The Value of the Soluable Intercellular Adhesion Molecule-1 Levels in Matermal Serum for Determination of Occult Chorioamnionitis in Premature Rupture of Membranes 被引量:6

To compare the diagnostic value of soluble intercellular adhesion molecule 1 (sICAM-1) with that of c-reactive protein (CRP) for detecting chorioamnionitis (CAM) in serum of women with premature rupture of membranes (PROM), 55 pregnant women with PROM, including 18 pregnant women with preterm premature rupture of membranes (PPROM) and 20 normal pregnant women at term (TPROM) were studied. Maternal serum were measured by Sandwish enzyme-linked immunoabsorbent assay (ELISA) for sICAM. CAM was histologically confirmed after delivery. The results revealed that (1) maternal serum levels of sICAM-1 and CRP were significantly higher in women with PROM than those without it; (2) maternal serum levels of sICAM-1 and CRP were significantly higher in women with CAM than those without it; (3) serum levels of sICAM-1 in PPROM women were similar to those in TPROM women, whereas serum levels of CRP in PPROM women were significantly higher than those in TPROM women; (4) the sensitivity, specificity, positive predictive value, negative predictive value, Kappa index and area under receiver operating characteristic (ROC) curve of maternal serum sICAM-1 (cutoff 104.7 ng/ml) and CRP(cutoff 1.03 mg/dl) for diagnosing CAM were 100 %, 91.2 %, 87.5 %, 100 %, 0.20, 0.995 and 81.0 %, 73.5 %, 65.4 %, 86.2 %, 0.13, 0.811, respectively; (5) among the mild histological CAM group, severe histological CAM group and clinical CAM group, the difference in maternal serum levels of sICAM-1 were significantly (P<0.001), with the order of concentration from high level to low level corresponding to the severity of CAM. It is concluded that maternal serum level of ICAM-1 is superior to that of CRP as biomarker for diagnosing intraamniotic infection in pregnant women with PROM. 展开更多

BACKGROUND: Previous studies have confirmed the neuroprotective effect of mild hypothermia on ischemic brain injury. OBJECTIVE: To investigate the effects of mild hypothermia on intercellular adhesion molecule-1 expression and serum interleukin-6 levels in ischemic brain tissues of focal brain ischemia rats, and to explore the neuroprotective effects of mild hypothermia on ischemic brain injury. DESIGN, TIME AND SETTING: A randomized, controlled, neurobiological experiment was performed at the Central Laboratory, First Affiliated Hospital, Xinxiang Medical College, China from February to July 2006. MATERIALS: Thirty healthy, adult, Sprague Dawley rats were used to establish middle cerebral artery occlusion models using the suture method, The immunohistochemistry (streptavidin-biotin-peroxidase complex method) kit was purchased from Boster, China. Interleukin-6 radioimmunoassay was supplied by Institute of Radioimmunity, Technology Development Center, General Hospital of Chinese PLA. METHODS: The rats were equally and randomly assigned into mild hypothermia and control groups, and middle cerebral artery occlusion models were established. The rectal temperature was maintained at (37 ±0.5)℃ in the control group. In the mild hypothermia group, the rectal temperature was maintained at (33±1)℃. MAIN OUTCOME MEASURES: At 12 hours after model establishment, the ischemic brain hemispheres were coronally sliced at the level of the optic chiasm. The number of intercellular adhesion molecule-1-positive vessels per high-power field was observed with an optical microscope. Serum interleukin-6 levels were measured by radioimmunoassay. RESULTS: Compared with the control group, intercellular adhesion molecule-1 and serum interleukin-6 expressions were significantly decreased in ischemic brain tissues of the mild hypothermia group (P 〈 0.01). CONCLUSION: Mild hypothermia exhibits a neuroprotective effect by reducing serum interleukin-6 and intercellular adhesion molecule-1 expression following cerebral ischemia. 展开更多

