However, the immaturity of its stable transformation system and its low transformation efficiency limit gene function research on non-heading Chinese cabbage. Agrobacterium rhizogenes-mediated(ARM) transgenic technology is a rapid and effective transformation method that has not yet been established for non-heading Chinese cabbage plants. Here, we optimized conventional ARM approaches(one-step and two-step transformation methods) suitable for living non-heading Chinese cabbage plants in nonsterile environments. Transgenic roots in composite non-heading Chinese cabbage plants were identified using phenotypic detection, fluorescence observation, and PCR analysis. The transformation efficiency of a two-step method on four five-day-old non-heading Chinese cabbage seedlings(Suzhouqing, Huangmeigui, Wuyueman, and Sijiu Caixin) was 43.33%-51.09%, whereas using the stout hypocotyl resulted in a transformation efficiency of 54.88% for the 30-day-old Sijiu Caixin. The one-step method outperformed the two-step method;the transformation efficiency of different varieties was above 60%, and both methods can be used to obtain transgenic roots for functional studies within one month. Finally, optimized ARM transformation methods can easily, quickly, and effectively produce composite non-heading Chinese cabbage plants with transgenic roots, providing a reliable foundation for gene function research and non-heading Chinese cabbage genetic improvement breeding.展开更多

Wheat(Triticum aestivum L.)–maize(Zea mays L.)(W–M) rotation is the dominant planting pattern in the Huang-HuaiHai Plain and is important for food security in China. However, the soil quality is deteriorating due to the W–M rotation’s long-term, intensive, and continuous cultivation. Introducing legumes into the W–M rotation system may be an effective way to improve soil quality. In this study, we aimed to verify this hypothesis by exploring efficient planting systems(wheat–peanut(Arachis hypogaea L.)(W–P) rotation and wheat rotated with maize and peanut intercropping(W–M/P)) to achieve higher agricultural production in the Huang-Huai-Hai Plain. Using traditional W–M rotation as the control, we evaluated crop productivity, net returns, soil microorganisms(SMs), and soil organic carbon(SOC) fractions for three consecutive years. The results indicated that wheat yields were significantly increased under W–P and W–M/P(382.5–579.0 and 179.8–513.1 kg ha-1, respectively) compared with W–M. W–P and W–M/P provided significantly higher net returns(58.2 and 70.4%, respectively) than W–M. W–M/P and W–M retained the SOC stock more efficiently than W–P, increasing by 25.46–31.03 and 14.47–27.64%, respectively, in the 0–20 cm soil layer. Compared with W–M, W–M/P improved labile carbon fractions;the sensitivity index of potentially mineralizable carbon, microbial biomass carbon(MBC), and dissolved organic carbon was 31.5, 96.5–157.2, and 17.8% in 20–40, 10–40, and 10–20 cm soil layers, respectively. The bacterial community composition and bacteria function were altered as per the soil depth and planting pattern. W–M/P and W–M exhibited similar bacterial community composition and function in 0–20 and 20–40 cm soil layers. Compared with W–P, a higher abundance of functional genes, namely, contains mobile elements and stress-tolerant, and a lower abundance of genes, namely, potentially pathogenic, were observed in the 10–20 cm soil layer of W–M and the 0–20 cm soil layer of W–M/P. SOC and MBC were the main factors affecting soil bacterial communities, positively correlated with Sphingomonadales and Gemmatimonadales and negatively correlated with Blastocatellales. Organic input was the main factor affecting SOC and SMs, which exhibited feedback effects on crop productivity. In summary, W–M/P improved productivity, net returns, and SOC pool compared with traditional W–M rotation systems, and it is recommended that plant–soil–microbial interactions be considered while designing high-yield cropping systems.展开更多

We show how detailed vegetation plot data can be used to assess the potential impact of inundation by a proposed hydroelectricity dam in the Mokihinui gorge, New Zealand, on representation of natural forests. Specifically we ask: 1) How well are the types of forest represented Locally, regionally, and nationally; and 2) How does the number of distinct communities (i.e. beta diversity) in the target catchment compare with other catchments nationally? Methods: For local and regional comparisons plant species composition was recorded on 45 objectively located 400 m2 vegetation plots established in each of three gorges, with one being the proposed inundation area of the Mokihinui lower gorge. The fuzzy classification framework of noise clustering was used to assign these plots to a specific alliance and association of a pre-existing national-scale classification. NationaLly, we examined the relationship between the number of alliances and associations in a catchment and either catchment size or the number of plots per catchment by fitting Generalised Additive Models. Results: The four alliances and five associations that were observed in the Mokihinui lower gorge arepresent in the region but limited locally. One association was narrowly distributed nationally, but is the mostfrequent association in the Mokihinui lower gorge; inundation may have consequences of national importance to its long-term persistence. That the Mokihinui lower gorge area had nearly twice as many plots that could not be assigned to pre- existing alliances and associations than either the Mokihinui upper or the Karamea lower gorges and proportionally more than the national dataset emphasises the compositional distinctiveness of this gorge. These outlier plots in the Mokihinui lower gorge may be unsorted assemblages of species or reflect sampling bias or that native- dominated woody riparian vegetation is rare on the landscape. At a national scale, the Mokihinui catchment has a higher diversity of forest alliances and associations (i.e. beta-diversity) than predicted based on catchment size and sampling intensity. Conclusions: Our analytical approach demonstrates one transparent solution to a common conservation planning problem: assessing how well ecosystems that will be destroyed by a proposed land-use change are represented using a multi-scale spatial and compositional framework. We provide a useful tool for assessing potential consequences of land-use change that can help guide decision making.展开更多

