Global climate change can affect tree growth and carbon sink function by influencing plant carbohydrate synthesis and utilization, while elevation can be used as an ideal setting under natural conditions to simulate climate change effects. The effect of elevation on tree growth may depend on organ type. However, the allocation patterns of nonstructural and structural carbohydrates(NSCs and SCs,respectively)in different tree organs and their response to elevation remain unclear. We selected four dominant tree species,Schima superba,Castanopsis eyrei,Castanopsis fargesii and Michelia maudiae, along an elevation gradient from 609 to 1,207 m in subtropical evergreen broad-leaved forests and analyzed leaf, trunk, and fine root NSCs,carbon(C),nitrogen(N)and phosphorus(P)concentrations and the relative abundance of SCs.Leaf NSCs increased initially and then decreased, and trunk NSCs increased with increasing elevation. However, root NSCs decreased with increasing elevation. The relative abundance of SCs in leaves and trunks decreased, while the relative abundance of root SCs increased with increasing elevation. No significant correlations between SCs and NSCs in leaves were detected, while there were negative correlations between SCs and NSCs in trunks, roots, and all organs.Hierarchical partitioning analysis indicated that plant C/N and C/P were the main predictors of changes in SCs and NSCs.Our results suggest that tree organs have divergent responses to elevation and that increasing elevation will inhibit the aboveground part growth and enhance the root growth of trees.A tradeoff between the C distribution used for growth and storage was confirmed along the elevation gradient, which is mainly manifested in the"sink"organs of NSCs.Our results provide insight into tree growth in the context of global climate change scenarios in subtropical forest ecosystems. 展开更多

To study non-structural carbohydrate character-istics and nutrient utilization strategies of Pinus yunnanen-sis under continuous drought conditions,2-year-old seed-lings were planted in pots with appropriate water, light and moderate and severe drought treatments[(80±5),(65±5),(50±5),and(35±5)%of field water-holding capacity].Non-structural carbohydrates, carbon(C),nitrogen(N),and phosphorus(P)concentrations were measured in each plant component. The results show that:(1)With increasing drought,non-structural carbohydrates gradually increased in leaves, stems, and coarse roots, while gradually decreased in fine roots;(2)C concentrations of all were relatively stable under different stress levels.Phosphorous utilization of each component increased under light and moderate drought conditions, while N and P utilization efficiency of each plant component decreased under severe drought.Growth was mainly restricted by N,first decreasing and then increasing with increased drought;(3)There was a correlation between the levels of non-structural carbohydrates and C,N,and P in each component.Changes in N concentration affected the interconversion between soluble sugar and starch, which play a regulatory role in the fluctuation of the concentration of non-structural carbohydrates;and,(4)Plasticity analysis showed that P.yunnanensis seedlings responded to drought mainly by altering starch concentration, the ratio of soluble sugar to starch in leaves and stems, and further by alter-ing N and P utilization efficiencies.Overall, these results suggest that the physiological activities of all organs of P.yunnanensis seedlings are restricted under drought and that trade-offs exist between different physiological indicators and organs. Our findings are helpful in understanding non-structural carbohydrate and nutrient adaptation mechanisms under drought in P.yunnanensis seedlings. 展开更多

