Pyrogallol(pyrogallic acid)is easy to be autooxidized,forming a free radical under alkaline condition,but the mechanism has not been clear.The mechanism of the autooxidation of pyrogallol was studied with ESR,UV and NMR techniques and it was found that pyrogallol was autooxidized to a quinone radical under alkaline condition.And at the same time oxygen recieved an electron from the reaction to form superoxide anions,the hydroxyl free radical was then produced by HaberWeiss reaction.The oxygen free radicals generated from this process promoted the autooxidation of pyrogallol. 展开更多

In this study, a new sensor b... Isoproterenol (ISPR) is an important catecholamine‐based drug that is widely used in the treatment of heart disease. However, overdose of this drug is very dangerous to the human body. In this study, a new sensor based on a pyrogallol red modified‐multiwalled carbon nanotube paste electrode (PGRMMWCNTPE) was prepared and used for high sensitivity determination of ISPR in aqueous solution. Electrocatalytic oxidation of ISPR at the PGRMMWCNTPE was investigated by chronoam‐perometry, cyclic voltammetry, and square‐wave voltammetry. The values of the catalytic rate con‐stant, electron transfer coefficient, and diffusion coefficient for ISPR oxidation were then calculated using voltammetric data. A linear calibration curve was constructed for ISPR concentration in the range 0.8–570μmol/L with a detection limit of 0.47μmol/L ISPR. The sensor was then applied to the determination of ISPR in urine and drug samples with satisfactory results. 展开更多

The interactions of pyrogallol red (PR) with proteins and a derivative method for the determination of proteins were proposed. In the pH range 2 56-4 10, PR interacts with proteins, including bovine serum albumin, human serum albumin, lysozyme, chymotrypsin, resulting in enhanced resonance light scattering(RLS) with the maximal peak at 360 0 nm. With the enhanced RLS, proteins in the range 0-5 0(μg/mL can be determined with the determination limits between 10 3 and 32 2 ng/mL depending on different protein. The effects of foreign substances were tested. The method has been applied to the determination of protein in the synthetic samples and human urine with satisfaction results. 展开更多

研究焦棓酸对猪链球菌的体外抗菌活性及对生物被膜的抑制作用,并初步探讨其抑菌机制。采用96孔微量稀释法测定常用抗菌药物及焦棓酸对猪链球菌的最小抑菌浓度和最小杀菌浓度;平板活菌计数法和分光光度计测定焦棓酸对猪链球菌作用的时间... 研究焦棓酸对猪链球菌的体外抗菌活性及对生物被膜的抑制作用,并初步探讨其抑菌机制。采用96孔微量稀释法测定常用抗菌药物及焦棓酸对猪链球菌的最小抑菌浓度和最小杀菌浓度;平板活菌计数法和分光光度计测定焦棓酸对猪链球菌作用的时间-杀菌曲线;采用结晶紫染色法检测焦棓酸对猪链球菌生物被膜形成的影响;通过荧光定量PCR从转录水平分析焦棓酸对猪链球菌生长与生物被膜形成相关基因表达的情况。结果显示,猪链球菌2型HB菌株对磺胺异恶唑、磺胺甲恶唑耐药严重,焦棓酸对猪链球菌2型HB菌株的最小抑菌浓度为32μg/mL,最小杀菌浓度为64μg/mL,并且焦棓酸对猪链球菌的抑制作用呈剂量依赖性;焦棓酸在16μg/mL能显著抑制猪链球菌2型HB菌株生物被膜形成的能力;另外,经焦棓酸处理后,发现猪链球菌的生长、生物被膜形成、毒力相关基因ftsZ、stK、mrp、fbps、gapdh、ccpA、luxS下调表达。综上所述,焦棓酸可为耐药猪链球菌在临床上感染的治疗提供新思路。 展开更多

