[Method] With ellagic acid yield as an indicator, ultrasound extraction method was adopted to extract the ellagic acid from pomegranate peels, and the concentration of ellagic acid was measured by using ul- traviolet spectrophotometry; L9(34) orthogonal experiment was designed with four fac- tors at three levels, to investigate the effect of extraction temperature, extraction du- ration, solid-liquid ratio and ultrasound power on extraction rate of ellagic acid. [Re- sult] The optimal extraction condition for ellagic acid is extraction temperature of 30 ~C, solid-liquid ratio of 1:200, extraction duration of 20 min, and ultrasound power of 50 W. The relationship between primary and secondary factors affecting experimental indicators was solid-liquid ratio 〉 extraction duration 〉 ultrasound power 〉 extraction temperature. [Conclusion] Solid-liquid ratio has the maximum effect on extraction rate of ellagic acid. The ellagic acid extraction technology identified in this study is rea- sonable and feasible.展开更多

[Methods] The effects of four factors including ultrasonic extraction time, liquid-to-material ratio, ultrasonic extraction temperature and ethanol concentration on extraction yield of luteolin from Ternate Grape Fern Herb were investigated by the ultrasonic-assisted extraction technology, through a single-factor experiment and an orthogonal experiment based on the single-factor experiment, and the experimental data was subjected to variance analysis and SSR analysis of the significance of differences between groups by SPSS20.0 software to select the optimal extraction process conditions. [Results] The optimal extraction process conditions were as follows: ultrasonic extraction time 25 min, liquid-to-material ratio 25∶1 ml/g, ethanol volume fraction 60%, ultrasonic extraction temperature 85 ℃. Methods The key extraction parameters that influenced the yield of flavonid from Fructus Gardeniae were optimized by employing an orthogonal experiment [L9(3)4], including the ratio of buffer solution (Na2B4O7· 10H2O) to raw material, concentration of Fructus Gardeniae in extracting solution, extraction time and pH of buffer solution. An UV/Vis detector was used to perform the qualitative and quantitative analyses of the extracted flavonid with the using of the standard sample. Results The maximum extraction yield of the crude extract was 5.0533 (mg/g) after 20 min when the mass ratio of Na2B4O7 · 10H2O to raw material was 0.4%, the concentration of Fructus Gardeniae in the extraction solution was 1/12 (g/mL), and pH of buffer solution was 4.5. The positive reactions to the Molish and HCI-Mg tests suggested that the extracted compound was flavonoid, and FTIR measurements also identified the presence of flavonoid in the extracts. Conclusion This work is expected to provide a basis for further research, development, and utilization of Fructus gardenia in flavonid extraction.展开更多

The results of orthogonal experiments indicate that LI...An experimental investigation was presented on the separation of Cu(Ⅱ), Zn(Ⅱ), and Cd(Ⅱ) from a rich sulfate leachate of zinc slag by solvent extraction. The results of orthogonal experiments indicate that LIX 984N is highly selective and very efficient in the extraction of Cu(Ⅱ), and the analysis of variance indicates that the sequence of parameters according to their influence on the separation efficiency is phase ratio 〉 LIX 984N concentration 〉 pH value 〉 extraction time. The optimal condition for copper extraction is obtained as 25% of LIX 984N concentration, 7 rain of extraction time, 3:2 of phase ratio O/A, and pH = 1.7. The separation of Zn(Ⅱ) and Cd(Ⅱ) was performed after the copper extraction from the raffinate. Comparative analysis of the separation with di-2-ethylhexyl phosphoric acid (D2EHPA), D2EHPA-tributyl- phosophate (TBP) synergistic extracting system, and 2-ethylhexyl phosphonic acid mono 2-ethylhexyl ester (HEHEHP) was made at pH = 2.0. It is demonstrated that the extraction efficiency with D2EHPA is improved after being saponified by sodium hydroxide, and D2EHPA-TBP synergistic extracting, as well as HEHEHP, has a superior selectivity to Zn(Ⅱ) over Cd(Ⅱ).展开更多

