[Objectives]To study and optimize the process conditions of enzymatic hydrolysis technology for extracting polysaccharides from Pseudostellaria heterophylla fibrous roots and its application in workshop pilot tests.[M...[Objectives]To study and optimize the process conditions of enzymatic hydrolysis technology for extracting polysaccharides from Pseudostellaria heterophylla fibrous roots and its application in workshop pilot tests.[Methods]P.heterophylla fibrous roots were taken as the matrix material,and Box Behnken design was used to analyze the extraction time,composite enzyme addition amount,and liquid-solid ratio for response surface optimization experiments,and then applied to the pilot extraction of P.heterophylla fibrous roots.[Results]Response surface analysis showed that all factors had a significant impact on the experimental indicators.The optimal extraction process conditions for polysaccharides from P.heterophylla fibrous roots were extraction time of 2.7 h,compound enzyme addition of 2.5%,and liquid-solid ratio of 32.The yield of polysaccharides from P.heterophylla fibrous roots was 4.83%.The water extraction process of P.heterophylla fibrous roots extraction pilot was used as the control group for response surface optimization of the pilot experiment.The optimization results showed that the extraction time was 3 h,the amount of composite enzyme added was 2.5%,and the liquid-solid ratio was 28.The polysaccharide yield was 4.75%,an increase of 4.63%compared to the control group.[Conclusions]This paper could provide feasibility for the innovation of enzy-matic hydrolysis technology for P.heterophylla fibrous roots and its workshop pilot practice application,as well as a reference for the industrial application of its medicinal resources.展开更多
[Objectives]Polysaccharide was extracted from Lactarius deliciosus by ultrasonic-assisted method to improve polysaccharide yield.[Methods]Five variables including extraction temperature,extraction time,ultrasonic powe...[Objectives]Polysaccharide was extracted from Lactarius deliciosus by ultrasonic-assisted method to improve polysaccharide yield.[Methods]Five variables including extraction temperature,extraction time,ultrasonic power,ultrasonic time and material-to-liquid ratio were selected for single factor experiments.The extraction process of L.deliciosus polysaccharide was optimized by response surface analysis,and its antioxidant activity was evaluated by measuring its total reduction ability and DPPH free radical scavenging capacity.[Results]The optimal extraction conditions were determined as follows:material-to-liquid ratio 1:35 g:ml,ultrasonic power 462 W,ultrasonic time 10 min,extraction time 110 min and extraction temperature 90℃.Under these conditions,the extraction rate of polysaccharide was(10.83±0.03)%.The antioxidant test results showed that when the mass concentration of L.deliciosus polysaccharide was 0.5 mg/ml,its absorbance and DPPH free radical scavenging rate reached their maximum values,which were 3.274%and 41.27%,respectively.The L.deliciosus polysaccharide had good antioxidant properties.[Conclusions]This study provides a theoretical basis for further development and utilization of L.deliciosus polysaccharide in the future.展开更多
Objective:To establish an optimized aqueous extraction process for polysaccharides from Physalis alkekengi L.peel and to preliminarily explore its in vitro anti-inflammatory activity against colorectal cancer SW620 ce...Objective:To establish an optimized aqueous extraction process for polysaccharides from Physalis alkekengi L.peel and to preliminarily explore its in vitro anti-inflammatory activity against colorectal cancer SW620 cells.Methods:A single-factor test combined with orthogonal test analysis was used to evaluate the effects of the material-to-liquid ratio,extraction temperature,and extraction time on the yield of polysaccharides from Physalis alkekengi L.peel.The antioxidant activity of the polysaccharides was assessed by analyzing their free radical scavenging ability in vitro,and the anti-inflammatory effect was evaluated using SW620 cells.Results:The optimal extraction conditions were a material-to-liquid ratio of m(g):V(mL)=1:30,an extraction temperature of 100℃,and an extraction time of 40 minutes,with a predicted polysaccharide yield of 25.7%.The polysaccharides from Physalis peruviana peel effectively scavenged DPPH,superoxide anion,and hydroxyl radicals.After treatment with Physalis peruviana polysaccharides,the levels of IL-1β,IL-18,and TNF-αin the cell culture medium were significantly reduced,and the phosphorylation level of P65 protein in SW620 cells was decreased.Conclusion:This extraction method is stable and reliable,and the prepared Physalis alkekengi L.polysaccharides exhibit significant in vitro antioxidant and anti-inflammatory activities.This study provides a theoretical basis for developing drugs for the prevention and treatment of colorectal cancer.展开更多

Polysaccharides from T. amurensis flowers were isolated and determined by spectrophotometry. Results show that the optimal ultrasonic temperature was 75℃ and ex tracting time 52 min. The content of polysaccharides in T. amurensis flowers measured by anthrone--H2SO4 colorimetry under 580 nm, was 9.74% with 0.47% of relative standard deviation (RSD, n=3). This study demonstrated that ultrasonic extraction method was simple, and the color of the treated samples was stable in 4 h. The average recovery value for the polysaccharides measured was 99.48%±1.01%, with 0.112% of RSD (n=3).展开更多

[Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. [Result] The results of response surface analysis showed that the optimal extraction process was solid-liquid ratio of 1:17, extraction temperature of 80 ℃ and extraction duration of 78.5 min, and the Pu-erh tea polysaccharide yield was 12.72%. [Conclu- sion] Using response surface methodology (RSM) is feasible for the optimization of Pu-erh tea polysaccharide extraction process, and the tea polysaccharide yield increased significantly.展开更多

[Method] The polysaccha- rides were directly extracted from the fresh algae mat or dry powder of Spirulina in boiling water. The contents and quality of the extracted polysaccharides were measured. [Result] On average, 236.06 g of polysaccharide was extracted from 25 kg of fresh Spirulina mat, with a yield rate of 0.94%, while 191.95 g of polysac- charide was extracted from 2.5 kg of dry powder of Spirulina with a yield rate of 0.77%. The polysaccharide content in the extract of fresh Spirulina mat was 12.56% (according to glucose content), while that of the dry powder was 12.38% (according to glucose content); the glucose was produced during the hydrolysis of Spirulina polysaccharide. [Conclusion] Extraction polysaccharide from Spirulina with boiling wa- ter greatly reduces the use of ethanol, and the possibility of pollution from the ex- ogenous non-food chemical reagents, so that the extracted polysaccharides can be used as food materials. This method makes it possible to establish the production line for Spirulina polysaccharide.展开更多

Background:Ampelopsis grossedentata,vine tea,which is the tea alternative beverages in China.In vine tea processing,a large amount of broken tea is produced,which has low commercial value.Methods:This study investigat...Background:Ampelopsis grossedentata,vine tea,which is the tea alternative beverages in China.In vine tea processing,a large amount of broken tea is produced,which has low commercial value.Methods:This study investigates the influence of different extraction methods(room temperature water extraction,boiling water extraction,ultrasonic-assisted room temperature water extraction,and ultrasonic-assisted boiling water extraction,referred to as room temperature water extraction(RE),boiling water extraction(BE),ultrasonic assistance at room temperature water extraction(URE),and ultrasonic assistance in boiling water extraction(UBE))on the yield,dihydromyricetin(DMY)content,free amino acid composition,volatile aroma components,and antioxidant properties of vine tea extracts.Results:A notable influence of extraction temperature on the yield of vine tea extracts(P<0.05),with BE yielding the highest at 43.13±0.26%,higher than that of RE(34.29±0.81%).Ultrasound-assisted extraction significantly increased the DMY content of the extracts(P<0.05),whereas DMY content in the RE extracts was 59.94±1.70%,that of URE reached 66.14±2.78%.Analysis revealed 17 amino acids,with L-serine and aspartic acid being the most abundant in the extracts,nevertheless ultrasound-assisted extraction reduced total free amino acid content.Gas chromatography-mass spectrometry analysis demonstrated an increase in the diversity and quantity of compounds in the vine tea water extracts obtained through ultrasonic-assisted extraction.Specifically,69 and 68 volatile compounds were found in URE and UBE extracts,which were higher than the number found in RE and BE extracts.In vitro,antioxidant activity assessments revealed varying antioxidant capacities among different extraction methods,with RE exhibiting the highest DPPH scavenging rate,URE leading in ABTS•+free radical scavenging,and BE demonstrating superior ferric ion reducing antioxidant activity.Conclusion:The findings suggest that extraction methods significantly influence the chemical composition and antioxidant properties of vine tea extracts.Ultrasonic-assisted extraction proved instrumental in elevating the DMY content in vine tea extracts,thereby enriching its flavor profile while maintaining its antioxidant properties.展开更多