BACKGROUND Pancreatic ductal adenocarcinoma(PDAC)is a common cancer with increasing morbidity and mortality due to changes of social environment.AIM To evaluate the significance of serum carbohydrate antigen 19-9(CA19-9)and tumor size changes pre-and post-neoadjuvant therapy(NAT).METHODS This retrospective study was conducted at the Chongqing Key Laboratory of Translational Research for Cancer Metastasis and Individualized Treatment,Chongqing University Cancer Hospital. This study specifically assessed CA19-9 levels and tumor size before and after NAT.RESULTS A total of 156 patients who completed NAT and subsequently underwent tumor resection were included in this study. The average age was 65.4±10.6 years and 72(46.2%)patients were female. Before survival analysis,we defined the post-NAT serum CA19-9 level/pre-NAT serum CA19-9 level as the CA19-9 ratio(CR).The patients were divided into three groups:CR<0.5,CR>0.5 and<1 and CR>1.With regard to tumor size measured by both computed tomography and magnetic resonance imaging,we defined the post-NAT tumor size/pre-NAT tumor size as the tumor size ratio(TR).The patients were then divided into three groups:TR<0.5,TR>0.5 and<1 and TR>1.Based on these groups divided according to CR and TR,we performed both overall survival(OS)and disease-free survival(DFS)analyses.Log-rank tests showed that both OS and DFS were significantly different among the groups according to CR and TR(P<0.05).CR and TR after NAT were associated with increased odds of achieving a complete or near-complete pathologic response. Moreover,CR(hazard ratio:1.721,95%CI:1.373-3.762;P=0.006),and TR(hazard ratio:1.435,95%CI:1.275-4.363;P=0.014)were identified as independent factors associated with OS.CONCLUSION This study demonstrated that post-NAT serum CA19-9 level/pre-NAT serum CA19-9 level and post-NAT tumor size/pre-NAT tumor size were independent factors associated with OS in patients with PDAC who received NAT and subsequent surgical resection. 展开更多

Recently developed ‘super’ rice cultivars with greater yield potentials often suffer from the problem of poor grain filling, especially in inferior spikelets. Here, we studied the activities of enzymes related to starch metabolism in rice stems and grains, and the microstructures related to carbohydrate accumulation and transportation to investigate the effects of different water regimes on grain filling. Two ‘super’ rice cultivars were grown under two irrigation regimes of well-watered(WW) and alternate wetting and moderate soil drying(AWMD). Compared with the WW treatment, the activities of ADP glucose pyrophosphorylase(AGPase), starch synthase(StSase) and starch branching enzyme(SBE), and the accumulation of non-structural carbohydrates(NSCs) in the stems before heading were significantly improved, and more starch granules were stored in the stems in the AWMD treatment. After heading, the activities of α-amylase, β-amylase, sucrose phosphate synthase(SPS) and sucrose synthase in the synthetic direction(SSs)were increased in the stems to promote the remobilization of NSCs for grain filling under AWMD. During grain filling, the enzymatic activities of sucrose synthase in the cleavage direction(SSc), AGPase, StSase and SBE in the inferior spikelets were increased, which promoted grain filling, especially for the inferior spikelets under AWMD.However, there were no significant differences in vascular microstructures. The grain yield and grain weight could be improved by 13.1 and 7.5%, respectively, by optimizing of the irrigation regime. We concluded that the low activities of key enzymes in carbon metabolism is the key limitation for the poor grain filling, as opposed to the vascular microstructures, and AWMD can increase the amount of NSC accumulation in the stems before heading, improve the utilization rate of NSCs after heading, and increase the grain filling, especially in the inferior spikelets, by altering the activities of key enzymes in carbon metabolism. 展开更多

