The influence of high and low methoxyl pectins on empirical and dynamic rheological properties of dough and Chinese steamed bread(CSB)quality were investigated. Different concentrations of high methoxyl apple pectin(HMAP), low methoxyl apple pectin(LMAP),and amidation apple pectin(ALMAP) were tested. Results from dynamic and empirical rheological tests indicated that all concentrations of pectins tested(0.2%, 0.6%, and 1%) negatively affected the gluten network. The low methoxyl pectins, LMAP and ALMAP, which had higher anionic densities, resulted in a pronounced weakening of the dough. Meanwhile, dough fermentation properties improved in the presence of appropriate pectins concentrations, and higher maximum dough height, stability of dough pore space, and gas retention were recorded. HMAP was the pectin most effective in influencing dough fermentation properties at all tested concentrations. Regarding CSB quality, textural properties, specific volume, moisture content, and water-holding capacity were analyzed. Generally, the textural properties of CSB were improved, including improved springiness and decreased hardness and chewiness, when 0.2%–0.6% HMAP or LMAP was used. The specific volume of CSB was increased by adding 0.2%–1% HMAP,0.6%–1% LMAP, or 0.2%–0.6% ALMAP. Moreover, CSB moisture loss was progressively reduced in the presence of different pectins at all tested concentrations during storage. In conclusion, pectins, especially HMAP, are good additives for improving the fermentation properties of dough and overall quality of CSB.展开更多

Furthermore, a growing number of mutants affecting cell wall composi- tion have begun to reveal the distinct contributions of different pectins to plant development. In the stem apex meristem (SAM) of the cucumber, epitopes of MAC 207, JIM 7, and JIM 5 were localized in the cells from second to third peripheral layers when the sex organ primodium began to differentiate; epitopes of MAC 207 and JIM 5 were also detected in the ragged edge cells. A very dense labeling signal with MAC 207 was observed in the carpel and pistil primodium. The AGP epitopes recognized by JIM 8 were localized in the anther of the male flower and the anther-like portion of the stagnant stamen of the female flower. This suggests that the AGPs and pectins in the SAM of the cucumber are closely associated with the differentiation of the SAM, from meristematic cells to floral primodium. This study aimed to characterize the physical-chemical and biological properties of pectin(PC)/chia seed mucilage(CM)membranes.PC/CM[100/0(control),80/20%,60/40%,and 40/60%w/w]membranes were prepared using the casting...This study aimed to characterize the physical-chemical and biological properties of pectin(PC)/chia seed mucilage(CM)membranes.PC/CM[100/0(control),80/20%,60/40%,and 40/60%w/w]membranes were prepared using the casting method.The membranes(PC/CM)were thin,yellow,lightly opaque(≈10%)and capable of blocking light UVB(between 66 at 52%).SEM analysis showed the presence of aggregates in the shape of a sphere(≈13μm)and ovoid(≈25μm).The proportion of 80/20 showed an increase in tensile strength(29%)and elastic modulus(19%)when compared to the control.FTIR analysis exhibited intermolecular interactions between PCPC,PC-CM,and CM-CM in the membranes.The thermal analysis(600°C)showed a slight improvement in the percentage of residual mass-loss of 3.31%(80/20)that control.The 40/60 membrane showed the lowest percentage of hemolysis(2.94%)but limited human albumin adsorption capacity.These results suggested that the blend PC/CM may be considered as a biomaterial for medical applications.展开更多

T herefore, the aim of this research was to explore whether there exist any bound lipases in rice bran(Oryza sativa). Three physical treatments(grinding, homogenizing and ultrasound crush)and 6 enzymatic treatments(cellulase, hemicellulase, pectinase, complex cellulase, glucoamylase and α-amylase)were applied to rice bran in order to investigate this bound lipase. The relative catalytic activities of extraction supernatant and residue for pectinase group were(437.63 ± 22.54)% and(159.26 ± 2.12)%, respectively, which were significantly higher(P < 0.05)than other groups. This phenomenon demonstrated that lipase was the most likely to combine with pectin. Molecular simulation proved that pectin could combine with two rice bran lipases(lipase 315 and lipase 308)and cover the catalytic centers so as to prevent the lipases from encountering the substrate and inhibiting their catalytic activities. During combination, pectin could make the lipases more compact and reduce the solvent accessible surface area of lipases, which would make the lipases inactive to molecular interaction. In summary, part of rice bran lipase was proved to exist in bound form and combined with the pectin.展开更多
Three pectin fractions(water-soluble fraction(WSF),chelator-soluble fraction(CSF),and sodium carbonatesoluble fraction(NSF))were obtained from Chinese dwarf cherry(Cerasus humilis)fruits.All of them were branched low ...Three pectin fractions(water-soluble fraction(WSF),chelator-soluble fraction(CSF),and sodium carbonatesoluble fraction(NSF))were obtained from Chinese dwarf cherry(Cerasus humilis)fruits.All of them were branched low methoxylated pectins with an amorphous or partially nanocrystalline nature and eight neutral monosaccharides(arabinose and galactose were most abundant).WSF,CSF and NSF had a degree of methylation(DM)of 35.82%,14.85%and 7.13%,uronic acid(UA)content of 76.02%,83.71%and 69.01%,and total protein content of 2.4%,2.1%and 8.8%,respectively.Their molecular weights were 340.31,330.16 and 141.31 kg/mol,respectively(analyzed by gel permeation chromatography(GPC)).WSF,CSF and NSF exhibited good rheological,thermal,emulsifying,emulsion-stabilizing,water-adsorbing,oil-binding,cholesterol-binding and antioxidant properties.NSF had the highest emulsifying,emulsion stabilizing,water-/oil-/cholesterol-binding and antioxidant capacities,followed by CSF.NSF had the highest viscosity(406.77 m Pa·s),flowability,and resistance to heat-induced changes/damage,which may be related to its lowest polydispersity index,DM and UA content and highest protein content.The three pectin fractions with desirable characteristics can be used as food additives/ingredients and dietary supplements.展开更多

