Methods Ten infertile men with immotile sperm were enrolled into this study (group A). The membrane integrity in the head and tail regions of individual spermatozoon of immotile sperm was examined by using the combined hypo-osmotic swelling-eosin Y exclusion test (HOS-EY test). Sperm morphology was observed by light, scanning and transmission electron microscopy. Ten semen samples from normospermic donors were used as the control (group B). Results The percentage of sperm with intact both head and tail membranes in group A was significantly lower than that in group B (P〈0.01), whereas the value of sperm with defective head membrane but intact tail membrane in group A was significantly higher than that in group B (P〈0.01). Abnormal sperm morphology in group A had a high incidence, and immotile sperm with viability and normal morphology could be observed in some cases. Most sperm had multiple ultrastructural defects. Conclussion Some immotile sperm had intact tail membrane but defective head membrane. Immotile sperm with viability and normal morphology could exist in some cases though abnormal sperm were in a great proportion. Carefully evaluating immotile sperm membrane integrity and morphology should benefit the treatment of patients with immotile sperm.展开更多

Membrane operation modes generally consist of filtration and backwash processes in a normal mode, a pressure decay test as an integrity test and a chemical circulation through pipe system in a cleaning mode. Thus factors effecting on membrane performance should be sufficiently considered before design and operation. This study evaluated flow analysis for vibration diagnostic and evaluation of the fatigue lifetime in the microfiltration system applied for a drinking water treatment plant. Vibration of main membrane pipelines was measured to identify the source of vibration. Also natural frequency and fluid dynamics was calculated by computational fluid dynamics. It showed that maximum magnitude frequencies were at 12 Hz and 22 Hz, respectively at water and air pipeline during filtration and backwash. Backwash process caused mainly vibration on the backwash water pipe. The calculated frequency from analysis of frequency response and CFD was in a good agreement with the measured frequency. Fatigue analysis showed that pipelines were getting little damage caused by vibration. Fatigue lifetime was predicted more than 15 years under the operation condition of daily filtration, and more than 27 years under the operation condition of a daily backwash mode, resulting in minor damage on the pipe lifetime.展开更多

Methods: Tes...Aim: To examine the expression and regulation of integral membrane protein 2b (Itm2b) in rat male reproductive tissues during sexual maturation and under different treatments by in situ hybridization. Methods: Testis, epididymis, and vas deferens were collected on days 1-70 to examine Itm2b expression during sexual maturation. To further examine the regulation of Itm2b, adult rats underwent surgical castration and cryptorchidism. Ethylene dimethane sulfonate and busulfan treatments were carried out to test the regulation of Itm2b after destruction of Leydig cells and germ cells. Results: In testis, Itm2b expression was moderately detected in the adluminal area of seminiferous cords on days 1-10, and detected at a low level in the spermatogonia on days 20 and 30. The Itm2b level was markedly increased in Leydig cells from day 20 to day 70. In epididymis and vas deferens, Itm2b was detected from neonate to adults, and the signal gradually increased in accordance with sexual maturation. Itm2b expression was significantly downregulated in epididymis and vas deferens of castrated rats, and strongly stimulated when castrated rats were treated with testosterone. Cryptorchidism led to a significant decline of Itm2b expression in testis and caput epididymis. Itm2b expression in epididymis and vas deferens was significantly decreased after the Leydig ceils were destroyed by ethylene dimethane sulfonate. Busulfan treatment produced no obvious change in Itm2b expression in epididymis or vas deferens. Conelusion: Our data suggested that Itm2b expression is upregulated by testosterone and might play a role in rat male reproduction.展开更多
The shape equation of lipid membranes is a fourth-order partial differential equation.Under the axisymmetric condi-tion,this equation was transformed into a second-order ordinary differential equation(ODE)by Zheng and...The shape equation of lipid membranes is a fourth-order partial differential equation.Under the axisymmetric condi-tion,this equation was transformed into a second-order ordinary differential equation(ODE)by Zheng and Liu(Phys.Rev.E 482856(1993)).Here we try to further reduce this second-order ODE to a first-order ODE.First,we invert the usual process of variational calculus,that is,we construct a Lagrangian for which the ODE is the corresponding Euler-Lagrange equation.Then,we seek symmetries of this Lagrangian according to the Noether theorem.Under a certain restriction on Lie groups of the shape equation,we find that the first integral only exists when the shape equation is identical to the Will-more equation,in which case the symmetry leading to the first integral is scale invariance.We also obtain the mechanical interpretation of the first integral by using the membrane stress tensor.展开更多

