The results of UV-visible spectra show that the HB and MCHB could intercalate into the base-stacking domain of the CT-DNA double helix. OBJECTIVE To investigate the killing effect of the combined treatment of ultrasound and hypocrellin B,an active component isolated from a natural product Hypocrella bambuase,on Candida albicans.METHODS At first,Candid...OBJECTIVE To investigate the killing effect of the combined treatment of ultrasound and hypocrellin B,an active component isolated from a natural product Hypocrella bambuase,on Candida albicans.METHODS At first,Candida albicans are incubated with hypcrellin B and the uptake of hypocrellin B in Candida albicans was determined using fluorescence intensity analysis.And then the sensitized Candida albicans were exposed to low-intensity of ultrasound.After the combined treatment of ultrasound and hypocellin B,the growth of Candida albicans was evaluated using the colony counting method,and membrane integrity of Candida albicans was also analyzed by a flow cytometry with propidium iodide staining.RESULTS Hypocrellin B was fast absorbed by candida albicans.The growth of candida albicans was more significantly inhibited by the combined treatment of ultrasound and hypocrellin B than the controls(P<0.05).The more marked damages to membrane integrity were found in the combined treatment than the controls(P<0.05).CONCLUSION The combined treatment of ultrasound and hypocrellin B had significant killing effect on Candida albicans,indicating sonodynamic therapy with hypocrellin B from natural product might be a potential approach for combating fungal infections.展开更多

In ...Liposome is well known as not only a drug-delivery system but also a simple model for biological membranes. It was reported that fluorescence properties of hypocrellins were changeable over some extreme pH values. In the current work, the effects of the microenvironments on the fluorescence properties of HB in liposome, including approximately physiological range of pH values pH = 6.0-8.0, concentration of cholesterols and ionic strength of the solution, were studied. It was found that the fluorescence intensity of HB was sensitive to and also regulated by the microenvironments. When concentration of cholesterols and ionic strength keep invariable in PBS solution, there exists the maximum for the fluorescence of HB-liposome at pH 7.4 while the minimum for that of HB at pH 7.0. In addition, when pH value keeps constant (7.2), there exists the maximum at the ionic strength of 0.12 mol/kg while that at the concentration of 6×10?4 mol/L for cholesterols. On the other hand, in PBS solution, the lower the ionic strength is, the higher the fluorescence intensity is. The environment-sensitive fluorescence may be potentially applicable to probe some specific environmental features in cells or tissues.展开更多

The experimental results show that the quenching of fluorescence in membrane protein by HB can be determined by the principle of dynamic quenching. The experimental process of fluorescence quenching was ob-served in detail by using the ESR technique. It brought to light that the essence of the changes of erythrocyte membranes’ functions caused by membrane protein cross linking and membrane lipid peroxidation, including increase of fluidity and ion permeability of membranes, etc., was that the orderly structure of erythrocyte membranes had been damaged by the active oxygen ( 1O 2, O 2 .- and .OH) generated by HB, including the decrease of α helix, β sheet and the increase of random coil in the main chain of membrane proteins, the decrease of mercapto groups, indole rings, p hydroxy phenyl rings, monosubstituted phenyl rings, etc. in the side chain, and the changes of the conformations of membrane lipids as well. With the increase of the irradiation time, the trans conformation of membrane lipids increased first, then decreased. On the contrary, the gauche conformation decreased first, then increased. In addition, the decrease of the intensities of the lines assigned to bending vibration of the conformation insensitive CH 2 and CH 3 of membrane proteins and lipids suggested that break of their chains had occurred.展开更多

Using Raman spectroscopy, the microcosmic photo...Hypocrellin B (HB) is a highly effective photosensitizer. One of its derivatives, 5_brominated_hypocrellin B (5_Br_HB) has better photosensitive characteristics than it. Using Raman spectroscopy, the microcosmic photodamage to DNA sensitized by HB and 5_Br_HB was studied at molecular level. The results showed that when HB and 5_Br_HB were added to the solution of DNA and irradiated, the characteristic Raman frequencies and intensities of DNA changed to various degrees. The photodamage occurred on the whole DNA molecule, including phosphate backbone,deoxyribose and four bases. Not only the conformation but also the configuration of DNA was photodamaged: breakage of some H_bonds, disappearance of B_form conformation , scission of double or single strand and serious damage of four bases. The damage to A_T pairs was stronger than that to C_G pairs and DNA became polynucleotides finally. The 5_Br_HB_photosensitized damage to DNA was stronger than HB , which suggested the better photosensitive characteristics of 5_Br_HB in bioactive environment. The molecular mechanisms of photodamage to DNA sensitized by 5_Br_HB and HB were also elucidated.展开更多
