BACKGROUND In vitro expansion to increase numbers of hematopoietic stem cells(HSCs)in cord blood could improve clinical efficacy of this vital resource.Nicotinamide(NAM)can promote HSC expansion ex vivo,but its effect...BACKGROUND In vitro expansion to increase numbers of hematopoietic stem cells(HSCs)in cord blood could improve clinical efficacy of this vital resource.Nicotinamide(NAM)can promote HSC expansion ex vivo,but its effect on hematopoietic stem and progenitor cells(HSPCs,CD34^(+)CD38)and functional subtypes of HSCs-shortterm repopulating HSCs(ST-HSCs,CD34^(+)CD38CD45RACD49f^(+))and long-term repopulating HSCs(LT-HSCs,CD34^(+)CD38CD45RACD49f^(+)CD90^(+))is not yet known.As a sirtuin 1(SIRT1)inhibitor,NAM participates in regulating cell adhesion,polarity,migration,proliferation,and differentiation.However,SIRT1 exhibits dual effects by promoting or inhibiting differentiation in different tissues or cells.We propose that the concentration of NAM may influence proliferation,differentiation,and SIRT1 signaling of HSCs.AIM To evaluate the effects and underlying mechanisms of action of different concentrations of NAM on HSC proliferation and differentiation.METHODS CD34^(+)cells were purified from umbilical cord blood using MacsCD34 beads,and cultured for 10-12 d in a serum-free medium supplemented with cytokines,with different concentrations of NAM added according to experimental requirements.Flow cytometry was used to detect phenotype,cell cycle distribution,and apoptosis of the cultured cells.Real-time polymerase chain reaction was used to detect the transcription levels of target genes encoding stemness-related factors,che mokines,components of hypoxia pathways,and antioxidant enzymes.Dichloro-dihydro-fluorescein diacetate probes were used to evaluate intracellular production of reactive oxygen species(ROS).Determination of the effect of different culture conditions on the balance of cytokine by cytometric bead array.RESULTS Compared with the control group,the proportion and expansion folds of HSPCs(CD34^(+)CD38)incubated with 5 mmol/L or 10 mmol/L NAM were significantly increased(all P<0.05).The ST-HSCs ratio and fold expansion of the 5 mmol/L NAM group were significantly higher than those of the control and 10 mmol/L NAM groups(all P<0.001),whereas the LT-HSCs ratio and fold expansion of the 10 mmol/L NAM group were significantly higher than those of the other two groups(all P<0.05).When the NAM concentration was>10 mmol/L,cell viability significantly decreased.In addition,compared with the 5 mmol/L NAM group,the proportion of apoptotic cells in the 10 mmol/L NAM group increased and the proportion of cells in S and G2 phase decreased.Compared with the 5 mmol/L NAM group,the HSCs incubated with 10 mmol/L NAM exhibited significantly inhibited SIRT1 expression,increased intracellular ROS content,and downregulated expression of genes encoding antioxidant enzymes(superoxide dismutase 1,peroxiredoxin 1).CONCLUSION Low concentrations(5 mmol/L)of NAM can better regulate the balance between proliferation and differentiation,thereby promoting expansion of HSCs.These findings allow adjustment of NAM concentrations according to expansion needs.展开更多

Millimeter-sized epoxy resin polymer(ERP) cylindrical lens array concentrators are fabricated by the soft imprint technique based on polydimethylsiloxane stamps. The photovoltaic measurements show that millimeter-sized ERP cylindrical lens array concentrators can considerably improve the power conversion efficiency of silicon solar cells. The validity of the proposed method is proved by the coupled optical and electrical simulations. The designed solar cell devices with the advantages of high-efficiency and convenient cleaning are very useful in practical applications.展开更多

The effects of the thickness and the radius of the metal plate, and the air environment on the temperature of the solar cells were analyzed in detail. It is shown that the temperature of the solar cells decreased sharply at the beginning, with the increase in the thickness of the metal plate, and then changed more smoothly. When the radius of the metal plate was 4 cm and the thickness increased to 2 mm or thicker, the temperature of the solar cell basically stabilized at about 53℃. Increasing the radius of the metal plate and the convective transfer coefficient made the temperature of the solar cell decrease remarkably. The effects of A1 and Cu as the metal plate material on cooling were analyzed contrastively, and demonstrated the superiority of A1 material for the cooling system. Furthermore, considering cost reduction, space holding and the stress of the system, we optimized the structural design of the metal plate. The simulated results can be referred to the design of the structure for the metal plate. Finally, a method to devise the structure of the metal plate for single concentrator solar cells was given.展开更多

The proposed SCAM consisted mainly of a silicon-wafer strip-cell sparse array and low-concentration- ratio array concentrator based on an epoxy resin polymer (ERP) cylindrical piano-convex lens. A polymer replication process based on a polydimethylsiloxane mold was used to fabricate the ERP lens array concentrator. The results show that 46.94% of the silicon-wafer cell was saved in the designed SCAM. Moreover, the output power of the SCAM with a low concentration ratio of 8 suns was improved by 8.6%, compared with a whole piece of a conventional silicon-wafer solar cell with the same area as the module. The proposed method encapsulating solar cells provides a means to reduce the usage of silicon cells in modules as well as improving the output power of modules.展开更多

