慢性乙型肝炎患者HBV DNA载量及血清IL-2R、GP73、miR-21与HBVM表达模式、肝组织病理学改变的相关性

目的探讨慢性乙型肝炎患者乙型肝炎病毒(HBV)DNA载量及血清白细胞介素-2受体(IL-2R)、高尔基体蛋白73(GP73)、微小RNA-21(miR-21)与乙型肝炎病毒血清标志物(HBVM)表达模式、肝组织病理学改变的相关性。方法回顾性选取2020年1月至2022年1... 目的探讨慢性乙型肝炎患者乙型肝炎病毒(HBV)DNA载量及血清白细胞介素-2受体(IL-2R)、高尔基体蛋白73(GP73)、微小RNA-21(miR-21)与乙型肝炎病毒血清标志物(HBVM)表达模式、肝组织病理学改变的相关性。方法回顾性选取2020年1月至2022年12月四川绵阳四0四医院(绵阳市第一人民医院)收治的106例慢性乙型肝炎患者作为观察组,另纳入本院125名同期健康体检者为对照组。观察组根据肝脏炎症程度分为炎症较轻组(G_(0)~G_(2)期,n=61)和炎症较重组(G_(3)~G_(4)期,n=45);根据肝纤维化程度分为纤维化较轻组(S_(0)~S_(2)期,n=80)和纤维化较重组(S_(3)~S_(4)期,n=26);根据外周血中是否产生乙肝表面抗体(抗-HBs)分为抗-HBs阳性组(n=77)和抗-HBs阴性组(n=29)。比较观察组与对照组血清IL-2R、GP73、miR-21水平,比较不同肝脏炎症程度、不同肝脏纤维化程度、不同HBVM表达模式的慢性乙型肝炎患者HBV DNA载量、血清IL-2R、GP73、miR-21水平,并采用Spearman相关性分析分析HBV DNA载量、血清IL-2R、GP73、miR-21与肝组织病理学改变的相关性。结果观察组血清IL-2R、GP73、miR-21水平分别为(559.65±46.65)U/mL、(149.87±31.65)ng/mL、1.98±0.65,均高于对照组[(272.54±28.65)U/mL、(48.76±9.65)ng/mL、1.23±0.36],差异均有统计学意义(P<0.05)。炎症较重组HBV DNA载量、血清IL-2R、GP73、miR-21水平分别为(5.98±0.73)loads、(703.54±57.54)U/mL、(243.65±38.65)ng/mL、1.99±0.54,均高于炎症较轻组[(4.65±0.68)loads、(450.65±36.65)U/mL、(80.43±20.34)ng/mL、1.54±0.45],差异均有统计学意义(P<0.05)。纤维化较重组HBV DNA载量、血清IL-2R、GP73、miR-21水平分别为(6.03±0.98)loads、(675.65±58.04)U/mL、(189.65±41.65)ng/mL、2.34±0.34,均高于纤维化较轻组[(4.48±0.79)loads、(522.65±31.54)U/mL、(135.76±26.54)ng/mL、1.31±0.28],差异均有统计学意义(P<0.05)。抗-HBs阳性组和抗-HBs阴性组HBV DNA载量、血清IL-2R、GP73、miR-21水平比较,差异均无统计学意义(P>0.05)。HBV DNA载量、血清IL-2R、GP73、miR-21均与肝脏炎症程度呈正比(r=0.546、0.498、0.657、0.508,P<0.05),与肝脏纤维化程度呈正比(r=0.487、0.506、0.673、0.703,P<0.05)。结论慢性乙型肝炎患者血清中IL-2R、GP73、miR-21表达较高,且患者HBV DNA载量、血清IL-2R、GP73、miR-21水平与肝脏炎症及纤维化程度呈正比,但其与HBVM表达模式无明显相关性。 展开更多