The optimal conditions for ultrasonic extraction of total flavonoids from A. keiskei were extraction time of 25 min, ethanol concentration of 80%, ultrasonic power of 60 W, solid-to-liquid ratio of 1:20 (g/ml), under which the yield of total flavonoids reached 1.56%. Extraction temperature, extrac- tion duration, solution pH and solid to liquid ratio were selected as technical parameters which required to be optimized. With the extraction rate of seaweed poly- saccharides as an index, single-factor experiment and orthogonal experiment were carried out to determine the optimal extraction technique. Seaweed polysaceharides were extracted with the optimal technical parameters to analyze the biological activities. Savage method was used to process seaweed polysaceharide extract. In vitro biological activities of preliminarily purified seaweed polysaeeharides were investigated successively, including reducing ability, antioxidant ability and antibacterial activity. The results showed that there was a positive correlation between polysaccharide concentration and its reducing ability, antioxidant ability and antibacterial activity. Furthermore, there was also a correlation among the reducing ability, antioxidant ability and antibacterial activity of seaweed polysaecharides; the relation- ship between each two of these three indicators was similar to positive linear correlation.展开更多
[Objectives]To establish a qualitative identification and content determination method for psoralen and bergapten in the roots and leaves of Ficus pandurata,and the multi-indicator scoring method to optimize the ethan...[Objectives]To establish a qualitative identification and content determination method for psoralen and bergapten in the roots and leaves of Ficus pandurata,and the multi-indicator scoring method to optimize the ethanol extraction process for the effective parts of F.pandurata.[Methods]Qualitative identification was carried out by thin-layer chromatography(TLC)and content determination was made by high-performance liquid chromatography(HPLC);the content of psoralen and bergapten and extract yield were used as indicators,to investigate the effects of ethanol volume fraction,solid-to-liquid ratio,extraction time and extraction times on the ethanol extraction process of F.pandurata;multi-indicator scoring method was adopted,orthogonal test method was designed to optimize the extraction process,and verification test was performed.[Results]TLC chart of F.pandurata shows clear spots and good separation;the detected concentrations of psoralen and bergapten in the roots are in the range of 1.02-32.64μg/mL and shows a good linear relationship with the peak area(r=0.9997);these components have not been detected in the leaves,and the precision,RSD of stability and repeatability test is less than 2%;the average recovery rate was 99.8%-100.2%,and the RSD value is 1.12%-1.13%(n=6);the optimized extraction process is to use 50%ethanol as the extraction solvent,the solid-to-liquid ratio of 1∶10,reflux extraction of 3 times,and 1.5 h each time;the results of the three batches of verification tests show that the content of the two indicator components obtained is high,and the average value of the total amount is 0.34 mg/g(RSD=0.30%,n=3).[Conclusions]The established quality control method for F.pandurata is simple,easy,accurate and reproducible;the preferred extraction process is stable and feasible,suitable for the extraction and purification of coumarin effective parts in F.pandurata roots,so it is expected to provide references for further development of F.pandurata.展开更多

[Methods]Selenium-enriched rice was used as materials to extract selenoproteins by phosphate buffer extraction method and to optimize the extraction process of selenoproteins by using the orthogonal experiment. Proteins and selenium content was measured by Coomassie Brilliant Blue G-250 reagent and AFS( atomic fluorescence spectrometry) respectively. [Results] The most significant factor affecting extraction of rice Selenoproteins was extraction NaO H concentration,followed by the ratio of solid-liquid,temperature and then extraction time. The optimum extraction conditions of selenoproteins from rice were extraction temperature of 50 ℃,NaO H concentration of 0. 14 mol/L,extraction time of 5 h,and solid-liquid ratio of 1∶ 30. [Conclusions]The alkali extraction process optimized by orthogonal test could effectively improve the extraction rate of selenoproteins,and the optimized process parameters could be popularized and applied in practical production.展开更多

[Results]The oxysophocarpine was determined by Shim-pack VP-ODS chromatographic column( 4. 6 mm × 250 mm,5 μm),mobile phase was methanol-0. 2% phosphoric acid aqueous solution( 7∶ 93),gradient elution,flow rate was 1 m L/min,the sample size was 5 μm,column temperature was 30℃,detection wavelength was 221 nm. Aqueous phase and organic phase volume ratio was 1∶ 1,hydrochloric acid concentration was 0. 3 mol/L,sodium hydroxide concentration was 0. 75 mol/L,water pump flow rate was 6 m L/min,60 min cycle time. The extraction rate of oxysophocarpine 98. 21 % within 60 min under the best experimental conditions. Oxysophocarpine has good linearity relationship within the range of 0. 01-0. 7 mg/m L( r^2= 0. 9978,n = 6),the respective average recovery rate was 97. 47%( RSD = 1. 95%). [Conclusions] This extraction process is simple in operation,the organic solvent has low consumption,and can be used for extraction of alkaloid.展开更多

