A method was developed for content determination of Na, Mg, A1, Si, P,S, C1, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Ga, Nb, Zr, Y, Sr, Rb, Ba, La and Ce etc. covering 26 major, minor, and trace elements in marine sedim...A method was developed for content determination of Na, Mg, A1, Si, P,S, C1, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Ga, Nb, Zr, Y, Sr, Rb, Ba, La and Ce etc. covering 26 major, minor, and trace elements in marine sediment samples using fused glass disc by X-ray Fluorescence spectrometry. Calibration was made using marine sediment certified reference materials and the synthetic standard samples prepared by mixing several marine sediments with stream sediment and carbonate standard samples in different proportions. The matrix effect was corrected using theoretical alpha coefficients, experience coefficients and the scattered radiation as the internal standard (for the trace elements). The accuracy of the method was evaluated by analysis of certified reference materials GBW07314, GBW07334 and GSMS6. The results are in good agreement with the certified values of the standards with RSD less than 2.60%, except for Y, Cr, Ga, Ce, La, Nb, Rb, and V with RSD less than 9.0% (n=12).展开更多

In this research, making use of a well known phenomenon of capillary flow, we have proposed an image analysis method of nucleic acids at the price of a small amount of sample. When a droplet of the supramolecular complex solution, formed by neutral red and nucleic acids(NA) under an approximate neutral condition, was placed on the hydrophobic surface of dimethyl dichlorosilane pretreated glass slides, and it was evaporated, the supramolecular complex exhibited the periphery of the droplet due to the capillary effect, and accumulated there to form a red capillary flow directed assembly ring(CFDAR). A typical CFDAR has an outer diameter of (2 r ) about 1.18 mm and a ring width(2 δ ) of about 41 μm. Depending on the experimental conditions, a variety of CFDAR can be assembled. The experimental results are in agreement with our former theoretical discussion. It was found that when a droplet volume is 0.1 μL, the fluorescence intensity of the CFDAR formed by the NR NA is in proportion to the content of calf thymus DNA in the range of 0-0.28 ng, fish sperm DNA of 0-0.24 ng and yeast RNA of 0-0.16 ng with the limit of detection(3 σ ) of 1 7, 1.4 and 0.9 pg, respectively for the three nucleic acids.展开更多

Clear 4', 6-diamidino-2-phenylindole (DAPI) multiple bands like (J-hands could be produced in mouse. The Meta- Morph software was then used to generate linescans of pixel intensity for the banded chromosomes from short arm to long arm. These linescans were sufficient not only to identify each individual chromosome but also analyze the physical sites of bands in chromosome. Based on the results, the clear and accurate karyotype of mouse metaphase chromosomes was established. The technique is therefore considered to he a new method for cytological studies of mouse.展开更多

This is the way of making many visual inspection assays more objective and less time and labor consuming. Also, it can provide new visually inaccessible information on relation between some optical parameters and various biological features of the microbial cul-tures. Of special interest is application of image analysis in fluorescence microscopy as it opens new ways of using fluorescence based methodology for single microbial cell studies. Examples of using image analysis in the studies of both the macroscopic and the microscopic microbiological objects obtained by various imaging techniques are presented and discussed.展开更多

In the PC, an oscillation of local field potential (LFP) with 0.5 - 1 Hz is observed by electrophysiological extracellular recording. Additionally, spatiotemporal neural activities in the PC have been examined using optical recordings. However, extracellular recording is preferable to measure neural activities for a long time with a high speed, while it is not abundant in spatial information. In this study, we therefore attempted to elicit spatial information from extracellular recording. For this purpose, we evaluated spatial information included in the LFP compared with the spatiotemporal neural activities measured by the fluorescent voltage imaging. As a result, aversive odors induced the coherent spatiotemporal neural activities in the PC, and the increase in coherency was observed as a change in the LFP waveform. It was also evaluated as a decrease in entropy by analyzing the LFP oscillation patterns and wavelet analysis. Thus, although the LFP provides only one series of signals, the coherency of the spatiotemporal neural activities in the PC can be evaluated by applying extracellular recording with wavelet analysis.展开更多
Laminar optical tomography(LOT)is a mesoscopic tomographic imaging technique ranging between confocal microscopy and diffuse optical tomography(DOT).Fluorescence LOT(FLOT)provides depth-resolved molecular information ...Laminar optical tomography(LOT)is a mesoscopic tomographic imaging technique ranging between confocal microscopy and diffuse optical tomography(DOT).Fluorescence LOT(FLOT)provides depth-resolved molecular information with 100-200μm resolution over 2-3mm depth.In this study,we use Monte Carlo simulation and singular-value analysis(SVA)to optimize the source-detector configurations for potential enhancement of FLOT imaging performance.The effects of different design parameters,including source incidence and detector collection angles,detector number,and sampling density,are presented.The results indicate that angled incidence/detection configuration might improve the imaging resolution and depth sensitivity,especially for low-scattering medium.Increasing the number of detectors and the number of scanning steps will also result in enhanced imaging performance.We also demonstrate that the optimal imaging performance depends upon the background scattering coe±cient.Our result might provide an optimization strategy for FLOT or LOT experimental setup.展开更多

The weak native fluorescence imaging for Zephyranthes candida (Lindl.) Herb. Pollen cells can be realized within 1 min. The spatial resolution of 0 6 μm per pixel has been achieved with a 40× objective. The imaging speed, sensitivity and spatial resolution are satisfactory, hence the system has a good application prospect in cell biology and medicine. Some factors that influence imaging speed and quality have been discussed preliminarily.展开更多
