In this study, segregating populations were constructed between EHel with extremely low ear height and B73. Through the SNP-based genotyping and phenotypic characterization, 13 QTL distributed on the chromosomes (Chrs) of Chr1, Chr2, Chr5-Chr8 were detected for four traits of internode no. above ear (INa), average internode length above ear (ILaa), internode no. below ear (INb), and average internode length below ear (ILab). Phenotypic variation explained (PVE) by a single QTL ranged from 6.82% (qILab2-2) to 12.99% (qILaa5). Zm00001d016823 within the physical region of qILaa5, the major QTL for ILaa with the largest PVE was determined as the candidate through the genomic annotation and sequence alignment between EHel and B73. Product of Zm00001d016823 was annotated as a WEB family protein homogenous to At1g75720. qRT-PCR assay showed that Zm00001d016823 highly expressed within the tissue of internode, exhibiting statistically higher expression levels among internodes of IN4 to IN7 in EHel than those in B73 (P Zm00001d016823 might provide novel insight into molecular mechanism beyond phytohormones controlling internode development in maize.展开更多

In recent years, CPE of type OXA-48 (Ambler class D) have been identified in Dakar. The aim of this study was to evaluate the phenotypic detection of OXA-48 CPE using a temocillin disc (30 μg). Methodology: A retrospective study was carried out at Medical Biology Laboratory of Pasteur Institute in Dakar on Ertapenem-Resistant Enterobacterales (ERE) strains isolated from 2015 to 2017. These strains were then tested with a 30 μg temocillin disc. Any strain resistant to temocillin (inhibition diameter Results: Forty-one ERE isolated during the study period were tested, of which 34 (82.9%) were OXA-48 based on phenotypic detection using temocillin disc and confirmed by PCR (100%). OXA-48 CPE strains detected were composed of Klebsiella pneumoniae (n = 14;41.2%), Enterobacter cloacae (n = 8;23.5%), Escherichia coli (n = 7, 20.5%), Citrobacter freundii (n = 3;8.8%), Cronobacter sakazakii (n = 1;3%) and Morganella morganii (n = 1;3%). Conclusion: Temocillin resistance has a good positive predictive value for detecting OXA-48 CPE by phenotypic method, confirmed by PCR. Temocillin is therefore a good marker for detection of OXA-48 CPE except Hafnia alvei.展开更多

The results showed that malic enzyme exists in heart, gonads, eyes and gills tissues in both female and male V. macrolepis. There was only one ME-2 in heart and ME-1 in eye of female fish, while there were ME-1 and ME-2 in heart and ME-2 in eyes of male fish. Ovary contained ME-1 and ME-2, while spermary only contained ME-1. In kidney and spleen tissues, malic enzyme was found only in female fish, while in muscle, malic enzyme existed only in male fish. The phenotypic of malic enzyme had obvious differences no matter in the same organ of different sexes in V. macrolepis or different organs in the same sex. But there were no differences among different individuals of the same sex. The results will provide basic data for developmental genetics, breed improvement and directed breeding of V. macrolepis, and lay a foundation for its development and protection.展开更多

To investigate the phenotypic plasticity in response to the heterogeneous soil water supply, ramet pairs of the species were subjected to heterogeneous water supply by which either mother ramets or daughter ramets were in high or low soil water supply, respectively, in the Maowusu (Mu Us) Sandy Land of Nei Mongol. The results showed that the phenotypic characteristics of the individual ramets of C epigejos were greatly influenced by the heterogeneous water supply. The ramets treated with high water supply significantly produced more new rhizomes and more offspring (ramets), and accumulated more shoot biomass, and allocated more biomass to their shoots than those treated with low water supply. In comparison with the daughter ramets in homogeneous soil water supply, phenotypic characteristics, in terms of new rhizome growth, the production of new offspring, and the biomass allocation pattern, of the daughter ramets within the pairs of the species were not significantly changed, no matter that high or low soil water supply to mother ramets. The phenotypic responses of mother ramets to soil water supply were similar to those of daughter ramets. From these results, it is inferred that the interconnected ramets of C epigejos response phenotypically to their local soil water rather than to the soil water experienced by the interconnected ramets. The interconnected ramets of C epigejos might be independent of each other in water relationship, although they are physically interconnected with rhizome segments. The physiological independence of interconnected ramets might facilitate the risk spreading and thus enhance the genet survivorship under the frequent drought stresses in Mu Us Sandland.展开更多

In December of 2006, 200 individuals were randomly sampled from the base population and subjected for correlation and path coefficient analysis. It was found that there were statistically significant phenotypic correlations among the traits (P 〈 0.01). Total weight was significantly and positively correlated with the shell length (r = 0.934 3), shell height (r = 0.895 9), shell width (r = 0.899 1 ), muscle weight (r = 0.882 0) and shell weight (r = 0.937 9), respectively. Shell length, shell width, muscle weight, shell height and shell weight had positive and direct effects on the total weight, with values of 0.397 1, 0.321 9, 0.172 1, 0.089 6 and 0.066 9, respectively. Shell length, shell width and muscle weight had higher direct effects on the total weight than shell height and shell weight. A combined evaluation of correlation, direct effects and indirect effects showed that direct selection for shell length, shell width, muscle weight, shell height and shell weight would be effective to improving the total weight. It was concluded that these traits could be regarded as the selection criteria in breeding programs of the species.展开更多

[Method] Phenotypic traits and genetic diversity were evaluated based on 5 quantitative traits and 21 qualitative traits of 162 accessions of Erianthus arundinaceum collected from nine provinces in China (74 in Yunnan, 15 in Fujian, 19 in Guizhou, 18 in Hainan, 14 in Sichuan, 10 in Jiangxi, 4 in Guangdong, 4 in Guangxi and 4 in Zhejiang). [Result] The Shannon-Wiener indices of qualitative traits among the E. arundinaceum populations from different provinces were generally low. The accessions from Fujian exhibited the highest genetic diversity index (0.762 4), while those from Guangxi showed the lowest one (0.294 2). There were great genetic variances in quantitative traits, with mean variation coefficients ranging from 14.95% to 32.15%. The accessions from Yunnan showed the highest variation coefficient (32.15%) and those from Guangxi showed the lowest one (14.95%). Brix exhibited extremely significant nega- tive correlation with altitude and plant height showed extremely significant negative correlation with latitude. Coefficients of genetic divergence indicated that a high proportion of total genetic variation was retained within the populations from different regions, and the high gene flow showed that there were active genetic exchanges among these populations, suggesting no significant genetic divergence among these populations. According to genetic distance and UPGMA, there was a certain correlation between genetic distance and different sampling regions of E. arundinaceum. [Conclusion] The results provide theoretical references for resource collection , het- erosis application and development of excellent genes of sugarcane germplasm.展开更多