It is of great significance to find ...[Objectives] The chlorogenic acid in Sambucus chinensis Lindl is a kind of biologically active substance with multiple pharmacological effects, but the content is relatively small. It is of great significance to find the best extraction process. Therefore, we designed single factor experiments and orthogonal experiments to screen the best conditions for extracting chlorogenic acid from S. chinensis. [Methods] With ethanol concentration, ultrasonic power, and extraction time as three single factors, single factor experiments were performed on S. chinensis, and its chlorogenic acid content was calculated from a standard curve. The contents of chlorogenic acid obtained under various conditions were compared, and then, orthogonal experiments were designed according to the orthogonal table to analyze the optimal extraction conditions of chlorogenic acid. [Results] The optimal extraction conditions of chlorogenic acid for S. chinensis were: the ethanol concentration 60%, the ultrasonic power 36 W, and the extraction time 40 min.[Conclusions] The factor that had the greatest effect on the extraction rate of chlorogenic acid in S. chinensis was ethanol concentration, followed by extraction time, and the least influencing factor was ultrasonic power. With 60% ethanol solution serving as the extractant, the ultrasonic power of 36 W, and the extraction time of 40 min, a better extraction rate was obtained. These conditions can be used as the best extraction conditions for chlorogenic acid.展开更多

文摘[Objectives]To optimize the water extraction process of Fagopyri Dibotryis Rhizoma.[Methods]The entropy weight method was used to determine the weight of epicatechin extraction rate and dry extract rate and calculate the comprehensive score.The water extraction process of Fagopyri Dibotryis Rhizoma was optimized by orthogonal design with the comprehensive score as the indicator and the amount of water,extraction time and extraction times as the factors.[Results]The optimum extraction process of Fagopyri Dibotryis Rhizoma was as follows:adding 10 times of water,extracting 3 times,and extracting for 60 min each time.[Conclusions]The optimized extraction process is stable and feasible,and can be used for the extraction of Fagopyri Dibotryis Rhizoma.

文摘[Objective] This study aimed to explore optimum conditions for papain-as- sisted extraction of polysaccharides from Agrocybe aegirit. [Method] Because enzyme is capable of destroying organizational structure of cells, papain was selected to cat- alyze cells to let polysaccharides out. An orthogonal experiment was carried out to investigate the effects of hydrolysis temperature, hydrolysis time, enzyme concentra- tion and solid to liquid ratio on extraction of polysaccharides from A. aegirit. [Result] Through the orthogonal experiment, the optimum papain-assisted extraction conditions were obtained: hydrolysis temperature, 45 ℃; hydrolysis time, 2 h; enzyme concen- tration 2%; solid to liquid ratio, 1:80. [Conclusion] This study will provide a reference for extraction of polysaccharides from A. aegirit.

文摘In this study, by using mulberry leaves as raw materials, ultrasonic extraction process of total flavonoids from mulberry leaves was optimized by orthogonal experiment. Results showed that the maximum yield of total flavonoids (6.1% ) was obtained from mulberry leaves with addition of 45 times the volume of water by ultrasonic extraction at 55 ℃ for three times, 55 min each. According to the verification test, the optimized technical process was reasonable and feasible with high stability.

文摘[Objectives] This study was conducted to optimize the extraction process of luteolin, a natural product of Ternate Grape Fern Herb, so as to provide a theoretical research basis for the chemical composition, pharmacological research and preparation development of Ternate Grape Fern Herb. [Methods] The effects of four factors including ultrasonic extraction time, liquid-to-material ratio, ultrasonic extraction temperature and ethanol concentration on extraction yield of luteolin from Ternate Grape Fern Herb were investigated by the ultrasonic-assisted extraction technology, through a single-factor experiment and an orthogonal experiment based on the single-factor experiment, and the experimental data was subjected to variance analysis and SSR analysis of the significance of differences between groups by SPSS20.0 software to select the optimal extraction process conditions. [Results] The optimal extraction process conditions were as follows: ultrasonic extraction time 25 min, liquid-to-material ratio 25∶1 ml/g, ethanol volume fraction 60%, ultrasonic extraction temperature 85 ℃. [Conclusions] The results were verified, and it was determined that the ultrasonic-assisted extraction process established in this study is reasonable and reliable, and can be used for the extraction of luteolin.

