In this study the numerical simulation has been first carried out on fluctuation and undulation of thermal layer under influence of tide (simultaneous input of 8 tidal components). The study reveals the geographic distribution of thermal layer fluctuation in the entire study region and temporal and spatial variations of the undulation in tidal period superposing on the fluctuation. Especially, the wave with large amplitude simulated is consistent with observation in the channal and the sea areas with a convex coastline and complex variation of depth, internal relations of tide, tidal current, residual current as well as the factors such as geography, and the fluctuation of thermal layer is induced by residual current due to unsymmetry which occurs as a result of the tidal movement in lower layer influenced by friction and geography, meanwhile, analysis indicates that the fluctuation of thermal layer and tidal oscillation are different undulations in character.展开更多

In ...As a short-day plant species, maize requires an optimal photoperiod for inducing reproductive growth. However, there is a lack of information regarding photoperiod-induced changes in maize mRNA and protein levels. In this study, a photoperiod-insensitive maize inbred line and its near isogenic photoperiod-sensitive line were used. By integrating RNAbased transcriptomic and iTRAQ LC-MS/MS-based proteomic approaches, we generated a comprehensive inventory of the transcripts and proteins with altered abundances in response to a long photoperiod(LP) during growth stage transitions. We detected 22 000 transcripts in RNA-sequence runs and 5 259 proteins from an iTRAQ-based analysis. A weak correlation between mRNA-and protein-level changes was observed, suggesting the LP-induced transition between maize growth stages is largely regulated post-transcriptionally. Differentially expressed genes influenced by LP conditions were associated with several regulatory processes in both maize inbred lines, especially phosphate ion transport and the circadian rhythm. Additionally, 31 transcripts and six proteins related to photoperiodic flowering in maize were identified by comparing transcriptomic and proteomic data. This transcriptomic and proteomic analysis represents the first comprehensive and comparative study of gene/protein-level changes occurring in photoperiod-sensitive and-insensitive maize inbred lines during growth stage transitions under LP conditions.展开更多
Underground hydrogen storage(UHS)and compressed air energy storage(CAES)are two viable largescale energy storage technologies for mitigating the intermittency of wind and solar power.Therefore,it is meaningful to comp...Underground hydrogen storage(UHS)and compressed air energy storage(CAES)are two viable largescale energy storage technologies for mitigating the intermittency of wind and solar power.Therefore,it is meaningful to compare the properties of hydrogen and air with typical thermodynamic storage processes.This study employs a multi-physical coupling model to compare the operations of CAES and UHS,integrating gas thermodynamics within caverns,thermal conduction,and mechanical deformation around rock caverns.Gas thermodynamic responses are validated using additional simulations and the field test data.Temperature and pressure variations of air and hydrogen within rock caverns exhibit similarities under both adiabatic and diabatic simulation modes.Hydrogen reaches higher temperature and pressure following gas charging stage compared to air,and the ideal gas assumption may lead to overestimation of gas temperature and pressure.Unlike steel lining of CAES,the sealing layer(fibre-reinforced plastic FRP)in UHS is prone to deformation but can effectively mitigates stress in the sealing layer.In CAES,the first principal stress on the surface of the sealing layer and concrete lining is tensile stress,whereas UHS exhibits compressive stress in the same areas.Our present research can provide references for the selection of energy storage methods.展开更多

This paper quantitatively studies the transient dynamic response of a semi-submersible production platform with the loss of one or several positioning mooring lines.A semi-submersible platform,production risers,and po...This paper quantitatively studies the transient dynamic response of a semi-submersible production platform with the loss of one or several positioning mooring lines.A semi-submersible platform,production risers,and positioning mooring lines are all included in the numerical simulation.Increased motion of the semi-submersible platform,tension variation of the remaining mooring lines/risers and the risk of mooring line or riser clashing are all investigated through fully coupled time-domain analysis.Combined environmental loads are selected from irregular waves and the steady current varying from very rough to extreme sea conditions.Three dimension radiation/diffraction theories and Morison’s equation are applied to calculate first-order wave force and second-order mean drift force of floating semi-submersible platform.Nonlinear time-domain finite element methods are employed to analyze the behavior of mooring lines and risers.Results show that the failure of mooring lines seriously reduce the platform’s stability performance.The tension of the rest lines is also increased accordingly.Remaining lines which are closer to the failed lines will have larger tension increase to compensate.Line-Line distance provides practical information for the risk of clashing investigation.展开更多