AIM:To explore the effect and mechanism of Lycium barbarum polysaccharide(LBP)inhibiting retinal neovascularization.METHODS:In vitro tests were performed on human retinal microvascular endothelial cells(HRECs)from thr...AIM:To explore the effect and mechanism of Lycium barbarum polysaccharide(LBP)inhibiting retinal neovascularization.METHODS:In vitro tests were performed on human retinal microvascular endothelial cells(HRECs)from three groups,including control group(normal oxygen),hypoxic group(hypoxia at 37℃,1%O_(2),5%CO_(2),and 94%N2),and LBP group(hypoxic group with LBP 100μg/mL).In vivo experiments,C57 mice were divided into three groups:control group(normal rearing group),the oxygen-induced ischemic retinopathy(OIR)group,and the OIR with 50 mg/kg LBP group.Retinal neovascularization was observed by fluorescein angiography and quantified.Retinal thickness was evaluated by Hematoxylin and eosin(HE)stain.The expression of epidermal growth factor receptor(EGFR),phosphatidylinositol 3-kinase(PI3K),mammalian target of rapamycin(mTOR),phosphorylated mammalian target of rapamycin(p-mTOR),protein kinase B(AKT),phosphorylated protein kinase B(p-AKT),interleukin-1β(IL-1β),inducible nitric oxide synthase(iNOS),and tumor necrosis factor-α(TNF-α)in each group were analyzed by Western blot.IL-1βlevel in retina was analyzed using immunohistochemical staining.RESULTS:The increased area of neovascular clusters in OIR mice was significantly decreased by LBP.Retinal thickness of OIR mice was significantly thinner compared with normal oxygenated mice and was increased in LBP group.Compared with those in the hypoxic groups,Western blotting of HRECs and retinal tissues revealed that the expression of EGFR,PI3K,p-mTOR,p-AKT,IL-1β,iNOS,and TNF-αdecreased in the LBP group but was still greater than that in control group.Moreover,IL-1βwas reduced in retinal sections treated with LBP.In the scratch test,the cell migration of the hypoxic group was significantly greater than that of the control group,while LBP treatment attenuated this increase in migration.CONCLUSION:LBP reduces retinal neovascularization and inflammation in vivo and inhibits the migration of HRECs in vitro by regulating the EGFR/PI3K/Akt/mTOR signaling pathway.展开更多
AIM:To evaluate the effect of femtosecond laser small incision lenticule extraction(SMILE)on the binocular visual function in myopic patients with glasses-free threedimensional(3D)technique.METHODS:Totally 50 myopic p...AIM:To evaluate the effect of femtosecond laser small incision lenticule extraction(SMILE)on the binocular visual function in myopic patients with glasses-free threedimensional(3D)technique.METHODS:Totally 50 myopic patients(39 females and 11 males)with SMILE were enrolled in this prospective study.The glasses-free 3D technique was used to evaluate the binocular visual function in these subjects including static stereopsis,dynamic stereopsis,foveal suppression,and binocular balance point of signal to noise ratio(s/n ratio).All subjects received measurements in 1d before operation,and 1d,1wk,and 1mo postoperatively.RESULTS:Both static and dynamic stereopsis showed no significant difference after SMILE.The foveal suppression improved significantly 1wk and 1mo after SMILE(P=0.005 and P=0.007 respectively).The binocular balance point of signal to noise ratio showed a significant improvement 1d,1wk and 1mo after SMILE for both eyes(P<0.001 for each eye respectively).CONCLUSION:Glasses-free 3D technique can be used to evaluate the effect of SMILE on the binocular visual function in myopic patients perceptively,and SMILE can improve both foveal suppression and binocular imbalance in these patients.展开更多

This research optimized the structure of lithium extraction solar ponds to enhance the crystallization rate and yield of Li_(2)CO_(3).Using the response surface methodology in Design-Expert 10.0.3,the authors conducte...This research optimized the structure of lithium extraction solar ponds to enhance the crystallization rate and yield of Li_(2)CO_(3).Using the response surface methodology in Design-Expert 10.0.3,the authors conducted experiments to investigate the influence of four factors related to solar pond structure on the crystallization of Li_(2)CO_(3) and their pairwise interactions.Computational Fluid Dynamics(CFD)simulations of the flow field within the solar pond were performed using COMSOL Multiphysics software to compare temperature distributions before and after optimization.The results indicate that the optimal structure for lithium extraction from the Zabuye Salt Lake solar ponds includes UCZ(Upper Convective Zone)thickness of 53.63 cm,an LCZ(Lower Convective Zone)direct heating temperature of 57.39℃,a CO32−concentration of 32.21 g/L,and an added soda ash concentration of 6.52 g/L.Following this optimized pathway,the Li_(2)CO_(3) precipitation increased by 7.34% compared to the initial solar pond process,with a 33.33% improvement in lithium carbonate crystallization rate.This study demonstrates the feasibility of optimizing lithium extraction solar pond structures,offering a new approach for constructing such ponds in salt lakes.It provides valuable guidance for the efficient extraction of lithium resources from carbonate-type salt lake brines.展开更多

BACKGROUND Bletilla striata polysaccharides(BSP)have antioxidant,immune regulation,and anti-fibrotic activities.However,the therapeutic effect and mechanisms underlying the action of BSP in metabolic dysfunction-assoc...BACKGROUND Bletilla striata polysaccharides(BSP)have antioxidant,immune regulation,and anti-fibrotic activities.However,the therapeutic effect and mechanisms underlying the action of BSP in metabolic dysfunction-associated steatotic liver disease(MASLD)have not been fully understood.AIMTo investigate the therapeutic effects and mechanisms of BSP on MASLD by centering on the hepatocyte nuclearfactor kappa B p65(RelA)/hepatocyte nuclear factor-1 alpha(HNF1α)signaling.METHODSA mouse model of MASLD was induced by feeding with a high-fat-diet(HFD)and a hepatocyte model of steatosiswas induced by treatment with sodium oleate(SO)and sodium palmitate(SP).The therapeutic effects of BSP onMASLD were examined in vivo and in vitro.The mechanisms underlying the action of BSP were analyzed for theireffect on lipid metabolism disorder,endoplasmic reticulum(ER)stress,and the RelA/HNF1αsignaling.RESULTSHFD feeding reduced hepatocyte RelA and HNF1αexpression,induced ER stress,lipid metabolism disorder,andnecroptosis in mice,which were significantly mitigated by treatment with BSP.Furthermore,treatment with BSP orBSP-containing conditional rat serum significantly attenuated the sodium oleate/sodium palmitate(SO/SP)-induced hepatocyte steatosis by decreasing lipid accumulation,and lipid peroxidation,and enhancing theexpression of RelA,and HNF1α.The therapeutic effects of BSP on MASLD were partially abrogated by RELAsilencing in mice and RELA knockout in hepatocytes.RELA silencing or knockout significantly down-regulatedHNF1αexpression,and remodeled ER stress and oxidative stress responses during hepatic steatosis.CONCLUSIONTreatment with BSP ameliorates MASLD,associated with enhancing the RelA/HNF1αsignaling,remodeling ERstress and oxidative stress responses in hepatocytes.展开更多

