This study demonstrates the feasibility of producing three polysaccharides(neutral LJP-1,acidic LJP-2 and acidic LJP-3)with significant in vitro and in vivo anti-inflammatory activities from the flowers of Lonicera ja...This study demonstrates the feasibility of producing three polysaccharides(neutral LJP-1,acidic LJP-2 and acidic LJP-3)with significant in vitro and in vivo anti-inflammatory activities from the flowers of Lonicera japonica.The three polysaccharides differed in chemical composition,molecular weight(Mw)distribution,glycosidic linkage pattern,functional groups and morphology.They exhibited excellent protective effects(in a dose-dependent manner)in lipopolysaccharide-injured RAW264.7 macrophages and Cu SO4-damaged zebrafish via reducing NO production and inhibiting the overexpressions of inflammation-related transcription factors,inflammatory proteins and cytokines in the NF-κB/MAPK signaling pathways.Their antiinflammatory effects varied owing to their different molecular characteristics and chemical compositions.Overall,LJP-2 at 400μg/m L was the most effective.LJP-2 consisted mainly of→5)-α-L-Araf(1→,→4)-α-LGalp A(1→and→2)-α-L-Rhap(1→residues with terminal T-β-D-Glcp.Thus,honeysuckle flowers are good sources of anti-inflammatory polysaccharides,and precise fractionation enables the production of potent antiinflammatory agents for the development of functional foods and healthcare products.展开更多
By a sequentially selective extraction procedure, polysaccharides (PS) from cyanobacterium Spirulina (Arthrospira) maxima that can be bio functionally healthy components were fractionated in four parts including the c...By a sequentially selective extraction procedure, polysaccharides (PS) from cyanobacterium Spirulina (Arthrospira) maxima that can be bio functionally healthy components were fractionated in four parts including the culture medium (PSCM), the external layers of the cell (PSEL), the cell wall (PSCW) and storage granules (PSSG). The four fractionated parts of polysaccharides were characterized by gas liquid chromatography (GLC) equipped with FID and fused silica capillary column (15 m×0.53 mm i. d.). The contents of PSCM, PSEL, PSCW and PSSG were about 2.0%, 2.6%, 10.0% and 52.0% to cell dry matter, respectively. Glucose was almost the only monosaccharide in PSSG and PSCW and most predominant in PSEL, while in PSCM, xylose, rhamnose and glucose were the main ones. Two uronic acids represented by glucuronic acid and galacturoic acid, six neutral monosaccharides including fucose, rhamnose, xylose, mannose, galactose and glucose, and two possible unidentified sugars were found in PSEL and PSCM. A. Meyer were totally fractionated into one neutral fraction(GLPN-1), six acidic fractions(GLPN-2, GLPA-1a, GLPA-1b, GLPA-1c, GLPA-2 and GLPA-3) by a combination of ethanol precipitation, ion-exchange and gel permeation chromatography. All the fractions were analyzed by determining monosaccharide composition, molecular weight distribution. The resuits show that GLPN-1 was mainly composed of Gal(38.1%) and Ara(33.6%) and presumed to be an arabinogalactan. GLPA-1a, GLPA-1b and GLPA-2 mainly contained Gal, Ara, Rha and GalA. The amount of Gal and Ara acounted for over 50% of total sugar and the amount of Rha was more than 10%. The molar ratios of Rha/GalA in GLPA-1a, GLPA-1b and GLPA-2 were 1:1, 1:2 and 1:2.7, respectively, indicating that they belong to type-1 rhamnogalacturonan (RG-1)-rich pectin. Fortunella margarita polysaccharides(FMPS)are composed of 4 polysaccharide fragments,namely mannogalactoglucan,galactoglucan,glucan and arabinoglucan.This study investigated the in vitro fermentation of FMPS fractions...Fortunella margarita polysaccharides(FMPS)are composed of 4 polysaccharide fragments,namely mannogalactoglucan,galactoglucan,glucan and arabinoglucan.This study investigated the in vitro fermentation of FMPS fractions and their effect on the fecal microbiota of mice and short chain fatty acids(SCFAs)production.The results showed that the structure of the fecal microbiota was changed after FMPS fermentation.At the phylum level,the polysaccharide fractions reduced the relative abundance of Bacteroidetes compared with the inulin group.At the genus level,the polysaccharide fractions increased the abundance of Lactobacillus,and decreased the abundance of Granulicatella,Proteus and Rummeliibacillus.Polysaccharide fractions could promote the proliferation of beneficial bacteria(Lactobacillus),and decreased harmful bacteria(Granulicatella,Proteus,Rummeliibacillus and Serratia).Furthermore,there was a significant distinction in the genus level flora processed by different polysaccharide fractions,especially mannogalactoglucan.Polysaccharide fractions showed potential prebiotic effects,with mannogalactoglucan,in particular,promoting proliferation of SCFA-producing bacteria.SCFAs were regulated by monosaccharide composition,molecular weight of the FMPS fractions,and the composition of the fecal bacteria.Polysaccharide fractions could modulate metabolic function of the fecal microbiota,altering the levels of SCFAs.These results suggested that FMPS are important functional components in gut health,especially mannogalactoglucan.展开更多
