目的基于氧化低密度脂蛋白(ox-LDL)/人凝集素样氧化低密度脂蛋白受体1(LOX-1)信号通路探讨脂质代谢紊乱促进肺癌进展的机制。方法收集81个已鉴定的具有成对相邻非癌组织(离肿瘤至少5 cm)的肺腺癌组织,使用免疫组织化学检测LOX-1表达。... 目的基于氧化低密度脂蛋白(ox-LDL)/人凝集素样氧化低密度脂蛋白受体1(LOX-1)信号通路探讨脂质代谢紊乱促进肺癌进展的机制。方法收集81个已鉴定的具有成对相邻非癌组织(离肿瘤至少5 cm)的肺腺癌组织,使用免疫组织化学检测LOX-1表达。肺腺癌细胞系(A549、H1299细胞)中过表达LOX-1。用Transwell测定细胞侵袭能力。用不同浓度oxLDL处理细胞,并检测细胞中LOX-1表达情况。结果在包含原发性人肺癌和匹配的邻近非癌组织中,肿瘤中LOX-1染色比非癌组织样品明显增强(中值H分数99.4 vs.16.2,P<0.001)。高LOX-1表达与低生存显著相关(P<0.001)。与无淋巴结转移的患者相比,发生淋巴结转移患者的癌组织具有更高的LOX-1水平(中值H分数83.2 vs.121.1,P<0.01)。LOX-1过表达显著促进肺癌细胞的侵袭转移细胞数(P<0.01)。此外,LOX-1是ox-LDL诱导的肺癌细胞转移所必需的功能靶点。伊他替尼抑制LOX-1过表达的A549在体外的转移能力。结论LOX-1的表达随着oxLDL水平的升高而增加,并且LOX-1的表达上调促进了肺癌细胞的转移,其作用机制可能与激活Janus激酶/转录因子激活子(JAK1/STAT6)信号通路有关。 展开更多

急性脑梗死患者不同性质颈动脉粥样斑块及预后的sdLDL-C、Lp-PLA2水平观察

目的 观察急性脑梗死(ACI)患者的小而密低密度脂蛋白胆固醇(sdLDL-C)、脂蛋白相关磷脂酶A2(Lp-PLA2)水平,探讨其与颈动脉粥样斑块稳定性的关系,以期为早期识别和改善患者预后提供思路。方法 依据颈部血管超声检查粥样斑块性质结果,164... 目的 观察急性脑梗死(ACI)患者的小而密低密度脂蛋白胆固醇(sdLDL-C)、脂蛋白相关磷脂酶A2(Lp-PLA2)水平,探讨其与颈动脉粥样斑块稳定性的关系,以期为早期识别和改善患者预后提供思路。方法 依据颈部血管超声检查粥样斑块性质结果,164例急性脑梗死患者(观察组)中无粥样斑块者49例、稳定粥样斑块者58例、不稳定粥样斑块者57例,以47例健康体检者为对照,比较2组的同型半胱氨酸(Hcy)、sdLDL-C、Lp-PLA2表达水平。统计患者入院时的美国国立卫生院卒中量表(NIHSS)得分,随访患者发病30 d的预后情况,并分析影响ACI预后的相关因素。结果 脑梗死患者的空腹血糖、甘油三酯、sdLDL-C/LDL-C、sdLDL-C/TC、Lp-PLA2水平均高于对照组,HDL-C水平低于对照组;有粥样斑块者的Hcy、sdLDL-C水平高于无粥样斑块者及对照组;不稳定粥样斑块者的TC、LDL-C水平高于无粥样斑块者及对照组,差异均有统计学意义(P<0.05)。预后不良组的不稳定粥样斑块发现率(45.78%)高于预后良好组(23.46%),入院时NIHSS评分及血糖、sdLDL-C、sdLDL-C/LDL-C、sdLDL-C/TC、Lp-PLA2水平均高于预后良好组,差异均有统计学意义(P<0.05)。结论 sdLDL-C、Lp-PLA2水平与急性脑梗死患者颈动脉粥样斑块的稳定性密切相关,对预后判断有积极意义。 展开更多