文摘The aim of this study was to investigate the extraction process and antioxidant activity of polyphenols from Yunnan Mengzi pomegranate peel.Single factor and orthogonal test were used as optimize the extract rate(ER)of polyphenols from pomegranate peel(PGP).The polyphenols antioxidant activity was evaluated by DPPH·,·OH and total reducing ability,and compared with that of Vc.The optimum extraction conditions of polyphenols were as follows:ethanol concentration of 50%,liquid-solid ratio of 50:1,extraction temperature of 90°C,extraction for 4 h.Under these conditions,the ER of polyphenols from GPG was 8.15%.The results showed that the scavenging effects of polyphenols and Vc on DPPH·,·OH increased with increasing concentration,and the scavenging effect of polyphenols on DPPH·was higher than Vc in rang of 0.1-1.6µg/mL.When the concentration is greater than 0.6µg/mL,the scavenging effects of plyphenols on·OH is higher than Vc.The total reducing ability of polyphenols from PGP improved with the increase of concentration,and when the concentration was 8µg/mL,the total reducing ability was 114 U/mL.

文摘[Objectives] To screen the optimal extraction conditions of flavonoids from leaves of Paliurus ramosissimus( Lour.) Poir.,measure the content of flavonoids in leaves of P. ramosissimus. using the Microplate Reader,and provide experimental basis for the development of medicinal resources of P. ramosissimus. [Methods] The ultrasonic extraction method was adopted and orthogonal design was carried out to study the effects of ethanol concentration( A),ultrasonic time( B),the solid-to-liquid ratio( C) on the extraction of flavonoids from leaves of P. ramosissimus. The Microplate Reader was used to measure the content of flavonoids in leaves of P. ramosissimus. in different extraction conditions,P. ramosissimus. and the optimal extraction conditions were used to measure the content of flavonoids from leaves of P. ramosissimus.[Results]Effects of the ultrasonic extraction method on the extraction rate of flavonoids from leaves of P. ramosissimus: extraction times >ethanol concentration > Ultrasound time > solid-liquid ratio. The screened optimal extraction process was: ethanol concentration of 65%,the solid-to-liquid ratio of 1∶ 25,ultrasonic time of 2 h and extraction times Under this condition,the extraction rate of total flavonoids in leaves of P. ramosissimus. was 2. 56%. [Conclusions] This method is simple and feasibility,reliable,and suitable for rapid measurement with large samples.

文摘Objective It is imperative to provide some consistent experimental results for the extraction of flavonid from Fructus Gardeniae. Methods The key extraction parameters that influenced the yield of flavonid from Fructus Gardeniae were optimized by employing an orthogonal experiment [L9(3)4], including the ratio of buffer solution (Na2B4O7· 10H2O) to raw material, concentration of Fructus Gardeniae in extracting solution, extraction time and pH of buffer solution. An UV/Vis detector was used to perform the qualitative and quantitative analyses of the extracted flavonid with the using of the standard sample. Results The maximum extraction yield of the crude extract was 5.0533 (mg/g) after 20 min when the mass ratio of Na2B4O7 · 10H2O to raw material was 0.4%, the concentration of Fructus Gardeniae in the extraction solution was 1/12 (g/mL), and pH of buffer solution was 4.5. The positive reactions to the Molish and HCI-Mg tests suggested that the extracted compound was flavonoid, and FTIR measurements also identified the presence of flavonoid in the extracts. Conclusion This work is expected to provide a basis for further research, development, and utilization of Fructus gardenia in flavonid extraction.

