The content of polyphenol compound in 10 processing apple varieties (4 cider and 6 juice varieties) were analyzed using the Folin-Ciocalteu method and HPLC. The degree of browning and the activities of polyphenol oxidase were also studied. The content and proportion of the polyphenol varied depending on the variety. Bitter varieties globally showed a higher polyphenol concentration than sweet or acid varieties. Proanthocyanidins, chlorogenic acid, (+)-catechin, (-)-epicatechin were high-concentrated polyphenols in apple fruits. Phloridzin, the unique polyphenol of apple, was abundant in the bitter variety Frequin rouge fruit. Total polyphenols, proanthocyanidins, (+)-catechin, and phloridzin had higher correlations with browning. The correlation was low between chlorogenic acid and browning. The polyphenolic profiles were correlated with the apple types. Cider apples contained more polyphenol than juice apple varieties. The content of flavan-3-ol has a close relationship with fruit browning.展开更多

Response surface methodology and Box–Behnken design were used to obtain predictive model (multivariate quadratic regression equation) and optimization of the extraction process. The response surface was analyzed by using the yields of total polyphenolic content as response value. Results: Based on the obtained results the optimum extraction condition, including microwave power of 30% Watts, extraction time of 18.5 min, the ionic liquid concen-tration of 0.79 mol/L, and the liquid–solid ratio of 10.72 mL/g 1-Buthyl-3-methylimidazolium tetrafluoroborate ([bmim]BF4) as a solvent was selected. The regression model was obtained to predicts the yields from Peperomia pellucida:Y = 30.250 – 1.356X1 + 2.655X2 + 2.252X3 – 0.565X4 + 0.990 X1X3 – 8.172 X1X4 – 3.439 X3X4 – 4.178 X12 – 3.210 X32 – 6.786 X42 – 7.290 X12X3 + 5.575 X1X32 – 4.843 X32X4 with R2 = 0.82519. Scale-up confirmation test was obtained the maximum yields of total polyphenolics content with the amount of 31.1725μg GAE/g. The total polyphenolic compounds content was determined by the Folin Ciocalteu and butanol-HCI methods.In vitro antioxidant activity was performed using ABTS and ferric reducing antioxidant power methods.Results: Mexican rambutan husk showed a total polyphenolic content of 582 mg/g and an evident antioxidant activity by ABTS and ferric reducing antioxidant power analysis.The HPLC/ESI/MS assay allowed the identification of 13 compounds, most of which belong to ellagitannins. Geraniin, corilagin and ellagic acid were present in the sample;the mineral composition was also evaluated.Conclusions: Rambutan husk cultivated in Mexico is a promising source for the recovery of added value bioactive compounds with antioxidant activity, which have potential applications as bioactive antioxidant agents for the treatment of diseases.展开更多
Objective:To evaluate the antioxidant and radical scavenging activities of Solanum anguivi fruit(SAG)and its possible effect on mitochondrial permeability transition pore as well as mitochondrial membrane potential(△...Objective:To evaluate the antioxidant and radical scavenging activities of Solanum anguivi fruit(SAG)and its possible effect on mitochondrial permeability transition pore as well as mitochondrial membrane potential(△Ψ_m)isolated from rat liver.Methods:Antioxidant activity of SAG was assayed by using 2,2-diphenyl-1-picrylhydrazyl(DPPH),reducing power,iron chelation and ability to inhibit lipid peroxidation in both liver and brain homogenate of rats.Also,the effect of SAG on mitochondrial membrane potential and mitochondrial swelling were determined.Identification and quantification of bioactive polyphenolics was done by HPLC-DAD.Results:SAG exhibited potent and concentration dependent free radical-scavenging activity(IC_(50)/DPPH=275.03±7.8μg/mL).Reductive and iron chelation abilities also increase with increase in SAG concentration.SAG also inhibited peroxidation of cerebral and hepatic lipids subjected to iron oxidative assault.SAG protected against Ca^(2+)(110μmol/L)-induced mitochondrial swelling and maintained the△Ψ_m.HPLC analysis revealed the presence of gallic acid[(17.54±0.04)mg/g],chlorogenic acid(21.90±0.02 mg/g),caffeic acid(16.64±0.01 mg/g).rutin[(14.71±0.03)mg/g]and quercetin[(7.39±0.05)mg/g].Conclusions:These effects could be attributed to the bioactive polyphenolic compounds present in the extract.Our results suggest that SAG extract is a potential source of natural antioxidants that may be used not only in pharmaceutical and food industry but also in the treatment of diseases associated with oxidative stress.展开更多