BACKGROUND Gastric cancer(GC)is a common malignant tumor of the digestive system with a high degree of malignancy. It usually develops insidiously without any specific symptoms in the early stages. As one of the diseases caused by abnormal gene changes,GC has abnormal expression of various oncogenes and products during its development.Tumor markers such as carcinoembryonic antigen(CEA),carbohydrate antigen 199(CA199)and carbohydrate antigen 724(CA724)are not expressed or lowly expressed in normal people, but significantly increased after carcinogenesis.Monitoring the changes in the levels of tumor markers such as CEA,CA199 and CA724 is conducive to early diagnosis and evaluation of the occurrence of some solid tumors.AIM To investigate the expression of CEA,CA199 and CA724 in GC and their correlation with clinical features, hoping to provide more effective markers for the early preventive diagnosis of GC.METHODS Of 87 patients with GC admitted to our hospital from September 2020 to December 2021 were included in the GC group, and another 80 healthy people who came to our hospital for physical examination with normal results during the same period were selected as the control group. The serum CEA,CA199,and CA724 levels were compared between the two groups, and the serum CEA,CA199,and CA724 levels were compared in patients with GC at different TNM stages, and the differences in the positive rates of CEA,CA199,and CA724 alone and in combination in detecting TNM stages of GC and GC were compared. In addition, the relationship between the levels of tumor markers CEA,CA199 and CA724 and the clinicopathological characteristics of GC patients was also analyzed. The relationship between the serum levels of CEA,CA199 and CA724 and the survival period of GC patients was analyzed by Pearson.RESULTS The serum levels of CEA,CA199 and CA724 in GC group were significantly higher than those in control group(P<0.05).With the increase of TNM stage, the serum CEA,CA199 and CA724 expression levels in GC patients increased significantly, and the differences between groups were statistically significant(P<0.05).The positive rate of the CA724 single test was higher than that of CEA and CA199 single test(P<0.05).The positive rate of the three combined tests was 95.40%(83/87),which was higher than that of CEA,CA199 and CA724 single tests. The difference was statistically significant(P<0.05).The combined detection positive rates of CEA,CA199,and CA724 in stages I,II,III,and IV of GC were 89.66%,93.10%,98.85%,and 100.00%respectively,all of which were higher than the individual detection rates of CEA,CA199,and CA724.The differences were statistically significant(P<0.05).There was no significant difference in serum CEA,CA199 and CA724 levels between GC patients with different genders, smoking history and alcohol history(P>0.05).However,the serum CEA,CA199 and CA724 levels were significantly higher in GC patients aged≥45 years,TNM stage III-IV,with lymph node metastasis and tumor diameter≥5 cm than in GC patients aged<45 years,TNM stage I-II,without lymph node metastasis and tumor diameter<5 cm(P<0.05).CONCLUSION The expression levels of serum tumor markers CEA,CA199 and CA724 in patients with GC are high and rise with the increase of TNM stage. The levels of CEA,CA199 and CA724 are related to age,TNM stage, lymph node metastasis and tumor diameter. The combined detection of CEA,CA199 and CA724 is helpful to improve the diagnostic accuracy of GC with high clinical guidance value. 展开更多

Crop yield and quality are often limited by the amount of phosphate fertilizer added to infertile soils,a key limiting factor for sustainable development in modern agriculture. The polyphosphate kinase(ppk)gene-expressing transgenic rice with a single-copy line(ETRS)is constructed to improve phosphate fertilizer utilization efficiency for phosphorus resource conservation. To investigate the potential mechanisms of the increased biomass in ETRS in low phosphate culture,ETRS was cultivated in a low inorganic phosphate(Pi)culture medium(15μmol/L Pi,LP)and a normal Pi culture medium(300μmol/L Pi,CP),respectively. After 89 d of cultivation in different concentrations of phosphate culture media, the total phosphorus, polyphosphate(polyP),biomass, photosynthetic rate, nonstructural carbohydrate(NSC)contents, related enzyme activities, and related gene expression levels were analyzed. The results showed that ETRS had a high polyP amount to promote the photosynthetic rate in LP,and its biomass was almost the same as the wild type(WT)in CP.The NSC content of ETRS in LP was higher than that of WT in LP,but slightly lower than that of WT in CP.PolyP notably promoted the sucrose phosphate synthase activities of ETRS and significantly down-regulated the expression levels of sucrose transporter genes(OsSUT3 and OsSUT4),resulting in inhibiting the transport of sucrose from shoot to root in ETRS.It was concluded that polyP can stimulate the synthesis of NSCs in LP,which improved the growth of ETRS and triggered the biological activities of ETRS to save phosphate fertilizer. Our study provides a new way to improve the utilization rate of phosphate fertilizer in rice production. 展开更多