To understand the role of intercellular adhesion molecule-1 (ICAM-1) in immune response of the inner ear, inner ear immune response was induced in rats by inoculation of keyhole limpet hemocyanine (KLH) into the scala tympani of the animals who had been systemically sensitized. The expression of ICAM-1 in the inner ear was immunohistochemically examined. ICAM-1 was found in the epithelium of the spiral modiolar vein (SMV) with its collecting venules (CVs) as early as 6 h after challenge. Expression of ICAM-1 was observed on the epithelium of the endolymphatic sac (ES) and perisaccular region at 12 h. The intensity of ICAM-1 staining reached its peak within 24 - 48 h in these sites of the inner ear. By day 28, most specimens were devoid of appreciable staining for ICAM-1. Our study demonstrates that adhesion molecules play an important role in extravasation of inflammatory cells from the systemic circulation in the process of inner ear immune response. It also shows that cytokines that control expression of adhesion molecules may be released by cells outside ES, besides those cells in the ES. 展开更多

Methods: The concen-tration of sICAM-1 in the serum of 91 patients with ACI was determined with ELISA and then the results were compared wlth those of 43 patients with cerebral hemorrhage and 30 healthy individuals. Results: In the 24th hour after infarction. the concentration of sICAMu-1 in the serum was significantly higher in patients with ACI than in patients with cerebral hemorrhage and normal controls (P< 0. 01). In the patients with ACI, the concentration exhibited an decreasing tendency in the period from the 24th hour to the 14th day andwas correlated with the focal size of cerebral infarction. During the first 14 days after infarction, the concen-tration was significantly higher in the patients with the complication of infection than in those without. Objective:To explore the relationship of plasma homocysteine(Hcy),soluble intercellular adhesion molecule-1(sICAM-1)and high mobility group box 1 protein(HMGB1)with carotid intima-media thickness(c-IMT)in elderly patients with type 2 diabetes mellitus.Methods:A total of 100 elderly patients who were diagnosed as type 2 diabetes mellitus in Baogang Hospital of Inner Mongolia from June 2017 to May 2020 were chosen as research objects.According to c-IMT,they were divided into the normal group(n=35),the mild to moderate group(n=41)and the severe group(n=24).The expression levels of plasma Hcy,sICAM-1 and HMGB1 were compared between groups respectively.Pearson’s correlation coefficient was used to analyze the relationship of plasma Hcy,sICAM-1,HMGB1 with c-IMT.Results:The comparison in plasma Hcy,sICAM-1,HMGB1 and c-IMT among the three groups of patients was of statistical significance(p<.05).The results of correlation analysis showed that the expression levels of plasma Hcy,sICAM-1 and HMGB1 were positively correlated with c-IMT in elderly patients with type 2 diabetes mellitus(r=.627,.598,.614;p<.05).Conclusions:The expression levels of plasma Hcy,sICAM-1 and HMGB1 are abnormally increased in elderly patients with type 2 diabetes mellitus,and related to c-IMT,which can provide a strong evidence for clinical diagnosis and treatment by detecting their levels in clinical practice. 展开更多

目的探讨血浆内皮素-1(ET-1)、D-二聚体(D-D)、血浆可溶性血管细胞黏附分子-1(sVCAM-1)、细胞间黏附因子-1(ICAM-1)在预测突发性耳聋患者预后方面的价值。方法选取2020年6月—2022年6月上海市杨浦区控江医院收治的老年突发性耳聋患者130... 目的探讨血浆内皮素-1(ET-1)、D-二聚体(D-D)、血浆可溶性血管细胞黏附分子-1(sVCAM-1)、细胞间黏附因子-1(ICAM-1)在预测突发性耳聋患者预后方面的价值。方法选取2020年6月—2022年6月上海市杨浦区控江医院收治的老年突发性耳聋患者130例,作为观察组。选取同期检查的健康体检者120例,作为对照组。采用酶联免疫法(ELISA)测量2组患者血浆ET-1、D-D、sVCAM-1、ICAM-1水平。采用受试者工作特征(ROC)曲线评估血浆ET-1、D-D、sVCAM-1、ICAM-1水平预测突发性耳聋患者预后的价值。结果观察组血浆ET-1、D-D、sVCAM-1、ICAM-1水平显著高于对照组,差异有统计学意义(P<0.05)。与高频突发性耳聋相比,低频突发性耳聋患者血浆ET-1、D-D、sVCAM-1、ICAM-1水平更高,差异有统计学意义(P<0.05)。随着突发性耳聋患者听力损失的加重,血浆ET-1、D-D、sVCAM-1、ICAM-1水平显著升高,差异有统计学意义(P<0.05)。与治疗有效组相比,治疗无效组血浆ET-1、D-D、sVCAM-1、ICAM-1水平显著提升,差异有统计学意义(P<0.05)。血浆ET-1、D-D、sVCAM-1和ICAM-1联合应用时预测治疗有效的灵敏度、特异度高于单一指标预测(P<0.05)。结论血浆ET-1、D-D、sVCAM-1、ICAM-1水平升高与突发性耳聋的发生及预后有一定的关系。上述治疗联合检测将有助于临床更好地评价突发性耳聋患者的预后。 展开更多