Pancreatic islet cells, which were isolated from Kunming mice, were cultured with different concentrations of glucose in DMEM, and divided into the following groups: G1, G2, G3, G4, G5, and G6 groups, corresponding to the glucose concentrations of 5.6, 7.8, 11.1, 16.7, 22.5, and 27.6 mmol/L, respectively. After culture for 120 h, insulin secretion was evaluated by radioimmunoassay, and the NF-rd3 expression was detected by immunocytochemistry. Bax activity and Cyt C release were measured by immunofluorescence, and apoptosis was examined by Hoechst33342 assay. The results showed that in GI, G2 and G3 groups, insulin secretion was enhanced with the increase of glucose concentration, and the NF-κB expression was also increased (P〈0.05), but Bax activity, Cyt C release and apoptosis rate showed no significant difference among them. However, in G4, G5, and G6 groups, apoptosis rate of islet cells, NF-rd3 expression, Bax activity, and Cyt C release were all significantly increased, and insulin secretion was impaired as compared with G1, G2, and G3 groups (P〈0.05). It was concluded that the exposure of islet cells to high glucose could induce islet cells apoptosis as well as impaired insulin secretion. The NF-κB signaling pathway and mitochondria pathway in islet cells might play some roles in the progressive loss of islet cells in diabetes. The inhibition of the NF-κB expression could be an effective strategy for protecting pancreatic islet cells.展开更多

In this paper, we focused on concentrated photovoltaic cells (CPV) which are promising ways of converting solar energy to electricity. However, it is not well known how serum factors affect the adhesion of monocytes. Methods: We have studied the effect of fetal calf serum (FCS), which we considered a source of LDL, on the adhesion of monocytes to endothelial cells (ECs) by using human monocytic THP-1 cells and both a monolayer of cultured bovine aortic endothelial cells (EC monoculture) and a co-culture with bovine aortic smooth muscle cells (EC-SMC co-culture). Results: It was found that the addition of FCS to the medium greatly affected the adhesion of THP-1 cells, and the higher the concentration of FCS in the medium, the greater the adhesion of THP-1 cells to endothelial cells. Adhesion of THP-1 cells to an EC-SMC co-culture was approximately twofold greater than that to an EC monoculture, and after adhering to endothelial cells, many THP-1 cells trans-migrated into the layer of smooth muscle cells. Conclusion: The results suggest that the elevation of the LDL (cholesterol) level in blood provides a favorable condition for the development of intimal hyperplasia and atherosclerosis by promoting the adhesion of monocytes to the endothelium and their subsequent migration into subendothelial spaces.展开更多
Objective The main purpose of this study was to investigate whether the folliculo stellate cells (FSC) respond to angiotensin(Ang)Ⅱ by increasing intracellular free Ca 2+ concentration ([Ca 2+ ]i),and where the origi...Objective The main purpose of this study was to investigate whether the folliculo stellate cells (FSC) respond to angiotensin(Ang)Ⅱ by increasing intracellular free Ca 2+ concentration ([Ca 2+ ]i),and where the origin of Ca 2+ mobilization is if that has occurred.Methods Pituitary cells in primary culture were prepared from male Wister rats(250g) by a conventional method and cultured in MEM supplemented with 4% normal rat serum.After 2 days in culutre,cells were loaded with 1 μmol/L fura PE3/AM for 1 h and subjected to a Ca 2+ imaging experiment with Quanti Cell 700 system.Excitation wavelengths of 340 and 380 nm were selected by means of a computer controlled filterwheel.Results The [Ca 2+ ]i of FSC in the rat anterior pituitary was elevated by Ang Ⅱ.The elevation of [Ca 2+ ]i of FSC induced by 0.1,1.0,10 and 100 nmol/L Ang Ⅱ was (56.33±6.18)( ±s ),(117.07±36.07),(175.59±40.01) and (216.02±11.52) nmol/L,respectively.The increase of [Ca 2+ ]i of FSC induced by 100 nmol/L Ang Ⅱ was not influenced by the medium without Ca 2+ (0Ca),but significantly suppressed by thapsigargin(TG),an inhibitor of ATPase.The rate of responsive FSC to Ang Ⅱ (100 nmol/L) was 61.84% which was obviously higher than that of pituitary endocrine cells(43.49%).Conclusion The present experiment demonstrates that the FSC in the rat anterior pituitary responds to Ang Ⅱ by increasing [Ca 2+ ]i,which raises the possibility that Ang Ⅱ released from either lactotrophs or gonadotrophs affects FSC through paracrine mechanism.The elevation of [Ca 2+ ]i induced by Ang Ⅱ presents a dosage dependent relation, and is possibly because of the release of Ca 2+ from an intracellular Ca 2+ pool(s).Fashions of Ca 2+ release are relative to the concentration of Ang Ⅱ.The results indicate that Ang Ⅱ functions as a paracrine factor among pituitary cells including FSC.展开更多
BACKGROUND There is still no consensus on which concentration of mesenchymal stem cells(MSCs)to use for promoting fracture healing in a rat model of long bone fracture.AIM To assess the optimal concentration of MSCs f...BACKGROUND There is still no consensus on which concentration of mesenchymal stem cells(MSCs)to use for promoting fracture healing in a rat model of long bone fracture.AIM To assess the optimal concentration of MSCs for promoting fracture healing in a rat model.METHODS Wistar rats were divided into four groups according to MSC concentrations:Normal saline(C),2.5×10^(6)(L),5.0×10^(6)(M),and 10.0×10^(6)(H)groups.The MSCs were injected directly into the fracture site.The rats were sacrificed at 2 and 6 wk post-fracture.New bone formation[bone volume(BV)and percentage BV(PBV)]was evaluated using micro-computed tomography(CT).Histological analysis was performed to evaluate fracture healing score.The protein expression of factors related to MSC migration[stromal cell-derived factor 1(SDF-1),transforming growth factor-beta 1(TGF-β1)]and angiogenesis[vascular endothelial growth factor(VEGF)]was evaluated using western blot analysis.The expression of cytokines associated with osteogenesis[bone morphogenetic protein-2(BMP-2),TGF-β1 and VEGF]was evaluated using real-time polymerase chain reaction.RESULTS Micro-CT showed that BV and PBV was significantly increased in groups M and H compared to that in group C at 6 wk post-fracture(P=0.040,P=0.009;P=0.004,P=0.001,respectively).Significantly more cartilaginous tissue and immature bone were formed in groups M and H than in group C at 2 and 6 wk post-fracture(P=0.018,P=0.010;P=0.032,P=0.050,respectively).At 2 wk post fracture,SDF-1,TGF-β1 and VEGF expression were significantly higher in groups M and H than in group L(P=0.031,P=0.014;P<0.001,P<0.001;P=0.025,P<0.001,respectively).BMP-2 and VEGF expression were significantly higher in groups M and H than in group C at 6 wk postfracture(P=0.037,P=0.038;P=0.021,P=0.010).Compared to group L,TGF-β1 expression was significantly higher in groups H(P=0.016).There were no significant differences in expression levels of chemokines related to MSC migration,angiogenesis and cytokines associated with osteogenesis between M and H groups at 2 and 6 wk post-fracture.CONCLUSION The administration of at least 5.0×10^(6)MSCs was optimal to promote fracture healing in a rat model of long bone fractures.展开更多

