Summary: The expression of DNA ploidy, the cell cycle and Ki67 antigen in nasopharyngeal carcinoma (NPC) were studied and their relationship with the clinical biological behaviors and prognosis of NPC was evaluated. Biopsied specimens of NPC were made into cell suspension. By using cytometric double labeling Ki67 and DNA method, the expression of DNA ploidy, the cell cycle and Ki67 antigen were analyzed. The patients were followed-up for about 3 years and the relationship between the above-mentioned parameters and the clinical biological behavior and prognosis of NPC were evaluated. Of the 62 cases of NPC, the DNA aneuploid accounted for 29.03 %. The S phase cells accounted for 0 to 54 % in the cell cycle and the positive expression of Ki67 ranged from 0 to 52 %. There were 40 cases of LPI (64.5 %) including 15 negative cases and 22 cases of HPI (35 5 %) respectively. The DNA anueploid content was positively related to the S phase cells. The patients having a low expression of Ki67 or DNA aneuploid in tumor cells were not sensitive to chemotherapy, liable to metastasis to distant organs and had a poor prognosis, while Ki67 showed no correlation with DNA ploidy and the cell cycle. It was suggested that DNA ploidy and Ki67 could be used as an independent and objective marker to evaluate the radiosensitivity and prognosis of NPC. 展开更多

Background: Evaluating the tumor proliferative index helps predict clinical behavior and provides prognostic insights for cutaneous basal cell carcinoma (cBCC) and squamous cell carcinoma (cSCC). Objective: This study aimed to identify differences in the proliferative indices among variants of cBCC and cSCC diagnosed at a tertiary healthcare center. Method: Skin biopsies histologically diagnosed as cBCC and cSCC between 2012 and 2018 at the Federal Medical Centre (FMC) Umuahia, Abia State, Nigeria, were analyzed. Archival formalin-fixed, paraffin-embedded (FFPE) tissue blocks were retrieved along with clinical data, and were prepared on charged microscope slides and the immunohistochemical staining was carried out. The primary antibody used in this study was clone BioCare CRM325C (RM) and adenotonsillar tissue blocks/slides served as positive controls. Ki-67 immunohistochemistry was performed on fresh 4µm sections of the tumor specimens. Results: The application of Ki-67 immunoperoxidase on both BCC and SCC cohort, yielded an intense observable brownish nuclear stain in areas of dense proliferating tumour cells on both cutaneous tumours. The average Ki-67 index for all cSCC cases was 24.7%, with a range of 2.3% - 80%, while the mean for cBCC was 15.8%, ranging from 1.2% - 45.6%. Variants with high proliferative indices were observed in 11.9% of cBCC cases and 29.1% of cSCC cases. Among the low proliferative index category, cSCC accounted for 5.4%, while cBCC represented 14.3%. For mild proliferative indices, cSCC cases made up 7.3% and cBCC, 11.9%. The majority of cases showed moderate proliferative indices, with 61.9% for cBCC and 58.2% for cSCC. Overall, there was a significant difference in proliferative indices between cSCC, cBCC, and their variants. Conclusion: The study found a significantly higher rate of cell proliferation, measured by Ki-67 immunostaining, in cSCC and its variants compared to cBCC. However, certain variants of cBCC also exhibited high Ki-67 expression, indicating they can be as aggressive as some cSCC variants. 展开更多

BACKGROUND: Gallbladder carcinoma is a highly lethal and aggressive disease with early metastasis, strong invasion and poor prognosis. Most patients with this disease are at the advanced and un-resectable stage and should be consi- dered for palliative treatment such as chemotherapy and ra- diotherapy. Unfortunately, reports of chemotherapy and radiotherapy for gallbladder carcinoma are disappointing. We investigated the influence of norcantharidin (NCTD) on proliferation, proliferation-related gene proteins PCNA and Ki-67 of human gallbladder carcinoma GBC-SD cells in vitro. METHODS: GBC-SD cell lines of human gallbladder carci- noma were cultured by the cell culture technique. The ex- periment was divided into NCTD group and control group. The tetrazolium-based colorimetric assay was used to evaluate cell growth. The streptavidin-biotin complex method was used to determine the expressions of prolifera- tion-related gene proteins PCNA and Ki-67 of human gall- bladder carcinoma GBC-SD cells. RESULTS: NCTD inhibited the growth and proliferation of GBC-SD cells from 10 mg/L or after 6 hours in a dose- and time-dependent manner, with the IC50 value of 56.18 μg/ ml at 48 hours. After treatment with NCTD, the expression of PCNA (0.932 ±0.031 vs. 0.318 ±0.023, P<0.001) and Ki-67 (0.964 ±0.092 vs. 0.297 ±0.018, P<0.001) proteins were decreased significantly. CONCLUSION: NCTD inhibits the proliferation of human gallbladder carcinoma GBC-SD cells in vitro and the expres- sion of their proliferation-related gene proteins PCNA and Ki-67. 展开更多