高氏盆炎方二号方对慢性盆腔炎大鼠的抗炎抗粘连作用及对ICAM-1和caspase-8表达的影响

目的探讨高氏盆炎方二号方对慢性盆腔炎(CPID)大鼠的抗炎抗粘连作用及对细胞间黏附分子-1(ICAM-1)和半胱天冬酶-8(caspase-8)表达的影响。方法首先通过体外抑菌试验观察高氏盆炎方二号方药液的最低抑菌浓度(MIC)。将75只雌性SD大鼠随机... 目的探讨高氏盆炎方二号方对慢性盆腔炎(CPID)大鼠的抗炎抗粘连作用及对细胞间黏附分子-1(ICAM-1)和半胱天冬酶-8(caspase-8)表达的影响。方法首先通过体外抑菌试验观察高氏盆炎方二号方药液的最低抑菌浓度(MIC)。将75只雌性SD大鼠随机分为空白组(n=10)和造模组(n=65)。造模10 d后,在造模组随机抽取5只大鼠处死,进行子宫组织HE染色后镜下呈慢性炎症改变,提示造模成功。随后将60只大鼠再随机分为模型组、中药组、中成药组及人胎盘组织液组,其中中药组又分为低、中、高剂量组,每组10只。模型组给予生理盐水2 mL/只灌胃,中药低、中、高剂量组分别给予高氏盆炎方二号方7、14、28 g/kg灌胃,中成药组给予蒲苓盆炎康颗粒3 g/kg灌胃,人胎盘组织液组给予人胎盘组织液1 mL/kg腹腔注射。以上各给药组均每日1次给药,连续给药20 d。末次给药24 h后处死大鼠并取子宫组织称重,计算子宫肿胀度及子宫系数;光镜下观察大鼠子宫组织病理学改变情况;用免疫组化法及Western-blot法检测大鼠子宫组织中ICAM-1和caspase-8蛋白表达水平。结果高氏盆炎方二号方对金黄色葡萄球菌、大肠杆菌及乙型溶血性链球菌有明显的抑菌作用。与空白组比较,模型组的子宫肿胀度升高,子宫系数降低(P<0.05),提示模型建立成功;与模型组比较,各给药组的子宫肿胀度降低(P<0.05),尤以中药组高、中剂量组及人胎盘组织液组改善显著。与模型组比较,各给药组大鼠子宫组织中的ICAM-1蛋白表达水平降低,caspase-8蛋白表达水平升高(P<0.05),提示给药治疗有效;空白组与中药高剂量组、中药中剂量组及人胎盘组织液组的ICAM-1、caspase-8蛋白表达水平比较,差异无统计学意义(P>0.05),说明以上三组疗效相当。结论高氏盆炎方二号方具有抑菌的作用,能够有效改善疾病症状、控制宫腔粘连的发展,修复受损的子宫组织形态及结构,其治疗机制可能与ICAM-1表达下调、caspase-8表达上调相关,可促进炎症细胞凋亡,增强机体抵抗力。 展开更多