目的探讨miR-21、雌激素(E2)及其受体(ER)在子宫腺肌病发病中的具体机制。方法采用qRT-PCR检测miR-21在子宫腺肌病组织中水平,以宫颈病变为对照组。体外原代培养子宫腺肌病细胞,qRT-PCR检测E2激活组、ICI82780(ER抑制剂)+E2激活组、E2... 目的探讨miR-21、雌激素(E2)及其受体(ER)在子宫腺肌病发病中的具体机制。方法采用qRT-PCR检测miR-21在子宫腺肌病组织中水平,以宫颈病变为对照组。体外原代培养子宫腺肌病细胞,qRT-PCR检测E2激活组、ICI82780(ER抑制剂)+E2激活组、E2剥夺组、E2剥夺+ER抑制组及对照组各组miR-21水平。miR-21mimic、miR-21inhibitor转染子宫腺肌病细胞,分别过表达和干扰miR-21,从正反两方面验证miR-21对子宫腺肌病细胞增殖、凋亡、迁移及细胞自噬等超微结构的影响。结果miR-21在子宫腺肌病病灶组织中的表达水平明显高于正常子宫肌层中表达水平(P<0.05),与其余各组的差异无统计学意义(P>0.05);E2激活状态下,E2激活组miR-21表达水平较ER抑制+E2激活组及对照组升高(P<0.05);E2剥夺状态下,E2剥夺+ER抑制组miR-21表达水平较E2剥夺组及对照组降低(P<0.05);干扰miR-21可以抑制子宫腺肌病细胞增殖和迁移,使线粒体内质网不同程度扩张、溶酶体增多、自噬小体出现,促进细胞的凋亡;而过表达miR-21则发挥相反的作用。结论MiR-21在子宫腺肌病细胞中通过改变细胞超微结构而发挥促增殖、迁移及抗凋亡的作用,可能与疾病的早期形成有关;ER除与E2结合调控miR-21外还可通过其他途径调控miR-21,这可能是子宫腺肌病的发病机制之一,其中ER对miR-21的调控作用大于E2。 展开更多

目的 探讨股腘型下肢动脉硬化闭塞症(LEASO)患者血清微小核糖核酸(miR)-21、miR-126水平与高迁移率族蛋白B1(HMGB1)/Toll样受体4(TLR4)信号通路活性和术后复发的关系。方法 选取行介入手术的股腘型LEASO患者120例,根据术后是否复发将股... 目的 探讨股腘型下肢动脉硬化闭塞症(LEASO)患者血清微小核糖核酸(miR)-21、miR-126水平与高迁移率族蛋白B1(HMGB1)/Toll样受体4(TLR4)信号通路活性和术后复发的关系。方法 选取行介入手术的股腘型LEASO患者120例,根据术后是否复发将股腘型LEASO患者分为复发组和未复发组。RF-qPCR剂检测血清miR-21、miR-126表达和外周血单个核细胞HMGB1 mRNA、TLR4 mRNA表达。采用Pearson相关性分析法分析股腘型LEASO患者血清miR-21、miR-126与外周血单个核细胞HMGB1 mRNA、TLR4 mRNA表达的相关性。多因素Logistic回归分析股腘型LEASO患者介入术后复发的影响因素。结果 120例股腘型LEASO患者随访2年,术后复发率为34.17%。与未复发组比较,复发组血清miR-21和外周血单个核细胞HMGB1 mRNA、TLR4 mRNA表达升高,血清miR-126表达降低(P均<0.05)。Pearson相关性分析显示,股腘型LEASO患者血清miR-21与外周血单个核细胞HMGB1 mRNA、TLR4 mRNA表达呈正相关(r分别为0.660、0.649,P均<0.05),miR-126与外周血单个核细胞HMGB1 mRNA、TLR4 mRNA表达呈负相关(r分别为-0.632、-0.641,P均<0.05),外周血单个核细胞HMGB1mRNA与TLR4 mRNA表达呈正相关(r=0.742,P<0.05)。多因素Logistic回归分析显示,高血压、糖尿病和miR-21、HMGB1 mRNA、TLR4 mRNA升高为股腘型LEASO患者介入术后复发的独立危险因素,miR-126升高为独立保护因素(P均<0.05)。结论 股腘型LEASO患者血清miR-21高表达和miR-126低表达与HMGB1/TLR4信号通路激活有关,是股腘型LEASO患者术后复发的独立影响因素。 展开更多