The Tet-On system is a ...AIM: Artificial beta-cell lines may offer an abundant source of cells for the treatment of type I diabetes, but insulin secretion in beta-cells is tightly regulated in physiological conditions. The Tet-On system is a "gene switch" system, which can induce gene expression by administration of tetracycline (Tet) derivatives such as doxcycline (Dox). Using this system, we established 293 cells to an artificial cell line secreting insulin in response to stimulation by Dox. METHODS: The mutated proinsulin cDNA was obtained from plasmid pcDNA3.1/C-mINS by the polymerase chain reaction (PCR), and was inserted downstream from the promoter on the expression vector pTRE2, to construct a recombined expression vector pTRE2mINS. The promoter on pTRE2 consists of the tetracycline-response element and the CMV minimal promoter and is thus activated by the reverse tetracycline-controlled transactivator (rtTA) when Dox is administrated. pTRE2mINS and plasmid pTK-Hyg encoding hygromycin were co-transfected in the tet293 cells, which express rtTA stably. Following hygromycin screening, the survived cells expressing insulin were selected and enriched. Dox was used to control the expression of insulin in these cells. At the levels of mRNA and protein, the regulating effect of Dox in culture medium on the expression of proinsulin gene was estimated respectively with Northern blot, RT-PCR, and radioimmunoassay. RESULTS: From the 28 hygromycin-resistant cell strains, we selected one cell strain (tet293/Ins6) secreting insulin not only automatically, but in response to stimulation by Dox. The amount on insulin secretion was dependent on the Dox dose (0,10,100,200,400,800 and 1000 microg.L(-1)), the level of insulin secreted by the cells treated with Dox (1000 microg.L(-1)) was 241.0pU.d(-1).cell(-1) , which was 25-fold that of 9.7pU.d(-1).cell(-1) without Dox treatment. Northern blot analyses and RT-PCR further confirmed that the transcription of insulin gene had already been up-regulated after exposing tet293/Ins6 cells to Dox for 15 minutes, and was also induced in a dose-dependent manner. However, the concentration of insulin in the media did not increase significantly until 5 hours following the addition of Dox. CONCLUSION: Human proinsulin gene was transfected successfully and expressed efficiently in 293 cells, and the expression was modulated by tetracycline and its derivatives, improving the accuracy, safety, and reliability of gene therapy, suggesting that conditional establishment of artificial beta-cells may be a useful approach to develop cellular therapy for diabetes mellitus.展开更多
The influence of the interaction between surrounding rock and lining on the long-term behaviour of a tunnel in service is significant.In this paper,we proposed a mechanical model of the circular lined tunnel with the ...The influence of the interaction between surrounding rock and lining on the long-term behaviour of a tunnel in service is significant.In this paper,we proposed a mechanical model of the circular lined tunnel with the alterable mechanical property under hydrostatic stress and radially inner surface pressure of the lining.The alterable mechanical properties of the surrounding rock and the lining are embodied by the changing of their elasticity modulus with service time and radial direction of the tunnel,respectively.The proposed mechanical model is successfully validated by comparison with the existing theoretical models and the numerical simulation,respectively.The influences of the main parameters of the proposed mechanical model,such as the radial power-law indexes and the time-varying coefficients of the surrounding rock and the lining,as well as the radially inner surface pressure of the lining,on the interface displacement and pressure between surrounding rock and lining are investigated.The research results can provide some valuable references for timely diagnosis and correct evaluation of the long-term behaviours of a tunnel in service.展开更多

Primarily cultured cells grew...A novel continuous ovary cell line from barfin flounder(Verasper moseri)(BFO cell line) was established with its primitive application in transgenic expression demonstrated in this study. Primarily cultured cells grew well at 22℃ in Dulbecco's modified Eagle medium/F12 medium(DMEM/F12, 1:1; p H 7.2) supplemented with 20% fetal bovine serum(FBS), carboxymethyl chitooligosaccharide, basic fibroblast growth factor(b FGF) and insulin-like growth factor-I(IGF-I). The primary BFO cells in fibroblastic morphology proliferated into a confluent monolayer about 2 weeks later, and were able to be subcultured. Impacts of medium and temperature on the growth of the cells were examined. The optimum growth was found in DMEM/F12 with 20% FBS and at 22℃. The BFO cells can be continuously subcultured to Passage 120 steadily with a population doubling time of 32.7 h at Passage 60. Chromosome analysis revealed that 72% of BFO cells at Passage 60 maintained the normal diploid chromosome number(46) with a normal karyotype of 2st+44t. The results of gene transformation indicated that green fluorescence protein(GFP) positively expressed in these cells after being transformed with pc DNA3.1-GFP. Therefore, a continuous and transformable BFO cell line was successfully established, which may serve as a useful tool for cytotechnological manipulation and transgenic modification of this fish.展开更多