血清ESM-1、sICAM-1水平对帕金森病患者发生认知障碍的预测价值

目的:分析血清内皮细胞特异性分子-1(ESM-1)、可溶性细胞间黏附分子-1(sICAM-1)水平对帕金森病(PD)患者发生认知障碍的预测价值。方法:选取2020年8月至2022年12月该院收治的93例PD患者进行前瞻性研究,根据蒙特利尔认知评估量表(MoCA)评... 目的:分析血清内皮细胞特异性分子-1(ESM-1)、可溶性细胞间黏附分子-1(sICAM-1)水平对帕金森病(PD)患者发生认知障碍的预测价值。方法:选取2020年8月至2022年12月该院收治的93例PD患者进行前瞻性研究,根据蒙特利尔认知评估量表(MoCA)评分将其分为认知正常组(n=50)和认知障碍组(n=43),另选取同期50名健康体检者,设为对照组。比较三组ESM-1、sICAM-1水平及MoCA评分,采用Pearson相关性分析血清ESM-1、sICAM-1水平与认知障碍的相关性,并绘制受试者工作特征(ROC)曲线分析血清ESM-1、sICAM-1单项及联合检测预测PD患者发生认知障碍的价值。结果:认知障碍组、认知正常组血清ESM-1水平、MoCA评分均低于对照组,且认知障碍组低于认知正常组,差异有统计学意义(P<0.05);认知障碍组、认知正常组血清sICAM-1水平均高于对照组,且认知障碍组高于认知正常组,差异有统计学意义(P<0.05);Pearson相关性分析结果显示,血清ESM-1水平与认知功能呈正相关(r>0,P<0.05);血清sICAM-1水平与认知功能呈负相关(r<0,P<0.05);ROC曲线结果显示,血清ESM-1、sICAM-1水平单项及联合预测PD患者发生认知障碍的曲线下面积分别为0.643、0.605、0.911,其中联合检测预测价值最高。结论:血清ESM-1水平与认知功能呈正相关,血清sICAM-1水平与认知功能呈负相关,且血清ESM-1、sICAM-1水平联合检测预测PD患者发生认知障碍的价值高于两者单项检测预测价值。 展开更多

BACKGROUND: Traditional Chinese medicine is a potent agent in the management of clinical and experimental acute pancreatitis (AP), but the molecular mechanism of its the- rapeutic action is unclear. Numerous experimental and clinical studies have shown that platelet endothelial cell ad- hesion molecule-1 (PECAM-1) is pivotal to leukocyte re- cruitment, which results in microcirculatory injury during inflammation, but its role in acute pancreatitis is poorly un- derstood. We investigated the effects of a compound of tra- ditional Chinese medicine pancreatitis-1 (TCMP-1) on the changes of platelet endothelial cell adhesion molecule-1 (PECAM-1) expression on polymorphonuclear leukocytes (PMNs) in acute edematous pancreatitis (AEP). METHODS: The model of acute pancreatitis was estab- lished by subcutaneous injection of caerulein, and TCMP-1 treated groups were given TCMP-1 by catheterization from mouth to stomach (20 ml/kg) immediately after first time subcutaneous injection of caerulein. The changes of expres- sion of PECAM-1 on leukocytes from the blood of the splenic vein and inferior vena cava were determined by flow cytometry. RESULTS: In the AEP group, expression of PECAM-1 on PMNs was not significantly different between pancreatic microcirculation and systemic circulation at AEP2h and AEP4h time point. Then from AEP4h time point to AEP8h time point, expression of PECAM-1 was up-regulated in systemic circulation while it was down-regulated in pancre- atic microcirculation and was significantly different be- tween pancreatic microcirculation and systemic circulation at AEP8h time point (P<0.05). In the TCMP-1 treated group, compared with the AEP group, expression of PE-CAM-1 on PMNs decreased in different levels between pan- creatic microcirculation and systemic circulation and was of significant difference at AEP8h time point (P <0.05). CONCLUSION: Inhibition of PECAM-1 expression on PMNs may prevent PMNs from transmigration through the endo- thelium and may be one of the treatment mechanisms of TCMP-1 decoction on AEP. 展开更多

