A highly selective and stability-indicating HPLC-method, combined with appropriate sample preparation steps, is developed for β-artemether assay and profiling of related impurities, including possible degradants, in ...A highly selective and stability-indicating HPLC-method, combined with appropriate sample preparation steps, is developed for β-artemether assay and profiling of related impurities, including possible degradants, in a complex powder for oral suspension. Following HPLC conditions allowed the required selectivity: a Prevail organic acid (OA) column (250 mm×4.6 mm, 5μm), flow rate set at 1.5 mL/min combined with a linear gradient (where A ? 25 mM phosphate buffer (pH 2.5), and B ? acetonitrile) from 30% to 75% B in a runtime of 60 min. Quantitative UV-detection was performed at 210 nm. Acetonitrile was applied as extraction solvent for sample preparation. Using acetonitrile-water mixtures as extraction solvent, a compartmental behaviour by a non-solving excipient-bound fraction and an artemether-solubilising free fraction of solvent was demonstrated, making a mobile phase based extraction not a good choice. Method validation showed that the developed HPLC-method is considered to be suitable for its intended regulatory stability-quality characterisation of β-artemether paediatric formulations. Furthermore, LC-MS on references as well as on stability samples was performed allowing identity confirmation of the β-artemether related impurities. MS-fragmentation scheme of β-artemether and its related substances is proposed, explaining the m/z values of the in-source fragments obtained.展开更多

Moreover, P governs crucial role in plant as it stimulates root development and growth, gives plant rapid and vigorous start leading to better tillering and essential for many metabolic processes for seed formation. Soil microbes play very important role in bio-weathering and biodegradation. The microorganisms produce low molecular mass organic acids, which attack the phosphate structure and transform phosphorus from non-utilizable to the utilizable for the plants form. The test of the relative efficiency of isolated strains is carried out by selecting the microorganisms that are capable of producing a halo/clear zone on a plate owing to the production of organic acids into the surrounding medium. It is a well-known fact that as the particle size of rock phosphate decreases, the microbe mediated solubilization of rock phosphate increases in soil. In the present investigation, microbial solubilization of nano rock phosphate (〈 100 nm) particles was studied. Experimental results revealed that Pseudomonas striata solubilized 11.45% of the total P after 24 h of incubation from nano rock phosphate particles while 28.95% and 21.19% of the total P was solubilized by Aspergillus niger (black pigmented) and Aspergillus niger (green pigmented), respectively. It was also observed that Aspergillus niger has the higher ability to dissolve Udaipur rock phosphate than Pseudomonas striata.展开更多
The effect of zinc(Zn) deficiency and excessive bicarbonate on the allocation and exudation of organic acids in plant organs(root, stem, and leaf) and root exudates of two Moraceae plants(Broussonetia papyrifera and M...The effect of zinc(Zn) deficiency and excessive bicarbonate on the allocation and exudation of organic acids in plant organs(root, stem, and leaf) and root exudates of two Moraceae plants(Broussonetia papyrifera and Morus alba) were investigated. Two Moraceae plants were hydroponically grown and cultured in nutrient solution in four different treatments with 0.02 mM Zn or no Zn,combined with no or 10 mM bicarbonate. The variations of organic acids in different plant organs were similar to those of root exudates in the four treatments except B. papyrifera, which was in a treatment that was a combination of 0.02 mM Zn and no bicarbonate. The response characteristics in the production, translocation, and allocation of organic acids in the plant organs and root exudates varied with species and treatments. Organic acids in plant organs and root exudates increased under Zn-deficient conditions,excessive bicarbonate, or both. An increase of organic acids in the leaves resulted in an increase of root-exuded organic acids. B. papyrifera translocated more oxalate and citrate from the roots to the rhizosphere than M. alba under the dual influence of 10 mM bicarbonate and Zn deficiency. Organic acids of leaves may be derived from dark respiration and photorespiration. By comparison, organic acids in stems, roots, and root exudates may be derived from dark respiration and organic acid translocation from the leaves. These results provide evidence for the selective adaptation of plants to environments with low Zn levels or high bicarbonate levels such as a karst ecosystem.展开更多

The j...This study identifies the potential role in heat-stress mitigation of phytohormones and other secondary metabolites produced by the endophytic fungus Paecilomyces formosus LWL1 in japonica rice cultivar Dongjin. The japonica rice was grown in controlled chamber conditions with and without P. formosus LWL1 under no stress (NS) and prolonged heat stress (HS) conditions. Endophytic association under NS and HS conditions significantly improved plant growth attributes, such as plant height, fresh weight, dry weight, and chlorophyll content. Furthermore, P. for- mosus LWL1 protected the rice plants from HS compared with controls, indicated by the lower endogenous level of stress-signaling compounds such as abscisic acid (25.71%) and jasmonic acid (34.57%) and the increase in total protein content (18.76%-33.22%). Such fungal endophytes may be helpful for sustainable crop production under high environmental temperatures.展开更多

文摘采用乙醇/盐双水相体系对牡丹花色苷进行萃取精制,分别考察分相盐和乙醇的添加量对花色苷和多糖分离效果的影响。结果表明,选择硫酸铵作为分相盐,添加量为5.5 g,乙醇与水的体积比为1∶1,在此条件下花色苷和多糖分离效果最好,花色苷萃取率为96.43%。进一步考察有机酸、金属离子、p H对牡丹花色苷稳定性的影响。研究表明,丙二酸和Mn2+对花色苷有辅色效果,可以提高花色苷的稳定性,低p H条件(p H 2,3)下花色苷的稳定性较好;以花色苷保存率为响应值,在单因素试验基础上进行响应面分析试验。试验优化结果为:丙二酸浓度88 mmol/L,Mn2+浓度6.2 mmol/L,p H 2.6,在此条件下花色苷保存率为79.21%,较空白对照组稳定性提高了近2倍。
