Methods: Antioxidant activities of aqueous infusion of green tea and Ocimum gratissimum (leaves) alone or in combination at various proportions (3:1, 2:1, 1:1, 1:2, 1:3) were determined by DPPH, ABTS, NO and ex-vivo assays including lipid peroxidation and haemolysis. Total phenolic content and flavonoid content was calculated by Folin-Ciocalteu reagent and aluminum chloride colorimetry method, respectively. A correlation study was also conducted between the antioxidant activity and total phenolic/flavonoid content of various infusions. The interactions were analyzed by combination index applying CompuSyn software. Results: Green tea exhibited high radical scavenging ability as compared to Ocimum gratissimum infusion. Combination of green tea and Ocimum gratissimum exhibited moderate antagonism to strong synergistic interaction at various ratios. A strong correlation was found between total phenolic content/total flavonoid content and antioxidant activities of individual infusions (green tea and Ocimum gratissimum). For binary mixture at different ratios, a weak to strong correlation was observed between total phenolic content and antioxidant activity and almost no correlation between total flavonoid content and antioxidant potential. Conclusions: Overall, green tea and Ocimum gratissimum combination (1:1) displayed the highest antioxidant potential and maximum synergism.展开更多

[ Method] A total of 270 broilers at 1 day old were fed on the basic diets supplemented with TP and VE at different proportions. The malondialdehyde (MDA) content, total antioxidant capacity (T-AOC) as well as activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were measured. The whole weight gain and feed conversion rate were also determined. [ Result] The broilers fed on TP and VE had remarkably higher serum activities of SOD and GSH-Px, stronger T-AOC and lower MDA content than the control ones. The best effects were achieved when the supplementary dosage of -rP and VE was respectively 60 and 80 mg/kg · Diet, because the experi- mental broilers had significantly higher antioxidant indices than the control ones ( P 〈 0.01 ). Additionally, the boilers at the age of 21 or 42 days old had significantly higher body weight and lower feed conversion rate than the control ones ( P 〈 0.01 ). [ Conclusion] The study provides a theoreti- cal basis for the applications of TP and VE in animal production.展开更多

Histology activity index (HAI) score at the end of treatment was reduced in 48% of patients who received antioxidant therapy vs 26% of patients who received placebo (P = 0.21). HCV-RNA levels decreased by l-log or more in 28% of patients who received antioxidant therapy vs 12% who received placebo (P = NS). In part 11 of the trial, oral administration of antioxidants was not associated with significant alterations in any of the end points.CONCLUSION: Antioxidant therapy has a mild beneficial effect on the inflammatory response of chronic HCV infection patients who are non-responders to interferon. Combined antiviral and antioxidant therapy may be beneficial for these patients.展开更多

There is dearth of data comparing OS homeostasis of severely obese adults with and without MetS, and need for biomarkers to help in differential diagnosis. Erythrocytic lipid and protein damage markers, malondialdehyde (MDA) and protein carbonyl (PCO), antioxidant enzymes erythrocytic superoxide dismutase(SOD), catalase (CAT), plasma glutathione peroxidase (GPX), and total antioxidant capacity (TAC) as ferric-reducing-ability-of-plasma (FRAP) were compared to understand OS homeostasis among 102 severely Ob (body mass index > 30), 102 Ob with severe (z-score > 2) MetS as per National Cholesterol Education Program-Adult Treatment Panel III guidelines and 100 healthy non-obese Controls. MDA/PCO and all antioxidant enzymes were lowest for ObMetS, followed by Ob, indicating greater damage to protein moieties of the erythrocytic membrane. Multiple regression analysis confirmed z-scores > 2 as significant predictor of lowered enzymes and TAC. Receiver Operator Curve analysis predicted that TAC was the most potential biomarker for the diagnosis and prognosis of MetS with an Odds Ratio of 88.5 indicating the high probability that FRAP would be low for ObMetS (z-score > 2) than for Ob with BMI > 30, but z-scores < 1. TAC is qualified as the most effective biomarker to distinguish between severely obese respondents with and without metabolic syndrome, and as a useful candidate for study of homeostatic breakdown in metabolic syndrome and the importance of z-score in assessment of MetS in obese respondents.展开更多