BACKGROUND:Platelet endothelial cell adhesion molecule-1(PECAM-1),also known as CD31,is mainly distributed in vascular endothelial cells.Studies have shown that PECAM-1 is a very significant indicator of angiogenesis,and has been used as an indicator for vascular endothelial cells.The present study aimed to explore the relationship between the expression of PECAM-1 and the degree of acute lung injury(ALI) and fibrosis in paraquat(PQ) induced lung injury in rabbits.METHODS:Thirty-six adult New Zealand rabbits were randomly divided into three groups(12rabbits in each group) according to PQ dosage:8 mg/kg(group A),16 mg/kg(group B),and 32 mg/kg(group C).After PQ infusion,the rabbits were monitored for 7 days and then euthanized.The lungs were removed for histological evaluation.Masson staining was used to determine the degree of lung fibrosis(LF),and semi-quantitative immune-histochemistry analysis to determine the expression of PECAM-1.Pearson's product-moment correlation analysis was performed to evaluate the relationship between the expression of PECAM-1 and the extent of lung injuries expressed by ALI score and degree of LF.RESULTS:Rabbits in the three groups showed apparent poisoning.The rabbits survived longer in group A than in groups B and C(6.47±0.99 days vs.6.09±1.04 days vs.4.77±2.04 days)(P<0.05).ALI score was lower in group A than in groups B and C(8.33±1.03 vs.9.83±1.17 vs.11.50±1.38)(P<0.05),and there was statistically significant difference between group B and group C(P=0.03).LF was slighter in group A than in groups B and C(31.09%±2.05%vs.34.37%±1.62%vs.36.54%±0.44%)(P<0.05),and there was statistically significant difference between group B and group C(P=0.026).The PEACAM-1 expression was higher in group A than in groups B and C(20.31%±0.70%vs.19.34%±0.68%vs.18.37%±0.46%)(P<0.05),and there was statistically significant difference between group B and group C(P=0.017).Pearson's correlation analysis showed that the expression of PECAM-1 was negatively correlated to both ALI score(Coe=-0.732,P=0.001)and degree of LF(Coe=-0.779,P<0.001).CONCLUSIONS:The PECAM-1 expression significantly decreases in New Zealand rabbits after PQ poisoning,and the decrease is dose-dependent.The PECAM-1 expression is negatively correlated with ALI score and LF,showing a significant role in the development of lung injuries induced by PQ. 展开更多

目的探讨血清心脏肌球蛋白结合蛋白C(cMyBP-C)、细胞间黏附分子-1(ICAM-1)、Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶(ADAMTS-1)水平与急性心肌梗死(AMI)患者预后的相关性。方法选取2021年4月至2023年4月于郑州市惠济区人民医院... 目的探讨血清心脏肌球蛋白结合蛋白C(cMyBP-C)、细胞间黏附分子-1(ICAM-1)、Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶(ADAMTS-1)水平与急性心肌梗死(AMI)患者预后的相关性。方法选取2021年4月至2023年4月于郑州市惠济区人民医院行经皮冠状动脉介入(PCI)术治疗的142例AMI患者作为研究对象,根据入院时Gensini积分将患者分为轻度狭窄组(49例)、中度狭窄组(55例)、重度狭窄组(38例)。比较三组术前血清cMyBP-C、ICAM-1、ADAMTS-1水平,比较不同预后患者(术前、术后7 d、术后14 d)血清cMyBP-C、ICAM-1、ADAMTS-1水平,采用Pearson分析相关性,采用绘制受试者工作特征(ROC)曲线评估预测价值,分析各指标不同水平患者预后不良的危险度。结果三组术前血清cMyBP-C、ICAM-1、ADAMTS-1水平比较:重度狭窄组>中度狭窄组>轻度狭窄组(P<0.05);术后7 d、术后14 d预后不良患者血清cMyBP-C、ICAM-1、ADAMTS-1水平高于预后良好患者(P<0.05);预后良好患者术后血清cMyBP-C、ICAM-1、ADAMTS-1水平呈下降趋势(P<0.05);术前血清cMyBP-C、ICAM-1、ADAMTS-1水平与Gensini积分均呈正相关(P<0.05);术后14 d联合预测AMI患者PCI术后预后不良AUC为0.813;术后14 d血清cMyBP-C、ICAM-1、ADAMTS-1高水平患者PCI术后预后不良的危险度是低水平的4.410、5.217、3.638倍(P<0.05)。结论血清cMyBP-C、ICAM-1、ADAMTS-1水平与AMI患者PCI术后转归情况密切相关,临床可根据各指标水平变化给予针对性干预,进而改善患者预后。 展开更多