To analyze the degree and pattern of phenotypic variation in leaves of Tetracentron sinense Oliv from the perspective of genetic and environmental adaptation and thus contribute to effective evidence-based conservatio...To analyze the degree and pattern of phenotypic variation in leaves of Tetracentron sinense Oliv from the perspective of genetic and environmental adaptation and thus contribute to effective evidence-based conservation and management strategies for germplasm resources,we measured 17 morphological and epidermal micromorphological leaf traits from 24 natural populations of T.sinense.Nested analysis of variance,multiple comparison,principal component analysis(PCA),cluster analysis,and correlation analysis were used to explore phenotypic leaf variation among and within populations and potential correlations with geographic and environmental factors.There were significant differences in 17 leaf phenotypic traits among and within populations.The mean phenotypic differentiation coefficient of the 17 traits was 56.34%,and the variation among populations(36.4%)was greater than that within populations(27.2%).The coefficient of variation(CV)of each trait ranged from 4.6 to 23.8%,and the mean was 11.8%.Phenotypic variation of leaves was related to environmental factors such as average annual sunshine hours,average July temperature,and average annual rainfall.The variation changed along gradients of longitude,latitude,and altitude.The PCA clustered the 24 natural populations into four groups.Our study suggests that phenotypic variation in T.sinense occurred primarily among populations,with moderate levels of phenotypic differentiation among populations and low levels of phenotypic variation within populations.The plant’s poor adaptability to the environment is likely an important contributor to its endangerment.Accordingly,conservation strategies are proposed to protect and manage the natural populations of T.sinense.展开更多

Yang is a rare and protected plant in China. This study was conducted to determine the phenotypic variation in this species and to document phenotypic variation within and among populations of P. bournei. Nested analysis of variance, coefficient of variation, multiple comparisons, principal component analysis(PCA) and correlation analysis were used to analyze ten phenotypic traits in ten natural populations of P.bournei from both the northeastern and the primary region of the range of this species. Significant differences among and within populations were observed in leaf and seed phenotypic traits. Variation among populations(34.92%)was greater than that within populations(26.19%). The mean phenotypic differentiation coefficient was 53.77% among populations, indicating that variation among populations comprised the majority of the phenotypic variation of P. bournei. The coefficient of variance(CV) of ten traits varied from 6.44 to 18.45%, with an average of 12.03%.The CV of leaf traits among populations(15.64%) was higher than that of seed traits(8.60%), indicating that seed traits were more stable. The results from CV and PCA indicated that leaf area, leaf length and thousand seed weight were the main factors accounting for the observed phenotypic variations. Significant or highly significant correlations were observed among most leaf and/or in seed phenotypic traits, whereas no significant correlations were observed between phenotypic traits and geographic factors.Based on cluster analysis, the ten populations can be divided into three clusters. METHODS: Cultures of MAPC and MSC were estab...AIM: To compare the phenotypic and neural differentiation potential of human bone marrow derived multipotent adult progenitor cells (MAPC) and mesenchymal stem cells (MSC). METHODS: Cultures of MAPC and MSC were established in parallel from same samples of human bone marrow (n = 5). Both stem cell types were evaluated for expression of pluripotency markers including Oct-4 and Nanog by immunocytochemistry and reversetranscription polymerase chain reaction (RT-PCR) and expression of standard mesenchymal markers including CD14, CD34, CD44, CD45, CD73, CD90, CD105 andhuman leukocyte antigen (HLA)-ABC by flow cytometry. After treatment with neural induction medium both MAPC and MSC were evaluated for expression of neural proteins [neuronal filament-200 (NF-200) and glial fibrillar acidic protein (GFAP)] by immunocytochemistry and Western blotting and neural genes [NF-200, GFAP, Tau, microtubule-associated protein (MAP)-1B, MAP-2, neuron-specific enolase (NSE) and oligodendrocyte-1 (Olig-1)] by quantitative real-time-PCR. RESULTS: MAPC had small trigonal shaped while MSC had elongated spindle-shaped morphology. The MAPC expressed Oct-4 and Nanog both at gene and protein levels, whereas MSC were negative for these pluripotent markers. MAPC were negative for HLA-ABC while MSC had high expression of HLA-ABC. In addition, MAPC as compared to MSC had significantly lower expression of CD44 (36.56% ± 1.92% vs 98.23% ± 0.51%), CD73 (15.11% ± 2.24% vs 98.53% ± 2.22%) and CD105 (13.81% ± 3.82%vs 95.12% ± 5.65%) (P < 0.001, for all) MAPC cultures compared to MSC cultures treated with neural induction medium had significantly higher fold change expression of NF-200 (0.64), GFAP (0.52), Tau (0.59), MAP-2 (0.72), Olig-1 (0.18) and NSE (0.29) proteins (P < 0.01 for Olig-1 and P < 0.001 for rest) as well as higher fold change expression of genes of NF-200 (1.34),GFAP (1.12),Tau (1.08),MAP-1B (0.92), MAP-2 (1.14) andNSE (0.4) (P < 0.001 for all). CONCLUSION: MAPC can be differentially characterized from MSC as Oct-4 and Nanog positive stem cells with no expression of HLA-ABC and low expression of mesenchymal markers CD44, CD73 and CD105 and when compared to MSC they possess greater predilection for differentiation into neuro-ectodermal lineage.展开更多
Germplasm resources are an important basis for genetic breeding and analysis of complex traits,and research on genetic diversity is conducive to the exploration and creation of new types of germplasm.In this study,the...Germplasm resources are an important basis for genetic breeding and analysis of complex traits,and research on genetic diversity is conducive to the exploration and creation of new types of germplasm.In this study,the distribution frequency,coefficient of variation,Shannon-Wiener index,and variance and cluster analyses were used to analyze the diversity and trait differences of 39 fruit phenotypic traits from 570 pear accessions,which included 456 pear accessions from 11 species and 114 interspecific hybrid cultivars that had been stored in the National Germplasm Repository of Apple and Pear(Xingcheng,China).The comprehensive evaluation indices were screened by correlation,principal component and regression analyses.A total of 132 variant types were detected in 28 categorical traits of pear germplasm fruit,which indicate a rich diversity.The diversity indices in decreasing order were:fruit shape(1.949),attitude of calyx(1.908),flesh texture type(1.700),persistency of calyx(1.681),russet location(1.658),relief of area around eye basin(1.644),flavor(1.610)and ground color(1.592).The coefficient of variation of titratable acidity in the 11 numerical traits of pear germplasm fruit was as high as 128.43%,which could more effectively reflect the differences between pear accessions.The phenotypic differentiation coefficient V_(st)(66.4%)among the five cultivated pear species,including Pyrus bretschneideri(White Pear),P.pyrifolia(Sand Pear),P.ussuriensis(Ussurian Pear),P.sinkiangensis(Xinjiang Pear),and P.communis(European Pear),was higher than the within population phenotypic differentiation coefficient V_(st)(33.6%).The variation among populations was the main source of variation in pear fruit traits.A hierarchical cluster analysis divided the 389 accessions of six cultivated pear species,including P.pashia(Himalayan Pear),into six categories.There were certain characteristics within the populations,and the differences between populations were not completely clustered by region.For example,Sand Pear cultivars from Japan and the Korean Peninsula clustered together with those from China.Most of the White Pear cultivars clustered with the Sand Pear,and a few clustered with the Ussurian Pear cultivars.The Ussurian Pear and European Pear cultivars clustered separately.The Xinjiang Pear and Himalayan Pear did not cluster together,and neither did the cultivars.Seventeen traits,three describing fruit weight and edible rate(fruit diameter,fruit length and fruit core size),five describing outer quality and morphological characteristics(over color,amount of russeting,dot obviousness,fruit shape,and stalk length),and nine describing inner quality(flesh color,juiciness of flesh,aroma,flavor,flesh texture,flesh texture type,soluble solid contents,titratable acidity,and eating quality)were selected from the 39 traits by principal component and stepwise regression analyses.These 17 traits could reflect 99.3%of the total variation and can be used as a comprehensive evaluation index for pear germplasm resources.展开更多

