BACKGROUND Pigmented villonodular synovitis(PVNS)is a benign proliferative disorder that affects the synovial joints,bursae,and tendon sheaths.To date,few studies have reported on the treatment of postoperative pain a...BACKGROUND Pigmented villonodular synovitis(PVNS)is a benign proliferative disorder that affects the synovial joints,bursae,and tendon sheaths.To date,few studies have reported on the treatment of postoperative pain and edema in patients with PVNS.Herein,we present the case of a woman who developed pain and edema in the left lower limb 1 wk after synovectomy and arthroscopic partial meniscectomy and was unable to walk due to limited flexion and extension of the left knee.CASE SUMMARY A 32-year-old woman underwent synovectomy and arthroscopic partial meniscectomy successively and was treated with a combination of manual lymphatic drainage(MLD)and kinesio taping(KT)in our hospital to alleviate postoperative pain and edema.The following parameters were assessed at 2 wk post-treatment and 1 wk post-discharge follow up:suprapatellar circumference,infrapatellar circumference,visual analog scale score,knee range of motion,pittsburgh sleep quality index score,hamilton anxiety rating scale(HAMA)score,and hamilton depression rating scale(HAMD)score.After treatment,the postoperative pain and edema in the patient’s left knee were effectively relieved,resulting in improved sleep quality and remarkably attenuated HAMA and HAMD scores.CONCLUSION Combined MLD and KT may be an effective approach for relieving postoperative pain and edema in patients with PVNS.展开更多

In this work, the orientation of the pigments in the isolated photosystem Ⅱ (PSⅡ) sub_core reaction center complexes was analyzed and characterized by means of low temperature absorption and LD spectroscopy. The preparations containing different amounts of CP47 isolated from spinach (Spinacia oleracea L.) chloroplast were used in order to investigate the orientation of pigments in the PSⅡ sub_core CP47/D1/D2/Cyt b_559 (CP47/D1/D2) complexes. Chlorophyll a (Chl a) absorbing at 680 nm in CP47/D1/D2/Cyt b_559 complex showed an orientation of the Q y transition parallel to the membrane plane. It is proposed that there are two forms of β_carotene (β_Car) in CP47/D1/D2/Cyt b_559 complex, denoted as β_Car (Ⅰ) and β_Car (Ⅱ), with different orientations, β_Car (Ⅰ) at 470 and 505 nm is roughly parallel to the membrane plane, and β_Car (Ⅱ) at 460 and 490 nm seems to be perpendicular orientation. Upon the photoinhibitory experiment β_Car (Ⅱ) was found to be photosensitive and easily photodamaged. It also showed that the positive LD signal observed at 680 nm was quite complicated. This signal is tentatively attributed to P680 and some Chl a of antenna in CP47 protein based upon our measurements.展开更多

Methods Primary porcine RPE cells were harvested by enzyme digestion and expanded in culture medium. Determine the levels ofdopamine (DA) and homovanillic acid (HVA) by high performance liquid chromatography electrochemical (HPLC) assay, and the levels of brain-derived neurotrophic factor (BDNF) and glial-derived neurotrophic factor (GDNF) were detected by ELISA. Alginate-polylysine-alginate (APA) microencapsulated cells were produced by using a high voltage electrostatic system. PD rat model was established by unilateral injection of 6-hydroxydopamine (6-OHDA) into the medial forebrain bundle (MFB). After that, the RPE-M was transplanted into the corpus striatum of PD rat, and then the rotation test scores were recorded and biochemical changes of the corpus striatum were tested. Results The levels of DA, HVA, BDNF and GDNF secreted by RPE were stable in the RPE culture supernatant and were not changed by the microencapsulation. Eighty-three percent rats developed PD by unilateral lesion of 6-OHDA in the MFB. The RPE-M transplantation had therapeutic effect on 33% PD rats. Conclusion Porcine RPE cells grow actively in vitro and could secrete DA, HVA, BDNF, and GDNF constantly, which does not be affected by the passage culture and the APA miroencapsulation. RPE-M transplantation of may be a curative therapy for PD.展开更多
The effects of polishing on proximate compositions,physico-chemical characteristics,mineral compositions and antioxidant properties of the rice flours obtained from three different pigmented rice varieties(Chak-hao An...The effects of polishing on proximate compositions,physico-chemical characteristics,mineral compositions and antioxidant properties of the rice flours obtained from three different pigmented rice varieties(Chak-hao Angangba,Chak-hao Amubi and Chak-hao Poireiton) were investigated.The rice varieties were significantly(P < 0.05) different in the contents of the test characteristics.Lipids,ash,minerals,phytochemicals(phenolic acids and flavonoids) and 2,2-diphenyl-1-picrylhydrazyl(DPPH) activity of rice flours were decreased after polishing(9% degree of milling),while amylose content and lightness were increased.X-ray diffraction pattern of rice flours exhibited A-type crystalline pattern with reflections at 15.1o,17.1o,18.2o and 23.0o.Pasting properties and transition temperatures were decreased after polishing treatment.Polishing resulted in changes in the crystallinity,enthalpy and morphology of rice flours.展开更多

METHODS: Sixteen pigmented rabbits were monocularly deprived (MD) by suturing the ...AIM: To determine the effect of 7-methylxanthine (7-MX) on the posterior sclera of form-deprivation myopia (FDM) in pigmented rabbits. METHODS: Sixteen pigmented rabbits were monocularly deprived (MD) by suturing the right eyelids after natural eye opening (ten-day old) for a period of 30 days. Two groups of pigmented rabbits were fed either 7-MX (30 mg per kg body weight; n=8) or vehicle control (saline equal volume with 7-MX; n=8). Ocular refractions, axial lengths and body weights were measured at the start and the end of the experiment 30 days later. Electron microscopy was used to measure and determine the collagen fibril diameters in the posterior pole of sclera. RESULTS: In vehicle control MD pigmented rabbits, 30 days of MD produced -1.10D+/- 0.78D of myopia and the axial length increased 0.51mm+/- 0.09mm. In MD pigmented rabbits fed with 7-MX, 30 days of MD induced only -0.21D+/- 0.11D of myopia and the axial length increased 0.07mm+/- 0.10mm. There was significant change in axial length of vehicle control MD pigmented rabbits (13.11mm +/- 0.19mm versus 12.60mm+/- 0.06mm; P =0.03). The changes in refraction and axial length of two MD groups' contalateral eyes during the 30 days were not significantly different (2.75D+/- 0.27D versus 2.75D 0.35D, P>0.05; 12.60mm+/- 0.06mm versus 12.45mm +/- 0.14mm, P >0.05). The weights of the two groups pigmented rabbits had no significant changes (187g+/- 22.1g versus 189g+/- 19.3g, P>0.05). The diameter cif scleral collagen fibers increased in both eyes of 7-MX treated pigmented rabbits. There was significant difference in collagen fibril diameters of inner layer (111.34nm+/- 28.30nm versus 94.80nm +/- 27.52nm, P =0.002) and outer layer (167.92nm +/- 55.82nm versus 144.04nm +/- 47.02nm P =0.016) in the posterior sclera between the myopic eyes of vehicle control MD group and contralateral eyes of 7-MX treated MD group. CONCLUSION: 7-MX appears to prevent FDM in pigmented rabbits by remodeling the posterior sclera.展开更多

