, gene pool "abyssinicum" mainly consisted of P. sativum ssp. abyssinicum, and gene pool "arvense" mainly consisted of P. sativum ssp. sativum var. arvense. While gene pool "sativum" were composed by 5 botanical taxon units, they are P. sativum ssp. asiaticum, P. sativum ssp. elatius var. elatius, P. sativum ssp. transcaucasicum, P. sativum ssp. elatius var. pumilio and P. sativum ssp. sativum var. sativum. "sativum" gene pool constructed the primary gene pool of cultivated genetic resources; "fulvum" gene pool, "abyssinicum" gene pool and "arvense" gene pool together constructed the secondary gene pool of cultivated genetic resources. Pairwise Nei78 genetic distance among botanical taxon based groups of pea genetic resources ranged from 7.531 to 35.956, 3 large cluster groups were identified based on the UPGMA dendrogram. Group Ⅰ equals to "sativum" and "arvense" gene pools, Group Ⅱ equals to "abyssinicum" gene pool, and Group Ⅲ equals to "fulvum" gene pool. The UPGMA clustering results generally supporting the PCA clusting results. There were significant differences among most botanical groups under Pisum genus, with clear separation of four gene pools for genetic diversity structure. The research results partially support the traditional botanical taxonomy under Pisum genus, and pointed out its advantage and shortcoming. In order to broaden the genetic bases of pea varieties, the genetic potentials in the four gene pools should be thoroughly exploited. 展开更多

Epizoic diatoms on marine copepods are common in nature and may have a special ecological relationship with their hosts.However,this special ecological group is not well known,and it has only rarely been studied in the China seas.To address this knowledge gap,the species diversity and classification of epizoic diatoms on planktonic copepods were studied with samples collected from the East China Sea.In the present study,a marine araphid diatom genus Protoraphis and its type species,Pr.hustedtiana,were observed and identified by light and electron microscopy,thus representing the first record of this genus and its type species in China.This genus is characterized by a median sternum strongly bent to opposite sides and terminate in two transapical grooves at the valve ends.Protoraphis hustedtiana was found to be epizoic on the posterior body appendages and segments of the marine calanoid copepod Candacia bradyi.An internal view shows a complex,ear-shaped process that is close to the apical slit field.The ecological habitats and geographical distributions of Protoraphis were also discussed,and,together with complementary morphological studies,our results have increased the number of records for marine epizoic diatoms to three genera with three species in China,including Pseudohimantidium and Pseudofalcula. 展开更多