The more frequent occurrence and severer drought events resulting from climate change are increasingly affecting the physiological performance of trees and ecosystem carbon sequestration in many regions of the world. However, our understanding of the mechanisms underlying the responses and adaption of forest trees to prolonged and multi-year drought is still limited. To address this problem,we conducted a long-term manipulative throughfall reduction(TFR,reduction of natural throughfall by 50%–70%during growing seasons)experiment in a natural oriental white oak(Quercus aliena var.acuteserrata Maxim.)forest under warm-temperate climate. After seven years of continuous TFR treatment, the aboveground growth in Q.aliena var.acuteserrata started to decline.Compared with the control plots, trees in the TFR treatment significantly reduced growth increments of stems(14.2%)and leaf area index(6.8%).The rate of net photosynthesis appeared to be more susceptible to changes in soil water in trees subjected to the TFR than in the control. The TFR-treated trees allocated significantly more photosynthates to belowground, leading to enhanced growth and nonstructural carbohydrates(NSC)storage in roots. The 7-year continuous TFR treatment increased the biomass, the production and the NSC concentration in the fine roots by 53.6%,153.6%and 9.6%,respectively.There were clear trade-offs between the aboveground growth and the fine root biomass and NSC storage in Q.aliena var.acuteserrata trees in response to the multi-year TFR treatment.A negative correlation between the fine root NSC concentration and soil water suggested a strategy of preferential C storage over growth when soil water became deficient;the stored NSC during water limitation would then help promote root growth when drought stress is released. Our findings demonstrate the warm-temperate oak forest adopted a more conservative NSC use strategy in response to long-term drought stress, with enhanced root growth and NSC storage at the expenses of above-ground growth to mitigate climate changeinduced drought. 展开更多

The functional lack of 17β-estradiol in postmenopausal women could create a dietary imbalance and cause body weight gain. This study aimed to better understand the interferences that could exist between 17β-estradiol, D2 receptors and the selection of carbohydrate, fat and protein consumption, as well as their consequences on body weight gain by using an animal model of the menopause. Ovariectomy exacerbates the consumption of foods rich in lipids. Thus confirming an inhibitory action of 17β-estradiol (E2) on the consumption of these types of foods. This consumption stimulates body weight gain, which is promoted by the high caloric content of these foods and not by the amount consumed. Our results showed a direct involvement of D2 receptors in food choice. This choice would be made according to the two (2) isoforms of the D2 receptors. The D2/BR isoform directs towards a high carbohydrate consumption, without causing a gain in body weight. While D2/SUL, promotes high fat food consumption, causing an increase in body weight. In women, 17β-estradiol modulates the activity ratio between these two D2 receptor isoforms to ensure energy and homeostatic balance, stabilizing food intake and body weight. 展开更多

Hypoxia is a common environmental stress factor in aquatic organisms, which varies among fish species. However, the mechanisms underlying the ability of fish species to tolerate hypoxia are not well known. Here,we showed that hypoxia response in different fish species was affected by lipid catabolism and preference for lipid or carbohydrate energy sources.Activation of biochemical lipid catabolism through peroxisome proliferator-activated receptor alpha(Pparα)or increasing mitochondrial fat oxidation in tilapia decreased tolerance to acute hypoxia by increasing oxygen consumption and oxidative damage and reducing carbohydrate catabolism as an energy source.Conversely, lipid catabolism inhibition by suppressing entry of lipids into mitochondria in tilapia or individually knocking out three key genes of lipid catabolism in zebrafish increased tolerance to acute hypoxia by decreasing oxygen consumption and oxidative damage and promoting carbohydrate catabolism. However, anaerobic glycolysis suppression eliminated lipid catabolism inhibition-promoted hypoxia tolerance in adipose triglyceride lipase(atgl)mutant zebrafish.Using 14 fish species with different trophic levels and taxonomic status, the fish preferentially using lipids for energy were more intolerant to acute hypoxia than those preferentially using carbohydrates. Our study shows that hypoxia tolerance in fish depends on catabolic preference for lipids or carbohydrates, which can be modified by regulating lipid catabolism. 展开更多

Interactions between water and carbon dynamics underlie drought-related tree mortality. While whole-tree water relations have been shown to play a key role in the response to and recovery from drought, the role of nonstructural carbohydrates(NSC) and how their storage and allocation changes surrounding drought events deserves further attention and is critical for understanding tree survival. Here,we quantified in situ NSC responses of temperate forest trees to the 2016 drought in the northeastern United States.Sugar and starch concentrations were measured in the stemwood of five tree species from 2014 to 2019,which allowed us to monitor NSCs in relation to climatic conditions before, during, and after the natural drought. We found that immediately following the drought, measured stemwood NSC concentrations decreased. However,NSC concentrations rebounded quickly within three years.Notably, trees allocated proportionally more to starch than to sugars following the 2016 drought. In winter 2017,starch comprised 45% of total stemwood stores, whereas starch made up only 1-2% in other years.Further,we modeled and assessed the climatic drivers of total NSC concentrations in the stem.Variation in total NSC concentrations was significantly predicted by the previous year’s temperature, precipitation, and standardized precipitation-evapotranspiration index(SPEI),with stemwood concentrations decreasing following hotter, drier periods and increasing following cooler, wetter periods.Overall, our work provides insight into the climatic drivers of NSC storage and highlights the important role that a tree’s carbon economy may play in its response and recovery to environmental stress. 展开更多

