Its unique life cycle requires a balanced "conquer and compromise" strategy to deal with the host anti-viral defenses. One of HSV-1 α(immediate early) gene products, infected cell protein 0(ICP0), is a multifunctional protein that interacts with and modulates a wide range of cellular defensive pathways. These pathways may locate in different cell compartments, which then migrate or exchange factors upon stimulation, for the purpose of a concerted and effective defense. ICP0 is able to simultaneously attack multiple host pathways by either degrading key restrictive factors or modifying repressive complexes. This is a viral protein that contains an E3 ubiquitin ligase, translocates among different cell compartments and interacts with major defensive complexes. The multiple functional domains of ICP0 can work independently and at the same time coordinate with each other. Dissecting the functional domains of ICP0 and delineating the coordination of these domains will help us understand HSV-1 pathogenicity as well as host defense mechanisms. This article focuses on describing individual ICP0 domains, their biochemical properties and their implication in HSV-1 infection. By putting individual domain functions back into the picture of host anti-viral defense network, this review seeks to elaborate the complex interactions between HSV-1 and its host.展开更多
Objective: To evaluate the transduction efficiency of a recombinant adenovirus carrying the gene for green fluorescent protein (Ad-GFP) into the primary cultures of fetal neural stem cells (NSCs) by the expression of ...Objective: To evaluate the transduction efficiency of a recombinant adenovirus carrying the gene for green fluorescent protein (Ad-GFP) into the primary cultures of fetal neural stem cells (NSCs) by the expression of GFP. Methods: The Ad-GFP was constructed by homologous recombination in bacteria with the AdEasy system; NSCs were isolated from rat fetal hippocampus and cultured as neurosphere suspensions. After infection with the recombinant Ad-GFP, NSCs were examined with a fluorescent microscopy and a flow cytometry for their expression of GFP. Results: After the viral infection, flow cytometry analysis revealed that the percentage of GFP-positive cells was as high as 97.05%. The infected NSCs sustained the GFP expression for above 4 weeks. After differentiated into astrocytes or neurons, they continued to express GFP efficiently. Conclusion: We have success- fully constructed a viral vector Ad-GFP that can efficiently infect the primary NSCs. The reporter gene was showed fully and sustained expression in the infected cells as well as their differentiated progenies.展开更多

目的探讨血清降钙素原(procalcitonin,PCT)、C反应蛋白(C-reactive protein,CRP)、高迁移率族蛋白B1(recombinant high mobility group protein 1,HMGB1)水平、白细胞计数(white blood cell,WBC)与肺癌合并肺部感染患者的关系,为临床治...目的探讨血清降钙素原(procalcitonin,PCT)、C反应蛋白(C-reactive protein,CRP)、高迁移率族蛋白B1(recombinant high mobility group protein 1,HMGB1)水平、白细胞计数(white blood cell,WBC)与肺癌合并肺部感染患者的关系,为临床治疗提供参考。方法选取2021年1月—2022年12月广西壮族自治区民族医院收治的159例患者为对象。其中肺癌未合并肺部感染者95例作为对照组,肺癌合并肺部感染者64例作为观察组。根据肺部感染评分(clinical pulmonary infection score,CPIS)将其分为轻度组(n=22)、中度组(n=21)、重度组(n=21)。分析肺癌合并肺部感染者病原菌分布,比较2组炎症因子水平、通气功能,不同程度肺部感染患者炎症因子水平、通气功能,并分析其相关性。结果64例肺癌合并肺部感染者痰液标本共检出69株病原菌,其中有18株革兰阳性菌、42株革兰阴性菌、9株真菌;观察组CRP、PCT、HMGB1、WBC、气道峰压(peak inspiratory pressure,PIP)、呼吸做功(work of breath,WOB)、响应频率(frequency response,Fres)高于对照组,动态顺应性(dynamic pulmonary compliance,Cdyn)低于对照组,差异有统计学意义(P<0.05);轻度组、中度组、重度组血清CRP、PCT、HMGB1、WBC、PIP、WOB、Fres呈依次升高趋势,差异有统计学意义(P<0.05);轻度组、中度组、重度组Cdyn呈依次降低趋势,差异有统计学意义(P<0.05);血清PCT、WBC水平与感染程度呈正相关(r=0.363、0.382,P<0.05)。结论肺癌合并肺部感染患者病原菌主要以革兰阴性菌为主,与肺癌患者相比,其血清炎症因子水平更高,通气功能指标较差,且其水平变化与感染程度存在一定关系,临床上可对相关指标进行监测,为临床诊治肺癌合并肺部感染提供参考依据。展开更多
目的分析B族链球菌(group B Streptococcus,GBS)筛查阳性孕母所分娩新生儿的转归及早期炎性指标检测的临床应用价值。方法选取2017年10月至2021年10月解放军总医院第五医学中心收治的GBS筛查阳性孕妇所分娩的135例高危新生儿为研究对象...目的分析B族链球菌(group B Streptococcus,GBS)筛查阳性孕母所分娩新生儿的转归及早期炎性指标检测的临床应用价值。方法选取2017年10月至2021年10月解放军总医院第五医学中心收治的GBS筛查阳性孕妇所分娩的135例高危新生儿为研究对象,根据孕母产前是否应用抗生素分为抗生素组(n=77)及对照组(n=58)。比较两组孕母情况、患儿一般情况、病理性黄疸发生率、不同时间点白细胞(white blood cell,WBC)计数、C反应蛋白(C-reactive protein,CRP)、降钙素原(procalcitonin,PCT)水平。通过受试者操作特征(receiver operating characteristic,ROC)曲线分析不同时间点WBC、CRP、PCT的临床应用价值。统计学方法采用独立样本t检验、χ^(2)检验、秩和检验。结果抗生素组新生儿炎性指标异常率明显低于对照组[26.0%(20/77)与48.3%(28/55),χ^(2)=6.311,P=0.012]。两组患儿分娩方式、羊水污染率、胎膜早破率、病理性黄疸发生率比较,差异均无统计学意义(P值均>0.05)。抗生素组6、24 h CRP水平均低于对照组[0.2(0.1,0.3)mg/L与0.2(0.1,0.4)mg/L,2.3(1.0,6.0)mg/L与4.0(2.8,10.5)mg/L,t=-3.137、-3.010,P值均<0.05]。135例高危新生儿中,发生早发型GBS败血症2例(1.5%)。48例(35.6%)患儿炎性指标异常,平均住院时间(6.3±1.3)d,均痊愈出院。生后24 h WBC及CRP联合检测对于发现新生儿感染意义最大,曲线下面积(area under the curve,AUC)为0.882。结论对GBS筛查阳性孕母分娩前应用抗生素预防治疗可以减少新生儿感染的发生,生后24 h WBC及CRP联合检测对于早期发现新生儿感染有重要意义。展开更多