阿帕替尼联合埃克替尼治疗EGFR基因21号外显子L858R突变型NSCLC的临床观察

目的探究对表皮生长因子受体(EGFR)基因21号外显子L858R突变型非小细胞肺癌(NSCLC)采用阿帕替尼联合埃克替尼治疗的临床效果。方法将EGFR基因21号外显子L858R突变型NSCLC患者101例按随机数表法分为观察组(n=51)与对照组(n=50)。观察组... 目的探究对表皮生长因子受体(EGFR)基因21号外显子L858R突变型非小细胞肺癌(NSCLC)采用阿帕替尼联合埃克替尼治疗的临床效果。方法将EGFR基因21号外显子L858R突变型NSCLC患者101例按随机数表法分为观察组(n=51)与对照组(n=50)。观察组采用阿帕替尼联合埃克替尼治疗,对照组仅采用埃克替尼治疗,治疗时间为56 d,2个周期。比较2组近期疗效、不良反应,并采用K-M生存分析曲线图分析2组远期疗效。结果观察组与对照组的客观缓解率(ORR)(74.51%vs 74.00%)与疾病控制率(DCR)(92.16%vs 94.00%)差异均无统计学意义(P>0.05);治疗期间2组未出现2级以上不良反应,观察组高血压和蛋白尿发生率分别为52.94%、50.98%,分别高于对照组的16.00%、10.00%,差异有统计学意义(P<0.05)。观察组生存率显著高于对照组(69.3%vs 43.1%),差异有统计学意义(P<0.05)。结论EGFR基因21号外显子L858R突变型NSCLC采用阿帕替尼联合埃克替尼治疗,可延长患者的无进展生存时间,不良反应的发生可接受。 展开更多

目的探究基于Toll样受体4(Toll-like receptor 4,TLR4)/核转录因子-κB(nuclear factor kappa B,NF-κB)信号通路沉默微小RNA-21(MicroRNA-21,miR-21)对糖尿病肾病大鼠的干预效果。方法选取45只大鼠,10只作为空白组,其余35只建立糖尿病... 目的探究基于Toll样受体4(Toll-like receptor 4,TLR4)/核转录因子-κB(nuclear factor kappa B,NF-κB)信号通路沉默微小RNA-21(MicroRNA-21,miR-21)对糖尿病肾病大鼠的干预效果。方法选取45只大鼠,10只作为空白组,其余35只建立糖尿病肾病模型,最终有30只建模成功,将最终建模成功的30只大鼠分为模型组、上调组和下调组,每组10只。做miR-21转染,将10μl miR-21过表达慢病毒悬液、miR-21沉默慢病毒悬液分别注射于上调组、下调组大鼠胃组织中,空白组、模型组注射同剂量的蒸馏水灌胃,注射24 h后观察大鼠变化。结果与空白相比,模型组、上调组、下调组白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1(interleukin-1,IL-1)、C-C基序趋化因子2(CC chemokine ligand 2,CCL2)、内皮素-1(endothelin 1,ET-1)、结缔组织生长因子(connective tissue growth factor,CTGF)、转化生长因子-β1(transforming the growth factor-β1,TGF-β1)水平、TLR4、NF-κB表达量上升,一氧化氮(nitric oxide,NO)水平下降(P<0.05);与模型组相比,上调组IL-6、IL-1、CCL2、ET-1、CTGF、TGF-β1水平、TLR4、NF-κB表达量上升(P<0.05),NO水平下降(P<0.05),下调组IL-6、IL-1、CCL2、ET-1、CTGF、TGF-β1水平、TLR4、NF-κB表达量下降(P<0.05),NO水平上升(P<0.05);与上调组相比,下调组IL-6、IL-1、CCL2、ET-1、CTGF、TGF-β1水平、TLR4、NF-κB表达量下降(P<0.05),NO水平上升(P<0.05)。结论沉默miR-21可缓解糖尿病肾病大鼠炎性反应、肾纤维化,改善内皮功能,其机制可能与TLR4/NF-κB信号通路被抑制有关。 展开更多