Atherosclerotic cardiovascular disease is the main cause of mortality and morbidity in the world. Plasma levels of low density lipoprotein cholesterol (LDL-C) are positively correlated with the risk of atherosclerosis. High plasma LDL concentrations in patients with hypercholesterolemia lead to build-up of LDL in the inner walls of the arteries, which becomes oxidized and promotes the formation of foam cells, consequently initiating atherosclerosis. Plasma LDL is mainly cleared through the LDL receptor (LDLR) pathway. Mutations in the LDLR cause familiar hyperch- olesterolemia and increase the risk of premature coronary heart disease. The expression of LDLR is regulated at the transcriptional level via the sterol regulatory element binding protein 2 (SREBP-2) and at the posttranslational levels mainly through proprotein convertase subtilisin/kexin-type 9 (PCSK9) and inducible degrader of the LDLR (IDOL). The effects of oxidized low density lipoprotein (ox-LDL) on the proliferation of cultured human vascular smooth muscle cells (vSMC) were investigated in vitro. By using NaBr density gradient centrifugation, LDL was isolated and purified from human plasma. Ox-LDL was produced from LDL by being incubated with CuSO4. ox-LDL was then added to the culture medium at different concentrations (35, 60, 85, 110, 135 and 160μg/mL) for 7 days. The influence of ox-LDL on vSMC proliferation was observed in growth curve, mitosis index, and in situ determination of apoptosis. The data were analyzed with SPSS 10.0 software. The results showed that the ox-LDL produced in vitro had a good purity and optimal oxidative degree, which was similar to the intrinsic ox-LDL in atherosclerotic plaque, ox-LDL at a concentration of 35 μg/mL demonstrated the strongest proliferation inducement, and at a concentration of 135 μg/mL, ox-LDL could inhibit the growth of vSMC. ox-LDL at concentrations of 35 and 50 μg/mL presented powerful mitotic trigger, and with the increase of ox-LDL concentration, the mitotic index of vSMC was decreased gradually, ox-LDL at higher concentrations promoted more apoptotic vSMCs, ox-LDL at lower concentrations triggered proliferation of vSMCs, and at higher concentrations induced apoptosis in vSMCs, ox-LDL played a promotional role in the pathogenesis and development of atherosclerosis by affecting vSMC proliferation and apoptosis. 展开更多

Apolipoprotein B (apoB) is the main protein component of very low density lipoprotein (VLDL) and is necessary for the assembly and secretion of these triglyceride (TG)-rich particles. Following release from the liver, VLDL is converted to low density lipoprotein (LDL) in the plasma and increased production of VLDL can therefore play a detrimental role in cardiovascular disease. Increasing evidence has helped to establish VLDL assembly as a target for the treatment of dyslipidemias. Multiple factors are involved in the folding of the apoB protein and the formation of a secretion-competent VLDL particle. Failed VLDL assembly can initiate quality control mechanisms in the hepatocyte that target apoB for degradation. ApoB is a substrate for endoplasmic reticulum associated degradation (ERAD) by the ubiquitin proteasome system and for autophagy. Efficient targeting and disposal of apoB is a regu- lated process that modulates VLDL secretion and partitioning of TG. Emerging evidence suggests that significant overlap exists between these degradative pathways. For example, the insulin-mediated targeting of apoB to autop- hagy and postprandial activation of the unfolded protein response (UPR) may employ the same cellular machinery and regulatory cues. Changes in the quality control mechanisms for apoB impact hepatic physiology and pathology states, including insulin resistance and fatty liver. Insulin signaling, lipid metabolism and the hepatic UPR may impact VLDL production, particularly during the postprandial state. In this review we summarize our current understanding of VLDL assembly, apoB degradation, quality control mechanisms and the role of these processes in liver physiology and in pathologic states. 展开更多

