This review compiles information from the literature on the chemical composition, pharmacological effects, and molecular mechanisms of earthworm extract(EE)and suggests possibilities for clinical translation of EE.We also consider future trends and concerns in this domain. We summarize the bioactive components of EE,including G-90,lysenin,lumbrokinase,antimicrobial peptides, earthworm serine protease(ESP),and polyphenols, and detail the antitumor,antithrombotic,antiviral,antibacterial,anti-i nflammatory,analgesic,antioxidant,wound-healing,antifibrotic,and hypoglycemic activities and mechanisms of action of EE based on existing in vitro and in vivo studies. We further propose the potential of EE for clinical translation in anticancer and lipid-modifying therapies, and its promise as source of a novel agent for wound healing and resistance to antibiotic tolerance. The earthworm enzyme lumbrokinase embodies highly effective anticoagulant and thrombolytic properties and has the advantage of not causing bleeding phenomena due to hyperfibrinolysis. Its antifibrotic properties can reduce the excessive accumulation of extracellular matrix. The glycolipoprotein extract G-90 can effectively scavenge reactive oxygen groups and protect cellular tissues from oxidative damage.Earthworms have evolved a well-developed defense mechanism to fight against microbial infections, and the bioactive agents in EE have shown good antibacterial, fungal, and viral properties in in vitro and in vivo experiments and can alleviate inflammatory responses caused by infections, effectively reducing pain.Recent studies have also highlighted the role of EE in lowering blood glucose.EE shows high medicinal value and is expected to be a source of many bioactive compounds. 展开更多

The degree of processing is rarely considered an independent factor in the health effects of fruit juices and beverages(FJBs)consumption. In fact, the consumption of ultra-processed foods has been shown to pose health risks. In this study,we first integrated 4 systems used to classify the degree of food processing and then classified FJBs into three major categories, low(minimal),moderate and high.Second,we compared the differences in attitudes towards FJBs in dietary guidelines.Third,we integrated the results of existing epidemiological surveys, randomized controlled trials, and animal experiments to explore the health risks associated with consuming FJBs.Deepening the processing of FJBs has been found to lead to an increased risk of diseases.Dietary pattern, nutrients, addition agents and consumer preferences may be influential factors. Finally,we investigated whether there were any changes in the health benefits of 100%fruit juices produced by different processing methods. In conclusion,minimally/moderately processed 100%fruit juices provide more health benefits than highly processed fruit beverages. The results support the need to consider the extent of FJBs processing in future studies to adjust official nutritional recommendations for beverage consumption. 展开更多

Denervation-induced skeletal muscle atrophy can potentially cause the decline in the quality of life of patients and an increased risk of mortality.Complex pathophysiological mechanisms with dynamic alterations have been documented in skeletal muscle atrophy resulting from innervation loss.Hence,an in-depth comprehension of the key mechanisms and molecules governing skeletal muscle atrophy at varying stages, along with targeted treatment and protection, becomes essential for effective atrophy management. Our preliminary research categorizes the skeletal muscle atrophy process into four stages using microarray analysis. This review extensively discusses the pathways and molecules potentially implicated in regulating the four stages of denervation and muscle atrophy.Notably, drugs targeting the reactivare oxygen species stage and the inflammation stage assume critical roles.Timely intervention during the initial atrophy stages can expedite protection against skeletal muscle atrophy.Additionally, pharmaceutical intervention in the ubiquitin-proteasome pathway associated with atrophy and autophagy lysosomes can effectively slow down skeletal muscle atrophy.Key molecules within this stage encompass MuRF1,MAFbx,LC3II,p62/SQSTM1,etc.This review also compiles a profile of drugs with protective effects against skeletal muscle atrophy at distinct postdenervation stages, thereby augmenting the evidence base for denervation-induced skeletal muscle atrophy treatment. 展开更多

Through exploring the effects of low pH on the composite system of desulfurization gypsum(DG)enhanced by melamine-formaldehyde resin(MF),it is found that the inducing of sulfate-ion,in contrast to chloride and oxalate ions, favors the longitudinal growth of the crystalline form of the hydration product, which was relatively simple and had the highest length to width(L/D)ratio.At the same time,MF can also improve L/D ratio of gypsum hydration products, which favors the formation of hydrated whiskers. Finally,in a composite system composed of hemihydrate gypsum,MF,and glass fibers, when dilute sulfuric acid was used to regulate pH=3-4,the tight binding formed among the components of the composite system compared to pH=5-6.The hydration product of gypsum adheres tightly to glass fiber surface and produces a good cross-linking and binding effect with MF.The flexural strength, compressive strength, elastic modulus, and water absorption of the desulphurized gypsum composite board is 22.7 MPa,39.8 MPa,5608 MPa,and 1.8%,respectively. 展开更多