Methods: Rambutan husk extracts were obtained by ultrasound-assisted extraction, mass/volume ratio, water/ethanol percentage and extraction time were evaluated. Once the best extraction condition of polyphenolic compounds was defined, a polyphenolic fraction was recovered using Ambetlite XAD-16. The total content of antioxidant polyphenolic compounds was determined by summation of the total hydrolysable polyphenol and total condensed polyphenol contents. Recovered compounds were identified by FTIR(ATR) spectroscopy and HPLC/ESI/MS. The antioxidant activity was carried out by ABTS, DPPH and lipid oxidation inhibition in vitro methods. Results: In Mexican variety rambutan husk, the total polyphenolic content was 487.67 mg/g, after ultrasound-assisted extraction. According to the HPLC/ESI/MS analysis 12 antioxidant polyphenolic compounds were identified, mostly ellagitannins such as geraniin, corilagin and ellagic acid. The antioxidant activity determined by ABTS, DPPH and lipid oxidation inhibition methods was demonstrated. The main functional groups of the identified compounds were determined by FTIR analysis. Conclusions: It was demonstrated that ultrasound-assisted extraction was effective and allowed the extraction and recovery of antioxidant polyphenolic compounds. Furthermore Mexican variety rambutan husk is an important source for recovering polyphenolic compounds with antioxidant activity, these compounds have potential application for the treatment/prevention of various diseases related to cancer and pathogenic microorganisms.展开更多

Moreover, both the size of LDL particle...The occurrence of Metabolic Syndrome (MS) represents an independent risk factor for developing cardiovascular disease states in patients suffering from type 2 diabetes mellitus. Moreover, both the size of LDL particles and liver dysfunction identified as non alcoholic fatty liver disease (NAFLD) represent important biomarkers for the development of cardiometabolic risk in patients with MS. Here we studied the effect of bergamot polyphenolic fraction (BPF) in patients with MS and NAFLD. 107 patients were enrolled at the San Raffaele IRCCS (Rome). All of them showed ultrasonografic evidences of NAFLD and at least three out of five previous identified criteria for the diagnosis of MS. Patients were divided into two groups: one receiving placebo and the second receiving BPF 650 mg twice a day for 120 consecutive days. In the group receiving BPF 650 mg twice a day, a significant reduction of fasting plasma glucose, serum LDL cholesterol and triglycerides alongside with an increase of HDL cholesterol was found. This effect was accompanied by significant reduction of both ultrasonographic and metabolic biomarkers of NAFLD. Moreover, a significant reduction of small dense LDL particles, as detected via proton NMR Spectroscopy, was found after BPF treatment. In conclusion, our data confirm the beneficial effect of bergamot-extract in patients with MS an effect highlighted by significant reduction of small dense LDL particles and by improvement of NAFLD biomarkers. This suggests a potential preventive role of bergamot derivatives in reducing cardiometabolic risk.展开更多

The design of experiment (DoE) was conducted to evaluate the influence of acetone concentration % (v/v), solid-to solvent ratio % (w/v), temperature (˚C) and extraction time (min) and their interaction on phenolic contents, using the Central Composite Rotatable Design (CCRD). The experimental data were analysed to fit statistical models for recovery of phenolic compounds. The selected models were significant (P 0.05), except for Chlorogenic acid and Quercetin 3-glucoside which had significant lack of fits (P R2 > 0.9000 and adjusted? ?reasonable agrees with predicted . Coefficient of variation < 5% for each determination at the 95% confidence interval. These models could be relied upon to achieve optimal concentrations of polyphenolic compounds for applications in nutraceutical, pharmaceutical and cosmetic industries.展开更多
Polyphenolic compounds with relatively high antioxidant activity obtained from subcritical water extraction of apple pomace were assessed for encapsulation by spray drying technique, making use of polymeric substances...Polyphenolic compounds with relatively high antioxidant activity obtained from subcritical water extraction of apple pomace were assessed for encapsulation by spray drying technique, making use of polymeric substances co-extracted with the polyphenolic compounds. Comparative assessments were carried out of the directly encapsulated subcritical water extract (SWE) products with particles formed when encapsulated with the addition of hydroxyl propyl-β-Cyclodextrin (SWE + HPβ-CD). The powders were characterized for their physico-chemical properties such as, moisture content, density, particle size, hygroscopicity to assess their suitability within cosmetic formulations. The SWE and SWE + HPβ-CD encapsulated products resulted in different physical properties. Although the particle size was less than 4 μm for both products, the direct encapsulation (SWE) was highly hygroscopic and this property was significantly reduced with addition of HPβ-Cyclodextrin (SWE + HPβ-CD). Scanning electron microscopy (SEM) and Fourier Transform Infrared (FT-IR) spectroscopic were em-ployed to analyse the micronised powders to support evidence of encapsulation. Both techniques revealed the interaction between compounds in extract and the carrier HPβ-Cyclodextrin suggesting successful encapsulation. The effect of storage conditions on retention of antioxidant activity of the subcritical water extract was evaluated within 35 days for extracts with and without the carrier HPβ-Cyclodextrin. Hydroxyl propyl-β-Cyclodextrin offered protection against degradation of antioxidant compounds thereby potentially extending the shelf-life and making the encapsulated powder suitable for incorporation in cosmetic and pharmaceutical applications.展开更多