The expression of intercellular adhesion molecule-1 (ICAM-1) in the pathogenesis of heroin-induced acute lung injury (ALI) in rats was investigated. The model of ALI was established by intravenous injection of heroin into tail vein in rats. Thirty-six rats were randomly divided into heroin-treated groups (1 h, 2 h, 4 h, 6 h and 24 h) and normal control group. Changes in histopathologic morphology and biological markers of ALI were measured. The expression of ICAM-1 in lung tissue was detected by using immunohistochemistry and RT-PCR. The results showed that the W/D ratio and protein contents in BALF of the heroin-treated groups were significantly higher than that of the control group (P<0.01). The histopathological changes in the lung tissue were more obvious in heroin-treated groups. The ICAM-1 protein and mRNA expression in the lung tissue of heroin-treated groups were significantly increased as compared with that of the control group (P<0.01), and correlated with the ALI parameters in a time-dependent manner. Increasing of ICAM-1 expression was involved in the formation of heroin-induced lung injury. Furthermore, the level of expression was positively correlated with the severity of lung injury. 展开更多

目的:探讨308 nm准分子激光联合复方甘草酸苷片对白癜风患者T细胞亚群及血清细胞间黏附分子-1(ICAM-1)、超氧化物歧化酶(SOD)的影响。方法:选取2021年1月至2022年3月在我院诊治的白癜风患者106例,采用随机数表法分为研究组和对照组两组... 目的:探讨308 nm准分子激光联合复方甘草酸苷片对白癜风患者T细胞亚群及血清细胞间黏附分子-1(ICAM-1)、超氧化物歧化酶(SOD)的影响。方法:选取2021年1月至2022年3月在我院诊治的白癜风患者106例,采用随机数表法分为研究组和对照组两组,每组各53例。对照组患者给予308 nm准分子激光治疗,观察组患者在对照组的基础上给予复方甘草酸苷片治疗。比较两组患者治疗前后的T细胞亚群,血清ICAM-1、SOD,以及临床治疗疗效、不良反应发生情况。结果:治疗后两组患者CD3^(+)、CD4^(+)、CD4^(+)/CD8^(+)较治疗前均升高(P<0.05),且研究组高于对照组(P<0.05)。治疗后两组患者血清ICAM-1水平较治疗前均降低(P<0.05),血清SOD水平较治疗前均升高(P<0.05),且研究组患者血清ICAM-1水平低于对照组(P<0.05),血清SOD高于对照组(P<0.05)。研究组患者治疗总有效率92.45%高于对照组的75.47%(P<0.05)。研究组患者不良反应发生率13.21%高于对照组的9.43%,但差异无统计学意义(P>0.05)。结论:308 nm准分子激光联合复方甘草酸苷片治疗对白癜风,可有效增强机体免疫功能,降低血清ICAM-1,升高血清SOD,临床治疗疗效好,且安全性高。 展开更多