Maint...Background: Due to worldwide increases in the prevalence of antibiotic-resistant bacteria, it is necessary to develop an active drug delivery system that can enable therapeutics to reach their molecular targets. Maintaining the concentration of any drug in the blood at a certain level for a long time is critical in the practice of drug therapy. With the increased frequency of drug use, the blood concentration of drugs exceeds the therapeutic level, leading to toxicity or ineffectiveness. To solve these problems, in recent years, much attention has been given to developing micro/nano preparations by encapsulating biologically active compounds on polymeric carriers. Therefore, we aimed to extract pectin from sea buckthorn peel, prepare microcapsules containing antibiotics, and determine their physical and chemical properties. Methods: Wastes were separated from sea buckthorn under “Medical raw materials Dry fruit of Hippophae rhamnoides MNS 5225:2002”. Pectin was isolated from sea buckthorn waste according to the “method for determination of pectins MNS3080:1981” standard. The degree of esterification was determined according to ISO 7623:2016. Antibiotic encapsulation with coacervates and water-based emulsions was performed. Antibiotic sensitivity was determined by microdilution according to the Clinical Laboratory Standard Institute (МТ100-S27) method. The results were determined between standard strains of Staphylococcus aureus ATCC 29213 and MRSA ATCC 2758 at different dilution concentrations. Result: Pectin is a brown powder with a sour taste and no odor. There was 71.4% esterification of pectin, 8.9% yield, 1.3% free carboxyl group, 3.2% methylated carboxyl group, 4.5% total carboxyl group, 3.5% ash, and 0.1% nitrogen. A study of the antibacterial activity of pectin containing doxycycline hyclate found that the inhibition of bacterial growth was 0.8 times less than that of pure pectin. It was 1 time less than that of doxycycline alone, and 33 times smaller than that of wontaxime when compared to pure pectin. Pectin containing doxycycline hyclate inhibited MRSA growth at a concentration 6 times lower than pure pectin. This was 2 times lower than doxycycline alone, and 8 times lower than wontaxime. Conclusion: Pectin yields 1.3 after 60 minutes of separation at a sediment concentration ratio of 1:1.15 and pH = 2. Pectin itself is antibacterial against MRSA.展开更多
AIM: To compare the effectiveness of standard triple, bismuth pectin quadruple and sequential therapies for Helicobacter pylori (H. pylori ) eradication in a randomized, double-blinded, comparative clinical trial in C...AIM: To compare the effectiveness of standard triple, bismuth pectin quadruple and sequential therapies for Helicobacter pylori (H. pylori ) eradication in a randomized, double-blinded, comparative clinical trial in China. METHODS: A total of 215 H. pylori -positive patients were enrolled in the study and randomly allocated into three groups: group A (n = 72) received a 10-d bismuth pectin quadruple therapy (20 mg rabeprazole bid , 1000 mg amoxicillin bid , 100 mg bismuth pectin qid , and 500 mg levofloxacin qd ); group B (n = 72) received the sequential therapy (20 mg omeprazole bid , 1000 mg amoxicillin bid , in 5 d, followed by 20 mg omeprazole bid , 500 mg tinidazole bid , 500 mg clarithromycin bid , for another 5 d); group C (n = 71) received a standard 1-wk triple therapy (20 mg omeprazole bid , 1000 mg amoxicillin bid , 500 mg clarithromycin bid ). After all these treatments, 20 mg omeprazole bid was administrated for 3 wk. H. pylori status was assessed by histology, 13C-urea breath test and rapid urease test at baseline and 4-6 wk after completion of treatment. Ulcer cicatrization was assessed by gastroscopy. χ 2 test (P < 0.05) was used to compare the eradication rates and ulcer cicatrisation rates among the three groups. RESULTS: The eradication rate was 83.33% (60/72) in group A, 88.89% (64/72) in group B, and 80.56% (58/71) in group C. The ulcer cicatrisation rate was 86.44% (51/59) in group A, 90.16% (55/61) in group B, and 84.91% (45/53) in group C. The sequential therapy yielded a higher eradication rate and ulcer cicatrisation rate than the standard triple and bismuth pectin quadruple therapies. Statistically, the eradication rate of group B was significantly different from groups A and C (P < 0.05), but the difference of ulcer cicatrisation rate and side effects was not statistically significant among the three groups (P > 0.05). The three protocols were generally well tolerated. CONCLUSION: The sequential therapy has achieved a significantly higher eradication rate, and is a more suitable first-line alternative protocol for anti-H. pylori infection compared with the standard triple and bismuth pectin quadruple therapies.展开更多