背景:DCLK1是在有丝分裂后的神经元中发现的一种跨膜微管相关蛋白激酶,近些年来发现其可能是肠道肿瘤干细胞的标志物。目的:观察DCLK1与Ki67在结直肠肿瘤中的表达及其病理学意义,探讨是否可以将DCLK1+/Ki67-作为结直肠癌肿瘤干细胞的标... 背景:DCLK1是在有丝分裂后的神经元中发现的一种跨膜微管相关蛋白激酶,近些年来发现其可能是肠道肿瘤干细胞的标志物。目的:观察DCLK1与Ki67在结直肠肿瘤中的表达及其病理学意义,探讨是否可以将DCLK1+/Ki67-作为结直肠癌肿瘤干细胞的标志物。方法:运用免疫组织化学方法检测150例结直肠癌组织中DCLK1与Ki67的表达,并与正常肠黏膜组织、癌旁组织、腺瘤组织比较。结果与结论:DCLK1和Ki67在癌组织中的表达率分别为36.7%,34.7%,其数量明显高于正常肠黏膜和腺瘤组织;DCLK1的表达与部位、浸润深度、淋巴结转移有关(P<0.05),而Ki67的表达仅与浸润深度有关(P<0.05);DCLK1与Ki67的表达呈负相关(r=-0.460,P=0.000)。结直肠癌组织中DCLK1+/Ki67-细胞数量约为2.01%,主要分布于肠黏膜隐窝的基底部及共壁腺管的共壁处,细胞核大深染且呈圆形或卵圆形,核仁明显,核分裂象少见,细胞质较少。DCLK1+/Ki67-细胞在不同分化程度的腺癌组织中均存在,且分化程度越低,细胞数量越多,故无论从细胞数量和位置,还是细胞形态,都表明DCLK1+/Ki67-细胞符合肿瘤干细胞的特征,可以将DCLK1+/Ki67-作为结直肠癌肿瘤干细胞的标志物。 展开更多

P53、P16及Ki-67与早期食管癌患者ESD术后复发的关系分析

目的 分析研究抑制蛋白基因P53(P53)、抑制蛋白基因P16(P16)及细胞增殖核抗原Ki-67(Ki-67)与早期食管癌患者内镜黏膜下剥离术(ESD)术后复发的关系。方法 选取2019年1月至2023年1月三门峡市中心医院收治80例的早期食管癌患者作为研究对象... 目的 分析研究抑制蛋白基因P53(P53)、抑制蛋白基因P16(P16)及细胞增殖核抗原Ki-67(Ki-67)与早期食管癌患者内镜黏膜下剥离术(ESD)术后复发的关系。方法 选取2019年1月至2023年1月三门峡市中心医院收治80例的早期食管癌患者作为研究对象,均进行ESD治疗。比较所有患者癌组织与癌旁组织(距肿瘤边缘>3 cm,镜下未见肿瘤组织或不典型增生组织)P53、P16、Ki-67蛋白表达。ESD术后对患者随访1年,根据有无术后复发食管癌,将患者分为复发组(n=20)与无复发组(n=60)。比较两组癌组织P53、P16、Ki-67蛋白表达。采用多因素Logistic回归分析早期食管癌患者ESD术后复发的影响因素,并绘制ROC曲线分析P53、P16、Ki-67蛋白表达与早期食管癌患者ESD术后复发。结果 与癌旁组织相比,癌组织P53、Ki-67蛋白表达升高,P16蛋白表达降低(t=9.276、13.987、10.595,均P<0.05);复发组的癌组织P53、Ki-67蛋白表达均高于无复发组,P16蛋白表达低于无复发组(t=5.086、4.648、5.139,均P<0.05);多因素Logistic回归分析显示,肿瘤低分化(OR=1.870)、肿瘤浸润侵犯黏膜下层(OR=1.808)、有淋巴结转移(OR=2.089)、P53蛋白高表达(OR=2.046)、P16蛋白低表达(OR=1.988)及Ki-67蛋白高表达(OR=1.761)均是早期食管癌患者ESD术后复发的独立危险因素(P<0.05);ROC曲线分析显示,P53、P16、Ki-67蛋白表达及联合检测的曲线下面积(AUC)分别为0.803、0.828、0.834、0.942,联合检测优于单一检测(P<0.05)。结论 P53、P16及Ki-67蛋白与早期食管癌患者ESD术后复发情况相关,可以作为辅助预测的相关诊断指标。 展开更多

