跨膜蛋白Crumbs(Crb)是细胞顶部的决定因子,对上皮细胞顶?底极性的建立和维持起着关键的作用。其胞内域虽然仅有37个氨基酸,但对Crb的功能必不可少。在果蝇(Drosophila melanogaster)中,如果胞内域发生突变,将造成胚胎发育异常、上皮细... 跨膜蛋白Crumbs(Crb)是细胞顶部的决定因子,对上皮细胞顶?底极性的建立和维持起着关键的作用。其胞内域虽然仅有37个氨基酸,但对Crb的功能必不可少。在果蝇(Drosophila melanogaster)中,如果胞内域发生突变,将造成胚胎发育异常、上皮细胞顶底极性丧失等严重后果。Crb胞内域从果蝇到小鼠(Mus musculus)和人类(Homo sapiens)具有很高的同源性,但线虫(Caenorhabditis elegans)两个Crb蛋白的胞内域与果蝇和哺乳动物却较为不同。为验证线虫Crb蛋白胞内域是否功能保守,本文利用基因组工程法(Genomic engineering),将果蝇基因组中Crb基因编码胞内域的部分替换为一致性和相似性较远的线虫Crb2基因的相应区段。与其他Crb胞内域突变果蝇不同,替换突变体胚胎发育正常,Crb及其他极性蛋白的表达和定位正常,胚胎上皮细胞顶底极性能够正确的建立和维持。这些结果证实虽然线虫和果蝇Crb蛋白胞内域之间存在大量序列变异,但重要的氨基酸位点和功能模块则完全保守。 展开更多

A novel soluble, reactive ladder-like polysilsesquioxane (L) with lateral bromophenyl groups was synthesized successfully by stepwise coupling polymerization (SCP) method including preaminolysis, hydrolysis and polycondensation steps. The monomer 3-trichlorosilylpropoxy-4-bromobenzene (M) was first prepared via hydrosilylation reaction catalyzed by dicyclopentadienylplatinum dichloride (Cp2PtCl2). The title polymer (L) was characterized by 1H-NMR, 29Si-NMR, FTIR, X-ray diffraction (XRD), DSC and vapor pressure osmometry (VPO). The experimental results indicate that the polymer (L) possesses a typical ladder-like structure. 展开更多

The crystal is of triclinic, space group P1 with a = 10.266(2), b = 11.573(2), c = 12.651(3) A, α = 89.95(3), β= 74.64(3),γ = 68.54(3)°, C50H44Cd4N12O18, Mr = 1550.57, V= 1341.1(5)A3, Dc = 1.920 g/cm3, F(000) = 764, p = 1.651 cm-1 and Z = 1. The final refinement gave R = 0.0342 and wR = 0.0738 for 5925 observed reflections with I 〉 2σ(I). The crystal structure involves two different ladder-like chains, which are further connected to form a ladder-like double chain architecture via hydrogen bonding interactions, extending along the b axis. The weak interactions including hydrogen bonds and π-π staking interactions contribute to the alignment of the complex in the crystalline state. 展开更多

BACKGROUND Crumbs homolog 2(CRB2)is a recently discovered gene that is closely related to the maintenance of normal polarity in podocytes;mutations can directly lead to steroid-resistant nephrotic syndrome(SRNS).However,the characteristics of nephrotic syndrome(NS)caused by CRB2 mutations have not been described.CASE SUMMARY We report a novel compound heterozygous mutation of the CRB2 gene in two siblings with SRNS.The two siblings had edema,proteinuria,hypoproteinemia and hyperlipidemia.Both their father and mother had normal phenotypes(no history of NS).Whole exon sequencing(WES)of the family showed a novel compound heterozygous mutation,c.2290(exon 8)C>T and c.3613(exon 12)G>A.Glucocorticoid therapy(methylprednisolone pulse therapy or oral prednisone)and immunosuppressive agents(tacrolimus)had no effect.During a 3-year follow-up after genetic diagnosis by WES,proteinuria persisted,but the patient was healthy.CONCLUSION CRB2 mutations related to SRNS often occur in exons 7,10,and 12.Clinical manifestations of SRNS caused by CRB2 mutations are often less severe than in other forms of SRNS. 展开更多

Compound 1 crystallizes in monoclinic, space group P21/c with a = 7.474(1), b = 17.807(1), c = 18.851(1) A, β = 108.32(1)°, V = 2381.7(2) A3, Z = 4, C16H32CI2Cu2Ns, Mr = 534.48, Dc = 1.491 g.cm-3,μ = 2.03 mm-1, F(000) = 1104, GOOF = 1.050, the final R = 0.0445 and wR = 0.1162 for 3162 observed reflections (I 〉 2σ(I)). Compound 1 shows discrete dimeric structures (A and B) containing inversion centers and the Cu(l) ions are coordinated in triangle geometries. The isomers are connected by N-H…CI hydrogen bonds, chains with graph-set C(7) and rings R22(14) and C-H..'Jr interactions into stair-step chains (Tapes A and B) running parallel to the [01-1] direction. The N-H…C1 hydrogen bonds result in chain and cyclic structures with graph-sets C22(17) and R34(18) linking tapes A and B to form two-dimensional networks along the [031] direction. Packing of crystal 1 is stabilized by rings R34(18) and weak C-H…C1 hydrogen bonds parallel to the [01-2] direction. Dynamic Behaviour of Pond Ash Mixed with Coir and Crumb Rubber