Vass is a dwarf clonal shrubby legume developed on the windward slopes of mobile or semi-fixed sand dunes of Central Asia. It is assumed that E. songoricum must possess a high degree of phenotypic plasticity for such a heterogeneous habitat. The variations of E. songoricum plants growing in two typical microhabitats (the upper slope and the lower slope of semi-mobile dune) were investigated. The morphological characteristics and the biomass allocation patterns were measured and compared at the clonal fragment level. Compared with the clonal fragment on the lower slope of dune, the clonal fragment on the upper slope of the dune (a) declined in total biomass and ramet height, (b) increased the length of rhizomes and the number of roots, (c) increased the degree of asym- metry, and (d) heightened allocation to the belowground biomass. Our results confirmed the hypothesis of high phenotypic adjustment capacity of E. songoricum to habitat moisture availability. Phenotypic plasticity of E. songoricum contributed to reduce the damage risk, led to an environmentally induced specialization in function of resources exploitation, resulted in its persistence in heterogeneous environments and was adaptive in sand dune environment.展开更多

A total of 290 alleles were detec...Diversity of 60 conventional japonica rice accessions with good eating quality at home and abroad was analyzed using SSR molecular markers, agronomic traits and taste characteristics. A total of 290 alleles were detected in the 60 accessions at 72 SSR loci with the high similarity coefficients varying between 0.600 and 0.924. The loci on chromosome 5 showed the greatest value in average allele number. Additionally, most of the SSR loci could detect 3 to 4 alleles. An UPGMA dendrogram based on the cluster analysis of the genetic similarity coefficients showed that the grouping trend of part of the rice accessions was geographic-related and most of the rice accessions in Jiangsu Province, China were clustered together. Furthermore, many domestic accessions from south and north origins in China were close to the foreign japonica rice varieties, as proved by their pedigree origin from the foreign high-quality sources. For taste characteristics, part of the accessions with excellent taste were clearly clustered into one category though they came from different geographical regions, which indicates that taste characteristics of some varieties were mainly genetically determined. In addition, the agronomic traits of japonica rice with good taste might be closely related with their geographical origins, but the relationship between superior taste characteristics and agronomic traits should be further clarified.展开更多

The aim was to assess the extent and patterns of agronomically important phenotypic variation across districts of origin and altitude classes for major qualitative traits using diversity index and multivariate methods. Eight qualitative and three quantitative traits were scored for 2740 plants and analyzed for diversity. The Shannon–Weaver diversity(H′) index was used to estimate phenotypic diversity. The estimated H′ ranged from monomorphic for glume hairiness to highly polymorphic for other traits. The highest(0.86) H′ was obtained for seed degree of shriveling, possibly indicating the differential responses of the genotypes to water deficit during later growth stages. With respect to district of origin, the highest(0.72) and lowest(0.44) H′ values were obtained for the Bale and SNNP districts, respectively. With respect to altitude, the highest(0.76) and lowest(0.62) H′ values were recorded for altitudes 1600–2000and > 3000 m above sea levels, respectively. Principal components analysis explained substantial variation contributed by district of origin and altitude range. Genotypes were clustered into three groups by districts of origin and altitude class, with relatively strong bootstrap values of 57 and 62 for the former and latter, respectively. It could be concluded that Ethiopian durum wheat landraces are very diverse both within and among districts of origin and altitude classes. This wealth of genetic diversity should be exploited for wheat improvement of yield and for resistance to biotic and abiotic stresses, particularly terminal drought.展开更多