The obtained...The aim of this study was to investigate the effect of high-pressure homogenization on the droplet size and physical stability of different formulations of pectin–zein stabilized rice bran oil emulsions. The obtained emulsions, both before and after passing through highpressure homogenizer, were subjected to stability test under environmental stress conditions,that is, temperature cycling at 4 °C/40 °C for 6 cycles and centrifugal test at 3000 rpm for 10 min. Applying high-pressure homogenization after mechanical homogenization caused only a small additional decrease in emulsion droplet size. The droplet size of emulsions was influenced by the type of pectin used;emulsions using high methoxy pectin(HMP) were smaller than that using low methoxy pectin(LMP). This is due to a greater emulsifying property of HMP than LMP. The emulsions stabilized by HMP–zein showed good physical stability with lower percent creaming index than those using LMP, both before and after passing through high-pressure homogenizer. The stability of emulsions after passing through high-pressure homogenizer was slightly higher when using higher zein concentration, resulting from stronger pectin–zein complexes that could rearrange and adsorb onto the emulsion droplets.展开更多

The microbeads were fabricated by ionotropic gelation with...Novel oral microbeads were developed based on a biopolymer–drug conjugate of doxorubicin(DOX) conjugated with thiolated pectin via reducible disulfide bonds. The microbeads were fabricated by ionotropic gelation with cations such as Al3+, Ca2+ and Zn2+. The results showed that using zinc acetate can produce the strongest microbeads with spherical shape.However, the microbeads prepared from thiolated pectin–DOX conjugate were very soft and irregular in shape. To produce more spherical microbeads with suitable strength, the native pectin was then added to the formulations. The particle size of the microbeads ranged from 0.87 to 1.14 mm. The morphology of the microbeads was characterized by optical and scanning electron microscopy. DOX was still in crystalline form when used in preparing the microbeads, as confirmed by powder X-ray diffractometry. Drug release profiles showed that the microbeads containing thiolated pectin–DOX conjugate exhibited reduction-responsive character;in reducing environments, the thiolated pectin–DOX conjugate could uncouple resulting from a cleavage of the disulfide linkers and consequently release the DOX. The best-fit release kinetics of the microbeads containing thiolated pectin–DOX conjugate, in the medium without reducing agent, fit the Korsmeyer–Peppas model while those in the medium with reducing agent fit a zero-order release model. These results suggested that the microbeads containing thiolated pectin–DOX conjugate may be a promising platform for cancer-targeted delivery of DOX, exploiting the reducing environment typically found in tumors.展开更多

Furthermore, it can be used as in-feed antibiotic alternative. Chicory (Cichorium intybus L.) forage with high content of pectin (uronic acids as building blocks) is a novel class of dietary fiber that is chemically different from cereal grains (with high content of arabinoxylans). In the present study, we investigated effects of dietary inclusion of chicory forage on digestibility, gut morphology and microbiota in broilers and young pigs. In the chicken experiment, 160 1-d old broiler chicks were fed 3 nutritionally balanced diets for 30 d including a cereal-based diet and 2 diets with part of the cereals substituted with 60 and 120 g/k9 chicory forage (CF60 and CF120], whereas in the pig experiment, 18 seven-wk old Yorkshire pigs were fed 3 diets for 18 d including a cereal-based diet and 2 diets with 80 and 160 g/kg chicory forage inclusion (CF80 and CF160). Our results showed that young pigs were capable to utilize chicory forage well with higher total tract apparent digestibility (TTAD) of all fiber fractions, particularly uronic acid, compared with the control (P 〈 0.01). In contrast, a decreased TTAD of all fiber fractions was observed in chickens fed on diet CF120 (P 〈 0.05). Moreover, diet induced changes in gut morphology were observed in the large intestine of chickens. The alteration of cecal mucosal thickness was further positively correlated with TTAD of non-starch polysaccharides (NSP) and its constituent sugars (P 〈 0.05). In addition, in pigs, terminal restriction fragment length polymorphism (T-RFLP) analysis of intestinal microbiota revealed substantial dietary effects (cereal control diet vs. chicory forage inclusion) on the relative abundance of 2 dominant bacterial phylotypes (Prevotella sp. vs. Roseburia sp.) respectively (P 〈 0.05). In conclusion, our data showed that chicory forage (Cichorium intybus L.), a novel dietary fiber source in animal nutrition, have potential beneficial properties as fiber ingredient in diets for both pigs and chickens.展开更多

B. campestris ssp. chinensis). In the present paper, a 455-bp antisense cDNA fragment of BcMF3 was introduced to binary vector pB1121, and then was mobilized into Agrobacterium tumefaciens strain LBA4404. The A. tumefaciens harboring the BcMF3 antisense fragment was transformed to Arabidopsis thaliana by floral dip. Scanning electronic microscopy examination demonstrated that 47.8% of BcMF3 antisense pollen grains exhibited abnormal shape, which might lead to decreased germination of pollens, suggesting that the product of BcMF3 gene plays an important role during microsporogenesis. The evidence on burst of 45.7% of BcMF3 antisense pollen tubes in vitro and a majority of BcMF3 antisense pollens restricted within the stigmatic tissue revealed that BcMF3 is involved in aiding the growth of pollen tubes. The results suggest that BcMF3 acts at both stages of microsporogensis and pollen tube growth.展开更多

