In order to evaluate the permeability and predict the absorpti...The intestinal permeability of two flavonoid compounds liquiritin (LQ) and isoliquiritin (ILQ) was investigated using the Caco-2 cell monolayer model. In order to evaluate the permeability and predict the absorption mechanism of the two compounds, the study on bidirectional permeability from the apical (AP) side to the basolateral (BL) side as well as from the BL side to the AP side was carried out. The determination was performed by HPLC-UV method. And the permeability parameters, especially the apparent permeability coefficients (Papp), were then calculated. The Papp values of LQ and ILQ are (5.40±0.16)× 10^-7 and (8.69±0.15)× 10^-7 cm/s, respectively. The results of time- and concentration-dependent transport experiments indicate that both LQ and ILQ are poor absorbed mainly through passive diffusion.展开更多
Selective adsorption of active ingredients liquiritin and isoliquiritin from Glycyrrhiza uralensis Fisch with multi-walled carbon nanotubes(MWNTs) has been studied.Distribution coefficients of liquiritin between ethan...Selective adsorption of active ingredients liquiritin and isoliquiritin from Glycyrrhiza uralensis Fisch with multi-walled carbon nanotubes(MWNTs) has been studied.Distribution coefficients of liquiritin between ethanol solvent and r-MWNTs or o-MWNTs in 293K is 37.5 and 43.3,while the distribution coefficients of isoliquiritin between ethanol solvent and r-MWNTs or o-MWNTs is 717 and 1080,respectively.It was indicated that the distribution coefficient of isoliquiritin adsorbed by MWNTs was much larger than that of liquiritin,and oxidation treatment of MWNTs could obviously enhance their adsorption ability.The possible reasons that MWNTs can selectively adsorb isoliquiritin other than liquiritin were discussed on the bases of molecular structure of the active ingredients and their interaction with nanotubes.展开更多

However,rare studies in the field of isoliquiritin antidepressant-like effects have been carried out,the molecular mechanism also remains unclear. In this study,the model of corticosterone-damaged rat adrenal pheochromocytoma(PC12)cells has been used to generate an in vitro experimental model of depression. The aim was to investigate thecytoprotective efficiency and potential mechanisms of isoliquiritin in corticosterone-damaged PC12 cells. METHODS The cells were treated with 400 μmol·L^(-1)corticosterone in the absence or presence of isoliquiritin for 24 h; cells viability and lactate dehydrogenase(LDH) leakage were then determined,Hoechst 33342/PI and Annexin V/PI double staining were performed to evaluate the cytoprotective efficiency of isoliquiritin. Next,intracellular calcium([Ca^(2+)]_i),mitochondrial membrane potential(MMP),reactive oxygen species(ROS),malondialdehyde(MDA),the activity of superoxide dismutase(SOD)and catalase(CAT),and Western blotting analysed for the expression of Bcl,Bax,caspase-3 and cytochrome C(Cyt-C) were investigated to explore the potential mechanisms. RESULTS The results of the present study showed that pretreatment of PC12 cells with isoliquiritinsignificantly prevented the corticosterone-induced cell apoptosis. In addition,isoliquiritin increased the activity of SOD and CAT,decreased the contents of ROS and MDA. These findings suggest that isoliquiritin provided a protective action against corticosterone-induced celldamage by reducing oxidative stress. Furthermore pretreatmented with isoliquiritin reduced corticosterone-induced mitochondrial dysfunction by preventing mitochondrial membrane potentials dissipation.Our findings indicated that isoliquiritin might exert its therapeutic effects viaregulating mitochondrial dysfunction. Moreover isoliquiritin strongly attenuated [Ca^(2+)]_i overload and down-regulation of Bax,caspase-3 and Cyt-C protein expression,up-regulation of Bcl protein expression. CONCLUSION Isoliquiritin has acytoprotective effect on corticosterone-induced cytotoxicity in PC12 cells,which may be related to its antioxidant action,inhibition of [Ca^(2+)]_ioverload and inhibition of the mitochondrial apoptotic pathway.展开更多