miR-4534对宫颈癌细胞增殖的影响及其临床病理意义

目的:研究宫颈鳞状细胞癌(CSCC)中miR-4534的表达水平、病理意义以及对细胞增殖的影响。方法:通过qRT-PCR实验检测71例CSCC组织中miR-4534的表达水平,分析miR-4534表达与患者临床病理特征的相关性。运用CCK-8实验检测miR-4534对增殖活... 目的:研究宫颈鳞状细胞癌(CSCC)中miR-4534的表达水平、病理意义以及对细胞增殖的影响。方法:通过qRT-PCR实验检测71例CSCC组织中miR-4534的表达水平,分析miR-4534表达与患者临床病理特征的相关性。运用CCK-8实验检测miR-4534对增殖活性的影响。通过荷瘤实验观察减低miR-4534表达后对移植瘤生长的影响;并通过增殖指标Ki-67免疫组化染色验证miR-4534对CSCC增殖的影响。结果:qRT-PCR检测出miR-4534在CSCC组织中表达水平升高,明显高于邻近正常宫颈组织,有统计学差异(P<0.05)。高表达的miR-4534与肿瘤直径和分化程度密切相关(均P<0.05),而与患者年龄、HPV感染、淋巴结转移和TNM分期无相关性(均P>0.05)。这一结果提示miR-4534参与了CSCC的生长增殖过程。CCK-8实验结果显示,在72h时与过表达对照组(0.67±0.05)相比,过表达miR-4534促进了C33A细胞的增殖活性(0.92±0.04);与减低表达对照组(0.70±0.06)相比,降低miR-4534表达则抑制了C33A细胞的增殖活性(0.48±0.03),差异均有统计学意义(P均<0.05)。裸鼠皮下移植瘤实验中,降低miR-4534表达后移植瘤的重量和体积均小于对照组,差异有统计学意义(均P<0.05)。细胞增殖指数Ki-67免疫组化染色显示减低miR-4534表达后Ki-67阳性细胞数目明显低于对照组,差异有统计学意义(P<0.05)。结论:miR-4534是一个新的CSCC促癌基因,能促进CSCC细胞的增殖生长,本研究为未来治疗CSCC提供新的策略。 展开更多

三阴性乳腺癌EGFR、Ki-67、P53及CTC表达与预后的关系研究

目的探讨三阴性乳腺癌表皮生长因子受体(EGFR)、细胞核增殖相关抗原Ki-67(Ki-67)、P53及循环肿瘤细胞(CTC)表达与预后的关系。方法选取95例三阴性乳腺癌患者,免疫组化检测病理组织标本中EGFR、Ki-67、P53表达;所有患者接受8个周期化疗,... 目的探讨三阴性乳腺癌表皮生长因子受体(EGFR)、细胞核增殖相关抗原Ki-67(Ki-67)、P53及循环肿瘤细胞(CTC)表达与预后的关系。方法选取95例三阴性乳腺癌患者,免疫组化检测病理组织标本中EGFR、Ki-67、P53表达;所有患者接受8个周期化疗,采用膜滤过分离肿瘤细胞技术(ISET)检测化疗前后的CTC表达,并分析化疗前后CTC表达与化疗疗效的关系;分析CTC与EGFR、Ki-67、P53表达的关联性;随访患者无进展生存期(PFS),采用COX回归分析三阴性乳腺癌进展的危险因素。结果EGFR、Ki-67、P53阳性检出率分别为44.21%(42/95)、63.16%(60/95)、56.84%(54/95);化疗后患者的CTC阳性检出率(14.74%)低于化疗前(61.05%,P<0.05);化疗疗效与化疗后CTC阳性表达呈负相关(P<0.001);COX回归分析显示,临床分期为Ⅲ期、EGFR阳性、化疗后CTC阳性是三阴性乳腺癌进展的独立危险因素(P<0.05);不同临床分期患者PFS比较,Ⅰ期>Ⅱ期>Ⅲ期,EGFR阳性患者PFS短于阴性患者,化疗后CTC阳性患者PFS短于阴性患者(P<0.05)。结论化疗前EGFR阳性表达、化疗后CTC阳性表达与三阴性乳腺癌患者预后差有关,化疗后CTC阳性率越低患者化疗疗效越好。 展开更多