However, the dysfunction of DCs can be possibly reversed by the stimulation of antigen peptides. In this study, DCs were cultured from peripheral blood monocytes (PBMCs) in patients with CHB in vitro, and the expression of phenotypic molecules on DCs loaded by different concentrations of HBsAg was observed. METHODS: Forty patients with CHB were divided randomly into 4 groups(10 patients in each group). PBMCs were isolated, and DCs were cultured after addition of granulocyte/macrophage colony-stimulating factor (GMCSF) and interleukin 4(IL-4). On the 9th day, DCs of the experimental groups were loaded at HBsAg concentrations of 2.5mg/L, 5mg/L and 10mg/L for 24 hours, whereas those of the control group were not loaded. An electron microscope was used to analyze the morphological changes of the DCs. The expression of phenotypic molecules on DCs in different groups was detected with flow cytometry. RESULTS: A combination of GM-CSF and IL-4 produced DCs from PBMCs in patients with CHB after being cultured for 9 days, whose morphological changes were tested by an electron microscope. The expression of phenotypic molecules on DCs in the control group was as low as CD83 (8.02±3.99)%, CD80(8.77±2.06)%, and MHC-DR (14.05±2.66)%. Loaded by different concentrations of HBsAg, the up-regulation of phenotypic molecules on DCs was found, with CD83(18.35±2.93)%, CD80(42.63±7.15)% and MHC-DR(47.49±6.59)% in 2.5mg/L HBsAg loading group, CD83(17.88±3.12)%, CD80(45.24± 10.93)% and MHC-DR(47.07±8.52)% in 5mg/L HBsAg loading group and CD83(16.74±2.86)%, CD80(44.59±6.99)% and MHC-DR(48.59±7.42)% in 10mg/L HBsAg loading group, respectively. Compared with the control group, the phenotypic molecules in the experimental groups were all different significantly (P<0.01), but among them, there were no differences (P>0.05). CONCLUSIONS: DCs cultured from PBMCs in the patients with CHB under the conditions of GM-CSF and IL-4 present on the typical dendritic morphology but are immature for expressing low phenotypic molecules. Loaded by different concentrations of HBsAg, the immature DCs can differentiate to mature DCs for expressing increasing phenotypic molecules.展开更多
Prior knowledge of the correlations among commercially important traits in Glyptocidaris crenularis can be used to guide development of breeding and aquaculture programs for this species.We evaluated the phenotypic co...Prior knowledge of the correlations among commercially important traits in Glyptocidaris crenularis can be used to guide development of breeding and aquaculture programs for this species.We evaluated the phenotypic correlations between somatic and gonadal traits in G.crenularis during two sample periods(October 2010 and April 2011).The coeffi cients of variation(CV%)for body weight(BW),redness(a*),yellowness(b*),gonad wet weight(GW),and gonad index(GI)were>20% in both periods,while those of other traits were very low.GW was signifi cantly correlated with somatic traits(diameter,height,and body weight)in both periods,although the correlation coeffi cients ranged from 0.473 to 0.636(P<0.01).Gonad moisture content(GC),a*,and lightness(L*)were not signifi cantly correlated with somatic traits(P>0.05).The color trait b* was negatively correlated with somatic traits(test diameter,test height,and body weight)during the period when the gonads were well developed(April 2011,P<0.05).We observed little difference in the nature of the phenotypic correlations between the two sample periods.The high variation in the somatic and gonad weight traits in G.crenularis suggests there is considerable potential for selective breeding.However,our results suggest that gonad quality traits cannot be selected indirectly by selecting for body weight.Our study provides an insight into the method for improving the breeding and aquaculture of G.crenulari.展开更多
In order to reveal the genetic diversity and variation sources of leaf phenotypic traits in walnut,the coefficient of variation,Shannon-weaver index,phenotypic differentiation coefficient,and the correlation of 9 leaf...In order to reveal the genetic diversity and variation sources of leaf phenotypic traits in walnut,the coefficient of variation,Shannon-weaver index,phenotypic differentiation coefficient,and the correlation of 9 leaf phenotypic traits for 251 walnut germplasm resources from two walnut populations were analyzed.The results showed that the averages of the variation coefficients and Shannon-weaver indexes of the leaf phenotype traits were 17.45%and 1.86 respectively.The F values of most inter-population traits and all intra-population traits were highly significant,and the F values of inter-population leaf shape index,leaflet number,length and leaf length were very large.The averages of inter-population and intra-population differentiation coefficients were 9.15%and 90.85%respectively.The leaf size,leaf thickness,and leaflet number were highly correlated with trunk perimeters.According to the results of principal component analyses,the 9 traits can be simplified into four principal components,which represent leaf size,petiole,leaflet number and leaf shape,respectively.There were extensive variations and rich genetic diversities in leaf phenotypic traits of walnut,and the intra-population variation was the leading source of phenotypic trait variation.Leaf shape index,leaflet number,internode length and leaf length can be used for population classification.The size and thickness of leaves and the number of leaflets will affect the accumulation of nutrients in walnut tree,and then affect its trunk perimeter.展开更多
Background The genomes of worldwide poultry breeds divergently selected for performance and other phenotypic traits may also be affected by,and formed due to,past and current admixture events.Adaptation to diverse env...Background The genomes of worldwide poultry breeds divergently selected for performance and other phenotypic traits may also be affected by,and formed due to,past and current admixture events.Adaptation to diverse environments,including acclimation to harsh climatic conditions,has also left selection footprints in breed genomes.Results Using the Chicken 50K_CobbCons SNP chip,we genotyped four divergently selected breeds:two aboriginal,cold tolerant Ushanka and Orloff Mille Fleur,one egg-type Russian White subjected to artificial selection for cold tolerance,and one meat-type White Cornish.Signals of selective sweeps were determined in the studied breeds using three methods:(1)assessment of runs of homozygosity islands,(2)F_(ST) based population differential analysis,and(3)haplotype differentiation analysis.Genomic regions of true selection signatures were identified by two or more methods or in two or more breeds.In these regions,we detected 540 prioritized candidate genes supplemented them with those that occurred in one breed using one statistic and were suggested in other studies.Amongst them,SOX5,ME3,ZNF536,WWP1,RIPK2,OSGIN2,DECR1,TPO,PPARGC1A,BDNF,MSTN,and beta-keratin genes can be especially mentioned as candidates for cold adaptation.Epigenetic factors may be involved in regulating some of these important genes(e.g.,TPO and BDNF).Conclusion Based on a genome-wide scan,our findings can help dissect the genetic architecture underlying various phenotypic traits in chicken breeds.These include genes representing the sine qua non for adaptation to harsh environments.Cold tolerance in acclimated chicken breeds may be developed following one of few specific gene expression mechanisms or more than one overlapping response known in cold-exposed individuals,and this warrants further investigation.展开更多

Walp) cultivars that grown in different sites of Fezzan (Southern part of Libya) ...Symbiotic and phenotypic characteristics of thirty rhiobial isolates obtained from root nodules of two cowpea (Vigna unguiculata L. Walp) cultivars that grown in different sites of Fezzan (Southern part of Libya) were studied. Cultural characteristics and cross-nodulation with Arachis hypogega and Faidherbia albida showed that they were slow-growing rhizobia. Each isolate was found to coexist with non-symbiotic bacteria similar in their cultural characteristics to fast-growing rhizobia. All isolates formed symbiosis with the test plants, but different in their nitrogen-fixation efficiency. Numerical analysis of phenotypic characteristics showed that at boundary level of 70% average similarity, the isolates formed four distinguished groups and two isolates remained separate. Most isolates exhibited wide tolerance to acidity, alkalinity and extreme temperatures. They also resistant to some heavy metals such as mercury, copper, zinc, lead, cadmium and aluminum at low concentrations and antibiotics like polymyxin, colistin, bacitracin and nalidixic acid. Isolates displayed different response to salinity ranging from sensitive, which unable to grow in 1% NaCI to resistant and grow at 2% NaCl or above. Urea was hydrolyzed by most of them and carbohydrates utilizations were different. Sucrose and maltose were metabolized by most of the test isolates, whereas, monosaccharide and sugar alcohols were poorly utilized.展开更多
文摘Internode number and length are the foundation to constitute plant height, ear height and the above-ground spatial structure of maize plant. In this study, segregating populations were constructed between EHel with extremely low ear height and B73. Through the SNP-based genotyping and phenotypic characterization, 13 QTL distributed on the chromosomes (Chrs) of Chr1, Chr2, Chr5-Chr8 were detected for four traits of internode no. above ear (INa), average internode length above ear (ILaa), internode no. below ear (INb), and average internode length below ear (ILab). Phenotypic variation explained (PVE) by a single QTL ranged from 6.82% (qILab2-2) to 12.99% (qILaa5). Zm00001d016823 within the physical region of qILaa5, the major QTL for ILaa with the largest PVE was determined as the candidate through the genomic annotation and sequence alignment between EHel and B73. Product of Zm00001d016823 was annotated as a WEB family protein homogenous to At1g75720. qRT-PCR assay showed that Zm00001d016823 highly expressed within the tissue of internode, exhibiting statistically higher expression levels among internodes of IN4 to IN7 in EHel than those in B73 (P Zm00001d016823 might provide novel insight into molecular mechanism beyond phytohormones controlling internode development in maize.
文摘Background: Nowadays, emergence of Carbapenemase-Producing Enterobacterales (CPE) throughout the world has become a public health problem, especially in countries with limited resources. In recent years, CPE of type OXA-48 (Ambler class D) have been identified in Dakar. The aim of this study was to evaluate the phenotypic detection of OXA-48 CPE using a temocillin disc (30 μg). Methodology: A retrospective study was carried out at Medical Biology Laboratory of Pasteur Institute in Dakar on Ertapenem-Resistant Enterobacterales (ERE) strains isolated from 2015 to 2017. These strains were then tested with a 30 μg temocillin disc. Any strain resistant to temocillin (inhibition diameter Results: Forty-one ERE isolated during the study period were tested, of which 34 (82.9%) were OXA-48 based on phenotypic detection using temocillin disc and confirmed by PCR (100%). OXA-48 CPE strains detected were composed of Klebsiella pneumoniae (n = 14;41.2%), Enterobacter cloacae (n = 8;23.5%), Escherichia coli (n = 7, 20.5%), Citrobacter freundii (n = 3;8.8%), Cronobacter sakazakii (n = 1;3%) and Morganella morganii (n = 1;3%). Conclusion: Temocillin resistance has a good positive predictive value for detecting OXA-48 CPE by phenotypic method, confirmed by PCR. Temocillin is therefore a good marker for detection of OXA-48 CPE except Hafnia alvei.