The coating morphology was observed using scanning electron microscopy (SEM), and the electrochemical properties were investigated by electrochemical impedance spectroscopy (EIS). Immersion test and neutral salt spray test were also conducted to investigate the corrosion resistance of the coating. It is demonstrated that the critical pigment volume content (CPVC) value is between 30% and 40%. The coating with PVC of 30% exhibits good corrosion resistance in 3.5% (mass fraction) NaCl solution.展开更多

AIM:To evaluate the effects of LIN28A(human)on high glucose-induced retinal pigmented epithelium(RPE)cell injury and its possible mechanism.METHODS:Diabetic retinopathy model was generated following 48h of exposure to...AIM:To evaluate the effects of LIN28A(human)on high glucose-induced retinal pigmented epithelium(RPE)cell injury and its possible mechanism.METHODS:Diabetic retinopathy model was generated following 48h of exposure to 30 mmol/L high glucose(HG)in ARPE-19 cells.Quantitative real-time polymerase chain reaction(qRT-PCR)and Western blot tested the expression of the corresponding genes and proteins.Cell viability as well as apoptosis was determined through cell counting kit-8(CCK-8)and flow cytometry assays.Immunofluorescence assay was adopted to evaluate autophagy activity.Caspase 3 activity,oxidative stress markers,and cytokines were appraised adopting their commercial kits,respectively.Finally,ARPE-19 cells were preincubated with EX527,a Sirtuin 1(SIRT1)inhibitor,prior to HG stimulation to validate the regulatory mechanism.RESULTS:LIN28A was downregulated in HG-challenged ARPE-19 cells.LIN28A overexpression greatly inhibited HGinduced ARPE-19 cell viability loss,apoptosis,oxidative damage as well as inflammatory response.Meanwhile,the repressed autophagy and SIRT1 in ARPE-19 cells challenged with HG were elevated after LIN28A overexpression.In addition,treatment of EX527 greatly inhibited the activated autophagy following LIN28A overexpression and partly abolished the protective role of LIN28A against HG-elicited apoptosis,oxidative damage as well as inflammation in ARPE-19 cells.CONCLUSION:LIN28A exerts a protective role against HG-elicited RPE oxidative damage,inflammation,as well as apoptosis via regulating SIRT1/autophagy.展开更多

This causes variations in phytoplankton community composition in the upwelling zone compared with the surrounding offshore area. Based on pigments analysis with subsequent calculations of group contributions to total chlorophyll a(Chl a) using CHEMTAX, we studied the distribution and composition of phytoplankton assemblages in the subsurface chlorophyll maximum along the south coast of Java and the influence of upwelling. Nineteen phytoplankton pigments were identified using high-performance liquid chromatography, and CHEMTAX analysis associated these to ten major phytoplankton groups. The phytoplankton community in the coastal area influenced by upwelling was characterized by high Chl a and fucoxanthin concentrations, indicating the dominance of diatoms. In contrast, in the offshore area, the Chl a and fucoxanthin concentrations declined to very low levels and the community was dominated by haptophytes represented by 19′-Hexanoyloxyfucoxanthin. Accordingly, microphytoplankton was found to be the major size class in the coastal area influenced by upwelling, while nanophytoplankton was most abundant in the offshore area. Low concentrations of other accessory pigments indicated less contribution from dinoflagellates,prasinophytes, chlorophytes and cryptophytes. Photo-pigment indices revealed that photosynthetic carotenoids(PSCs) were the largest component of the pigment pool, exceeding the proportion of Chl a, with the average PSCTP up to 0.62. These distribution trends can mainly be explained by phytoplankton adaption strategies to upwelling and subsurface conditions by changing species composition and adjusting the pigment pool.展开更多
The study of training hyperparameters optimisation problems remains underexplored in skin lesion research.This is the first report of using hierarchical optimisation to improve computational effort in a four-dimension...The study of training hyperparameters optimisation problems remains underexplored in skin lesion research.This is the first report of using hierarchical optimisation to improve computational effort in a four-dimensional search space for the problem.The authors explore training parameters selection in optimising the learning process of a model to differentiate pigmented lesions characteristics.In the authors'demonstration,pretrained GoogleNet is fine-tuned with a full training set by varying hyperparameters,namely epoch,mini-batch value,initial learning rate,and gradient threshold.The iterative search of the optimal global-local solution is by using the derivative-based method.The authors used non-parametric one-way ANOVA to test whether the classification accuracies differed for the variation in the training parameters.The authors identified the mini-batch size and initial learning rate as parameters that significantly influence the model's learning capability.The authors'results showed that a small fraction of combinations(5%)from constrained global search space,in contrarily to 82%at the local level,can converge with early stopping conditions.The mean(standard deviation,SD)validation accuracies increased from 78.4(4.44)%to 82.9(1.8)%using the authors'system.The fine-tuned model's performance measures evaluated on a testing dataset showed classification accuracy,precision,sensitivity,and specificity of 85.3%,75.6%,64.4%,and 97.2%,respectively.The authors'system achieves an overall better diagnosis performance than four state-of-the-art approaches via an improved search of parameters for a good adaptation of the model to the authors'dataset.The extended experiments also showed its superior performance consistency across different deep networks,where the overall classification accuracy increased by 5%with this technique.This approach reduces the risk of search being trapped in a suboptimal solution,and its use may be expanded to network architecture optimisation for enhanced diagnostic performance.展开更多
Bacterial abundance, phytoplankton community structure and environmental parameters were investigated to study the relationships between bacteria and phytoplankton during giant jellyfish Nemopilema nomurai blooms in t...Bacterial abundance, phytoplankton community structure and environmental parameters were investigated to study the relationships between bacteria and phytoplankton during giant jellyfish Nemopilema nomurai blooms in the central Yellow Sea during 2013. N. nomurai appeared in June, increased in August, reached a peak and began to degrade in September 2013. Results showed that phosphate was possible a key nutrient for both phytoplankton and bacteria in June, but it changed to nitrate in August and September. Phytoplankton composition significantly changed that pico-phytoplankton relative biomass significantly increased, whereas other size phytoplankton significantly decreased during jellyfish bloom. In June, a significantly positive correlation was observed between chlorophyll a concentration and bacterial abundance(r=0.67, P0.001, n=34).During jellyfish outbreak in August, there was no significant correlation between phytoplankton and bacteria(r=0.11, P0.05, n=25), but the relationship(r=0.71, P0.001, n=31) was rebuilt with jellyfish degradation in September. In August, small size phytoplankton occupied the mixed layer in offshore stations, while bacteria almost distributed evenly in vertical. Chlorophyll a concentration significantly increased from(0.42±0.056) μg/L in June to(0.74±0.174) μg/L in August, while bacterial abundance just slightly increased. Additionally, the negative net community production indicated that community respiration was not entirely determined by the local primary productivity in August. These results indicated that jellyfish blooms potentially affect coupling of phytoplankton and bacteria in marine ecosystems.展开更多

