Traditional Chinese Medicine Erhuang Suppository for Treatment of Persistent High-risk Human Papillomavirus Infection and Its Impact on Transcriptome of Uterine Cervix

Objective High-risk human papillomavirus(HR-HPV)infection is the chief cause of cervical intraepithelial neoplasia(CIN)and cervical carcinoma.The Erhuang suppository(EHS)is a traditional Chinese medicine(TCM)prepared from realgar(As_(2)S_(2)),Coptidis rhizoma,alumen,and borneolum syntheticum and has been used for antiviral and antitumor purposes.However,whether EHS can efficiently alleviate HR-HPV infection remains unclear.This study was conducted to evaluate the efficacy of EHS for the treatment of persistent HR-HPV infection in the uterine cervix.Methods In this study,we evaluated the therapeutic efficacy of EHS in a randomized controlled clinical trial with a 3-month follow-up.Totally,70 patients with persistent HR-HPV infection were randomly assigned to receive intravaginal administration of EHS or placebo.HPV DNA,ThinPrep cytologic test(TCT),colposcopy,and safety evaluation were carried out after treatment.Microarray analysis was performed to compare transcriptome profiles before and after EHS treatment.A K14-HPV16 mouse model was generated to confirm the efficiency of EHS.Results After 3 months,74.3%(26/35)of the patients in the treatment group were HPV negative,compared to 6.9%(2/29)in the placebo group.High-throughput microarrays revealed distinct transcriptome profiles after treatment.The differentially expressed genes were significantly enriched in complement activation,immune response,and apoptotic processes.The K14-HPV16 mouse model also validated the remarkable efficacy of EHS.Conclusion This study demonstrated that EHS is effective against HR-HPV infection and cervical lesions.Additionally,no obvious systemic toxicity was observed in patients during the trial.The superior efficacy and safety of EHS demonstrated its considerable value as a potential cost-effective drug for the treatment of HPV infection and HPV-related cervical diseases.

Rooted in Chinese traditional culture, Traditional Chinese Medicine contains abundant humanistic thoughts, for instance, pursuing the unity of human and nature, advocating the vital importance of life, sticking to the virtue of "medicine being humane art", abiding by the principle of "Benevolence prior to interest", complying with the medical rule of "Respecting peers". These are the very concrete reflections of medical humanistic spirit. In this article we aim to explore the humanistic thoughts contained in Traditional Chinese Medicine, and try to inherit and carry forward the spirit to better relieve the tensions between doctors and patients and improve the quality of medical services.

Efficacy and mechanisms of Traditional Chinese Medicine prevention and treatment for respiratory viruses

Respiratory virus infection was the most common viral infection in clinical practice with the greatest impact,including the Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),posing a huge threat to the world's public health and human life safety.Commonly used antiviral drugs have obvious side effects and a narrow scope of application.Respiratory viruses are susceptible to infection,mutation,and prevalence,which also pose challenges to traditional antiviral drugs and vaccine development.Traditional Chinese Medicine(TCM)has a long history of treating infectious diseases,with many herbs and compounds.Its multi-component,multi-target and multi-path characteristics have made it have great advantages and potential in the development of new anti-respiratory virus drugs.This review summarized TCM for the prevention and treatment of common respiratory viruses,and provided new strategies for the research and development of new TCM antiviral drugs and for responding to infectious respiratory virus diseases.

In vitro study on the antiviral activity of 9 extracts of traditional Chinese herbal medicine against the human respiratory syncytial virus