A single concentrator solar cell model with a heat sink is established to simulate the thermal performance of the system by varying the number, height, and thickness of fins, the base thickness and thermal resistance ...A single concentrator solar cell model with a heat sink is established to simulate the thermal performance of the system by varying the number, height, and thickness of fins, the base thickness and thermal resistance of the thermal conductive adhesive. Influence disciplines of those parameters on temperatures of the solar cell and heat sink are obtained. With optimized number, height and thickness of fins, and the thickness values of base of 8, 1.4 cm, 1.5 mm, and 2 mm, the lowest temperatures of the solar cell and heat sink are 41.7 ~C and 36.3 ~C respectively. A concentrator solar cell prototype with a heat sink fabricated based on the simulation optimized structure is built. Outdoor temperatures of the prototype are tested. Temperatures of the solar cell and heat sink are stabilized with time continuing at about 37 ℃-38 ℃ and 35 ℃-36 ℃respectively, slightly lower than the simulation results because of effects of the wind and cloud. Thus the simulation model enables to predict the thermal performance of the system, and the simulation results can be a reference for designing heat sinks in the field of single concentrator solar cells.展开更多

Methods Rat mesangial cells were cultured. lntracellular calcium concentrations were measured by confocal Laser Scanning Microscopy and Fura-3 fluorescence dyeing techniques. Cell growth was measured by MTT assay. Results PDGF-BB increased intracellular calcium concentrations in a dose-dependent manner, and at the same time promote the proliferation of mesangial cells. After preincubation with calcium channel blocker nifedipine or angiotensin converting enzyme inhibitor captopril, both the increase of intracellular calcium concentrations and cell proliferations induced by PDGF-BB were inhibited. Tripteriglum Wilfordii Glycosides (TMG) significantly inhibited the mesangial cell proliferations, but it had no significant effect on intracellular calcium concentrations. Conclusion There was a positive relationship between the elevation of intracellular calcium concentration and cell proliferation in glomerular mesangial cells, but the increase of intracellular calcium concentrations wasn't the only way for proliferation.展开更多