文摘[ Objective ] This study aimed to establish the optimal extraction technology of total flavonoids from Vitex rotundifolia and provide a theoretical basis for the utilization and development of V. rotundifolia. [ Method ] With the extraction temperature, extraction time, solid-liquid ratio and ethanol concentration as various factors, the content of total flavonoids as an indicator, the extraction technology of total flavonoids from V. rotundifolia was optimized by orthogonal experiment. [ Result] The optimal combination of extraction conditions was extraction temperature 70℃, extraction time 1 h, solid-liquid ratio 1: 12, ethanol concentration 70%. [Condusion] The optimized extraction technology was simple and efficient for extracting total flavonoids from V. rotundifolia.

文摘[Objectives]This study was conducted to optimize the extraction process of total flavonoids from Penthorum chinense Pursh and compare their contents from different parts.[Methods]Single factor and orthogonal experiments were designed to optimize the extraction process of total flavonoids from P.chinense Pursh with the volume fraction of ethanol,the ratio of material to liquid,heating reflux extraction time and extraction times as factors,and the content of total flavonoids as the index.A verification test was carried out.The optimized extraction process was adopted to compare the contents of total flavonoids from different parts of P.chinense Pursh.[Results]The best extraction process was extracting the powder of P.chinense Pursh for 2.0 h with 20 times of 55%ethanol by reflux twice.Under this condition,the contents of total flavonoids were 3.63%,8.90%,11.28%,and 4.36%from stems,leaves,flowers and whole grass of P.chinense Pursh,respectively.[Conclusions]The process is reasonable,feasible and stable,and can effectively extract total flavonoids from P.chinense Pursh.The contents of total flavonoids from different parts of P.chinense Pursh were quite different,and the value was higher in the leaves and flowers,so the proportions of leaves and flowers should be paid attention to in the industrial processing of P.chinense Pursh.
文摘[Objectives]The extraction conditions of formula oolong tea were investigated by an orthogonal experiment.[Methods]The technical conditions were optimized by the 4C method,and the application of formula oolong tea extract in cigarettes was studied.[Results]①In the experimental range,the best sensory evaluation effect of formula oolong tea extract was obtained with extraction conditions of 70%ethanol as extraction solvent,extraction time h,extraction temperature 25℃,and ultrasonic frequency 80 kHz,and follow-up low-temperature concentration,low-temperature sedimentation and low-temperature centrifugation.②The effects of different centrifugal speeds on the quality of formula oolong tea extract were explored.The formula oolong tea extract obtained under the conditions of 3000 r/min and centrifugal time of 10 min showed the best evaluation effect with soft and delicate smoke,rich smoke fragrance,good comfort and refreshing mouthfeel.③The effective aroma components in the formula oolong tea extract were qualitatively analyzed by GC-MS.[Conclusions]This study provides high-quality raw materials and a theoretical basis for the research of independent flavor blending in cigarette industry enterprises.
文摘An experimental investigation was presented on the separation of Cu(Ⅱ), Zn(Ⅱ), and Cd(Ⅱ) from a rich sulfate leachate of zinc slag by solvent extraction. The results of orthogonal experiments indicate that LIX 984N is highly selective and very efficient in the extraction of Cu(Ⅱ), and the analysis of variance indicates that the sequence of parameters according to their influence on the separation efficiency is phase ratio 〉 LIX 984N concentration 〉 pH value 〉 extraction time. The optimal condition for copper extraction is obtained as 25% of LIX 984N concentration, 7 rain of extraction time, 3:2 of phase ratio O/A, and pH = 1.7. The separation of Zn(Ⅱ) and Cd(Ⅱ) was performed after the copper extraction from the raffinate. Comparative analysis of the separation with di-2-ethylhexyl phosphoric acid (D2EHPA), D2EHPA-tributyl- phosophate (TBP) synergistic extracting system, and 2-ethylhexyl phosphonic acid mono 2-ethylhexyl ester (HEHEHP) was made at pH = 2.0. It is demonstrated that the extraction efficiency with D2EHPA is improved after being saponified by sodium hydroxide, and D2EHPA-TBP synergistic extracting, as well as HEHEHP, has a superior selectivity to Zn(Ⅱ) over Cd(Ⅱ).