BACKGROUND It is challenging to distinguish intestinal tuberculosis from Crohn’s disease due to dynamic changes in epidemiology and similar clinical characteristics. Recent studies have shown that polymorphisms in genes involved in the interleukin (IL)- 23/IL-17 axis may affect intestinal mucosal immunity by affecting the differentiation of Th17 cells. AIM To investigate the specific single-nucleotide polymorphisms (SNPs) in genes involved in the IL-23/IL-17 axis and possible pathways that affect susceptibility to intestinal tuberculosis and Crohn's disease. METHODS We analysed 133 patients with intestinal tuberculosis, 128 with Crohn’s disease, and 500 normal controls. DNA was extracted from paraffin-embedded specimens or whole blood. Four SNPs in the IL23/Th17 axis (IL22 rs2227473, IL1β rs1143627, TGFβ rs4803455, and IL17 rs8193036) were genotyped with TaqMan assays. The transcriptional activity levels of different genotypes of rs2227473 were detected by dual luciferase reporter gene assay. The expression of IL-22R1 in different intestinal diseases was detected by immunohistochemistry. RESULTS The A allele frequency of rs2227473 (P = 0.030, odds ratio = 0.60, 95% confidence interval: 0.37-0.95) showed an abnormal distribution between intestinal tuberculosis and healthy controls. The presence of the A allele was associated with a higher IL-22 transcriptional activity (P < 0.05). In addition, IL-22R1 was expressed in intestinal lymphoid tissues, especially under conditions of intestinal tuberculosis, and highly expressed in macrophage-derived Langhans giant cells. The results of immunohistochemistry showed that the expression of IL-22R1 in patients with Crohn's disease and intestinal tuberculosis was significantly higher than that in patients with intestinal polyps and colon cancer (P < 0.01). CONCLUSION High IL-22 expression seems to be a protective factor for intestinal tuberculosis. IL-22R1 is expressed in Langhans giant cells, suggesting that the IL-22/IL-22R1 system links adaptive and innate immunity. 展开更多

In the gut of patients with Crohn's disease and patients with ulcerative colitis,the major forms of inflammatory bowel diseases(IBD) in humans,the tissue-damaging immune response is mediated by an active cross-talk between immune and non-immune cells.Accumulating evidence indicates also that cytokines produced by these cells play a major role in initiating and shaping this pathologic process.One such cytokine seems to be interleukin(IL)-21,a member of the common γ-chainreceptor family.IL-21 is produced in excess in the in-flamed intestine of patients with IBD mostly by activated CD4+ T helper cells co-expressing interferon-γ and follicular T helper cells.Moreover,both in vitro and in vivo studies indicate that excessive IL-21 production leads to the activation of multiple signaling pathways that expand and sustain the ongoing mucosal inflammation.In this article,we review the available data supporting the pathogenic role of IL-21 in IBD. 展开更多

Methods: Thesoluble interleukin-2 receptor (sIL-2R) level in serum was analyzed by ELISA and Na^+ -K^+ -exchanging ATPase activi-ty in semen by phosphorus (Pi) assay. Results: The slL-2R level in serum was significantly higher and the Na^+ -K^+ -exchanging ATPase activity in semen significantly lower in AsAb positive infertile men when compared with thecontrols. Conclusion: The AsAb titer varies with the slL-2R level in serum. A decrease in Na^+ -K^+ -exchangingATPase activity in semen may play a role in male infertility caused by AsAb. 展开更多

AIM:To elucidate the effect of rapamycin on regulating the production of interleukin(IL)-1β in Aspergillus fumigatus(A.fumigatus)-induced keratitis and to verify whether the expression of IL-1β in A.fumigatus keratitis is associated with the mammalian target of rapamycin(mT OR)/Toll-like receptor 4(TLR4) signaling pathway.METHODS:Fungal keratitis mouse models of susceptible C57 BL/6 mice were established using A.fumigatus.The mice were subsequently treated with rapamycin.The protein levels of p-mT OR,TLR4,and IL-1β in normal and infected corneal tissue were measured by Western blot.The TLR4 and IL-1β m RNA levels were determined by real-time polymerase chain reaction(PCR).RESULTS:In C57 BL/6 mice,rapamycin treatment decreased the clinical scores and production of the pro-inflammatory cytokine,IL-1β.The expression of TLR4,stimulated by A.fumigatus,was reduced as well when the mT OR signaling pathway was suppressed by rapamycin.CONCLUSION:Rapamycin is beneficial for the outcome of fungal keratitis and has an inhibitory effect expression of the inflammatory cytokine IL-1β.The inhibitory effect on IL-1β expression can be associated with the mT OR/TLR4 signaling pathway in A.fumigatus infection in mice. 展开更多

