This work quantified changes in tissue Vc levels during winter hibernation in the soft-shelled turtle Pelodiscus sinensis , to reveal the stress response pattern of tissue Vc during hibernation and contribute basic data for turtle culture. We sampled juvenile soft-shelled turtles at pre-hibernation (17.0°C mud temperature;MT), during hibernation (5.8°C MT) and after arousal (20.1°C MT) in the field. The transcript levels of the gene encoding L-gulonolactone oxidase (GLO), the key enzyme for Vc synthesis, decreased significantly during hibernation. However, GLO activity did not match the GLO transcription patterns and remained stable during hibernation, and showed temperature-dependent kinetic characteristics. Vitamin C levels in the brain, liver, kidney, and spleen (but not muscle) all decreased significantly during hibernation, but recovered to pre-hibernation levels or even higher levels after arousal. The soft-shelled turtle endured 5 months of hibernation with no significant oxidative damage in most tissues, except in the spleen. Splenic Vc was nearly exhausted during hibernation, accompanied by a significantly elevated malonaldehyde (MDA) level. Although the high level of oxidative damage quickly decreased after arousal, the potential tissue damage in the spleen during hibernation might account for the weakened immune capacity of turtles after hibernation.展开更多
To study the morphology and physiology of blood cells from five species of heterotherm vertebrates(carp Cyprinus carpio,crucian Carassius auratus,catfish Silurus asotus,toad Bufo bufo gar-garizans and Chinese soft-sh...To study the morphology and physiology of blood cells from five species of heterotherm vertebrates(carp Cyprinus carpio,crucian Carassius auratus,catfish Silurus asotus,toad Bufo bufo gar-garizans and Chinese soft-shelled turtle Pelodiscus sinensis),three kinds of living cells(thrombocytes,small lymphocytes,erythrocytes)were obtained from the experimental animals.Plasma samples were partly anticoagulated with trisodium citrate,potassium oxalate and magnesium sulphate(MgSO4)at 23℃ in vitro,and observed in the light microscope and scanning electron microscope.Results showed that:1)small lymphocytes were typically spherical and normally dispersed in oxalate-plasma from all of the experiment animals and in citrated-plasma from carp and crucian.However,they were aggregated in MgSO4-plasma from all of the experiment animals and in citrated-plasma from catfish,toad and Chinese soft-shelled turtle.2)thrombocytes were dispersive and spindle or elliptical in shape in oxalate-plasma from the five species of experimental animals;in citrated-plasma from carp,crucian and catfish;in MgSO4-plasma from carp,crucian,catfish and Chinese soft-shelled turtle.However,they were aggregated in citrated-plasma from toad and Chinese soft-shelled turtle,and in the MgSO4-plasma from toad.3)erythrocytes were typically normal,and not haemolytic in citrated-plasma and in MgSO4-plasma from five species of experimental animals,but they were completely haemolytic in oxalate-plasma from experimental animals except for toads.Rouleaux formation of erythrocytes,such as occurs in mammalian plasma,was not observed in the experimental animals.We conclude that:1)the same of small lymphocytes or the thrombocytes in heterotherm vertebrate plasma possess the ability to aggregate together in vitro,the process of the aggregation may relate to plasma Ca 2+ and Mg 2+ concentration;2)the process of erythrocyte sedimentation caused by their specific gravity in plasma of these experimental animals,is different from that caused by rouleaux formation of erythrocytes in mammalian plasma.展开更多
In this paper, RT PCR analysis on SOX gene of seven tissues from the Chinese soft shelled turtle (Pelodiscus sinensis) was studied, and SOX gene fragments of expression from the testicle, brain, heart, kidney and sple...In this paper, RT PCR analysis on SOX gene of seven tissues from the Chinese soft shelled turtle (Pelodiscus sinensis) was studied, and SOX gene fragments of expression from the testicle, brain, heart, kidney and spleen were cloned using RT PCR products.The results show that SOX genes has specific expression in the testicle, brain, spleen, cardiac muscle and kidney and isn’t expression in muscle, liver and ovary of femel.The results of sequence reveal that the SOX genes of expression in the testicle are TSSOX1、TSSOX4 TSSOX5 and TSSOX9, and those are TSSOX2 and TSSOX4 in the brain, and this is TSSOX4 in the spleen and heart tissue, and those are TSSOX2 and TSSOX3 in the kidney tissue.This suggests that the SOX gene act important role not only on the sex determination, but also on the development of neural system, immunocyte system and the differentiation of male germ cell.展开更多