METHODS: Human AB serum, knock-out serum replacement(KSR) and B27 were supplemented as a substitute of fetal bovine serum(FBS) in culture medium of human fRPE cells. Cell morphology was examined by light microscope and transmission electron microscope. Proliferation ability was detected by cell cycle analysis and examination of KI67 expression. Apoptosis was analyzed using FACS. The expression ofRPE-specific markers was demonstrated by quantitative real-time polymerase chain reaction(qPCR), Western blot(WB) and immunocytochemistry. Paracrine function was determined using enzyme-linked immunosorbent assay method.RESULTS: Our results indicated that the optimum concentration of KSR was 15%, the optimum concentration of B27 was 2%, and the optimum concentration of human AB serum was 10%. fRPE cells cultured in 15% KSR and 2% B27 media showed repressed propagation and differentiation ability, and gradually lost epithelial morphology and RPE function. While fRPE cells cultured in 10% human AB serum media showed a typical cobblestone morphology with pigmentation, elevated proliferation ability, satisfying paracrine function and expressed RPE-specific markers. CONCLUSION: Our study indicates that culturing fRPE cells in 10% human AB serum-supplemented medium is more favorable compared with KSR, B27 and traditional FBS-supplemented mediums when fRPE cells are to be applied in cell-based therapy.展开更多
BACKGROUND Pigmented villonodular synovitis(PVNS)is an uncommon disease that usually occurs in large joints,and involvement of the subtalar joint is rare.The lack of comprehensive knowledge of subtalar joint PVNS coul...BACKGROUND Pigmented villonodular synovitis(PVNS)is an uncommon disease that usually occurs in large joints,and involvement of the subtalar joint is rare.The lack of comprehensive knowledge of subtalar joint PVNS could lead to misdiagnosis.CASE SUMMARY We present a 64-year-old woman who,at her first visit,complained of discomfort in the right ankle when she walked.Based on the physical signs and X-ray report,the physician failed to make the suspected diagnosis of PVNS.Eighteen months later,the patient returned with a complaint of a mass in her right lateral malleolus with intermittent blunt pain.The X-ray presented an osteophyte formation and soft tissue calcification at the margin of the subtalar joint.The laboratory tests were normal,whereas magnetic resonance imaging(MRI)showed a low-intensity area on both T1-and T2-weighted images.A suspected diagnosis of PVNS was made and later confirmed by postoperative pathology.Subsequently,the patient received radiotherapy with 32 Gy in 16 fractions.At 6 mo postoperatively,the patient only complained of discomfort after walking three blocks.The American Orthopedic Foot and Ankle Society Ankle-Hindfoot score was 97.CONCLUSION MRI is necessary for the diagnosis of PVNS.Early soft tissue calcification and painless joint swelling should be of concern.展开更多

AIM: To demonstrate the feasibility of mesenchymal stem cell(MSC)-mediated nano drug delivery, which was characterized by the “Trojan horse”-like transport of hypoxiainducible factor-1α small interfering RNA(HIF-1...AIM: To demonstrate the feasibility of mesenchymal stem cell(MSC)-mediated nano drug delivery, which was characterized by the “Trojan horse”-like transport of hypoxiainducible factor-1α small interfering RNA(HIF-1α si RNA) between MSCs and retinal pigment epithelial cells(RPE) under hypoxia environment.METHODS: Plasmid and lentivirus targeting the human HIF-1α gene were designed and constructed. HIF-1α si RNA was encapsulated into poly(lactic-co-glycolic acid) nanoparticles(PLGA-NPs) through the water-in-oil-in-water(w/o/w) multiple emulsion technique. The effect of PLGANPs uptake on the expression of HIF-1α m RNA was tested in RPE cells by real-time quantitative polymerase chain reaction(q PCR) and additional transfected conditions were used as control, including lentivirus group, nude plasmid group and blank PLGA group. MSCs were transfected with the NPs and the transfection efficacy was evaluated by flow cytometry. Transwell co-culture system of transfected MSCs and RPE cells was constructed under hypoxia environment. The effects of MSC-loaded HIF-1α si RNA PLGA-NPs on proliferation, apoptosis, and migration of RPE cells were then evaluated. The effect of transfected MSCs on HIF-1α expression of RPE cells was analyzed by using q PCR at the time points 24h, 3d, and 7d.RESULTS: The average diameter of PLGA-NPs loaded with HIF si RNA was 314.1 nm and the zeta potential was-0.36 m V. The transfection efficiency of PLGA-NPs was 67.3%±5.2% into MSCs by using flow cytometry. Compared with the lentivirus group, the PLGA-NPs loaded with HIF-1α si RNA can effectively reduce the expression of HIF-1α m RNA up to 7d in RPE(0.63±0.05 at 7d, P<0.001). In the Transwell co-culture system of transfected MSCs and RPE, the abilities of proliferation(2.34±0.17, 2.40±0.28, 2.47±0.24 at 48h, F=0.23, P=0.80), apoptosis(14.83%±2.43%, 12.94%±2.19%, 12.39%±3.21%;F=0.70, P=0.53) and migration(124.5±7.78, 119.5±5.32, 130±9.89, F=1.33, P=0.33) of the RPE cells had no differences between MSCloaded HIF-1α si RNA PLGA-NPs and other groups. The inhibition of PLGA on the HIF-1α m RNA expression in RPE cells could continue until the 7th day, the level of HIF-1α m RNA was lower than that of other groups(F=171.98, P<0.001). CONCLUSION: The delivery of PLGA-NPs loaded with HIF-1α si RNA carried by MSCs is found to be beneficial temporally for HIF-1α m RNA inhibition in RPE cells under hypoxia environment. The MSC-based bio-mimetic delivery of HIF-1α si RNA nanoparticles is a potential method for therapy against choroidal neovascularization.展开更多