Strawberries are rich in polyphenols which impart health benefits when metabolized by the gut microbiome,including anti-inflammatory,neuroprotective,and antiproliferative effects.In addition,polyphenolic anthocyanins ...Strawberries are rich in polyphenols which impart health benefits when metabolized by the gut microbiome,including anti-inflammatory,neuroprotective,and antiproliferative effects.In addition,polyphenolic anthocyanins contribute to the attractive color of strawberry fruits.However,the genetic basis of polyphenol biosynthesis has not been extensively studied in strawberry.In this investigation,ripe fruits from three cultivated strawberry populations were characterized for polyphenol content using HPLC-DAD-MSn and genotyped using the iStraw35k array.GWAS and QTL analyses identified genetic loci controlling polyphenol biosynthesis.QTL were identified on four chromosomes for pelargonidin-3-O-malonylglucoside,pelargonidin-3-O-acetylglucoside,cinnamoyl glucose,and ellagic acid deoxyhexoside biosynthesis.Presence/absence of ellagic acid deoxyhexoside and pelargonidin-3-O-malonylglucoside was found to be under the control of major gene loci on LG1X2 and LG6b,respectively,on the F.×ananassa linkage maps.Interrogation of gene predictions in the F.vesca reference genome sequence identified a single candidate gene for ellagic acid deoxyhexoside biosynthesis,while seven malonyltransferase genes were identified as candidates for pelargonidin-3-O-malonylglucoside biosynthesis.Homologous malonyltransferase genes were identified in the F.×ananassa‘Camarosa’genome sequence but the candidate for ellagic acid deoxyhexoside biosynthesis was absent from the‘Camarosa’sequence.This study demonstrated that polyphenol biosynthesis in strawberry is,in some cases,under simple genetic control,supporting previous observations of the presence or absence of these compounds in strawberry fruits.It has also shed light on the mechanisms controlling polyphenol biosynthesis and enhanced the knowledge of these biosynthesis pathways in strawberry.The above findings will facilitate breeding for strawberries enriched in compounds with beneficial health effects.展开更多

The made...This study was carried out to characterize total residual catechins and their fractions, polyphenolic contents and antioxidant activities of black teas enriched with high levels of theaflavin-3,3’-digallate. The made teas were processed from eleven selected cultivars. A comparative study was carried out between the processed teas and those from commercially grown Kenyan cultivars in relation to the above chemical parameters. A correlation matrix analysis was also conducted to find out whether a relationship existed between the antioxidant activities and the said chemical parameters. The total residual catechins were found to range between 3.10% and 8.08%. The total polyphenol levels varied between 19.00% and 28.90%, while the antioxidant activities of the teas ranged from 82.70% to 91.70%. There was a significant p < 0.001 correlation between the antioxidant activity and total polyphenols (r = 0.8948). There was also a high correlation p < 0.001 between the antioxidant activity and total catechins (r = 0.8878). Out of the four catechin fractions, the antioxidant activity correlated most with EGCG (r = 0.8774). The total polyphenolic contents and antioxidant activities for most of the cultivars were comparable to those of the green tea reference standard. From the figures obtained, it can be concluded that the most of the newly developed black teas of the selected cultivars have higher quality and enhanced antioxidant activities and that they can be recommended for commercial production.展开更多

A paired match approach was used to switch 20 pigs (range of initial body weight, BW: 113 - 121 kg) to two experimental diets, for four weeks: ten pigs (pair housed) were fed with an experimental acorn based diet (acorns: 50% in the diet, as fed;total polyphenols, 78.1 g TP/Kg DM in the diet;tannic acid equivalent, 25.8 g TAE/kg DM);the remainder ten, received a pelleted complete diet for finishers (0% acorns in the diet). Daily feed intake in the last two weeks of the experimental feeding was recorded per pair of pigs in both groups of animals, showing an average intake of 610 mg TAE/kg BW/d. At an average final BW of between 127 to 137 kg, all pigs were slaughtered and LNs from different anatomical regions of the carcass were removed and analysed. At gross inspection, LNs from both groups displayed different grades of intensity and diffusion of pigmentation: a partial and incidental pigmentation was randomly detected in renal or sub-iliac LNs in the control group;a constant and uniform pigmentation of LNs was observed in acorns fed pigs: a dark brown staining diffused to the whole LN associated with a brownish colour of the muscles was found systematically. At light microscope intracytoplasmic granules were found in macrophages and dendritic cells from both groups, but, at confocal laser analysis, an intense auto-fluorescence was observed in medial-iliac LNs from the carcasses of acorn-fed pigs (green emission). However, intracellular sources of blue and green fluorescence at different wavelengths, likely due to tryptophan, indoleamine and derivates were also found in medial-iliac and inguinal LNs from the control group. A dietary origin was attributed to the different discoloration of LNs between the carcasses of the two groups: such acquired