Methods:A cell culture model, [^3H]-thymidine([^3H]-TdR) incorporation test and confocal microscope were used to observe proliferation and intracellular free calcium concentration([Ca^2±]) of rabbit PASMC induced by ET-1 in vitro. Results:The value of [^3H]-TdR incorporation in ET-1 group was increased 1.468 times higher than that in control group. Iptakalim at the concentration of 10^-7mol/L, 10^-6mol/L ,10^-5 mol/L lowered [^3H]-TdR incorporation by (19.8 ± 4.6)%, (41.2 ± 9.5)%, (54.7 ± 10.1)%, respectively, compared with the value of the cells treated with ET-1(P〈 0.01); The intracellular fluorescence intensity of PASMC in ET-1 group was increased from 73.70 ± 10.12 to 143.84 ± 28.23, significantly higher than that in control group(P 〈 0.01); whereas with Iptakalim,the fluorescence intensity(FI) was only increased from 74.30 ± 10.20 to 86.03 ± 9.82, significantly lower than that in ET-1 group(P 〈 0.01). Conclusion:Iptakalim inhibited proliferation of PASMC and decreased intracellular free calcium concentration of cultured rabbit PASMC induced by ET-1.展开更多

Starting from the thermal balance of the model, the equation is solved and simulated with a MATLAB code, considering air as the cooling fluid. This enabled us to evaluate some of the parameters influencing the electrical and thermal performance of this device. The results showed that the temperature, thermal efficiency and electrical efficiency delivered depend on the air mass flow rate. The electrical and thermal efficiencies for different values of air mass flow are encouraging, and demonstrate the benefits of cooling photovoltaic cells. The results show that thermal efficiency decreases air flow rate greater than 0.7 kg/s, whatever the value of the light concentration used. The thermal efficiency of the solar cell increases as the light concentration increases, whatever the air flow rate used. For a concentration equal to 30 sun, the thermal efficiency is 0.16 with an air flow rate equal to 0.005 kg/s;the thermal efficiency increases to 0.19 with an air flow rate equal to 0.1 kg/s at the same concentration. An interesting and useful finding was that the proposed numerical model allows the determination of the electrical as well as thermal efficiency of the hybrid CPV/T with air flow as cooling fluid.展开更多
Bifacial PV cells have the capacity to produce solar electricity from both sides and, thus, amongst other advantages, allow a significantly increase both in peak and annual power output while utilizing the same amount...Bifacial PV cells have the capacity to produce solar electricity from both sides and, thus, amongst other advantages, allow a significantly increase both in peak and annual power output while utilizing the same amount of silicone. According to the manufacturer, the bifacial cells are around 1.3 times more expensive than the single-sided cells. This way, bifacial PV cells can effectively reduce the cost of solar power for certain applications. Today, the most common application for these cells is in stationary vertical collectors which are exposed to sunlight from both sides, as the relative position of the sun changes throughout the day. Another possible application is to utilize these cells in concentrating collectors. Three test prototypes utilizing bifacial PV cells were built. The initial two prototypes were built for indoor testing and differed only in geometry of the reflector, one being asymmetric and the other symmetric. Both prototypes were evaluated in an indoor solar simulator. Both reflector designs yielded positive electrical performance results and similar efficiencies from both sides of the cell were achieved. However, lower fill factor than expected was achieved for both designs when compared to the single cell tests. The results are discussed and suggestions for further testing are presented. A third prototype was built in order to perform outdoor evaluations. This prototype utilized a bifacial PV cells string laminated in silicone enclosed between 2 standard glass panes and a collector box with an asymmetric CPC concentrator. The prototype peak electrical efficiency and temperature dependence were evaluated. A comparison between the performance of the bottom and top sides of the asymmetric collector is also presented. Additionally, the incidence modifier angle (IAM) is also briefly discussed.展开更多

The samples are characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), high-resolution transmission electron microscopy (HRTEM), and selected-area electron diffraction (SAED). It is found that dye-sensitized solar cells (DSSCs) assembled by the as-prepared Tit2 single-crystal NRs exhibit different trends under the condition of different nucleation and growth concentrations. Optimum cell performance is obtained with high nucleation concentration and low growth cycle concentration. The efficiency enhancement is mainly attributed to the improved specific surface area of the nanorod.展开更多