Bacillus megaterium HDYM-09 produced pectin lyase that exhibited the highest activity of 2116.71 ± 11.55 U/mL. Bacillus licheniformis HDYM-03 produced pectate lyase that exhibited the highest activity of 611.21 ± 14.54 U/mL. Based on these fi ndings, we constructed four retting systems to degrade the pectin substance. The results showed that the content of galacturonic acid in the mixed system was 529.21 μg/mL, the content of reducing sugar was 98.14 mg/mL, and the weight loss ratio of cells reached 19.49%, which were signifi cantly higher than those in other systems. The mixed system has more advantages, and the utilization rate of degumming was higher, which further ensured that the degumming can be carried out effi ciently and quickly. The mixed system exhibits feasible applications in the fi ber and textile industry.展开更多

The GC analysis indicated that it contains the monosaccharides of GalA, Gal, Ara and Rha. Their molar ratio is 2.10∶1.00∶0.12∶0.13. Partial hydrolysis with acid, pectinase treatment, periodate oxidation, Smith degradation, methylation analyses, GC/MS analyses and NMR analyses were used for the structure analyses of SB_~1-1 . The results reveal that SB_~1-1 has a lower branched structure. The main chain is composed of GalA and Gal; the inner part is α-1,4-linked-GalA; the border is 1,4-linked-Gal. FT-IR and HPGC are used individually to determine the structures and monosaccharide composition of apple pectin with different relative molecular weights. In the case of transmembrane pressure difference of 0.08 MPa, material concentration of 1 g·L^-1, and temperature of 50℃, 6 kinds of apple pectins with different molecular weights have been obtained. Molecular weight of apple pectin is correspondingly related to its physicochemical properties. The galacturonic acid contents and esterification and gelatination degrees increase relatively with an increase in molecular weight, and the monosaccharide composition relatively increase much more. And at the same time, the ultrafiltration has a better role to play in decoloring apple pectin solution. Accordingly, this research can be used as a new method for in-depth exploration of apple pectin.展开更多

文摘Dough preparation and performance are two critical factors influencing the final quality of wheat products. The influence of high and low methoxyl pectins on empirical and dynamic rheological properties of dough and Chinese steamed bread(CSB)quality were investigated. Different concentrations of high methoxyl apple pectin(HMAP), low methoxyl apple pectin(LMAP),and amidation apple pectin(ALMAP) were tested. Results from dynamic and empirical rheological tests indicated that all concentrations of pectins tested(0.2%, 0.6%, and 1%) negatively affected the gluten network. The low methoxyl pectins, LMAP and ALMAP, which had higher anionic densities, resulted in a pronounced weakening of the dough. Meanwhile, dough fermentation properties improved in the presence of appropriate pectins concentrations, and higher maximum dough height, stability of dough pore space, and gas retention were recorded. HMAP was the pectin most effective in influencing dough fermentation properties at all tested concentrations. Regarding CSB quality, textural properties, specific volume, moisture content, and water-holding capacity were analyzed. Generally, the textural properties of CSB were improved, including improved springiness and decreased hardness and chewiness, when 0.2%–0.6% HMAP or LMAP was used. The specific volume of CSB was increased by adding 0.2%–1% HMAP,0.6%–1% LMAP, or 0.2%–0.6% ALMAP. Moreover, CSB moisture loss was progressively reduced in the presence of different pectins at all tested concentrations during storage. In conclusion, pectins, especially HMAP, are good additives for improving the fermentation properties of dough and overall quality of CSB.

文摘Pectins are complex cell wall polysaccharides important for many aspects of plant development. Recent studies have discovered extensive physical interactions between pectins and other cell wall components,implicating pectins in new molecular functions. Pectins are often localized in spatially-restricted patterns, and some of these non-uniform pectin distributions contribute to multiple aspects of plant development, including the morphogenesis of cells and organs. Furthermore, a growing number of mutants affecting cell wall composi- tion have begun to reveal the distinct contributions of different pectins to plant development. This review discusses the interactions of pectins with other cell wall components, the functions of pectins in controlling cellular morphology, and how non-uniform pectin composition can be an important determinant of developmental processes.
文摘Abstract: Arabinogalactan proteins (AGPs) and pectins were detected in the floral buds of cucumber (Cucumis sativus L.) during its sex determination using the following monoclonal antibodies: MAC 207 (recognizes AGP epitopes); JIM 8 (recognizes a subset of AGP epitopes); and JIM 5 and JIM 7 (epitopes of pectins esterified to various degrees). In the stem apex meristem (SAM) of the cucumber, epitopes of MAC 207, JIM 7, and JIM 5 were localized in the cells from second to third peripheral layers when the sex organ primodium began to differentiate; epitopes of MAC 207 and JIM 5 were also detected in the ragged edge cells. A very dense labeling signal with MAC 207 was observed in the carpel and pistil primodium. The AGP epitopes recognized by JIM 8 were localized in the anther of the male flower and the anther-like portion of the stagnant stamen of the female flower. This suggests that the AGPs and pectins in the SAM of the cucumber are closely associated with the differentiation of the SAM, from meristematic cells to floral primodium. The subset of AGPs recognized by JIM 8 may play an important role in stamen formation.