This study optimized the alkali-extraction conditions and antioxidant activities of soluble polysaccharides from the body wall of Sipunculus nudus. The effects of solid-liquid ratio, extraction duration, extraction temperature, and alkali concentration on the yield of S. nudus polysaccharides(SNP) were examined, according to which the optimal combination of extraction parameters was obtained by an orthogonal test. The relative influencing importance of different extraction parameters on the yield of SNP followed the order as solid-liquid ratio > extraction temperature > alkali concentration > extraction duration. The highest extraction yield, 1.98%, was achieved under an optimal extraction condition: temperature 60℃, solid-liquid ratio 1:6 g mL^(-1), duration 5 h, and alkali(Na OH) mass fraction 6%. The in vitro antioxidant activities examination showed that extracted SNP under this optimized condition had strong power in reducing certain hydroxyl and superoxide radical scavenging abilities. The promising results showed that extracted SNP could be a potent natural antioxidant.展开更多
With the development of the new energy industry and the depletion of nickel sulfide ore resources,laterite nickel ore has become the main source of primary nickel,and nickel for power batteries has become a new growth...With the development of the new energy industry and the depletion of nickel sulfide ore resources,laterite nickel ore has become the main source of primary nickel,and nickel for power batteries has become a new growth point in consumption.This paper systematically summarizes the processes,parameters,products,recovery rates,environmental indicators,costs,advantages,disadvantages and the latest research progress of mainstream nickel extraction processes from laterite nickel ore.It also provides a comparative analysis of the environmental impact and economic efficiency of different nickel extraction processes.It is found that the current nickel extraction processes from laterite nickel ore globally for commercial production mainly include the RKEF process for producing ferronickel and the HPAL process for producing intermediate products.The former accounts for about 80%of laterite nickel ore production.Compared to each other,the investment cost per ton of nickel metal production capacity for the RKEF is about 43000$,with an operational cost of about 16000$per ton of nickel metal and a total nickel recovery rate of 77%–90%.Its products are mainly used in stainless steels.For the HPAL process,the investment cost per ton of nickel metal production capacity is about 56000$,with an operational cost of about 15000$per ton of nickel metal and a total nickel recovery rate of 83%–90%.Its products are mainly used in power batteries.The significant differences between the two lies in energy consumption and carbon emissions,with the RKEF being 2.18 and 2.37 times that of the HPAL,respectively.Although the use of clean energy can greatly reduce the operational cost and environmental impact of RKEF,if RKEF is converted to producing high Ni matte,its economic and environmental performance still cannot match that of the HPAL and oxygen-enriched side-blown processes.Therefore,it can be inferred that with the increasing demand for nickel in power batteries,HPAL and oxygen-enriched side blowing processes will play a greater role in laterite nickel extraction.展开更多

文摘Morchella septimelata polysaccharide(MSP)possesses multiple biological activities.In this study,double enzyme method was used to extract polysaccharides from Morchella septimelata.The basis of single factor experiment and response surface design was used to optimize the extraction process of polysaccharides.The optimal extraction conditions of polysaccharide were as follows:extraction time 2 h,extraction temperature 40.5℃,solid-liquid ratio 1:15(w/v),and the amount of double enzyme(cellulase:β-glucanase,mass ratio 1:2)2.0%.Under these conditions,the extraction yield of polysaccharide reached(14.86±0.12)%.

文摘[Objectives]To study and optimize the process conditions of enzymatic hydrolysis technology for extracting polysaccharides from Pseudostellaria heterophylla fibrous roots and its application in workshop pilot tests.[Methods]P.heterophylla fibrous roots were taken as the matrix material,and Box Behnken design was used to analyze the extraction time,composite enzyme addition amount,and liquid-solid ratio for response surface optimization experiments,and then applied to the pilot extraction of P.heterophylla fibrous roots.[Results]Response surface analysis showed that all factors had a significant impact on the experimental indicators.The optimal extraction process conditions for polysaccharides from P.heterophylla fibrous roots were extraction time of 2.7 h,compound enzyme addition of 2.5%,and liquid-solid ratio of 32.The yield of polysaccharides from P.heterophylla fibrous roots was 4.83%.The water extraction process of P.heterophylla fibrous roots extraction pilot was used as the control group for response surface optimization of the pilot experiment.The optimization results showed that the extraction time was 3 h,the amount of composite enzyme added was 2.5%,and the liquid-solid ratio was 28.The polysaccharide yield was 4.75%,an increase of 4.63%compared to the control group.[Conclusions]This paper could provide feasibility for the innovation of enzy-matic hydrolysis technology for P.heterophylla fibrous roots and its workshop pilot practice application,as well as a reference for the industrial application of its medicinal resources.