文摘BACKGROUND In vitro expansion to increase numbers of hematopoietic stem cells(HSCs)in cord blood could improve clinical efficacy of this vital resource.Nicotinamide(NAM)can promote HSC expansion ex vivo,but its effect on hematopoietic stem and progenitor cells(HSPCs,CD34^(+)CD38)and functional subtypes of HSCs-shortterm repopulating HSCs(ST-HSCs,CD34^(+)CD38CD45RACD49f^(+))and long-term repopulating HSCs(LT-HSCs,CD34^(+)CD38CD45RACD49f^(+)CD90^(+))is not yet known.As a sirtuin 1(SIRT1)inhibitor,NAM participates in regulating cell adhesion,polarity,migration,proliferation,and differentiation.However,SIRT1 exhibits dual effects by promoting or inhibiting differentiation in different tissues or cells.We propose that the concentration of NAM may influence proliferation,differentiation,and SIRT1 signaling of HSCs.AIM To evaluate the effects and underlying mechanisms of action of different concentrations of NAM on HSC proliferation and differentiation.METHODS CD34^(+)cells were purified from umbilical cord blood using MacsCD34 beads,and cultured for 10-12 d in a serum-free medium supplemented with cytokines,with different concentrations of NAM added according to experimental requirements.Flow cytometry was used to detect phenotype,cell cycle distribution,and apoptosis of the cultured cells.Real-time polymerase chain reaction was used to detect the transcription levels of target genes encoding stemness-related factors,che mokines,components of hypoxia pathways,and antioxidant enzymes.Dichloro-dihydro-fluorescein diacetate probes were used to evaluate intracellular production of reactive oxygen species(ROS).Determination of the effect of different culture conditions on the balance of cytokine by cytometric bead array.RESULTS Compared with the control group,the proportion and expansion folds of HSPCs(CD34^(+)CD38)incubated with 5 mmol/L or 10 mmol/L NAM were significantly increased(all P<0.05).The ST-HSCs ratio and fold expansion of the 5 mmol/L NAM group were significantly higher than those of the control and 10 mmol/L NAM groups(all P<0.001),whereas the LT-HSCs ratio and fold expansion of the 10 mmol/L NAM group were significantly higher than those of the other two groups(all P<0.05).When the NAM concentration was>10 mmol/L,cell viability significantly decreased.In addition,compared with the 5 mmol/L NAM group,the proportion of apoptotic cells in the 10 mmol/L NAM group increased and the proportion of cells in S and G2 phase decreased.Compared with the 5 mmol/L NAM group,the HSCs incubated with 10 mmol/L NAM exhibited significantly inhibited SIRT1 expression,increased intracellular ROS content,and downregulated expression of genes encoding antioxidant enzymes(superoxide dismutase 1,peroxiredoxin 1).CONCLUSION Low concentrations(5 mmol/L)of NAM can better regulate the balance between proliferation and differentiation,thereby promoting expansion of HSCs.These findings allow adjustment of NAM concentrations according to expansion needs.

文摘A unique method is proposed to encapsulate solar cells and improve their power conversion efficiency by using a millimeter-sized cylindrical lens array concentrator. Millimeter-sized epoxy resin polymer(ERP) cylindrical lens array concentrators are fabricated by the soft imprint technique based on polydimethylsiloxane stamps. The photovoltaic measurements show that millimeter-sized ERP cylindrical lens array concentrators can considerably improve the power conversion efficiency of silicon solar cells. The validity of the proposed method is proved by the coupled optical and electrical simulations. The designed solar cell devices with the advantages of high-efficiency and convenient cleaning are very useful in practical applications.

文摘A metal plate cooling model for 400~ single concentrator solar cells was established. The effects of the thickness and the radius of the metal plate, and the air environment on the temperature of the solar cells were analyzed in detail. It is shown that the temperature of the solar cells decreased sharply at the beginning, with the increase in the thickness of the metal plate, and then changed more smoothly. When the radius of the metal plate was 4 cm and the thickness increased to 2 mm or thicker, the temperature of the solar cell basically stabilized at about 53℃. Increasing the radius of the metal plate and the convective transfer coefficient made the temperature of the solar cell decrease remarkably. The effects of A1 and Cu as the metal plate material on cooling were analyzed contrastively, and demonstrated the superiority of A1 material for the cooling system. Furthermore, considering cost reduction, space holding and the stress of the system, we optimized the structural design of the metal plate. The simulated results can be referred to the design of the structure for the metal plate. Finally, a method to devise the structure of the metal plate for single concentrator solar cells was given.

文摘A compact, low-cost and high-output-power silicon-wafer solar strip-cells-array module (SCAM) wus experimentally demonstrated. The proposed SCAM consisted mainly of a silicon-wafer strip-cell sparse array and low-concentration- ratio array concentrator based on an epoxy resin polymer (ERP) cylindrical piano-convex lens. A polymer replication process based on a polydimethylsiloxane mold was used to fabricate the ERP lens array concentrator. The results show that 46.94% of the silicon-wafer cell was saved in the designed SCAM. Moreover, the output power of the SCAM with a low concentration ratio of 8 suns was improved by 8.6%, compared with a whole piece of a conventional silicon-wafer solar cell with the same area as the module. The proposed method encapsulating solar cells provides a means to reduce the usage of silicon cells in modules as well as improving the output power of modules.