AIM:To investigate the effect of Tenascin C(TNC)on the expression of pro-inflammatory cytokines and matrixmetalloproteinases in human cardiac myofibroblasts(CMF).METHODS:CMF were isolated and cultured from patients undergoing coronary artery bypass grafting.Cultured cells were treated with either TNC(0.1μmol/L,24 h)or a recombinant protein corresponding to different domains of the TNC protein;fibrinogen-like globe(FBG)and fibronectin typeⅢ-like repeats(TNⅢ5-7)(both 1μmol/L,24 h).The expression of the proinflammatory cytokines;interleukin(IL)-6,IL-1β,TNFαand the matrix metalloproteinases;MMPs(MMP1,2,3,9,10,MT1-MMP)was assessed using real time RT-PCR and western blot analysis.RESULTS:TNC increased both IL-6 and MMP3(P<0.01)mR NA levels in cultured human CMF but had no significant effect on the other markers studied.The increase in IL-6 mR NA expression was mirrored by an increase in protein secretion as assessed by enzymelinked immunosorbant assay(P<0.01).Treating CMF with the recombinant protein FBG increased IL-6mR NA and protein(P<0.01)whereas the recombinant protein TNⅢ5-7 had no effect.Neither FBG nor TNⅢ5-7 had any significant effect on MMP3 expression.The expression of toll-like receptor 4(TLR4)in human CMF was confirmed by real time RT-PCR,western blot and immunohistochemistry.Pre-incubation of cells with TLR4neutralising antisera attenuated the effect of both TNC and FBG on IL-6 mR NA and protein expression.CONCLUSION:TNC up-regulates IL-6 expression in human CMF,an effect mediated through the FBG domain of TNC and via the TLR4 receptor. 展开更多

Interleukin-34(IL-34)is a novel cytokine that plays an important role in innate immunity and inflammatory processes by binding to the colonystimulating factor-1 receptor(CSF-1R).However,information on the function of IL-34 in fish remains limited.In the present study,we identified an IL-34 homolog from mudskippers(Boleophthalmus pectinirostris).In silico analysis showed that the mudskipper IL-34(BpIL-34)was similar to other known IL-34 variants in sequence and structure and was most closely related to an orange-spotted grouper(Epinephelus coioides)homolog.BpIL-34 transcripts were constitutively expressed in various tissues,with the highest level of expression found in the brain.Edwardsiella tarda infection significantly up-regulated the mRNA expression of BpIL-34 in the mudskipper tissues.The recombinant mature BpIL-34 peptide(rBpIL-34)was purified and used to produce anti-rBpIL-34 IgG.Western blot analysis combined with PNGase F digestion revealed that native BpIL-34 in monocytes/macrophages(MOs/MФs)was N-glycosylated.In vitro,rBpIL-34 treatment enhanced the phagocytotic and bactericidal activity of mudskipper MOs/MФs,as well as the mRNA expression of pro-inflammatory cytokines like tumor necrosis factorα(BpTNF-α)and BpIL-1βin these cells.Furthermore,the knockdown of mudskipper CSF-1R1(BpCSF-1R1),but not mudskipper BpCSF-1R2,significantly inhibited the rBpIL-34-mediated enhanced effect on MO/MФfunction.In conclusion,our results indicate that mudskipper BpIL-34 modulates the functions of MOs/MФs via BpCSF-1R1. 展开更多