文摘[Objectives]Polysaccharide was extracted from Lactarius deliciosus by ultrasonic-assisted method to improve polysaccharide yield.[Methods]Five variables including extraction temperature,extraction time,ultrasonic power,ultrasonic time and material-to-liquid ratio were selected for single factor experiments.The extraction process of L.deliciosus polysaccharide was optimized by response surface analysis,and its antioxidant activity was evaluated by measuring its total reduction ability and DPPH free radical scavenging capacity.[Results]The optimal extraction conditions were determined as follows:material-to-liquid ratio 1:35 g:ml,ultrasonic power 462 W,ultrasonic time 10 min,extraction time 110 min and extraction temperature 90℃.Under these conditions,the extraction rate of polysaccharide was(10.83±0.03)%.The antioxidant test results showed that when the mass concentration of L.deliciosus polysaccharide was 0.5 mg/ml,its absorbance and DPPH free radical scavenging rate reached their maximum values,which were 3.274%and 41.27%,respectively.The L.deliciosus polysaccharide had good antioxidant properties.[Conclusions]This study provides a theoretical basis for further development and utilization of L.deliciosus polysaccharide in the future.
文摘Objective:To establish an optimized aqueous extraction process for polysaccharides from Physalis alkekengi L.peel and to preliminarily explore its in vitro anti-inflammatory activity against colorectal cancer SW620 cells.Methods:A single-factor test combined with orthogonal test analysis was used to evaluate the effects of the material-to-liquid ratio,extraction temperature,and extraction time on the yield of polysaccharides from Physalis alkekengi L.peel.The antioxidant activity of the polysaccharides was assessed by analyzing their free radical scavenging ability in vitro,and the anti-inflammatory effect was evaluated using SW620 cells.Results:The optimal extraction conditions were a material-to-liquid ratio of m(g):V(mL)=1:30,an extraction temperature of 100℃,and an extraction time of 40 minutes,with a predicted polysaccharide yield of 25.7%.The polysaccharides from Physalis peruviana peel effectively scavenged DPPH,superoxide anion,and hydroxyl radicals.After treatment with Physalis peruviana polysaccharides,the levels of IL-1β,IL-18,and TNF-αin the cell culture medium were significantly reduced,and the phosphorylation level of P65 protein in SW620 cells was decreased.Conclusion:This extraction method is stable and reliable,and the prepared Physalis alkekengi L.polysaccharides exhibit significant in vitro antioxidant and anti-inflammatory activities.This study provides a theoretical basis for developing drugs for the prevention and treatment of colorectal cancer.

文摘Seven extracting temperatures (25, 35, 45, 55, 65, 75, 85℃) and extracting time (30, 40, 50, 60, 70, 80, 90 min) were designed for selecting the optimal extracting time and extracting temperature for ultrasonic extraction of T. amurensis flowers. Polysaccharides from T. amurensis flowers were isolated and determined by spectrophotometry. Results show that the optimal ultrasonic temperature was 75℃ and ex tracting time 52 min. The content of polysaccharides in T. amurensis flowers measured by anthrone--H2SO4 colorimetry under 580 nm, was 9.74% with 0.47% of relative standard deviation (RSD, n=3). This study demonstrated that ultrasonic extraction method was simple, and the color of the treated samples was stable in 4 h. The average recovery value for the polysaccharides measured was 99.48%±1.01%, with 0.112% of RSD (n=3).

文摘[Objective] This paper aims to study a new method of extracting pumpkin polysaccharide from pumpkin. Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. [Conclusion] This study provided basic data for research and application of Pumpkin polysaccharide.

文摘[Objective] This study aimed to optimize the extraction process parameters of Pu-erh tea polysaccharide. [Method] Single-factor experiment was carried out to analyze the influences of three main factors, including extraction temperature, ex- traction duration and solid-liquid ratio, on the extraction yield of tea polysaccharide. Box-Behnken central composite design and response surface methodology were adopted to determine the optimal extraction process of Pu-erh tea polysaccharide. [Result] The results of response surface analysis showed that the optimal extraction process was solid-liquid ratio of 1:17, extraction temperature of 80 ℃ and extraction duration of 78.5 min, and the Pu-erh tea polysaccharide yield was 12.72%. [Conclu- sion] Using response surface methodology (RSM) is feasible for the optimization of Pu-erh tea polysaccharide extraction process, and the tea polysaccharide yield increased significantly.