文摘The roles of NF-kappaB (NF-κB) expression, Bax activity and cytochrome C (Cyt C) release, apoptosis of islet cells induced by high concentration glucose were explored in vitro. Pancreatic islet cells, which were isolated from Kunming mice, were cultured with different concentrations of glucose in DMEM, and divided into the following groups: G1, G2, G3, G4, G5, and G6 groups, corresponding to the glucose concentrations of 5.6, 7.8, 11.1, 16.7, 22.5, and 27.6 mmol/L, respectively. After culture for 120 h, insulin secretion was evaluated by radioimmunoassay, and the NF-rd3 expression was detected by immunocytochemistry. Bax activity and Cyt C release were measured by immunofluorescence, and apoptosis was examined by Hoechst33342 assay. The results showed that in GI, G2 and G3 groups, insulin secretion was enhanced with the increase of glucose concentration, and the NF-κB expression was also increased (P〈0.05), but Bax activity, Cyt C release and apoptosis rate showed no significant difference among them. However, in G4, G5, and G6 groups, apoptosis rate of islet cells, NF-rd3 expression, Bax activity, and Cyt C release were all significantly increased, and insulin secretion was impaired as compared with G1, G2, and G3 groups (P〈0.05). It was concluded that the exposure of islet cells to high glucose could induce islet cells apoptosis as well as impaired insulin secretion. The NF-κB signaling pathway and mitochondria pathway in islet cells might play some roles in the progressive loss of islet cells in diabetes. The inhibition of the NF-κB expression could be an effective strategy for protecting pancreatic islet cells.
文摘Previous studies have shown that renewable energy is one of the effective ways to fight global climate change and emissions not to mention the increasing price of fossil fuels. Among the various renewable energy sources which include wind, solar, biofuel, geothermal and tidal waves, solar power has attracted much attention especially here in Africa because of the abundance of solar radiation. A lot of studies have been done on various photovoltaic (PV) cells ranging from silicon to thin film and most recently multi-junction solar cells. In this paper, we focused on concentrated photovoltaic cells (CPV) which are promising ways of converting solar energy to electricity. It is expected that if the most cost effective ways of converting solar energy to electricity is used, both the cost of installation and the running cost of concentrated photovoltaic cells will equate the utility grid electricity cost in a few years to come.

文摘Background:The adhesion of monocytes to the endothelium following accumulation of low-density lipoprotein (LDL) in subendothelial spaces is an important step in the development of intimal hyperplasia in arterially implanted vein grafts and atherosclerosis in both animals and humans. However, it is not well known how serum factors affect the adhesion of monocytes. Methods: We have studied the effect of fetal calf serum (FCS), which we considered a source of LDL, on the adhesion of monocytes to endothelial cells (ECs) by using human monocytic THP-1 cells and both a monolayer of cultured bovine aortic endothelial cells (EC monoculture) and a co-culture with bovine aortic smooth muscle cells (EC-SMC co-culture). Results: It was found that the addition of FCS to the medium greatly affected the adhesion of THP-1 cells, and the higher the concentration of FCS in the medium, the greater the adhesion of THP-1 cells to endothelial cells. Adhesion of THP-1 cells to an EC-SMC co-culture was approximately twofold greater than that to an EC monoculture, and after adhering to endothelial cells, many THP-1 cells trans-migrated into the layer of smooth muscle cells. Conclusion: The results suggest that the elevation of the LDL (cholesterol) level in blood provides a favorable condition for the development of intimal hyperplasia and atherosclerosis by promoting the adhesion of monocytes to the endothelium and their subsequent migration into subendothelial spaces.

文摘Objective The main purpose of this study was to investigate whether the folliculo stellate cells (FSC) respond to angiotensin(Ang)Ⅱ by increasing intracellular free Ca 2+ concentration ([Ca 2+ ]i),and where the origin of Ca 2+ mobilization is if that has occurred.Methods Pituitary cells in primary culture were prepared from male Wister rats(250g) by a conventional method and cultured in MEM supplemented with 4% normal rat serum.After 2 days in culutre,cells were loaded with 1 μmol/L fura PE3/AM for 1 h and subjected to a Ca 2+ imaging experiment with Quanti Cell 700 system.Excitation wavelengths of 340 and 380 nm were selected by means of a computer controlled filterwheel.Results The [Ca 2+ ]i of FSC in the rat anterior pituitary was elevated by Ang Ⅱ.The elevation of [Ca 2+ ]i of FSC induced by 0.1,1.0,10 and 100 nmol/L Ang Ⅱ was (56.33±6.18)( ±s ),(117.07±36.07),(175.59±40.01) and (216.02±11.52) nmol/L,respectively.The increase of [Ca 2+ ]i of FSC induced by 100 nmol/L Ang Ⅱ was not influenced by the medium without Ca 2+ (0Ca),but significantly suppressed by thapsigargin(TG),an inhibitor of ATPase.The rate of responsive FSC to Ang Ⅱ (100 nmol/L) was 61.84% which was obviously higher than that of pituitary endocrine cells(43.49%).Conclusion The present experiment demonstrates that the FSC in the rat anterior pituitary responds to Ang Ⅱ by increasing [Ca 2+ ]i,which raises the possibility that Ang Ⅱ released from either lactotrophs or gonadotrophs affects FSC through paracrine mechanism.The elevation of [Ca 2+ ]i induced by Ang Ⅱ presents a dosage dependent relation, and is possibly because of the release of Ca 2+ from an intracellular Ca 2+ pool(s).Fashions of Ca 2+ release are relative to the concentration of Ang Ⅱ.The results indicate that Ang Ⅱ functions as a paracrine factor among pituitary cells including FSC.

