Brucellosis is one of the most widespread zoonotic diseases, with the most frequent complication being osteoarticular changes. The aim of this study was to assess the changes of C-terminal telopeptide of type II collagen (CTX-II) in patients infected with brucellosis. A total of 84 brucellosis patients and 43 volunteers were selected and divided into brucellosis vs. control groups. Serum samples were subjected to serological tests for brucellosis, 展开更多

The methods and strategies used to screen for syphilis and to confirm initially reactive results can vary significantly across clinical laboratories. While the performance characteristics of these different approaches have been evaluated by multiple studies, there is not,as of yet,a single, universally recommende dalgorithm for syphilis testing. To clarify the currently available options for syphilis testing, this update will summarize the clinical challenges to diagnosis, review the specific performance characteristics of treponemal and non-treponemal tests, and finally, summarize select studies published over the past decade which have evaluated these approaches.Specifically, this review will discuss the traditional and reverse sequence syphilis screening algorithms commonly used in the United States, alongside a discussion of the European Centre for Disease Prevention and Control syphilis algorithm.Ultimately,in the United States, the decision of which algorithm to use is largely dependent on laboratory resources, the local incidence of syphilis and patient demographics. 展开更多

Background Few literatures pertain to the 16S ribosomal DNA (16S rDNA) analysis of bacteria contributing to primary and persistent endodontic lesions, with no information available for the Chinese population. As such, we investigated endodontic bacteria associated with primary and persistent endodontic lesions in adult Chinese patients living in Beijing, China using 16S rDNA gene sequencing techniques. Methods Endodontic microbial samples were obtained from fourteen adult Chinese patients and subjected to DNA extraction. Pllymerase chain reaction (PCR) products were cloned and 100 clones from each generated library were randomly selected. Purified plasmid DNA with 16S rDNA gene inserts was sequenced, and the sequences were searched against GenBank databases using the BLASTN algorithm. Only significant identification with the highest-scored BLAST result and 99% minimum similarity was considered for phylotyping. Results More than 150 taxa were obtained. Primary endodontic infection was mainly associated with Burkholderia cepacia, Actinomyces, Aranicola spp. and Streptococcus sanguinis, whilst Burkholderia cepacia was predominant in the persistent endodontic infections. Conclusion There is a difference in the species profile associated with endodontic infections of Chinese patients living in Beijing in comparison to other geographical or ethnic reports. 展开更多

Woodchuck hepatitis virus (WHV) is molecularly and pathogenically closely related to hepatitis B virus (HBV).Both viruses display tropism towards hepatocytes and cells of the immune system and cause similar liver pathology, where acute hepatitis can progress to chronic hepatitis and to hepatocellular carcinoma (HCC).Two forms of occult hepadnaviral persistence were identified in the woodchuck-WHV model:secondary occult infection (SOI) and primary occult infection (POI).SOI occurs after resolution of a serologically apparent infection with hepatitis or after subclinical serologically evident virus exposure.POI is caused by small amounts of virus and progresses without serological infection markers, but the virus genome and its replication are detectable in the immune system and with time in the liver.SOI can be accompanied by minimal hepatitis, while the hallmark of POI is normal liver morphology.Nonetheless,HCC develops in about 20% of animals with SOI or POI within 3 to 5 years. The virus persists throughout the lifespan in both SOI and POI at serum levels rarely greater than 100 copies/mL,causes hepatitis and HCC when concentrated and administered to virus-na(i)ve woodchucks.SOI is accompanied by virusspecific T and B cell immune responses, while only virusspecific T cells are detected in POI.SOI coincides with protection against reinfection, while POI does not and hepatitis develops after challenge with liver pathogenic doses >1000 virions. Both SOI and POI are associated with virus DNA integration into the liver and the immune system genomes.Overall,SOI and POI are two distinct forms of silent hepadnaviral persistence that share common characteristics. Here,we review findings from the woodchuck model and discuss the relevant observations made in human occult HBV infection (OBI). 展开更多

Multiple Omicron sub-lineages have emerged, with Omicron XBB and XBB.1.5 subvariants becoming the dominant variants globally at the time of this study. The key feature of new variants is their ability to escape humoral immunity despite the fact that there are limited genetic changes from their preceding variants. This raises the question of whether Omicron should be regarded as a separate serotype from viruses serologically clustered with the ancestral severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)virus.Here,we present cross-neutralization data based on a pseudovirus neutralization test using convalescent sera from naïve individuals who had recovered from primary infection by SARS-CoV-1 and SARS-CoV-2 strains/variants including the ancestral virus and variants Beta,Delta,Omicron BA.1,Omicron BA.2 and Omicron BA.5.The results revealed no significant cross-neutralization in any of the three-way testing for SARS-CoV-1,ancestral SARS-CoV-2 and SARS-CoV-2 Omicron subvariants. The data argue for the assignment of three distinct serotypes for the currently known human-infecting SARS-related coronaviruses. 展开更多

Prolonged antiretroviral therapy(ART)-free remission post-treatment has been observed and reported in human immunodeficiency virus-1(HIV-1)infection.The primary factors for such achievement have been linked to the transcriptional inactivation of HIV-1 DNA and immune recovery. Here,we reported a patient with acute HIV-1 infection who immediately received intensified quadruple ART.At month 9(M9),an old CFDA-approved immuno-suppressive herbal medicine,Tripterygium Wilfordii Hook F(TwHF),was used in addition to ART.The patient was closely monitored.Virological and immunological tests as well as transcriptome analysis were carried out at each visit. The results showed that TwHF reduced serum IP-10 level and inhibited T cell activation. Both ART and TwHF were discontinued in M24,and levels of peripheral blood HIV-1 RNA and DNA remained suppressed for consecutive 12months.With transcriptome analysis,we found pattern changes linking immuno-activation and amino acid metabolism with viral suppression and rebound. This indicates that the intentional suppression of immuno-activation is a promising approach for a functional cure of HIV-1 infection. 展开更多

A nonlinear SEIR mathematical model is developed to investigate the impact of migrated population, infected with Ebola virus,on human-to-human transmission of Ebola Virus Disease(EVD)in a disease-free area.In view of the dynamics of Ebola virus disease, here, the infected class is supposed to be divided into subclasses, viz.primary and secondary infected. The proposed model is analyzed qualitatively using the stability theory of differential equations and quantitatively using numerical simulation. The obtained results, qualitatively and quantitatively, suggest that migration and contact rates play an important role in controlling the spreading of disease.Critical values for migration and contact rates are evaluated and it is revealed that if these rates go beyond their critical values,it leads to delay in the stabilization of the system. It is also found that primary reproductive number increases with increase in migration rate.Besides this, the approximate time required to attain stability of the disease model system is also determined. The model analysis recommends quarantining the noninfected from the secondary infected in order to control the spreading out of disease. 展开更多