文摘[Objective] This study aimed to optimize the ultrasonic extraction process of total flavonoids from Angelica keiskei Koidzumi. [Method] Using the yield of total flavonoids as an indicator, the ultrasonic extraction process of total flavonoids from A. keiskei was optimized by orthogonal experimental design. [ Result ] Among four factors in orthogonal experimental design, extraction time exhibited the most significant effect on the extraction result; ethanol concentration exhib- ited a significant effect on the extraction result; ultrasonic power and solid-to-liquid ratio had no significant effect. The optimal conditions for ultrasonic extraction of total flavonoids from A. keiskei were extraction time of 25 min, ethanol concentration of 80%, ultrasonic power of 60 W, solid-to-liquid ratio of 1:20 (g/ml), under which the yield of total flavonoids reached 1.56%. [ Conclusion] This study provides the basis for further development and utilization of total flavonoids from A. keiskei.
文摘Water-soluble polysaccharide was isolated from grapefruit with the method of water-dissolving and ethanol-precipitating. By single factor test and orthogonal experimental method, the extraction conditions of the water-soluble polysaccharide were investigated, which contained liquid-solid ratio, temperature, time and pH. The results showed that pH was the most important factor of polysaccharide extraction, followed by temperature. The optimized extraction technology conditions were obtained. The liquid-solid ratio was 1:40, temperature was 90?C, time was 2.0 h, and pH was 2. The highest extraction rate of crude polysaccharide was 8.30%.

文摘Dried seaweed powder was used as the raw material to extract polysaccharides by ethanol precipitation after water bath. Extraction temperature, extrac- tion duration, solution pH and solid to liquid ratio were selected as technical parameters which required to be optimized. With the extraction rate of seaweed poly- saccharides as an index, single-factor experiment and orthogonal experiment were carried out to determine the optimal extraction technique. Seaweed polysaceharides were extracted with the optimal technical parameters to analyze the biological activities. Savage method was used to process seaweed polysaceharide extract. In vitro biological activities of preliminarily purified seaweed polysaeeharides were investigated successively, including reducing ability, antioxidant ability and antibacterial activity. The results showed that there was a positive correlation between polysaccharide concentration and its reducing ability, antioxidant ability and antibacterial activity. Furthermore, there was also a correlation among the reducing ability, antioxidant ability and antibacterial activity of seaweed polysaecharides; the relation- ship between each two of these three indicators was similar to positive linear correlation.

文摘[Objectives]To establish a qualitative identification and content determination method for psoralen and bergapten in the roots and leaves of Ficus pandurata,and the multi-indicator scoring method to optimize the ethanol extraction process for the effective parts of F.pandurata.[Methods]Qualitative identification was carried out by thin-layer chromatography(TLC)and content determination was made by high-performance liquid chromatography(HPLC);the content of psoralen and bergapten and extract yield were used as indicators,to investigate the effects of ethanol volume fraction,solid-to-liquid ratio,extraction time and extraction times on the ethanol extraction process of F.pandurata;multi-indicator scoring method was adopted,orthogonal test method was designed to optimize the extraction process,and verification test was performed.[Results]TLC chart of F.pandurata shows clear spots and good separation;the detected concentrations of psoralen and bergapten in the roots are in the range of 1.02-32.64μg/mL and shows a good linear relationship with the peak area(r=0.9997);these components have not been detected in the leaves,and the precision,RSD of stability and repeatability test is less than 2%;the average recovery rate was 99.8%-100.2%,and the RSD value is 1.12%-1.13%(n=6);the optimized extraction process is to use 50%ethanol as the extraction solvent,the solid-to-liquid ratio of 1∶10,reflux extraction of 3 times,and 1.5 h each time;the results of the three batches of verification tests show that the content of the two indicator components obtained is high,and the average value of the total amount is 0.34 mg/g(RSD=0.30%,n=3).[Conclusions]The established quality control method for F.pandurata is simple,easy,accurate and reproducible;the preferred extraction process is stable and feasible,suitable for the extraction and purification of coumarin effective parts in F.pandurata roots,so it is expected to provide references for further development of F.pandurata.

