[Objective] The aim was to establish the optimal rye-specific PCR reaction system for rye.[Method] The ordinary wheat "Chinese Spring",S165,rye,octoploid triticale and hexaploid triticale were used as materials to c...[Objective] The aim was to establish the optimal rye-specific PCR reaction system for rye.[Method] The ordinary wheat "Chinese Spring",S165,rye,octoploid triticale and hexaploid triticale were used as materials to carry out study on the effect of the amount of template DNA,primers,dNTPs,Mg2+ concentrations,Taq DNA polymerase and annealing temperature on the rye-specific PCR reaction system of rye.[Result] The genomic DNA extracted by modified CTAB DNA extraction method showed high quality,which was satisfied for the PCR reaction template.The rye-specific PCR reaction system was 25 μl,including 10 × buffer solution,1.5 mmol/L MgCl2,200 μmol/L dNTP,40 ng primers,40-60 ng DNA template and 1 U Taq DNA polymerase.[Conclusion] The optimal rye-specific PCR reaction system was established,which provided basis for the identification of exogenous germplasm of rye in wheat background.展开更多

Time required and reagent cost for both assays were evaluated.RESULTS: Real-time PCR detected 100, 50, 10, 1 and 0.1% of YIDD plasmid, whereas LDR detected 100, 50, 10, 1, 0.1, and 0.01% of YIDD plasmid, in mixtures with YMDD plasmid of 106 copies/mL. Among the 52 clinical serum samples, completely concordant results were obtained for all samples by both assays, and 39 YIDD, 9 YVDD, and 4 YIDD/YVDD were detected. Cost and time required for LDR and real-time PCR are 60/80 CNY (8/10.7 US dollars) and 4.5/2.5 h, respectively.CONCLUSION: LDR and real-time PCR are both sensitive and inexpensive methods for monitoring low abundant YMDD mutants during lamivudine therapy in patients with chronic hepatitis B. Forty cases...A technique has been developed using PCR to detect monoclonality of B-lymphoproliferative disorders. DNA was extracted from the blood, tissue and paraffin embedded sections by biochemical means or boiling. Forty cases of B-non Hodgkin's lymphoma (NHL), 15 cases of T-NHL, 8 cases of chronic lymphocytic leukemia, 17 cases of reactive lymphadenopathy and 12 cases of various non-lym-phocytic tumor were examined. Monoclonality of B-lymphocytes was detected in 86-92% of cases with B-lymphoproliferative diseases, but none in T-NHL, reactive disorders and non-lymphatic tumors. This technique provides a new molecular biologic method to diagnose malignant B-lymphoproliferative dicor-ders. It may be useful in Ig gene rearrangement study, differential diagnosis and retrospective investigation of lymphoproliferative disorders.展开更多