The effect of curing time on K/S values was also investigated. It has been found that butadiene based binder shows good performance in terms of crocking fastness, formaldehyde release and PVC migra-tion. It has also been observed that by increasing the binder concentration, the release of formaldehyde decreased and by increasing the curing time, the K/S values of printed fabric were decreased.展开更多

Malfunctioning of RPE contributes to the onset and advance of retinal degenerative diseases. Up to date, RPE replacement therapy is the only possible method to completely reverse retinal degeneration. Transplantation of human RPE stem cell-derived RPE(h RPESC-RPE) has shown some good results in animal models. With promising results in terms of safety and visual improvement, human embryonic stem cell-derived RPE(h ESC-RPE) can be expected in clinical settings in the near future. Despite twists and turns, induced pluripotent stem cell-derived RPE(i PSC-RPE) is now being intensely investigated to overcome genetic and epigenetic instability. By far, only one patient has received i PSCRPE transplant, which is a hallmark of i PSC technology development. During follow-up, no major complications such as immunogenicity or tumorigenesis have been observed. Future trials should keep focusing on the safety of stem cell-derived RPE(SC-RPE) especially in long period, and better understanding of the nature of stem cell and the molecular events in the process to generate SC-RPE is necessary to the prosperity of SC-RPE clinical application.展开更多

The...<strong>Introduction:</strong> Intense Pulsed Light (IPL) technology is well accepted in the medical aesthetic field for the treatment of various skin lesions, including pigmented and vascular lesions. The light penetrates into the skin and is selectively absorbed by lesion chromophore. Absorbed energy is converted into heat, coagulating the lesion, which naturally fades following the treatment. The current article presents a retrospective efficacy evaluation of an IPL device with high peak power. <strong>Methods:</strong> Representative treatment results were collected from several clinics based on photographs taken at baseline and after treatments. Photos were evaluated and analyzed for aesthetic improvement of the various skin conditions in different facial and body areas. <strong>Results:</strong> Analysis included cases of pigmented and vascular lesions, textural lesions, and more specific conditions such as melasma and rosacea. The two evaluators’ scoring demonstrated improvement in all cases according to the Global Aesthetic Improvement Scale (GAIS) scale. <strong>Conclusion:</strong> The vast experience gathered from the market in treating various skin lesions supports the safety and efficacy of the investigated IPL device. The device’s particular specifications contribute to the successful results and ease of treatment for the practitioner and the patient.展开更多
Objective:To study the inhibitory effect of rice bran extracts of Thai black Kam Muang and red Hawm Dawk Mali Deang on oxidative stress factors including superoxide(O2·-),nitric oxide(NO·),and inducible nitr...Objective:To study the inhibitory effect of rice bran extracts of Thai black Kam Muang and red Hawm Dawk Mali Deang on oxidative stress factors including superoxide(O2·-),nitric oxide(NO·),and inducible nitric oxide synthase(iNOS).Methods:Bran extracts(40%ethanol)of Kam Muang and Hawm Dawk Mali Deang were obtained and evaluated for in vitro 2-2′-azino-di-(3-ethylbenzthiazoline sulfonate)(ABTS)and NO·scavenging activity.Their inhibitory effects on cellular O2·-and NO·were measured in phorbol 12-myristate 13-acetate-stimulated neutrophil-like HL-60 cells and lipopolysaccharidestimulated RAW264.7 macrophages,respectively,and their viability was monitored using the MTT assay.The effect on iNOS expression was also assessed by the Western blotting assay.Total contents of phenolics,flavonoids,and subtypes were also determined.Results:Hawm Dawk Mali Deang exhibited about 3.5-fold greater cellular O2·-inhibitory activity than Kam Muang[EC50 values of(23.57±4.54)and(81.98±1.45)μg/mL,respectively]in phorbol 12-myristate 13-acetate-stimulated HL-60 cells.Hawm Dawk Mali Deang exhibited about 2-fold higher in vitro ABTS·+and NO·scavenging activity than Kam Muang,but it exerted cellular NO·inhibitory activity of only about 26%(undetermined EC50 value)in lipopolysaccharide-stimulated RAW264.7 cells.Conversely,Kam Muang exerted potent cellular NO·inhibitory activity[EC50 value:(281.13±59.18)μg/mL]and dose-dependently decreased iNOS levels.No cytotoxicity of both extracts was detected in both cell types.As for corresponding contents,Hawm Dawk Mali Deang contained higher contents of phenolics and flavonoids than Kam Muang.Moreover,Kam Muang and Hawm Dawk Mali Deang had a high content of total anthocyanins[(14.73±0.52)mg C3GE/g of extract]and total proanthocyanidins[(115.13±1.47)mg CE/g of extract],respectively.Conclusions:Based on these data,bran extracts of Thai black Kam Muang and red rice Hawm Dawk Mali Deang can help lower oxidative stress and inflammation attributed partly to O2·-and NO·.展开更多