Methods: The produce of VDAC3 recombinant protein encoded by cDNA sequence of human VDAC3 exon 5-8, based on experimental design of VDAC3 knock-out mice study. And after the purification of various human sperm VDAC3 recombinant proteins, epitope has been predicted in our recombinant protein determined by ElliPro program. Polyclonal antibody was produced for 14 wk. Then anti-VDAC3-exon 5-8 recombinant antiserum was inoculated to human sperm. After the process, antibody VDAC3 protein in human sperm was incubation with anti-VDAC3 recombinant antibody. Finally evaluation the effect of VDAC3 antiserum to human sperm motility and plasma membrane integrity was proceeded.Results: Human VDAC3 recombinant protein was successfully over-expressed in Escherichia coli and purified by affinity chromatography method. Purified human sperm VDAC3 recombinant protein could stimulate immune response in rabbit producing an antibody against VDAC3. Anti-VDAC3 recombinant antibody recognized VDAC3 antigen in human sperm could decrease human sperm motility and membrane integrity significantly.Conclusions:Anti-VDAC3 recombinant polyclonal antibody that we produced in rabbit by ourselves could decrease sperm motility and sperm membrane integrity. The authors suggest this polyclonal antibody could be used as a candidate agent for male contraception in the future. Furthermore, the authors intend to explore the effect of this antibody into sperm function aiming at male contraceptive vaccine development.展开更多

Based on the PARAFAC model, we found that the pea...This study proposed the design, fabrication, and assembly of membrane integrity detection instruments in membrane bioreactors (MBR) based on fluorescence spectroscopy. Based on the PARAFAC model, we found that the peak at 280/335 nm strengthened after membrane breakage. The peak at 340/430 nm reflected the sludge concentration in the MBR and reduced the influence of internal filtration effects on detection. Therefore, we determined that the dual-LED light source excitation detection system can detect tryptophan-like substances at 280 nm (T-peak) and humic acid at 340 nm (C-peak). T-peak was identified as the core index indicating membrane integrity. Moreover, the C-peak is the reference indicator factor for a sensitive response to changes in the sludge concentration. The portable fluorescence instrument exhibited high sensitivity and good feedback accuracy compared to particle counting and turbidity detection, where the log reduction value was greater than 3.5. This overcomes the disadvantage of false alarms in particle counters and is not affected by the position of the pump system. This portable instrument provides a flexible and highly sensitive method for the assessment of industrial membrane integrity.展开更多
The blood-brain barrier is a unique property of central nervous system blood vessels that protects sensitive central nervous system cells from potentially harmful blood components.The mechanistic basis of this barrier...The blood-brain barrier is a unique property of central nervous system blood vessels that protects sensitive central nervous system cells from potentially harmful blood components.The mechanistic basis of this barrier is found at multiple levels,including the adherens and tight junction proteins that tightly bind adjacent endothelial cells and the influence of neighboring pericytes,microglia,and astrocyte endfeet.In addition,extracellular matrix components of the vascular basement membrane play a critical role in establishing and maintaining blood-brain barrier integrity,not only by providing an adhesive substrate for blood-brain barrier cells to adhere to,but also by providing guidance cues that strongly influence vascular cell behavior.The extracellular matrix protein laminin is one of the most abundant components of the basement membrane,and several lines of evidence suggest that it plays a key role in directing blood-brain barrier behavior.In this review,we describe the basic structure of laminin and its receptors,the expression patterns of these molecules in central nervous system blood vessels and how they are altered in disease states,and most importantly,how genetic deletion of different laminin isoforms or their receptors reveals the contribution of these molecules to blood-brain barrier function and integrity.Finally,we discuss some of the important unanswered questions in the field and provide a“to-do”list of some of the critical outstanding experiments.展开更多
This study was performed to investigate the availability of forward osmosis(FO)for microalgae harvesting using sulfonated polyethersulfone(SPES)/PES porous membranes.In FO process,porous membranes(<25.0 L m^−2 h^−1...This study was performed to investigate the availability of forward osmosis(FO)for microalgae harvesting using sulfonated polyethersulfone(SPES)/PES porous membranes.In FO process,porous membranes(<25.0 L m^−2 h^−1)exhibited more superior water flux than TFC FO membranes(<2.6 L m^−2 h^−1).Furthermore,the incorporation of SPES has been demonstrated to enhance membrane performance.The effects of SPES content on pore structure and separation performance were investigated.Compared with pure PES porous membranes,porous membranes with 40%SPES yielded an improved hydrophilicity and greater porosity.It exhibited two times higher water fluxes than the pure PES porous membrane.For microalgae harvesting,AL-FS mode(active layer facing the feed solution)was more favourable than AL-DS mode(active layer facing the draw solution)because less deposited microalgae on the active layer mitigate the membrane biofouling.FO operation combined with SPES/PES porous membranes is conducive to preserving microalgae cell integrity under the mild condition.In addition,FO membrane can be cleaned by a simple water rinse.Potential implications were highlighted as a sustainable method for microalgae harvesting because of no pressure input and less chemical cleaning demand.展开更多