文摘By using polyacrylamide gel electrophoresis and biochemical straining method, malic enzyme isozyme in 10 organs of male or female Varicorhinus macrolepis were analyzed. The results showed that malic enzyme exists in heart, gonads, eyes and gills tissues in both female and male V. macrolepis. There was only one ME-2 in heart and ME-1 in eye of female fish, while there were ME-1 and ME-2 in heart and ME-2 in eyes of male fish. Ovary contained ME-1 and ME-2, while spermary only contained ME-1. In kidney and spleen tissues, malic enzyme was found only in female fish, while in muscle, malic enzyme existed only in male fish. The phenotypic of malic enzyme had obvious differences no matter in the same organ of different sexes in V. macrolepis or different organs in the same sex. But there were no differences among different individuals of the same sex. The results will provide basic data for developmental genetics, breed improvement and directed breeding of V. macrolepis, and lay a foundation for its development and protection.
文摘Calamagrostis epigejos (L.) Roth. is a perennial grass with slender and long rhizome segments between interconnected neighbor ramets. To investigate the phenotypic plasticity in response to the heterogeneous soil water supply, ramet pairs of the species were subjected to heterogeneous water supply by which either mother ramets or daughter ramets were in high or low soil water supply, respectively, in the Maowusu (Mu Us) Sandy Land of Nei Mongol. The results showed that the phenotypic characteristics of the individual ramets of C epigejos were greatly influenced by the heterogeneous water supply. The ramets treated with high water supply significantly produced more new rhizomes and more offspring (ramets), and accumulated more shoot biomass, and allocated more biomass to their shoots than those treated with low water supply. In comparison with the daughter ramets in homogeneous soil water supply, phenotypic characteristics, in terms of new rhizome growth, the production of new offspring, and the biomass allocation pattern, of the daughter ramets within the pairs of the species were not significantly changed, no matter that high or low soil water supply to mother ramets. The phenotypic responses of mother ramets to soil water supply were similar to those of daughter ramets. From these results, it is inferred that the interconnected ramets of C epigejos response phenotypically to their local soil water rather than to the soil water experienced by the interconnected ramets. The interconnected ramets of C epigejos might be independent of each other in water relationship, although they are physically interconnected with rhizome segments. The physiological independence of interconnected ramets might facilitate the risk spreading and thus enhance the genet survivorship under the frequent drought stresses in Mu Us Sandland.
文摘In April of 2006, a base population of the noble scallop Chlamys nobilis was established by collecting parental breeders from the stocks in Wushi, Zhanjiang. In December of 2006, 200 individuals were randomly sampled from the base population and subjected for correlation and path coefficient analysis. It was found that there were statistically significant phenotypic correlations among the traits (P 〈 0.01). Total weight was significantly and positively correlated with the shell length (r = 0.934 3), shell height (r = 0.895 9), shell width (r = 0.899 1 ), muscle weight (r = 0.882 0) and shell weight (r = 0.937 9), respectively. Shell length, shell width, muscle weight, shell height and shell weight had positive and direct effects on the total weight, with values of 0.397 1, 0.321 9, 0.172 1, 0.089 6 and 0.066 9, respectively. Shell length, shell width and muscle weight had higher direct effects on the total weight than shell height and shell weight. A combined evaluation of correlation, direct effects and indirect effects showed that direct selection for shell length, shell width, muscle weight, shell height and shell weight would be effective to improving the total weight. It was concluded that these traits could be regarded as the selection criteria in breeding programs of the species.

文摘[Objective] This study was conducted to utilize and estimate Erianthus arundinaceum resources and to develop their elite traits. [Method] Phenotypic traits and genetic diversity were evaluated based on 5 quantitative traits and 21 qualitative traits of 162 accessions of Erianthus arundinaceum collected from nine provinces in China (74 in Yunnan, 15 in Fujian, 19 in Guizhou, 18 in Hainan, 14 in Sichuan, 10 in Jiangxi, 4 in Guangdong, 4 in Guangxi and 4 in Zhejiang). [Result] The Shannon-Wiener indices of qualitative traits among the E. arundinaceum populations from different provinces were generally low. The accessions from Fujian exhibited the highest genetic diversity index (0.762 4), while those from Guangxi showed the lowest one (0.294 2). There were great genetic variances in quantitative traits, with mean variation coefficients ranging from 14.95% to 32.15%. The accessions from Yunnan showed the highest variation coefficient (32.15%) and those from Guangxi showed the lowest one (14.95%). Brix exhibited extremely significant nega- tive correlation with altitude and plant height showed extremely significant negative correlation with latitude. Coefficients of genetic divergence indicated that a high proportion of total genetic variation was retained within the populations from different regions, and the high gene flow showed that there were active genetic exchanges among these populations, suggesting no significant genetic divergence among these populations. According to genetic distance and UPGMA, there was a certain correlation between genetic distance and different sampling regions of E. arundinaceum. [Conclusion] The results provide theoretical references for resource collection , het- erosis application and development of excellent genes of sugarcane germplasm.

文摘[Objective] This paper aimed at revealing the rules of phenotypic variation of Leymus chinensis in Zhalong wetlands and providing the scientific and theoretical bases for devising protection strategies of Leymus chinensis populations in Zhalong National Nature Reserve.[Method] Four phenotypic traits of five Leymus chinensis populations in Zhalong Wetlands were comparison and analysis using principal component analysis method and UPGMA cluster analysis method.[Result] There were significant differences among the four phenotypic traits of five populations,the order of coefficients of variation were node number〉plant height〉leaf width〉leaf length;the order of variation degrees of the populations from large to small was KQH,MD,JZ,ZK,TTG population.Results of principal component analysis suggested that the four traits were all factors led to the phenotypic differences among the populations of Leymus chinensis.Leymus chinensis of five populations were divided into three groups by cluster analysis,MD,JZ and TTG were classified as one group,KQH and ZK population were individually classified as one group.[Conclusion] Leymus chinensis had large-scale phenotypic variations and showed strong adaptability to different habitats,which was very important for selection,breeding of fine varieties and germplasm conservation.