文摘BACKGROUND There is still no consensus on which concentration of mesenchymal stem cells(MSCs)to use for promoting fracture healing in a rat model of long bone fracture.AIM To assess the optimal concentration of MSCs for promoting fracture healing in a rat model.METHODS Wistar rats were divided into four groups according to MSC concentrations:Normal saline(C),2.5×10^(6)(L),5.0×10^(6)(M),and 10.0×10^(6)(H)groups.The MSCs were injected directly into the fracture site.The rats were sacrificed at 2 and 6 wk post-fracture.New bone formation[bone volume(BV)and percentage BV(PBV)]was evaluated using micro-computed tomography(CT).Histological analysis was performed to evaluate fracture healing score.The protein expression of factors related to MSC migration[stromal cell-derived factor 1(SDF-1),transforming growth factor-beta 1(TGF-β1)]and angiogenesis[vascular endothelial growth factor(VEGF)]was evaluated using western blot analysis.The expression of cytokines associated with osteogenesis[bone morphogenetic protein-2(BMP-2),TGF-β1 and VEGF]was evaluated using real-time polymerase chain reaction.RESULTS Micro-CT showed that BV and PBV was significantly increased in groups M and H compared to that in group C at 6 wk post-fracture(P=0.040,P=0.009;P=0.004,P=0.001,respectively).Significantly more cartilaginous tissue and immature bone were formed in groups M and H than in group C at 2 and 6 wk post-fracture(P=0.018,P=0.010;P=0.032,P=0.050,respectively).At 2 wk post fracture,SDF-1,TGF-β1 and VEGF expression were significantly higher in groups M and H than in group L(P=0.031,P=0.014;P<0.001,P<0.001;P=0.025,P<0.001,respectively).BMP-2 and VEGF expression were significantly higher in groups M and H than in group C at 6 wk postfracture(P=0.037,P=0.038;P=0.021,P=0.010).Compared to group L,TGF-β1 expression was significantly higher in groups H(P=0.016).There were no significant differences in expression levels of chemokines related to MSC migration,angiogenesis and cytokines associated with osteogenesis between M and H groups at 2 and 6 wk post-fracture.CONCLUSION The administration of at least 5.0×10^(6)MSCs was optimal to promote fracture healing in a rat model of long bone fractures.
文摘A new transparent photovoltaic panel composed of a luminescent solar concentrator and Al/BaTiO3/ZnO/Pt ferroelectric solar cells is presented,in which a portion of the incoming solar illumination is converted by the fluorophores to ultraviolet(UV)light which is then absorbed by ZnO.Firstly,the solar cells are simulated using Atlas-Silvaco.Then,the panel is modelled based on the obtained solar cell characteristics.This panel would be of great importance for building integrated photovoltaics domain because of its high transparency.

文摘Two improved algorithms are proposed to extend a diffractive optical element (DOE) to work under the broad spec- trum of sunlight. An optimum design has been found for the DOE, with a weighted average optical efficiency of about 6.8% better than that of the previous design. The optimization of designing high optical efficiency DOEs will pave the way for future designs of high-efficiency, low-cost lateral multijunction solar cells based on such a DOE.

文摘A single concentrator solar cell model with a heat sink is established to simulate the thermal performance of the system by varying the number, height, and thickness of fins, the base thickness and thermal resistance of the thermal conductive adhesive. Influence disciplines of those parameters on temperatures of the solar cell and heat sink are obtained. With optimized number, height and thickness of fins, and the thickness values of base of 8, 1.4 cm, 1.5 mm, and 2 mm, the lowest temperatures of the solar cell and heat sink are 41.7 ~C and 36.3 ~C respectively. A concentrator solar cell prototype with a heat sink fabricated based on the simulation optimized structure is built. Outdoor temperatures of the prototype are tested. Temperatures of the solar cell and heat sink are stabilized with time continuing at about 37 ℃-38 ℃ and 35 ℃-36 ℃respectively, slightly lower than the simulation results because of effects of the wind and cloud. Thus the simulation model enables to predict the thermal performance of the system, and the simulation results can be a reference for designing heat sinks in the field of single concentrator solar cells.
文摘Objective To investigate the relationship between the alteration of intracellular calcium concentration and proliferation in cultured glomerular mesangial cells. Methods Rat mesangial cells were cultured. lntracellular calcium concentrations were measured by confocal Laser Scanning Microscopy and Fura-3 fluorescence dyeing techniques. Cell growth was measured by MTT assay. Results PDGF-BB increased intracellular calcium concentrations in a dose-dependent manner, and at the same time promote the proliferation of mesangial cells. After preincubation with calcium channel blocker nifedipine or angiotensin converting enzyme inhibitor captopril, both the increase of intracellular calcium concentrations and cell proliferations induced by PDGF-BB were inhibited. Tripteriglum Wilfordii Glycosides (TMG) significantly inhibited the mesangial cell proliferations, but it had no significant effect on intracellular calcium concentrations. Conclusion There was a positive relationship between the elevation of intracellular calcium concentration and cell proliferation in glomerular mesangial cells, but the increase of intracellular calcium concentrations wasn't the only way for proliferation.