The cytotoxicity studies of fractions were evaluated in vitro using tetrazolium-based calorimetric assay against human dermal fibroblast, colon adenocarcinoma(Caco-2), breast cancer(MCF-7) and lung cancer(A547) cell lines. The anti-oxidant activity of fractions was determined in vitro against 2,2-diphenyl-1-picrylhydrazyl(DPPH), 2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt(ABTS) and chelation of iron(Fe2+). Gas chromatography mass spectrometry analysis was performed to detect phytochemical constituents in fraction with potent biological activity. Results: Fraction 2 of Jatropha zeyheri roots exhibited the lowest minimum inhibitory concentration of 40 μg/mL against Klebsiella pneumoniae and 80 μg/mL against Candida albicans, Staphylococcus aureus and Mycoplasma hominis. The fractions revealed some varying degrees of cytotoxicity against human dermal fibroblasts yielding LC50 values ranging from 28.96 to 166.52 μg/mL. Fraction 3 exhibited the highest selectivity index value of 2.08 against Klebsiella pneumoniae. Moreover, fraction 2 selectively inhibited the growth of Caco-2 with LC50 of 8.83 μg/mL, compared to other cancerous cell lines. Fraction 2 of Jatropha zeyheri further exhibited IC50 of 19.66, 22.63 and 1.82 μg/mL against DPPH, ABTS and Fe2+, respectively. Gas chromatography mass spectrometry analysis revealed the presence of cyclotetracosane(10.08%), 9-hexacosene(9.40%), hexadecanoic acid(3.90%),(Z)-9-octadecenamide(3.63%), octacosane(2.27%), 11-n-decylheneicosane(2.23%), ethyl vallesiachotamate(2.17%), heneicosanoic acid(2.10%), and octadecanoic acid(2.08%) in fraction 2 of Jatropha zeyheri. Conclusions: These identified compounds, particularly cyclotetracosane(hydrocarbon), may well explain the biological activity of fraction 2 of Jatropha zeyheri, which possesses higher biological activity than other fractions. These compounds can be further investigated for use in treating various bacterial and fungal opportunistic infections associated with HIV-AIDS and related cancers.展开更多

[Method] Totally 500 individuals of 1-day-old Guangxi Partridge chickens were randomly divided into five groups, five replicates each group and 20 chicks each replicate. Groups A, B and C were treatment groups supplemented with 0.70%, 0.35% and0.175% P. felterrae extracts in the basal diet, respectively; group D was a medical control group supplemented with 0.01% colistin sulfate premix in the basal diet; and group E was a control group with basal diet. Serum samples were collected from 10 chickens in each group at 21, 35 and 49 days of age, to analyze the biochemical and immune indexes. [Result] It had no significant impact on serum TP and GLU contents of chickens by adding different levels of P. felterrae extracts in the basal diet(P 〉0.05). However, at 21 and 35 days of age, the TBILI content of chickens in groups A, B and C were extremely lower or significantly lower than that in group E( P〈0.01; P〈0.05). At 35 days of age, the serum AKP activities in groups A, B and C were extremely higher than that in group E(P〈0.01), and the serum GOT activities at 21 and 35 days of age in group C were significantly higher than that in group E(P〈0.05). At 49 days of age, the serum Ig G content in groups A, B and C and the IL-2 content in group B and group C were significantly lower or extremely lower than that in group E( P〈0.05; P〈0.01); the T-AOC content in group B was significantly higher than that in group E(P〈0.05); the GSH content in groups A, B and C were significantly higher than that in group E( P〈0.05). Adding different levels of P. felterrae extracts in the diet significantly decreased the TBILI content, reduced the Ig G, IL-2 and IL-6 contents, and improved the AKP and GOT activities and the T-AOC and GSH contents of Guangxi Partridge chicken. [Conclusion] Adding 0.35% P. felterrae extracts in the basal diet received the best effort.展开更多