文摘To analyze the degree and pattern of phenotypic variation in leaves of Tetracentron sinense Oliv from the perspective of genetic and environmental adaptation and thus contribute to effective evidence-based conservation and management strategies for germplasm resources,we measured 17 morphological and epidermal micromorphological leaf traits from 24 natural populations of T.sinense.Nested analysis of variance,multiple comparison,principal component analysis(PCA),cluster analysis,and correlation analysis were used to explore phenotypic leaf variation among and within populations and potential correlations with geographic and environmental factors.There were significant differences in 17 leaf phenotypic traits among and within populations.The mean phenotypic differentiation coefficient of the 17 traits was 56.34%,and the variation among populations(36.4%)was greater than that within populations(27.2%).The coefficient of variation(CV)of each trait ranged from 4.6 to 23.8%,and the mean was 11.8%.Phenotypic variation of leaves was related to environmental factors such as average annual sunshine hours,average July temperature,and average annual rainfall.The variation changed along gradients of longitude,latitude,and altitude.The PCA clustered the 24 natural populations into four groups.Our study suggests that phenotypic variation in T.sinense occurred primarily among populations,with moderate levels of phenotypic differentiation among populations and low levels of phenotypic variation within populations.The plant’s poor adaptability to the environment is likely an important contributor to its endangerment.Accordingly,conservation strategies are proposed to protect and manage the natural populations of T.sinense.

文摘Phoebe bournei(Hemsl.) Yang is a rare and protected plant in China. This study was conducted to determine the phenotypic variation in this species and to document phenotypic variation within and among populations of P. bournei. Nested analysis of variance, coefficient of variation, multiple comparisons, principal component analysis(PCA) and correlation analysis were used to analyze ten phenotypic traits in ten natural populations of P.bournei from both the northeastern and the primary region of the range of this species. Significant differences among and within populations were observed in leaf and seed phenotypic traits. Variation among populations(34.92%)was greater than that within populations(26.19%). The mean phenotypic differentiation coefficient was 53.77% among populations, indicating that variation among populations comprised the majority of the phenotypic variation of P. bournei. The coefficient of variance(CV) of ten traits varied from 6.44 to 18.45%, with an average of 12.03%.The CV of leaf traits among populations(15.64%) was higher than that of seed traits(8.60%), indicating that seed traits were more stable. The results from CV and PCA indicated that leaf area, leaf length and thousand seed weight were the main factors accounting for the observed phenotypic variations. Significant or highly significant correlations were observed among most leaf and/or in seed phenotypic traits, whereas no significant correlations were observed between phenotypic traits and geographic factors.Based on cluster analysis, the ten populations can be divided into three clusters. These clusters were not a result of geographic distances.

文摘AIM: To compare the phenotypic and neural differentiation potential of human bone marrow derived multipotent adult progenitor cells (MAPC) and mesenchymal stem cells (MSC). METHODS: Cultures of MAPC and MSC were established in parallel from same samples of human bone marrow (n = 5). Both stem cell types were evaluated for expression of pluripotency markers including Oct-4 and Nanog by immunocytochemistry and reversetranscription polymerase chain reaction (RT-PCR) and expression of standard mesenchymal markers including CD14, CD34, CD44, CD45, CD73, CD90, CD105 andhuman leukocyte antigen (HLA)-ABC by flow cytometry. After treatment with neural induction medium both MAPC and MSC were evaluated for expression of neural proteins [neuronal filament-200 (NF-200) and glial fibrillar acidic protein (GFAP)] by immunocytochemistry and Western blotting and neural genes [NF-200, GFAP, Tau, microtubule-associated protein (MAP)-1B, MAP-2, neuron-specific enolase (NSE) and oligodendrocyte-1 (Olig-1)] by quantitative real-time-PCR. RESULTS: MAPC had small trigonal shaped while MSC had elongated spindle-shaped morphology. The MAPC expressed Oct-4 and Nanog both at gene and protein levels, whereas MSC were negative for these pluripotent markers. MAPC were negative for HLA-ABC while MSC had high expression of HLA-ABC. In addition, MAPC as compared to MSC had significantly lower expression of CD44 (36.56% ± 1.92% vs 98.23% ± 0.51%), CD73 (15.11% ± 2.24% vs 98.53% ± 2.22%) and CD105 (13.81% ± 3.82%vs 95.12% ± 5.65%) (P < 0.001, for all) MAPC cultures compared to MSC cultures treated with neural induction medium had significantly higher fold change expression of NF-200 (0.64), GFAP (0.52), Tau (0.59), MAP-2 (0.72), Olig-1 (0.18) and NSE (0.29) proteins (P < 0.01 for Olig-1 and P < 0.001 for rest) as well as higher fold change expression of genes of NF-200 (1.34),GFAP (1.12),Tau (1.08),MAP-1B (0.92), MAP-2 (1.14) andNSE (0.4) (P < 0.001 for all). CONCLUSION: MAPC can be differentially characterized from MSC as Oct-4 and Nanog positive stem cells with no expression of HLA-ABC and low expression of mesenchymal markers CD44, CD73 and CD105 and when compared to MSC they possess greater predilection for differentiation into neuro-ectodermal lineage.