It was found that the cell growth rate was markedly reduced by profenofos at the concentrations of 2.5, 5 and 7.5 mg/L tested; and that the fine structures of the cells were also altered by profenofos, as evidenced by dilation of nuclear membranes and mitochondria cristae, and presence of enlarged lysosomes with engulfed organelles and numerous vacuoles in the cytoplasm. Probably, mitochondria, the cell energy generating sites, are the most prominent sites of profenofos cytotoxity in the cells. This seems to be the first report of the use of marine fish cell line for evaluation of the acute in vitro cytotoxicity of organophosphorus pesticide.展开更多
A high-efficiency propeller can enable a long mission duration for autonomous underwater vehicles(AUVs).In this study,a new method with OpenProp coupled with computational fluid dynamics was developed to design a prop...A high-efficiency propeller can enable a long mission duration for autonomous underwater vehicles(AUVs).In this study,a new method with OpenProp coupled with computational fluid dynamics was developed to design a propeller for an Explorer100 AUV.The towed system simulation of the AUV was used to measure the nominal wake,and a self-propulsion simulation was used to measure the effective wake at the disc plane just in front of a propeller.Two propellers referring to the nominal wake(propeller 1)and effective wake(propeller 2)were designed with OpenProp and appended with the AUV for self-propulsion simulations,respectively.Through the numerical simulation of the AUV self-propulsion tests,the cruising velocity of AUV was obtained.The flow characteristics of the self-propulsion in pressure and velocity contours were also analyzed.The propeller designed with an effective wake improved the thrust,velocity,and efficiency by approximately 11.3%,6.7%,and 2.5%,respectively,as compared with those with a nominal wake.The cruising velocity of the final designed propeller for the Explorer100 AUV improved by 21.8%,as compared to that of the original propeller from the AUV free-running tests.展开更多
Close-distance coal seams are widely distributed over China,and the coal pillars left by the overlying coal seams afect the retracement channel of the underlying coal seam in the stopping stage.Based on the engineerin...Close-distance coal seams are widely distributed over China,and the coal pillars left by the overlying coal seams afect the retracement channel of the underlying coal seam in the stopping stage.Based on the engineering background of close-distance seam mining in a coal mine,the reasonable position of the underlying coal seam's stopping line and the support method of the large section roadway during stopping are investigated using feld measurements,similar simulation experiments,and numerical simulations.There are three types of location relationships between the stopping line of the underlying coal seam and the stopping line of the overlying coal seam:"externally staggered with the upper stopping line"(ESUL,stops mining under the overlying goaf),"overlapped with upper stopping line"(OUL),and"internally staggered with the upper stopping line"(ISUL,ISUL-SD for shorter internal staggered distances,ISUL-LD for longer ones).There are diferent stress arch structures in the overlying strata of the above three positions,and the stress arch evolution process exists in the process of ESUL→OUL→ISUL-SD→ISUL-LD:a front and rear double stress arch structure→the front arch gradually decreases→the front arch dies out,and the double arch synthesizes the single arch→the single-arch range expands→the nested double arch.The relationship between the stress arch structure and the position of the stopping line is evaluated as follows:(1)ESUL:the stress concentration in the roof plate of the retracement channel of the underlying coal seam is the highest,because the overburden block of the extensive collapse zone acts directly on the roof plate of the retracement channel,resulting in relative difculties in roof support.(2)OUL:although the retracement channel roof pressure is minimal,the overlying rock structure has the potential for rotation or slippage instability.(3)ISUL-SD:the pressure on the roof of the retracement channel is small and the overburden structure is stable,which is conducive to the safe retraction of the support and not limited by the width of the end-mining coal pillar.(4)ISUL-LD:it is basically the same as the condition of stopping under the non-goaf;however,it has a limitation on the width of the end-mining coal pillar.The location of the stopping line is selected as ISUL-SD,and the retraction process of the self-excavating retraction channel was adopted.A partition asymmetric support scheme which is proven by feld practice is proposed,through a comprehensive analysis of the pre-stress feld simulation of the support scheme,based on the diferent control requirements of the roof above the support and the roof of the retracement channel in the stopping area.This method realizes safe and smooth withdrawal of the support.展开更多
Electrical pollution is a worldwide concern,because it is potentially harmful to human health.Trees not only play a significant role in moderating the climate,but also can be used as shields against electrical polluti...Electrical pollution is a worldwide concern,because it is potentially harmful to human health.Trees not only play a significant role in moderating the climate,but also can be used as shields against electrical pollution.Shielding effects on the electric field strength under transmission lines by two tree species,Populus alba and Larix gmelinii,were examined in this study.The electrical resistivity at different heights of trees was measured using a PiCUS sonic tomograph,which can image the electrical impedance for trees.The electric field strength around the trees was measured with an elf field strength measurement system,HI-3604,and combined with tree resistivity to develop a model for calculating the electric field intensity around trees using the finite element method.In addition,the feasibility of the finite element method was confirmed by comparing the calculated results and experimental data.The results showed that the trees did reduce the electric field strength.The electric field intensity was reduced by 95.6%,and P.alba was better than L.gmelinii at shielding.展开更多