A novel and facile seed-mediated method for the preparation of monodispersed gold nanorods(GNRs) is presented by introducing pyrogallol as a reductant. Fast Fourier transformation of high-resolution transmission electron microscopy reveals that the synthesized GNRs are single crystalline. The longitudinal surface plasmon resonance of GNRs can be finely tuned by varying silver ion concentrations or seed amounts. Also, both thick(diameter >30 nm) and thin(diameter <10 nm) GNRs with exceptional monodispersity can be well prepared by this method. These findings indicate that this method has a greater performance in controlling the morphology of GNRs than that of traditional approach with ascorbic acid as a reductant. 展开更多

The controlled self-assembly of discrete metal-organic nanocapsules(MONCs),and metal-organic frameworks(MOFs)based on the MONCs are achieved.Specifically,the solvothermal reaction of nickel nitrate hexahydrate and C-methylpyrogallol[4]arene in mixed DMF/MeOH solution leads to the unexpected form of discrete nickel-seamed hexameric pyrogallol[4]arene MONCs,and MONC-based three-dimensional(3D)MOF.Notably,the latter MOF is constructed from the aforementioned nickelseamed MONC building blocks and formate linkers in-situ generated from the hydrolysis of DMF solve nt.Interestingly,introducing pyridine and formic acid in the reaction conditions leads to the controlled assemblies of the discrete MONC and MONC-based 3D MOF structures.Moreover,the variabletemperature magnetic susceptibilities of both the abovementioned compounds have been investigated,indicating typical antiferromagnetic interactions between the metal centers. 展开更多

The roasting process of pyrogallol,a polyphenol compound distributed in coffee beverages,significantly enhanced itsα-glucosidase inhibitory activity.In this study,a bioassay-guided isolation of the thermal reaction products of pyrogallol led to the identification of two potentα-glucosidase inhibitors,4-4′dimer of pyrogallol(4,4′-DP)and 4-5′dimer of pyrogallol(4,5′-DP).Theirα-glucosidase inhibitory activity was higher than that of pyrogallol,as evidenced by comparing the IC_(50)values(206.2±1.2μM for 4,4′-DP,187.6±2.6μM for 4,5′-DP,2660±60.1μM for pyrogallol).And the roasting products were more potentα-glucosidase inhibitors compared to acarbose(IC_(50)=695±12.7μM).Enzyme kinetics demonstrated that 4,4′-DP and 4,5′-DP inhibitedα-glucosidase in an uncompetitive and a non-competitive manner,respectively.Docking simulations revealed that the main interaction forces between these two compounds andα-glucosidase were hydrogen bonding and hydrophobic effect.These results suggested that a simple roasting process might increase theα-glucosidase inhibitory activity of pyrogallol-containing foods such as coffee beverages. 展开更多

The content of flavones of the seabuckthorn ( Hippophae rhamnoides L. )root bark,branch bark and leaves were determined using UV vis spectrophotometry.In the meantime,the scavenging hydroxyl free radical(OH ·)and superoxide anion free radical(O 2 ·- )of the extracts of seabuckthorn root bark,branch bark and leaves were measured through Fenton reaction and pyrogallol autoxidation method.The results showed that the seabuckthorn root bark,branch bark and leaves had a strong capacity on scavenging free radical.The scavenging rates of hydroxyl free radical were 69.2%,62.2% and 74.1%,respectively.The inhibition rates of superoxide anion free radical were 10.5%,12.3% and 33.3%,respectively.Finally the antioxidant mechanism of seabuckthorn flavones were tentatively explored. 展开更多