文摘Germplasm resources are an important basis for genetic breeding and analysis of complex traits,and research on genetic diversity is conducive to the exploration and creation of new types of germplasm.In this study,the distribution frequency,coefficient of variation,Shannon-Wiener index,and variance and cluster analyses were used to analyze the diversity and trait differences of 39 fruit phenotypic traits from 570 pear accessions,which included 456 pear accessions from 11 species and 114 interspecific hybrid cultivars that had been stored in the National Germplasm Repository of Apple and Pear(Xingcheng,China).The comprehensive evaluation indices were screened by correlation,principal component and regression analyses.A total of 132 variant types were detected in 28 categorical traits of pear germplasm fruit,which indicate a rich diversity.The diversity indices in decreasing order were:fruit shape(1.949),attitude of calyx(1.908),flesh texture type(1.700),persistency of calyx(1.681),russet location(1.658),relief of area around eye basin(1.644),flavor(1.610)and ground color(1.592).The coefficient of variation of titratable acidity in the 11 numerical traits of pear germplasm fruit was as high as 128.43%,which could more effectively reflect the differences between pear accessions.The phenotypic differentiation coefficient V_(st)(66.4%)among the five cultivated pear species,including Pyrus bretschneideri(White Pear),P.pyrifolia(Sand Pear),P.ussuriensis(Ussurian Pear),P.sinkiangensis(Xinjiang Pear),and P.communis(European Pear),was higher than the within population phenotypic differentiation coefficient V_(st)(33.6%).The variation among populations was the main source of variation in pear fruit traits.A hierarchical cluster analysis divided the 389 accessions of six cultivated pear species,including P.pashia(Himalayan Pear),into six categories.There were certain characteristics within the populations,and the differences between populations were not completely clustered by region.For example,Sand Pear cultivars from Japan and the Korean Peninsula clustered together with those from China.Most of the White Pear cultivars clustered with the Sand Pear,and a few clustered with the Ussurian Pear cultivars.The Ussurian Pear and European Pear cultivars clustered separately.The Xinjiang Pear and Himalayan Pear did not cluster together,and neither did the cultivars.Seventeen traits,three describing fruit weight and edible rate(fruit diameter,fruit length and fruit core size),five describing outer quality and morphological characteristics(over color,amount of russeting,dot obviousness,fruit shape,and stalk length),and nine describing inner quality(flesh color,juiciness of flesh,aroma,flavor,flesh texture,flesh texture type,soluble solid contents,titratable acidity,and eating quality)were selected from the 39 traits by principal component and stepwise regression analyses.These 17 traits could reflect 99.3%of the total variation and can be used as a comprehensive evaluation index for pear germplasm resources.

文摘Mauremys mutica(Cantor,1842)is an endangered species in China.Main phenotypic variations inbody color,body weight,body shape,clutch size,egg size,and hatchling size were revealed betweenthe southern and northern populations.Both populations have the phenomenon of'larger male'sexualsize dimorphism(SSD),especially in the southern population.Furthermore,genetic variations betweenthe two populations were analyzed by RAPD band patterns of 30 random individuals in each population.The average genetic distance was 0.299±0.108 among the samples of two populations.The average ge-netic distance between southern and northern populations was 0.305±0.046.Cluster analysis indicatedthat all the individuals from the southern and northern populations were clustered among themselves toform two distinct clades.A total of 20 population-specific RAPD fragments were scored from 16 primers,and could be used as RAPD markers for distinguishing the southern and the northern population.Basedon the nucleotide sequences of two RAPD markers,two pairs of SCAR primers(SC1-S and SC2-S)weredesigned,which could be used as SCAR markers for the southern population.According to the significantphenotypic and genetic variations,we suggested that the northern population and southern populationmight be considered as two separate taxa,the'northern taxon'and the'southern taxon',and the con-servation should be respectively conducted on the two taxa.

文摘Eremosparton songoricum (Litv.) Vass is a dwarf clonal shrubby legume developed on the windward slopes of mobile or semi-fixed sand dunes of Central Asia. It is assumed that E. songoricum must possess a high degree of phenotypic plasticity for such a heterogeneous habitat. The variations of E. songoricum plants growing in two typical microhabitats (the upper slope and the lower slope of semi-mobile dune) were investigated. The morphological characteristics and the biomass allocation patterns were measured and compared at the clonal fragment level. Compared with the clonal fragment on the lower slope of dune, the clonal fragment on the upper slope of the dune (a) declined in total biomass and ramet height, (b) increased the length of rhizomes and the number of roots, (c) increased the degree of asym- metry, and (d) heightened allocation to the belowground biomass. Our results confirmed the hypothesis of high phenotypic adjustment capacity of E. songoricum to habitat moisture availability. Phenotypic plasticity of E. songoricum contributed to reduce the damage risk, led to an environmentally induced specialization in function of resources exploitation, resulted in its persistence in heterogeneous environments and was adaptive in sand dune environment.

文摘Diversity of 60 conventional japonica rice accessions with good eating quality at home and abroad was analyzed using SSR molecular markers, agronomic traits and taste characteristics. A total of 290 alleles were detected in the 60 accessions at 72 SSR loci with the high similarity coefficients varying between 0.600 and 0.924. The loci on chromosome 5 showed the greatest value in average allele number. Additionally, most of the SSR loci could detect 3 to 4 alleles. An UPGMA dendrogram based on the cluster analysis of the genetic similarity coefficients showed that the grouping trend of part of the rice accessions was geographic-related and most of the rice accessions in Jiangsu Province, China were clustered together. Furthermore, many domestic accessions from south and north origins in China were close to the foreign japonica rice varieties, as proved by their pedigree origin from the foreign high-quality sources. For taste characteristics, part of the accessions with excellent taste were clearly clustered into one category though they came from different geographical regions, which indicates that taste characteristics of some varieties were mainly genetically determined. In addition, the agronomic traits of japonica rice with good taste might be closely related with their geographical origins, but the relationship between superior taste characteristics and agronomic traits should be further clarified.

文摘The phenotypic diversity of 274 Ethiopian durum wheat accessions was analyzed, taking their geographic origins into account. The aim was to assess the extent and patterns of agronomically important phenotypic variation across districts of origin and altitude classes for major qualitative traits using diversity index and multivariate methods. Eight qualitative and three quantitative traits were scored for 2740 plants and analyzed for diversity. The Shannon–Weaver diversity(H′) index was used to estimate phenotypic diversity. The estimated H′ ranged from monomorphic for glume hairiness to highly polymorphic for other traits. The highest(0.86) H′ was obtained for seed degree of shriveling, possibly indicating the differential responses of the genotypes to water deficit during later growth stages. With respect to district of origin, the highest(0.72) and lowest(0.44) H′ values were obtained for the Bale and SNNP districts, respectively. With respect to altitude, the highest(0.76) and lowest(0.62) H′ values were recorded for altitudes 1600–2000and > 3000 m above sea levels, respectively. Principal components analysis explained substantial variation contributed by district of origin and altitude range. Genotypes were clustered into three groups by districts of origin and altitude class, with relatively strong bootstrap values of 57 and 62 for the former and latter, respectively. It could be concluded that Ethiopian durum wheat landraces are very diverse both within and among districts of origin and altitude classes. This wealth of genetic diversity should be exploited for wheat improvement of yield and for resistance to biotic and abiotic stresses, particularly terminal drought.
文摘BACKGROUND: The antigen reducing ability of dentritic cells (DCs), a kind of antigen presenting cells (APCs) initiating immune response, is associated with the specific immune tolerance of chronic hepatitis B(CHB) patients. However, the dysfunction of DCs can be possibly reversed by the stimulation of antigen peptides. In this study, DCs were cultured from peripheral blood monocytes (PBMCs) in patients with CHB in vitro, and the expression of phenotypic molecules on DCs loaded by different concentrations of HBsAg was observed. METHODS: Forty patients with CHB were divided randomly into 4 groups(10 patients in each group). PBMCs were isolated, and DCs were cultured after addition of granulocyte/macrophage colony-stimulating factor (GMCSF) and interleukin 4(IL-4). On the 9th day, DCs of the experimental groups were loaded at HBsAg concentrations of 2.5mg/L, 5mg/L and 10mg/L for 24 hours, whereas those of the control group were not loaded. An electron microscope was used to analyze the morphological changes of the DCs. The expression of phenotypic molecules on DCs in different groups was detected with flow cytometry. RESULTS: A combination of GM-CSF and IL-4 produced DCs from PBMCs in patients with CHB after being cultured for 9 days, whose morphological changes were tested by an electron microscope. The expression of phenotypic molecules on DCs in the control group was as low as CD83 (8.02±3.99)%, CD80(8.77±2.06)%, and MHC-DR (14.05±2.66)%. Loaded by different concentrations of HBsAg, the up-regulation of phenotypic molecules on DCs was found, with CD83(18.35±2.93)%, CD80(42.63±7.15)% and MHC-DR(47.49±6.59)% in 2.5mg/L HBsAg loading group, CD83(17.88±3.12)%, CD80(45.24± 10.93)% and MHC-DR(47.07±8.52)% in 5mg/L HBsAg loading group and CD83(16.74±2.86)%, CD80(44.59±6.99)% and MHC-DR(48.59±7.42)% in 10mg/L HBsAg loading group, respectively. Compared with the control group, the phenotypic molecules in the experimental groups were all different significantly (P<0.01), but among them, there were no differences (P>0.05). CONCLUSIONS: DCs cultured from PBMCs in the patients with CHB under the conditions of GM-CSF and IL-4 present on the typical dendritic morphology but are immature for expressing low phenotypic molecules. Loaded by different concentrations of HBsAg, the immature DCs can differentiate to mature DCs for expressing increasing phenotypic molecules.