The construction of this platform is based on the user pro...In this paper, a hybrid automatic optimization strategy is proposed for the design of underwater robot lines. Isight is introduced as an integration platform. The construction of this platform is based on the user programming and several commercial software including UG6.0, GAMBIT2.4.6 and FLUENT12.0. An intelligent parameter optimization method, the particle swarm optimization, is incorporated into the platform. To verify the strategy proposed, a simulation is conducted on the underwater robot model 5470, which originates from the DTRC SUBOFF project. With the automatic optimization platform, the minimal resistance is taken as the optimization goal;the wet surface area as the constraint condition; the length of the fore-body, maximum body radius and after-body's minimum radius as the design variables. With the CFD calculation, the RANS equations and the standard turbulence model are used for direct numerical simulation. By analyses of the simulation results, it is concluded that the platform is of high efficiency and feasibility. Through the platform, a variety of schemes for the design of the lines are generated and the optimal solution is achieved. The combination of the intelligent optimization algorithm and the numerical simulation ensures a global optimal solution and improves the efficiency of the searching solutions.展开更多
The in vitro acute cytotoxicity of the neonicotinoid insecticide acetamiprid to FG cell line, derived from the gill of flounder Paralichthys olizaceus, was tested by the three widely used endpoint bioassays-neutral r...The in vitro acute cytotoxicity of the neonicotinoid insecticide acetamiprid to FG cell line, derived from the gill of flounder Paralichthys olizaceus, was tested by the three widely used endpoint bioassays-neutral red (NR) assay, tetrazolium (MTT) assay and cell protein assay. It was found that acetamiprid was increasingly toxic to FG cells at concentrations of 1 μg/cm^3 or above, and the inhibitory concentration 50% values for NR, MTF, and cell protein assays were 38.38, 36.27 and 32.03 μg/cm^3, respectively. This appeared to be the first report on the in vitro cytotoxicity of acetamiprid to non-mammalian vertebrate cells. Ultrastructural examination revealed that for the cells exposed to 60 μg/cm^3 acetamiprid for 48 h, their mitochondria were severely damaged with the cristae swelled up or disrupted, while their nuclei and rough endoplasmic reticlum (RER) appeared to be still normal. This suggests that mitochondria are possibly the primary target of acetamiprid.展开更多

Neurogenesis occurs continuously in different areas of the central neural system in adult teleosts compared with adult mammals. Therefore, NSC cell lines must be established to offer a valuable in vitro system for studies on neurogenesis and other related functions. In this study, four cell lines designated as PoB1, PoB2, PoBf and PoBh were established from the brain of the Japanese flounder Paralichthys olivaceus. The cell lines were sub-cultured over 150 times and still grew well in DMEM/F12 medium at 24℃. PoB1, PoB2, PoBf and PoBh were identified as neural stem/progenitor cell lines on the basis of the mRNA expression of nestin and/or aldh111 or slc6 a4 and the formation of neurospheres. The cells transfected with the pEGFP-C1 plasmid showed fluorescent signals with distinct reagent dependencies. The established cell lines from the brain of P. olivaceus offer a valuable system in vitro for the study of neurogenesis, fish neural regulation and endocrinology-related functions.展开更多

In this paper,the simultaneous resonance of a ferromagnetic thin plate in a time-varying magnetic field,having axial speed and being subjected to a periodic line load,is studied.Based on the large deflection theory of...In this paper,the simultaneous resonance of a ferromagnetic thin plate in a time-varying magnetic field,having axial speed and being subjected to a periodic line load,is studied.Based on the large deflection theory of thin plates and electromagnetic field theory,the nonlinear vibration differential equation of the plate is obtained by using the Hamilton′s principle and the Galerkin method.Then the boundary condition in which the longer opposite sides are clamped and hinged is considered.The dimensionless nonlinear differential equations are solved by using the method of multiple scales,and the analytical solution is given.In addition,the stability analysis is also carried out by using Lyapunov stability theory.Through numerical analysis,the variation curves of system resonance amplitude with frequency tuning parameter,magnetic field strength and external excitation amplitude are obtained.Different parameters that have significant effects on the response of the system,such as the thickness,the axial velocity,the magnetic field intensity,the position,and the frequency of external excitation,are considered and analyzed.The results show that the system has multiple solution regions and obvious nonlinear coupled characteristics.展开更多

The multiaxial fatigue criteria proposed by Wang and Brown, together with the ralnflow counting method and Miner- Palmgren's rule, are applied to the cumulative damage estimation and life prediction. As the axis line deflection of overlong kilns generally results in asymmetric load distribution on each roller, the load ratio is introduced to describe the deflection for quantitative stress analyses. The stress analyses are performed within the finite element code ANSYS. The tangential friction stress is calculated in terms :of the condition of the rolling contact area. By taking one roller as an example, the plotted fatigue life versus load ratio curve discovers how the axis line deflection affects the fatigue life. This study is significant to prevent the fatigue failure of the roller and can provide basis to adjust and optimize the axis line of the rotary kiln.展开更多
The tower line system will be in an unsafe status due to uniform or uneven fall of ice coating which is attached to the surface of tower and lines.The fall of ice could be caused by wind action or thermal force.In ord...The tower line system will be in an unsafe status due to uniform or uneven fall of ice coating which is attached to the surface of tower and lines.The fall of ice could be caused by wind action or thermal force.In order to study the dynamic characteristics of the self-failure of the transmission line under the action of dynamicwind load,a finite elementmodel of the two-span transmission tower line system was established.The birth and death element methods are used to simulate the icing and shedding of the line.Tensile failure strength is the shedding criterion for ice coating.The fluctuating wind speed time history of the tower line systemis first simulated,and then the fluctuating wind and the average wind are superimposed to generate the instantaneous wind speed and converted into wind load.The dynamic response of the transmission tower line systemunder iced coupling with different wind speeds and different thicknesses of ice coating was studied.This is the first attempt that the coupling dynamic response of the icing shedding and wind load for the transmission tower-line system is discussed in this paper.In addition,the dynamic characteristics of wind are included.In particular,the limiting mechanical conditions are considered.According to the simulation results,it is found:because of the ice shedding,the stress of the conductor changes obviously in the first 20 seconds,and the ground wire changes sharply in the first two seconds;the icing of the conductor(ground)wire is gradually deicing under the action of wind vibration;the displacement of tower top increases with the increase of wind speed and icing thickness.展开更多