文摘BACKGROUND Pigmented villonodular synovitis(PVNS)is a benign proliferative disorder that affects the synovial joints,bursae,and tendon sheaths.To date,few studies have reported on the treatment of postoperative pain and edema in patients with PVNS.Herein,we present the case of a woman who developed pain and edema in the left lower limb 1 wk after synovectomy and arthroscopic partial meniscectomy and was unable to walk due to limited flexion and extension of the left knee.CASE SUMMARY A 32-year-old woman underwent synovectomy and arthroscopic partial meniscectomy successively and was treated with a combination of manual lymphatic drainage(MLD)and kinesio taping(KT)in our hospital to alleviate postoperative pain and edema.The following parameters were assessed at 2 wk post-treatment and 1 wk post-discharge follow up:suprapatellar circumference,infrapatellar circumference,visual analog scale score,knee range of motion,pittsburgh sleep quality index score,hamilton anxiety rating scale(HAMA)score,and hamilton depression rating scale(HAMD)score.After treatment,the postoperative pain and edema in the patient’s left knee were effectively relieved,resulting in improved sleep quality and remarkably attenuated HAMA and HAMD scores.CONCLUSION Combined MLD and KT may be an effective approach for relieving postoperative pain and edema in patients with PVNS.

文摘Linear dichroism (LD) spectroscopy is an important technique in the study of the orientation and organization of pigments in the photosynthetic membrane complexes in vivo and in vitro . In this work, the orientation of the pigments in the isolated photosystem Ⅱ (PSⅡ) sub_core reaction center complexes was analyzed and characterized by means of low temperature absorption and LD spectroscopy. The preparations containing different amounts of CP47 isolated from spinach (Spinacia oleracea L.) chloroplast were used in order to investigate the orientation of pigments in the PSⅡ sub_core CP47/D1/D2/Cyt b_559 (CP47/D1/D2) complexes. Chlorophyll a (Chl a) absorbing at 680 nm in CP47/D1/D2/Cyt b_559 complex showed an orientation of the Q y transition parallel to the membrane plane. It is proposed that there are two forms of β_carotene (β_Car) in CP47/D1/D2/Cyt b_559 complex, denoted as β_Car (Ⅰ) and β_Car (Ⅱ), with different orientations, β_Car (Ⅰ) at 470 and 505 nm is roughly parallel to the membrane plane, and β_Car (Ⅱ) at 460 and 490 nm seems to be perpendicular orientation. Upon the photoinhibitory experiment β_Car (Ⅱ) was found to be photosensitive and easily photodamaged. It also showed that the positive LD signal observed at 680 nm was quite complicated. This signal is tentatively attributed to P680 and some Chl a of antenna in CP47 protein based upon our measurements.
文摘Object To investigate the therapeutic effect of microencapsulated porcine retinal pigmented epithelial cells (RPE-M) transplantation on rat model of Parkinson's disease (PD). Methods Primary porcine RPE cells were harvested by enzyme digestion and expanded in culture medium. Determine the levels ofdopamine (DA) and homovanillic acid (HVA) by high performance liquid chromatography electrochemical (HPLC) assay, and the levels of brain-derived neurotrophic factor (BDNF) and glial-derived neurotrophic factor (GDNF) were detected by ELISA. Alginate-polylysine-alginate (APA) microencapsulated cells were produced by using a high voltage electrostatic system. PD rat model was established by unilateral injection of 6-hydroxydopamine (6-OHDA) into the medial forebrain bundle (MFB). After that, the RPE-M was transplanted into the corpus striatum of PD rat, and then the rotation test scores were recorded and biochemical changes of the corpus striatum were tested. Results The levels of DA, HVA, BDNF and GDNF secreted by RPE were stable in the RPE culture supernatant and were not changed by the microencapsulation. Eighty-three percent rats developed PD by unilateral lesion of 6-OHDA in the MFB. The RPE-M transplantation had therapeutic effect on 33% PD rats. Conclusion Porcine RPE cells grow actively in vitro and could secrete DA, HVA, BDNF, and GDNF constantly, which does not be affected by the passage culture and the APA miroencapsulation. RPE-M transplantation of may be a curative therapy for PD.

文摘The effects of polishing on proximate compositions,physico-chemical characteristics,mineral compositions and antioxidant properties of the rice flours obtained from three different pigmented rice varieties(Chak-hao Angangba,Chak-hao Amubi and Chak-hao Poireiton) were investigated.The rice varieties were significantly(P < 0.05) different in the contents of the test characteristics.Lipids,ash,minerals,phytochemicals(phenolic acids and flavonoids) and 2,2-diphenyl-1-picrylhydrazyl(DPPH) activity of rice flours were decreased after polishing(9% degree of milling),while amylose content and lightness were increased.X-ray diffraction pattern of rice flours exhibited A-type crystalline pattern with reflections at 15.1o,17.1o,18.2o and 23.0o.Pasting properties and transition temperatures were decreased after polishing treatment.Polishing resulted in changes in the crystallinity,enthalpy and morphology of rice flours.

文摘AIM: To determine the effect of 7-methylxanthine (7-MX) on the posterior sclera of form-deprivation myopia (FDM) in pigmented rabbits. METHODS: Sixteen pigmented rabbits were monocularly deprived (MD) by suturing the right eyelids after natural eye opening (ten-day old) for a period of 30 days. Two groups of pigmented rabbits were fed either 7-MX (30 mg per kg body weight; n=8) or vehicle control (saline equal volume with 7-MX; n=8). Ocular refractions, axial lengths and body weights were measured at the start and the end of the experiment 30 days later. Electron microscopy was used to measure and determine the collagen fibril diameters in the posterior pole of sclera. RESULTS: In vehicle control MD pigmented rabbits, 30 days of MD produced -1.10D+/- 0.78D of myopia and the axial length increased 0.51mm+/- 0.09mm. In MD pigmented rabbits fed with 7-MX, 30 days of MD induced only -0.21D+/- 0.11D of myopia and the axial length increased 0.07mm+/- 0.10mm. There was significant change in axial length of vehicle control MD pigmented rabbits (13.11mm +/- 0.19mm versus 12.60mm+/- 0.06mm; P =0.03). The changes in refraction and axial length of two MD groups' contalateral eyes during the 30 days were not significantly different (2.75D+/- 0.27D versus 2.75D 0.35D, P>0.05; 12.60mm+/- 0.06mm versus 12.45mm +/- 0.14mm, P >0.05). The weights of the two groups pigmented rabbits had no significant changes (187g+/- 22.1g versus 189g+/- 19.3g, P>0.05). The diameter cif scleral collagen fibers increased in both eyes of 7-MX treated pigmented rabbits. There was significant difference in collagen fibril diameters of inner layer (111.34nm+/- 28.30nm versus 94.80nm +/- 27.52nm, P =0.002) and outer layer (167.92nm +/- 55.82nm versus 144.04nm +/- 47.02nm P =0.016) in the posterior sclera between the myopic eyes of vehicle control MD group and contralateral eyes of 7-MX treated MD group. CONCLUSION: 7-MX appears to prevent FDM in pigmented rabbits by remodeling the posterior sclera.