Objective:To study the in vitro virucidal activity of 9 extracts of traditional Chinese herbal medicine(the water extracts of Evodia lepta,Clausena lansium,Clerodendrum cyrtophyllum,Callicarpa nudiflora,Nauclea officinalis and Elaeagnus gonyanthes,the alcohol extracts of Nauclea officinalis,Elaeagnus gonyanthes and Zanthoxylumarmatum)on human respiratory syncytial virus(HRSV).Methods:The cytotoxic effect of the extracts on cells was evaluated by a cell viability assay using the CCK-8 method,a concentration of the extracts with cell viability greater than 50%was selected for the follow-up anti-HRSV effect assay,the 50%effective concentration(EC50)was assessed by an in vitro cell infection model.Results:The EC50s of the water extract from Clerodendrum cyrtophyllum,Callicarpa nudiflora and Elaeagnus gonyanthes were 0.05 mg/mL,0.03 mg/mL and 0.05 mg/mL,and the therapeutic index(TI)of them were 18.60,21.67 and 56.80 respectively.Conclusion:The water extracts of Clerodendrum cyrtophyllum,Callicarpa nudiflora and Elaeagnus gonyanthes possess the activity of anti-HRSV virus.

Treating Principles and Methods of Traditional Chinese Medicine in Treatment of Peripheral Vascular Diseases

Peripheral vascular disease embraces a variety of conditions such as thromboangiitis obliterans, Raynaud's disease, thrombophlebitis, aorto-arteritis, obliterate atherosclerosis, varix or phlebothrombosis of the lower limb. Although the affected blood vessel may be arterial or venous one and caused either by inflammation or degenerate changes, they share common symptoms and signs, such as blood stasis, ischemia, thrombosis, ecchymosis, swelling, constriction or obliteration of blood vessels due to circulatory impairment. Since they share common characteristics, the treating principles and methods will be discussed as follows.

Promoting the Development of Traditional Chinese Medicine in the New Century

Reviewing the past, traditional Chinese medicine (TCM) in the 20th century experienced vicissitude. Since the founding the People's Republic of China, especially for the past twenty years, traditional Chinese medicine undertaking, full of vigor and vitality, has made an unprecedented development in China. The legal status and scientific status of traditional Chinese medicine and pharmacology have already been established. The medical system of traditional Chinese medicine, which is composed of traditional Chinese medicine hospitals, ethnic minority medicine hospitals, integration of traditional and western medicine hospitals and comprehensive hospitals, is playing an important role in preventing, treating diseases and enhancing people's health level. The system of production and circulation for traditional Chinese medicine has basically guaranteed the people's demand for Chinese medicine, with the principles of producing medicinal materials as basis, taking Chinese medicine industry as pillar and Chinese medicine as link. Remarkable achievements have been scored in the research of TCM development as well as research of TCM basic theory and clinical treatment. A number of scientific and technological results have been transformed into productive forces. TCM education has formed a rational pattern based on inheriting traditional way of transmission from master to disciple as well as taking the university and college education as the main part. Minority medical science has been further explored and sorted out; the level of medical treatment, teaching and scientific research for ethnic minority medicines has been improved. As the contingent of integrated traditional and western medicine is growing steadily, medical treatment system, scientific research system and teaching system for integrated traditional and western medicine takes shape progressively. International exchange and cooperation on traditional Chinese medicine has been greatly promoted. Anticancer effect of Jinlongshe granules on in situ-transplanted human MKN-45 gastric cancer in nude mice and xenografted sarcoma 180 in Kunming mice and its mechanism