文摘Background Pectin is a heteropolysaccharide that acts as an intestinal immunomodulator,promoting intestinal development and regulating intestinal flora in the gut.However,the relevant mechanisms remain obscure.In this study,pigs were fed a corn-soybean meal-based diet supplemented with either 5%microcrystalline cellulose(MCC)or 5%pectin for 3 weeks,to investigate the metabolites and anti-inflammatory properties of the jejunum.Result The results showed that dietary pectin supplementation improved intestinal integrity(Claudin-1,Occludin)and inflammatory response[interleukin(IL)-10],and the expression of proinflammatory cytokines(IL-1β,IL-6,IL-8,TNF-α)was down-regulated in the jejunum.Moreover,pectin supplementation altered the jejunal microbiome and tryptophan-related metabolites in piglets.Pectin specifically increased the abundance of Lactococcus,Enterococcus,and the microbiota-derived metabolites(skatole(ST),3-indoleacetic acid(IAA),3-indolepropionic acid(IPA),5-hydroxyindole-3-acetic acid(HIAA),and tryptamine(Tpm)),which activated the aryl hydrocarbon receptor(AhR)pathway.AhR activation modulates IL-22 and its downstream pathways.Correlation analysis revealed the potential relationship between metabolites and intestinal morphology,intestinal gene expression,and cytokine levels.Conclusion In conclusion,these results indicated that pectin inhibits the inflammatory response by enhancing the AhR-IL22-signal transducer and activator of transcription 3 signaling pathway,which is activated through tryptophan metabolites.

文摘This study aimed to characterize the physical-chemical and biological properties of pectin(PC)/chia seed mucilage(CM)membranes.PC/CM[100/0(control),80/20%,60/40%,and 40/60%w/w]membranes were prepared using the casting method.The membranes(PC/CM)were thin,yellow,lightly opaque(≈10%)and capable of blocking light UVB(between 66 at 52%).SEM analysis showed the presence of aggregates in the shape of a sphere(≈13μm)and ovoid(≈25μm).The proportion of 80/20 showed an increase in tensile strength(29%)and elastic modulus(19%)when compared to the control.FTIR analysis exhibited intermolecular interactions between PCPC,PC-CM,and CM-CM in the membranes.The thermal analysis(600°C)showed a slight improvement in the percentage of residual mass-loss of 3.31%(80/20)that control.The 40/60 membrane showed the lowest percentage of hemolysis(2.94%)but limited human albumin adsorption capacity.These results suggested that the blend PC/CM may be considered as a biomaterial for medical applications.

