BACKGROUND Diabetic intracerebral hemorrhage(ICH)is a serious complication of diabetes.The role and mechanism of bone marrow mesenchymal stem cell(BMSC)-derived exosomes(BMSC-exo)in neuroinflammation post-ICH in patients with diabetes are unknown.In this study,we investigated the regulation of BMSC-exo on hyperglycemia-induced neuroinflammation.AIM To study the mechanism of BMSC-exo on nerve function damage after diabetes complicated with cerebral hemorrhage.METHODS BMSC-exo were isolated from mouse BMSC media.This was followed by transfection with microRNA-129-5p(miR-129-5p).BMSC-exo or miR-129-5poverexpressing BMSC-exo were intravitreally injected into a diabetes mouse model with ICH for in vivo analyses and were cocultured with high glucoseaffected BV2 cells for in vitro analyses.The dual luciferase test and RNA immunoprecipitation test verified the targeted binding relationship between miR-129-5p and high-mobility group box 1(HMGB1).Quantitative polymerase chain reaction,western blotting,and enzyme-linked immunosorbent assay were conducted to assess the levels of some inflammation factors,such as HMGB1,interleukin 6,interleukin 1β,toll-like receptor 4,and tumor necrosis factorα.Brain water content,neural function deficit score,and Evans blue were used to measure the neural function of mice.RESULTS Our findings indicated that BMSC-exo can promote neuroinflammation and functional recovery.MicroRNA chip analysis of BMSC-exo identified miR-129-5p as the specific microRNA with a protective role in neuroinflammation.Overexpression of miR-129-5p in BMSC-exo reduced the inflammatory response and neurological impairment in comorbid diabetes and ICH cases.Furthermore,we found that miR-129-5p had a targeted binding relationship with HMGB1 mRNA.CONCLUSION We demonstrated that BMSC-exo can reduce the inflammatory response after ICH with diabetes,thereby improving the neurological function of the brain. 展开更多

在大语言模型教育应用中,人机会话反馈形式单一问题一直未得到较好解决,这严重影响了学生的高阶思维能力培养。开发具有智能群体多维感知能力的会话机器人,是解决这一问题的重要途径。为此,研究者以群体感知、同伴反馈和研究性学习理论... 在大语言模型教育应用中,人机会话反馈形式单一问题一直未得到较好解决,这严重影响了学生的高阶思维能力培养。开发具有智能群体多维感知能力的会话机器人,是解决这一问题的重要途径。为此,研究者以群体感知、同伴反馈和研究性学习理论为基础,利用文心一言、飞书开发平台和学习分析等技术,开发了智能群体多维感知会话机器人“孔子”,构建了人机协同研学教学模式。教学实验以“现代教育技术应用”课程为教学内容,采用准实验研究设计,邀请数学专业学生参与实验。结果表明,与直接使用文心一言的教学相比,人机协同研学教学模式能显著提升学生的问题解决能力、批判性思维能力、创新能力和外在学习动机,但对学习绩效、内在动机和协作学习能力效果欠佳。为了提高教学效果,研究者需提高多维感知评价精准性、感知维度全面性、情境学习多样性和教师教学技能。 展开更多

Objective Innate lymphoid cells(ILCs)are a class of newly discovered immunocytes.Group 1 ILCs(ILC1s)are identified in the decidua of humans and mice.High mobility group box 1(HMGB1)is predicted to be one of the target genes of miR-142-3p,which is closely related to pregnancy-related diseases.Furthermore,miR-142-3p and HMGB1 are involved in regulating the NF-κB signaling pathway.This study aimed to examine the regulatory effect of miR-142-3p on ILC1s and the underlying mechanism involving HMGB1 and the NF-κB signaling pathway.Methods Mouse models of normal pregnancy and abortion were constructed,and the alterations of ILC1s,miR-142-3p,ILC1 transcription factor(T-bet),and pro-inflammatory cytokines of ILC1s(TNF-α,IFN-γand IL-2)were detected in mice from different groups.The targeting regulation of HMGB1 by miR-142-3p in ILC1s,and the expression of HMGB1 in normal pregnant mice and abortive mice were investigated.In addition,the regulatory effects of miR-142-3p and HMGB1 on ILC1s were detected in vitro by CCK-8,Annexin-V/PI,ELISA,and RT-PCR,respectively.Furthermore,changes of the NF-κB signaling pathway in ILC1s were examined in the different groups.For the in vivo studies,miR-142-3p-Agomir was injected in the uterus of abortive mice to evaluate the abortion rate and alterations of ILC1s at the maternal-fetal interface,and further detect the expression of HMGB1,pro-inflammatory cytokines,and the NF-κB signaling pathway.Results The number of ILC1s was significantly increased,the level of HMGB1 was significantly upregulated,and that of miR-142-3p was considerably downregulated in the abortive mice as compared with the normal pregnant mice(all P<0.05).In addition,miR-142-3p was found to drastically inhibit the activation of the NF-κB signaling pathway(P<0.05).The number of ILC1s and the levels of pro-inflammatory cytokines were significantly downregulated and the activation of the NF-κB signaling pathway was inhibited in the miR-142-3p Agomir group(all P<0.05).Conclusion miR-142-3p can regulate ILC1s by targeting HMGB1 via the NF-κB signaling pathway,and attenuate the inflammation at the maternal-fetal interface in abortive mice. 展开更多