AIM： To study the antitumor effect of Chinese compound Jinlongshe （JLS） granules on sarcoma 180 and MKN-45 human gastric cancer cell lines in vivo and its mechanism. METHODS： After establishment of S180 sarcoma （S180） and MKN-45 gastric cancer model of nude mice, the tumor-bearing mice were divided into 5 groups at random. Three experimental groups were respectively given the aqueous extract of JLS granules at doses of 120 g, 60 g and 20 g/（kg per 6/wk,i.g） for 3 wk in S180 and 6 wk in nude mice model. Positive control was given cyclophosphamide （Cy） at a dose of 50 mg/（kg per 3/wk, i.g） for 3 wk in S180 models and 5-Fluorouracil （5-FU） 20 mg/（kg per 3/wk, i.g） for 3 wk in nude mice model. Negative control was given normal saline （NS） at a dose of 0.18 g/（kg per 6/wk, i.g） respectively. After 3 wk in mice bearing S180 tumor and 6 wk in nude mice model, the experimental animals were sacrificed and the masses of tumor were weighed, and the rates of tumor inhibition of each treated group were calculated respectively. To determine the antitumor mechanisms, the morphological changes, cell cycle and apoptosis were observed in MKN-45 nude mice model. Annexin V-FITC/PI double staining FCM assay was used to further determine the live cells, apoptotic cells, necrotic cells and debris. RESULTS： The inhibitory rates of JLS granules at the doses of 20 g/kg, 60 g/kg and 120 g/kg were 50.31%, 55.94% and 68.13% （P 〈 0.01） in nude mice models and 40.90%, 50.32% and 58.46% （P 〈 0.01） in S180 model. The inhibitory rate of Cy was 85.22% in S180 models and the inhibitory rate of 5-FU was 53.43% in nude mice model （P 〈 0.01）. Nuclear chromatin and margination were observed under a transmission electron microscope （TEM）. The G0/G1 phase was arrested, typical apoptotic peak appeared, the apoptotic rate was 22.81%-38.54% in three JLS granule-treated groups. Annexin V-FITC/PI double staining FCM assay showed that the apoptotic cells were 4.36%, 3.08% and 7.08% in three dosages, most cells were localized in the low right quadrant. CONCLUSION：Jinlongshe granules possess anti-tumor effects on experimental tumor models in vivo, and apoptosis induction is one of its anti-tumor mechanisms.

Inhibitory effect of Fuzheng Yiliuyin in combination with chemotherapeutics on human gastric carcinoma cell strain

AIM： To study the inhibitory effects of Fuzheng Yiliuyin （Decoction for Suppressing Tumors by Strengthening the Body Resistance） in combination with chemotherapeutics on human gastric carcinoma cell strain. METHODS： Fuzheng Yiliuyin （ZY） combined with various kinds of chemotherapeutics was put into two kinds of cultivated human gastric carcinoma cell strains, then its inhibitory effects on human gastric carcinoma cell strains were determind by the MTT method. Flow cytorneter was used to assay the apoptosis rate, and the ultrastructure of gastric carcinoma cells was observed under transmission electron microscope. RESULTS： Obvious apoptosis was seen in gastric carcinoma cells after treatment with ZY for 72 h. ZY and chemical drugs had synergistic inhibition effects on the cultivated gastric carcinoma cells, but the effects were different on various cell strains. The inhibitory effects of ZY could be strengthened by cytotoxic action and apoptosis. ZY combined with tluorouracil, etoposide and cisplatin （EFP） chemotherapeutics had better inhibitory effects on SGC-7901, while ZY combined with EFP or with DDP chemotherapeutics had better inhibitory effects than other drugs on MGC-803. CONCLUSION： ZY induces apoptosis and inhibits the growth of gastric carcinoma cells. ZY has the synergistic function of chemotherapeutics.

Quantum theory-based physical model of the human body in TCM

In the study,a quantum resonant cavity model based on wave-particle duality was proposed for the explanation of the dynamic processes of essence,vigor,and spirit in the human body in traditional Chinese medicine(TCM).It is assumed that there is a macro human order parameter(wave function),and its dynamics are governed by a macro potential field reflecting influences from heaven,earth,and society,and satisfy the generalized Schrodinger equation.This proposed model was applied in the study to interpret basic concepts of human body in TCM,with an aim to unfold the TCM development in the future.

Unity of form and spirit in Huangdi Neijing

Traditional Chinese medicine has many core contents,one of which is the unity of form and spirit.To inherit and develop traditional Chinese medicine,we need to reexamine traditional Chinese medicine with philosophy and medicine.Taking Huangdi Neijing as an example,we restudy the unity of form and spirit in traditional Chinese medicine from two aspects of medicine and philosophy.