pigmentation is relevant in the left sub-iliac LN, but the confocal laser microscopic test to elicit auto-fluorescence of polyphenolic compounds (biological markers) displayed a 76.9% specificity, despite a 100% of sensitivity for the univocal identification of the carcass from acorn-fed pigs. A rapid method was presented for the determination of 19 polyphenols in tea by ultra-high performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF MS).Tea samples were extr...A rapid method was presented for the determination of 19 polyphenols in tea by ultra-high performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF MS).Tea samples were extracted by 50%(V/V)ethanol,then separated by Waters Acquity BEH C18 column using a binary solvent system composed of acetonitrile and water(0.1%formic acid)by gradient elution.The analytes were determined by Q-TOF MS in TOF MS and information dependent acquisition(IDA)-MS/MS mode.The results showed that mass accuracy error of the 19 polyphenols were lower than 5.0×10^(-6),good linear relationship was got in range of 0.2–500μg/L and correlation coefficient was higher than 0.9990.The LOD was in the range of 0.002–0.100 mg/kg and the LOQ was in the range of 0.004–0.200 mg/kg.Recovery of the method was in range of 78.4%–109.2%with spike levels of 0.004–2.000 mg/kg,relative standard deviations were lower than 10%.The method was simple,rapid and accurate.It could be used for the rapid screening and quantitative analysis of 19 polyphenols in tea.展开更多
文摘Polyphenolic compound in processing apple (Malus domestica Borkh.) varieties and the relationship between polyphenol content and enzymatic browning were studied to provide reference for raw material selection and processing method optimization. The content of polyphenol compound in 10 processing apple varieties (4 cider and 6 juice varieties) were analyzed using the Folin-Ciocalteu method and HPLC. The degree of browning and the activities of polyphenol oxidase were also studied. The content and proportion of the polyphenol varied depending on the variety. Bitter varieties globally showed a higher polyphenol concentration than sweet or acid varieties. Proanthocyanidins, chlorogenic acid, (+)-catechin, (-)-epicatechin were high-concentrated polyphenols in apple fruits. Phloridzin, the unique polyphenol of apple, was abundant in the bitter variety Frequin rouge fruit. Total polyphenols, proanthocyanidins, (+)-catechin, and phloridzin had higher correlations with browning. The correlation was low between chlorogenic acid and browning. The polyphenolic profiles were correlated with the apple types. Cider apples contained more polyphenol than juice apple varieties. The content of flavan-3-ol has a close relationship with fruit browning.

文摘Objective: To optimize the ionic liquid based microwave-assisted extraction (IL-MAE) of polyphenolic content from Peperomia pellucida (L) Kunth. Methods: The IL-MAE factors as experimental design parameters, including microwave power, extraction time, ionic liquid concentration, and liquid–solid ratio had been involved. Response surface methodology and Box–Behnken design were used to obtain predictive model (multivariate quadratic regression equation) and optimization of the extraction process. The response surface was analyzed by using the yields of total polyphenolic content as response value. Results: Based on the obtained results the optimum extraction condition, including microwave power of 30% Watts, extraction time of 18.5 min, the ionic liquid concen-tration of 0.79 mol/L, and the liquid–solid ratio of 10.72 mL/g 1-Buthyl-3-methylimidazolium tetrafluoroborate ([bmim]BF4) as a solvent was selected. The regression model was obtained to predicts the yields from Peperomia pellucida:Y = 30.250 – 1.356X1 + 2.655X2 + 2.252X3 – 0.565X4 + 0.990 X1X3 – 8.172 X1X4 – 3.439 X3X4 – 4.178 X12 – 3.210 X32 – 6.786 X42 – 7.290 X12X3 + 5.575 X1X32 – 4.843 X32X4 with R2 = 0.82519. Scale-up confirmation test was obtained the maximum yields of total polyphenolics content with the amount of 31.1725μg GAE/g. Conclusions: The IL-MAE method produced a higher extraction polyphenolic and performed rapidly, easily and efficiently.
文摘Objective: To determinate the recovery of total polyphenolic compounds content,in vitro antioxidant activity and HPLC/ESI/MS characterization of extract from Nephelium lappaceum L.(Mexican rambutan).Methods: The rambutan husk extract was obtained by aqueous extraction and a polyphenolic fraction was recovered using Amberlite XAD-16. The total polyphenolic compounds content was determined by the Folin Ciocalteu and butanol-HCI methods.In vitro antioxidant activity was performed using ABTS and ferric reducing antioxidant power methods.Results: Mexican rambutan husk showed a total polyphenolic content of 582 mg/g and an evident antioxidant activity by ABTS and ferric reducing antioxidant power analysis.The HPLC/ESI/MS assay allowed the identification of 13 compounds, most of which belong to ellagitannins. Geraniin, corilagin and ellagic acid were present in the sample;the mineral composition was also evaluated.Conclusions: Rambutan husk cultivated in Mexico is a promising source for the recovery of added value bioactive compounds with antioxidant activity, which have potential applications as bioactive antioxidant agents for the treatment of diseases.