文摘A novel kind of waterborne epoxy coating pigmented by nano-sized aluminium powders on high strength steel was formulated. Several coatings with different pigment volume content (PVC) were prepared. The coating morphology was observed using scanning electron microscopy (SEM), and the electrochemical properties were investigated by electrochemical impedance spectroscopy (EIS). Immersion test and neutral salt spray test were also conducted to investigate the corrosion resistance of the coating. It is demonstrated that the critical pigment volume content (CPVC) value is between 30% and 40%. The coating with PVC of 30% exhibits good corrosion resistance in 3.5% (mass fraction) NaCl solution.

文摘AIM:To evaluate the effects of LIN28A(human)on high glucose-induced retinal pigmented epithelium(RPE)cell injury and its possible mechanism.METHODS:Diabetic retinopathy model was generated following 48h of exposure to 30 mmol/L high glucose(HG)in ARPE-19 cells.Quantitative real-time polymerase chain reaction(qRT-PCR)and Western blot tested the expression of the corresponding genes and proteins.Cell viability as well as apoptosis was determined through cell counting kit-8(CCK-8)and flow cytometry assays.Immunofluorescence assay was adopted to evaluate autophagy activity.Caspase 3 activity,oxidative stress markers,and cytokines were appraised adopting their commercial kits,respectively.Finally,ARPE-19 cells were preincubated with EX527,a Sirtuin 1(SIRT1)inhibitor,prior to HG stimulation to validate the regulatory mechanism.RESULTS:LIN28A was downregulated in HG-challenged ARPE-19 cells.LIN28A overexpression greatly inhibited HGinduced ARPE-19 cell viability loss,apoptosis,oxidative damage as well as inflammatory response.Meanwhile,the repressed autophagy and SIRT1 in ARPE-19 cells challenged with HG were elevated after LIN28A overexpression.In addition,treatment of EX527 greatly inhibited the activated autophagy following LIN28A overexpression and partly abolished the protective role of LIN28A against HG-elicited apoptosis,oxidative damage as well as inflammation in ARPE-19 cells.CONCLUSION:LIN28A exerts a protective role against HG-elicited RPE oxidative damage,inflammation,as well as apoptosis via regulating SIRT1/autophagy.

文摘Upwelling occurs on the coast of Java between June and October, forced by local alongshore winds associated with the southeasterly monsoon. This causes variations in phytoplankton community composition in the upwelling zone compared with the surrounding offshore area. Based on pigments analysis with subsequent calculations of group contributions to total chlorophyll a(Chl a) using CHEMTAX, we studied the distribution and composition of phytoplankton assemblages in the subsurface chlorophyll maximum along the south coast of Java and the influence of upwelling. Nineteen phytoplankton pigments were identified using high-performance liquid chromatography, and CHEMTAX analysis associated these to ten major phytoplankton groups. The phytoplankton community in the coastal area influenced by upwelling was characterized by high Chl a and fucoxanthin concentrations, indicating the dominance of diatoms. In contrast, in the offshore area, the Chl a and fucoxanthin concentrations declined to very low levels and the community was dominated by haptophytes represented by 19′-Hexanoyloxyfucoxanthin. Accordingly, microphytoplankton was found to be the major size class in the coastal area influenced by upwelling, while nanophytoplankton was most abundant in the offshore area. Low concentrations of other accessory pigments indicated less contribution from dinoflagellates,prasinophytes, chlorophytes and cryptophytes. Photo-pigment indices revealed that photosynthetic carotenoids(PSCs) were the largest component of the pigment pool, exceeding the proportion of Chl a, with the average PSCTP up to 0.62. These distribution trends can mainly be explained by phytoplankton adaption strategies to upwelling and subsurface conditions by changing species composition and adjusting the pigment pool.

文摘The study of training hyperparameters optimisation problems remains underexplored in skin lesion research.This is the first report of using hierarchical optimisation to improve computational effort in a four-dimensional search space for the problem.The authors explore training parameters selection in optimising the learning process of a model to differentiate pigmented lesions characteristics.In the authors'demonstration,pretrained GoogleNet is fine-tuned with a full training set by varying hyperparameters,namely epoch,mini-batch value,initial learning rate,and gradient threshold.The iterative search of the optimal global-local solution is by using the derivative-based method.The authors used non-parametric one-way ANOVA to test whether the classification accuracies differed for the variation in the training parameters.The authors identified the mini-batch size and initial learning rate as parameters that significantly influence the model's learning capability.The authors'results showed that a small fraction of combinations(5%)from constrained global search space,in contrarily to 82%at the local level,can converge with early stopping conditions.The mean(standard deviation,SD)validation accuracies increased from 78.4(4.44)%to 82.9(1.8)%using the authors'system.The fine-tuned model's performance measures evaluated on a testing dataset showed classification accuracy,precision,sensitivity,and specificity of 85.3%,75.6%,64.4%,and 97.2%,respectively.The authors'system achieves an overall better diagnosis performance than four state-of-the-art approaches via an improved search of parameters for a good adaptation of the model to the authors'dataset.The extended experiments also showed its superior performance consistency across different deep networks,where the overall classification accuracy increased by 5%with this technique.This approach reduces the risk of search being trapped in a suboptimal solution,and its use may be expanded to network architecture optimisation for enhanced diagnostic performance.