AIM: To evaluate the effect of preoperative transcatheter arterial chemoembolization (TACE) on proliferation of hepatocellular carcinoma (HCC) cells. METHODS: A total of 136 patients with HCC underwent liver resection. Of 136 patients, 79 patients received 1 to 5 courses of TACE prior to liver resection (TACE group), who were further subdivided into four groups: Group A (n = 11) who received i to 4 courses of chemotherapy alone; Group B (n = 33) who received i to 5 courses of chemotherapy combined with iodized oil; Group C (n = 23) who received i to 3 courses of chemotherapy combined with iodized oil and gelatin sponge; and Group D (n = 12) who received 1 to 3 courses of chemotherapy combined with iodized oil, ethanol and gelatin sponge. The other 57 patients only received liver resection (non- TACE group). The expressions of Ki-67 and proliferating cell nuclear antigen (PCNA) protein were detected in the liver cancer tissues by immunohistochemical method. RESULTS: The Ki-67 protein expression was significantly lower in Groups C and D as compared with non-TACE group (31.35% ± 10.85% vs 44.43% ± 20.70%, 30.93% ± 18.10% vs 44.43% ± 20.70%, respectively, P 〈 0.05). The PCNA protein expression was significantly lower in Groups C and D as compared with non-TACE group (49.61% ± 15.11% vs 62.92% ± 17.21%, 41.16% ± 11.83% vs 62.92% ± 17.21%, respectively, P 〈 0.05). The Ki-67 protein expression was significantly higher in Group A as compared with non-TACE group (55.44% ± 13.72% vs 44.43% ± 20.70%, P 〈 0.05). The PCNA protein expression was significantly higher in Groups A and B as compared with non-TACE group (72.22% ± 8.71% vs 62.92% ± 17.21%, 69.91% ± 13.38% vs 62.92% ± 17.21%, respectively, P 〈0.05). CONCLUSION: Preoperative multi-material TACE suppresses the proliferation of HCC cells, while a single material embolization and chemotherapy alone enhance the proliferation of HCC cells. 展开更多

To investigate the effects of anti-sense peptide nucleic acids (PNAs) targeting Ki-67 gene on modulation of the proliferation and apoptosis of human renal carcinoma cell lines, human renal carcinoma cell line 786-0 cells were treated with anti-sense PNAs at different concentrations (1.0 μmol/L, 2.0 μmol/L, 10.0 μmol/L). The Ki-67 expression of 786-0 cells was detected by immunohistochemical technique and Western blot method respectively. The proliferation of 786-0 cells was studied by cell growth curves and ^3H-thymidine incorporation. The apoptosis of 786-0 cells was detected by TUNEL assay. The control groups were treated with anti-sense oligonucleotide (ASODNs) targeting Ki-67 gene. Our results showed that the Ki-67 expression of 786-0 cells treated with anti-sense PNAs (16.9±0.7) was significantly inhibited as compared with that of the control groups (28.6±0.4) (P〈0.01). The Ki-67 protein rate of 786-0 cells treated with anti-sense PNAs (42.1 ±2.2) was significantly reduced when compared with that of the control groups (83.6± 1.4) (P〈0.01). Proliferation of 786-0 cells treated with anti-sense PNAs (20.7 ± 1.5) was significantly inhibited as compared with that of the control groups (58.6± 1.4) (P〈0.01). The apoptosis rate of 786-0 cells treated with anti-sense PNAs (28.7 ± 2.3) was significantly increased higher compared with that of the control groups (13.8 ±1.0) (P〈0.01). From these finds we are led to conclude that anti-sense PNAs targeting Ki-67 gene have stronger effects on the inhibition of the proliferation and induction of apoptosis of human renal carcinoma cells than ASODNs targeting Ki-67 gene. The strategies using anti-sense PNAs targeting Ki-67 gene may be a promising approach for the treatment of renal cell carcinoma. 展开更多