文摘As a short-day plant species, maize requires an optimal photoperiod for inducing reproductive growth. However, there is a lack of information regarding photoperiod-induced changes in maize mRNA and protein levels. In this study, a photoperiod-insensitive maize inbred line and its near isogenic photoperiod-sensitive line were used. By integrating RNAbased transcriptomic and iTRAQ LC-MS/MS-based proteomic approaches, we generated a comprehensive inventory of the transcripts and proteins with altered abundances in response to a long photoperiod(LP) during growth stage transitions. We detected 22 000 transcripts in RNA-sequence runs and 5 259 proteins from an iTRAQ-based analysis. A weak correlation between mRNA-and protein-level changes was observed, suggesting the LP-induced transition between maize growth stages is largely regulated post-transcriptionally. Differentially expressed genes influenced by LP conditions were associated with several regulatory processes in both maize inbred lines, especially phosphate ion transport and the circadian rhythm. Additionally, 31 transcripts and six proteins related to photoperiodic flowering in maize were identified by comparing transcriptomic and proteomic data. This transcriptomic and proteomic analysis represents the first comprehensive and comparative study of gene/protein-level changes occurring in photoperiod-sensitive and-insensitive maize inbred lines during growth stage transitions under LP conditions.

文摘Underground hydrogen storage(UHS)and compressed air energy storage(CAES)are two viable largescale energy storage technologies for mitigating the intermittency of wind and solar power.Therefore,it is meaningful to compare the properties of hydrogen and air with typical thermodynamic storage processes.This study employs a multi-physical coupling model to compare the operations of CAES and UHS,integrating gas thermodynamics within caverns,thermal conduction,and mechanical deformation around rock caverns.Gas thermodynamic responses are validated using additional simulations and the field test data.Temperature and pressure variations of air and hydrogen within rock caverns exhibit similarities under both adiabatic and diabatic simulation modes.Hydrogen reaches higher temperature and pressure following gas charging stage compared to air,and the ideal gas assumption may lead to overestimation of gas temperature and pressure.Unlike steel lining of CAES,the sealing layer(fibre-reinforced plastic FRP)in UHS is prone to deformation but can effectively mitigates stress in the sealing layer.In CAES,the first principal stress on the surface of the sealing layer and concrete lining is tensile stress,whereas UHS exhibits compressive stress in the same areas.Our present research can provide references for the selection of energy storage methods.

Numerical averaging method is used to extract bi-soliton from a couple of Gaussian pulses and its stability is studied by a free propagation for a long distance.
文摘Crack line field analysis method has become an independent method for crack elastic-plastic analysis, which greatly simplifies the complexity of crack elastic-plastic problems and overcomes the corresponding mathematical difficulty. With this method, the precise elastic-plastic solutions near crack lines for variety of crack problems can be obtained. But up to now all solutions obtained by this method were for different concrete problems, no general steps and no general form of matching equations near crack line are given out. With crack line analysis method, this paper proposes the general steps of elastic plastic analysis near crack line for mode I crack in elastic-perfectly plastic solids under plane stress condition, and in turn given out the solving process and result for a specific problem.

文摘This paper quantitatively studies the transient dynamic response of a semi-submersible production platform with the loss of one or several positioning mooring lines.A semi-submersible platform,production risers,and positioning mooring lines are all included in the numerical simulation.Increased motion of the semi-submersible platform,tension variation of the remaining mooring lines/risers and the risk of mooring line or riser clashing are all investigated through fully coupled time-domain analysis.Combined environmental loads are selected from irregular waves and the steady current varying from very rough to extreme sea conditions.Three dimension radiation/diffraction theories and Morison’s equation are applied to calculate first-order wave force and second-order mean drift force of floating semi-submersible platform.Nonlinear time-domain finite element methods are employed to analyze the behavior of mooring lines and risers.Results show that the failure of mooring lines seriously reduce the platform’s stability performance.The tension of the rest lines is also increased accordingly.Remaining lines which are closer to the failed lines will have larger tension increase to compensate.Line-Line distance provides practical information for the risk of clashing investigation.