文摘Objective:To explore the effects of Iptakalim on intracellular free calcium concentration and on the proliferation of cultured rabbit pulmonary arterial smooth muscle cells induced by endothelin-1 (ET-1) in vitro. Methods:A cell culture model, [^3H]-thymidine([^3H]-TdR) incorporation test and confocal microscope were used to observe proliferation and intracellular free calcium concentration([Ca^2±]) of rabbit PASMC induced by ET-1 in vitro. Results:The value of [^3H]-TdR incorporation in ET-1 group was increased 1.468 times higher than that in control group. Iptakalim at the concentration of 10^-7mol/L, 10^-6mol/L ,10^-5 mol/L lowered [^3H]-TdR incorporation by (19.8 ± 4.6)%, (41.2 ± 9.5)%, (54.7 ± 10.1)%, respectively, compared with the value of the cells treated with ET-1(P〈 0.01); The intracellular fluorescence intensity of PASMC in ET-1 group was increased from 73.70 ± 10.12 to 143.84 ± 28.23, significantly higher than that in control group(P 〈 0.01); whereas with Iptakalim,the fluorescence intensity(FI) was only increased from 74.30 ± 10.20 to 86.03 ± 9.82, significantly lower than that in ET-1 group(P 〈 0.01). Conclusion:Iptakalim inhibited proliferation of PASMC and decreased intracellular free calcium concentration of cultured rabbit PASMC induced by ET-1.
文摘In this paper, we propose a thermal model of a hybrid photovoltaic/thermal concentration system. Starting from the thermal balance of the model, the equation is solved and simulated with a MATLAB code, considering air as the cooling fluid. This enabled us to evaluate some of the parameters influencing the electrical and thermal performance of this device. The results showed that the temperature, thermal efficiency and electrical efficiency delivered depend on the air mass flow rate. The electrical and thermal efficiencies for different values of air mass flow are encouraging, and demonstrate the benefits of cooling photovoltaic cells. The results show that thermal efficiency decreases air flow rate greater than 0.7 kg/s, whatever the value of the light concentration used. The thermal efficiency of the solar cell increases as the light concentration increases, whatever the air flow rate used. For a concentration equal to 30 sun, the thermal efficiency is 0.16 with an air flow rate equal to 0.005 kg/s;the thermal efficiency increases to 0.19 with an air flow rate equal to 0.1 kg/s at the same concentration. An interesting and useful finding was that the proposed numerical model allows the determination of the electrical as well as thermal efficiency of the hybrid CPV/T with air flow as cooling fluid.
文摘Bifacial PV cells have the capacity to produce solar electricity from both sides and, thus, amongst other advantages, allow a significantly increase both in peak and annual power output while utilizing the same amount of silicone. According to the manufacturer, the bifacial cells are around 1.3 times more expensive than the single-sided cells. This way, bifacial PV cells can effectively reduce the cost of solar power for certain applications. Today, the most common application for these cells is in stationary vertical collectors which are exposed to sunlight from both sides, as the relative position of the sun changes throughout the day. Another possible application is to utilize these cells in concentrating collectors. Three test prototypes utilizing bifacial PV cells were built. The initial two prototypes were built for indoor testing and differed only in geometry of the reflector, one being asymmetric and the other symmetric. Both prototypes were evaluated in an indoor solar simulator. Both reflector designs yielded positive electrical performance results and similar efficiencies from both sides of the cell were achieved. However, lower fill factor than expected was achieved for both designs when compared to the single cell tests. The results are discussed and suggestions for further testing are presented. A third prototype was built in order to perform outdoor evaluations. This prototype utilized a bifacial PV cells string laminated in silicone enclosed between 2 standard glass panes and a collector box with an asymmetric CPC concentrator. The prototype peak electrical efficiency and temperature dependence were evaluated. A comparison between the performance of the bottom and top sides of the asymmetric collector is also presented. Additionally, the incidence modifier angle (IAM) is also briefly discussed.

文摘An excellent organolead halide perovskite film is important for the good performance of perovskite solar cells(PSCs).However,defects in perovskite crystals can affect the photovoltaic properties and stability of solar cells.To solve this problem,this study incorporated a complex of Cd S and Cd(SCN_(2)H_(4))_(2)Cl_(2)into the CH_(3)NH_(3)Pb I_(3)active layer.The effects of different doping concentrations of Cd S and Cd(SCN_(2)H_(4))_(2)Cl_(2)on the performance and stability of PSCs were analyzed.Results showed that doping appropriate incorporation concentrations of Cd S and Cd(SCN_(2)H_(4))_(2)Cl_(2)in CH_(3)NH_(3)Pb I_(3)can improve the performance of the prepared solar cells.In specific,Cd S and Cd(SCN_(2)H_(4))_(2)Cl_(2)can effectively passivate the defects in perovskite crystals,thereby suppressing the charge recombination in PSCs and promoting the charge extraction at the TiO_(2)/perovskite interface.Due to the reduction of perovskite crystal defects and the enhancement of compactness of the Cd S:Cd(SCN_(2)H4)_(2)Cl_(2):CH_(3)NH_(3)Pb I_(3)composite film,the stability of PSCs is significantly improved.