文摘In this study,an orthogonal experiment was performed to explore the optimal extraction technique of water-soluble polysaccharides from pumpkin.Moreover,the polysaccharides' capabilities to scavenge ·OH and DPPH· and their gross reducing power was measured with Vc and BHT as references.In the orthogonal experiment,various factors including temperature,extraction time,solid-liquid ratio,were optimized.The results showed that the polysaccharides extracted from pumpkin are endowed with strong in vitro antioxidant properties.Our study provided references for the extraction of pumpkin polysaccharides and for their use as antioxidants.
文摘[Objective] The aim of this study was to develop a simple and effective method for the polysaccharide extraction from Spirulina. [Method] The polysaccha- rides were directly extracted from the fresh algae mat or dry powder of Spirulina in boiling water. The contents and quality of the extracted polysaccharides were measured. [Result] On average, 236.06 g of polysaccharide was extracted from 25 kg of fresh Spirulina mat, with a yield rate of 0.94%, while 191.95 g of polysac- charide was extracted from 2.5 kg of dry powder of Spirulina with a yield rate of 0.77%. The polysaccharide content in the extract of fresh Spirulina mat was 12.56% (according to glucose content), while that of the dry powder was 12.38% (according to glucose content); the glucose was produced during the hydrolysis of Spirulina polysaccharide. [Conclusion] Extraction polysaccharide from Spirulina with boiling wa- ter greatly reduces the use of ethanol, and the possibility of pollution from the ex- ogenous non-food chemical reagents, so that the extracted polysaccharides can be used as food materials. This method makes it possible to establish the production line for Spirulina polysaccharide.

[Method] Because enzyme is capable of destroying organizational structure of cells, papain was selected to cat- alyze cells to let polysaccharides out. An orthogonal experiment was carried out to investigate the effects of hydrolysis temperature, hydrolysis time, enzyme concentra- tion and solid to liquid ratio on extraction of polysaccharides from A. aegirit. [Result] Through the orthogonal experiment, the optimum papain-assisted extraction conditions were obtained: hydrolysis temperature, 45 ℃; hydrolysis time, 2 h; enzyme concen- tration 2%; solid to liquid ratio, 1:80. [Conclusion] This study will provide a reference for extraction of polysaccharides from A. aegirit.

文摘Background:Ampelopsis grossedentata,vine tea,which is the tea alternative beverages in China.In vine tea processing,a large amount of broken tea is produced,which has low commercial value.Methods:This study investigates the influence of different extraction methods(room temperature water extraction,boiling water extraction,ultrasonic-assisted room temperature water extraction,and ultrasonic-assisted boiling water extraction,referred to as room temperature water extraction(RE),boiling water extraction(BE),ultrasonic assistance at room temperature water extraction(URE),and ultrasonic assistance in boiling water extraction(UBE))on the yield,dihydromyricetin(DMY)content,free amino acid composition,volatile aroma components,and antioxidant properties of vine tea extracts.Results:A notable influence of extraction temperature on the yield of vine tea extracts(P<0.05),with BE yielding the highest at 43.13±0.26%,higher than that of RE(34.29±0.81%).Ultrasound-assisted extraction significantly increased the DMY content of the extracts(P<0.05),whereas DMY content in the RE extracts was 59.94±1.70%,that of URE reached 66.14±2.78%.Analysis revealed 17 amino acids,with L-serine and aspartic acid being the most abundant in the extracts,nevertheless ultrasound-assisted extraction reduced total free amino acid content.Gas chromatography-mass spectrometry analysis demonstrated an increase in the diversity and quantity of compounds in the vine tea water extracts obtained through ultrasonic-assisted extraction.Specifically,69 and 68 volatile compounds were found in URE and UBE extracts,which were higher than the number found in RE and BE extracts.In vitro,antioxidant activity assessments revealed varying antioxidant capacities among different extraction methods,with RE exhibiting the highest DPPH scavenging rate,URE leading in ABTS•+free radical scavenging,and BE demonstrating superior ferric ion reducing antioxidant activity.Conclusion:The findings suggest that extraction methods significantly influence the chemical composition and antioxidant properties of vine tea extracts.Ultrasonic-assisted extraction proved instrumental in elevating the DMY content in vine tea extracts,thereby enriching its flavor profile while maintaining its antioxidant properties.

The results showed that the optimum extraction process for alkali-soluble polysacchadde from poria was as follows: solid-liquid ratio of 1:50, alkali concentration of 0.5 mol/L, and extraction time of 8 h.