文摘AIM: Artificial beta-cell lines may offer an abundant source of cells for the treatment of type I diabetes, but insulin secretion in beta-cells is tightly regulated in physiological conditions. The Tet-On system is a "gene switch" system, which can induce gene expression by administration of tetracycline (Tet) derivatives such as doxcycline (Dox). Using this system, we established 293 cells to an artificial cell line secreting insulin in response to stimulation by Dox. METHODS: The mutated proinsulin cDNA was obtained from plasmid pcDNA3.1/C-mINS by the polymerase chain reaction (PCR), and was inserted downstream from the promoter on the expression vector pTRE2, to construct a recombined expression vector pTRE2mINS. The promoter on pTRE2 consists of the tetracycline-response element and the CMV minimal promoter and is thus activated by the reverse tetracycline-controlled transactivator (rtTA) when Dox is administrated. pTRE2mINS and plasmid pTK-Hyg encoding hygromycin were co-transfected in the tet293 cells, which express rtTA stably. Following hygromycin screening, the survived cells expressing insulin were selected and enriched. Dox was used to control the expression of insulin in these cells. At the levels of mRNA and protein, the regulating effect of Dox in culture medium on the expression of proinsulin gene was estimated respectively with Northern blot, RT-PCR, and radioimmunoassay. RESULTS: From the 28 hygromycin-resistant cell strains, we selected one cell strain (tet293/Ins6) secreting insulin not only automatically, but in response to stimulation by Dox. The amount on insulin secretion was dependent on the Dox dose (0,10,100,200,400,800 and 1000 microg.L(-1)), the level of insulin secreted by the cells treated with Dox (1000 microg.L(-1)) was 241.0pU.d(-1).cell(-1) , which was 25-fold that of 9.7pU.d(-1).cell(-1) without Dox treatment. Northern blot analyses and RT-PCR further confirmed that the transcription of insulin gene had already been up-regulated after exposing tet293/Ins6 cells to Dox for 15 minutes, and was also induced in a dose-dependent manner. However, the concentration of insulin in the media did not increase significantly until 5 hours following the addition of Dox. CONCLUSION: Human proinsulin gene was transfected successfully and expressed efficiently in 293 cells, and the expression was modulated by tetracycline and its derivatives, improving the accuracy, safety, and reliability of gene therapy, suggesting that conditional establishment of artificial beta-cells may be a useful approach to develop cellular therapy for diabetes mellitus.

文摘The influence of the interaction between surrounding rock and lining on the long-term behaviour of a tunnel in service is significant.In this paper,we proposed a mechanical model of the circular lined tunnel with the alterable mechanical property under hydrostatic stress and radially inner surface pressure of the lining.The alterable mechanical properties of the surrounding rock and the lining are embodied by the changing of their elasticity modulus with service time and radial direction of the tunnel,respectively.The proposed mechanical model is successfully validated by comparison with the existing theoretical models and the numerical simulation,respectively.The influences of the main parameters of the proposed mechanical model,such as the radial power-law indexes and the time-varying coefficients of the surrounding rock and the lining,as well as the radially inner surface pressure of the lining,on the interface displacement and pressure between surrounding rock and lining are investigated.The research results can provide some valuable references for timely diagnosis and correct evaluation of the long-term behaviours of a tunnel in service.

文摘A novel continuous ovary cell line from barfin flounder(Verasper moseri)(BFO cell line) was established with its primitive application in transgenic expression demonstrated in this study. Primarily cultured cells grew well at 22℃ in Dulbecco's modified Eagle medium/F12 medium(DMEM/F12, 1:1; p H 7.2) supplemented with 20% fetal bovine serum(FBS), carboxymethyl chitooligosaccharide, basic fibroblast growth factor(b FGF) and insulin-like growth factor-I(IGF-I). The primary BFO cells in fibroblastic morphology proliferated into a confluent monolayer about 2 weeks later, and were able to be subcultured. Impacts of medium and temperature on the growth of the cells were examined. The optimum growth was found in DMEM/F12 with 20% FBS and at 22℃. The BFO cells can be continuously subcultured to Passage 120 steadily with a population doubling time of 32.7 h at Passage 60. Chromosome analysis revealed that 72% of BFO cells at Passage 60 maintained the normal diploid chromosome number(46) with a normal karyotype of 2st+44t. The results of gene transformation indicated that green fluorescence protein(GFP) positively expressed in these cells after being transformed with pc DNA3.1-GFP. Therefore, a continuous and transformable BFO cell line was successfully established, which may serve as a useful tool for cytotechnological manipulation and transgenic modification of this fish.
文摘The FG 9307 cell line derived from the gill of flounder Paralichthys olivaceus was used in the present study to determine the acute cytotoxic effects of the organophosphorous pesticide, profenofos. It was found that the cell growth rate was markedly reduced by profenofos at the concentrations of 2.5, 5 and 7.5 mg/L tested; and that the fine structures of the cells were also altered by profenofos, as evidenced by dilation of nuclear membranes and mitochondria cristae, and presence of enlarged lysosomes with engulfed organelles and numerous vacuoles in the cytoplasm. Probably, mitochondria, the cell energy generating sites, are the most prominent sites of profenofos cytotoxity in the cells. This seems to be the first report of the use of marine fish cell line for evaluation of the acute in vitro cytotoxicity of organophosphorus pesticide.