目的 探讨SKAP1通过调控细胞间黏附分子-1(ICAM-1)对宫颈癌模型小鼠肿瘤生长、转移及免疫调节的机制。方法 培养人宫颈癌细胞He La,构建SKAP1-mi RNA慢病毒干扰质粒和阴性对照Neg-mi RNA空白慢病毒干扰质粒,分别感染He La细胞,随机分为... 目的 探讨SKAP1通过调控细胞间黏附分子-1(ICAM-1)对宫颈癌模型小鼠肿瘤生长、转移及免疫调节的机制。方法 培养人宫颈癌细胞He La,构建SKAP1-mi RNA慢病毒干扰质粒和阴性对照Neg-mi RNA空白慢病毒干扰质粒,分别感染He La细胞,随机分为对照组(细胞未做处理)、空质粒组(Neg-mi RNA组)和SKAP1基因沉默组(SKAP1-mi RNA组),每组10只。将稳定表达的3组细胞配制成单细胞悬液分别接种于对应组BALB/c裸鼠右前腋窝中部外侧皮下,记录各组肿瘤生长情况,30 d后采用活体荧光成像技术检测肿瘤转移情况。处死裸鼠,取肿瘤组织,采用RealTime PCR法检测各组裸鼠肿瘤组织SKAP1、ICAM-1 m RNA相对表达量,免疫组化法检测SKAP1、ICAM-1蛋白表达情况,Western blot法检测SKAP1、ICAM蛋白相对表达量,流式细胞术检测免疫细胞CD3+、CD4+、CD8+水平,ELISA法检测Th1和Th2细胞因子水平。结果 细胞接种后饲养期间内,对照组和空质粒组裸鼠各死亡1只,SKAP1基因沉默组死亡2只。各组瘤体体积均随时间延长而增大(P<0.05);与对照组、空质粒组比较,SKAP1基因沉默组各时间段瘤体体积减小、肿瘤细胞转移面积减小、荧光信号强度降低、肿瘤组织SKAP1和ICAM-1 m RNA及其蛋白相对表达量均降低,差异有统计学意义(P<0.05);与对照组、空质粒组比较,SKAP1基因沉默组肿瘤组织免疫细胞CD3+、CD4+、CD4+/CD8+及Th1细胞因子IL-2、IFN-γ水平均升高,CD8+及Th2细胞因子IL-4、IL-10水平均降低(P<0.05)。结论 沉默SKAP1基因可以抑制He La细胞宫颈癌裸鼠移植瘤的生长及转移,逆转小鼠Th1向Th2漂移,恢复Th1表达优势,这一作用可能是通过下调ICAM-1表达实现的。 展开更多

目的 探讨血清可溶性细胞间黏附分子-1(sICAM-1)、可溶性血管细胞黏附分子-1(sVCAM-1)、内皮素-1(ET-1)水平对过敏性紫癜(HSP)的临床意义。方法 回顾性选取2020年5月至2022年5月泉州市儿童医院收治的85例HSP患儿,检测入院时血清sICAM-1... 目的 探讨血清可溶性细胞间黏附分子-1(sICAM-1)、可溶性血管细胞黏附分子-1(sVCAM-1)、内皮素-1(ET-1)水平对过敏性紫癜(HSP)的临床意义。方法 回顾性选取2020年5月至2022年5月泉州市儿童医院收治的85例HSP患儿,检测入院时血清sICAM-1、sVCAM-1、ET-1水平。根据有无肾脏受累将其分为紫癜性肾炎(HSPN)组(n=28)与非HSPN(n HSPN)组(n=57),根据治疗后有无复发将其分为无复发组(n=69)与复发组(n=16)。另选取同期行健康体检的30名儿童作为健康对照组。结果 与健康对照比较,n HSPN组及HSPN组血清s ICAM-1、s VCAM-1、ET-1水平均显著升高,差异有统计学意义(P <0.05);与n HSPN组比较,HSPN组血清s ICAM-1、s VCAM-1、ET-1水平亦显著升高,差异有统计学意义(P <0.05)。相关性分析显示,血清sICAM-1与sVCAM-1呈正相关(r=0.682,P <0.05),sICAM-1与ET-1呈正相关(r=0.536,P<0.05),sVCAM-1与ET-1呈正相关(r=0.614,P<0.05)。与无复发组比较,复发组血清sICAM-1显著升高,差异有统计学意义(P <0.05);两组sVCAM-1、ET-1水平比较,差异无统计学意义(P> 0.05)。受试者工作特征(ROC)曲线分析显示,血清sICAM-1、sVCAM-1、ET-1及三者联合预测HSPN的曲线下面积(AUC)分别为0.745、0.839、0.668、0.904,显著高于机会参考线下面积,差异有统计学意义(P <0.05),但预测疾病复发的AUC分别为0.533、0.512、0.504、0.569,与机会参考线下面积比较,差异均无统计学意义(P> 0.05)。结论 sICAM-1、sVCAM-1、ET-1参与HSP的发生发展,并且可作为预测HSPN的潜能标志物。 展开更多