文摘AIM:To explore the effect and mechanism of Lycium barbarum polysaccharide(LBP)inhibiting retinal neovascularization.METHODS:In vitro tests were performed on human retinal microvascular endothelial cells(HRECs)from three groups,including control group(normal oxygen),hypoxic group(hypoxia at 37℃,1%O_(2),5%CO_(2),and 94%N2),and LBP group(hypoxic group with LBP 100μg/mL).In vivo experiments,C57 mice were divided into three groups:control group(normal rearing group),the oxygen-induced ischemic retinopathy(OIR)group,and the OIR with 50 mg/kg LBP group.Retinal neovascularization was observed by fluorescein angiography and quantified.Retinal thickness was evaluated by Hematoxylin and eosin(HE)stain.The expression of epidermal growth factor receptor(EGFR),phosphatidylinositol 3-kinase(PI3K),mammalian target of rapamycin(mTOR),phosphorylated mammalian target of rapamycin(p-mTOR),protein kinase B(AKT),phosphorylated protein kinase B(p-AKT),interleukin-1β(IL-1β),inducible nitric oxide synthase(iNOS),and tumor necrosis factor-α(TNF-α)in each group were analyzed by Western blot.IL-1βlevel in retina was analyzed using immunohistochemical staining.RESULTS:The increased area of neovascular clusters in OIR mice was significantly decreased by LBP.Retinal thickness of OIR mice was significantly thinner compared with normal oxygenated mice and was increased in LBP group.Compared with those in the hypoxic groups,Western blotting of HRECs and retinal tissues revealed that the expression of EGFR,PI3K,p-mTOR,p-AKT,IL-1β,iNOS,and TNF-αdecreased in the LBP group but was still greater than that in control group.Moreover,IL-1βwas reduced in retinal sections treated with LBP.In the scratch test,the cell migration of the hypoxic group was significantly greater than that of the control group,while LBP treatment attenuated this increase in migration.CONCLUSION:LBP reduces retinal neovascularization and inflammation in vivo and inhibits the migration of HRECs in vitro by regulating the EGFR/PI3K/Akt/mTOR signaling pathway.

文摘AIM:To evaluate the effect of femtosecond laser small incision lenticule extraction(SMILE)on the binocular visual function in myopic patients with glasses-free threedimensional(3D)technique.METHODS:Totally 50 myopic patients(39 females and 11 males)with SMILE were enrolled in this prospective study.The glasses-free 3D technique was used to evaluate the binocular visual function in these subjects including static stereopsis,dynamic stereopsis,foveal suppression,and binocular balance point of signal to noise ratio(s/n ratio).All subjects received measurements in 1d before operation,and 1d,1wk,and 1mo postoperatively.RESULTS:Both static and dynamic stereopsis showed no significant difference after SMILE.The foveal suppression improved significantly 1wk and 1mo after SMILE(P=0.005 and P=0.007 respectively).The binocular balance point of signal to noise ratio showed a significant improvement 1d,1wk and 1mo after SMILE for both eyes(P<0.001 for each eye respectively).CONCLUSION:Glasses-free 3D technique can be used to evaluate the effect of SMILE on the binocular visual function in myopic patients perceptively,and SMILE can improve both foveal suppression and binocular imbalance in these patients.
文摘The effect of material granularity, extraction times, extraction time, mi-crowave power and solid-liquid ratio on the yield of polysaccharide of Dendrobium huoshanense in microwave-assisted extraction was discussed through single factor experiment. The key factors in polysaccharide extraction and the advantages over the traditional extraction method were studied based on orthogonal experiment. The results showed that the influences became unobvious with the promotion of the fac-tor’s level. The orthogonal experiment showed that the optimal factor in the mi-crowave-assisted extraction was that the sample was extracted for four times with an extraction time of 40 min under 550 W. Al these results would lay a founda-tion for exploring a new method for the extraction of polysaccharide from Dendrobi-um huoshanense.

文摘This research optimized the structure of lithium extraction solar ponds to enhance the crystallization rate and yield of Li_(2)CO_(3).Using the response surface methodology in Design-Expert 10.0.3,the authors conducted experiments to investigate the influence of four factors related to solar pond structure on the crystallization of Li_(2)CO_(3) and their pairwise interactions.Computational Fluid Dynamics(CFD)simulations of the flow field within the solar pond were performed using COMSOL Multiphysics software to compare temperature distributions before and after optimization.The results indicate that the optimal structure for lithium extraction from the Zabuye Salt Lake solar ponds includes UCZ(Upper Convective Zone)thickness of 53.63 cm,an LCZ(Lower Convective Zone)direct heating temperature of 57.39℃,a CO32−concentration of 32.21 g/L,and an added soda ash concentration of 6.52 g/L.Following this optimized pathway,the Li_(2)CO_(3) precipitation increased by 7.34% compared to the initial solar pond process,with a 33.33% improvement in lithium carbonate crystallization rate.This study demonstrates the feasibility of optimizing lithium extraction solar pond structures,offering a new approach for constructing such ponds in salt lakes.It provides valuable guidance for the efficient extraction of lithium resources from carbonate-type salt lake brines.
文摘Ultrasonic extraction (UE) was employed for the extraction of bamboo leaf polysaccharides (BLP). The influential parameters of UE procedure including extraction time, ultrasonic power and solid/liquid ratio were optimized by orthogonal experiments. DEAE-cellulose col- umn chromatography was applied to purify BLP and then the radical scavenging activity of BLP was also evaluated. Optimal extraction con- ditions were: extraction time .of 15 min, ultrasonic power of 300 W, and solid/liquid ratio of 1:15. Four kinds of polysaccharides were obtained by DEAE-cellulose colunm chromatography; the maximum superoxide radical scavenging rate (20.4%) of BLP was inferior to that of vitamin C (Vc, the control) and the hydroxyl radical scavenging rate (50%) was equivalent to that of Vc.