文摘A high-efficiency propeller can enable a long mission duration for autonomous underwater vehicles(AUVs).In this study,a new method with OpenProp coupled with computational fluid dynamics was developed to design a propeller for an Explorer100 AUV.The towed system simulation of the AUV was used to measure the nominal wake,and a self-propulsion simulation was used to measure the effective wake at the disc plane just in front of a propeller.Two propellers referring to the nominal wake(propeller 1)and effective wake(propeller 2)were designed with OpenProp and appended with the AUV for self-propulsion simulations,respectively.Through the numerical simulation of the AUV self-propulsion tests,the cruising velocity of AUV was obtained.The flow characteristics of the self-propulsion in pressure and velocity contours were also analyzed.The propeller designed with an effective wake improved the thrust,velocity,and efficiency by approximately 11.3%,6.7%,and 2.5%,respectively,as compared with those with a nominal wake.The cruising velocity of the final designed propeller for the Explorer100 AUV improved by 21.8%,as compared to that of the original propeller from the AUV free-running tests.

文摘Close-distance coal seams are widely distributed over China,and the coal pillars left by the overlying coal seams afect the retracement channel of the underlying coal seam in the stopping stage.Based on the engineering background of close-distance seam mining in a coal mine,the reasonable position of the underlying coal seam's stopping line and the support method of the large section roadway during stopping are investigated using feld measurements,similar simulation experiments,and numerical simulations.There are three types of location relationships between the stopping line of the underlying coal seam and the stopping line of the overlying coal seam:"externally staggered with the upper stopping line"(ESUL,stops mining under the overlying goaf),"overlapped with upper stopping line"(OUL),and"internally staggered with the upper stopping line"(ISUL,ISUL-SD for shorter internal staggered distances,ISUL-LD for longer ones).There are diferent stress arch structures in the overlying strata of the above three positions,and the stress arch evolution process exists in the process of ESUL→OUL→ISUL-SD→ISUL-LD:a front and rear double stress arch structure→the front arch gradually decreases→the front arch dies out,and the double arch synthesizes the single arch→the single-arch range expands→the nested double arch.The relationship between the stress arch structure and the position of the stopping line is evaluated as follows:(1)ESUL:the stress concentration in the roof plate of the retracement channel of the underlying coal seam is the highest,because the overburden block of the extensive collapse zone acts directly on the roof plate of the retracement channel,resulting in relative difculties in roof support.(2)OUL:although the retracement channel roof pressure is minimal,the overlying rock structure has the potential for rotation or slippage instability.(3)ISUL-SD:the pressure on the roof of the retracement channel is small and the overburden structure is stable,which is conducive to the safe retraction of the support and not limited by the width of the end-mining coal pillar.(4)ISUL-LD:it is basically the same as the condition of stopping under the non-goaf;however,it has a limitation on the width of the end-mining coal pillar.The location of the stopping line is selected as ISUL-SD,and the retraction process of the self-excavating retraction channel was adopted.A partition asymmetric support scheme which is proven by feld practice is proposed,through a comprehensive analysis of the pre-stress feld simulation of the support scheme,based on the diferent control requirements of the roof above the support and the roof of the retracement channel in the stopping area.This method realizes safe and smooth withdrawal of the support.

文摘Electrical pollution is a worldwide concern,because it is potentially harmful to human health.Trees not only play a significant role in moderating the climate,but also can be used as shields against electrical pollution.Shielding effects on the electric field strength under transmission lines by two tree species,Populus alba and Larix gmelinii,were examined in this study.The electrical resistivity at different heights of trees was measured using a PiCUS sonic tomograph,which can image the electrical impedance for trees.The electric field strength around the trees was measured with an elf field strength measurement system,HI-3604,and combined with tree resistivity to develop a model for calculating the electric field intensity around trees using the finite element method.In addition,the feasibility of the finite element method was confirmed by comparing the calculated results and experimental data.The results showed that the trees did reduce the electric field strength.The electric field intensity was reduced by 95.6%,and P.alba was better than L.gmelinii at shielding.
文摘In this paper, a hybrid automatic optimization strategy is proposed for the design of underwater robot lines. Isight is introduced as an integration platform. The construction of this platform is based on the user programming and several commercial software including UG6.0, GAMBIT2.4.6 and FLUENT12.0. An intelligent parameter optimization method, the particle swarm optimization, is incorporated into the platform. To verify the strategy proposed, a simulation is conducted on the underwater robot model 5470, which originates from the DTRC SUBOFF project. With the automatic optimization platform, the minimal resistance is taken as the optimization goal;the wet surface area as the constraint condition; the length of the fore-body, maximum body radius and after-body's minimum radius as the design variables. With the CFD calculation, the RANS equations and the standard turbulence model are used for direct numerical simulation. By analyses of the simulation results, it is concluded that the platform is of high efficiency and feasibility. Through the platform, a variety of schemes for the design of the lines are generated and the optimal solution is achieved. The combination of the intelligent optimization algorithm and the numerical simulation ensures a global optimal solution and improves the efficiency of the searching solutions.
文摘The in vitro acute cytotoxicity of the neonicotinoid insecticide acetamiprid to FG cell line, derived from the gill of flounder Paralichthys olizaceus, was tested by the three widely used endpoint bioassays-neutral red (NR) assay, tetrazolium (MTT) assay and cell protein assay. It was found that acetamiprid was increasingly toxic to FG cells at concentrations of 1 μg/cm^3 or above, and the inhibitory concentration 50% values for NR, MTF, and cell protein assays were 38.38, 36.27 and 32.03 μg/cm^3, respectively. This appeared to be the first report on the in vitro cytotoxicity of acetamiprid to non-mammalian vertebrate cells. Ultrastructural examination revealed that for the cells exposed to 60 μg/cm^3 acetamiprid for 48 h, their mitochondria were severely damaged with the cristae swelled up or disrupted, while their nuclei and rough endoplasmic reticlum (RER) appeared to be still normal. This suggests that mitochondria are possibly the primary target of acetamiprid.

