[Objectives]This study was conducted to establish a rapid and effective method for simultaneous extraction of 54 kinds of veterinary drug residues in animal-derived food, including sulfonamides, quinolones, tetracycli...[Objectives]This study was conducted to establish a rapid and effective method for simultaneous extraction of 54 kinds of veterinary drug residues in animal-derived food, including sulfonamides, quinolones, tetracyclines, malachite greens, penicillins, nitroimidazoles, tranquilizers and macrolides, by HPLC-MS. [Methods] The samples were extracted with 80% acetonitrile water(containing 0.1% formic acid), combined with QuEChERS extraction technology and C18 and PSA purification, analyzed by high performance liquid chromatography-mass spectrometry, and quantified by external standard method. The target substances were analyzed on ZORBAX Eclipse C18 chromatographic column using 0.2% formic acid water and 0.2% methanol as mobile phases. The gradient elution mode was used for chromatographic separation and multiple reaction detection. [Results] In the linear range of 0.5-50.0 ng/ml, the linear relationship of the 54 kinds of veterinary drug residues was good, with correlation coefficients(r~2) greater than 0.995, and the detection limits ranged from 0.30 to 1.00 μg/kg. The results showed that the recovery ranged from 75.4% to 118.2% when different matrixes were added for recovery. [Conclusions] This method is simple, efficient, accurate, stable, and highly operable. It is applicable to simultaneous batch screening of veterinary drug residues in animal-derived food, and has high practical application value.展开更多

By the determination of the metal residues in the samples, the calculated actual daily exposure and concentration of the metal Pd, Ir, Rh, Ru, Mo, Ni, Cr, V, Cu, Mn, Fe and Zn that were less than the permitted daily exposures (PDE) and the limited concentration permitted in the EMEA guideline on the specification limits for residues of metal catalysts or metal reagents [1] . The metal residues can de adequately removed from the active pharmaceutical ingredients and the corresponding drugs. The redox behavior of spironolactone(SP) drug in Britton-Robinson(BR) buffer of pH 2-11 was investigated by differential pulse cathodic stripping voltammetry(DPCSV) and cyclic voltammetry(CV) at hanging mercur...The redox behavior of spironolactone(SP) drug in Britton-Robinson(BR) buffer of pH 2-11 was investigated by differential pulse cathodic stripping voltammetry(DPCSV) and cyclic voltammetry(CV) at hanging mercury dropping electrode(HMDE).At pH 9-10.5,the DPCSV of SP drug showed two cathodic peaks at1.15 and1.38 V at the HMDE vs.Ag/AgCl reference electrode.In the CV,at pH 9-10,the dependence of the cathodic peak current,Ip,c and peak potential,Ep,c of the second peak(Ep,c2) on the scan rate(n) and on the depolizer(SP) concentrations was typical of an electrode coupled(EC) chemical reaction type mechanism.The plot of Ip,cat 1.380 V of the DPCSV vs.SP concentration at pH 9 was linear over the concentration range of 1.2×1010-9.6×107 M.The lower limit of detection(LLOD) and limit of quantification(LOQ) of the drug were 1.1×1011 and 4.14×1011 M,respectively.The method was successfully applied for the analysis of SP residues in industrial wastewater,in pure form(98.273.1%) and in drug formulations e.g.Aldactones tablet(98.3572.9%).The method was validated by comparison with HPLC and the official data methods.展开更多

[Method]Total three different concentrations of cimaterol were administered to pigs, and the residue amounts of cimaterol in pig hair, urine and flesh were monitored at different raising stage. [Result] During the administration period, the residue amount of cimaterol was highest in urine, so urine is the suitable sample for rapid detection of cimaterol in manufacturing enterprises. Cimaterol could be accumulated in pig hair,where cimaterol was metabolized slowly. Thus, pig hair can be used as the test sample for tracing illegal use of veterinary drugs and for vivo detection. Flesh can be used as test sample for direct judgment whether cimaterol residue exceeds the relevant standard. [Conclusion] This study will provide certain theoretical basis for drug monitor in animal husbandry.展开更多
In drug discovery research, residual solvent measurement is an integral part of purity analysis for synthesis of a drug candidate before it is used for toxicity testing. This is usually carried out using gas chromatog...In drug discovery research, residual solvent measurement is an integral part of purity analysis for synthesis of a drug candidate before it is used for toxicity testing. This is usually carried out using gas chromatography(GC)with direct injection sample introduction. This method requires testing compounds to be soluble at high concentrations( > 50 mg/mL, usually in DMSO) to achieve acceptable sensitivity, a hurdle which is not always achievable for some samples such as cyclic peptides and oligonucleotides. To overcome the limitation associated with the direct injection approach, a new method using the Chromatoprobe thermal extraction device was developed for quantifying residual solvents of drug discovery compounds. This method not only consumes significantly less material(less than 1 mg), but also shows higher sensitivity than the direct injection approach.In addition, because no diluent is required with the Chromatoprobe thermal extraction, all residual solvents can be detected and measured without further method optimization. In our study, we compared data from GC residual solvent analysis using the Chromatoprobe solid sample introduction to those of the direct injection method for seven in-house samples. Our results showed a good agreement between the data from these two sample introduction methods. Thus, the Chromatoprobe sample introduction method provided a samplesparing alternative to the direct injection method for the measurement of residual solvents in drug discovery.This method can be particularly useful for residual solvent analysis in samples that are available only in limited amounts, poorly soluble, and/or unstable in the diluents used for the direct injection method.展开更多

Amodel that can obtain rapid and accurate detection of coronavirus disease 2019(COVID-19)plays a significant role in treating and preventing the spread of disease transmission.However,designing such amodel that can ba...Amodel that can obtain rapid and accurate detection of coronavirus disease 2019(COVID-19)plays a significant role in treating and preventing the spread of disease transmission.However,designing such amodel that can balance the detection accuracy andweight parameters ofmemorywell to deploy a mobile device is challenging.Taking this point into account,this paper fuses the convolutional neural network and residual learning operations to build a multi-class classification model,which improves COVID-19 pneumonia detection performance and keeps a trade-off between the weight parameters and accuracy.The convolutional neural network can extract the COVID-19 feature information by repeated convolutional operations.The residual learning operations alleviate the gradient problems caused by stacking convolutional layers and enhance the ability of feature extraction.The ability further enables the proposed model to acquire effective feature information at a lowcost,which canmake ourmodel keep smallweight parameters.Extensive validation and comparison with other models of COVID-19 pneumonia detection on the well-known COVIDx dataset show that(1)the sensitivity of COVID-19 pneumonia detection is improved from 88.2%(non-COVID-19)and 77.5%(COVID-19)to 95.3%(non-COVID-19)and 96.5%(COVID-19),respectively.The positive predictive value is also respectively increased from72.8%(non-COVID-19)and 89.0%(COVID-19)to 88.8%(non-COVID-19)and 95.1%(COVID-19).(2)Compared with the weight parameters of the COVIDNet-small network,the value of the proposed model is 13 M,which is slightly higher than that(11.37 M)of the COVIDNet-small network.But,the corresponding accuracy is improved from 85.2%to 93.0%.The above results illustrate the proposed model can gain an efficient balance between accuracy and weight parameters.展开更多