Rice yield stability is a breeding goal, particularly for short-growth duration rice, but its underlying mechanisms remain unclear. In an attempt to identify the relationship between yield stability and source–sink characteristics in short-growth duration rice,a field experiment was conducted at three sites(Yueyang,Liuyang, and Hengyang)in 2021 and 2022.This study compared yield, yield components, source–sink characteristics, and their stability between two stable-yielding short-growth duration rice cultivars,Zhongzao 39(Z-39)and Lingliangyou 268(L-268),and two unstable-yielding short-growth duration rice cultivars,Zhongjiazao 17(Z-17)and Zhuliangyou 819(Z-819).The stability of agronomic parameters was represented by the coefficient of variation(CV).The respective CVs of yield in Z-17,Z-819,Z-39,and L-268 were 10.2%,10.1%,4.5%,and 5.7%in 2021 and 19.7%,15.0%,5.4%,and 6.5%in 2022.The respective CVs of grain weight were 6.3%,5.7%,3.4%,and 4.5%in Z-17,Z-819,Z-39,and L-268 in 2021,and 8.1%,6.3%,1.5%,and 0.8%in 2022.The mean source capacity per spikelet and pre-heading non-structural carbohydrate reserves per spikelet(NSC_(pre))were 7%–43%and7%–72%lower in Z-819 and Z-17than in L-268 and Z-39 in 2021 and 2022.The mean quantum yield of photosystem II photochemistry of leaf, leaf area index, and specific leaf weight of L-268 and Z-39 were higher than those of Z-819 and Z-17 at the heading stage. This study suggests that high NSC_(pre),caused by great leaf traits before heading, increases source capacity per spikelet and its stability, thereby increasing the stability of grain weight and yield.Increasing NSC_(pre)is critical for achieving grain weight and yield stability in short-growth duration rice. 展开更多

In Bacillus thuringenesis(Bt) transgenic cotton, the cotton boll has the lowest insecticidal protein content when compared to the other organs. The present study investigated the effects of amino acid spray application at the peak flowering stage on the cotton boll Bt toxin concentration and yield formation. Boll protein synthesis and carbohydrate conversion were also studied to reveal the fundamental mechanism. Three treatments(i.e., CK, the untreated control;LA1, five amino acids;LA2, 21 amino acids) were applied to two Bt cultivars of G. hirsutum(i.e., the hybrid Sikang 3 and the conventional Sikang 1) in the cotton-growing seasons during 2017 and 2018. Amino acid spray application at the peak flowering stage resulted in an increase of 5.2–16.4% in the boll Bt protein concentration and an increase of 5.5–11.3%in the seed cotton yield, but there was no difference between the two amino acid treatments. In addition, amino acid applications led to increases in the amino acid content, soluble protein content, glutamate pyruvate transaminase(GPT)activity, glutamate oxaloacetate transaminase(GOT) activity, glucose content, fructose content and soluble acid invertase(SAI) activity. This study also found that Bt protein content, enhanced boll number and the weight of opened bolls were closely related to carbon and nitrogen metabolism. The Bt protein content had significant linear positive correlations with amino acid and soluble protein contents. Enhanced boll number had significant linear positive correlations with the GPT and GOT activities from 15–25 days after flowering(DAF). The weight of opened bolls from 55–65 DAF had a significant linear positive correlation with the SAI activity. These results indicate that the enhancement of boll protein synthesis and carbohydrate conversion by amino acid application resulted in a simultaneous increase in the boll Bt protein concentration and cotton lint yield. 展开更多