Melanin, synthesized by melanocyte, is transferred to neighboring keratinocyte and finally accumulates in perinuclear site. Except functioning as an internal sunscreen to protect from UV damage, the potential effect of melanin on modulating the bioactivity of keratinocyte has not yet been fully investigated. In this study, we added melanin directly to the culture of human epidermal keratinocytes and the uptake of melanin was found to be dose- and time-dependent as determined by spectrophotometric method. The uptaken melanin accumulated perinuclearly in keratinocytes that is similar to the pattern observed in human solar lentigo tissue by microscopic examination. Pretreatment of keratinocytes with either niacinamide or trypsin inhibitor reduced the uptake of melanin dose-dependently, indicating a PAR-2-dependent pathway involved. Melanin uptake by keratinocytes inhibited cell proliferation as demonstrated both by the decrease of cell number and nuclear Ki-67 expression. Inhibited Ki-67 expression in melanin-containing keratinocyte was also found in human lentigo tissue. The cell cycle arrested at G1 phase in melanin-uptaken keratinocytes was confirmed by flow cytometric method. The protein expressions of cyclin-dependent kinase 1 (CDK1), CDK2, cyclin E, cyclin A and cyclin B were significantly reduced by melanin treatment. Microarray analysis, RT/real-time PCR and western blot demonstrated the inhibited expression of DKK1, a protein known to reduce skin pigmentation, in melanin-uptaken keratinocytes. Together, the direct incubation of keratinocyte with melanin might serve as a useful model to study the potential mechanisms involved in melanin uptake and pigmentation process. 展开更多

目的:探讨超声造影定量参数在Luminal型乳腺癌诊断中的价值,分析Luminal型乳腺癌患者超声造影定量参数与其雄激素受体(AR)及肿瘤增殖细胞相关抗原Ki-67的相关性。方法:选取医院收治的90例Luminal型乳腺癌患者,并根据病理及免疫组化检测... 目的:探讨超声造影定量参数在Luminal型乳腺癌诊断中的价值,分析Luminal型乳腺癌患者超声造影定量参数与其雄激素受体(AR)及肿瘤增殖细胞相关抗原Ki-67的相关性。方法:选取医院收治的90例Luminal型乳腺癌患者,并根据病理及免疫组化检测结果将其分为LuminalA型组(32例)和LuminalB型组(58例),比较两组患者超声造影表现差异及定量参数,同时比较定量参数与Ki-67及AR表达水平的相关性。结果:两组患者的超声造影增强强度、灌注缺损、增强边界及穿入血流表现比较,差异均有统计学意义(x^(2)=5.204,x^(2)=11.880,x^(2)=6.556,x^(2)=7.249;P<0.05)。LuminalB型组患者与LuminalA型组相比,病灶区域血流灌注的峰值强度(PI)上升,达峰时间(TTP)及平均渡越时间(TOF)下降,差异均有统计学意义(t=8.084,t=8.639,t=7.945;P<0.05)。LuminalA型组患者Ki-67及AR阳性检出率低于LuminalB型组,差异有统计学意义(x^(2)=4.224,x^(2)=4.262;P<0.05)。90例Luminal型乳腺癌患者PI与Ki-67和AR表达水平均呈正相关(r=0.159,r=0.657;P<0.05);TTP、TOF与Ki-67和AR表达水平均呈负相关(r=-0.537,r=-0.407;r=-0.503,r=-0.023;P<0.05)。结论:超声造影定量参数对Luminal型乳腺癌的临床诊断价值较高,并与AR、Ki-67具有一定的相关性,可用于对Luminal型乳腺癌状况变化的诊断评估。 展开更多