AIM: To study the efficacy of marrow mesenchymal stem cells (MSCs) transplantation combined with interleukin-1 receptor antagonist (IL-1Ra) for acute liver failure (ALF). METHODS: Chinese experimental miniature swine were randomly divided into four groups (n = 7), and all animals were given D-galactosamine (D-gal) to induce ALF. Group A animals were then injected with 40 mL saline via the portal vein 24 h after D-gal induction;Group B animals were injected with 2 mg/kg IL-1Ra via the ear vein 18 h, 2 d and 4 d after D-gal induction; Group C received approximately 1 × 108 green fluorescence protein (GFP)-labeled MSCs (GFP-MSCs) suspended in 40 mL normal saline via the portal vein 24 h after D-gal induction; Group D animals were injected with 2 mg/kg IL-1Ra via the ear vein 18 h after D-gal induction, MSCs transplantation was then carried out at 24 h after D-gal induction, and finally 2 mg/kg IL-1Ra was injected via the ear vein 1 d and 3 d after surgery as before. Liver function, serum inflammatory parameters and pathological changes were measured and the fate of MSCs was determined.RESULTS: The optimal efficiency of transfection (97%) was achieved at an multiplicity of infection of 80, as observed by fluorescence microscopy and flow cytometry (FCM). Over 90% of GFP-MSCs were identified as CD44+ CD90+ CD45-MSCs by FCM, which indicated that most GFP-MSCs retained MSCs characteristics. Biochemical assays, the levels of serum inflammatory parameters and histological results in Group D all showed a significant improvement in liver injury compared with the other groups (P < 0.05). The number of GFP-MSCs in Group D was also greater than that in Group B, and the long-term cell proliferation rate was also better in Group D than in the other groups.CONCLUSION: MSCs transplantation is useful in ALF, IL-1Ra plays an important role in alleviating the inflammatory condition, and combination therapy with MSCs transplantation and IL-1Ra is a promising treatment for ALF. 展开更多

Objective The current study was designed to find out the effect of Glucosidorum Tripterygii Totorum (GTT) on the serum level of soluble interleukin-2 receptor (sIL-2R) in patient with ankylosing spondylitis (AS). Method 29 patients with active AS were selected to take GTT (1mg per kg) three times a day for one year. After that, its curative effect was evaluated. The serum level of sIL-2R of these patients was measured by sandwich ELISA method and was compared with that of normal subjects. Result The serum level of sIL-2R in active AS patients was obviously higher than that of the non-active AS patients. (P<0.01). The total effective rate of GTT on AS was 89.6%, while clinical relief rate 27.6%, obvious effective rate 44.8%, effective rate 17.2% and non-effective rate 10.4%. The patients’ serum level of sIL-2R after therapy was significantly lower than that before therapy except patients with no effect. (P<0.05). Conclusion GTT has positive curative effect on active AS patients and could cause obvious decrease of the serum level of sIL-2R. The serum level of sIL-2R can be used as an important index of activity of AS and as a guide of therapy. 展开更多

Methods: Forty-four patients with SLE were randomly divided into conventional corticosteroid treated group (control group, n =20) and conventional treatment group with leeching intervention added (leeching group, n =24). Before and after treatment the concentration of plasma endothelin (ET) and soluble interleukin-2 receptor (sIL-2R) were determined. Results: Before treatment the level of plasma ET and sIL-2R in the SLE patients were all higher than those in the normal healthy group, ( P <0.01). But after treatment the level of these in both groups were significantly improved than those of before treatment ( P <0.05), and comparison between these two treated groups showed that the difference between them was significant ( P <0.05). Conclusion: Leeching added to conventional treatment of SLE could be more effective in improving the level of plasma ET and sIL-2R, and ameliorating the impairment of renal tissues. 展开更多

The aim of this study is to examine the ability of interferon-γ (IFN-γ), and tumor necrosis factor-or (TNF-α to induce the production of human interleukin-15 (hIL-15) and IL-15 receptor (IL-15Rα by human umbilical vein endothelial cells (HUVECs). The data are summarized as follows: 1. Northern blot revealed that IL-15 mRNA was up-regulated by IFN-γ and TNF-α 2. Intracellular IL-15 protein was visualized by fluorescence microscopy, whereas the expression of IL-15 on the surface of HUVECs was detected by fluorescence activated cell sorting (FACS), and no detectable IL-15 in the medium was verified by ELISA. 3. IL-15Rα was detected on the surface of HUVECs by FACS after IFN-α and TNF-α stimulation, whereas Western blotting revealed that the elevated expression on surface IL-15Rα was not due to the increased protein expression. The conclusion demonstrated from our results is that IFN-α and TNF-α play an important role in regulating the expression of IL-15 and IL-15Rα on the surface of HUVECs. 展开更多