Stored nonstructural carbohydrates(NSC)indicate a balance between photosynthetic carbon(C)assimilation and growth investment or loss through respiration and root exudation. They play an important role in plant function and whole-plant level C cycling.CO_(2)elevation and nitrogen(N)deposition, which are two major environmental issues worldwide, aff ect plant photosynthetic C assimilation and C release in forest ecosystems. However, information regarding the eff ect of CO_(2)elevation and N deposition on NSC storage in diff erent organs remains limited, especially regarding the trade-off between growth and NSC reserves. Therefore, here we analyzed the variations in the NSC storage in diff erent organs of Chinese fi r(Cunninghamia lanceolata)under CO_(2)elevation and N addition and found that NSC concentrations and contents in all organs of Chinese fi r saplings increased remarkably under CO_(2)elevation.However,N addition induced diff erential accumulation of NSC among various organs.Specifi cally,N addition decreased the NSC concentrations of needles, branches, stems, and fi ne roots, but increased the NSC contents of branches and coarse roots. The increase in the NSC contents of roots was more pronounced than that in the NSC content of aboveground organs under CO_(2)elevation.The role of N addition in the increase in the structural biomass of aboveground organs was greater than that in the increase in the structural biomass of roots. This result indicated that a diff erent tradeoff between growth and NSC storage occurred to alleviate resource limitations under CO_(2)elevation and N addition and highlights the importance of separating biomass into structural biomass and NSC reserves when investigating the eff ects of environmental change on biomass allocation. 展开更多

Nonalcoholic fatty liver disease(NAFLD)is a high-incidence lipid disorder that affects more than a quarter of the population worldwide, and dietary intervention is the recognized treatment.Starch is the main component of staple foods that are consumed daily, and the effects, metabolic pathway, and molecular mechanism of starch in the context of NAFLD remain unclear. Our study showed that a high-starch carbohydrate diet(HCD)led to the occurrence and exacerbation of NAFLD in mice.Transcriptomics and metabonomic analyses showed that the increased fatty acid influx mediated by NADPH oxidase 2(NOX2)exacerbated NAFLD.Knocking down NOX2 specifically alleviated HCD-induced NAFLD in vivo and in vitro. Moreover, the large amounts of ROS produced by NOX2 further exacerbated insulin resistance and increased lipolysis in perirenal white adipose tissue(periWAT),thereby providing fatty acids for hepatic lipid synthesis. In addition, the interaction between AMPKα1 and p47phox was the pathway that mediated the high expression of NOX2 induced by a HCD.Our study systematically demonstrated the effect of a HCD on NAFLD.Elevated fatty acid influx is a unique molecular regulatory pathway that mediates HCD-induced NAFLD exacerbation, which is different from the effect of simple sugars.Additionally,NOX2 was suggested to be a specific and effective drug target for NAFLD. 展开更多

Carnivorous fish have poor tolerance to carbohydrate in feed and low utilization rate of carbohydrate. Therefore, the balance of carbohydrate and lipids in the nutrient metabolism of carnivorous fish, the ef fective conversion and utilization of carbohydrate and lipids, and the feedback regulation of feeding are the key links for the e fficient utilization of carnivorous fish feed.Carbohydrate response element binding protein(ChREBP)is a new transcription factor found in recent years in the glucose signaling pathway, and can also participate in feeding regulation. We performed in-vivo and in-vitro experiments to reveal the role of ChREBP in the glucose metabolism and feeding in mandarin fish. The mRNA expression of ChREBP and appetite regulatory factors were measured after intraperitoneal injection of glucose in mandarin fish Siniperca chuatsi and cotransfection with glucose and glucose+siRNA in the hypothalamic cells in mandarin fish. The results reveal that at hour 2 and 4 post intraperitoneal injection with 1 mg/g BW glucose, the blood glucose level of the mandarin fish increased significantly, but food intake decreased significantly, and it also displayed a significantly increased ChREBP mRNA expression levels in liver. At hour 4 post injection, hypothalamic ChREBP mRNA level was significantly increased, whereas the mRNA expression levels of appetite genes neuropeptide Y(npy)and agouti-related peptide(AgRP)were decreased significantly. When the glucose concentration was 40 mmol/L,the expression level of ChREBP mRNA in mandarin fish hypothalamic cells was significantly up-regulated,but the expression level of appetite gene npy mRNA was down-regulated.When siRNA and glucose were co-transfected into mandarin fish brain cells, the expression level of chrebp mRNA was significantly decreased, and the appetite gene npy mRNA was significantly increased. The results indicated that glucose regulated food intake through the modulation of appetite gene npy by ChREBP. 展开更多