目的:探讨细胞周期蛋白D1(Cyclin D1)、细胞增殖抗原-67(Ki-67)、细胞角蛋白19(CK19)联合评估甲状腺乳头状癌(PTC)的效能,分析三者与颈部淋巴结转移的相关性。方法:选取我院2017年9月至2021年9月收治的91例PTC患者为PTC组,取同期收治的... 目的:探讨细胞周期蛋白D1(Cyclin D1)、细胞增殖抗原-67(Ki-67)、细胞角蛋白19(CK19)联合评估甲状腺乳头状癌(PTC)的效能,分析三者与颈部淋巴结转移的相关性。方法:选取我院2017年9月至2021年9月收治的91例PTC患者为PTC组,取同期收治的结节性甲状腺肿患者52例为对照组。两组均通过手术获取病变组织,检测Cyclin D1、Ki-67、CK19表达情况,分析三者对PTC的诊断价值。根据PTC患者有无颈部淋巴结转移,分成转移组、非转移组,比较两组病理组织中Cyclin D1、Ki-67、CK19表达情况,经多元Logistic回归模型分析三者与PTC患者颈部淋巴结转移的关系。结果:PTC组Cyclin D1、CK19阳性表达率分别为83.52%、85.71%,高于对照组的17.31%、17.31%(P<0.05);PTC组Ki-67>1%占比为93.41%,高于对照组的13.46%(P<0.05)。Cyclin D1、Ki-67、CK19单独诊断PTC的敏感度分别为83.52%、93.41%、85.71%,特异度分别为82.69%、86.54%、82.69%,三者联合诊断的敏感度、特异度分别为91.21%、96.15%。转移组的Cyclin D1、CK19阳性表达率分别为97.06%、97.06%,高于非转移组的75.44%、78.95%(P<0.05)。多元Logistic回归分析显示,Cyclin D1阳性表达会增加PTC患者颈部淋巴结转移风险(OR:3.446,95%CI:1.041~11.407,P<0.05)。结论:PTC患者的Cyclin D1、Ki-67、CK19阳性表达率较高,三者联合对PTC有较好的诊断效能,其中Cyclin D1阳性表达与颈部淋巴结转移相关。 展开更多

目的 观察子宫内膜癌(EC)组织中增殖细胞核抗原-67(Ki-67)、抑癌基因p16蛋白表达,分析EC组织中Ki-67、p16蛋白表达对手术治疗预后的评估价值。方法 选择137例实施手术治疗的EC患者作为研究对象,收集患者的EC组织,检测Ki-67、p16在EC组... 目的 观察子宫内膜癌(EC)组织中增殖细胞核抗原-67(Ki-67)、抑癌基因p16蛋白表达,分析EC组织中Ki-67、p16蛋白表达对手术治疗预后的评估价值。方法 选择137例实施手术治疗的EC患者作为研究对象,收集患者的EC组织,检测Ki-67、p16在EC组织中的表达,随访2年,评估患者预后(复发、转移发生)情况,比较不同预后EC患者基线资料、实验室指标,重点分析EC患者组织中Ki-67、p16蛋白表达对手术治疗预后的预测。结果 随访2年,137例EC患者中17例复发,4例转移,预后不良发生率为15.33%(21/137)。经Logistic回归分析结果显示,国际妇产科协会(FIGO)分期高、Ki-67表达高可能是EC患者淋巴结转移的风险因子(OR>1,P<0.05),p16表达高可能是EC患者手术治疗预后的保护因子(OR<1,P<0.05)。绘制ROC曲线图,结果显示,Ki-67、p16蛋白表达预测EC患者手术治疗预后不良的AUC分别为0.833、0.827,均有一定预测价值;绘制决策曲线结果显示,在阈值0.2~1.0范围内,联合子宫内膜癌患者组织中Ki-67、p16蛋白表达预测EC患者手术治疗预后不良的净受益率优于单独某一指标,且在高风险阈值0.0~1.0内的净受益率始终大于0,始终有临床意义,净受益率最大值为0.153。结论 子宫内膜癌组织中Ki-67、p16蛋白表达与手术治疗预后有关,用于预测预后不良具有一定的价值,且联合预测获得的净受益率更高。 展开更多