文摘Objective:To evaluate the antioxidant and radical scavenging activities of Solanum anguivi fruit(SAG)and its possible effect on mitochondrial permeability transition pore as well as mitochondrial membrane potential(△Ψ_m)isolated from rat liver.Methods:Antioxidant activity of SAG was assayed by using 2,2-diphenyl-1-picrylhydrazyl(DPPH),reducing power,iron chelation and ability to inhibit lipid peroxidation in both liver and brain homogenate of rats.Also,the effect of SAG on mitochondrial membrane potential and mitochondrial swelling were determined.Identification and quantification of bioactive polyphenolics was done by HPLC-DAD.Results:SAG exhibited potent and concentration dependent free radical-scavenging activity(IC_(50)/DPPH=275.03±7.8μg/mL).Reductive and iron chelation abilities also increase with increase in SAG concentration.SAG also inhibited peroxidation of cerebral and hepatic lipids subjected to iron oxidative assault.SAG protected against Ca^(2+)(110μmol/L)-induced mitochondrial swelling and maintained the△Ψ_m.HPLC analysis revealed the presence of gallic acid[(17.54±0.04)mg/g],chlorogenic acid(21.90±0.02 mg/g),caffeic acid(16.64±0.01 mg/g).rutin[(14.71±0.03)mg/g]and quercetin[(7.39±0.05)mg/g].Conclusions:These effects could be attributed to the bioactive polyphenolic compounds present in the extract.Our results suggest that SAG extract is a potential source of natural antioxidants that may be used not only in pharmaceutical and food industry but also in the treatment of diseases associated with oxidative stress.

文摘Present work was undertaken to screen some drought tolerant indigenous aromatic rice cultivars(IARCs),commonly cultivated in West Bengal,India,based on their capacity to produce osmolytes,redox-sensitive phenolic acids and flavonoids,as contrivances for redox-regulation under drought stress.Polyethylene glycol induced post imbibitional dehydration stress mediated changes in redox regulatory properties of the germinating seeds of the four IARCs(Jamainadu,Tulaipanji,Sitabhog,Badshabhog),which were assessed in terms of changes in prooxidant accumulation(in-situ localization of reactive oxygen species(ROS)by confocal microscopy,DCFDA(2′,7′-dichlorofluorescin diacetate)oxidation,O2-and H2O2 accumulation),cumulative antioxidative defense(radical scavenging property and total thiol content),ROS scavenging phenolic acids(gallic acid,protocatechuic acid,gentisic acid,para-hydroxy benzoic acid,chlorogenic acid,caffeic acid,syringic acid,salicylic acid,sinapic acid and p-coumaric acid)and flavonoids(catechin,naringin,rutin,quercetin,kaempferol,myricetin and apigenin).The capability of germinating seeds to accumulate osmolytes(like glycinebetaine,proline,soluble carbohydrates and K+ion)and polyphenolic compounds was also correlated with their corresponding redox status and redox biomarkers(conjugated diene,hydroperoxide,thiobarbituric acid reactive substances and free carbonyl content)produced under the same conditions.The results in general showed that accumulation of osmolytes along with the redox-sensitive phenolics and flavonoids conferred the ability to maintain the redox homeostasis under drought stress for the tolerant IARCs(Badshabhog and Tulaipanji).
文摘A mild, cascade type methodology was developed for the synthesis of polyphenolic ethers by the palladium-catalyzed cross coupling of phenols and halo compounds under microwave heating. In most cases, reactions run in neat conditions and in some cases, IPA/water mixture, and 1,4-dioxane were employed as solvents to furnish the products. By applying this new method, we were able to synthesize and purify a good number of polyether compounds with complete spectral data.

文摘Salvia polyphenolic acid for injection is a new generation of Chinese medicine injectable,which is developed by extracting and separating the water-soluble phenolic acid active ingredients of Salvia divinorum by using modern advanced technology as raw material.Pharmacological research shows that salvia polyphenolic acid has anti-inflammatory,anti-oxidative stress,neurotrophic,regenerative and protective effects on ischemic brain injury and can reduce myocardial ischemic injury.In clinical practice,it is mainly used in the treatment of stroke(mild to moderate cerebral infarction)in the recovery period of fetish blood obstruction.In recent years,it has also been gradually extended to treating acute cerebral infarction and respiratory syndrome.Since its launch,the pharmacological effects and clinical applications of Danshen polyphenolic acid for injection are reviewed to provide research clues for further essential trials and reference basis for clinical promotion and application.