Background Mepiquat chloride(MC)application and plant population density(PPD)increasing are required for modern cotton production. However, their interactive effects on leaf physiology and carbohydrate metabolism remain obscure. This study aimed to examine whether and how MC and PPD affect the leaf morpho-physiological characteristics, and thus final cotton yield.PPD of three levels(D1:2.25 plants·m^(-2),D2:4.5 plants·m^(-2),and D3:6.75 plants·m^(-2))and MC dosage of two levels(MC0:0 g·ha^(-2),MC1:82.5 g·ha^(-2))were combined to create six treatments. The dynamics of nonstructual carbohydrate concentration, carbon metabolism-related enzyme activity, and photosynthetic attributes in cotton leaves were examined during reproductive growth in 2019 and 2020.Results Among six treatments, the high PPD of 6.75 plants·m^(-2)combined with MC application(MC1D3)exhibited the greatest seed cotton yield and biological yield. The sucrose, hexose, starch, and total nonstructural carbohydrate(TNC)concentrations peaked at the first flowering(FF)stage and then declined to a minimum at the first boll opening(FBO)stage.Compared with other treatments,MC1D3 improved starch and TNC concentration by 5.4%~88.4%,7.8%~52.0% in 2019,and by 14.6%~55.9%,13.5%~39.7% in 2020 at the FF stage,respectively.Additionally,MC1D3 produced higher transformation rates of starch and TNC from the FF to FBO stages, indicating greater carbon production and utilization efficiency.MC1D3 displayed the maximal specific leaf weight(SLW)at the FBO stage, and the highest chlorophyll a(Chl a),Chl b,and Chl a+b concentration at the mid-late growth phase in both years. The Rubisco activity with MC1D3 was 2.6%~53.2% higher at the flowering and boll setting stages in both years, and 2.4%~52.7% higher at the FBO stage in 2020 than those in other treatments. These results provided a explanation of higher leaf senescence-resistant ability in MC1D3.Conclusion Increasing PPD coupled with MC application improves cotton yield by enhancing leaf carbohydrate production and utilization efficiency and delaying leaf senescence. 展开更多

The detrimental effects of both diabetes mellitus(DM)and hyperglycemia in the perioperative period are well established and have driven extensive efforts to control blood glucose concentration(BGC)in a variety of clinical settings. It is now appreciated that acute BGC spikes, hypoglycemia, and high glycemic variability(GV)lead to more endothelial dysfunction and oxidative stress than uncomplicated, chronically elevated BGC.In the perioperative setting, fasting is the primary approach to reducing the risk for pulmonary aspiration;however, prolonged fasting drives the body into a catabolic state and therefore may increase GV.Elevated GV in the perioperative period is associated with an increased risk for postoperative complications, including morbidity and mortality. These challenges pose a conundrum for the management of patients typically instructed to fast for at least 8 h before surgery.Preliminary evidence suggests that the administration of an oral preoperative carbohydrate load(PCL)to stimulate endogenous insulin production and reduce GV in the perioperative period may attenuate BGC spikes and ultimately decrease postoperative morbidity, without significantly increasing the risk of pulmonary aspiration. The aim of this scoping review is to summarize the available evidence on the impact of PCL on perioperative GV and surgical outcomes, with an emphasis on evidence pertaining to patients with DM.The clinical relevance of GV will be summarized, the relationship between GV and postoperative course will be explored, and the impact of PCL on GV and surgical outcomes will be presented.A total of 13 articles, presented in three sections, were chosen for inclusion. This scoping review concludes that the benefits of a PCL outweigh the risks in most patients, even in those with well controlled type 2 DM.The administration of a PCL might effectively minimize metabolic derangements such as GV and ultimately result in reduced postoperative morbidity and mortality, but this remains to be proven.Future efforts to standardize the content and timing of a PCL are needed.Ultimately,a rigorous data-driven consensus opinion regarding PCL administration that identifies optimal carbohydrate content, volume, and timing of ingestion should be established. 展开更多