文摘Rice bran residue possessed a steady lipase activity((26.68 ± 3.69)%)after its endogenous lipase was extracted continuously by phosphate buffer solution(PBS)for 24 h. T herefore, the aim of this research was to explore whether there exist any bound lipases in rice bran(Oryza sativa). Three physical treatments(grinding, homogenizing and ultrasound crush)and 6 enzymatic treatments(cellulase, hemicellulase, pectinase, complex cellulase, glucoamylase and α-amylase)were applied to rice bran in order to investigate this bound lipase. The relative catalytic activities of extraction supernatant and residue for pectinase group were(437.63 ± 22.54)% and(159.26 ± 2.12)%, respectively, which were significantly higher(P < 0.05)than other groups. This phenomenon demonstrated that lipase was the most likely to combine with pectin. Molecular simulation proved that pectin could combine with two rice bran lipases(lipase 315 and lipase 308)and cover the catalytic centers so as to prevent the lipases from encountering the substrate and inhibiting their catalytic activities. During combination, pectin could make the lipases more compact and reduce the solvent accessible surface area of lipases, which would make the lipases inactive to molecular interaction. In summary, part of rice bran lipase was proved to exist in bound form and combined with the pectin.
文摘Three pectin fractions(water-soluble fraction(WSF),chelator-soluble fraction(CSF),and sodium carbonatesoluble fraction(NSF))were obtained from Chinese dwarf cherry(Cerasus humilis)fruits.All of them were branched low methoxylated pectins with an amorphous or partially nanocrystalline nature and eight neutral monosaccharides(arabinose and galactose were most abundant).WSF,CSF and NSF had a degree of methylation(DM)of 35.82%,14.85%and 7.13%,uronic acid(UA)content of 76.02%,83.71%and 69.01%,and total protein content of 2.4%,2.1%and 8.8%,respectively.Their molecular weights were 340.31,330.16 and 141.31 kg/mol,respectively(analyzed by gel permeation chromatography(GPC)).WSF,CSF and NSF exhibited good rheological,thermal,emulsifying,emulsion-stabilizing,water-adsorbing,oil-binding,cholesterol-binding and antioxidant properties.NSF had the highest emulsifying,emulsion stabilizing,water-/oil-/cholesterol-binding and antioxidant capacities,followed by CSF.NSF had the highest viscosity(406.77 m Pa·s),flowability,and resistance to heat-induced changes/damage,which may be related to its lowest polydispersity index,DM and UA content and highest protein content.The three pectin fractions with desirable characteristics can be used as food additives/ingredients and dietary supplements.
文摘Background: Due to worldwide increases in the prevalence of antibiotic-resistant bacteria, it is necessary to develop an active drug delivery system that can enable therapeutics to reach their molecular targets. Maintaining the concentration of any drug in the blood at a certain level for a long time is critical in the practice of drug therapy. With the increased frequency of drug use, the blood concentration of drugs exceeds the therapeutic level, leading to toxicity or ineffectiveness. To solve these problems, in recent years, much attention has been given to developing micro/nano preparations by encapsulating biologically active compounds on polymeric carriers. Therefore, we aimed to extract pectin from sea buckthorn peel, prepare microcapsules containing antibiotics, and determine their physical and chemical properties. Methods: Wastes were separated from sea buckthorn under “Medical raw materials Dry fruit of Hippophae rhamnoides MNS 5225:2002”. Pectin was isolated from sea buckthorn waste according to the “method for determination of pectins MNS3080:1981” standard. The degree of esterification was determined according to ISO 7623:2016. Antibiotic encapsulation with coacervates and water-based emulsions was performed. Antibiotic sensitivity was determined by microdilution according to the Clinical Laboratory Standard Institute (МТ100-S27) method. The results were determined between standard strains of Staphylococcus aureus ATCC 29213 and MRSA ATCC 2758 at different dilution concentrations. Result: Pectin is a brown powder with a sour taste and no odor. There was 71.4% esterification of pectin, 8.9% yield, 1.3% free carboxyl group, 3.2% methylated carboxyl group, 4.5% total carboxyl group, 3.5% ash, and 0.1% nitrogen. A study of the antibacterial activity of pectin containing doxycycline hyclate found that the inhibition of bacterial growth was 0.8 times less than that of pure pectin. It was 1 time less than that of doxycycline alone, and 33 times smaller than that of wontaxime when compared to pure pectin. Pectin containing doxycycline hyclate inhibited MRSA growth at a concentration 6 times lower than pure pectin. This was 2 times lower than doxycycline alone, and 8 times lower than wontaxime. Conclusion: Pectin yields 1.3 after 60 minutes of separation at a sediment concentration ratio of 1:1.15 and pH = 2. Pectin itself is antibacterial against MRSA.
文摘AIM: To compare the effectiveness of standard triple, bismuth pectin quadruple and sequential therapies for Helicobacter pylori (H. pylori ) eradication in a randomized, double-blinded, comparative clinical trial in China. METHODS: A total of 215 H. pylori -positive patients were enrolled in the study and randomly allocated into three groups: group A (n = 72) received a 10-d bismuth pectin quadruple therapy (20 mg rabeprazole bid , 1000 mg amoxicillin bid , 100 mg bismuth pectin qid , and 500 mg levofloxacin qd ); group B (n = 72) received the sequential therapy (20 mg omeprazole bid , 1000 mg amoxicillin bid , in 5 d, followed by 20 mg omeprazole bid , 500 mg tinidazole bid , 500 mg clarithromycin bid , for another 5 d); group C (n = 71) received a standard 1-wk triple therapy (20 mg omeprazole bid , 1000 mg amoxicillin bid , 500 mg clarithromycin bid ). After all these treatments, 20 mg omeprazole bid was administrated for 3 wk. H. pylori status was assessed by histology, 13C-urea breath test and rapid urease test at baseline and 4-6 wk after completion of treatment. Ulcer cicatrization was assessed by gastroscopy. χ 2 test (P < 0.05) was used to compare the eradication rates and ulcer cicatrisation rates among the three groups. RESULTS: The eradication rate was 83.33% (60/72) in group A, 88.89% (64/72) in group B, and 80.56% (58/71) in group C. The ulcer cicatrisation rate was 86.44% (51/59) in group A, 90.16% (55/61) in group B, and 84.91% (45/53) in group C. The sequential therapy yielded a higher eradication rate and ulcer cicatrisation rate than the standard triple and bismuth pectin quadruple therapies. Statistically, the eradication rate of group B was significantly different from groups A and C (P < 0.05), but the difference of ulcer cicatrisation rate and side effects was not statistically significant among the three groups (P > 0.05). The three protocols were generally well tolerated. CONCLUSION: The sequential therapy has achieved a significantly higher eradication rate, and is a more suitable first-line alternative protocol for anti-H. pylori infection compared with the standard triple and bismuth pectin quadruple therapies.
文摘Modified pectin cross-linked with adipic acid, was synthesized and used for heavy metal removal from wastewater. SEM and FTIR were used to investigate its structure and morphology. The modified pectin had a rough, porous phase covered with carboxy groups, resulting a high adsorption capacity. And at the room temperature, the saturated loading capacity for Pb^2+, Cu^2+ and Zn^2+ reached 1.82 retool/g, 1.794 mmol/g and 0.964 retool/g, respectively. The results proved its potential application to remove of the heavy metal.