文摘Objective: To reach the recovery and identification of antioxidant polyphenolic compounds from Nephelium lappaceum L.(Mexican variety) husk using ultrasound-assisted extraction and liquid chromatography/mass spectrometry as well as the in vitro antioxidant activity. Methods: Rambutan husk extracts were obtained by ultrasound-assisted extraction, mass/volume ratio, water/ethanol percentage and extraction time were evaluated. Once the best extraction condition of polyphenolic compounds was defined, a polyphenolic fraction was recovered using Ambetlite XAD-16. The total content of antioxidant polyphenolic compounds was determined by summation of the total hydrolysable polyphenol and total condensed polyphenol contents. Recovered compounds were identified by FTIR(ATR) spectroscopy and HPLC/ESI/MS. The antioxidant activity was carried out by ABTS, DPPH and lipid oxidation inhibition in vitro methods. Results: In Mexican variety rambutan husk, the total polyphenolic content was 487.67 mg/g, after ultrasound-assisted extraction. According to the HPLC/ESI/MS analysis 12 antioxidant polyphenolic compounds were identified, mostly ellagitannins such as geraniin, corilagin and ellagic acid. The antioxidant activity determined by ABTS, DPPH and lipid oxidation inhibition methods was demonstrated. The main functional groups of the identified compounds were determined by FTIR analysis. Conclusions: It was demonstrated that ultrasound-assisted extraction was effective and allowed the extraction and recovery of antioxidant polyphenolic compounds. Furthermore Mexican variety rambutan husk is an important source for recovering polyphenolic compounds with antioxidant activity, these compounds have potential application for the treatment/prevention of various diseases related to cancer and pathogenic microorganisms.
文摘The occurrence of Metabolic Syndrome (MS) represents an independent risk factor for developing cardiovascular disease states in patients suffering from type 2 diabetes mellitus. Moreover, both the size of LDL particles and liver dysfunction identified as non alcoholic fatty liver disease (NAFLD) represent important biomarkers for the development of cardiometabolic risk in patients with MS. Here we studied the effect of bergamot polyphenolic fraction (BPF) in patients with MS and NAFLD. 107 patients were enrolled at the San Raffaele IRCCS (Rome). All of them showed ultrasonografic evidences of NAFLD and at least three out of five previous identified criteria for the diagnosis of MS. Patients were divided into two groups: one receiving placebo and the second receiving BPF 650 mg twice a day for 120 consecutive days. In the group receiving BPF 650 mg twice a day, a significant reduction of fasting plasma glucose, serum LDL cholesterol and triglycerides alongside with an increase of HDL cholesterol was found. This effect was accompanied by significant reduction of both ultrasonographic and metabolic biomarkers of NAFLD. Moreover, a significant reduction of small dense LDL particles, as detected via proton NMR Spectroscopy, was found after BPF treatment. In conclusion, our data confirm the beneficial effect of bergamot-extract in patients with MS an effect highlighted by significant reduction of small dense LDL particles and by improvement of NAFLD biomarkers. This suggests a potential preventive role of bergamot derivatives in reducing cardiometabolic risk.
文摘Response surface methodology (RSM) was applied to provide predictive models for optimisation of extraction of selected polyphenolic compounds from cider apple pomace under aqueous acetone. The design of experiment (DoE) was conducted to evaluate the influence of acetone concentration % (v/v), solid-to solvent ratio % (w/v), temperature (˚C) and extraction time (min) and their interaction on phenolic contents, using the Central Composite Rotatable Design (CCRD). The experimental data were analysed to fit statistical models for recovery of phenolic compounds. The selected models were significant (P 0.05), except for Chlorogenic acid and Quercetin 3-glucoside which had significant lack of fits (P R2 > 0.9000 and adjusted? ?reasonable agrees with predicted . Coefficient of variation < 5% for each determination at the 95% confidence interval. These models could be relied upon to achieve optimal concentrations of polyphenolic compounds for applications in nutraceutical, pharmaceutical and cosmetic industries.
文摘Polyphenolic compounds with relatively high antioxidant activity obtained from subcritical water extraction of apple pomace were assessed for encapsulation by spray drying technique, making use of polymeric substances co-extracted with the polyphenolic compounds. Comparative assessments were carried out of the directly encapsulated subcritical water extract (SWE) products with particles formed when encapsulated with the addition of hydroxyl propyl-β-Cyclodextrin (SWE + HPβ-CD). The powders were characterized for their physico-chemical properties such as, moisture content, density, particle size, hygroscopicity to assess their suitability within cosmetic formulations. The SWE and SWE + HPβ-CD encapsulated products resulted in different physical properties. Although the particle size was less than 4 μm for both products, the direct encapsulation (SWE) was highly hygroscopic and this property was significantly reduced with addition of HPβ-Cyclodextrin (SWE + HPβ-CD). Scanning electron microscopy (SEM) and Fourier Transform Infrared (FT-IR) spectroscopic were em-ployed to analyse the micronised powders to support evidence of encapsulation. Both techniques revealed the interaction between compounds in extract and the carrier HPβ-Cyclodextrin suggesting successful encapsulation. The effect of storage conditions on retention of antioxidant activity of the subcritical water extract was evaluated within 35 days for extracts with and without the carrier HPβ-Cyclodextrin. Hydroxyl propyl-β-Cyclodextrin offered protection against degradation of antioxidant compounds thereby potentially extending the shelf-life and making the encapsulated powder suitable for incorporation in cosmetic and pharmaceutical applications.