The contamination removal efficiency and their trends in the wetland treatment system were studied under different hydraulic loading rates(HLR). The contamination removal efficiencies were compared according to the seasonal change under optimum HLR. The result shows that in the same season, under different hydraulic loadings ranging from 2 to 6 m3/(m2·d) at the same period, the best HLR is 4 m3/(m2·d) in the experimental system. The average removal rates of COD, TN, ammoniacal nitrogen(NH4+-N), and TP in the constructed wetland are 38.37%, 45.97%, 39.86% and 41.69%, respectively. According to China Standard for Surface Water Resources (GB3838-2002), mean effluent of COD, TN, NH4+-N and TP can nearly reach Grade Ⅲ, GradeⅤ, GradeⅠand GradeⅠ, respectively. Furthermore, treatment efficiency of the system in summer is obvious higher than that in other seasons. The expenditure of constructing the constructed wetland with the average treating capacity of 176 m3/d and lifetime of 20 years is 17075.00 RMB. The average disposal cost is summed up to 0.17 RMB/m3, which shows that the pretreatment of the micro-polluted Yellow River raw water by constructed wetland is feasible.展开更多

Plants are exposed to different stresses, which may change lignin content and composition. In many cases, particularly for plant-microbe interactions, this has been suggested as defence responses of plants to the stress. Thus, understanding how a stressor modulates expression of the genes related with lignin biosynthesis may allow us to develop study-models to increase our knowledge on the metabolic control of lignin deposition in the cell wall. This review focuses on recent literature reporting on the main types of abiotic and biotic stresses that alter the biosynthesis of lignin in plants.展开更多

We systematically investigated n-alkane δD values from 51 submerged plants(39 Potamogeton, 1 Myriophyllum, and 11 Ruppia), 13 algae(5 Chara, 3 Cladophora, and 5 Spirogyra) and 20 terrestrial plants(10 grasses and 10 shrubs) in and around 15 lakes on the Tibetan Plateau. Our results demonstrate that δD values of C_(29) nalkane are correlated significantly with the lake water δD values both for algae(R^2=0.85, p<0.01, n=9) and submerged plants(R^2=0.90, p<0.01, n=25), indicating that δD values of these algae and submerged plants reflect the δD variation of lake water. We find that apparent hydrogen isotope fractionation factors between individual n-alkanes and water(εa/w) are not constant among different algae and submerged plants, as well as in a single genus under different liminological conditions, indicating that the biosynthesis or environmental conditions(e.g. salinity) may affect their δD values. The δD values of submerged plant Ruppia in the Xiligou Lake(a closed lake) are significant enriched in D than those of terrestrial grasses around the lake(one-way ANOVA,p<0.01), but the algae Chara in the Keluke Lake(an open lake) display similar δD values with grasses around the lake(one-way ANOVA, p=0.826>0.05), suggesting that the n-alkane δD values of the algae and submerged plants record the signal of D enrichment in lake water relative to precipitation only in closed lakes in arid and semi-arid area. For each algae and submerged plant sample, we find uniformed δD values of different chain length n-alkanes, implying that, in combination with other proxies such as Paq and Average Chain Length, the offset between the δD values of different chain length n-alkanes can help determine the source of sedimentary n-alkanes as well as inferring the hydrological characteristics of an ancient lake basin(open vs closed lake).展开更多

文摘Lignin is a polymer of phenylpropanoid compounds formed through a complex biosynthesis route, represented by a metabolic grid for which most of the genes involved have been sequenced in several plants, mainly in the model-plants Arabidopsis thaliana and Populus. This review focuses on recent literature reporting on the main types of abiotic and biotic stresses that alter the biosynthesis of lignin in plants.

文摘Aims Species diversity–productivity relationships in natural ecosystems have been well documented in the literature. However, biotic and abiotic factors that determine their relationships are still poorly understood, especially under future climate change scenarios. Methods Randomized block factorial experiments were performed in three meadows along an elevational gradient on Yulong Mountain,China, where open-top chambers and urea fertilizer manipulations were used to simulate warming and nitrogen addition, respectively.Besides species diversity,we measured functional diversity based on five traits:plant height, specific leaf area and leaf carbon, nitrogen and phosphorus contents.Several abiotic factors relating to climate(air temperature and precipitation)and soil chemistry(pH,organic carbon concentration, total nitrogen concentration and phosphorus concentration)were also measured.Generalized linear mixed-effect models were used to investigate the responses of species diversity and productivity to elevation, warming, nitrogen addition and their interactions. The effects of biotic and abiotic factors on the direction and magnitude of their relationship were also assessed.Important Findings Species diversity decreased with increasing elevation and declined under warming at mid-elevation,while productivity decreased with increasing elevation.Functional richness, maximum air temperature, soil pH and their interactions showed strong but negative influences on the species diversity–productivity relationship;the relationship shifted from positive to neutral and then to slightly negative as these sources of variation increased. Our study highlights the negative effects of short-term warming on species diversity and emphasizes the importance of both biotic and abiotic drivers of species diversity–productivity relationships in mountain meadow communities.

文摘The hydrogen isotopic composition(δD) of n-alkanes in lacustrine sediments is widely used in palaeoenvironmental studies, but the heterogeneous origins and relative contributions of these lipids provide challenges for the interpretation of the increasing dataset as an environment and climatic proxy. For each algae and submerged plant sample, we find uniformed δD values of different chain length n-alkanes, implying that, in combination with other proxies such as Paq and Average Chain Length, the offset between the δD values of different chain length n-alkanes can help determine the source of sedimentary n-alkanes as well as inferring the hydrological characteristics of an ancient lake basin(open vs closed lake).