文摘Prior knowledge of the correlations among commercially important traits in Glyptocidaris crenularis can be used to guide development of breeding and aquaculture programs for this species.We evaluated the phenotypic correlations between somatic and gonadal traits in G.crenularis during two sample periods(October 2010 and April 2011).The coeffi cients of variation(CV%)for body weight(BW),redness(a*),yellowness(b*),gonad wet weight(GW),and gonad index(GI)were>20% in both periods,while those of other traits were very low.GW was signifi cantly correlated with somatic traits(diameter,height,and body weight)in both periods,although the correlation coeffi cients ranged from 0.473 to 0.636(P<0.01).Gonad moisture content(GC),a*,and lightness(L*)were not signifi cantly correlated with somatic traits(P>0.05).The color trait b* was negatively correlated with somatic traits(test diameter,test height,and body weight)during the period when the gonads were well developed(April 2011,P<0.05).We observed little difference in the nature of the phenotypic correlations between the two sample periods.The high variation in the somatic and gonad weight traits in G.crenularis suggests there is considerable potential for selective breeding.However,our results suggest that gonad quality traits cannot be selected indirectly by selecting for body weight.Our study provides an insight into the method for improving the breeding and aquaculture of G.crenulari.
文摘In order to reveal the genetic diversity and variation sources of leaf phenotypic traits in walnut,the coefficient of variation,Shannon-weaver index,phenotypic differentiation coefficient,and the correlation of 9 leaf phenotypic traits for 251 walnut germplasm resources from two walnut populations were analyzed.The results showed that the averages of the variation coefficients and Shannon-weaver indexes of the leaf phenotype traits were 17.45%and 1.86 respectively.The F values of most inter-population traits and all intra-population traits were highly significant,and the F values of inter-population leaf shape index,leaflet number,length and leaf length were very large.The averages of inter-population and intra-population differentiation coefficients were 9.15%and 90.85%respectively.The leaf size,leaf thickness,and leaflet number were highly correlated with trunk perimeters.According to the results of principal component analyses,the 9 traits can be simplified into four principal components,which represent leaf size,petiole,leaflet number and leaf shape,respectively.There were extensive variations and rich genetic diversities in leaf phenotypic traits of walnut,and the intra-population variation was the leading source of phenotypic trait variation.Leaf shape index,leaflet number,internode length and leaf length can be used for population classification.The size and thickness of leaves and the number of leaflets will affect the accumulation of nutrients in walnut tree,and then affect its trunk perimeter.

文摘Background The genomes of worldwide poultry breeds divergently selected for performance and other phenotypic traits may also be affected by,and formed due to,past and current admixture events.Adaptation to diverse environments,including acclimation to harsh climatic conditions,has also left selection footprints in breed genomes.Results Using the Chicken 50K_CobbCons SNP chip,we genotyped four divergently selected breeds:two aboriginal,cold tolerant Ushanka and Orloff Mille Fleur,one egg-type Russian White subjected to artificial selection for cold tolerance,and one meat-type White Cornish.Signals of selective sweeps were determined in the studied breeds using three methods:(1)assessment of runs of homozygosity islands,(2)F_(ST) based population differential analysis,and(3)haplotype differentiation analysis.Genomic regions of true selection signatures were identified by two or more methods or in two or more breeds.In these regions,we detected 540 prioritized candidate genes supplemented them with those that occurred in one breed using one statistic and were suggested in other studies.Amongst them,SOX5,ME3,ZNF536,WWP1,RIPK2,OSGIN2,DECR1,TPO,PPARGC1A,BDNF,MSTN,and beta-keratin genes can be especially mentioned as candidates for cold adaptation.Epigenetic factors may be involved in regulating some of these important genes(e.g.,TPO and BDNF).Conclusion Based on a genome-wide scan,our findings can help dissect the genetic architecture underlying various phenotypic traits in chicken breeds.These include genes representing the sine qua non for adaptation to harsh environments.Cold tolerance in acclimated chicken breeds may be developed following one of few specific gene expression mechanisms or more than one overlapping response known in cold-exposed individuals,and this warrants further investigation.
文摘Symbiotic and phenotypic characteristics of thirty rhiobial isolates obtained from root nodules of two cowpea (Vigna unguiculata L. Walp) cultivars that grown in different sites of Fezzan (Southern part of Libya) were studied. Cultural characteristics and cross-nodulation with Arachis hypogega and Faidherbia albida showed that they were slow-growing rhizobia. Each isolate was found to coexist with non-symbiotic bacteria similar in their cultural characteristics to fast-growing rhizobia. All isolates formed symbiosis with the test plants, but different in their nitrogen-fixation efficiency. Numerical analysis of phenotypic characteristics showed that at boundary level of 70% average similarity, the isolates formed four distinguished groups and two isolates remained separate. Most isolates exhibited wide tolerance to acidity, alkalinity and extreme temperatures. They also resistant to some heavy metals such as mercury, copper, zinc, lead, cadmium and aluminum at low concentrations and antibiotics like polymyxin, colistin, bacitracin and nalidixic acid. Isolates displayed different response to salinity ranging from sensitive, which unable to grow in 1% NaCI to resistant and grow at 2% NaCl or above. Urea was hydrolyzed by most of them and carbohydrates utilizations were different. Sucrose and maltose were metabolized by most of the test isolates, whereas, monosaccharide and sugar alcohols were poorly utilized.