Integrated flotation therapy for emotional regulation during the viral recovery period: Applying “form and spirit co-nourishing” in Chinese medicine

Objective:The objective of this study was to evaluate the effectiveness of flotation therapy combined with psychological resilience training in improving emotional regulation and sleep quality in patients recovering from viral infections and to assess the application of the traditional Chinese medicine(TCM)theory of“form and spirit co-nourishing”in modern healthcare interventions.Methods:A total of 20 patients recovering from viral infections with anxiety and sleep disturbances were randomly assigned to the experimental group(n=10)and the control group(n=10).The experimental group received a combination of resilience training,targeting five key areas:physiological regulation,cognitive restructuring,emotional management,self-efficacy,and social support,alongside flotation therapy,with 45-min weekly sessions.The control group received conventional medical treatment.Anxiety levels were measured using the Hamilton Anxiety Scale,while sleep quality was assessed using the Pittsburgh Sleep Quality Index,both before and after the intervention.Results:Compared with intervention before,the resilience level of the experimental group significantly increased after the intervention(P<0.001),while no notable changes were detected in the control group(P>0.05);the experimental group demonstrated significantly lower anxiety level and sleep disorder level and higher resilience level compared to the control group after the intervention(all P<0.001).Conclusion:The combined use of flotation therapy and resilience training,grounded in the TCM principle of“form and spirit co-nourishing,”significantly improves emotional regulation and sleep quality in patients recovering from viral infections.This integrative approach supports both physical and psychological healing,aligning with the TCM holistic treatment model,and offers a promising avenue for modern mental health interventions.

基于对中医药院校学生工匠精神培育的实践经验和理论研究,构建一套科学可行的工匠精神培育成效评价体系,并通过实证研究验证其有效性和可靠性。该评价体系由思想理念、技能水平、品德素养、责任意识和创新能力等方面构成,每个方面包括若干个具体的评价指标,通过权重分配和综合评价方法,得出中医药院校学生工匠精神培育的总体成效评价结果。该评价体系具有科学性、公正性、实用性和可行性,可以为中医药院校学生工匠精神培育提供有效的评价工具和参考依据。

Effect of Traditional Chinese Medicine for Treating Human Immunodeficiency Virus Infections and Acquired Immune Deficiency Syndrome:Boosting Immune and Alleviating Symptoms

To respond to the human immunodeficiency virus （HIV）/acquired immune deficiency syndrome （AIDS） epidemic in China, the integration of antiretroviral therapy （ART） and traditional Chinese medicine （TCM） has important implications in health outcomes, especially in China where the use of TCM is widespread. The National Free TCM Pilot Program for HIV Infected People began in 5 provinces （Henan, Hebei, Anhui, Hubei, and Guangdong） in 2004, and quickly scaled up to 19 provinces, autonomous regions, and municipalities in China including some places with high prevalence, 26,276 adults have been treated thus far. Usually, people with HIV infection seek TCM for four main reasons： to enhance immune function, to treat symptoms, to improve quality of life, and to reduce side effects related to medications. Evidences from randomized controlled clinical trials suggested some beneficial effects of use of traditional Chinese herbal medicine forHIV infections and AIDS. More proofs from large, well-designed, rigorous trials is needed to give firm support. Challenges include interaction between herbs and antiretroviral drugs, stigma and discrimination. The Free TCM Program has made considerable progress in providing the necessary alternative care and treatment for HIV-infected people in China, and has strong government support for continued improvement and expansion, establishing and improving a work mechanism integrating Chinese and Western medicines.

医疗机构基于“三结合”中药新药研发及管理体系的构建

目的:在中药审评审批制度改革的政策背景下,围绕医疗机构,探索中医药理论、人用经验和临床试验相结合(简称“三结合”)的审评证据体系落地举措,为医疗机构中药制剂及中药新药研发提供参考。方法:根据“三结合”审评证据体系下中药新药研发的相关要求,从机构层面加强人用经验研究相关配套建设,从管理层面搭建中药研发、成果转化服务平台,从临床层面规范开展人用经验研究,构建医疗机构中药新药研发及管理体系。结果:医疗机构中药新药研发及管理体系的构建,有助于将临床诊疗数据转变为支持中药新药注册的人用经验证据,通过提高中药研发水平促进中药新药转化。结论:在医疗机构构建基于“三结合”的中药新药研发及管理体系,是“三结合”审评证据体系落地、政策引导研发实践的路径之一。