文摘BACKGROUND Bletilla striata polysaccharides(BSP)have antioxidant,immune regulation,and anti-fibrotic activities.However,the therapeutic effect and mechanisms underlying the action of BSP in metabolic dysfunction-associated steatotic liver disease(MASLD)have not been fully understood.AIMTo investigate the therapeutic effects and mechanisms of BSP on MASLD by centering on the hepatocyte nuclearfactor kappa B p65(RelA)/hepatocyte nuclear factor-1 alpha(HNF1α)signaling.METHODSA mouse model of MASLD was induced by feeding with a high-fat-diet(HFD)and a hepatocyte model of steatosiswas induced by treatment with sodium oleate(SO)and sodium palmitate(SP).The therapeutic effects of BSP onMASLD were examined in vivo and in vitro.The mechanisms underlying the action of BSP were analyzed for theireffect on lipid metabolism disorder,endoplasmic reticulum(ER)stress,and the RelA/HNF1αsignaling.RESULTSHFD feeding reduced hepatocyte RelA and HNF1αexpression,induced ER stress,lipid metabolism disorder,andnecroptosis in mice,which were significantly mitigated by treatment with BSP.Furthermore,treatment with BSP orBSP-containing conditional rat serum significantly attenuated the sodium oleate/sodium palmitate(SO/SP)-induced hepatocyte steatosis by decreasing lipid accumulation,and lipid peroxidation,and enhancing theexpression of RelA,and HNF1α.The therapeutic effects of BSP on MASLD were partially abrogated by RELAsilencing in mice and RELA knockout in hepatocytes.RELA silencing or knockout significantly down-regulatedHNF1αexpression,and remodeled ER stress and oxidative stress responses during hepatic steatosis.CONCLUSIONTreatment with BSP ameliorates MASLD,associated with enhancing the RelA/HNF1αsignaling,remodeling ERstress and oxidative stress responses in hepatocytes.

文摘Marine organisms constitute unlimited resource of bioactive substances due to their high biodiversity and represent a valuable source of new compounds. This study optimized the alkali-extraction conditions and antioxidant activities of soluble polysaccharides from the body wall of Sipunculus nudus. The effects of solid-liquid ratio, extraction duration, extraction temperature, and alkali concentration on the yield of S. nudus polysaccharides(SNP) were examined, according to which the optimal combination of extraction parameters was obtained by an orthogonal test. The relative influencing importance of different extraction parameters on the yield of SNP followed the order as solid-liquid ratio > extraction temperature > alkali concentration > extraction duration. The highest extraction yield, 1.98%, was achieved under an optimal extraction condition: temperature 60℃, solid-liquid ratio 1:6 g mL^(-1), duration 5 h, and alkali(Na OH) mass fraction 6%. The in vitro antioxidant activities examination showed that extracted SNP under this optimized condition had strong power in reducing certain hydroxyl and superoxide radical scavenging abilities. The promising results showed that extracted SNP could be a potent natural antioxidant.

文摘With the development of the new energy industry and the depletion of nickel sulfide ore resources,laterite nickel ore has become the main source of primary nickel,and nickel for power batteries has become a new growth point in consumption.This paper systematically summarizes the processes,parameters,products,recovery rates,environmental indicators,costs,advantages,disadvantages and the latest research progress of mainstream nickel extraction processes from laterite nickel ore.It also provides a comparative analysis of the environmental impact and economic efficiency of different nickel extraction processes.It is found that the current nickel extraction processes from laterite nickel ore globally for commercial production mainly include the RKEF process for producing ferronickel and the HPAL process for producing intermediate products.The former accounts for about 80%of laterite nickel ore production.Compared to each other,the investment cost per ton of nickel metal production capacity for the RKEF is about 43000$,with an operational cost of about 16000$per ton of nickel metal and a total nickel recovery rate of 77%–90%.Its products are mainly used in stainless steels.For the HPAL process,the investment cost per ton of nickel metal production capacity is about 56000$,with an operational cost of about 15000$per ton of nickel metal and a total nickel recovery rate of 83%–90%.Its products are mainly used in power batteries.The significant differences between the two lies in energy consumption and carbon emissions,with the RKEF being 2.18 and 2.37 times that of the HPAL,respectively.Although the use of clean energy can greatly reduce the operational cost and environmental impact of RKEF,if RKEF is converted to producing high Ni matte,its economic and environmental performance still cannot match that of the HPAL and oxygen-enriched side-blown processes.Therefore,it can be inferred that with the increasing demand for nickel in power batteries,HPAL and oxygen-enriched side blowing processes will play a greater role in laterite nickel extraction.