This study aims to identify a natural plant chemical with hypolipidemic effects that can be used to treat high cholesterol without adverse reactions.Through network pharmacology screening,it was found that Rosae Rugosae Flos(RF)flavonoids had potential therapeutic effects on hyperlipidemia and its mechanism of action was discussed.TCMSP and GeneCards databases were used to obtain active ingredients and disease targets.Venn diagrams were drawn to illustrate the findings. The interaction network diagram was created using Cytoscape 3.8.0 software. The PPI protein network was constructed using String.GO and KEGG enrichment analysis was performed using Metascape. The results revealed 2 active flavonoid ingredients and 60 potential targets in RF.The key targets, including CCL2,PPARG,and PPARA,were found to play a role in multiple pathways such as the AGE-RAGE signaling pathway, lipid and atherosclerosis, and cancer pathway in diabetic complications. The solvent extraction method was optimized for efficient flavonoid extraction based on network pharmacology prediction results. This was achieved through a single factor and orthogonal test, resulting in an optimum process with a reflux time of 1.5 h,a solid-liquid ratio of 1:13 g/mL,and an ethanol concentration of 50%. 展开更多

This study aimed to investigate the mechanism of action of Sophora Flos(SF)in the treatment of hyperlipidemia(HLP)using network pharmacology and molecular docking methods, and to optimize the extraction process of the predicted active components. The STRING database was used for protein interaction analysis and PPI network construction via Cytoscape 3.9.1.Pymol was employed for docking and visualization. An extensive review of SF identifi ed 6 active ingredients,297 related objectives,84 disease objectives, and 57 total objectives. After protein interaction and topology analysis,18 core targets were identified. These included 146 gene function entries(P<0.05).Active compounds, mainly flavonoids, can modulate the expression of various proteins such as TNF,IL-6,IL-1β,PPARG,and TGFB1 to achieve therapeutic effects on HLP.The network pharmacology and molecular docking results suggested that the active fl avonoids component in SF may be related to the treatment of hyperlipidemia. Therefore, the orthogonal experiment method was used to optimize the extraction process of total fl avonoid from SF using ethanol refl ux extraction, based on a single factor experiment. The effects of refl ux time,solid-liquid ratio, ethanol concentration, and other factors on the extraction of total fl avonoid from SF were investigated. The optimum process conditions were refl ux time of 1.25 h,solid-liquid ratio of 1:15 g/mL and ethanol concentration of 60%.Using these conditions, the purity of total fl avonoid extracted from SF was 70.33±0.22%. 展开更多

Effects on Sedimentary Processes via Upper Triassic Climate Forcing Caused by Multiple Impacting and Large Igneous Provinces (LIP)-Rifting/Degassing: Jordanian Platform/Arabian Plate and Germanic Basin/Central Europe

Upper Triassic sedimentary systems of both the Arabian Plate and the Germanic Basin reveal climate- and plate tectonic-forced effects through certain time-intervals experienced by architectural elements, lithofacies types, unconformities, flash flood deposits, maximum flooding surfaces/sequence boundary (MFS/SB), mineralogy, and isotope anomalies. Further, Moon recession and changes of Earth’s rotation velocity (core/mantle boundary) are associated with multiple impacting and large igneous provinces/Mid Oceanic Ridge Basalt, LIP/MORB-rifting/degassing. While acidification (by degassing, sturz-rain) does influence tectosilicates and carbonates, montmorillonite represents a key mineral as transformation of volcanic/impact glass (Tephra) to be found as co-components in and in certain pelite units as “boundary clay-suspicions” (mixture of eolian paleoloess, pelite, paleosol, and tephra → tuffite). Obviously, unconformities and sequence boundaries of both study areas separate and dislocate interrupted ∂13C and 87Sr/86Sr-data groups along the isotope curves. Both Proto-Arctic Ocean rifting/degassing comprising kimberlitic pyroclastic eruptions and Neotethys rifting/degassing as well as multiple impacting played the most important role during the Norian, followed by the incipient Central Atlantic Magmatic Provinces rifting since the Rhaetian. The following associations are encountered and dealt with in this study: Sequence boundaries-∂13C, maximum flooding surfaces-(FUCs)-∂13C, unconformities-plate motion, tephra-pelite-tuffite-montmorillonite. Norian: maximum flooding surfaces (MFSs)-“paleosol”/boundary clay?-rifting-volcanism, Moon/Earth data change. So the Norian (~221 - 206 Ma) hosts anomalous “amalgamated maximum flooding surfaces (MFSs)”, amalgamated paleosol (Jordanian Platform), multiple impacting (~219 - 214 Ma), the maximum opening of the Proto-Arctic Ocean (PAO) (~230 - 200 Ma), Neo-Tethys (NT)-subvolcanic (sills, dikes) in the NE Dead Sea area prior the Rhaetian, and a significant change of Earth/Moon relation data. The study concludes that rare and extreme events are very strongly shaping the geologic constellations in the Earth System. 展开更多