文摘The aim of this study was to investigate the effect of high-pressure homogenization on the droplet size and physical stability of different formulations of pectin–zein stabilized rice bran oil emulsions. The obtained emulsions, both before and after passing through highpressure homogenizer, were subjected to stability test under environmental stress conditions,that is, temperature cycling at 4 °C/40 °C for 6 cycles and centrifugal test at 3000 rpm for 10 min. Applying high-pressure homogenization after mechanical homogenization caused only a small additional decrease in emulsion droplet size. The droplet size of emulsions was influenced by the type of pectin used;emulsions using high methoxy pectin(HMP) were smaller than that using low methoxy pectin(LMP). This is due to a greater emulsifying property of HMP than LMP. The emulsions stabilized by HMP–zein showed good physical stability with lower percent creaming index than those using LMP, both before and after passing through high-pressure homogenizer. The stability of emulsions after passing through high-pressure homogenizer was slightly higher when using higher zein concentration, resulting from stronger pectin–zein complexes that could rearrange and adsorb onto the emulsion droplets.
文摘Novel oral microbeads were developed based on a biopolymer–drug conjugate of doxorubicin(DOX) conjugated with thiolated pectin via reducible disulfide bonds. The microbeads were fabricated by ionotropic gelation with cations such as Al3+, Ca2+ and Zn2+. The results showed that using zinc acetate can produce the strongest microbeads with spherical shape.However, the microbeads prepared from thiolated pectin–DOX conjugate were very soft and irregular in shape. To produce more spherical microbeads with suitable strength, the native pectin was then added to the formulations. The particle size of the microbeads ranged from 0.87 to 1.14 mm. The morphology of the microbeads was characterized by optical and scanning electron microscopy. DOX was still in crystalline form when used in preparing the microbeads, as confirmed by powder X-ray diffractometry. Drug release profiles showed that the microbeads containing thiolated pectin–DOX conjugate exhibited reduction-responsive character;in reducing environments, the thiolated pectin–DOX conjugate could uncouple resulting from a cleavage of the disulfide linkers and consequently release the DOX. The best-fit release kinetics of the microbeads containing thiolated pectin–DOX conjugate, in the medium without reducing agent, fit the Korsmeyer–Peppas model while those in the medium with reducing agent fit a zero-order release model. These results suggested that the microbeads containing thiolated pectin–DOX conjugate may be a promising platform for cancer-targeted delivery of DOX, exploiting the reducing environment typically found in tumors.

文摘Dietary fiber, resistant to host-mediated digestion in the small intestine due to lack of endogenous enzymes, impacts many facets of animal health and is associated with gut development especially in young monogastrics. Furthermore, it can be used as in-feed antibiotic alternative. Chicory (Cichorium intybus L.) forage with high content of pectin (uronic acids as building blocks) is a novel class of dietary fiber that is chemically different from cereal grains (with high content of arabinoxylans). In the present study, we investigated effects of dietary inclusion of chicory forage on digestibility, gut morphology and microbiota in broilers and young pigs. In the chicken experiment, 160 1-d old broiler chicks were fed 3 nutritionally balanced diets for 30 d including a cereal-based diet and 2 diets with part of the cereals substituted with 60 and 120 g/k9 chicory forage (CF60 and CF120], whereas in the pig experiment, 18 seven-wk old Yorkshire pigs were fed 3 diets for 18 d including a cereal-based diet and 2 diets with 80 and 160 g/kg chicory forage inclusion (CF80 and CF160). Our results showed that young pigs were capable to utilize chicory forage well with higher total tract apparent digestibility (TTAD) of all fiber fractions, particularly uronic acid, compared with the control (P 〈 0.01). In contrast, a decreased TTAD of all fiber fractions was observed in chickens fed on diet CF120 (P 〈 0.05). Moreover, diet induced changes in gut morphology were observed in the large intestine of chickens. The alteration of cecal mucosal thickness was further positively correlated with TTAD of non-starch polysaccharides (NSP) and its constituent sugars (P 〈 0.05). In addition, in pigs, terminal restriction fragment length polymorphism (T-RFLP) analysis of intestinal microbiota revealed substantial dietary effects (cereal control diet vs. chicory forage inclusion) on the relative abundance of 2 dominant bacterial phylotypes (Prevotella sp. vs. Roseburia sp.) respectively (P 〈 0.05). In conclusion, our data showed that chicory forage (Cichorium intybus L.), a novel dietary fiber source in animal nutrition, have potential beneficial properties as fiber ingredient in diets for both pigs and chickens.

文摘Pectins had been extracted from shaddock raw material with 0.01 mol·L^-1 HCI and with three enzymic preparations including cellulase from aspergillus oryzaei.β -glucosidase from almonds and protopectinases-T, a protopectin-solubilizing enzyme, which did not degrade polygalacturonic acid, The effects of enzymic and acid soluhilization of pectin from shaddock materials were compared, The yield of pectin preparations, degree of acetylation, gelling strength, molecular weight and viscosity were determined. Enzymic methods of extraction gave much bigger yields of pectic substances. Compared with the pectin extracted with hydrochloric acid, all the samples derived from shaddock raw material with enzymic preparations had a significant lower D-galacturonic acid content.