Background: Sugar moiety of macromolecules is today very well known for its implications in many biological recognition mechanisms including cell-cell, extracellular matrix-cell and/or bacteria-cell interactions. In this context lectins, which are carbohydrate-binding proteins displaying a high affinity for sugar groups of other molecules, are of a great importance, notably in immune response involving bacteria, viruses and fungi. As protein-carbohydrate interactions are often mediated by ions such as calcium, zinc or magnesium, we were prompted to study the effect of a thermal spring water (which contains this type of component) on interactions existing between: 1) osidic receptors of human normal keratinocytes and 2) two lectins greatly implicated in the immune response mechanisms (i.e. the dectin-1 and the langerin), and their ligands. Materials and Methods: In a first series of experiments, we studied the effect of increasing concentrations of a thermal spring water on interactions existing between glycosylated molecules and the osidic receptors expressed at the normal human keratinocytes surface. In a second step, and in order to better understand the putative effect of our thermal spring water on the immune response, we analyzed its effect on the interactions existing between the dectin-1 (implicated in the recognition of bacteria, viruses and fungi) and the langerin (expressed by Langerhans cells, the immune cells of the cutaneous tissue), and their ligands in a model using recombinant human lectins and appropriate binding molecules. Results: We showed here that our thermal spring water was able to reinforce interactions between keratinocytes osidic receptors and some of their ligands, in a dose-related manner: From 8% to 55% of increase with 10% to 30% (v/v) of thermal spring water. In the second part of our studies, we also showed that our thermal spring water was able to modulate interactions between dectin-1 and langerin and their ligands through a biphasic effect: Interactions were enhanced by more than 40% and 20% respectively with 10% of thermal spring water, and return to their basal level or lower for higher concentrations. Conclusion: The tested thermal spring water, probably due to its ionic composition, could significantly affect interactions of osidic receptors with their ligands. This property could be of a great interest to help immune system to maintain an appropriate “vigilance state” by using the thermal water at up to a concentration of 10%, and by avoiding any runaway reaction in case of aggression, by using concentrations higher than 10%. . 展开更多

Carbohydrate represents an important part of the soil labile organic carbon pool. Water soluble carbohydrate drives the C cycle in forest soil by affecting microbial activity and hot water extractable car- bohydrate is thought related to soil carbon sequestration due to the asso- ciation with soil aggregation. In a temperate forest region of northeast China, Changbai Mountain, we investigated the abundance, spatial dis- tribution, and seasonal dynamics of cool and hot-water extractable car- bohydrate in soils under mixed broad-leaved Korean pine forest. The concentrations of cool-water extractable carbohydrate (CWECH) in three soil layers (0-5, 5-10, 10-20 cm) ranged from 4.1 to 193.3 g.kg-1 dry soil, decreasing rapidly with soil depth. On an annual average, the CWECH concentrations in soils at depths of 5-10 and 10-20 cm were 54.2% and 24.0%, respectively, of that in the 0-5 cm soil layer. CWECH showed distinct seasonal dynamics with the highest concentrations in early spring, lowest in summer, and increasing concentrations in autumn. Hot-water extractable carbohydrate (HWECH) concentrations in three soil layers ranged from 121.4 to 2026.2 g.kgq dry soil, which were about one order of magnitude higher than CWECH. The abundance of HWECH was even more profile-dependent than CWECH, and decreased more rapidly with soil depth. On an annual average, the HWECH concentration in soils 10-20 cm deep was about one order of magnitude lower than that in the top 0-5 cm soil. The seasonality of HWECH roughly tracked that of CWECH but with seasonal fluctuations of smaller amplitude. The car- bohydrate concentrations in cool/hot water extracts of soil were positively correlated with UV254 and UV2s0 of the same solution, which has implications for predicting the leaching loss of water soluble organic carbon. 展开更多