目的 采用过氧化物酶法检测并建立郑州地区表观健康人群血清小而密低密度脂蛋白胆固醇(sdLDL-C)活性值参考区间,并进行验证。方法 选取2019年6月至2022年6月在该院特需中心体检健康的785例成年人为研究对象,并按照性别和年龄(18~<30... 目的 采用过氧化物酶法检测并建立郑州地区表观健康人群血清小而密低密度脂蛋白胆固醇(sdLDL-C)活性值参考区间,并进行验证。方法 选取2019年6月至2022年6月在该院特需中心体检健康的785例成年人为研究对象,并按照性别和年龄(18~<30岁、30~<40岁、40~<50岁、50~<60岁和≥60岁)进行分组。采用过氧化物酶法检测血清sdLDL-C活性值,采用Kolmogorov-Smirnov(K-S)法对各组数据进行正态性检验,采用双侧95%CI进行sdLDL-C活性值参考区间上下限的设定。另收集100例体检健康者血清样本进行参考区间验证,当比率值(R)≥90%,判断验证通过。结果 郑州地区表观健康人群sdLDL-C活性值呈正态分布,男性为(0.700±0.151)mmol/L,女性为(0.670±0.137)mmol/L,男女比较,差异有统计学意义(P<0.01)。女性各年龄组间血清sdLDL-C活性值比较,差异无统计学意义(P>0.05),男性各年龄组间血清sdLDL-C活性值比较,差异有统计学意义(P<0.05),进一步分析显示18~<30岁组、30~<40岁组和40~<50岁组间差异无统计学意义(P>0.05),50~<60岁组与≥60岁组间差异无统计学意义(P>0.05),因此将男性合并为18~<50岁组和≥50岁组。郑州地区表观健康人群血清sdLDL-C活性值参考区间为女性(18~80岁)0.400~0.938 mmol/L、男性(18~<50岁)0.440~1.004 mmol/L、男性(≥50岁)0.368~0.976 mmol/L。100例体检健康者的比率值为0.98,新建立的参考区间验证通过。结论 该研究初步建立和验证了郑州地区表观健康人群血清sdLDL-C活性值参考区间,为心血管疾病的发病风险评估以及诊断治疗提供参考依据。 展开更多

目的探讨25羟基维生素D、同型半胱氨酸(homocysteine,Hcy)和低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)对结直肠腺瘤的预测价值。方法选取2021年6月至2023年1月于新疆维吾尔自治区人民医院消化内科住院行结肠镜检... 目的探讨25羟基维生素D、同型半胱氨酸(homocysteine,Hcy)和低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)对结直肠腺瘤的预测价值。方法选取2021年6月至2023年1月于新疆维吾尔自治区人民医院消化内科住院行结肠镜检查的患者为研究对象,将患者依据结肠镜检查及病理结果纳入病例组(189例)和对照组(207例),检测并比较两组间25羟基维生素D、Hcy、LDL-C水平,以及三者联合检测对结直肠腺瘤的预测价值。结果血清25羟基维生素D(OR=0.940)、Hcy(OR=1.057)和LDL-C(OR=1.578)均为腺瘤性息肉的独立影响因素,25羟基维生素D、Hcy和LDL-C三者联合检测对腺瘤性息肉的预测价值最大(AUC=0.737),优于单独检测及两两联合检测。25羟基维生素D、Hcy、LDL-C预测合并结直肠腺瘤风险的临界值分别为23.5 ng/mL、17.5μmol/L、3.85 mmol/L。结论25羟基维生素D水平低于23.5 ng/mL、Hcy高于17.5μmol/L、LDL-C水平高于3.85 mmol/L的患者属于合并结直肠腺瘤的高风险人群,三者联合检测对结直肠腺瘤的预测价值最大。 展开更多

In order to investigate the roles of Wnt signal pathway in transformation of cardiac valvular myofibroblasts to the osteoblast-like phenotype, the primary cultured porcine aortic valve myofibroblasts were incubated with oxidized low density lipoprotein(ox-LDL, 50 mg/L), and divided into four groups according to the ox-LDL treatment time: control group, ox-LDL 24-h group, ox-LDL 48-h group, and ox-LDL 72-h group. Wnt signal pathway blocker Dickkopf-1(DDK-1, 100 μg/L) was added in ox-LDL 72-h group. The expression of α-smooth muscle actin(α-SMA), bone morphogenetic protein 2(BMP2), alkaline phosphatase(ALP), and osteogenic transcription factor Cbfa-1 was detected by Western blotting, and that of β-catenin, a key mediator of Wnt signal pathway by immunocytochemical staining method. The Wnt/β-catenin was observed and the transformation of myofibroblasts to the osteoblast-like phenotype was examined. The expression of α-SMA, BMP2, ALP and Cbfa-1 proteins in the control group was weaker than in the ox-LDL-treated groups. In ox-LDL-treated groups, the protein expression of α-SMA, BMP2, ALP, and Cbfa-1 was significantly increased in a time-dependent manner as compared with the control group, and there was significant difference among the three ox-LDL-treated groups(P〈0.05 for all); β-catenin protein was also up-regulated in the ox-LDL-treated groups in a time-dependent manner as compared with the control group(P〈0.05), and its transfer from cytoplasm to nucleus and accumulation in the nucleus were increased in the same fashion(P〈0.05). After addition of DKK-1, the expression of α-SMA, bone-related proteins and β-catenin protein was significantly reduced as compared with ox-LDL 72-h group(P〈0.05). The Wnt/ β-catenin signaling pathway may play an important role in transformation of valvular myofibroblasts to the osteoblast-like phenotype. 展开更多

