Iguratimod is a novel anti-rheumatic drug which is reported to have the capability of anti-cytokines. In this study, a rat model was constructed to investigate the effect of iguratimod on bone metastasis and it was found that iguratimod alleviated cancer-induced bone destruction. To further explore whether an anti-tumor activity of iguratimod contributes to the effect of bone resorption suppression, two human breast cancer cell lines MDA-MB-231 and MCF-7 were studied. The effect of iguratimod on tumor proliferation was detected by CCK-8 assay and flow cytometry. The effects of iguratimod on migration and invasion of cancer cells were determined by wound-healing and Transwell assays. Results showed that high dose(30 μg/m L) iguratimod slightly suppressed the proliferation of cancer cells but failed to inhibit their migration and invasion capacity. Interestingly, iguratimod decreased the transcription level of IL-6 in MDA-MB-231 cells in a concentration-dependent manner. Moreover, iguratimod partially impaired NF-κB signaling by suppressing the phosphorylation of NF-κB p65 subunit. Our findings indicated that iguratimod may alleviate bone destruction by partially decreasing the expression of IL-6 in an NF-κB-dependent manner, while it has little effect on the tumor proliferation and invasion.展开更多
This paper introduces a Takagi-Sugeno(T-S)fuzzy regulator design using the negative absolute eigenvalue(NAE)approach for a class of nonlinear and unstable systems.The open-loop system is initially embodied by the trad...This paper introduces a Takagi-Sugeno(T-S)fuzzy regulator design using the negative absolute eigenvalue(NAE)approach for a class of nonlinear and unstable systems.The open-loop system is initially embodied by the traditional T-S fuzzy model and then,all closed-loop subsystems are combined using the proposed Max-Min operator in place of traditional weighted average operator from the controller side to lessen the coupling virtually and simplify the proposed regulator design.For each virtually decoupled closed-loop subsystem,the composite regulators(i.e.,primary and secondary regulators)are designed by the NAE approach based on the enhanced eigenvalue analysis.The Lyapunov function is utilized to guarantee the asymptotic stability of the overall T-S fuzzy control system.The most popular and widely used nonlinear and unstable systems like the electromagnetic levitation system(EMLS)and the inverted cart pendulum(ICP)are simulated for the wide range of the initial conditions and the enormous variation in the disturbance.The transient and steady-state performance of the considered systems using the proposed design are analyzed in terms of the decay rate,settling time and integral errors as IAE,ISE,ITAE,and ITSE to validate the effectiveness of the proposed approach compared to the most popular and traditional parallel distributed compensation(PDC)approach.展开更多
Grain number per panicle(GNP)is an important agronomic trait that contributes to rice grain yield.Despite its importance in rice breeding,the molecular mechanism underlying GNP regulation remains largely unknown.In th...Grain number per panicle(GNP)is an important agronomic trait that contributes to rice grain yield.Despite its importance in rice breeding,the molecular mechanism underlying GNP regulation remains largely unknown.In this study,we identified a previously unrecognized regulatory gene that controls GNP in rice,Oryza sativa REPRODUCTIVE MERISTEM 20(OsREM20),which encodes a B3 domain transcription factor.Through genetic analysis and transgenic validation we found that genetic variation in the CArG box-containing inverted repeat(IR)sequence of the OsREM20 promoter alters its expression level and contributes to GNP variation among rice varieties.Furthermore,we revealed that the IR sequence regulates OsREM20 expression by affecting the direct binding of OsMADS34 to the CArG box within the IR sequence.Interestingly,the divergent pOsREM20IR and pOsREM20ΔIR alleles were found to originate from different Oryza rufipogon accessions,and were independently inherited into the japonica and indica subspecies,respectively,during domestication.Importantly,we demonstrated that IR sequence variations in the OsREM20 promoter can be utilized for germplasm improvement through either genome editing or traditional breeding.Taken together,our study characterizes novel genetic variations responsible for GNP diversity in rice,reveals the underlying molecular mechanism in the regulation of agronomically important gene expression,and provides a promising strategy for improving rice production by manipulating the cis-regulatory element-containing IR sequence.展开更多
A fuzzy robust path tracking strategy of an active pelagic trawl system with ship and winch regulation is proposed.First,nonlinear mathematic model of the pelagic trawl system was derived using Lagrange equation and f...A fuzzy robust path tracking strategy of an active pelagic trawl system with ship and winch regulation is proposed.First,nonlinear mathematic model of the pelagic trawl system was derived using Lagrange equation and further simplified as a low order model for the convenience of controller design.Then,an active path tracking strategy of pelagic trawl system was investigated to improve the catching efficiency of the target fish near the sea bottom.By means of the active tracking control,the pelagic trawl net can be positioned dynamically to follow a specified trajectory via the coordinated winch and ship regulation.In addition,considering the system nonlinearities,modeling uncertainties and the unknown exogenous disturbance of the trawl system model,a nonlinear robust H2 /H∞ controller based on Takagi-Sugeno(T-S) fuzzy model was presented,and the simulation comparison with linear robust H2 /H∞ controller and PID method was conducted for the validation of the nonlinear fuzzy robust controller.The nonlinear simulation results show that the average tracking error is 0.4 m for the fuzzy robust H2 /H∞ control and 125.8 m for the vertical and horizontal displacement,respectively,which is much smaller than linear H2 /H∞ controller and the PID controller.The investigation results illustrate that the fuzzy robust controller is effective for the active path tracking control of the pelagic trawl system.展开更多