目的探究血清CC家族趋化因子配体21(CCL21)、CXC趋化因子受体7(CXCR7)水平与阵发性房颤射频消融术后早期复发的相关性。方法选取2018年6月至2021年1月襄阳市中心医院收治的122例接受射频消融术治疗的房颤病人为研究对象,根据射频消融术... 目的探究血清CC家族趋化因子配体21(CCL21)、CXC趋化因子受体7(CXCR7)水平与阵发性房颤射频消融术后早期复发的相关性。方法选取2018年6月至2021年1月襄阳市中心医院收治的122例接受射频消融术治疗的房颤病人为研究对象,根据射频消融术后3个月病人是否出现复发分为早期复发组(19例)和未早期复发组(103例)。采用酶联免疫吸附(ELISA)法检测病人血清中CCL21、CXCR7水平,采用受试者操作特征(ROC)曲线评价血清CCL21、CXCR7预测阵发性房颤射频消融术后早期复发的价值,logistic回归分析影响阵发性房颤射频消融术后早期复发的危险因素。结果与未早期复发组相比,早期复发组病人左心房直径(LAD)、血清超敏C反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)及CCL21水平较高(P<0.05),左心室射血分数(LVEF)及血清CXCR7水平较低(P<0.05);ROC结果显示,血清CCL21、CXCR7水平预测术后早期复发的AUC分别为0.88、0.87,CCL21为138.36 ng/L时,CXCR7为2113.83μg/L,具有最优的预测能力,二者联合预测术后早期复发的AUC为0.94,灵敏度为84.20%,特异度为95.10%。logistic回归分析,结果显示高LAD值、CCL21≥138.36 ng/L、CXCR7≤2113.83μg/L是阵发性房颤射频消融术后早期复发的危险因素(P<0.05),高LVEF值是阵发性房颤射频消融术后早期复发的保护因素(P<0.05)。结论术后早期复发的阵发性房颤病人血清中CCL21水平上升,CXCR7水平下降,对阵发性房颤射频消融术后早期复发具有一定的预测价值。 展开更多

Interleukin-2 and its receptor are of importance in regulating immunity responses. The changes of interleukin-2 (IL-2) and soluble interleukin-2 receptor (IL-2R) during heart valve (s) replacement operation and effects of aprotinin on them were observed. Twenty patients undergoing heart valve (s) replacement were randomly divided into two groups: control group (n=10) and apro- tinin group (n=10). In aprotinin group, 1 000 000 KIU aprotinin was given by vein injection and then 2 000 000 KIU was given as a bolus in prime. Blood samples were collected before CPB, right after CPB and on the 1st, 3rd and 7th postoperative day (POD) for serum IL-2 and sIL-2R determination. Results showed that after CPB, IL-2 was reduced and slL-2R increased. Meanwhile, serum IL-2R was lower in aprotinin group than that of control. It is concluded that the immunity depression after CPB is associated with low level of IL-2 and high level of sIL-2R and aprotinin can ameliorate the situation. 展开更多

Objective: Recent studies have shown that the local expression of soluble interleukin (IL) -1 receptor type Ⅱ (slL-1 R Ⅱ ) in endometrial tissue of women with endometriosis is decreased, and the depression of IL-1 R Ⅱ was more significant in infertile women than that in fertile women with endometriosis. In this research, we investigated the remedial effect of slL-1-R Ⅱ administration on endometriosis in the nude mouse model. Methods: Nineteen nude model mice with endometriosis were randomly divided into three groups: group A was treated by intraperitoneal administration with only slL-1 R Ⅱ for two weeks, group B was similarly treated with only IL- 1, and group C (control) was administered saline. After 2 weeks, the size of the ectopic endometrial lesions was calculated, and the expression of vascular endothelial growth factor (VEGF) and B-cell lymphoma leukemia-2 (Bcl- 2) were detected by immunohistochemistry. The IL-8 and VEGF levels in the peritoneal fluid (PF) and serum were also measured by enzyme-linked immunosorbent assay (ELISA). Results: The mean size of ectopic endometrial lesion did not differ between the three groups (P 〉 0.05). Compared with the control, the expression of VEGF and Bcl-2 was significantly lower in group A, and higher in group B. In the three groups, the levels of IL-8 in the PF and serum were highest in group A, and lowest in group B. Conclusion: slL-1 R Ⅱ may suppresse hyperplasia of ectopic endometriosis, perhaps by reducing the expression of certain cytokines, such as VEGF, IL-8, and Bcl-2, which could provide a new clinical strategy for the treatment of endometriosis. 展开更多