文摘Strawberries are rich in polyphenols which impart health benefits when metabolized by the gut microbiome,including anti-inflammatory,neuroprotective,and antiproliferative effects.In addition,polyphenolic anthocyanins contribute to the attractive color of strawberry fruits.However,the genetic basis of polyphenol biosynthesis has not been extensively studied in strawberry.In this investigation,ripe fruits from three cultivated strawberry populations were characterized for polyphenol content using HPLC-DAD-MSn and genotyped using the iStraw35k array.GWAS and QTL analyses identified genetic loci controlling polyphenol biosynthesis.QTL were identified on four chromosomes for pelargonidin-3-O-malonylglucoside,pelargonidin-3-O-acetylglucoside,cinnamoyl glucose,and ellagic acid deoxyhexoside biosynthesis.Presence/absence of ellagic acid deoxyhexoside and pelargonidin-3-O-malonylglucoside was found to be under the control of major gene loci on LG1X2 and LG6b,respectively,on the F.×ananassa linkage maps.Interrogation of gene predictions in the F.vesca reference genome sequence identified a single candidate gene for ellagic acid deoxyhexoside biosynthesis,while seven malonyltransferase genes were identified as candidates for pelargonidin-3-O-malonylglucoside biosynthesis.Homologous malonyltransferase genes were identified in the F.×ananassa‘Camarosa’genome sequence but the candidate for ellagic acid deoxyhexoside biosynthesis was absent from the‘Camarosa’sequence.This study demonstrated that polyphenol biosynthesis in strawberry is,in some cases,under simple genetic control,supporting previous observations of the presence or absence of these compounds in strawberry fruits.It has also shed light on the mechanisms controlling polyphenol biosynthesis and enhanced the knowledge of these biosynthesis pathways in strawberry.The above findings will facilitate breeding for strawberries enriched in compounds with beneficial health effects.
文摘This study was carried out to characterize total residual catechins and their fractions, polyphenolic contents and antioxidant activities of black teas enriched with high levels of theaflavin-3,3’-digallate. The made teas were processed from eleven selected cultivars. A comparative study was carried out between the processed teas and those from commercially grown Kenyan cultivars in relation to the above chemical parameters. A correlation matrix analysis was also conducted to find out whether a relationship existed between the antioxidant activities and the said chemical parameters. The total residual catechins were found to range between 3.10% and 8.08%. The total polyphenol levels varied between 19.00% and 28.90%, while the antioxidant activities of the teas ranged from 82.70% to 91.70%. There was a significant p < 0.001 correlation between the antioxidant activity and total polyphenols (r = 0.8948). There was also a high correlation p < 0.001 between the antioxidant activity and total catechins (r = 0.8878). Out of the four catechin fractions, the antioxidant activity correlated most with EGCG (r = 0.8774). The total polyphenolic contents and antioxidant activities for most of the cultivars were comparable to those of the green tea reference standard. From the figures obtained, it can be concluded that the most of the newly developed black teas of the selected cultivars have higher quality and enhanced antioxidant activities and that they can be recommended for commercial production.

文摘Objective:To study the antioxidant properties of crude extract of different Asteraceae plants.Methods:The antioxidant properties of six extracts were evaluated using different antioxidant tests,including free radical scavenging,reducing power,metal chelation,superoxide anion radical scavenging,total antioxidant capacity and inhibition of lipid peroxidation activities.Results:Picris cyanocarpa(P.cyanocarpa)and Anthemis deserti(A.deserti)had powerful antioxidant properties as radical scavenger,reducing agent and superoxide anion radical scavenger while Achillia fragrantissima(A.fragrantissima)and Artemissia monosperma(A.monosperma)were the most efficient as ion chelator(100%at 100,200 and 400μg/mL)A.fragrantissima and Rhanlarium appoposum(R.appoposum.)showed 100%inhibition on peroxidation of linoleic acid emulsion at 200 and 400μg/mL,while butylatedhydroxy toluene and ascorinc acid showed 100and 95%inhibition percentage at 400μg/mL,respectively.Those various antioxidant activities were compared to standard antioxidants such as butylated hydroxyl toluene and ascorbic acid.Conclusions:In most tests P.cyanocarpa and A.deserti had powerful antioxidant properties as radical scavenger,reducing agent and superoxide anion radical scavenger.

文摘Objective:To evaluate the effects of phenolic acids(caffeic,ferulic,and coumaric acids)and flavones(luteolin and apigenin)on the proliferation and melanogenesis in murine melanoma B16-F10 cells.Methods:Cell proliferation was determined after 24 and 48 hours of incubation using MTT assay.The effects of these tested compounds on cell cycle progression were analyzed by flow cytometry.Moreover,the melanin content and tyrosinase activity were measured spectrophotometrically at 475 nm.Results:Luteolin and apigenin exhibited significant anti-proliferative activity against B16-F10 cells,while caffeic,ferulic,and coumaric acids induced slight inhibition after 24 and 48 hours of incubation.The tested compounds disturbed cell cycle progression of B16-F10,by a subsequent decrease in G1 and arrested cycle progression in either G1/S or G2/M phase.Furthermore,apigenin provoked an increase in melanin content of B16-F10 cells.In contrast,luteolin,caffeic,ferulic and coumaric acids induced a decrease in melanin content of B16-F10 cells by inhibiting tyrosinase activity.Conclusions:These active polyphenols may be used as skin whitening agents or natural tanning agents to treat skin pigmentation disorders.