文摘The gastrointestinal tract of livestock and poultry is prone to challenge by feedborne antigens,pathogens,and other stress factors in the farm environment.Excessive physiological inflammation and oxidative stress that arises firstly disrupts the intestinal epithelial barrier followed by other components of the gastrointestinal tract.In the present review,the interrelationship between intestinal barrier inflammation and oxidative stress that contributes to the pathogenesis of inflammatory bowel disease was described.Further,the role of naturally existing immunomodulatory nutrients such as the omega-3 polyunsaturated fatty acids,citrus pectin,and milk-derived exosomes in preventing intestinal barrier inflammation was discussed.Based on the existing evidence,the possible molecular mechanism of these bioactive nutrients in the intestinal barrier was outlined for application in animal diets.

文摘In an effort to provide some information relevant to the molecular mechanism of genic male sterility in plants, BcMF3 gene that encodes a pectin methylesterase was isolated from the fertile B line of Chinese cabbage-pak-choi (Brassica rapa ssp. chinensis, syn. B. campestris ssp. chinensis). In the present paper, a 455-bp antisense cDNA fragment of BcMF3 was introduced to binary vector pB1121, and then was mobilized into Agrobacterium tumefaciens strain LBA4404. The A. tumefaciens harboring the BcMF3 antisense fragment was transformed to Arabidopsis thaliana by floral dip. Scanning electronic microscopy examination demonstrated that 47.8% of BcMF3 antisense pollen grains exhibited abnormal shape, which might lead to decreased germination of pollens, suggesting that the product of BcMF3 gene plays an important role during microsporogenesis. The evidence on burst of 45.7% of BcMF3 antisense pollen tubes in vitro and a majority of BcMF3 antisense pollens restricted within the stigmatic tissue revealed that BcMF3 is involved in aiding the growth of pollen tubes. The results suggest that BcMF3 acts at both stages of microsporogensis and pollen tube growth.

文摘This study demonstrated that Bacillus licheniformis HDYM-03 and Bacillus megaterium HDYM-09, isolated from a liquid sample of fl ax retting pool, were able to produce pectinolytic enzymes using polysaccharides as substrates. Bacillus megaterium HDYM-09 produced pectin lyase that exhibited the highest activity of 2116.71 ± 11.55 U/mL. Bacillus licheniformis HDYM-03 produced pectate lyase that exhibited the highest activity of 611.21 ± 14.54 U/mL. Based on these fi ndings, we constructed four retting systems to degrade the pectin substance. The results showed that the content of galacturonic acid in the mixed system was 529.21 μg/mL, the content of reducing sugar was 98.14 mg/mL, and the weight loss ratio of cells reached 19.49%, which were signifi cantly higher than those in other systems. The mixed system has more advantages, and the utilization rate of degumming was higher, which further ensured that the degumming can be carried out effi ciently and quickly. The mixed system exhibits feasible applications in the fi ber and textile industry.
文摘A water-soluble pectin SB_~1-1 was isolated and purified from the root of Panax ginseng C. A. Mey. The HPLC analysis indicates that SB_~1-1 is homogenous. Its molecular weight was estimated via gel filtration to be 10000. The GC analysis indicated that it contains the monosaccharides of GalA, Gal, Ara and Rha. Their molar ratio is 2.10∶1.00∶0.12∶0.13. Partial hydrolysis with acid, pectinase treatment, periodate oxidation, Smith degradation, methylation analyses, GC/MS analyses and NMR analyses were used for the structure analyses of SB_~1-1 . The results reveal that SB_~1-1 has a lower branched structure. The main chain is composed of GalA and Gal; the inner part is α-1,4-linked-GalA; the border is 1,4-linked-Gal. Some of the 1,4-linked-GalA and 1,4-linked-Gal residues are substituted at O6. On an average, there is one branch for every ten hexose residues. The side chain is composed of 1,6-linked-Gal and 1,3,6-linked-Gal. The nonreduced end is composed of Rha, Ara and Gal. The main glycosidic link of SB_~1-1 has an α configuration.

文摘Separation conditions of ultrafiltration for apple pectin, weights and their structures and properties are studied and relations between apple pectin with different relative molecular Five kinds of ultrafiltration membranes with different molecular weight cut-offs are used to separate apple pectin. FT-IR and HPGC are used individually to determine the structures and monosaccharide composition of apple pectin with different relative molecular weights. In the case of transmembrane pressure difference of 0.08 MPa, material concentration of 1 g·L^-1, and temperature of 50℃, 6 kinds of apple pectins with different molecular weights have been obtained. Molecular weight of apple pectin is correspondingly related to its physicochemical properties. The galacturonic acid contents and esterification and gelatination degrees increase relatively with an increase in molecular weight, and the monosaccharide composition relatively increase much more. And at the same time, the ultrafiltration has a better role to play in decoloring apple pectin solution. Accordingly, this research can be used as a new method for in-depth exploration of apple pectin.

文摘Mixed gels of chitosan-pectin were prepared by varying the ratio of constituents in the presence of NaCl. Mixed gel at 3%of total polysaccharide concentration with addtion of 12%NaCl showed a synergistic maximum when the ratio of chitosan to pectin was 60: 40. The effect of the polysaccharide concentration, the preparation temperature (T p), the time of incubation, balk salt concentration, the molecular weight and the degree of deacetylation of chitosan on gelation have been studied. Interaction mechanism between molecules of both polysaccharides was investigated by FT-IR spectrometry.