Objective: To investigate the molecular mechanisms and effective target ponits of lipid-lowering drug, Rhizoma Curcumae Longae, and study the effect of curcumin on the expression of low density lipoprotein (LDL) receptors in macrophages in mice. Methods. Macrophages in mice were treated with curcumin, which was purified from the ethanolly extraction of Rhizoma Curcumae Longae for 24 h. The LDL receptors expressed in the macrophages were determined by enzyme-linked immunosorbent assay (ELISA) and assay of Dil labeled LDL uptake by flow cytometer. Results: It was found for the first time that 10 μmol/L-50 μmol/L curcumin could obviously up-regulate the expression of LDL receptor in macrophages in mice, and a dose-effect relationship was demonstrated. Conclusion: One of the lipid-lowering mechanisms of traditional Chinese medicine, Rhizoma Curcumae Longae, was completed by the effect of curcumin through the up-regulation of the expression of LDL receptor. 展开更多

Summary:In the present study, we examined the regulation of the expression and function of ABCA1 by modified LDL (ox-LDL) in vitro. After incubation with apoA-I for 24 h, RAW264.7 cells effluxed 37.65 % cholesterol loaded by acetyl LDL (ac-LDL), and 9.78 % cholesterol in ox-LDL group. The level of ABCA1 mRNA increased about three times either when cells were incubated with 100 μg /mL ac-LDL or with 100 μg /mL ox-LDL. However, the level of ABCA1 protein rose by 1.57 times in ac-LDL group and 1.26 times in ox-LDL group. These results demonstrated that ox-LDL had different effect on the expression and function of ABCA1, ox-LDL might decrease the cholesterol efflux mediated by ABCA1 through other unknown mechanisms. 展开更多

目的分析血清激肽释放酶6(KLK6)、CC趋化因子受体2(CCR2)、氧化修饰低密度脂蛋白(ox-LDL)与帕金森病的相关性。方法将2020年7月至2022年12月于该院诊治的帕金森病患者150例纳入研究作为患者组,按照Hoehn-Yahr(H-Y)分期进一步分为Ⅰ期27... 目的分析血清激肽释放酶6(KLK6)、CC趋化因子受体2(CCR2)、氧化修饰低密度脂蛋白(ox-LDL)与帕金森病的相关性。方法将2020年7月至2022年12月于该院诊治的帕金森病患者150例纳入研究作为患者组,按照Hoehn-Yahr(H-Y)分期进一步分为Ⅰ期27例、Ⅱ期42例、Ⅲ期47例、Ⅳ期34例。另外,选取同期健康体检者150例作为对照组。采用简易精神状态检查(MMSE)量表评估患者精神障碍情况。比较患者组与对照组及不同H-Y分期帕金森病患者血清KLK6、CCR2、ox-LDL水平。分析血清KLK6、CCR2、ox-LDL对帕金森病的诊断价值。分析血清KLK6、CCR2、ox-LDL与H-Y分期、MMSE评分的相关性。结果患者组血清KLK6、CCR2、ox-LDL水平高于对照组(P<0.05)。受试者工作特征(ROC)曲线分析显示,KLK6、CCR2、ox-LDL诊断帕金森病的曲线下面积(AUC)分别为0.813、0.847、0.826,最佳临界值对应的灵敏度、特异度:KLK6为66.7%、90.0%,CCR2为68.0、91.3%,ox-LDL为59.3%、100.0%。不同H-Y分期患者血清KLK6、CCR2、ox-LDL比较:Ⅰ期<Ⅱ期<Ⅲ期<Ⅳ期;MMSE评分比较:Ⅰ期>Ⅱ期>Ⅲ期>Ⅳ期;两两比较差异均有统计学意义(P<0.05)。血清KLK6、CCR2、ox-LDL水平与H-Y分期均呈正相关(r=0.559、0.716、0.722,P<0.05);血清KLK6、CCR2、ox-LDL水平与MMSE评分均呈负相关(r=-0.276、-0.448、-0.457,P<0.05)。结论血清KLK6、CCR2、ox-LDL对帕金森病患者具有一定的诊断价值,并且与帕金森病患者的病情严重程度和认知功能有关。 展开更多