文摘Bacterial abundance, phytoplankton community structure and environmental parameters were investigated to study the relationships between bacteria and phytoplankton during giant jellyfish Nemopilema nomurai blooms in the central Yellow Sea during 2013. N. nomurai appeared in June, increased in August, reached a peak and began to degrade in September 2013. Results showed that phosphate was possible a key nutrient for both phytoplankton and bacteria in June, but it changed to nitrate in August and September. Phytoplankton composition significantly changed that pico-phytoplankton relative biomass significantly increased, whereas other size phytoplankton significantly decreased during jellyfish bloom. In June, a significantly positive correlation was observed between chlorophyll a concentration and bacterial abundance(r=0.67, P0.001, n=34).During jellyfish outbreak in August, there was no significant correlation between phytoplankton and bacteria(r=0.11, P0.05, n=25), but the relationship(r=0.71, P0.001, n=31) was rebuilt with jellyfish degradation in September. In August, small size phytoplankton occupied the mixed layer in offshore stations, while bacteria almost distributed evenly in vertical. Chlorophyll a concentration significantly increased from(0.42±0.056) μg/L in June to(0.74±0.174) μg/L in August, while bacterial abundance just slightly increased. Additionally, the negative net community production indicated that community respiration was not entirely determined by the local primary productivity in August. These results indicated that jellyfish blooms potentially affect coupling of phytoplankton and bacteria in marine ecosystems.

文摘AIM: To find out an animal-free, xeno-free culture method for human fetal retinal pigment epithelium(fRPE) cells aiming for cell-replacement therapy. METHODS: Human AB serum, knock-out serum replacement(KSR) and B27 were supplemented as a substitute of fetal bovine serum(FBS) in culture medium of human fRPE cells. Cell morphology was examined by light microscope and transmission electron microscope. Proliferation ability was detected by cell cycle analysis and examination of KI67 expression. Apoptosis was analyzed using FACS. The expression ofRPE-specific markers was demonstrated by quantitative real-time polymerase chain reaction(qPCR), Western blot(WB) and immunocytochemistry. Paracrine function was determined using enzyme-linked immunosorbent assay method.RESULTS: Our results indicated that the optimum concentration of KSR was 15%, the optimum concentration of B27 was 2%, and the optimum concentration of human AB serum was 10%. fRPE cells cultured in 15% KSR and 2% B27 media showed repressed propagation and differentiation ability, and gradually lost epithelial morphology and RPE function. While fRPE cells cultured in 10% human AB serum media showed a typical cobblestone morphology with pigmentation, elevated proliferation ability, satisfying paracrine function and expressed RPE-specific markers. CONCLUSION: Our study indicates that culturing fRPE cells in 10% human AB serum-supplemented medium is more favorable compared with KSR, B27 and traditional FBS-supplemented mediums when fRPE cells are to be applied in cell-based therapy.
文摘BACKGROUND Pigmented villonodular synovitis(PVNS)is an uncommon disease that usually occurs in large joints,and involvement of the subtalar joint is rare.The lack of comprehensive knowledge of subtalar joint PVNS could lead to misdiagnosis.CASE SUMMARY We present a 64-year-old woman who,at her first visit,complained of discomfort in the right ankle when she walked.Based on the physical signs and X-ray report,the physician failed to make the suspected diagnosis of PVNS.Eighteen months later,the patient returned with a complaint of a mass in her right lateral malleolus with intermittent blunt pain.The X-ray presented an osteophyte formation and soft tissue calcification at the margin of the subtalar joint.The laboratory tests were normal,whereas magnetic resonance imaging(MRI)showed a low-intensity area on both T1-and T2-weighted images.A suspected diagnosis of PVNS was made and later confirmed by postoperative pathology.Subsequently,the patient received radiotherapy with 32 Gy in 16 fractions.At 6 mo postoperatively,the patient only complained of discomfort after walking three blocks.The American Orthopedic Foot and Ankle Society Ankle-Hindfoot score was 97.CONCLUSION MRI is necessary for the diagnosis of PVNS.Early soft tissue calcification and painless joint swelling should be of concern.