目的初步探讨肝衰竭患者外周血单个核细胞人瑞粒酶逆转录酶(hTERT)mRNA及血清血管生成素-2(Ang-2)和细胞间黏附分子-1(ICAM-1)水平变化的临床意义。方法2019年1月~2021年5月我院诊治的74例肝衰竭患者【亚急性肝衰竭(SALF)13例,慢加急性... 目的初步探讨肝衰竭患者外周血单个核细胞人瑞粒酶逆转录酶(hTERT)mRNA及血清血管生成素-2(Ang-2)和细胞间黏附分子-1(ICAM-1)水平变化的临床意义。方法2019年1月~2021年5月我院诊治的74例肝衰竭患者【亚急性肝衰竭(SALF)13例,慢加急性肝衰竭(ACLF)39例和慢性肝衰竭(CLF)22例】和同期健康体检者30例,分离并检测外周血单个核细胞(PBMC)hTERT mRNA水平,采用ELISA法检测血清Ang-2和ICAM-1水平。结果肝衰竭组PBMC hTERT mRNA、血清Ang-2和ICAM-1水平分别为(0.9±0.3)、(1344.6±55.3)ng/L和(540.2±22.4)ng/ml,显著高于健康人组【分别为(0.4±0.1)、(1062.5±36.2)ng/L和(167.3±15.6)ng/ml,P<0.05】;SALF患者PBMC hTERT mRNA、血清Ang-2和ICAM-1水平分别为(1.5±0.6)、(1507.8±38.2)ng/L和(647.3±26.2)ng/ml,显著高于ACLF患者【分别为(1.2±0.4)、(1398.6±35.8)ng/L和(582.7±24.1)ng/ml,P<0.05】或CLF患者【分别为(0.7±0.2)、(1280.5±46.3)ng/L和(468.9±20.3)ng/m,P<0.05】;经过1~3 m治疗,SALF组死亡3例,ACLF组死亡5例,CLF组死亡4例。结论肝衰竭患者PBMCs hTERT mRNA及血清Ang-2和ICAM-1水平显著升高,它们可能为判断肝损伤程度提供客观依据,值得进一步研究。 展开更多

血清sICAM-1和TGF-β1水平、RBC-ICR率及淋巴细胞比例与剖宫产术后发生产褥感染的关系

目的 分析血清可溶性细胞间黏附分子1(sICAM-1)和转化生长因子β1(TGF-β1)水平、红细胞免疫复合物花环(RBC-ICR)率、淋巴细胞比例与剖宫产术后发生产褥感染的关系。方法 选取108例剖宫产术后发生产褥感染的产妇作为研究组,依据感染程... 目的 分析血清可溶性细胞间黏附分子1(sICAM-1)和转化生长因子β1(TGF-β1)水平、红细胞免疫复合物花环(RBC-ICR)率、淋巴细胞比例与剖宫产术后发生产褥感染的关系。方法 选取108例剖宫产术后发生产褥感染的产妇作为研究组,依据感染程度将其分为轻度组(41例)、中度组(35例)与重度组(32例)。另选取剖宫产术后未发生产褥感染的60例产妇作为对照组。比较研究组与对照组之间,以及轻度组、中度组与重度组之间产前血清sICAM-1和TGF-β1水平、 RBC-ICR率、淋巴细胞比例。采用多因素Logistic回归模型分析影响剖宫产术后发生产褥感染的因素。结果 研究组的产前血清sICAM-1和TGF-β1水平、 RBC-ICR率、淋巴细胞比例高于对照组(P<0.05)。轻度组、中度组、重度组的产前血清sICAM-1和TGF-β1水平、 RBC-ICR率及淋巴细胞比例依次升高(P<0.05)。多因素Logistic回归分析结果显示,校正混杂因素后,产前血清TGF-β1水平、RBC-ICR率、淋巴细胞比例是剖宫产术后发生产褥感染的独立影响因素(P<0.05)。结论 剖宫产术后发生产褥感染的产妇产前血清sICAM-1和TGF-β1水平、 RBC-ICR率、淋巴细胞比例升高,且四项指标均随着感染程度的加重而升高。产前血清TGF-β1水平、RBC-ICR率、淋巴细胞比例升高的产妇,其剖宫产术后发生产褥感染的风险增加。 展开更多