目的研究恩格列净(EMP)对氧化低密度脂蛋白(ox-LDL)诱导内皮祖细胞(EPCs)损伤的保护作用及机制。方法通过密度梯度离心法提取、分离并培养小鼠骨髓来源的的EPCs。采用Dil标记乙酰化低密度脂蛋白(Dil-ac-LDL)联合FITC标记荆豆凝集素-1(FI... 目的研究恩格列净(EMP)对氧化低密度脂蛋白(ox-LDL)诱导内皮祖细胞(EPCs)损伤的保护作用及机制。方法通过密度梯度离心法提取、分离并培养小鼠骨髓来源的的EPCs。采用Dil标记乙酰化低密度脂蛋白(Dil-ac-LDL)联合FITC标记荆豆凝集素-1(FITC-UEA-1)双摄取法鉴定。将EPCs分为正常对照组,ox-LDL组以及ox-LDL联合不同浓度恩格列净实验组。CCK-8检测细胞活力,Transwell检测细胞迁移,FITC-Annexin V/PI检测细胞凋亡,ELISA检测细胞上清液中血管内皮细胞生长因子(VEGF)、基质细胞衍生因子-1α(SDF-1α)含量;流式细胞术检测一氧化氮(NO)的合成情况。Western blot检测AMPK、p-AMPK、eNOS、p-eNOS的蛋白表达。结果提取的EPCs诱导培养至第7天经鉴定为小鼠骨髓EPCs。与对照组比较,ox-LDL组细胞活力降低,迁移细胞减少,凋亡增加,VEGF、SDF-1α含量降低,NO合成减少(P<0.05);与ox-LDL组相比,不同浓度恩格列净组细胞活力有所提高,迁移细胞增多,凋亡减少,VEGF、SDF-1α含量升高,NO合成增加(P<0.05)。ox-LDL处理可明显抑制AMPK及eNOS磷酸化(P<0.05),恩格列净处理可以改善AMPK及eNOS磷酸化水平(P<0.05),而AMPK抑制剂Compound C可使恩格列净改善EPCs功能活性的作用受到明显的抑制(P<0.05)。结论恩格列净可改善ox-LDL诱导的EPCs功能障碍,其机制与调控AMPK/eNOS信号通路有关。 展开更多

目的探究氧化低密度脂蛋白(oxLDL)通过CXCL16/ERK1/2信号通路与川崎病(KD)冠状动脉损伤(CAL)的关系。方法构建KD的细胞模型,将实验分为3组:正常对照组(Con组)、KD冠状动脉正常组(NCAL组,加入10%NCAL患者血清)、KD冠状动脉损伤组(CAL组,... 目的探究氧化低密度脂蛋白(oxLDL)通过CXCL16/ERK1/2信号通路与川崎病(KD)冠状动脉损伤(CAL)的关系。方法构建KD的细胞模型,将实验分为3组:正常对照组(Con组)、KD冠状动脉正常组(NCAL组,加入10%NCAL患者血清)、KD冠状动脉损伤组(CAL组,加入10%CAL患者血清)。细胞培养24 h后,利用Elisa法检测细胞上清液中oxLDL的表达水平;另据公共数据库筛选oxLDL对KD病情的潜在相关靶点,用RT-qPCR检测CXCL16的表达水平;采用Western Blot技术检测细胞蛋白CXCL16、p-ERK的表达水平;通过CCK8试验和Edu法评估内皮细胞的活力和增殖活性,利用内皮细胞划痕试验检测细胞迁移能力。运用ERK1/2抑制剂(SCH772984)干预ERK1/2表达,建立了四个不同处理组别:NCAL组、NCAL+SCH组、CAL组、CAL+SCH组,并通过Western Blot技术检测CXCL16、p-ERK的表达水平变化。结果与Con组相比,NCAL组中oxLDL和CXCL16的表达水平较高,而在CAL组中这一趋势更为显著(P<0.05)。相较于Con组,NCAL组中的CXCL16、p-ERK的表达呈上升趋势,而在CAL组中这种趋势进一步增强(P<0.05)。CAL组内皮细胞死亡明显,CCK-8检测显示CAL患者血清刺激后细胞活性较低(P<0.05),Edu检测结果显示CAL组的Edu阳性细胞显著减少(P<0.05),细胞划痕愈合速度减缓(P<0.05)。结果显示抑制ERK1/2组在抑制ERK1/2表达后CXCL16的表达明显下降(P<0.05)。结论oxLDL可能透过CXCL16/ERK1/2信号通路加重内皮细胞功能受损,从而引发KD患者CAL的发生。 展开更多