目的探究亚太肝脏研究协会ACLF(AARC-ACLF)评分联合中性粒细胞与淋巴细胞比值(NLR)对HBV相关慢加急性肝衰竭(ACLF)短期预后的预测价值。方法选择94例HBV相关ACLF患者作为研究对象,于入组24 h内检测NLR,计算AARC-ACLF评分。根据临床疗效... 目的探究亚太肝脏研究协会ACLF(AARC-ACLF)评分联合中性粒细胞与淋巴细胞比值(NLR)对HBV相关慢加急性肝衰竭(ACLF)短期预后的预测价值。方法选择94例HBV相关ACLF患者作为研究对象,于入组24 h内检测NLR,计算AARC-ACLF评分。根据临床疗效及生存情况,分为转归良好组及不佳组。分析HBV相关ACLF患者病情转归的影响因素。评估AARC-ACLF评分和血清NLR水平对HBV相关ACLF预后的预测效能。结果94例受试者中,转归良好58例,转归不佳36例。转归不佳组凝血酶原时间(PT)、总胆红素(TBil)、白细胞(WBC)、NLR及AARC-ACLF评分分别为(26.68±6.91)s、(330.2±126.4)μmol/L、(6.67±2.06)×10^(9)/L、6.45±2.76、(10.14±1.29)分,转归良好组为(20.32±4.20)s、(180.1±83.7)μmol/L、(5.06±1.97)×10^(9)/L、2.85±1.67、(8.03±1.09)分,差异有统计学意义(P<0.05)。NLR及AARC-ACLF评分偏高均为HBV相关ACLF患者病情转归不佳的影响因素(P<0.05);血清NLR、AARC-ACLF评分、NLR联合AARC-ACLF评分预测HBV相关ACLF患者预后的AUC分别为0.878、0.892、0.943。结论HBV相关ACLF患者的血清NLR及AARC-ACLF评分均较高,对该病的短期预后具有一定的临床预测价值,且两者联合预测效能更高。 展开更多

Obesity is recognized as the second highest risk factor for cancer. The pathogenic mechanisms underlying tobaccorelated cancers are well characterized and efective programs have led to a decline in smoking and related cancers, but there is a global epidemic of obesity without a clear understanding of how obesity causes cancer. Obesity is heterogeneous, and approximately 25% of obese individuals remain healthy(metabolically healthy obese, MHO), so which fat deposition(subcutaneous versus visceral, adipose versus ectopic) is "malignant"? What is the mechanism of carcinogenesis? Is it by metabolic dysregulation or chronic inflammation? Through which chemokines/genes/signaling pathways does adipose tissue influence carcinogenesis? Can selective inhibition of these pathways uncouple obesity from cancers? Do all obesity related cancers(ORCs) share a molecular signature? Are there common(overlapping) genetic loci that make individuals susceptible to obesity, metabolic syndrome, and cancers? Can we identify precursor lesions of ORCs and will early intervention of high risk individuals alter the natural history? It appears unlikely that the obesity epidemic will be controlled anytime soon; answers to these questions will help to reduce the adverse efect of obesity on human condition. 展开更多

The technology on obtaining enterosorbent from local feedstock--hydrolyzed lignin of cotton seed hulls named Zerotox was developed. The purpose of this research is to conduct a complex study on its physical-chemical and pharmacological properties. Samples of cotton hydrolyzed lignin based enterosorbent Zerotox were investigated, wood hydrolyzed lignin based enterosorbent Polifepan (Russia) was used as a comparison drug. The technology of new medical enterosorbent Zerotox, based on cotton hydrolyzed lignin, has been developed with its physical-chemical and pharmacological characteristics being researched. Comparative research on adsorbing capacity of sorbent Zerotox (80 mg/g) and its analogue--Polipefan (55 mg/g), which is obtained from wood hydrolyzed lignin, has displayed advantages of the new sorbent. IR-spectroscopy research on lignin sorbent Zerotox has shown presence of oxygen-containing functional groups which determine its high absorbing capacity due to its chemisorption. Atomic-adsorptive method has proved high absorbing capacity of the sorbent towards heavy metals ions. Acute and chronic toxicity and antidiarrheal activity of enterosorbent Zerotox have also been studied. 展开更多