目的比较邻苯三酚自氧化法和氮蓝四唑(NBT)光还原法分别测定超氧化物歧化酶(SOD)酶活力的差异。方法采用两种方法分别测定SOD标准品、苜蓿和番茄两种植物提取物的不同来源SOD酶活性,比较其精确性和灵敏度。结果在标准品SOD酶活的测定中... 目的比较邻苯三酚自氧化法和氮蓝四唑(NBT)光还原法分别测定超氧化物歧化酶(SOD)酶活力的差异。方法采用两种方法分别测定SOD标准品、苜蓿和番茄两种植物提取物的不同来源SOD酶活性,比较其精确性和灵敏度。结果在标准品SOD酶活的测定中,邻苯三酚自氧化法和NBT光还原法测酶活性的变异系数分别为0.84、2.03,而进样量后者是前者的2.6倍,稀释倍数是前者的6倍,邻苯三酚自氧化法的精确度高于NBT光还原法,但灵敏度远不及后者。在测定两种植物SOD时,邻苯三酚自氧化法测得酶活性变异系数是NBT光还原反应测得酶活性变异系数的1.30~1.47倍,NBT光还原法具有稳定性、重现性好的特点,优于邻苯三酚自氧化法。结论 NBT光还原法是测定植物SOD及SOD标准品酶活的理想检测方法。 展开更多

目的与方法:通过醇提得到各种茶多酚提取液,酒石酸亚铁法测定了多酚的含量;水提得到各种茶多糖提取液,用硫酸-苯酚法测定其中多糖的含量。通过DPPH·法、水杨酸法及邻苯三酚法测定不同茶叶中多酚、多糖含量对自由基的清除作用的影响... 目的与方法:通过醇提得到各种茶多酚提取液,酒石酸亚铁法测定了多酚的含量;水提得到各种茶多糖提取液,用硫酸-苯酚法测定其中多糖的含量。通过DPPH·法、水杨酸法及邻苯三酚法测定不同茶叶中多酚、多糖含量对自由基的清除作用的影响,对比研究八种茶叶的抗氧化能力。结果:不同茶叶中多糖的含量从2.19%增大到2.89%(半发酵乌龙茶>红茶>绿茶),抗氧化能力:红茶>绿茶>乌龙茶(DPPH·法);红茶>绿茶>乌龙茶(水杨酸法);红茶>乌龙茶>绿茶(邻苯三酚法);不同茶叶中多酚的含量从1.71%增大到9.75%(绿茶>半发酵乌龙茶>红茶);结论:茶多糖和多酚是茶叶的主要抗氧化成分。绿茶的抗氧化作用主要来自茶多酚,红茶的抗氧化作用主要来自茶多糖。除乌龙茶多糖提取液外,两种抗氧化成分的抗氧化能力与其含量成量效关系。 展开更多

分别用DPPH法、邻苯三酚自氧化法测定了赤灵芝和黑灵芝提取物的抗氧化作用。结果表明:不同溶剂灵芝提取物对DPPH自由基均有清除作用,对邻苯三酚自氧化也均有抑制作用。不同溶剂提取物的抗氧化活性强弱为:95%乙醇>丙酮>去离子水>... 分别用DPPH法、邻苯三酚自氧化法测定了赤灵芝和黑灵芝提取物的抗氧化作用。结果表明:不同溶剂灵芝提取物对DPPH自由基均有清除作用,对邻苯三酚自氧化也均有抑制作用。不同溶剂提取物的抗氧化活性强弱为:95%乙醇>丙酮>去离子水>氯仿,提取物用量与抗氧化性成正相关,而黑灵芝的抗氧化作用相对赤灵芝要强。同BHT和抗坏血酸进行了比较,黑灵芝的丙酮提取物浓度达到1mg/ml时已超过10mg/mlBHT的抗氧化效果,相当于0.5mg/ml浓度抗坏血酸的抗氧化效果。 展开更多

The ability of water soluble β alanine C 60 adducts for superoxide anion radical scavenging has been studied by spectrophotometry and the modified pyrogallol auto oxidation method. It is found that β alanine C 60 adducts have an excellent efficiency in eliminating superoxide radical generated from auto oxidation of pyrogallol. At a mass concentration level of 150 μg/mL for β alanine C 60 adducts, the radical scavenging efficiency is approximately equivalent to one enzyme unit, in vitro experiments show that β alanine C 60 adducts can enhance the activity of mouse thymus cells and prolong their life time span. It reveals the potential use of these β alanine C 60 adducts as the novel potent free radical scavengers in biological systems. 展开更多