An experimental program was conducted to determine the dynamic properties of pond ash(PA)and pond ash mixed with admixtures like crumb rubber(CR)and coir fiber(CF)which are wastes of different industries.Additives CR and CF were mixed separately with pond ash in a percentage ranging from 0%–20%at an interval of 5%and 0%–2%at an interval of 0.5%by weight of pond ash,respectively.Dynamic properties(Shear modulus,Number of cycles to initiate liquefaction,Degradation index and Damping ratio)for pond ash mixes were determined using strain-controlled cyclic tri-axial apparatus.The shear strain was kept constant at 1.2%and the frequency varied from 0.4 to 1 Hz.Liquefaction resistance improves when crumb rubber is added to pond ash,however adding Coir fiber to pond ash has essentially no effect on the number of cycles required to initiate liquefaction.The dynamic shear modulus decreases when crumb rubber was mixed in pond ash whereas when coir fiber is added to pond ash,the dynamic shear modulus increases significantly.A mix of PA,CR,and CF was also tried to balance the number of cycles required to initiate liquefaction and shear modulus.The mixed sample in the proportion of PA(84%)+CR(15)+CF(1%)can be termed a balanced mix as the number of cycles to initiate liquefaction was more than all PA-CF mixes and also the shear modulus was more than all PA-CR mixes.Damping ratio increases with increasing frequency of loading(0.4 to 1 Hz). 展开更多

为确定橡胶混凝土的软化本构模型,对2种缝高比和5种橡胶掺量的混凝土试件进行楔入劈拉试验研究和理论分析。依据试验结果,采用规范公式及能量守恒定律计算混凝土试件的断裂韧度和断裂能,并求解出PETERSSON、欧洲混凝土规范、XU和REINHAR... 为确定橡胶混凝土的软化本构模型,对2种缝高比和5种橡胶掺量的混凝土试件进行楔入劈拉试验研究和理论分析。依据试验结果,采用规范公式及能量守恒定律计算混凝土试件的断裂韧度和断裂能,并求解出PETERSSON、欧洲混凝土规范、XU和REINHARDT3种软化曲线的相关参数。根据3种软化本构关系得到黏聚韧度理论值,并与黏聚韧度试验计算值进行了对比分析。研究结果表明:掺入适量的橡胶集料可提高混凝土的断裂失稳韧度和断裂能;3种典型的混凝土软化模型均不能很好地描述橡胶混凝土的黏聚韧度;对XU和REINHARDT软化关系式进行修正后,得到了橡胶混凝土的软化本构模型,可用于研究橡胶混凝土材料的抗裂特性。 展开更多

Conventional repairing methods for asphalt pavement have some inconveniences,such as insufficient strength,and are typically time-consuming.To address these issues,this study proposes a new technological method to design and prepare a high-performance assembled asphalt concrete block for fast repair of the potholes.A series of composite modified asphalt binders with 10%crumb rubber(CR)and different dosages(0%,1%,3%,5%)of polyurethane(PU)are examined to determine the optimized binder.Subsequently,the corresponding asphalt mixtures are prepared for further comparison and assessment of engineering properties,such as moistureinduced damage,high-temperature deformation,and low-temperature cracking characteristics.The test results show that PU can significantly improve the high-temperature performance and hardness of(crumb rubber modified asphalt)CRMA binder;3%PU contributes allowing the resistance of CRMA mixture to moisture-induced damage at higher levels,particularly under water whole immersion;as 3%PU is added,the high-temperature rutting deformation resistance of the CRMA mixture increases significantly,and the low-temperature anti-cracking properties are also improved slightly.Therefore,the innovatively designed high-quality assembled fast-repairing asphalt concrete block is recommended as an appropriate option for highway maintenance. 展开更多

以某公司加工的环烷基和中间基原油为原料,通过优化原油配比,采用常减压蒸馏工艺,通过控制减压蒸馏工艺条件,生产出符合技术标准要求且能够用于改性沥青的基质沥青原料。通过筛选与基质沥青配伍性好的胶粉、助剂添加比例及工艺条件优化... 以某公司加工的环烷基和中间基原油为原料,通过优化原油配比,采用常减压蒸馏工艺,通过控制减压蒸馏工艺条件,生产出符合技术标准要求且能够用于改性沥青的基质沥青原料。通过筛选与基质沥青配伍性好的胶粉、助剂添加比例及工艺条件优化,研制出符合技术规范要求的胶粉复合改性沥青并进行了工业试生产。对工业生产的产品检测及专业部门鉴定结果表明,胶粉复合改性沥青产品各项技术指标符合JT/T798-2019《路用废胎胶粉橡胶沥青》技术要求,成功实现了工业化生产和应用。 展开更多