文摘AIM: To demonstrate the feasibility of mesenchymal stem cell(MSC)-mediated nano drug delivery, which was characterized by the “Trojan horse”-like transport of hypoxiainducible factor-1α small interfering RNA(HIF-1α si RNA) between MSCs and retinal pigment epithelial cells(RPE) under hypoxia environment.METHODS: Plasmid and lentivirus targeting the human HIF-1α gene were designed and constructed. HIF-1α si RNA was encapsulated into poly(lactic-co-glycolic acid) nanoparticles(PLGA-NPs) through the water-in-oil-in-water(w/o/w) multiple emulsion technique. The effect of PLGANPs uptake on the expression of HIF-1α m RNA was tested in RPE cells by real-time quantitative polymerase chain reaction(q PCR) and additional transfected conditions were used as control, including lentivirus group, nude plasmid group and blank PLGA group. MSCs were transfected with the NPs and the transfection efficacy was evaluated by flow cytometry. Transwell co-culture system of transfected MSCs and RPE cells was constructed under hypoxia environment. The effects of MSC-loaded HIF-1α si RNA PLGA-NPs on proliferation, apoptosis, and migration of RPE cells were then evaluated. The effect of transfected MSCs on HIF-1α expression of RPE cells was analyzed by using q PCR at the time points 24h, 3d, and 7d.RESULTS: The average diameter of PLGA-NPs loaded with HIF si RNA was 314.1 nm and the zeta potential was-0.36 m V. The transfection efficiency of PLGA-NPs was 67.3%±5.2% into MSCs by using flow cytometry. Compared with the lentivirus group, the PLGA-NPs loaded with HIF-1α si RNA can effectively reduce the expression of HIF-1α m RNA up to 7d in RPE(0.63±0.05 at 7d, P<0.001). In the Transwell co-culture system of transfected MSCs and RPE, the abilities of proliferation(2.34±0.17, 2.40±0.28, 2.47±0.24 at 48h, F=0.23, P=0.80), apoptosis(14.83%±2.43%, 12.94%±2.19%, 12.39%±3.21%;F=0.70, P=0.53) and migration(124.5±7.78, 119.5±5.32, 130±9.89, F=1.33, P=0.33) of the RPE cells had no differences between MSCloaded HIF-1α si RNA PLGA-NPs and other groups. The inhibition of PLGA on the HIF-1α m RNA expression in RPE cells could continue until the 7th day, the level of HIF-1α m RNA was lower than that of other groups(F=171.98, P<0.001). CONCLUSION: The delivery of PLGA-NPs loaded with HIF-1α si RNA carried by MSCs is found to be beneficial temporally for HIF-1α m RNA inhibition in RPE cells under hypoxia environment. The MSC-based bio-mimetic delivery of HIF-1α si RNA nanoparticles is a potential method for therapy against choroidal neovascularization.
文摘Pigment printing was carried out on lab scale by simple screen-printing techniques. By the application of acrylate and butadiene based binder, the crocking fastness, formaldehyde release and PVC migration of fabric printed with Imparon red KB pigment was evaluated. The effect of curing time on K/S values was also investigated. It has been found that butadiene based binder shows good performance in terms of crocking fastness, formaldehyde release and PVC migra-tion. It has also been observed that by increasing the binder concentration, the release of formaldehyde decreased and by increasing the curing time, the K/S values of printed fabric were decreased.
文摘As a constituent of blood-retinal barrier and retinal outer segment(ROS) scavenger, retinal pigmented epithelium(RPE) is fundamental to normal function of retina. Malfunctioning of RPE contributes to the onset and advance of retinal degenerative diseases. Up to date, RPE replacement therapy is the only possible method to completely reverse retinal degeneration. Transplantation of human RPE stem cell-derived RPE(h RPESC-RPE) has shown some good results in animal models. With promising results in terms of safety and visual improvement, human embryonic stem cell-derived RPE(h ESC-RPE) can be expected in clinical settings in the near future. Despite twists and turns, induced pluripotent stem cell-derived RPE(i PSC-RPE) is now being intensely investigated to overcome genetic and epigenetic instability. By far, only one patient has received i PSCRPE transplant, which is a hallmark of i PSC technology development. During follow-up, no major complications such as immunogenicity or tumorigenesis have been observed. Future trials should keep focusing on the safety of stem cell-derived RPE(SC-RPE) especially in long period, and better understanding of the nature of stem cell and the molecular events in the process to generate SC-RPE is necessary to the prosperity of SC-RPE clinical application.
文摘Hidroacanthoma simplex(HAS) is a rare skin appendage tumor and is recognized as an intraepidermal variant of poroma. Pigmented HAS is an extremely rare variant with dendritic melanocytes and/or melanin pigment in the tumor cells and a few cases of pigmented HAS have been reported in the English literature. We herein report a case of pigmented HAS and discuss the clinicopathological features of pigmented HAS and the pitfall in the diagnosis of black lesions.
文摘<strong>Introduction:</strong> Intense Pulsed Light (IPL) technology is well accepted in the medical aesthetic field for the treatment of various skin lesions, including pigmented and vascular lesions. The light penetrates into the skin and is selectively absorbed by lesion chromophore. Absorbed energy is converted into heat, coagulating the lesion, which naturally fades following the treatment. The current article presents a retrospective efficacy evaluation of an IPL device with high peak power. <strong>Methods:</strong> Representative treatment results were collected from several clinics based on photographs taken at baseline and after treatments. Photos were evaluated and analyzed for aesthetic improvement of the various skin conditions in different facial and body areas. <strong>Results:</strong> Analysis included cases of pigmented and vascular lesions, textural lesions, and more specific conditions such as melasma and rosacea. The two evaluators’ scoring demonstrated improvement in all cases according to the Global Aesthetic Improvement Scale (GAIS) scale. <strong>Conclusion:</strong> The vast experience gathered from the market in treating various skin lesions supports the safety and efficacy of the investigated IPL device. The device’s particular specifications contribute to the successful results and ease of treatment for the practitioner and the patient.

文摘Objective:To study the inhibitory effect of rice bran extracts of Thai black Kam Muang and red Hawm Dawk Mali Deang on oxidative stress factors including superoxide(O2·-),nitric oxide(NO·),and inducible nitric oxide synthase(iNOS).Methods:Bran extracts(40%ethanol)of Kam Muang and Hawm Dawk Mali Deang were obtained and evaluated for in vitro 2-2′-azino-di-(3-ethylbenzthiazoline sulfonate)(ABTS)and NO·scavenging activity.Their inhibitory effects on cellular O2·-and NO·were measured in phorbol 12-myristate 13-acetate-stimulated neutrophil-like HL-60 cells and lipopolysaccharidestimulated RAW264.7 macrophages,respectively,and their viability was monitored using the MTT assay.The effect on iNOS expression was also assessed by the Western blotting assay.Total contents of phenolics,flavonoids,and subtypes were also determined.Results:Hawm Dawk Mali Deang exhibited about 3.5-fold greater cellular O2·-inhibitory activity than Kam Muang[EC50 values of(23.57±4.54)and(81.98±1.45)μg/mL,respectively]in phorbol 12-myristate 13-acetate-stimulated HL-60 cells.Hawm Dawk Mali Deang exhibited about 2-fold higher in vitro ABTS·+and NO·scavenging activity than Kam Muang,but it exerted cellular NO·inhibitory activity of only about 26%(undetermined EC50 value)in lipopolysaccharide-stimulated RAW264.7 cells.Conversely,Kam Muang exerted potent cellular NO·inhibitory activity[EC50 value:(281.13±59.18)μg/mL]and dose-dependently decreased iNOS levels.No cytotoxicity of both extracts was detected in both cell types.As for corresponding contents,Hawm Dawk Mali Deang contained higher contents of phenolics and flavonoids than Kam Muang.Moreover,Kam Muang and Hawm Dawk Mali Deang had a high content of total anthocyanins[(14.73±0.52)mg C3GE/g of extract]and total proanthocyanidins[(115.13±1.47)mg CE/g of extract],respectively.Conclusions:Based on these data,bran extracts of Thai black Kam Muang and red rice Hawm Dawk Mali Deang can help lower oxidative stress and inflammation attributed partly to O2·-and NO·.