目的 探讨低密度脂蛋白胆固醇(LDL-C)/高密度脂蛋白胆固醇(HDL-C)对急性非ST段抬高型急性心肌梗死(NSTEMI)患者介入治疗预后的预测价值。方法 选择医院2021年6月至2022年6月收治的105例接受经皮冠状动脉介入治疗的NSTEMI患者为研究对象... 目的 探讨低密度脂蛋白胆固醇(LDL-C)/高密度脂蛋白胆固醇(HDL-C)对急性非ST段抬高型急性心肌梗死(NSTEMI)患者介入治疗预后的预测价值。方法 选择医院2021年6月至2022年6月收治的105例接受经皮冠状动脉介入治疗的NSTEMI患者为研究对象,所有患者入院时记录一般资料,采集血液标本,检测血清LDL-C、HDL-C水平,并计算LDL-C/HDL-C值。介入治疗后均进行为期1 a的随访,观察患者预后情况,采用logistic回归分析NSTEMI患者介入治疗预后的危险因素,并绘制受试者工作特征(ROC)曲线,检验LDL-C/HDL-C对NSTEMI患者介入治疗预后的预测价值。结果 105例NSTEMI患者介入治疗出院后被随访1 a, 25例(23.81%)发生主要心血管不良事件,纳入预后不良组,80例未发生主要心血管不良事件,纳入预后良好组。预后不良组血清cTnT、LDL-C、LDL-C/HDL-C水平均高于预后良好组,HDL-C水平低于预后良好组(P<0.05);经logistic回归分析,cTnT、LDL-C、LDL-C/HDL-C是NSTEMI患者介入治疗预后不良的危险因素(OR>1,P<0.05),HDL-C是NSTEMI患者介入治疗预后不良的保护因素(OR<1,P<0.05)。LDL-C、HDL-C、LDL-C/HDL-C预测NSTEMI患者介入治疗预后情况的曲线下面积(AUC)分别为0.677、0.795、0.808。结论 LDL-C/HDL-C与NSTEMI患者介入治疗预后情况密切相关,其水平上调可提示介入治疗预后不良发生风险增加。 展开更多

Monocyte chemoattractant protein-1(MCP-1), a potent chemoattractant, is thought to play an important role in migration of monocytes into atherosclerotic lesions. The present study was designed to investigate the capacity of human peripheral blood monocytes to express MCP-1 and effects of native very low density lipoprotein (VLDL) and oxidized VLDL(OX-VLDL) on the expression. The total RNA was extracted from cultured monocytes, which were exposed to VLDL and OX-VLDL, and the media conditioned by monocytes were collected. MCP-1 mRNA expression was examined by Northern blot analysis. MCP-1 protein in conditioned media was determined by using sandwich ELISA. The results showed that monocytes can express MCP-1 after a 24 h incubation at 37℃,and the expression was markedly increased by a exposure to OX-VLDL, whereas the expression was slightly increased when exposed to VLDL. It suggests that the capacity of monocytes to produce MCP-1 that recruits and activates circulating monocytes may be of considerable importance in atherogenesis, and oxidation of VLDL enhances its potential to promote atherogenesis. 展开更多

To explore the functions of very low density lipoprotein receptor (VLDL-R) subtype II in lipoprotein metabolism and foam cells formation, the recombinant plasmid with the two subtypes cDNA was constructed respectively, the ldl-A7 cell lines were transfected and two cell lines expressing VLDL-R were obtained: one stably expressing the VLDLR with the O-linked sugar region (type I VLDLR) and the other without the O-linked sugar region (type II VLDLR). In the study on binding of VLDLR to their nuclein labeled natural ligands (VLDL and β-VLDL), it was found that surface binding of 125I-VLDL or 125I-β-VLDL of ldl-A7 cells transfected with type I VLDLR recombinant (ldl-A7-VRI) was more higher than that of ldl-A7 cells transfected with type II VLDLR recombinant (ldl-A7-VRII). After being incubated with VLDL for different time, the contents of triglyceride and total cholesterol in cells were mensurated, and the formation of foam cells and accumulation of lipid in cells was observed by oil-red O staining. The results showed that the contents of triglyceride and total cholesterol in ldl-A7-VR I were much higher than those in ldl-A7-VR II, and ldl-A7-VR I could transform into foam cells notably. It was suggested that type I VLDLR binds with relative higher affinity to VLDL and β-VLDL, and internalizes much more lipoprotein into cells. As a result, we can conclude that type I VLDLR plays a more important role in lipoprotein metabolism and foam cells formation than type II VLDLR. 展开更多