文摘Rubuphenol (1), a new polyphenolic compound, was isolated together with the known ellagic acid (2) as new cell cycle inhibitors from Rubus aleaefolius Poir. through a bioassay-guided separation procedure and the structure of 1 was elucidated by spectroscopic method. Compounds 1 and 2 inhibited the cell cycle of tsFT210 cells at the G0/G1 phase respectively with the MIC values of 14.6 mM and 10.3 mM.
文摘Polyphenols are a large family of more than 10,000 naturally occurring compounds,which exert countless pharmacological,biological and physiological benefits for human health including several chronic diseases such as cancer,diabetes,cardio-vascular,and neurological diseases.Their role in traditional medicine,such as the use of a wide range of remedial herbs(thyme,oregano,rosemary,sage,mint,basil),has been well and long known for treating common respiratory problems and cold infections.This review reports on the most highlighted polyphenolic compounds present in up to date literature and their specific antiviral perceptive properties that might enhance the body immunity facing COVID-19,and other viral infec-tious diseases.In fact,several studies and clinical trials increasingly proved the role of polyphenols in controlling numer-ous human pathogens including SARS and MERS,which are quite similar to COVID-19 through the enhancement of host immune response against viral infections by different biological mechanisms.Thus,polyphenols ought to be considered as a potential and valuable source for designing new drugs that could be used effectively in the combat against COVID‐19 and other rigorous diseases.
文摘The investigation aimed at exploring whether 1) high contents of natural polyphenols from the diet can induce pigment accumulation in lymph nodes (LNs);2) if so, whether polyphenolic compounds and derivates can be used as biological markers;3) and whether a lymph node from a specific anatomical region can be univocally identified, so as to be con sidered as a sentinel for the identification of the dietary origin of pigments. A paired match approach was used to switch 20 pigs (range of initial body weight, BW: 113 - 121 kg) to two experimental diets, for four weeks: ten pigs (pair housed) were fed with an experimental acorn based diet (acorns: 50% in the diet, as fed;total polyphenols, 78.1 g TP/Kg DM in the diet;tannic acid equivalent, 25.8 g TAE/kg DM);the remainder ten, received a pelleted complete diet for finishers (0% acorns in the diet). Daily feed intake in the last two weeks of the experimental feeding was recorded per pair of pigs in both groups of animals, showing an average intake of 610 mg TAE/kg BW/d. At an average final BW of between 127 to 137 kg, all pigs were slaughtered and LNs from different anatomical regions of the carcass were removed and analysed. At gross inspection, LNs from both groups displayed different grades of intensity and diffusion of pigmentation: a partial and incidental pigmentation was randomly detected in renal or sub-iliac LNs in the control group;a constant and uniform pigmentation of LNs was observed in acorns fed pigs: a dark brown staining diffused to the whole LN associated with a brownish colour of the muscles was found systematically. At light microscope intracytoplasmic granules were found in macrophages and dendritic cells from both groups, but, at confocal laser analysis, an intense auto-fluorescence was observed in medial-iliac LNs from the carcasses of acorn-fed pigs (green emission). However, intracellular sources of blue and green fluorescence at different wavelengths, likely due to tryptophan, indoleamine and derivates were also found in medial-iliac and inguinal LNs from the control group. A dietary origin was attributed to the different discoloration of LNs between the carcasses of the two groups: such acquired pigmentation is relevant in the left sub-iliac LN, but the confocal laser microscopic test to elicit auto-fluorescence of polyphenolic compounds (biological markers) displayed a 76.9% specificity, despite a 100% of sensitivity for the univocal identification of the carcass from acorn-fed pigs. Cranial sternal LNs resulted to suit the sentinel role in the distinction of carcass from acorns fed pigs at confocal laser microscopic analysis.

文摘A rapid method was presented for the determination of 19 polyphenols in tea by ultra-high performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF MS).Tea samples were extracted by 50%(V/V)ethanol,then separated by Waters Acquity BEH C18 column using a binary solvent system composed of acetonitrile and water(0.1%formic acid)by gradient elution.The analytes were determined by Q-TOF MS in TOF MS and information dependent acquisition(IDA)-MS/MS mode.The results showed that mass accuracy error of the 19 polyphenols were lower than 5.0×10^(-6),good linear relationship was got in range of 0.2–500μg/L and correlation coefficient was higher than 0.9990.The LOD was in the range of 0.002–0.100 mg/kg and the LOQ was in the range of 0.004–0.200 mg/kg.Recovery of the method was in range of 78.4%–109.2%with spike levels of 0.004–2.000 mg/kg,relative standard deviations were lower than 10%.The method was simple,rapid and accurate.It could be used for the rapid screening and quantitative analysis of 19 polyphenols in tea.