文摘Neurogenesis is an important progress wherein the neural stem cells(NSCs) differentiate into functional neurons under conductive conditions. Neurogenesis occurs continuously in different areas of the central neural system in adult teleosts compared with adult mammals. Therefore, NSC cell lines must be established to offer a valuable in vitro system for studies on neurogenesis and other related functions. In this study, four cell lines designated as PoB1, PoB2, PoBf and PoBh were established from the brain of the Japanese flounder Paralichthys olivaceus. The cell lines were sub-cultured over 150 times and still grew well in DMEM/F12 medium at 24℃. PoB1, PoB2, PoBf and PoBh were identified as neural stem/progenitor cell lines on the basis of the mRNA expression of nestin and/or aldh111 or slc6 a4 and the formation of neurospheres. The cells transfected with the pEGFP-C1 plasmid showed fluorescent signals with distinct reagent dependencies. The established cell lines from the brain of P. olivaceus offer a valuable system in vitro for the study of neurogenesis, fish neural regulation and endocrinology-related functions.
文摘In the past decade,several efforts around the world were started with the goal of introducing "smart metering" capabilities into the power grid.These efforts have spurred renewed interest in the design of next generation Narrowband Power Line Communications(NB-PLC) transceivers.In the past few years,ITU-T and IEEE have standardized a family of next generation OFDM-based NB-PLC transceivers some of which are today being considered for massive deployments in Europe and Asia.This paper addresses the important role that PLC has not only for smart metering but also for many other Smart Grid applications,and also gives an overview of the main differences between these next generation NB-PLC standards.

文摘In this paper,the simultaneous resonance of a ferromagnetic thin plate in a time-varying magnetic field,having axial speed and being subjected to a periodic line load,is studied.Based on the large deflection theory of thin plates and electromagnetic field theory,the nonlinear vibration differential equation of the plate is obtained by using the Hamilton′s principle and the Galerkin method.Then the boundary condition in which the longer opposite sides are clamped and hinged is considered.The dimensionless nonlinear differential equations are solved by using the method of multiple scales,and the analytical solution is given.In addition,the stability analysis is also carried out by using Lyapunov stability theory.Through numerical analysis,the variation curves of system resonance amplitude with frequency tuning parameter,magnetic field strength and external excitation amplitude are obtained.Different parameters that have significant effects on the response of the system,such as the thickness,the axial velocity,the magnetic field intensity,the position,and the frequency of external excitation,are considered and analyzed.The results show that the system has multiple solution regions and obvious nonlinear coupled characteristics.

文摘This paper investigates the multiaxial fatigue life of the roller in rolling contact with wheels with respect to axis line deflection. The multiaxial fatigue criteria proposed by Wang and Brown, together with the ralnflow counting method and Miner- Palmgren's rule, are applied to the cumulative damage estimation and life prediction. As the axis line deflection of overlong kilns generally results in asymmetric load distribution on each roller, the load ratio is introduced to describe the deflection for quantitative stress analyses. The stress analyses are performed within the finite element code ANSYS. The tangential friction stress is calculated in terms :of the condition of the rolling contact area. By taking one roller as an example, the plotted fatigue life versus load ratio curve discovers how the axis line deflection affects the fatigue life. This study is significant to prevent the fatigue failure of the roller and can provide basis to adjust and optimize the axis line of the rotary kiln.

文摘The tower line system will be in an unsafe status due to uniform or uneven fall of ice coating which is attached to the surface of tower and lines.The fall of ice could be caused by wind action or thermal force.In order to study the dynamic characteristics of the self-failure of the transmission line under the action of dynamicwind load,a finite elementmodel of the two-span transmission tower line system was established.The birth and death element methods are used to simulate the icing and shedding of the line.Tensile failure strength is the shedding criterion for ice coating.The fluctuating wind speed time history of the tower line systemis first simulated,and then the fluctuating wind and the average wind are superimposed to generate the instantaneous wind speed and converted into wind load.The dynamic response of the transmission tower line systemunder iced coupling with different wind speeds and different thicknesses of ice coating was studied.This is the first attempt that the coupling dynamic response of the icing shedding and wind load for the transmission tower-line system is discussed in this paper.In addition,the dynamic characteristics of wind are included.In particular,the limiting mechanical conditions are considered.According to the simulation results,it is found:because of the ice shedding,the stress of the conductor changes obviously in the first 20 seconds,and the ground wire changes sharply in the first two seconds;the icing of the conductor(ground)wire is gradually deicing under the action of wind vibration;the displacement of tower top increases with the increase of wind speed and icing thickness.