文摘[Objectives] The aims were to optimize the extraction process of selenoproteins from selenium-enriched rice in Guangxi and provide references for the intensive processing and comprehensive utilization of selenium protein resources. [Methods]Selenium-enriched rice was used as materials to extract selenoproteins by phosphate buffer extraction method and to optimize the extraction process of selenoproteins by using the orthogonal experiment. Proteins and selenium content was measured by Coomassie Brilliant Blue G-250 reagent and AFS( atomic fluorescence spectrometry) respectively. [Results] The most significant factor affecting extraction of rice Selenoproteins was extraction NaO H concentration,followed by the ratio of solid-liquid,temperature and then extraction time. The optimum extraction conditions of selenoproteins from rice were extraction temperature of 50 ℃,NaO H concentration of 0. 14 mol/L,extraction time of 5 h,and solid-liquid ratio of 1∶ 30. [Conclusions]The alkali extraction process optimized by orthogonal test could effectively improve the extraction rate of selenoproteins,and the optimized process parameters could be popularized and applied in practical production.

文摘[Objectives] To establish the high performance liquid chromatography( HPLC) determination method for oxysophocarpine and optimize the extraction and purification technology of oxysophocarpine from Sophora alopecuroides by inverse three-phase membrane cycle.[Methods]Based on the single-factor experiment,the effects of aqueous phase and organic phase volume ratio,the concentration of sodium hydroxide,concentration of hydrochloric acid and extraction cycle time on extraction process of oxysophocarpine were investigated using orthogonal design method,to determine the optimal extraction process. [Results]The oxysophocarpine was determined by Shim-pack VP-ODS chromatographic column( 4. 6 mm × 250 mm,5 μm),mobile phase was methanol-0. 2% phosphoric acid aqueous solution( 7∶ 93),gradient elution,flow rate was 1 m L/min,the sample size was 5 μm,column temperature was 30℃,detection wavelength was 221 nm. Aqueous phase and organic phase volume ratio was 1∶ 1,hydrochloric acid concentration was 0. 3 mol/L,sodium hydroxide concentration was 0. 75 mol/L,water pump flow rate was 6 m L/min,60 min cycle time. The extraction rate of oxysophocarpine 98. 21 % within 60 min under the best experimental conditions. Oxysophocarpine has good linearity relationship within the range of 0. 01-0. 7 mg/m L( r^2= 0. 9978,n = 6),the respective average recovery rate was 97. 47%( RSD = 1. 95%). [Conclusions] This extraction process is simple in operation,the organic solvent has low consumption,and can be used for extraction of alkaloid.

[Methods] Ethanol extraction method was adopted,spectrophotometry was used,and single factor experiment and orthogonal experiment were carried out to study the effects of ethanol percentage,extraction temperature,solid-to-liquid ratio and extraction time on the extraction of total flavonoids from leaves of C. lacryma-jobi L.[Results] The order of 4 factors influencing the extraction of flavonoids from leaves of C. lacryma-jobi L. was: solid-to-liquid ratio > extraction time > ethanol percentage > extraction temperature. When the extraction temperature was 70℃,the extraction time was 1. 5 h and the solid-liquid ratio was 1: 10,the ethanol percentage was 60%,the extraction effect was the best,extraction of flavonoids was 0. 107 5 mg/m L.[Conclusions] This study is expected to provide a theoretical basis for further development and utilization of C. lacryma-jobi L.
文摘[Objectives] The chlorogenic acid in Sambucus chinensis Lindl is a kind of biologically active substance with multiple pharmacological effects, but the content is relatively small. It is of great significance to find the best extraction process. Therefore, we designed single factor experiments and orthogonal experiments to screen the best conditions for extracting chlorogenic acid from S. chinensis. [Methods] With ethanol concentration, ultrasonic power, and extraction time as three single factors, single factor experiments were performed on S. chinensis, and its chlorogenic acid content was calculated from a standard curve. The contents of chlorogenic acid obtained under various conditions were compared, and then, orthogonal experiments were designed according to the orthogonal table to analyze the optimal extraction conditions of chlorogenic acid. [Results] The optimal extraction conditions of chlorogenic acid for S. chinensis were: the ethanol concentration 60%, the ultrasonic power 36 W, and the extraction time 40 min.[Conclusions] The factor that had the greatest effect on the extraction rate of chlorogenic acid in S. chinensis was ethanol concentration, followed by extraction time, and the least influencing factor was ultrasonic power. With 60% ethanol solution serving as the extractant, the ultrasonic power of 36 W, and the extraction time of 40 min, a better extraction rate was obtained. These conditions can be used as the best extraction conditions for chlorogenic acid.
